Abstract:A perfect mimic of human bone is very difficult. Still, the latest advancement in biomaterials makes it possible to design composite materials with morphologies merely the same as that of bone tissues. In the present work is the fabrication of selenium substituted Hydroxyapatite (HAP-Se) covered by lactic acid (LA)—Polyethylene glycol (PEG)—Aspartic acid (AS) composite with the loading of vincristine sulfate (VCR) drug (HAP-Se/LA-PEG-AS/VCR) for twin purposes of bone regenerations. The HAP-Se/LA-PEG-AS/VCR composite coated on titanium implant through electrophoretic deposition (EPD). The prepared composite characterized using FTIR, XRD techniques to rely on the composites' chemical nature and crystalline status. The morphology of the composite and the titanium plate with the composite coating was investigated by utilizing SEM, TEM instrument techniques, and it reveals the composite has porous morphology. The drug (VCR) load in HAP-Se/LA-PEG-AS and releasing nature were investigated through UV-Visible spectroscopy at the wavelength of 295 nm. In vitro study of SBF treatment shows excellent biocompatibility to form the HAP crystals. The viability against MG63 and toxicity against Saos- 2 cells have expressed the more exceptional biocompatibility in bone cells and toxicity with the cancer cells of prepared composites. The in-vivo study emphasizes prepared biomaterial suitable for implantation and helps accelerate bone regeneration on osteoporosis and osteosarcoma affected hard tissue.

Abstract:Osteogenic differentiation is great significance for improving the bone regeneration. Present study evaluates the osteogenic ability of lanthanum (La3+) and silicate (SiO44-) substituted hydroxyapatite (MHAP) - polymeric composite coated surface treated titanium (Ti) implant. The bio-ceramic MHAP was synthesized by hydrothermal process with assistance of calcium alginate template. For enhance the hydrophilicity, the polymer poly (vinyl pyrrolidone) (PVP) was included in the composite by ultra-sonication method. The negative zeta potential value -9.97 mV of Ca-alg/ La, Si-HAP was observed after the incorporation of PVP in the matrix. Incorporation of minerals and PVP polymer was confirmed and analyzed by Energy Dispersive X-ray analysis (EDX), Fourier Transform Infra-Red spectroscopy (FT-IR) and Electron Microscopy techniques. A compact coating of the composite with the thickness of 448 nm on Ti surface was achieved by Electrophoretic deposition (EPD) method. The in-vitro MTT assay method and alkaline phosphate ALP activity (94% and 0.94 a.u respectively for the optimized composite) were utilized to determine the cell viability and differentiation on human Bone Marrow-Derived Stem Cells (hBMSCs). The osteogenic ability of bio-composite coated Ti in hBMSCs and in-vivo rat model has strongly suggests the fabricated Ti plate with bio-composite coatings can act as promising biomaterial for orthopedics.

Abstract:INTRODUCTION Adequate hydration status in the sport is essential for good health, yet the relationship between hydration, proteinuria and sports is little studied. OBJECTIVE To analyze the influence of an isotonic sports drink as rehydration strategy on the hydration status and proteinuria after karate training. METHODS Ten athletes participated in this study. In the first session of standard training, called observation training session (STO), the athletes hydrated themselves according to their habits, and in the second session of standard training, called nutritional intervention training session (STIN), an ideal practice of hydration protocol was followed, using an isotonic sports drink as a rehydration liquid during the training. The hydration status was verified by monitoring the body weight before and after training, the urine specific gravity pre-and post-training and the urine volume post-training. To observe the influence of practice of hydration on the renal function post exercise proteinuria was measured. RESULTS We observed a statistically significant difference in urine density between the samples pre- and post-exercise only on STIN (p = 0.047). When we compare the sessions, there was a lower variation in body weight (p = 0.011) and higher urinary volume (p < 0.001), on nutritional intervention training. In STO, there was a higher percentage of athletes who showed proteinuria (70%) compared to the STIN (50%) in the urine sample after training. CONCLUSION The use of isotonic sports drink as practice of hydration by karate athletes promoted rehydration during one session of training and reduce post-training proteinuria.
Promotion of Osteogenic Differentiation of Stem Cells and Increase of Bone-Bonding Ability in Vivo Using Urease-Treated Titanium Coated with Calcium Phosphate and Gelatin

