Abstract:Although sedimentary ancient DNA (sedaDNA) has been increasingly used to study paleoecological dynamics (<a href="#B88">Schulte et al., 2020</a>), the approach has rarely been compared with the traditional method of pollen analysis for investigating past changes in the vegetation composition and diversity of Arctic treeline areas. Here, we provide a history of latitudinal floristic composition and species diversity based on a comparison of sedaDNA and pollen data archived in three Siberian lake sediment cores spanning the mid-Holocene to the present (7.6–0 cal ka BP), from northern typical tundra to southern open larch forest in the Omoloy region. Our results show that the sedaDNA approach identifies more plant taxa found in the local vegetation communities, while the corresponding pollen analysis mainly captures the regional vegetation development and has its limitations for plant diversity reconstruction. Measures of alpha diversity were calculated based on sedaDNA data recovered from along a tundra to forest tundra to open larch forest gradient. Across all sites, sedaDNA archives provide a complementary record of the vegetation transition within each lake's catchment, tracking a distinct latitudinal vegetation type range from larch tree/alder shrub (open larch forest site) to dwarf shrub-steppe (forest tundra) to wet sedge tundra (typical tundra site). By contrast, the pollen data reveal an open landscape, which cannot distinguish the temporal changes in compositional vegetation for the open larch forest site and forest-tundra site. Increasing Larix pollen percentages were recorded in the forest-tundra site in the last millenium although no Larix DNA was detected, suggesting that the sedaDNA approach performs better for tracking the local establishment of Larix. Highest species richness and diversity are found in the mid-Holocene (before 4.4 ka) at the typical tundra site with a diverse range of vegetational habitats, while lowest species richness is recorded for the forest tundra where dwarf-willow habitats dominated the lake's catchment. During the late Holocene, strong declines in species richness and diversity are found at the typical tundra site with the vegetation changing to relatively simple communities. Nevertheless, plant species richness is mostly higher than at the forest-tundra site, which shows a slightly decreasing trend. Plant species richness at the open larch forest site fluctuates through time and is higher than the other sites since around 2.5 ka. Taken together, there is no evidence to suggest that the latitudinal gradients in species diversity changes are present at a millennial scale. Additionally, a weak correlation between the principal component analysis (PCA) site scores of sedaDNA and species richness suggests that climate may not be a direct driver of species turnover within a lake's catchment. Our data suggest that sedaDNA and pollen have different but complementary abilities for reconstructing past vegetation and species diversity along a latitude.

Abstract:To reconstruct past vegetation from pollen or, more recently, lake sedimentary DNA (sedDNA) data is a common goal in palaeoecology. To overcome the bias of a researcher’s subjective assessment and to assign past assemblages to modern vegetation types quantitatively, the modern analogue technique (MAT) is often used for vegetation reconstruction. However, a rigorous comparison of MAT-derived pollen-based and sedDNA-based vegetation reconstruction is lacking. Here, we assess the dissimilarity between modern taxa assemblages from lake surface-sediments and fossil taxa assemblages from four lake sediment cores from the south-eastern Tibetan Plateau and northern Siberia using receiver operating characteristic (ROC) curves, ordination methods, and Procrustes analyses. Modern sedDNA samples from 190 lakes and pollen samples from 136 lakes were collected from a variety of vegetation types. Our results show that more modern analogues are found with sedDNA than pollen when applying similarly derived thresholds. In particular, there are few modern pollen analogues for open vegetation such as alpine or arctic tundra, limiting the ability of treeline shifts to be clearly reconstructed. In contrast, the shifts in the main vegetation communities are well captured by sedimentary ancient DNA (sedaDNA). For example, pronounced shifts from late-glacial alpine meadow/steppe to early–mid-Holocene coniferous forests to late Holocene Tibetan shrubland vegetation types are reconstructed for Lake Naleng on the south-eastern Tibetan Plateau. Procrustes and PROTEST analyses reveal that intertaxa relationships inferred from modern sedaDNA datasets align with past relationships generally, while intertaxa relationships derived from modern pollen spectra are mostly significantly different from fossil pollen relationships. Overall, we conclude that a quantitative sedaDNA-based vegetation reconstruction using MAT is more reliable than a pollen-based reconstruction, probably because of the more straightforward taphonomy that can relate sedDNA assemblages to the vegetation surrounding the lake.

