Abstract:Synaptic plasticity is a key component of neuronal dynamics, describing the process by which the connections between neurons change in response to experiences. In this study, we extend a network model of $\theta$-neuron oscillators to include a realistic form of adaptive plasticity. In place of the less tractable spike-timing-dependent plasticity, we employ recently validated phase-difference-dependent plasticity rules, which adjust coupling strengths based on the relative phases of $\theta$-neuron oscillators. We investigate two approaches for implementing this plasticity: pairwise coupling strength updates and global coupling strength updates. A mean-field approximation of the system is derived and we investigate its validity through comparison with the $\theta$-neuron simulations across various stability states. The synchrony of the system is examined using the Kuramoto order parameter. A bifurcation analysis, by means of numerical continuation and the calculation of maximal Lyapunov exponents, reveals interesting phenomena, including bistability and evidence of period-doubling and boundary crisis routes to chaos, that would otherwise not exist in the absence of adaptive coupling.

Synchronicity transitions determine connection fluctuations in a model of coupled oscillators with plasticity

Abstract:Sleep and rest are crucial for knowledge reorganization and creativity. During these periods, synapses between neurons are plastically altered and neuronal activities are collectively synchronized, accompanied by large differences in excitation-inhibition (EI) balance. These processes are assumed to be dissimilar from the learning process during task engagement. The detailed mechanism of how synchronized neuronal activities modify neural circuits via plasticity has yet to be fully understood. The Kuramoto model is utilized to study the collective synchronization of oscillators, including neurons. We previously proposed the EI-Kuramoto model, in which the EI balance was implemented in the Kuramoto model. The model alters its synchronicity based on the EI balance of the interaction strength. In this study, we developed this EI-Kuramoto model by implementing plasticity, leading to the plastic EI-Kuramoto (pEI-Kuramoto) model. Models with high inhibition displayed desynchronized dynamics and consistent connection strengths. Models with low inhibition exhibited bistable dynamics between synchronized and desynchronized states and fluctuation of interaction strengths in middle strength connections, while the strongest connections remained stable. These results, stabilizing a few strong connections and fluctuating the other connections in low inhibition conditions, could facilitate knowledge abstraction and reorganization. Our findings shed light on how varying inhibitory effects influence network stability and coupling, offering deeper insights into synaptic networks and knowledge reshaping.

Abstract:Transient or partial synchronization can be used to do computations, although a fully synchronized network is frequently related to epileptic seizures. Here, we propose a homeostatic mechanism that is capable of maintaining a neuronal network at the edge of a synchronization transition, thereby avoiding the harmful consequences of a fully synchronized network. We model neurons by maps since they are dynamically richer than integrate-and-fire models and more computationally efficient than conductance-based approaches. We first describe the synchronization phase transition of a dense network of neurons with different tonic spiking frequencies coupled by gap junctions. We show that at the transition critical point, inputs optimally reverberate through the network activity through transient synchronization. Then, we introduce a local homeostatic dynamic in the synaptic coupling and show that it produces a robust self-organization toward the edge of this phase transition. We discuss the potential biological consequences of this self-organization process, such as its relation to the Brain Criticality hypothesis, its input processing capacity, and how its malfunction could lead to pathological synchronization.

How effective delays shape oscillatory dynamics in neuronal networks

Abstract:Synaptic, dendritic and single-cell kinetics generate significant time delays that shape the dynamics of large networks of spiking neurons. Previous work has shown that such effective delays can be taken into account with a rate model through the addition of an explicit, fixed delay [Roxin et al. PRL 238103 (2005)]. Here we extend this work to account for arbitrary symmetric patterns of synaptic connectivity and generic nonlinear transfer functions. Specifically, we conduct a weakly nonlinear analysis of the dynamical states arising via primary instabilities of the asynchronous state. In this way we determine analytically how the nature and stability of these states depend on the choice of transfer function and connectivity. We arrive at two general observations of physiological relevance that could not be explained in previous works. These are: 1 - Fast oscillations are always supercritical for realistic transfer functions. 2 - Traveling waves are preferred over standing waves given plausible patterns of local connectivity. We finally demonstrate that these results show a good agreement with those obtained performing numerical simulations of a network of Hodgkin-Huxley neurons.

