Abstract:Ethnopharmacological Relevance: Pepper essential oils have potential immunomodulatory, anti-tumor, and anti-cancer activities. Pepper exhibits the potential to prevent or attenuate carcinogenesis as therapeutic tools. However, the related mechanism remains unelucidated. Aim of the Study: The present study aims to provide reasonable information for the explanation of the dissimilarity of the essential oils from white (WPEO) and black pepper (BPEO). Materials and Methods: WPEO, BPEO, and their single active component, as well as synthetic antioxidants, were compared by the cell model methods and chemical methods, including intracellular antioxidant activity (CAA), total antioxidant activities (TAA), superoxide radical (SR), hydroxyl radical (HR), DPPH radical (DR) scavenging activities and inhibition ability of lipoprotein lipid peroxidation (ILLP). Results: The median effective concentration (EC 50 ) values (mg/mL) of the WPEO and BPEO of SR, HR, DR, and ILLP were 0.437 and 0.327, 0.486 and 0.204, 7.332 and 6.348, 0.688, and 0.624 mg/mL, respectively. The CAA units of WPEO and BPEO were 50.644 and 54.806, respectively. CAA, DR, and TAA of BPEO were significantly higher than those of WPEO ( p < 0.05). The BPEO and WPEO can be differentiated as the former have higher correlations with 3-carene, α-pinene, β-pinene, and limonene while the latter has a higher caryophyllene correlation. The WPEO and BPEO show a good intracellular scavenging ability of reactive oxygen species in HeLa cells. Conclusion: Generally, pepper oil has stronger activities than single components, indicating that pepper is a broad-spectrum natural antioxidant.

Abstract:The crude ethanol extract of Pulsatilla chinensis (Bunge) Regel roots was extracted successively with light petroleum, dichloromethane and n-butanol. The light petroleum fraction (PEF) exhibited potent anti-proliferation activity on HL60 cells with an IC50 value of 14 mu g/mL. As a result, ICR mice transplanted with tumor strain S180 were employed for testing the effectiveness of drug administration of PER The tumor inhibitory rate was 36.7% at a dose of 20 mg/kg/d, which was higher than the positive control, which produced 31.5% inhibition. However, an unusual phenomenon was observed in that the tumor inhibitory response was reverse dose-dependent since tumor inhibition was only 19.5% at a dose of 180 mg/kg/d. GC-MS revealed that the main components of the PEF were C-19 and C-22 polyunsaturated fatty acids. Previous studies had revealed that polyunsaturated fatty acids exhibit either anti-tumor or tumor promoting activities, and so it is proposed that the effects of PEF on tumor growth is dependent on dosage.
Black pepper ( Piper nigrum L.) essential oil counteracts dexamethasone-induced hepatic injury via modulating PGC-1α/ PPAR-α pathway in rats

Abstract:We previously characterized the volatile compounds of black pepper ( Piper nigrum ) essential oil (BPEO) and demonstrated its mitigating effect against dexamethasone-induced pancreatic damage. Herein, we investigated the protective effect of the oil against dexamethasone-induced hepatotoxicity in rats and pinned the possible underlying mechanisms. In the in vivo experiment, male Wistar rats were divided into five groups. The first group served as the normal control receiving a saline solution (Control). The second group was subjected to hepatic damage through dexamethasone exposure (Dexa) at 10 mg/kg. The third group received both dexamethasone and metformin (Met) at a dose of 50 mg/kg. The remaining two groups were exposed to dexamethasone along with black pepper oil administration at low and high doses (BPEO, 0.5 mL/kg and 1 mL/kg). We showed that BPEO offset toxicity manifestations, as indicated by reducing monocyte infiltration, hepatocyte degeneration and vacuolation, and reduction of ALT and AST levels. Moreover, relative to the dexamethasone-treated rats, BPEO declined the number of PGC-1α positive hepatocytes and activated the expression of PPAR-α in the liver. BPEO showed comparable hepatoprotective potency to the standard drug, metformin, especially at the higher dose tested (1 mL/kg). This study showcases the promising potential of BPEO in thwarting dexamethasone-induced adverse effects on the liver by alleviating hepatic biomarkers hikes and preventing histopathological changes and fibrosis. Finally, it opens a new avenue on the potential role of BPEO in modulating steroid-induced hepatic injury through the PGC-1α/ PPAR-α pathway.