Abstract:Because of its excellent biocompatibility and low allergenicity, titanium has been widely used for bone replacement and tissue engineering. To produce a desirable composite with enhanced bone response and mechanical strength, in this study bioactive calcium phosphate (CaP) and gelatin composites were coated onto titanium (Ti) via a novel urease technique. The cellular responses to the CaP/gelatin/Ti (CaP/gel/Ti) and bone bonding ability were evaluated with proliferation and osteogenic differentiation of mesenchymal stem cells (MSCs) on CaP/gel/Ti and CaP/Ti in vitro. The results showed that the optical density values, alkaline phosphatase expression and genes expression of MSCs on CaP/gel/Ti were similar to those on CaP/Ti, yet significantly higher than those on pure Ti (p < 0.05). CaP/gel/Ti and CaP/Ti rods (2 mm in diameter, 10 mm in length) were also implanted into femoral shaft of rabbits and pure Ti rods served as control (n = 10). Histological examination, scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) measurements were performed at 4 and 8 weeks after the operation. The histological and SEM observations demonstrated clearly that more new bone formed on the surface of CaP/gel/Ti than in the other two groups at each time point. The CaP/gel/Ti bonded to the surrounding bone directly with no intervening soft tissue layer. An interfacial layer, containing Ti, Ca and P, was found to form at the interface between bone and the implant on all three groups by EDS analysis. However, the content of Ca, P in the surface of CaP/gel/Ti implants was more than in the other two groups at each time point. The CaP/gel/Ti modified by the urease method was not only beneficial for MSCs proliferation and osteogenic differentiation, but also favorable for bone bonding ability on Ti implants in vivo, suggesting that Ti functionalized with CaP and gelatin might have a great potential in clinical joint replacement or dental implants.

Abstract:For development of thin bioactive coatings on metal implants, a dense and uniform apatite layer was coated onto titanium (Ti) implants in situ by using a new biomimetic method, which is composed of apatite nucleation and growth steps in simulated body fluid (SBF). Analysis of the coatings by thin film X-ray diffraction and scanning electron microscopy-energy dispersive X-ray microanalysis (SEM-EMPA) before implantation showed that its characteristics were very similar to those of natural bone. The coated and uncoated rectangular plates were bilaterally implanted into the tibial proximal metaphyses of rabbits. After 6, 10 and 25 weeks post-implantation, the bone bonding and bone formation at the bone-implant interfaces were evaluated by a detachment test and undecalcified histological examination. Mechanical testing in tension showed that the failure load of apatite layer-coated Ti implants was significantly higher than that of uncoated control at each time period (all P < 0.001). Histologically, it was shown that bone was deposited directly onto the apatite coating without any intervening soft tissue, while in the paired controls, interpositional soft tissue was seen at the bone-implant interface. By SEM-EPMA, a uniform calcium- and phosphorus-rich layer was detected between the coated implants and bone, but not in uncoated controls at either earlier or later time periods. The results indicate that the apatite layer deposited on Ti in situ may significantly increase the bone bonding strength by providing a bioactive surface, which allows for an early bone apposition to the implant. In addition, the apatite layer-coated Ti produced by the biomimetic process may fulfil the requirements of favourable thin coatings and strong adhesion at the metal-coating interface.

