Abstract:Endowing biomaterials with functional elements enhances their biological properties effectively. However, improving bioactivity and biosafety simultaneously is still highly desirable. Herein, cerium (Ce) and copper (Cu) are incorporated into silicocarnotite (CPS) to modulate the constitution and microstructure for degradability, bioactivity and biosafety regulation. Our results demonstrated that introducing Ce suppressed scaffold degradation, while, co-incorporation of both Ce and Cu accelerated degradability. Osteogenic effect of CPS in vitro was promoted by Ce and optimized by Cu, and Ce-induced angiogenic inhibition could be mitigated by cell coculture method and reversed by Ce-Cu co-incorporation. Ce enhanced osteogenic and angiogenic properties of CPS in a dose-dependent manner in vivo, and Cu-Ce coexistence exhibited optimal bioactivity and satisfactory biosafety. This work demonstrated that coculture in vitro was more appropriately reflecting the behavior of implanted biomaterials in vivo. Interactive effects of multi-metal elements were promising to enhance bioactivity and biosafety concurrently. The present work provided a promising biomaterial for bone repair and regeneration, and offered a comprehensive strategy to design new biomaterials which aimed at adjustable degradation behavior, and enhanced bioactivity and biosafety.
Bioadaptable bone regeneration of Zn-containing silicocarnotite bioceramics with moderate biodegradation and antibacterial activity

Abstract:Preparing novel bioceramics with bioadaptability of high osteogenesis and moderate degradation is particularly challenging. In this work, zinc containing silicocarnotite (Ca5(PO4)2SiO4, CPS) scaffolds (Zn-CPS) were applied as a potential novel therapy for bone defect since the good antibacterial capacity and biocompatibility of Zn. The effects of ZnO content on antibacterial capacity, protein adsorption, osteogenesis and degradation behavior of Zn-CPS bioceramics were investigated and the related mechanism was also discussed. The obtained Zn-CPS bioceramics showed enhanced antibacterial capacity, osteogenesis and degradation. In general, the antibacterial capacity and osteogenesis of Zn-CPS improved with the increase of ZnO content, while the degradation behavior of Zn-CPS exhibited different laws in vitro and in vivo. Specifically, after implantation for 12 weeks, the bone formation percent and degradation percent of 5Zn-CPS could up to 43.0% and 50.9%, indicating an excellent bioadaptability. Moreover, proteomics analysis of adsorbed proteins on Zn-CPS indicated that the addition of ZnO into CPS may activate PI3K-Akt signaling pathway to promote osteogenesis, and degradation contrast in vitro and in vivo also demonstrated that in vivo related factors, such as proteins, cells and enzymes played the crucial role in degradation. These findings imply that Zn-CPS bioceramics are promising materials for bone defect repair with infections.

Evaluation of the antibacterial properties and in-vitro cell compatibilities of doped copper oxide/hydroxyapatite composites

Abstract:Mg, Zn and Ce-doped CuO/HA composites were prepared by a two-step sol-gel and hydrothermal process. SEM images showed a spherical appearance of HA and a needle-like morphology for doped CuO. XRD patterns revealed that all doped CuO/HA composites exhibited a hexagonal crystal structure of HA and a monoclinic crystal structure of CuO with no impurities. ICP analysis indicated that with the increase of loading amount of doped CuO, the concentrations of Cu2+ ions and doping ions released from composites increased. Moreover, CuO/HA composites exhibit improved antibacterial properties against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) as compared with HA. When the loading amount of doped CuO in composites increased to 15 wt%, the composites exhibited the best antibacterial activity and complete bacterial growth inhibition effect. Furthermore, the CCK-8 assay revealed that the doped CuO/HA composites are noncytotoxic and can promote the proliferation of osteosarcoma cells. This work highlights the potential of the doped CuO/HA composites with significant antibacterial activity, bioactivity and cell compatibility for potential biomedical applications in dental implants and bone regeneration.
Antibacterial and cell-friendly copper-substituted tricalcium phosphate ceramics for biomedical implant applications