Lake Water Depth Controlling Archaeal Tetraether Distributions in Mid‐latitude Asia: Implications for Paleo Lake‐level Reconstruction

Abstract:Lake-level reconstructions, related to terrestrial hydrological changes, are important for our understanding of past and future climates. Currently, however, reliable lake-level proxies are still limited. Here we report distributions of archaeal tetraether lipids in 70 surface sediment samples collected from 55 lakes in midlatitude Asia. We have found that among various lake physico-chemical characteristics, the relative abundances of crenarchaeol and Hydroxylated isoprenoid glycerol dialkyl glycerol tetraethers (%cren and %OH-GDGTs) are best correlated with lake water depth, due to a preference of Thaumarchaeota, the producer of these biomarkers, for a niche in subsurface lake water. This supports the recent hypothesis based on single-lake investigations that %cren and %OH-GDGTs are potentially novel lake-level proxies. Our results also suggest that %OH-GDGTs is less affected by soil input than %cren. Nevertheless, other confounding factors should be well constrained and local/site-specific calibrations are needed before the two molecular proxies are used quantitatively in down-core applications. Plain Language Summary Lake-level reconstructions can provide useful information about past changes in hydroclimate, which impact both ecosystems and socio-economic sustainable development in fundamental ways, but reliable lake-level proxies are still limited to date. Here we show that the %cren and %OH-GDGTs indices, calculated based on the distributions of archaeal membrane lipids, are controlled by lake water depth in lake surface sediments across a variety of lake conditions. This suggests that past lake level changes can be inferred by analyzing the ubiquitous archaeal lipids preserved in downcore lake sediments.

Abstract:Ancient DNA metabarcoding applied together with the investigations of the plant macro-remains, pollen, spores and non-pollen palynomorphs (NPP), open new perspectives and give better taxonomical resolution, allowing to obtain more precise and specific data on the local environment conditions and their changes. So far, only three multiproxy studies that involve both molecular and palaeobotanical/palynological methods are available for the high Arctic archipelago Svalbard. We intend to contribute filling this gap. Therefore, a field trip to Svalbard was undertaken in September, 2019, and three sediment cores were retrieved from the Tenndammen lake (N 78&#176;06.118; E 15&#176;02.024, 7 m asl) which is a small and shallow water body (ca 2.5&#160;m depth). The lake is located in the valley of Colesdalen, a well-known Svalbard&#8217;s biodiversity hot spots and a home for about seven to ten thermophilic plant species.To investigate the Holocene to modern vegetation history of this place, the 85cm core Te2019 was chosen, it was described for lithology, X-rayed, &#181;XRF-scanned, line-scan photographed with high resolution and sampled for sedaDNA, pollen, spores and NPP studies as well as for studies on plant macrofossils. Ten 14C AMS dates were taken in order to establish an age-depth model. The DNA record contains around 100 taxa, most findings of those are supported by pollen studies (Asteraceae, Betula, Brassicaceae, Salix, Saxifraga, Vaccinium/Ericaceae) and by spores (Equisetum and Bryophyta). In addition, various fungi spores were identified. Investigations of plant macro-remains well support findings of the aquatic (i.e. Warnstorfia fluitans) and terrestrial mosses (e.g. Aulacomnium conf. turgidum, Bryum spp., Distichium capillaceum, Calliergon richardsonii, Scorpidium cossonii, Sphagnum spp., Rhizomnium spp.). Besides, fragments of Salix and Betula leaves and fruit parts, various leaf, stem tissues and flower fragments of Saxifraga species were found within the samples from the same depths with the correspondence to DNA records. Three DNA zones (SvDNA 1 &#8211; SvDNA3) and &#160;one subzone within the earliest zone (SvDNA-1a &#8211; SvDNA-1b) were established. Relations between DNA, pollen and macrofossil zones were studied. This study is performed within the &#8220;Future ArcTic Ecosystems&#8221; (FATE) research program: Initiative for investigation on drivers of diversity and future scenarios from ethnoecology, contemporary ecology and ancient DNA.