Abstract:Surface modification of titanium (Ti) using biomolecules has attracted much attention recently. In this study, a new strategy has been employed to construct a stable and bioactive coating on Ti. To this end, a derivative of hyaluronic acid (HA), i.e. HA-GRGDSPC-(SH), was synthesized. The disulfide-crosslinked Arg–Gly–Asp (RGD)-containing collagen/hyaluronic acid polyelectrolyte membrane (PEM) coating was then fabricated on Ti through the alternate deposition of collagen and HA-GRGDSPC-(SH) with five assembly cycles and subsequent crosslinking via converting free sulphydryl groups into disulfide linkages (RGD–CHC-Ti group). The assembly processes for PEM coating and the physicochemical properties of the coating were carefully characterized. The stability of PEM coating in phosphate-buffered saline solution could be adjusted by the crosslinking degree, while its degradation behaviors in the presence of glutathione were glutathione concentration dependent. The adhesion and proliferation of MC3T3-E1 cells were significantly enhanced in the RGD–CHC-Ti group. Up-regulated bone specific genes, enhanced alkaline phosphatase activity and osteocalcin production, the increased areas of mineralization were also observed in the RGD–CHC-Ti group. These results indicate that the strategy employed herein may function as an effective way to construct stable, RGD-containing bioactive coatings on Ti.

Abstract:Hydroxyapatite (HA) coatings were introduced onto Poly L-lactic Acid (PLLA) polymer in a controlled manner by immobilized urease method with a shortened precipitation time. Osteoblastic-like cellular responses to the composite were examined in terms of cell proliferation, differentiation and cell morphology, as well as the expression of bone-associated genes. The cells exhibited higher cellular proliferation at 2 and 4 days on the HA/PLLA composite compared to PLLA scaffold, while no significant difference was observed later at 6 days. The alkaline phosphatase (ALP) activity by cells at 7 days was statistically higher on HA/PLLA scaffold than on PLLA. Moreover, the gene expression of ALP and osteocalcin (OC) was up regulated on HA/PLLA composite by RT-PCR analysis. The preliminary study suggested that the use of the controlled modification of hydroxyapatite coating on PLLA scaffold to produce HA/PLLA composite might enhance cellular activity, indicating the potential use for bone substitute in tissue engineering.
Longer-Term Mechanical And Biological Evaluation Of Titanium Alloy Coated With Apatite Layer

Abstract:The objective of this study was the development and use of a biomimetic coating method, which deposits a thin and uniform apatite layer onto titanium (Ti) implants in simulated body fluid (SBF), for improving their bone-bonding ability. The mechanical characteristics and histology of commercially pure Ti and apatite layer-coated Ti-6Al-4V alloy were investigated in rabbit tibiae. Interface failure load was determined using a detaching test after periods of 6, 10, 24, and 32 weeks. The apatite layer-coated implants exhibited significantly higher failure load than the uncoated control at each time period (all p < 0.001). Histologically, the coated-implants bonded directly to bone via the uniform coating, with no intervening soft tissue. In uncoated controls, there was fibrous tissue intervening at the interface even at longer periods. SEM-EPMA demonstrated a Ca-P-rich layer on the coated implants and direct bonding at the interface. No sign of degradation and delamination of the coating was seen after periods of up to 32 weeks. Our results suggest that apatite layer coating on Ti alloys in situ may improve bone-to-implant bonding and make it as a promising coating material.

Abstract:For dental/orthopedic implants to achieve better bone apposition and bone-implant bonding, various approaches to improve titanium surfaces have been developed. Recently, a fluoridated hydroxyapatite (FHA) coating on titanium (Ti) implants was made by sol–gel method and shown to be a possible applicative bone implant. The purpose of the current study was to evaluate biological responses and biomechanical bonding strength of FHA coated Ti implants as compared with that of the conventional Ti alloys and hydroxyapatite (HA) coated Ti implants. In vitro assays were made using human osteoblast-like cell (MG63) culture on different implants with cell attachment, morphology and differentiation evaluations. The implant plates were also implanted into the proximal metaphysis of New Zealand White rabbit tibiae. After 8 and 16 weeks implantation, mechanical and histological assessments were performed to evaluate biomechanical and biological behavior in vivo. The results showed that the cell adhesion and cell growth rate on the FHA and HA surface was higher than that on cp Ti surface (p<0.01), and insignificant difference was observed between two coated groups. Mechanical test demonstrated that the FHA implants had a higher interface shear strength than the both controls at 8 and 16 wks, with no significant difference with HA-Ti. Histologically, the coated implants revealed a significantly greater percentage of bone-implant contact when compared with the uncoated implants. Results demonstrated that the new FHA surface improved cell adhesion and proliferation. The coating exhibited a bioactive mechanical and histological behavior at bone-implant interface, suggesting that a useful approach by combined coating processes could optimize implant surfaces for bone deposition and early implant fixation.
Production of an eco-friendly, effective antimicrobial and antifungal from cerium/natural bio-hydroxyapatite coating for titanium plates suitable for bone augmentation