Comparative analysis of chemical composition and antioxidant activity of essential oil and hydrolate from black pepper fruit (Piper nigrum L.)

Abstract:The objective of this study was to analyze the chemical composition of the essential oil (EO) and hydrolate of black pepper fruit, as well as their antioxidant activity. The EO was obtained by Clevenger-type hydrodistillation with hydromodule 1:10 m/v for 240 minutes, and the hydrolate was collected after the hydrodistillation process. The qualitative composition of EO was determined by GC/MS and quantitative by GC/FID method, while the qualitative composition of a hydrolate was determined by HS-SPME-GC/MS and quantitative composition by HS-SPME-GC/FID method. The antioxidant activity was investigated by DPPH assay. Fifty-five compounds were identified from black pepper EO, where the most abundant compounds were (E)-caryophyllene (41.6 %), limonene (9.7 %), and sabinene (8.6 %). Twelve compounds were identified from black pepper hydrolate, where the most abundant compounds were α-terpineol (34.7 %), borneol (17.3 %), and terpinen-4-ol (13.9 %). The hydrolate showed higher antioxidant activity after 20 minutes of incubation with an EC50 value of 0.993 ± 0.011 mg/cm3 compared to the EO with an EC50 value of 67.72 ± 1.871 mg/cm3. According to the results obtained in this study, both the EO and hydrolate are good sources of natural antioxidants with potential uses in the food, organic agriculture, pharmaceutical, and cosmetic industries as a safer alternative to synthetic additives. Furthermore, the possibility of wider uses of the hydrolate should be investigated in more detail. Although researchers interested in hydrolate investigation focus mostly on their antioxidant activity, these "aromatic wastes" could also present promising cosmetic activities.

The antioxidant and radical scavenging activities of black pepper (Piper nigrum) seeds

Abstract:Water and ethanol crude extracts from black pepper (Piper nigrum) were investigated for their antioxidant and radical scavenging activities in six different assay, namely, total antioxidant activity, reducing power, 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, and metal chelating activities. Both water extract (WEBP) and ethanol extract (EEBP) of black pepper exhibited strong total antioxidant activity. The 75 microg/ml concentration of WEBP and EEBP showed 95.5% and 93.3% inhibition on peroxidation of linoleic acid emulsion, respectively. On the other hand, at the same concentration, standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and alpha-tocopherol exhibited 92.1%, 95.0%, and 70.4% inhibition on peroxidation of linoleic acid emulsion, respectively. Also, total phenolic content in both WEBP and EEBP were determined as gallic acid equivalents. The total phenolics content of water and ethanol extracts were determined by the Folin-Ciocalteu procedure and 54.3 and 42.8 microg gallic acid equivalent of phenols was detected in 1 mg WEBP and EEBP.

Abstract:Piper nigrum Linnaeus is often used as a treatment for chills, stomach diseases, and other ailments. Piperine has many biological functions; however, its mechanism for preventing gastric mucosal damage is still unclear. The objective of this study was to investigate the preventive effects of piperine on ethanol-induced gastric mucosal injury by using GES-1 cells and rats. SSOD, CAT, GSH-Px and malondialdehyde MDA were effectively regulated in GES-1 cells pre-treated with piperine. Piperine significantly increased SOD, CAT and GSH-Px activities, but decreased the ulcer area, MDA, ROS and MPO levels in the gastric tissues of rats. RT-PCR analysis showed that piperine downregulated the mRNA expression levels of keap1, JNK, ERK and p38, and upregulated the mRNA transcription levels of Nrf2 and HO-1. Western blotting results indicated that piperine could activate the protein expression levels of Nrf2 and HO-1 and inhibit the protein expression levels of keap1, p-JNK, p-ERK and p-p38. In conclusion, piperine suppressed ethanol-induced gastric ulcers in vitro and in vivo via oxidation inhibition and improving gastric-protecting activity by regulating the Nrf2/HO-1 and MAPK signalling pathways.