Abstract:The development of new materials with antibacterial properties and the scope to decrease or eliminate the excessive antibiotic use is an urgent priority due to the growing antibiotic resistance-related mortalities. New bone substitute materials with intrinsic antibacterial characteristics are highly requested for various clinical applications. In this study, the choice of copper ions as substitutes for calcium in tricalcium phosphate (TCP) has been justified by their pronounced broad-spectrum antibacterial properties. Copper-substituted TCP (Cu-TCP) ceramics with the copper content of 1.4 and 0.1 wt% were synthesized by mechano-chemical activation. X-ray diffraction (XRD) analyses established that both pure and copper-containing compounds adopted the structure of whitlockite (ll-TCP). XRD and electron paramagnetic resonance (EPR) spectroscopy revealed the partial isovalent substitution of calcium ions with copper ions in the ll-TCP lattice. With the use of infrared and EPR spectroscopies, it was detected that carbonate ions got incorporated into the ll-TCP structure during the synthesis procedure. By releasing the tension in the M(5)O6 octahedron consequential to the lower Ca-O bond length than the corresponding sum of ionic radii, the substitution of calcium with smaller copper ions stabilizes the structure of ll-TCP. As concluded form the thermal analyses, the introduction of Cu prevented the polymorphic transformation of ll- to a-TCP. At the same time, the introduction of Cu to the ll-TCP structure enhanced the crystal growth and porosity of the ceramics, which had a positive effect on the cytocompatibility of the material. The MTT colorimetric assay showed that the metabolic activity of the mouse fibroblast NCTC L929 cell line during 24 h of incubation with 3-day extracts from Cu-TCP (1.4 wt%) and ll-TCP pellets in the cell culture medium was similar to the negative control, indicating the absence of any inhibitory effects on cells. The seeding and the growth of human dental pulp stem cells on the surface of Cu-TCP (1.4 wt%) and ll-TCP ceramics also showed the absence of any signs of cytotoxicity. Finally, microbiological assays demonstrated the antibacterial activity of CuTCP ceramics against Escherichia coli and Salmonella enteritidis, whereas ll-TCP did not exhibit such an activity. Overall, the addition of Cu ions to ll-TCP improves its antibacterial properties without diminishing the biocompatibility of the material, thus making it more attractive than pure ll-TCP for clinical applications such as synthetic bone grafts and orthopaedic implant coatings.
Bioceramic scaffolds with two-step internal/external modification of copper-containing polydopamine enhance antibacterial and alveolar bone regeneration capability

Abstract:Magnesium-doped calcium silicate (CS) bioceramic scaffolds have unique advantages in mandibular defect repair; however, they lack antibacterial properties to cope with the complex oral microbiome. Herein, for the first time, the CS scaffold was functionally modified with a novel copper-containing polydopamine (PDA(Cu2+‍)) rapid deposition method, to construct internally modified (*P), externally modified (@PDA), and dually modified (*P@PDA) scaffolds. The morphology, degradation behavior, and mechanical properties of the obtained scaffolds were evaluated in vitro. The results showed that the CS*P@PDA had a unique micro-/nano-structural surface and appreciable mechanical resistance. During the prolonged immersion stage, the release of copper ions from the CS*P@PDA scaffolds was rapid in the early stage and exhibited long-term sustained release. The in vitro evaluation revealed that the release behavior of copper ions ascribed an excellent antibacterial effect to the CS*P@PDA, while the scaffolds retained good cytocompatibility with improved osteogenesis and angiogenesis effects. Finally, the PDA(Cu2+)-modified scaffolds showed effective early bone regeneration in a critical-size rabbit mandibular defect model. Overall, it was indicated that considerable antibacterial property along with the enhancement of alveolar bone regeneration can be imparted to the scaffold by the two-step PDA(Cu2+) modification, and the convenience and wide applicability of this technique make it a promising strategy to avoid bacterial infections on implants.
Osteogenic, Angiogenic, and Antibacterial Bioactive Nano-Hydroxyapatite Co-Synthesized Using γ-Polyglutamic Acid and Copper

Abstract:Nano-antibacterial calcium phosphate (CaP) has attracted intense attention with regard to its wide variety of medical and biological applications. The γ-polyglutamic acid and copper cosynthesized hydroxyapatite (γ-PGA/CuxHAp) was synthesized using the wet method. Structural and chemical characterizations demonstrate that copper was quantitatively incorporated into the hydroxyapatite structure, and the degree of Cu substitution was up to 20 mol % in the synthesized nanocrystals. Morphology characterization showed that the size of the γ-PGA/CuxHAp nanoparticles decreases with the increased copper content. γ-PGA/CuxHAp exhibited a steady release of Cu ions. Two experimental protocols were applied to compare the antibacterial activity of the γ-PGA/CuxHAp samples. A positive correlation was observed between Cu content and the inhibition of bacterial growth. The study also showed that nanoparticles with smaller particle sizes exhibited higher antibacterial activities than the larger particles. Endothelial and osteoblast cells rapidly proliferated on γ-PGA/CuxHAp, whereas high concentrations (20 mol %) of Cu ions reduced cell proliferation. In the rat calvarial defect model, some γ-PGA/CuxHAp samples such as γ-PGA/CuxHAp (x = 8, 16) showed efficient bone regeneration capacities at 12 weeks post implantation. Thus, the multibiofunctional γ-PGA/CuxHAp nanocomposite exhibited degradative, angiogenic, bactericidal and bone regenerative properties, providing a potential means to address some of the critical challenges in the field of bone tissue engineering.