Abstract:As a promising technique for rebuilding bone defects, materials having enhanced osteoductivity and antimicrobial and fungi effects have been used for bone replacement. Accordingly, the present study aims to develop a sintered titanium (Ti) plate prepared by the cold isostatic press (CIP) coated with nano bio-natural hydroxyapatite (n-HA) via freeze-drying technique for bone substitution. Moreover, the influence of cerium chloride heptahydrate doping with different concentrations of 0.5, 2, and 8 wt% was evaluated for enhancing the osteogeneration process and antibacterial activity on titanium (Ti) plates instead of antibiotic-loaded. Physical properties showed that the n-HA/2%CeCl 3 coat is the most suitable concentration with an appropriate porosity of 73.43 ± 3.52 compared to porous n-HA coating Ti plate with a porosity of 90.38 ± 0.27. The biological activity ( in-vitro ) study in terms of following up the apatite nucleation on the blank Ti plate and Ti/n-HA/2 % CeCl 3 surfaces after immersions in SBF for up to 28 days showed the formation of spherical clusters from carbonated apatite particles on the sample's surface, with a Ca/P ratio equal to 2.26. The cytotoxicity test was examined using a bone osteosarcoma cell line (HOS). The final results confirmed minimal cytotoxicity for the plate coated with n-HA/2 % CeCl 3 (3.6 %). Cell attachment results indicate the potential of the cells to adhere and spread on the surface. In addition, the antimicrobial assessment of n-HA/2 % CeCl 3 coated Ti plates showed broad-spectrum activity against not only the gram-positive or gram-negative but also fungal ones.

Abstract:In order to reduce the loosening of dental implants, surface modification with hydroxyapatite (HA) coating has shown promising results. Therefore, in this present study, the sol-gel technique has been employed to form a tantalum and strontium ion-doped hybrid HA layer coating onto the titanium (Ti)-alloy substrate. In this study, the surface modification was completed by using 3% tantalum pent oxide (Ta2O5), 3% strontium (Sr), and a combination of 1.5% Ta2O5 and 1.5% Sr as additives, along with HA gel by spin coating technique. These additives played a prominent role in producing a porous structure layer coating and further cell growth. The MG63 cell culture assay results indicated that due to the incorporation of strontium ions along with tantalum embedded in HA, cell proliferation increased significantly after a 48 h study. Therefore, the present results, including microstructure, crystal structure, binding energy, and cell proliferation, showed that the additives 1.5% Ta2O5 and 1.5% Sr embedded in HA on the Ti–substrate had an optimized porous coating structure, which will enhance bone in-growth in surface-modified Ti-implants. This material had a proper porous morphology with a roughness profile, which may be suitable for tissue in-growth between a surface-modified textured implant and bone interface and could be applicable for dental implants.

Abstract:There is a substantial global market for orthopedic implants, but these implants still face the problem of a high failure rate in the short and long term after implantation due to the complex physiological conditions in the body. The use of multifunctional coatings on orthopedic implants has been proposed as an effective way to overcome a range of difficulties. Here, a multifunctional (TA@HA/Lys) n coating composed of tannic acid (TA), hydroxyapatite (HA), and lysozyme (Lys) was fabricated in a layer-by-layer (LBL) manner, where TA deposited onto HA firmly stuck Lys and HA together. The deposition of TA onto HA, the growth of (TA@HA/Lys) n , and multiple related biofunctionalities were thoroughly investigated. Our data demonstrated that such a hybrid coating displayed antibacterial and antioxidant effects, and also facilitated the rapid attachment of cells [both mouse embryo osteoblast precursor cells (MC3T3-E1) and dental pulp stem cells (DPSCs)] in the early stage and their proliferation over a long period. This accelerated osteogenesis in vitro and promoted bone formation in vivo . We believe that our findings and the developed strategy here could pave the way for multifunctional coatings not only on orthopedic implants, but also for additional applications in catalysts, sensors, tissue engineering, etc.