Abstract:Black pepper (Piper nigrum L.) is the most widely used spice in the world. Blackening is considered to be beneficial and important in the processing of black pepper because it contributes to its color and flavor. The purpose of this paper is to investigate polyphenol oxidation as well as the chlorophyll and vitamin C (VC) degradation in the blackening of Piper nigrum L. Black pepper was produced by four methods, and changes in polyphenols, chlorophyll and VC were studied by high performance liquid chromatography (HPLC) and ultraviolet-visible and visible (UV-Vis) spectrophotometry. The results show that polyphenol oxidase activity significantly decreased during the preparation of black pepper, and the concentrations of phenolic compounds, VC, and chlorophyll a and b also significantly decreased. Polyphenol oxidation and chlorophyll and VC degradation contribute to the blackening. A crude extract of phenolic compounds from black pepper was prepared by the system solvent method. The greater the polarity of the extraction solvent, the higher the extraction rates of the phenolic compounds and the total phenol content. Pepper phenolic compounds were analyzed by HPLC analysis.

Abstract:Piperine is the main compound present in black pepper, and is the carrier of its specific pungent taste, which is responsible for centuries of human dietary utilization and worldwide popularity as a food ingredient. Along with the application as a food ingredient and food preservative, it is used in traditional medicine for many purposes, which has in most cases been justified by modern scientific studies on its biological effects. It has been confirmed that piperine has many bioactive effects, such as antimicrobial action, as well as many physiological effects that can contribute to general human health, including immunomodulatory, hepatoprotective, antioxidant, antimetastatic, antitumor, and many other activities. Clinical studies demonstrated remarkable antioxidant, antitumor, and drug availability-enhancing characteristics of this compound, together with immunomodulatory potential. All these facts point to the therapeutic potential of piperine and the need to incorporate this compound into general health-enhancing medical formulations, as well as into those that would be used as adjunctive therapy in order to enhance the bioavailability of various (chemo)therapeutic drugs.

Abstract:P. nigrum L. extracts and the piperine alkaloid have important antimicrobial, anti-inflammatory, and antioxidant properties. Therefore, in this study, we evaluated the antimicrobial activity and cytotoxicity of P. nigrum L. extracts and piperine, a compound isolated from the extracts of P. nigrum L. Extracts obtained via maceration, soxhlet, and purification steps, in addition to isolated piperine, were used in this study. Spectroscopic methods, such as nuclear magnetic resonance, scanning electron microscopy, X-ray diffraction, thermogravimetry, and differential scanning calorimetry, were used to characterize piperine. In the microbiological analyses, the extract obtained via maceration-derived sample showed high efficiency in inhibiting Salmonella spp. (MIC < 100 μg/mL). The extract obtained via a soxhlet-derived sample showed promising inhibitory activity against almost all microorganisms, with negligible inhibition of Pseudomonas aeruginosa. Favorable inhibition coefficients were also observed against Staphylococcus aureus and Salmonella spp. (MIC < 100 μg/mL) for the extract obtained via purification of the steps-derived sample. Piperine showed an excellent inhibition coefficient against most microorganisms, with inactivity only observed against P. aeruginosa. Cytotoxicity evaluation assays in cancer cell lines revealed that piperine exhibited inhibitory potential on all tested tumor cell lines, causing a decrease in cell viability and achieving an IC50 of less than 30 μg/mL. The analyzed extracts from P. nigrum L. seeds showed cytotoxic activity against tumor and non-tumor cell lines.