Geometric Analysis of Synchronization in Neuronal Networks with Global Inhibition and Coupling Delays

Abstract:We study synaptically coupled neuronal networks to identify the role of coupling delays in network's synchronized behaviors. We consider a network of excitable, relaxation oscillator neurons where two distinct populations, one excitatory and one inhibitory, are coupled and interact with each other. The excitatory population is uncoupled, while the inhibitory population is tightly coupled. A geometric singular perturbation analysis yields existence and stability conditions for synchronization states under different firing patterns between the two populations, along with formulas for the periods of such synchronous solutions. Our results demonstrate that the presence of coupling delays in the network promotes synchronization. Numerical simulations are conducted to supplement and validate analytical results. We show the results carry over to a model for spindle sleep rhythms in thalamocortical networks, one of the biological systems which motivated our study. The analysis helps to explain how coupling delays in either excitatory or inhibitory synapses contribute to producing synchronized rhythms.

Abstract:The dynamics of synaptic interactions within spiking neuron networks play a fundamental role in shaping emergent collective behavior. This paper studies a finite-size network of quadratic integrate-and-fire neurons interconnected via a general synaptic function that accounts for synaptic dynamics and time delays. Through asymptotic analysis, we transform this integrate-and-fire network into the Kuramoto-Sakaguchi model, whose parameters are explicitly expressed via synaptic function characteristics. This reduction yields analytical conditions on synaptic activation rates and time delays determining whether the synaptic coupling is attractive or repulsive. Our analysis reveals alternating stability regions for synchronous and partially synchronous firing, dependent on slow synaptic activation and time delay. We also demonstrate that the reduced microscopic model predicts the emergence of synchronization, weakly stable cyclops states, and non-stationary regimes remarkably well in the original integrate-and-fire network and its theta neuron counterpart. Our reduction approach promises to open the door to rigorous analysis of rhythmogenesis in networks with synaptic adaptation and plasticity.
Effects of Synaptic Connectivity Inhomogeneities for Propagation of Activity in Neural Tissue

Abstract:The study of traveling waves of activity in neural tissue can provide deep insights into the functions of the brain during sensory processing or during abnormal states such as epilepsy, migraines or hallucinations. Computational models of these systems usually describe the tissue as a vast interconnected network of neurons comprised of large number of units with similar properties, for example integrate and fire neurons. It is also widely assumed that while the strength of the connections between neurons changes as a function of distance, this interaction does not depend on other local parameters. These assumptions allow for formulation of a set of integro-differential equations describing the propagation of the traveling wave fronts in a one-dimensional integrate-and-fire network of synaptically coupled neurons, allowing for investigation of the network dynamics during wave initiation and propagation. Equations for the transition between initiation and transition toward constant speed traveling waves have been derived for Gaussian connectivity [1] and finite support connectivity [2]. These results have been also confirmed through numerical simulations, leading to methods for optimizing and improving simulations of large-scale networks [3]. These results have been extended beyond the simpler case of one-spike activity propagation, deriving equations for constant speed waves with a finite and infinite number of spikes [4]. This framework has produced insight on the mechanisms of stable constant-speed traveling wave solutions, but the study of inhomogeneities in synaptic connections likely to exist in the brain tissue has received much less attention since not surprisingly, the presence of inhomogeneities vastly increases the complexity of the mathematical models. However, recent work [5,6] used homogenization theory to determine how inhomogeneities can induce propagation failure. We extended our previous models that exhibit constant-speed traveling waves to investigate how the presence of these inhomogeneities affects the relationship between the speed of the activity propagation and its acceleration. We determine that the estimates from homogenization theory do not accurately capture the conditions for propagation failure. More precisely, just prior to stopping, the activity propagates at a higher average speed than predicted from the theoretical results of the homogenization theory. We derive more precise estimates for the conditions when propagation failure occurs. Furthermore, our study points to directions where researchers can obtain additional tools for analyzing experimental data in order to infer details of synaptic connectivity.
Stability of Remote Synchronization in Star Networks of Kuramoto Oscillators