Abstract:Lake sediments represent valuable and widely used archives for tracking environmental and biotic changes over time. Past aquatic communities are traditionally studied via morphological identification of the remains of organisms. However, molecular identification tools, such as DNA metabarcoding, have revolutionized the field of biomonitoring by enabling high-throughput and fast identification of organisms from environmental samples (e.g., sediments and soil). Sedimentary ancient DNA (sedaDNA) metabarcoding, an approach to track the biodiversity of target organisms from sediment cores, spanning thousands of years, has been successfully applied in many studies. However, researchers seldom explore how well the signals from sedaDNA data correlate with the fossil records of target organisms. This information is essential to infer past environmental conditions and community changes of bioindicators when the increasingly popular molecular identification method, metabarcoding, is desired instead of a morphological identification approach. In this study, we explore the correlations of diatom valve records across the last ∼940 years with the diatom sedaDNA metabarcoding data from the same sediment core from lake Nam Co (Tibetan Plateau). Overall, the results from valve vs. sedaDNA data revealed concordant diatom richness as well as community patterns. However, several mismatches in the diatom taxonomic composition existed between the data sets. In general, sedaDNA data harbored much higher diatom diversity, but due to the lack of reference sequences in public databases, many molecular units (amplicon sequence variants) remained unclassified to lower taxonomic levels. As our study lake, Nam Co, is characterized by brackish water and alkaline pH, some likely cases for the observed taxonomic composition mismatches may be due to a valve dissolution issue. Nevertheless, significant drivers for the diatom richness and community structure largely corresponded between data sets. Both valve and sedaDNA data demonstrated similar breakpoints for historical diatom community shifts. A particularly strong shift in the diatom community structure occurred after ∼1950 CE, which may be associated with abrupt environmental changes on the Tibetan Plateau. Altogether, our study indicates that environmentally driven signals reflected by the diatom communities are successfully recovered via microfossil as well as molecular identification methods.

Abstract:Environmental DNA (eDNA) analysis methods permit broad yet detailed biodiversity sampling to be performed with minimal field effort. However, considerable uncertainty remains regarding the spatial resolution necessary for effective sampling, especially in aquatic environments. Also, contemporary plant communities are under-investigated with eDNA methods relative to animals and microbes. We analyzed eDNA samples from six small temperate lakes to elucidate spatial patterns in the distributions of algae and aquatic and terrestrial plants, using metabarcoding of the Internal Transcribed Spacer-1 (ITS1) genomic region. Sampling locations were varied across horizontal and vertical space: sites in each lake included a mixture of nearshore and offshore positions, each of which was stratified into surface (shallow) and benthic (deep) samples. We detected the expected community variation (beta diversity) from lake to lake, but only small effects of offshore distance and sampling depth. Taxon richness (alpha diversity) was slightly higher in nearshore samples, but displayed no other significant spatial effects. These diversity metrics imply that plant eDNA is more evenly distributed than its generating organisms in these small lake environments. Read abundances were heavily weighted toward aquatic macrophytes, though taxon richness was greatest in the algae and other non-vascular plants. We also identified representatives of many phylogenetically and ecologically varied plant taxa, including terrestrial species from surrounding areas. We conclude that freshwater plant eDNA surveys successfully capture differences among lake communities, and that easily accessible, shore-based sampling may be a reliable technique for informing research and management in similar ecosystems.

Abstract:Sediments from Tibetan lakes in NW China are potentially sensitive recorders of climate change and its impact on ecosystem function. However, the important plankton members in many Tibetan Lakes do not make and leave microscopically diagnostic features in the sedimentary record. Here we established a taxon-specific molecular approach to specifically identify and quantify sedimentary ancient DNA (sedaDNA) of non-fossilized planktonic organisms preserved in a 5-m sediment core from Kusai Lake spanning the last 3100 years. The reliability of the approach was validated with multiple independent genetic markers. Parallel analyses of the geochemistry of the core and paleo-climate proxies revealed that Monsoon strength-driven changes in nutrient availability, temperature, and salinity as well as orbitally-driven changes in light intensity were all responsible for the observed temporal changes in the abundance of two dominant phytoplankton groups in the lake, Synechococcus (cyanobacteria) and Isochrysis (haptophyte algae). Collectively our data show that global and regional climatic events exhibited a strong influence on the paleoecology of phototrophic plankton in Kusai Lake.

Abstract:Summary Fungi are crucial organisms in most ecosystems as they exert ecological key functions and are closely associated with land plants. Fungal community changes may therefore help reveal biodiversity changes in past ecosystems. Lake sediments contain DNA of organisms in the catchment area, which allows reconstructing past biodiversity by using metabarcoding of ancient sedimentary DNA. We developed a novel PCR primer combination for fungal metabarcoding targeting a short amplicon to account for length bias of amplification due to ancient DNA degradation. In-silico PCRs showed higher diversity using this primer combination than using previously established fungal metabarcoding primers. We analyzed existing data from sediment cores from four artic and one boreal lake in Siberia. These cores had been stored for 2–22 years and examined degradation effects of ancient DNA and storage time-related bias in fungal communities. Amplicon size differed between fungal divisions, however, we observed no significant effect of sample age on amplicon length and GC content, suggesting robust results. We also found no indication of post-coring fungal growth during storage distorting ancient fungal communities. Terrestrial soil fungi, including mycorrhizal fungi and saprotrophs, were predominant in all lakes, which supports the use of lake sedimentary ancient DNA for reconstructing terrestrial communities.
Plant Sedimentary Ancient DNA From Far East Russia Covering the Last 28,000 Years Reveals Different Assembly Rules in Cold and Warm Climates