Abstract:Background: Cardiovascular diseases (CVDs) are the leading cause of death worldwide. Black pepper, the fruits of Piper nigrum L., is well known as "the king of spices" and used as seasoning and condiments globally. In addition to being an important food additive, black pepper is also used as a traditional medicine to treat vomiting, abdominal pain, and diarrhea, among others. Piperine is a major pungent alkaloid identified in the fruits of P. nigrum (black pepper), whose content is at a range of similar to 5-13%. Black pepper and piperine have shown protective effect on CVDs. Scope and approach: Literature search was conducted to systematically review the cardiovascular protective effect of both black pepper and its major bioactive constituent piperine. Key findings and conclusions: Black pepper was reported to regulate lipid metabolism, inflammation, and oxidation status in CVDs. Piperine exhibited beneficial effect by targeting many processes associated with atherosclerosis. Piperine is able to prevent lipid peroxidation, oxidized low-density lipoprotein uptake in macrophages, lipid droplet formation, and adhesion of inflammatory cells to endothelial monolayer, promote cholesterol efflux from macrophages, as well as improve lipid profile. Besides, piperine may ameliorate myocardial ischemia, cardiac injury, and cardiac fibrosis, exhibit antihypertensive and antithrombosis effect, as well as prevent arterial stenosis by inhibiting vascular smooth muscle cell proliferation. The summarized information could provide the basis to develop black pepper or piperine as a food additive to prevent or treat CVDs.
Anti-tyrosinase and anti-melanogenic effects of piperine isolated from Piper nigrum on B16F10 mouse melanoma cells

Abstract:The essential function of melanin is to protect our skin against harmful environmental factors. However, excessive melanin production can cause undesirable hyperpigmentation issues, such as freckles and melasma. Although several compounds are used to control melanin production by inhibiting tyrosinase (TYR), their efficacy is limited by skin-related adverse effects and cytotoxicity concerns. Consequently, searching for new natural compounds with an effective TYR inhibitor (TYR-I) activity but less harmful effects continues. Plant-based natural extracts are an alternative that are in great demand due to their safety and diverse biological properties. This study assessed ten isolated plant compounds for their TYR-I activities using an in vitro mushroom TYR inhibition assay. Among these compounds, piperine (400 μM) demonstrated the highest TYR-I activity, with a potency of 36.27 ± 1.96 %. Hence, this study examined the effect of piperine on melanogenesis in melanocyte stimulating hormone-treated B16F10 melanoma cells and using kojic acid as a positive reference. Cell viability was evaluated through the standard 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Measurements of cellular TYR activity and melanin content were performed and related to changes in the transcriptional expression levels of melanogenesis-related genes, assessed via quantitative real-time reverse transcriptase (RT-q)PCR analysis. The results revealed that piperine at a concentration of 44 μM significantly reduced cellular TYR activity by 21.51 ± 2.00 % without causing cytotoxicity. Additionally, at the same concentration, piperine significantly decreased the intracellular melanin content by 37.52 ± 2.53 % through downregulating transcription levels of TYR and TYR-related protein 1 (TRP-1) but not TRP-2. Kojic acid, at a concentration of 1407 μM, induced a significant decrease in the melanin content and cellular TYR activity by suppressing all three melanogenesis-related genes. These findings suggest that piperine has potential as a potent depigmenting agent.

Abstract:Nowadays, antibacterial resistance to synthetic antibiotics has become a big issue worldwide. Mentha piperita essential oil (EO) and its bioactive components with antibacterial, antibiofilm, and antioxidant properties are recommended for the treatment of microbial infections in traditional medicine. This study aimed to detect phytochemical components, antioxidant and cytotoxic activity; to evaluate antibacterial and antibiofilm effects of M. piperita EO on strains. Gas chromatography/mass spectrometry (GC-MS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests were used to determine chemical composition, antioxidant and cytotoxic potentials of EO, respectively. Antibacterial activity was carried out through disc diffusion and minimum inhibitory concentration (MIC) assays. The biofilm biomass of bacteria was determined by the crystal violet (CV) staining method. Menthone (27.28%), L-menthol (23.71%), and limonene (7.48%) were the main components. M. piperita EO was found to have strong antibacterial activity with the MIC-MBC values in the range of 0.125-16.00% (v/v) and moderate antioxidant activity. The peppermint EO showed an antibiofilm effect on all strains and a cytotoxic effect on NIH 3T3 cells. M. piperita EO may be used alone or in combination with antibiotics or carrier oils as a therapeutic agent against bacterial infections. Further in vivo experiments should be carried out to detect a safe dosage of EO.
Chemical composition and antioxidant activity of black pepper (Piper nigrum L.) fructus essential oil hydrodistillation fractions