Abstract:Titanium was functionalized by bionic micro/nano-structure and Sr-HA coating.Functionalized titanium could promote cell adhesion and inhibit osteoclast activity.Functionalized titanium could improve the bone-implant bonding strength.As the population ages, more and more people are suffering from osteoarthritis (OA), resulting in an increasing requirement for joint implants. Surface modification to improve the topology and composition of the implants has been proved to be an effective way to improve the primary stability and long-term success rate of joint implants. In this work, a bionic micro/nano-structure accompanied with a strontium-substituted hydroxyapatite (SrHA) coating was fabricated on titanium (Ti) surface via electrochemical corrosion, ultrasonic treatment, and hydrothermal deposition methods. The in vitro study demonstrated that the bionic structure and the bioactive apatite could synergistically increase the expressions of integrin-related gene (ITG α5β1) and osteoblastic genes (Col-I and OCN), and thus promote osteoblast growth. In addition, owing to the anti-bone resorption property of Sr2+, the coating could effectively inhibit osteoclast differentiation and proliferation. In a word, the prepared samples not only promoted osteogenesis but also inhibited osteoclastogenesis. The in vivo experiment via a rabbit model found that the bionic structured surface provided the pore for new bone ingrowth, which was beneficial to the mechanical interlocking between the implant and bone. Moreover, the bionic structure and bioactive SrHA coating had a synergistic effect on promoting bone formation, osseointegration, and bone-implant bonding strength. Preparation of Cu 2+ /TA/HAP composite coating with anti-bacterial and osteogenic potential on 3D-printed porous Ti alloy scaffolds for orthopedic applications

Abstract:Because of stress shielding effects, traditional titanium (Ti) alloy scaffolds have a high elastic modulus, which might promote looseness and bone disintegration surrounding the implant, increasing the likelihood of a second surgery. In contrast, 3D-printed porous Ti alloy scaffolds can reduce the scaffold weight while enhancing biocompatibility. Further, these scaffolds' porous nature allows bone tissue ingrowth as well as strong pore connectivity, which can improve nutrient absorption. Nevertheless, bare Ti alloy implants may fail because of inadequate bone integration; hence, adding a coating on the implant surface is an effective technique for improving implant stability. In this study, a composite coating comprising hydroxyapatite (HAP), chitosan (CS), tannic acid (TA) and copper ions (Cu 2+ ) (Cu 2+ /TA/HAP composite coating) was prepared on the surface of 3D printed porous Ti alloy scaffolds using electrophoretic deposition. Using the standard plate count method, Live/Dead bacteria staining assay, FITC Phalloidin and 4′,6-diamidino-2-phenylindole staining assay, and live/dead staining of cells we determined that the composite coating has better antibacterial properties and cytocompatibility as well as lower cytotoxicity. The Alkaline Phosphatase assay revealed that the coating results showed good osteogenesis potential. Overall, the composite coatings produced in this investigation give new potential for the application of Ti alloys in clinics.

Abstract:<p>In this study, we described the facile synthesis of hydroxyapatite (HAP) through thermal decomposition of Rogu fish bones <em>(Labeo Rohita)</em> and this prized biominerals is used for bone implant applications. The HAP coating on Ti6Al4V alloy was performed through electrophoretic deposition (EPD) along with carboxymethyl cellulose (CMC) extracted from rice husk and sodium alginate (SA). The physicochemical and structural behaviour of the HAP and its biocomposites were investigated through various characterization techniques such as fourier transform infrared spectroscopy (FTIR), X-ray powder diffraction (XRD) and field emission scanning electron microscopy (FESEM). The physicochemical characterizations confirmed the formation of phase pure apatite structure. The HAP and its biocomposites coated implants were investigated in simulated body fluid (SBF) solution to determine the biomineralization on the implant surface. The biomineralization process reveals that the HAP/CMC biocomposite containing 2 wt % SA exhibit better apatite formations on the implant surface than the other biocomposite coated implants (1 wt % and 3 wt % SA). Microhardness of the HAP and its biocomposites were investigated through Vicker's microhardness measurements. The antibacterial activity confirms that the as-synthesized HAP does not possess any bacterial activity. Furthermore, the high surface negative charge of SA containing HAP/CMC biocomposite shows better antibacterial activity against the gram-positive strain. The cell proliferation investigation for the 2 wt % SA containing HAP/CMC biocomposite against MG63 cell line exhibits high cell affinity, which reveals that the HAP/CMC/SA composite confirms the biocompatible nature of the biocomposite.</p>

