Abstract:A novel electrochemical sensor with capability of probing chiral amino acids with gold nanoparticle (n-Au) labels using bovine serum albumin (BSA) as a chiral selector and subsequent signal amplification step by silver enhancement is introduced. The assay relies on the stereoselectivity of BSA embedded in ultrathin gamma-alumina sol-gel film coated on the surface of the glassy carbon electrode (GCE). The recognition to the n-Au-labeled L- or D-amino acids for BSA-GCE could be monitored by the differential pulse voltammetry (DPV), while the DPV signal was greatly amplified by the anchored silver atoms on the n-Au, leading to a new way of quantitatively analysis of chiral amino acids electrochemically at sub-picomolar level. With L-tryptophan as the probe solute, the linear concentration range was from 1.33 x 10(-12) to 1 x 10(-9)mol L(-1) and detection limit was 5 x 10(-13)mol L(-1). For tryptophan enantiomers, the enantioselectivity coefficient 2.3 was obtained.
Optically Active Ultrafine Au-Ag Alloy Nanoparticles Used for Colorimetric Chiral Recognition and Circular Dichroism Sensing of Enantiomers

Abstract:Despite a significant surge in the number of investigations into chirality at the nanoscale, especially thiolated chiral molecules capping gold clusters, only limited knowledge is currently available to elaborate the alloying effect on chiroptical behavior of bimetallic nanoparticles (NPs). Also, few successful cases as to the efforts toward the development of chirality-dependent applications on the optically active nanomaterial have been made. Herein, as a positive test case for chiral alloy nanoparticle synthesis, the stable and large chiroptical ultrafine Au-Ag alloy NPs were prepared by reduction of different molar fractions of HAuCl4 and AgNO3 with NaBH4 in the presence of d/l-penicillamine (d/l-Pen). Compared with those of monometallic Au and Ag counterparts with comparable size, the Au-Ag alloy NPs (Ag mole fraction, 70%) obviously displayed the largest optical activities with the maximum g-factors of ∼1.6 × 10-3. Impressively, the Pen-mediated synthesis of chiral Au-Ag alloy NPs possesses a colorimetric self-recognition function and can be used as an incisive circular dichroism (CD) probe toward d- and l-Pen enantiomers. The plasmonic CD signal amplification (ΔICD) shows good linearity with the amount of Pen over the range of 5.0-80.0 μM with a detection limit (3σ) of 1.7 μM for l-Pen and 1.5 μM for d-Pen, respectively. In addition, the sensing system exhibits good selectivity toward d- and l-Pen in the presence of other enantiomers; therefore, it is highly expected that the approach described here would open new opportunities for design of more novel enantioselective analyses of important species related to biological processes.

Abstract:Chiral recognition of enantiomers with identical mirror-symmetric molecular structures is important for the analysis of biomolecules, and it conventionally relies on stereoselective interactions in chiral chemical environments. Here, we develop a magneto-electrochemical method for the enhanced detection of chiral amino acids (AAs), that combines the advantages of the high sensitivity of electrochemiluminescence (ECL) biosensors and chirality-induced effects under magnetic field. The ECL difference between L- and D-enantiomers can be amplified by over 35-fold under a field of 3.5 kG, and the chiral discrimination can be achieved in dilute AA solutions down to nM level. The field-dependent ECL and chronocoulometry measurements suggest that chiral AAs can lock the spins on their radicals and thus enlarge the ECL change under applied magnetic field (magneto-ECL, MECL), which explains the field-enhanced chiral discrimination of AA enantiomers. Finally, a detailed protocol is demonstrated for the identification of unknown AA solutions, in which the species, chirality and concentration of AAs can be determined simultaneously from the 2D plots of the ECL and MECL results. This work benefits the development of field-assisted detection methods and represents a promising and universal strategy for the comprehensive analysis of chiral biomolecules.

Abstract:In this work, we prepared gold nanoparticles (AuNPs) by employing gluconic acid (GlcA) as reducing-cum-stabilizing agent. The proposed GlcA-AuNPs successfully worked as a colorimetric sensor for visual chiral recognition of aromatic amino acid enantiomers, namely tyrosine (d/l-Tyr), phenylalanine (d/l-Phe), and tryptophan (d/l-Trp). After adding L-types to GlcA-AuNPs solution, the color of the mixture changed from red to purple (or gray), while no obvious color change occurred on the addition of D-types. The effect can be detected by naked eyes. The particles have been characterized by transmission electron microscopy, Fourier-transform infrared spectroscopy, zeta potential, the dynamic light scattering analysis as well as UV–Vis spectroscopy. This assay can be used to determine the enantiomeric excess of l-Trp in the range from 0 to + 100%. The method has advantages in simplicity, sensitivity, fast response, and low cost.