Abstract:Nano-sized copper(II) oxide (CuO) is an interesting material with proficient antibacterial activities but limited due to its toxicity to mammalian cells. In this study, CuO nano-sized needles were coated on the surface of nano-silicate platelets (NSP), which were exfoliated from natural montmorillonite clays and with the thickness of about 1 nm, to form nanohybrids. Our findings indicate that NSPs can effectively prevent the aggregation of CuO needles, and significantly enhance antibacterial efficiency without affecting biocompatibility. An optimal antibacterial activity against Staphylococcus aureus exceeding 99% could be obtained, while that for Escherichia coli was about 88%. Electron paramagnetic resonance results suggest that •OH radicals, one of the reactive oxygen species generated by CuO/NSP, might damage the cellular components and thus result in the death of bacterial.

Abstract:Journal of Biomaterials Applications, Ahead of Print. Atomic substitution or doping of a bioceramic material hydroxyapatite (HA) with specific ions is an appealing approach for improving its biocompatibility and activity, as well as imparting antibacterial properties. In this study, selenium- and/or copper-substituted hydroxyapatite powders were synthesized by an aqueous precipitation method and using the freeze-drying technique. The molar concentrations of constituents were calculated based on the proposed mechanism whereby selenium (Se4+) ions partially substitute phosphorus (P5+) sites, and copper (Cu2+) ions partially substitute (Ca2+) sites in the HA lattice. Dried precipitated samples were characterized using Inductively coupled plasma optical emission spectroscopy (ICP-OES), X-ray diffraction analysis (XRD), Fourier-transform infrared spectroscopy (FTIR) and Field-emission scanning electron microscopy with energy dispersive X-ray spectroscopy (FESEM-EDX). Accordingly, substitution of Se4+ and/or Cu2+ ions took place in the crystal lattice of HA without the formation of any impurities. The presence of sulphur (S2-) ions in the hydroxyapatite was detected by ICP-OES in all samples with copper substituted in the lattice. The cytotoxicity of the powders on osteoblastic (MC3T3-E1) cells was evaluated in vitro. Selenium substituted hydroxyapatite (SeHA), at the concentration (200 μg/mL), demonstrated higher populations of the live cells than that of control (cells without powders), suggesting that selenium may stimulate the proliferation of these cells. In addition, the copper substituted hydroxyapatite (CuHA) and the selenium and copper substituted hydroxyapatite (SeCuHA) at the concentrations (200 and 300 μg/mL) and (200 μg/mL), respectively demonstrated better results than the unsubstituted HA. Antimicrobial activity was assessed using a well-diffusion method against Streptococcus mutans and Candida albicans, and superior results has obtained with SeCuHA samples. Presented findings imply that selenium and/or copper substituted modified hydroxyapatite nanoparticles, may be an attractive antimicrobial and cytocompatible substrate to be considered for use in a range of translational applications.

Abstract:An ideal artificial bone will likely be multifunctional, combining different technologies to simultaneously promote bone regeneration while inhibiting microbial infection. In this study, copper- (Cu-) doped nano laponite (cnLAP) was prepared by a cation-exchanged method, and the cnLAP coating on poly(butylene succinate) (PBSu) scaffold was fabricated by poly(dopamine) modification. The results showed that incorporation of Cu ions into nano laponite (nLAP) did not have obvious effects on the morphology and surface area of cnLAP (compared with nLAP), which could be coated easily on macroporous PBSu scaffolds. In addition, the cnLAP-coated PBSu scaffolds could inhibit the growth of both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) , indicating good antibacterial activity. Moreover, the cnLAP-coated PBSu scaffolds significantly promoted proliferation and improved alkaline phosphatase (ALP) activity of bone mesenchymal stem cells (BMSCs) compared with PBSu scaffolds. Furthermore, no obvious differences in cell responses to cnLAP- and nLAP-coated PBSu scaffolds were found, indicating that incorporation of Cu into nLAP had no negative effects on its cytocompatibility. The results suggested that the cnLAP-coated PBSu scaffolds exhibited excellent cytocompatibility and antimicrobial activity, which might offer promising opportunities for promoting bone regeneration and prevention of infectious from bacteria and effective treatment of bone defects.