Abstract:Woody plants are expanding into the Arctic in response to the warming climate. The impact on arctic plant communities is not well understood due to the limited knowledge about plant assembly rules. Records of past plant diversity over long time series are rare. Here, we applied sedimentary ancient DNA metabarcoding targeting the P6 loop of the chloroplast trn L gene to a sediment record from Lake Ilirney (central Chukotka, Far Eastern Russia) covering the last 28 thousand years. Our results show that forb-rich steppe-tundra and dwarf-shrub tundra dominated during the cold climate before 14 ka, while deciduous erect-shrub tundra was abundant during the warm period since 14 ka. Larix invasion during the late Holocene substantially lagged behind the likely warmest period between 10 and 6 ka, where the vegetation biomass could be highest. We reveal highest richness during 28–23 ka and a second richness peak during 13–9 ka, with both periods being accompanied by low relative abundance of shrubs. During the cold period before 14 ka, rich plant assemblages were phylogenetically clustered, suggesting low genetic divergence in the assemblages despite the great number of species. This probably originates from environmental filtering along with niche differentiation due to limited resources under harsh environmental conditions. In contrast, during the warmer period after 14 ka, rich plant assemblages were phylogenetically overdispersed. This results from a high number of species which were found to harbor high genetic divergence, likely originating from an erratic recruitment process in the course of warming. Some of our evidence may be of relevance for inferring future arctic plant assembly rules and diversity changes. By analogy to the past, we expect a lagged response of tree invasion. Plant richness might overshoot in the short term; in the long-term, however, the ongoing expansion of deciduous shrubs will eventually result in a phylogenetically more diverse community.

Abstract:Long-chain alkenones in lacustrine settings are potentially excellent biomarkers for the reconstruction of past terrestrial environmental conditions, and have been found in many different types of lakes around the globe. A wider range of factors influence the occurrence and distribution of alkenones in lake sediments and waters when compared to marine systems. Lake environmental conditions, such as temperature (in particular) and salinity, are among the key factors controlling alkenone distributions in lacustrine settings. Here we investigated alkenone distribution patterns in lakes of the northern Qinghai-Tibetan Plateau, China, and their possible relationship with environmental conditions, by analyzing paired samples of suspended particulate matter in surface waters and surface sediments. Salinity of investigated lake waters ranges from almost 0 to ∼100g/L, while temperature variation among the lakes is minimal, effectively eliminating temperature effects on the alkenone distribution patterns observed here. We show that (1) alkenone concentrations vary substantially between the lakes, yet controlling mechanisms remain elusive; (2) C37/C38 ratios are substantially lower in the lakes of the Qaidam Basin than in the Lake Qinghai region, probably indicating different alkenone producers in the two regions; and (3) large variations in %C37:4 (the percentage of the C37:4 alkenone), determined from both surface waters and sediments, are negatively correlated with salinity. We suggest that the %C37:4 index could be used as a salinity indicator at least on a regional scale, with careful considerations of other potentially complicating factors. However, potential reasons for why salinity could significantly affect %C37:4 values need further investigation.

Abstract:The Air Force Research Laboratory (AFRL) is studying the application and utility of various ground-based and space-based optical sensors for improving surveillance of space objects in both Low Earth Orbit (LEO) and Geosynchronous Earth Orbit (GEO). This information can be used to improve our catalog of space objects and will be helpful in the resolution of satellite anomalies. At present, ground-based optical and radar sensors provide the bulk of remotely sensed information on satellites and space debris, and will continue to do so into the foreseeable future. However, in recent years, the Space-Based Visible (SBV) sensor was used to demonstrate that a synthesis of space-based visible data with ground-based sensor data could provide enhancements to information obtained from any one source in isolation. The incentives for space-based sensing include improved spatial resolution due to the absence of atmospheric effects and cloud cover and increased flexibility for observations. Though ground-based optical sensors can use adaptive optics to somewhat compensate for atmospheric turbulence, cloud cover and absorption are unavoidable. With recent advances in technology, we are in a far better position to consider what might constitute an ideal system to monitor our surroundings in space. This work has begun at the AFRL using detailed optical sensor simulations and analysis techniques to explore the trade space involved in acquiring and processing data from a variety of hypothetical space-based and ground-based sensor systems. In this paper, we briefly review the phenomenology and trade space aspects of what might be required in order to use multiple band-passes, sensor characteristics, and observation and illumination geometries to increase our awareness of objects in space.
Appraisal of Alkenone- and Archaeal Ether-Based Salinity Indicators in Mid-Latitude Asian Lakes