Abstract:The aim of this study was to examine the effect of hydrodistillation time on the chemical composition and antioxidant activity of collected essential oils' (EOs') fractions from black pepper fructus for five hydrodistillation periods: I (0-15 min), II (15-30 min), III (30-60 min), IV(60-90 min) and V (90-120 min). In this way, an essential oil with the desired chemical composition and antioxidant activity can be selected. The essential oil was obtained by Clevenger-type hydrodistillation (CHD). The qualitative and quantitative composition of the collected EOs' fractions was determined by GC-MS and GC-FID analysis. The efficiency of EOs' fractions to scavenge DPPH radicals was evaluated spectrophotometrically. Essential oil yield was 3.0 ± 0.050 ml/100 g of plant material after 120 min of CHD. The content of monoterpene hydrocarbons and aromatic compounds decreased, while the content of oxygenated monoterpenes, sesquiterpene hydrocarbons and oxygenated sesquiterpenes increased with the time of CHD. The most abundant compound of EOs' fractions was ( E )-caryophyllene, whose content increased with the time of CHD (I-V: 24.9-36.6%). Results indicated that all collected EOs' fractions exhibited antioxidant activity (EC 50 values were in the range I-V: 81.27±0.327–8.02±0.044 mg/ml), where fraction V showed the highest activity. This study proved that CHD time and fractionation affect the composition and antioxidant activity of black pepper essential oil. Future research may be extended to examine other biological activities of black pepper EOs' fractions, as well as synergistic effects of other identified active components of the essential oil, in addition to ( E )-caryophyllene, that would affect biological activities.

Safety Evaluation and Antihyperlipidemia Effect of Aqueous Extracts from Fermented Puerh Tea

Abstract:Fermented puerh tea, having undergone a long period of secondary oxidization and fermentation, has become more and more popular in recent years. In the present paper, a safety evaluation of aqueous extracts from fermented puerh tea (EFPT) was performed, including an oral acute toxicity study in rats and mice, mutation tests, a mouse micronucleus test, mouse sperm abnormality test and a 30 day feeding study in rats. Meanwhile, the antihyperlipidemia effect of EFPT was investigated as well. It was found that the oral maximum tolerated dose of EFPT was more than 10.0 g per kg body weight both in rats and mice. And it had no mutagenicity as judged by negative experimental results of the mutation test. No abnormal symptoms, clinical signs or deaths have been found in rats in each group throughout the experiments. In addition, EFPT in this study showed certain effects on hyperlipidemia.

In this paper, we discuss the results of the subchronic toxicity and mutagenicity studies conducted to understand the potential for adverse effects if any, of Black Pepper Extract Preparation (BPE).MethodsTo evaluate mutagenicity, an Ames test was conducted with BPE in the presence and absence of S9 metabolic activation. Long-term safety was inferred through a 90-day subchronic toxicology study using adult rats. Dose levels were selected with expected human intake levels of NFT (120 mg/kg/day), with an acceptable safety factor, for preclinical safety and tolerability. Sprague Dawley rats were fed diets targeting dietary intakes (doses) of 0, 125, 350, or 700 mg/kg/day of BPE for 90 days, an NFT dose level equivalent to 68, 190, and 380 mg/kg/day.ResultsIn vitro Ames test up to 5000 μg/plate with and without S9 metabolic activation showed no BPE-related increases in revertant colony numbers and was non-mutagenic. There were no BPE-related changes in viability, clinical signs, body weight, food consumption, and organ weights. BPE dietary administration did not induce any treatment-related changes in hematology, clinical chemistry, other macroscopic or microscopic endpoints.Discussion and conclusionThe highest dose tested with BPE (700 mg/kg/day) was the no-observed-adverse-effect level (NOAEL) that revealed no adverse effects. Based on toxicological endpoints evaluated, this NOAEL for BPE corresponded to a human equivalent NFT dose level of 380 mg/kg/day, dependent upon a (∼50%) concentration of NFT in BPE.