High performing additively manufactured bone scaffolds based on copper substituted diopside

Abstract:The inclusion of small amounts of copper is often reported to enhance the mechanical and biointegrative performance of bioceramics towards tissue engineering applications. In this work, 3D scaffolds were additively manufactured by robocasting of precipitation derived copper doped diopside. Compositions were chosen in which magnesium sites in diopside were substituted by copper up to 3 at.%. Microstructure, mechanical performance, bioactivity, biodegradability, drug release, biocompatibility, in vitro angiogenesis and antibacterial activity were studied. Results indicate that copper is incorporated in the diopside structure and improves materials fracture toughness. Scaffolds with more than 80% porosity exhibited compressive strengths exceeding that of cancellous bone. All compositions showed bioactivity and drug release functionalities. However, only samples with 0 to 1 at.% copper substitution showed favorable proliferation of osteogenic sarcoma cells, human umbilical vein endothelial cells and fibroblasts, while larger amounts of copper had cytotoxic behavior. In vitro angiogenesis was significantly enhanced by low levels of copper. Copper containing materials showed anti Escherichia coli activity, increasing with copper content. We show that across multiple indicators, copper substituted diopside of the composition CaMg0.99Cu0.01Si2O6, exhibits high performance as a synthetic bone substitute, comparing favorably with known bioceramics. These findings present a pathway for the enhancement of bioactivity and mechanical performance in printable bioceramics.
Bioactive and biocompatible copper containing glass-ceramics with remarkable antibacterial properties and high cell viability designed for future in vivo trials

Abstract:In the present study our interest is focused on finding the efficiency of 60SiO2·(32 - x)CaO·8P2O5·xCuO (mol%) glass-ceramics, with 0 ≤ x ≤ 4 mol%, in terms of bioactivity, biocompatibility, antibacterial properties and cell viability in order to determine the most appropriate composition for their further use in in vivo trials. The sol-gel synthesized samples show a preponderantly amorphous structure with a few crystallization centers associated with the formation of an apatite and calcium carbonate crystalline phases. The Fourier Transform Infrared (FT-IR) spectra revealed slightly modified absorption bands due to the addition of copper oxide, while the information derived from the measurements performed by transmission electron microscopy, UV-vis and electron paramagnetic resonance spectroscopy showed the presence of ions and metallic copper species. X-Ray photoelectron spectroscopic analysis indicated the presence of copper metallic species, in a reduced amount, only on the sample surface with the highest Cu content. Regarding in vitro assessment of bioactivity, the results obtained by X-ray diffraction, FT-IR spectroscopy and scanning electron microscopy, demonstrated the formation of a calcium phosphate layer on all investigated sample surfaces. The inhibitory effect of the investigated samples was more significant on the Pseudomonas aeruginosa than the Staphylococcus aureus strain, the sample with the lowest concentration of copper oxide (0.5 mol%) being also the most efficient in both bacterial cultures. This sample also exhibits a very good bactericidal activity, for the other samples it was necessary to use a higher quantity to inhibit and kill the bacterial species. The secondary structure of adsorbed albumin presents few minor changes, indicating the biocompatibility of the glass-ceramics. The cell viability assay shows a good proliferation rate on samples with 0.5 and 1.5 mol% CuO, although all glass-ceramic samples exhibited a good in vivo tolerance.
Copper sulfide anchored MXene improving photo-responsive self-healing polyurethane with enhanced mechanical and antibacterial properties