Abstract:Lake water salinity, an important indicator of lake hydrological conditions, is critical to deciphering terrestrial paleoclimatic and paleoenvironmental changes. The proportion of C-37:4 alkenone to total C-37 alkenones (%C-37:4) and the relative abundance of archaeol to caldarchaeol (ACE) are promising salinity indices in lacustrine settings. Developing regional calibrations for both indicators is challenging due to limited datasets and a poor understanding of the factors that influence them. Here we present %C-37:4 and ACE data collected from 55 lakes in mid-latitude Asia with a wide salinity range, to evaluate how well the two proxies can be correlated with salinity and to infer potential influencing factors. The %C-37:4 values only show a weak negative correlation with salinity from all investigated lakes. The occurrence of C-37:3 alkenone isomer and abnormal values in the alkenone C37/C38 ratio might signal species shifts or environmental factors that compromise the %C-37:4-salinity relationship in our data set. Seasonal bias in alkenone production could further obscure this %C-37:4-salinity relationship. A stronger relationship emerges after removing samples influenced by these factors: %C-37:4 = -8.56 * log(10) (salinity) + 80.6, r(2) = 0.62, n = 37; or %C-37:4 = -13.46 * log(10) (salinity) + 101.48, with uncertainty in both variables considered. The ACE values show a strong positive correlation with salinity from all investigated lakes: ACE = 2.27 * 10(-4)* salinity + 25.4, r(2) = 0.75, n = 68; or ACE = 1.86 * 10(-4)* salinity + 38.1, with uncertainty in both variables considered. However, substantial deviations of the ACE values in low salinity range (<60,000 mg/L) and majority of the ACE shift in the range of 60,000-100,000 mg/L suggest that the ACE primarily responds to Euryarchaeota/Archaea community changes when a salinity threshold is crossed. Accordingly, both lake salinity indicators have the potential to reconstruct past salinity changes when their influencing factors could be constrained, and the two independent proxies, when they are used together, could further refine salinity reconstructions. Abstract:Freshwaters worldwide are under increasing pressure from anthropogenic activities and changing climate. Unfortunately, many inland waters lack sufficient long-term monitoring to assess environmental trends. Analysis of sedimentary ancient DNA ( sedaDNA) is emerging as a means to reconstruct the past occurrence of microbial communities of inland waters. The purpose of this study was to assess a combination of high-throughput sequencing (16S rRNA) of sedaDNA and traditional paleolimnological analyses to explore multidecadal relationships among cyanobacterial community composition, the potential for cyanotoxin production, and paleoenvironmental proxies. DNA was extracted from two sediment cores collected from a northern Canadian Great Plains reservoir. Diversity indices illustrated significant community-level changes since reservoir formation. Furthermore, higher relative abundances in more recent years were observed for potentially toxic cyanobacterial genera including Dolichospermum. Correlation-based network analysis revealed this trend significantly and positively correlated to abundances of the microcystin synthetase gene ( mcyA) and other paleoproxies (nutrients, pigments, stanols, sterols, and certain diatom species), demonstrating synchrony between molecular and more standard proxies. These findings demonstrate a novel approach to infer long-term dynamics of cyanobacterial diversity in inland waters and highlight the power of high-throughput sequencing to reconstruct trends in environmental quality and inform lake and reservoir management and monitoring program design.