A Visual Chiroptical System with Chiral Assembly Graphene Quantum Dots for D-phenylalanine Detection

Abstract:Chirality is a fundamental phenomenon of nature, and the enantioselective recognition of amino acids isomers is especially important for life science. In this study, chiroptical system based on chiral assembly graphene quantum dots (GQDs) was developed for visual testing of D-phenylalanine (D-Phe). Here, GQDs were used as the fluorescent element, and chiral functional moieties of 1,3,5-triformylphloroglucinol-functionalized chiral ( +)-diacetyl-L-tartaric anhydride (TPTA) were used as the chiral recognition elements. Based on the formed chiral microenvironment, the fluorescence intensity of TPTA-assembled GQDs had a good linear relationship with D-Phe in the concentration range of 0.1–5 μM, and the detection limit was 0.023 μM. According to the variation in luminance of TPTA-assembled GQDs, visual testing to D-Phe was realized using a smartphone-assisted chiroptical system with a detection limit of 0.050 μM. The spiked recoveries of both chiroptical sensing methods based on TPTA-assembled GQDs from the food matrix ranged from 86.20 to 110.0%. Furthermore, TPTA-assembled GQDs were successfully applied to intracellular chiroptical imaging in response to D-Phe in vitro. The developed chiral nanomaterial TPTA-assembled GQDs with excellent photochemical stability, optical properties, and bioimaging capabilities provide a promising technique for the visual detection of amino acid isomers in the field of smart devices.

Abstract:The development of chiral analytical methods for tyrosine enantiomers, characterized by superior recognition performance and selectivity, holds significant potential for application in the biochemical and biomedical domains associated with tyrosine. Herein, a straightforward and effective strategy for tyrosine enantiomers identification and purity analysis by chiral carbon dots (CDs) fluorescence sensor were reported. The chiral CDs were designed by using L-cysteine and 2,4-diaminophenol hydrochloride through a gentle one-step synthesis strategy at room temperature. The resulting chiral CDs exhibited completely different fluorescence response to L-tyrosine and D-tyrosine, that is, L-tyrosine could enhance the fluorescence of the chiral CDs, but D-tyrosine made no difference. The fluorescent differential recognition factor for enantiomers was as high as 23.2, which was the highest be achievement to our best knowledge so far. The varying discriminatory capabilities of chiral CDs towards tyrosine enantiomers were also applicable to HeLa cells. To confirm the recognition mechanism of chiral CDs to tyrosine enantiomers, the chiral CDs-modified SiO 2 stationary phase were synthesized and applied to separate the tyrosine enantiomers in reverse-phase HPLC mode. Furthermore, density functional theory was combined with the fluorescence spectroscopy and chromatographic results to verify that the recognition ability was related to the difference of weak interaction energy and Gibbs free energy between the precursor molecule of the chiral CDs and the tyrosine enantiomers. This cost-effective and highly selectivity assay not only provided a valuable tool for tyrosine enantiomers identification and purity analysis, but also delivered an important reference for the recognition and purity identification of other chiral drugs.

Abstract:Ultrasensitive determination of the chiral nature of a molecule is important for fully understanding protein and DNA structures, and for the design, synthesis and purification of drug molecules. Consequently, development of ultrasensitive detection of chiral molecules is a grand challenge and at the heart of many of today’s research fields, including chemistry, biology, biotechnology and pharmacology. Here a novel method afforded by electrochemically tracing the Cu2+ ions that are replaced by L/D-carnitine on anL/D-cysteine modified gold electrode is described. The electrochemical signal of copper ionswas monitored, these were immobilized on the L/D-cysteine functionalized gold electrode surface via cooperative metal-ligand interaction, followed by replacing the Cu2+ with L-carnitine or D-carnitine chiral molecules based on the hetero- or homo- chiral interaction between carnitine and cysteine molecules. The results illustrated that the Cu2+ replacement ratio between L/D-carnitine and D/L-cysteine were higher than that between L/D-carnitine and L/D-cysteine, which is consistent with previous studies that hetero-chiral interaction is stronger than homo-chiral interaction. This study demonstrates a facile, but innovative electrochemical method to probe the chiral recognition of carnitine molecules.