Abstract:Synchrony of neuronal ensembles is believed to facilitate information exchange among cortical regions in the human brain. Recently, it has been observed that distant brain areas which are not directly connected by neural links also experience synchronization. Such synchronization between remote regions is sometimes due to the presence of a mediating region connecting them, e.g., \textit{the thalamus}. The underlying network structure of this phenomenon is star-like and motivates us to study the \textit{remote synchronization} of Kuramoto oscillators, {modeling neural dynamics}, coupled by a directed star network, for which peripheral oscillators get phase synchronized, remaining the accommodating central mediator at a different phase. We show that the symmetry of the coupling strengths of the outgoing links from the central oscillator plays a crucial role in enabling stable remote synchronization. We also consider the case when there is a phase shift in the model which results from synaptic and conduction delays. Sufficient conditions on the coupling strengths are obtained to ensure the stability of remotely synchronized states. To validate our obtained results, numerical simulations are also performed.

Neuromodulatory effects on synchrony and network reorganization in networks of coupled Kuramoto oscillators

Abstract:Neuromodulatory processes in the brain can critically change signal processing on a cellular level leading to dramatic changes in network level reorganization. Here, we use coupled non-identical Kuramoto oscillators to investigate how changes in the shape of phase response curves from Type 1 to Type 2, mediated by varying ACh levels, coupled with activity dependent plasticity may alter network reorganization. We first show that when plasticity is absent, the Type 1 networks, as expected, exhibit asynchronous dynamics with oscillators of the highest natural frequency robustly evolving faster in terms of their phase dynamics. At the same time, the Type 2 networks synchronize, with oscillators locked so that the ones with higher natural frequency have a constant phase lead as compared to the ones with lower natural frequency. This relationship establishes a robust mapping between the frequency and oscillators’ phases in the network, leading to structure/frequency mapping when plasticity is present. Further we show that while connection plasticity can produce stable synchrony (so called splay states) in Type 1 networks, the structure/frequency reorganization observed in Type 2 networks is not present.

Synaptic delay induced macroscopic dynamics of the large-scale network of Izhikevich neurons

Abstract:We consider a large network of Izhikevich neurons. Each neuron has a quadratic integrate-and-fire type model with a recovery variable modelling spike frequency adaptation (SFA). We introduce a biologically motivated synaptic current expression and a delay in the synaptic transmission. Following the Ott-Antonsen theory, we reduce the network model to a mean-field system of delayed differential equations. Numerical bifurcation analysis allows us to locate higher-codimension bifurcations and to identify the regions in the parameter space where the network exhibits changes in the macroscopic dynamics, including transitions between states where the individual neurons exhibit asynchronous tonic firing and different types of synchronous bursting. We investigate the impact of the heterogeneity of the quenched input current, the SFA mechanism and the synaptic delay on macroscopic dynamics. In the limit that the heterogeneity goes to zero, our perturbation and bifurcation analysis shows that the behaviour of the mean-field model remains consistent, although this limit breaks an assumption of the model reduction. For a single population of neurons with SFA, the synaptic delay has little effect on the generation of COs for weak coupling, but favours their emergence beyond that, and even induces new macroscopic dynamics. In particular, Torus bifurcations may occur, and these are a crucial mechanism for the emergence of population bursting with two nested frequencies. We discuss how these solutions may relate to cross-frequency coupling which is potentially relevant for understanding healthy and pathological brain functions.

Frustrated hierarchical synchronization and emergent complexity in the human connectome network

Abstract:The spontaneous emergence of coherent behavior through synchronization plays a key role in neural function, and its anomalies often lie at the basis of pathologies. Here we employ a parsimonious (mesoscopic) approach to study analytically and computationally the synchronization (Kuramoto) dynamics on the actual human-brain connectome network. We elucidate the existence of a so-far-uncovered intermediate phase, placed between the standard synchronous and asynchronous phases, i.e. between order and disorder. This novel phase stems from the hierarchical modular organization of the connectome. Where one would expect a hierarchical synchronization process, we show that the interplay between structural bottlenecks and quenched intrinsic frequency heterogeneities at many different scales, gives rise to frustrated synchronization, metastability, and chimera-like states, resulting in a very rich and complex phenomenology. We uncover the origin of the dynamic freezing behind these features by using spectral graph theory and discuss how the emerging complex synchronization patterns relate to the need for the brain to access --in a robust though flexible way-- a large variety of functional attractors and dynamical repertoires without ad hoc fine-tuning to a critical point.