Abstract:Piper tuberculatum, traditionally known as 'pimenta‐d'arda,' 'pimenta‐longa,' or 'pimenta de macaco,' has been used in Brazilian folk medicine to treat inflammatory symptoms, It is used for its sedative effects in pain relief. Considering this species' significant essential oil content, the present study aimed to evaluate the anti‐edematogenic and anti‐inflammatory effects of the essential oil Piper tuberculatum (EOPT) in vivo. To this end, Swiss mice (Mus musculus) of both sexes were treated orally with the EOPT at 50, 100, and 250 mg/Kg. The rotarod and open field evaluated the potential activity in the central nervous system. At the same time, formalin and abdominal writhing tests were carried out to perform the pharmacological screening of the essential oil. Next, the anti‐edematogenic effect was assessed using paw edema models induced by carrageenan, dextran, histamine, and arachidonic acid. The anti‐inflammatory activity was then characterized in peritonitis (acute) and granuloma (chronic) models. All the EOPT doses (50, 100, and 250 mg/kg) had analgesic effects associated with anti‐inflammatory mechanisms in screening models. Accordingly, the treatment (EOPT 100 mg/Kg) inhibited the inflammatory process in acute and chronic models. In conclusion, EOPT has analgesic, antiedematogenic, and anti‐inflammatory effects, highlighting its potential use in developing anti‐inflammatory drugs.

Abstract:The aim of this study was to investigate the antibacterial activity of black pepper essential oil (BPEO) on Escherichia coli, further evaluate the potential mechanism of action. Results showed that the minimum inhibition concentration (MIC) of BPEO was 1.0 μL/mL. The diameter of inhibition zone values were with range from 17.12 to 26.13 mm. 2 × MIC treatments had lower membrane potential and shorter kill-time than 1 × MIC, while control had the highest values. E. coli treated with BPEO became deformed, pitted, shriveled, adhesive, and broken. 2 × MIC exhibited the greatest electric conductivity at 1, 3, 5, 7, 9, 11, and 13 h, leaked DNA materials at 4, 8, 12, 16, 20, 24, and 28 h, proteins at 4, 6, 8, 10, 12, 14, and 16 h, potassium ion at 0, 0.5, 1, 1.5, and 2 h, phosphate ion at 0.5, 1, 1.5, and 2 h and ATP (P < 0.05); 1 × MIC had higher values than control. BPEO led to the leakage, disorder and death by breaking cell membrane. This study suggested that the BPEO has potential as the natural antibacterial agent in meat industry.