Abstract:The process of hydrogen plasma reacting with a Si(110) surface is investigated. The reaction process is measured with in situ infrared absorption spectroscopy (IRAS) in multiple internal reflection geometry. We monitor the evolution of hydride components in the Si(110) surface exposed to the hydrogen plasma. IRAS data show that the surface atomic arrangement of a Si(110) surface is distorted by plasma exposure. H is inserted into Si crystal and defects and vacancies are generated. Further, hydrogen plasma exposure creates silicon dihydride <formula formulatype="inline"><tex Notation="TeX">$({\rm SiH}_{2})$</tex></formula> components in an amorphous layer. The <formula formulatype="inline"><tex Notation="TeX">${\rm SiH}_{2}$</tex></formula> formation rate is 0.5, by a comparison with the hydrogen plasma exposure time.

Abstract:Chiral recognition of essential amino acids (EAAs) is a huge challenge that keeps plaguing analytical scientists due to their cryptochirality and limited steric interaction sites. Inspired by the superior enantioselectivity of functional supramolecular cyclodextrins (CDs) and strong signal amplification ability of field effect transistors (FETs), this work firstly reports a cationic supramolecular charge switch for facile enantiodiscrimination of EAAs based on extended-gate organic FET (EG-OFET). The cationic phenylcarbamoylated-CD single isomer acts as a charge switch via interacting with different enantiomers and the weak stereo-differentiation intermolecular interaction signals between the cationic perphenylcarbamoylated CDs and EAAs on the EG can be strongly and rapidly amplified through an OFET. Efficient chiral differentiation of six EAAs, including phenylalanine, tryptophan, leucine, isoleucine, lysine and valine, are successfully achieved without any derivation process and the detection limit for d-phenylalanine is down to 10−13 mol/L. We believe that this study provides a new and facile sensing perspective for natural amino acids and may afford deeper understanding of molecular chirality.

Abstract:A novel electrochemical chiral sensor was constructed for the discrimination of Tryptophan (Trp) by the modification of Au@Pt bimetallic nanoparticles, poly(cysteine) (PL-Cys), and Cu2+-modified β-cyclodextrin (Cu2-β-CD) on the surface of glassy carbon electrode. The constructed sensor was characterized by transmission electron microscopy, infrared spectroscopy, UV–vis adsorption spectra, and the electrochemical experiments. The modified Au@Pt endowed the sensor with high surface area and good conductivity. The electropolymerized PL-Cys facilitated the loading of Cu2-β-CD based on the electrostatic interaction. As a chiral selector, Cu2-β-CD enabled the sensor to effectively identify the Trp enantiomers based on the host–guest interaction with a high the oxidation peak current ratio (IL/ID, IL and ID were the oxidation peak currents of l-Trp and d-Trp measured on the sensor, respectively) of 2.38. Under the optimized conditions, IL/ID increased with the increase of l-Trp in the raceme solution, which was further employed for the detection of Trp in serum samples with satisfactory results. With the good selectivity, stability, and reproducibility, the present electrochemical sensor can be used for the simple, rapid, and efficient chiral recognition and real samples determination.

Dipeptide-Capped Copper Nanoparticles as Chiral Nanozymes for Colorimetric Enantioselective Recognition of 3,4-Dihydroxy-d,l-phenylalanine

Abstract:In pharmaceutical and biomedical applications, it is imperative to identify chiral molecules. However, colorimetric sensing enantiomers relying on chiral nanozymes is still a major challenge in chirality recognition. Herein, we report a facile and simple strategy to prepare copper nanoparticles (CuNPs) using d-cysteine-d-histidine (DCDH), d-cysteine-l-histidine, and l-cysteine-d-histidine as the capping agents. All of these CuNPs exhibited peroxidase-mimicking activity in 3,3',5,5'-tetramethylbenzidine oxidation and presented chiral selectivity toward 3,4-dihydroxy-d,l-phenylalanine (d,l-DOPA). More importantly, DCDH-modified CuNPs (DCDH@CuNPs) showed higher peroxidase-mimicking catalytic activity in the presence of d-DOPA than l-DOPA. This demonstrates that in the stereoselective recognition CuNPs play the catalytic center role and chiral dipeptide ligands play the inducer role. The insights obtained from this study not only provide information to deeply understand the molecular principles of colorimetric chiral recognition upon CuNPs but also guide the design of dipeptide-based chiral nanozymes toward enantiomers.