Impact of delays and rewiring on the dynamics of small-world neuronal networks with two types of coupling

Abstract:We study synchronization transitions and pattern formation on small-world networks consisting of Morris–Lecar excitable neurons in dependence on the information transmission delay and the rewiring probability. In addition, networks formed via gap junctional connections and coupling via chemical synapses are considered separately. For gap-junctionally coupled networks we show that short delays can induce zigzag fronts of excitations, whereas long delays can further detriment synchronization due to a dynamic clustering anti-phase synchronization transition. For the synaptically coupled networks, on the other hand, we find that the clustering anti-phase synchronization can appear as a direct consequence of the prolongation of information transmission delay, without being accompanied by zigzag excitatory fronts. Irrespective of the coupling type, however, we show that an appropriate small-world topology can always restore synchronized activity if only the information transmission delays are short or moderate at most. Long information transmission delays always evoke anti-phase synchronization and clustering, in which case the fine-tuning of the network topology fails to restore the synchronization of neuronal activity.

Abstract:Over long timescales, neuronal dynamics can be robust to quite large perturbations, such as changes in white matter connectivity and grey matter structure through processes including learning, aging, development and certain disease processes. One possible explanation is that robust dynamics are facilitated by homeostatic mechanisms that can dynamically rebalance brain networks. In this study, we simulate a cortical brain network using the Wilson-Cowan neural mass model with conduction delays and noise, and use inhibitory synaptic plasticity (ISP) to dynamically achieve a spatially local balance between excitation and inhibition. Using MEG data from 55 subjects we find that ISP enables us to simultaneously achieve high correlation with multiple measures of functional connectivity, including amplitude envelope correlation and phase locking. Further, we find that ISP successfully achieves local E/I balance, and can consistently predict the functional connectivity computed from real MEG data, for a much wider range of model parameters than is possible with a model without ISP.

Abstract:Synchronization transitions are investigated in small-world neuronal networks that are locally modeled by the Rulkov map with additive spatiotemporal noise. In particular, we investigate the impact of different information transmission delays and rewiring probability. We show that short delays induce zigzag fronts of excitations, whereas intermediate delays can further detriment synchrony in the network due to a dynamic clustering anti-phase synchronization transition. Detailed investigations reveal, however, that for longer delay lengths the synchrony of excitations in the network can again be enhanced due to the emergence of in-phase synchronization. In addition, we show that an appropriate small-world topology can restore synchronized behavior provided information transmission delays are either short or long. On the other hand, within the intermediate delay region, which is characterized by anti-phase synchronization and clustering, differences in the network topology do not notably affect the synchrony of neuronal activity. Abstract:In neural circuits, synaptic strengths influence neuronal activity by shaping network dynamics, and neuronal activity influences synaptic strengths through activity-dependent plasticity. Motivated by this fact, we study a recurrent-network model in which neuronal units and synaptic couplings are interacting dynamic variables, with couplings subject to Hebbian modification with decay around quenched random strengths. Rather than assigning a specific role to the plasticity, we use dynamical mean-field theory and other techniques to systematically characterize the neuronal-synaptic dynamics, revealing a rich phase diagram. Adding Hebbian plasticity slows activity in already chaotic networks and can induce chaos in otherwise quiescent networks. Anti-Hebbian plasticity quickens activity and produces an oscillatory component. Analysis of the Jacobian shows that Hebbian and anti-Hebbian plasticity push locally unstable modes toward the real and imaginary axes, respectively, explaining these behaviors. Both random-matrix and Lyapunov analysis show that strong Hebbian plasticity segregates network timescales into two bands, with a slow, synapse-dominated band driving the dynamics, suggesting a flipped view of the network as synapses connected by neurons. For increasing strength, Hebbian plasticity initially raises the complexity of the dynamics, measured by the maximum Lyapunov exponent and attractor dimension, but then decreases these metrics, likely due to the proliferation of stable fixed points. We compute the marginally stable spectra of such fixed points as well as their number, showing exponential growth with network size. Finally, in chaotic states with strong Hebbian plasticity, a stable fixed point of neuronal dynamics is destabilized by synaptic dynamics, allowing any neuronal state to be stored as a stable fixed point by halting the plasticity. Network Rewiring and Plasticity Promotes Synchronization of Suprachiasmatic Nucleus Neurons