Effect of Bioactivity of Surface Topography and Coating Forming by Infrared Light-Induced on Titanium for Bone Repair

Abstract:Calcium ions and phosphate ions are usually present in biological organisms and human bodies. Different ratios of calcium to phosphorus result in different types of calcium-to-phosphorus crystals. Hydroxyapatite (HA) is the main component of human hard bony tissues. It has good biocompatibility and is often used in bone repair. With the addition of cobalt ions, it can act as a hypoxia-inducing factor to accelerate the regeneration of hard bony tissues. At present, the laser-assisted biomimetic (LAB) method can very quickly deposit calcium phosphate coatings, which can be used on polymer and titanium surfaces. In this study, we first used anodization treatment (with TiO2 nanotubes (TNTs)), alkali treatment (with NaOH), and acid treatment (with HCl) to form nanopore structures on titanium surfaces in the laboratory. Subsequently, LAB treatment was used to deposit calcium phosphate and cobalt-substituted hydroxyapatite onto titanium pieces with different surface treatments. The results showed that smaller holes resulted in better deposition (TNTs), and controlling the pH value in the solution changed the crystal morphology. LAB treatment imbued the titanium surface with super-hydrophilic properties and improved biocompatibility. A human osteoblast cell line (MG-63) used for the cell viability test showed that LAB treatment can improve cell growth. In particular, TNT-CoHA (cobalt-substituted hydroxyapatite) cells grew the best. Immersion in simulated bodily fluid confirmed that LAB treatment with a CoHA solution improved the hydrophilicity, biocompatibility, and bioactivity of titanium surfaces. It is hoped that this study provides useful information for surface coating of biomedical materials in the future.

Abstract:Orthopedic and dental implant failure continues to be a significant concern due to localized bacterial infections. Previous studies have attempted to improve implant surfaces by modifying their texture and roughness or coating them with antibiotics to enhance antibacterial properties for implant longevity. However, these approaches have demonstrated limited effectiveness. In this study, we attempted to engineer the titanium (Ti) alloy surface biomimetically at the nanometer scale, inspired by the cicada wing nanostructure using alkaline hydrothermal treatment (AHT) to simultaneously confer antibacterial properties and support the adhesion and proliferation of mammalian cells. The two modified Ti surfaces were developed using a 4 h and 8 h AHT process in 1 N NaOH at 230 °C, followed by a 2-hour post-calcination at 600 °C. We found that the control plates showed a relatively smooth surface, while the treatment groups (4 h & 8 h AHT) displayed nanoflower structures containing randomly distributed nano-spikes. The results demonstrated a statistically significant decrease in the contact angle of the treatment groups, which increased wettability characteristics. The 8 h AHT group exhibited the highest wettability and significant increase in roughness 0.72 ± 0.08 μm ( P < 0.05), leading to more osteoblast cell attachment, reduced cytotoxicity effects, and enhanced relative survivability. The alkaline phosphatase activity measured in all different groups indicated that the 8 h AHT group exhibited the highest activity, suggesting that the surface roughness and wettability of the treatment groups may have facilitated cell adhesion and attachment and subsequently increased secretion of extracellular matrix. Overall, the findings indicate that biomimetic nanotextured surfaces created by the AHT process have the potential to be translated as implant coatings to enhance bone regeneration and implant integration.