Abstract:The burgeoning interest in rapid, simultaneous multi-target detection has propelled advancements in chiral electrochemiluminescence (ECL) assays. This study presents the design and implementation of a potential-resolved dual-color ECL sensor, engineered for the concurrent detection of aspartic acid (Asp) and phenylalanine (Phe) enantiomers. The sensor array was meticulously constructed by amalgamating anode chiral ECL probes Ru(phen)2(L-Cys) nanocrystals with cathode ECL probes ZnO nanoflowers (ZnO NFs). This research explored the potential of executing multianalyte assays via a potential-resolved ECL strategy, contributing to the advancements in the field of chiral ECL assays.

Abstract:Abstract Chirality exists everywhere in nature, so chiral recognition is also an extremely important field. This paper explores the recognition of amino acid configuration by an electrochemical chiral sensing platform constructed with carbon nanomaterials (carbon quantum dots (CQDs) as the substrate material) combined with chiral selectors (chitosan (CS)). CQDs/CS, which is bonded by intermolecular force, and CQDs‐CS, which is formed by covalent grafting through amidation, are selected as electrochemical chiral recognition materials, and a series of characterization methods are carried out to indicate the successful preparation of the materials, and then attached to the glass carbon electrode (GCE), forming a chiral sensing platform for amino acid enantiomers. The differential voltammetry (DPV) method was used to convert the chemical signal into electrical signal. The results showed that compared with CQDs/CS, CQDs‐CS had better enantiomer resolution to tryptophan (Trp), and the peak current ratio (I L /I D ) was 2.28.

Abstract:A novel chiral sensing platform, 6-O-α-maltosyl-β-cyclodextrin (Mal-βCD)-based film, is proposed for selective electrochemical recognition of tyrosine (Tyr) enantiomers. Black phosphorus nanosheets (BP NSs) and Mal-βCD modified glassy carbon electrode (Mal-βCD/BP NSs/GCE) were prepared by a layer-to-layer drop-casting method, and the platform was easy to fabricate and facile to operate. It is proposed that the amino and hydroxyl groups of the Tyr enantiomers and the chiral hydroxyl groups of Mal-βCD selectively form intermolecular hydrogen bonds to dominate effective chiral recognition. Two linear equations of Ip (μA) = 11.40 CL-Tyr (mM) + 0.28 (R2 = 0.99147) and Ip (μA) = 7.96 CD-Tyr (mM) + 0.22 (R2 = 0.99583) in the concentration range 0.01-1.00 mM have been obtained. The limits of detection (S/N=3) for L-Tyr and D-Tyr were 4.81 and 6.89 µM, respectively. An interesting phenomenon was that the value of IL-Tyr/ID-Tyr (1.51) in this work was slightly higher than the value of IL-Trp/ID-Trp (1.49) reported in our previous study, where tryptophan (Trp) enantiomers were electrochemically recognized by Nafion (NF)-stabilized BPNSs-G2-β-CD composite. The two similar sensors fabricated by different methods showed different recognition ability toward either Tyr or Trp enantiomers, and the underlying mechanism was discussed in detail. More importantly, the proposed chiral sensor enables prediction of the percentages of D-Tyr in racemic Tyr mixtures. The chiral sensor may provide a novel approach for the fabrication of novel chiral platforms in the practical detection of L- or D-enantiomer in racemic Tyr mixtures.Graphical abstract.

Abstract:Two amino-functionalized chiral poly(3,4-ethylenedioxythiophene) (PEDOT) derivatives, poly(N-(tert-butoxycarbonyl)-L-phenylalayl (3,4-ethylenedioxythiophene-2'-yl)methylamide) (PEDOT-Boc-Phe) and poly(L-phenylalayl (3,4-ethylenedioxythiophene-2'-yl)methylamide) (PEDOT-Phe), were synthesized electrochemically via potentiostatic polymerization of corresponding monomers N-(tert-butoxycarbonyl)-L-phenylalayl (3,4-ethylenedioxythiophene-2'-yl)methylamide (EDOT-Boc-Phe) and L-phenylalayl (3,4-ethylenedioxythiophene-2'-yl)methylamide (EDOT-Phe). The electrochemical behavior, structural characterization, spectroscopic properties, and surface morphology of two chiral conducting polymer films were systematically explored. They displayed excellent reversible redox activities, and rough and compact surface. Importantly, PEDOT-Boc-Phe and PEDOT-Phe modified electrodes were used to identify 3,4-dihydroxyphenylalanine (DOPA) enantiomers by square wave voltammetry (SWV) in sulphuric acid solution. It is interesting that the SWV peak currents of enantiomers were found to be quite different and hence the enantiomers could be successfully recognized but the enantioselectivity was not observed using the common method of cyclic voltammetry (CV). Satisfactory results implied that the obtained polymer films could play crucial roles in the development of practical value and analytical application prospects.