Abstract:Fungi are crucial organisms in most ecosystems as they exert ecological key functions and are closely associated with land plants. Fungal community changes may, therefore, help reveal biodiversity changes in past ecosystems. Lake sediments contain the DNA of organisms in the catchment area, which allows reconstructing past biodiversity by using metabarcoding of ancient sedimentary DNA. We re- evaluated various commonly used metabarcoding primers, and we developed a novel PCR primer combination for fungal metabarcoding to produce a short amplicon, thus accounting for amplification bias due to the degradation of ancient DNA. In silico PCRs showed higher diversity using this new primer combination, compared with previously established fungal metabarcoding primers. We analyzed data from sediment cores from four artic and one boreal lake in Siberia. These cores had been stored for 2– 22 years after coring; we, therefore, examined the degradation effects of ancient DNA and storage time- related bias affecting fungal communities. Amplicon lengths showed considerable variation within and between the major divisions of fungi, for example, amplicons of Basidiomycota were significantly longer than those of Mucoromycota; however, we observed no significant effect of sample age on amplicon length and GC content, suggesting the robustness of our results. We also found no indication of post- coring fungal growth during storage regarding the proportions of common mold taxa, which would otherwise distort conclusions on past fungal communities. Abstract:Human-induced transitions in vegetation community and plant diversity have shown far-reaching ecological and socioeconomic consequences worldwide during the Anthropocene. Sedimentary ancient DNA (sedaDNA) is increasingly used to study paleoecological dynamics; however, this promising approach has never been directly compared with pollen records for subtropical floristic reconstruction, particularly in intensively modified landscapes. Here, we conducted high-resolution sedaDNA and pollen analysis on Lake Taihu sediment archives from the eastern China floodplain, together with contemporary surveys, to examine the evolutionary dynamics in the terrestrial and aquatic plant communities spanning the past hundred years. Terrestrial plant DNA was successfully amplified and 57 taxa were well identified through the sediment core, but the molecular detection of aquatic macrophytes was subject to some issues associated with hydrological fluctuations and the reference library/primer used. The plant diversity recovered from sedaDNA showed a specific local signal and greater taxonomic resolution for crops and grasses related to cultivated land use, while the pollen records mainly captured regional terrestrial vegetation development especially forestry as well as mixed aquatic plant information mostly linked to littoral habitats. The results demonstrate that these two proxies are complementary rather than overlapping in the subtropical floodplain regions, and in combination reflect more detailed flora changes. Our comprehensive records reveal human-dominated terrestrial vegetation turnover and increase in diversity since the 1960s from the local to regional level. The succession and degradation of littoral aquatic plant community was probably attributed to hydrodynamic conditions and cultural eutrophication. Our finding highlights the broad prospects of combining morphological and molecular approaches for paleoecological research, and adds to the increasing evidence supporting that recent terrestrial and aquatic ecosystem dynamics are dominated by anthropogenic environmental changes.
Lake sedimentary DNA accurately records 20th Century introductions of exotic conifers in Scotland

Abstract:Sedimentary DNA (sedDNA) has recently emerged as a new proxy for reconstructing past vegetation, but its taphonomy, source area and representation biases need better assessment. We investigated how sedDNA in recent sediments of two small Scottish lakes reflects a major vegetation change, using well-documented 20th Century plantations of exotic conifers as an experimental system. We used next-generation sequencing to barcode sedDNA retrieved from subrecent lake sediments. For comparison, pollen was analysed from the same samples. The sedDNA record contains 73 taxa (mainly genus or species), all but one of which are present in the study area. Pollen and sedDNA shared 35% of taxa, which partly reflects a difference in source area. More aquatic taxa were recorded in sedDNA, whereas taxa assumed to be of regional rather than local origin were recorded only as pollen. The chronology of the sediments and planting records are well aligned, and sedDNA of exotic conifers appears in high quantities with the establishment of plantations around the lakes. SedDNA recorded other changes in local vegetation that accompanied afforestation. There were no signs of DNA leaching in the sediments or DNA originating from pollen.

Abstract:Fungi are ecologically important in several ecosystem processes, yet their community composition, ecophysiological roles, and responses to changing environmental factors in historical sediments are rarely studied. Here we explored ancient fungal DNA from lake Lielais Svētiņu sediment throughout the Holocene (10.5 kyr) using the ITS metabarcoding approach. Our data revealed diverse fungal taxa and smooth community changes during most of the Holocene with rapid changes occurring in the last few millennia. More precisely, plankton parasitic fungi became more diverse from the Late Holocene (2–4 kyr) which could be related to a shift towards a cooler climate. The Latest Holocene (~2 kyr) showed a distinct increase in the richness of plankton parasites, mycorrhizal, and plant pathogenic fungi which can be associated with an increased transfer rate of plant material into the lake and blooms of planktonic organisms influenced by increased, yet moderate, human impact. Thus, major community shifts in plankton parasites and mycorrhizal fungi could be utilized as potential paleo-variables that accompany host-substrate dynamics. Our work demonstrates that fungal aDNA with predicted ecophysiology and host specificity can be employed to reconstruct both aquatic and surrounding terrestrial ecosystems and to estimate the influence of environmental change.