Abstract:Biomaterials are synthetic materials that are utilized to restore or replace damaged or diseased human body parts, allowing them to regain their original form and function to improve the quality and longevity of human life. Titanium and its alloys have long been employed in biomedical applications due to its remarkable features, such as good biocompatibility, resistance to bodily fluid effects, tremendous tensile strength, flexibility, and high corrosion resistance. Titanium and its alloys have a unique combination of strength and biocompatibility that makes them suitable for medical or recreational purposes. If these materials are used as bio-implant, it releases toxic ions like aluminium and vanadium in the body fluid environment after implantation. [1-3] To overcome the problem, Ti6Al4V alloy was coated with hydroxyapatite (HAp), which provides better bioactivity, osteocompatibility, and antibacterial activity. This layer prevents the further passing of ions from the biomaterial and improves the tissue growth on the bone. The present work is to synthesize HAp from snail shells using a simple wet precipitation method.[4] The waste material of snail shells can be utilized for the development of the HAp, which was non-toxic, eco-friendly, and also to improve bioactivity and biocompatibility of the biomaterials. The prepared HAp was coated on the Ti6Al4V alloy by using the electro-deposition method.[5] Thermal analysis of obtained CaO powder was investigated by TG–DTA analysis. The coated alloy was characterized by various techniques such as FTIR, XRD, TG-DTA, FESEM, EDAX, AFM, and antibacterial activity.
Improved Cell Adhesion and Osteogenesis Using a PLTGA (poly L-lactide, 1,3-Trimethylene Carbonate, and Glycolide) Terpolymer by Gelatin-Assisted Hydroxyapatite Immobilization for Bone Regeneration

Abstract:A PLTGA (poly l-lactide, trimethylene carbonate, glycolide) terpolymer possesses great potential for orthopedic applications due to its excellent biocompatibility and controllable biodegradability. However, the unfavorable surface conditions of bare PLTGA such as its poor hydrophilicity and smooth morphology impede its clinical applications, highlighting the need for tailored surface modifications to improve its cytological behavior and osteogenic capacity. We herein develop a facile and effective strategy to deposit a gelatin/hydroxyapatite (GEL/HAP) hybrid coating onto the surface of PLTGA that involves consecutive chemical grafting and in situ reaction steps. Following the surface modification treatment, the resultant PLTGA scaffold with the GEL/HAP coating exhibited drastically improved hydrophilicity (79.1°vs. 48.2° in water contact angle) and increased surface roughness (18.4 vs. 267.9 nm, more than 14-fold, in root-mean-square roughness), respectively. In addition, preosteoblast (MC3T3-E1) cells were seeded onto the bare/modified PLTGA scaffold to evaluate biological performance, including cell adhesion, proliferation, mineralization and osteogenic differentiation. Based on these results, the GEL/HAP hybrid coating can endow the PLTGA terpolymer substrate with enhanced cell adhesion, proliferation and osteogenic functionality. Overall, post-treatment of PLTGA with the GEL/HAP hybrid coating may be a promising methodology in bone regeneration applications.

Abstract:In order to overcome the poor osteo-inductive properties of titanium implant, some methods have been used. The efforts to improve implant biocompatibility and durability by applying a hybrid technique of composite oxidation (pre-anodic and micro-arc oxidation) and hydrothermal treatment were described. Pure titanium was used as the substrate material. An oxalic acid was used as the electrolyte for the pre-anodic oxidation. A calcium and phosphate salt solution was acted as the electrolyte of micro-arc oxidation and the common pure water was used for hydrothermal treatment. X-ray diffraction (XRD), and scanning electron microscopy (SEM) have been used to investigate the microstructure and morphology of the coatings. The results show that a compact TiO2 film can be made by pre-anodic oxidation, which is effective as chemical barriers against the in-vivo release of metal ions from the implants. A porous TiO2 coating can be produced by micro-arc oxidation on titanium plate, which is beneficial to bone tissue growth and enhancing anchorage of implant to bone. De calciurn HA can be formed on the coating using hydrothermal treatment, which is similar with the primary component of bone and has a very good osteo-inductivity. The porous gradient titania coating made by the hybrid oxidation and hydrothermal treatment should show good biocompatibility in the environment of the human body.