Abstract:Chiral recognition of amino acid plays a significant role in pharmaceutical, medical, and food science. This study describes a chiral sensing system of β-cyclodextrin (β-CD)-coated sulfur quantum dots (CD-SQDs) for the selective fluorescence recognition of tryptophan (Trp) enantiomers. CD-SQDs were prepared by a facile assembly fission method and could selectively recognize L-Trp by the different binding ability between L/D-Trp and β-CD. The inclusion of L-Trp and the stereoselective catalysis of CD-SQDs enzyme mimics cause the increased fluorescence intensity of CD-SQDs, which has a linear response ranging from 10 to 500 nM and the detection limit as 2.3 nM. CD-SQDs also show great selectivity for L-Trp from the commercial compound amino acid injection. The study could provide an effective method for the chiral recognition of amino acid enantiomers based on the catalytic activity of nanoenzymes.

Development of a chiral electrochemical sensor based on copper–amino acid mercaptide nanorods for enantioselective discrimination of tryptophan enantiomers

Abstract:Chirality is an important property of nature and it regulates fundamental phenomena in nature and organisms. Here, we develop a chiral electrochemical sensor based on copper–amino acid mercaptide nanorods (L-CuCys NRs) to discriminate tryptophan (Trp) isomers. The chiral L-CuCys NRs are prepared in alkaline solution based on the facile coordination reaction between the sulfhydryl groups of L-Cys and copper ions. Since the stability constant ( K ) of L-CuCys NRs with L-Trp (752) are much higher than that of L-CuCys NRs with D-Trp (242), the cross-linking bonds between L-CuCys NRs and L-Trp are more stable than those between L-CuCys NRs and D-Trp. Consequently, this electrochemical sensor can selectively recognize the Trp isomers with an enantiomeric electrochemical difference ratio ( I L-Trp / I D-Trp ) of 3.22, and it exhibits a detection limit of 0.26 μM for L-Trp. Moreover, this electrochemical sensor can quantitatively measure Trp isomers in complex samples. Importantly, this electrochemical sensor has the characteristics of high stability, good repeatability, easy fabrication, low cost, and efficient discrimination of tryptophan (Trp) isomers.

Abstract:Chirality sensing is a very challenging task. Here, we report a method for ultrasensitive detection of chiral molecule L/D-carnitine based on changes in the recognition tunneling current across self-assembled core satellite gold nanoparticle (GNP) networks. The recognition tunneling technique has been demonstrated to work at the single molecule level where the binding between the reader molecules and the analytes in a nanojunction. This process was observed to generate a unique and sensitive change in tunneling current, which can be used to identify the analytes of interest. The molecular recognition mechanism between amino acid L-cysteine and L/D-carnitine has been studied with the aid of SERS. The different binding strength between homo- or heterochiral pairs can be effectively probed by the copper ion replacement fracture. The device resistance was measured before and after the sequential exposures to L/D-carnitine and copper ions. The normalized resistance change was found to be extremely sensitive to the chirality of carnitine molecule. The results suggested that a GNP networks device optimized for recognition tunneling was successfully built and that such a device can be used for ultrasensitive detection of chiral molecules.
Molecular Imprinted Polypyrrole Nanowires for Chiral Amino Acid Recognition

Abstract:Recognition and separation of chiral molecules is of great importance in the field of biotechnology, especially on pharmaceutics. Molecular imprinted polymers (MIP) have been proved as a powerful tool for the separation of target molecules. In this paper, polypyrrole (PPy) nanowires with an average diameter of ca. 100nm and few micrometers in length were successfully prepared by electrochemical polymerization, in which enantiomeric camphorsulfonic acid (d- or l-CSA) molecule acted as both the dopant and molecular imprinting pseudo-template. Both electrochemical impedance spectra (EIS) and circular dichroism (CD) spectra were employed to demonstrate the enantioselective interaction between de-doped PPy nanowires imprinted with CSA enantiomers (MIP-PPy) and chiral phenylalanine (d- or l-Phe). EIS leads to the characterization of interfacial electron resistance of the PPy coated electrode, while CD spectra provides spectral evidence for the enantioselective recognition event. Both methods proved that MIP-PPy nanowires have the capability for the recognition and separation of chiral phenylalanine. The sensitivity of the chiral recognition was further investigated by using various concentrations of chiral Phe and adsorption time. This approach provides a potential route for fabricating enantioselective recognition polypyrrole nanostructures that could be employed in enantiomeric sensoring or separating various chiral molecules.