Abstract:Background Piper nigrum essential oil (PnEO) possesses pleasant aroma, unique flavor, and various bioactivities; however, its role against colitis remains unclear. Purpose In this study, we investigated the role of PnEO in relieving colitis and explored its potential mechanisms in a mouse model of dextran sulfate sodium (DSS)–induced colitis. Methods Initially, we identified and quantified the components of PnEO by gas chromatography-mass spectrometry (GC-MS). Subsequently, we investigated the protective role of PnEO (50 and 200 mg/kg) in DSS-induced colitis in mice by evaluating disease activity index (DAI) scores and colon length, and performing histological analyses. Eyeball blood was collected and cytokines were determined using ELISA kits. The anti-inflammatory mechanisms of PnEO were analyzed by western blot (WB) and immunohistochemistry (IHC). The intestinal barrier function was evaluated according to tight junction (TJ) protein mRNA levels. We used 16S rRNA gene sequencing to analyze the intestinal microflora of mouse cecal contents. Results Supplementation with PnEO (50 and 200 mg/kg) increased colon length and improved colon histopathology. PnEO regulated inflammatory responses by downregulating TLR4/MAPKs activation, thereby reducing the release of cytokines and mediators. Moreover, it also protected the intestinal barrier through enhancing the expression of claudin-1, claudin-3, occludin, ZO-1, and mucin 2. 16S rRNA gene sequencing revealed that PnEO (200 mg/kg) decreased the abundance of Akkermansia in the gut microbiome. Conclusion PnEO treatment (50 and 200 mg/kg) relieved DSS-induced colitis by inhibiting TLR4/MAPK pathway and protecting intestinal barrier, and high-dose PnEO exhibited better effects. Moreover, PnEO (200 mg/kg) regulated key compositions of the gut microbiome, which indicated that it had therapeutic potential for sustaining gut health to lower the risk of colitis.

Abstract:Pepper (Piper nigrum L.) is a widely used spice plant known for its fruits and roots, which serve as flavor enhancers in culinary applications and hold significant economic value. Despite the popularity of pepper fruits, their roots remain relatively understudied, with limited research conducted on their bioactive components. This study focused on discovering and separating the primary bioactive amide alkaloids found in pepper roots. The process involved using the antioxidant activity of crude fractions and the Global Natural Products Social Molecular Networking analysis platform. The process led to the discovery of 23 previously unknown hydroxyl-amide alkaloids. Notably, compounds 11, 12, and 14 showed excellent antioxidant activity, while compound 11 exhibited significant inhibitory effects on mushroom tyrosinase. Theoretical exploration of enzyme-ligand interactions was conducted through molecular docking and molecular dynamics simulation. The findings of this study highlight the potential of hydroxyl-amide alkaloids as antioxidant products and natural food preservatives in the pharmaceutical and food cosmetic industries.
Acetic acid as an effective mucolytic in magnification with chromoendoscopy for pit patterns analysis

Abstract:We have read the article of Togashi et al. with great interest [1]. They have com− pared the accuracy of predicting polyp histology at magnification colonoscopy using acetic acid and indigo carmine, ei− ther alone or in combination. They have shown that use of acetic acid spray in− stantly improves the pit pattern image during magnification chromoendoscopy and has high accuracy in the histological prediction of colorectal polyps. They state that acetic acid is particularly effective because it rapidly removes the mucus that can make pit pattern analysis diffi− cult, and the aceto−white reaction accen− tuates the pit pattern. However, they have shown that acetic acid increases the accu− racy more for adenomas (76 % ± 94%) than for hyperplastic polyps (92 %± 100 %). Generally, hyperplastic polyps are found as diminutive sessile polyps especially in the rectum and sigmoid colon. Endoscop− ically, hyperplastic polyps rather than adenomas correspond to sessile, pale polyps with a smooth surface and a par− tial or complete surface covering of mu− cus. Therefore, if acetic acid is an effective mucolytic, as expected by the authors, it should improve the diagnostic accuracy for hyperplastic polyps rather than for adenomas. Furthermore, the authors have shown that acetic acid alone (95 %) achieves higher accuracy overall than indigo car− mine alone (83%), although this differ− ence did not reach statistical significance, possibly due to the small sample size. We conducted a prospective study to clarify whether using dye spraying and magnifi− cation at each step contributes to further endoscopic distinction in comparison with the conventional endoscopic view [2]. The overall diagnostic accuracy achieved with chromoendoscopy using indigo carmine with magnification was 95.6 % ± significantly better (by 10% and 5 % respectively) than the accuracy achieved with either the conventional view or with chromoendoscopy without magnification. Therefore, the result of Togashi et al. in relation to the diagnostic accuracy of indigo carmine with magnifi− cation seems to be lower than expected.