Abstract:Diseases caused by bacterial infection are becoming a major threat to human health. Therefore, developing efficient antibacterial materials is of great significance in improving medical care and protecting people's health. In this work, an accordion-like structural Ti3C2@CuS was synthesized by copper sulfide (CuS) nanospheres anchored firmly on the surface of Ti3C2Tx via the hydrothermal method. The multilayer Ti3C2@CuS becomes few-layered nanosheets after ultrasonic treatment, which have an enjoyable dispersion in the polyurethane (PU) matrix. PU and the released Cu2+ from Ti3C2@CuS are firmly linked by a coordination bond, which improves the mechanical properties and thermal stability of Ti3C2@CuS-PU and reduces the heavy metal ion pollution by blocking the Cu2+ released by forming coordination bonds. Moreover, Ti3C2@CuS-PU exhibits an excellent self-healing performance after 30 tensile cycles. Additionally, Ti3C2Tx and CuS could improve the separation efficiency of the electron-hole pairs of CuS to produce more reactive oxygen species (ROS) to kill bacteria. Ti3C2@CuS-PU maintains a highly long-term sterilization ability of more than 90 % in 30 days because of the synergistic effect of the sustained release of copper ions, the elevated ROS production ability, and the excellent dispersion of Ti3C2@CuS in PU. This work demonstrates a simple and promising route for designing multifunctional antibacterial self-healing materials.
Copper nanoparticles incorporated visible light-curing chitosan-based hydrogel membrane for enhancement of bone repair

Abstract:Alveolar bone defects caused by tumor, trauma and inflammation can lead to the loss of oral function and complicate denture restoration. Currently, guided bone regeneration (GBR) barrier membranes for repairing bone defect cannot effectively promote bone regeneration due to their unstable degradation rate and poor antibacterial properties. Furthermore, they require additional tailoring before implantation. Therefore, this study developed a visible light-curing hydrogel membrane (CF-Cu) comprising methacrylated carboxymethyl chitosan (CMCS-MA), silk fibroin (SF), and copper nanoparticles (Cu NPs) to address these shortcomings of commercial membranes. The CF-Cu hydrogel, characterized by scanning electron microscopy (SEM), a universal testing machine, and swelling and degradation tests, demonstrated a smooth porous network structure, suitable swelling ratio, biodegradability, and enhanced mechanical strength. Cytotoxicity and hemolysis tests in vitro demonstrated excellent cyto- and hemo-compatibility of the CF-Cu hydrogel extracts. Additionally, evaluation of antibacterial properties in vitro, including colony forming unit (CFU) counts, MTT assays, and live/dead fluorescence staining, showed that the CF-Cu hydrogel exhibited excellent antibacterial properties, inhibiting over 80% of S. aureus, S. mutans, and P. gingivalis with CF-1Cu hydrogel compared to the control group. Moreover, evaluation of osteogenic differentiation of rBMSCs in vitro suggested that the CF-1Cu hydrogel significantly improved alkaline phosphatase (ALP) activity and the mineralization of extracellular matrix, up-regulating the expressions of osteogenesis-related genes (Runx2, ALP, Col-1, OPN and BSP). In summary, these results indicated that CF-1Cu hydrogel exhibited excellent cytocompatibility, antibacterial and osteogenic properties in vitro. Therefore, the CF-1Cu hydrogel holds potential as a viable material for application in GBR procedures aimed at addressing bone defects.
Distinct mechanisms of iron and zinc metal ions on osteo-immunomodulation of silicocarnotite bioceramics

Abstract:The immunomodulatory of implants have drawn more and more attention these years. However, the immunomodulatory of different elements on the same biomaterials have been rarely investigated. In this work, two widely used biosafety elements, iron and zinc added silicocarnotite (Ca5(PO4)2SiO4, CPS) were applied to explore the routine of elements on immune response. The immune reactions over time of Fe-CPS and Zn-CPS were explored at genetic level and protein level, and the effects of their immune microenvironment with different time points on osteogenesis were also investigated in depth. The results confirmed that both Fe-CPS and Zn-CPS had favorable ability to secret anti-inflammatory cytokines. The immune microenvironment of Fe-CPS and Zn-CPS also could accelerate osteogenesis and osteogenic differentiation in vitro and in vivo. In terms of mechanism, RNA-seq analysis and Western-blot experiment revealed that PI3K-Akt signaling pathway and JAK-STAT signaling pathways were activated of Fe-CPS to promote macrophage polarization from M1 to M2, and its immune microenvironment induced osteogenic differentiation through the activation of Hippo signaling pathway. In comparison, Zn-CPS inhibited polarization of M1 macrophage via the up-regulation of Rap1 signaling pathway and complement and coagulation cascade pathway, while its osteogenic differentiation related pathway of immune environment was NF-κB signaling pathway.
Polyetheretherketone with Citrate Potentiated Influx of Copper Boosts Osteogenesis, Angiogenesis, and Bacteria-Triggered Antibacterial Abilities

Abstract:Because of the increasing use of biomedical implants, implants associated infections are becoming more prevalent and cause increased morbidity and significant higher healthcare cost. As it is a race for the surface between cells and microbes, achieving a rapid bone regeneration and osseointegration, especially when confronted with implants associated infections, is the primary goal. Herein, we develop a promising solution using porous polyetheretherketone (PEEK) implants incorporated with copper-citrate nanoclusters via bioinspired polydopamine, it can deliver a high dose of bactericides only when and where bacteria appear to eradicate bacteria and release a small amount of copper/citrate under physiological environment to promote osteogenesis and angiogenesis in vitro and in vivo. The synergistic effect between copper and citrate, which is that citrate can facilitate a 2-fold influx of copper into cells, not only heightened the bacteria-killing ability but also encouraged the osteogenesis and angiogenesis performances of implants. This present work provides an effective method to control infections while promoting osseointegration simultaneously, which will show tremendous clinical application and can be a solution to current challenges facing orthopedics.

Abstract:The present study was designed to investigate the relationship between auditory vigilance performance and speed of habituation of the GSR component of the orienting response (OR) to auditory stimuli. Fast habituators (n = 7) displayed a greater rate of vigilance decrement than slow habituators (n = 7), suggesting a more rapid onset of a phase of low arousal in these subjects.

Abstract:Bone cements were prepared by mixing calcium sulfate and copper sulfate in various proportions. We examined physical and physicochemical properties of the copper doped calcium sulfates and the effects of the cements on angiogenesis in vivo. Rod shaped calcium sulfate crystals were visible by scanning electron microscopy in the cement that contained no copper sulfate, whereas plate-like crystals covered the surface of the cement with high copper content. After immersion of the cements in simulated body fluid (SBF) for 1 day, X-ray diffractometric analysis showed that gypsum precipitates had formed in the copper doped calcium sulfate. The compressive strength of the cements increased from 3.3 MPa for pure calcium sulfate to 6.4 MPa for samples with copper sulfate added. Calcium ion release was greatest from pure calcium sulfate, and the rate of copper ion release was higher for cements containing the most copper. We found that 6 weeks after implantation, more blood vessels had formed around the high copper cement than for the copper-free cement. Copper doped calcium sulfate appears to be useful for application to regenerative medicine including wound healing and bone tissue engineering.

Abstract:Adding foreign ions to hydroxyapatite (HAp) is a popular approach for improving its properties. This study focuses on the effects of calcium substitution with copper in HAp. Instead of calcium, copper ions were doped into the structure of hydroxyapatite nanoparticles at 1%, 3%, and 5% concentrations. XRD analysis showed that the amount of substituted copper was less than needed to generate a distinct phase, yet its lattice parameters and crystallinity slightly decreased. Further, the results of degradation tests revealed that copper doping in hydroxyapatite doubled calcium ion release in water. The incorporation of copper into the apatite structure also boosted the HAp zeta potential and FBS protein adsorption onto powders. According to antibacterial investigations, a concentration of 200 mg/ml of hydroxyapatite containing 5% copper was sufficient to effectively eradicate E. coli and S. aureus bacteria. Furthermore, copper improved hydroxyapatite biocompatibility. Alkaline phosphatase activity and alizarin red tests showed that copper in hydroxyapatite did not inhibit stem cell differentiation into osteoblasts. Also, the scratch test demonstrated that copper-containing hydroxyapatite extract increased HUVEC cell migration. Overall, our findings demonstrated the utility of incorporating copper into the structure of hydroxyapatite from several perspectives, including the induction of antibacterial characteristics, biocompatibility, and angiogenesis.

Abstract:A facile method to prepare cuprous oxide/bovine serum albumin (Cu 2 O/BSA) hierarchical nanocomposite particles (NCPs) through imitating biomineralization is presented. BSA acted as the structure-directing agents, guiding the nucleation, growth and assembly of Cu(OH) 2 at binding sites to construct the hierarchical structure and then Cu 2 O/BSA NCPs were achieved. The synthesized nanocomposite with hierarchical structure exhibited excellent antibacterial performance. The products obtained were characterized with XRD, TEM, FTIR, EDX and XPS to investigate the formation mechanism of Cu 2 O/BSA hierarchical structure. Cu 2 O/BSA NCPs with size of 20–50 nm built up by several 6–7 nm Cu 2 O nanocrystals with BSA showed better antibacterial performance in the comparative experiments, which was attributed to the special structure and good biocompatibility of the Cu 2 O/BSA NCPs. And the possible mechanism was presented to explain the excellent antibacterial properties of Cu 2 O/BSA NCPs.