Abstract:In mammals, circadian rhythms throughout the body are orchestrated by the master clock in the hypothalamic suprachiasmatic nucleus (SCN), where SCN neurons are coupled with neurotransmitters to generate a uniform circadian rhythm. How the SCN circadian rhythm is so robust and flexible is, however, unclear. In this paper, we propose a temporal SCN network model and investigate the effects of dynamical rewiring and flexible coupling due to synaptic plasticity on the synchronization of the neural network in SCN. In networks consisting of simple Poincaré oscillators and complex circadian clocks, we found that dynamical rewiring and coupling plasticity enhance the synchronization in inhomogeneous networks. We verified the effect of enhanced synchronization in different architectures of random, scale-free, and small-world networks. A simple mean-field analysis for synchronization in plastic networks is proposed. Intuitively, the synchronization is greatly enhanced because both the random rewiring and coupling plasticity in the heterogeneous network have effectively increased the coupling strength in the whole network. Our results suggest that a proper network model for the master SCN circadian rhythm needs to take into account the effects of dynamical changes in topology and plasticity in neuron interactions that could help the brain to generate a robust circadian rhythm for the whole body.
The role of gap junctions and clustered connectivity in emergent synchronisation patterns of inhibitory neuronal networks

Abstract:Inhibitory interneurons, ubiquitous in the central nervous system, form networks connected through both chemical synapses and gap junctions. These networks are essential for regulating the activity of principal neurons, especially by inducing temporally patterned dynamic states. We aim to understand the dynamic mechanisms for synchronisation in networks of electrically and chemically coupled interneurons. We use the exact mean-field reduction to derive a neural mass model for both homogeneous and clustered networks. We first analyse a single population of neurons to understand how the two couplings interact with one another. We demonstrate that the network transitions from an asynchronous to a synchronous regime either by increasing the strength of the gap junction connectivity or the strength of the background input current. Conversely, the strength of inhibitory synapses affects the population firing rate, suggesting that electrical and chemical coupling strengths act as complementary mechanisms by which networks can tune synchronous oscillatory behavior. In line with previous work, we confirm that the depolarizing spikelet is crucial for the emergence of synchrony. Furthermore, find that the fast frequency component of the spikelet ensures robustness to heterogeneity. Next, inspired by the existence of multiple interconnected interneuron subtypes in the cerebellum, we analyse networks consisting of two clusters of cell types defined by differing chemical versus electrical coupling strengths. We show that breaking the electrical and chemical coupling symmetry between these clusters induces bistability, so that a transient external input can switch the network between synchronous and asynchronous firing. Together, our results shows the variety of cell-intrinsic and network properties that contribute to synchronisation of interneuronal networks with multiple types of coupling.

Abstract:We investigate the synchronization transition of the Shinomoto-Kuramoto model on networks of the fruit-fly and two large human connectomes. This model contains a force term, thus is capable of describing critical behavior in the presence of external excitation. By numerical solution we determine the crackling noise durations with and without thermal noise and show extended non-universal scaling tails characterized by the exponent 2 < τ t < 2.8, in contrast with the Hopf transition of the Kuramoto model, without the force τ t = 3.1(1). Comparing the phase and frequency order parameters we find different synchronization transition points and fluctuation peaks as in case of the Kuramoto model, related to a crossover at Widom lines. Using the local order parameter values we also determine the Hurst (phase) and β (frequency) exponents and compare them with recent experimental results obtained by fMRI. We show that these exponents, characterizing the auto-correlations are smaller in the excited system than in the resting state and exhibit module dependence.

The existence of semi-stable states within a cortical column is dependent on the type of interaction between the neuronal populations, i.e., the form of the synaptic kernels. Current-to-current coupling has been shown, in contrast to potential-to-current coupling, to create semi-stable states within a cortical column. The interaction between semi-stable states of the cortical columns is studied where we derive the dynamics for the collected activity. For small excitations the dynamics follow the Kuramoto model; however, in contrast to previous work we derive coupled equations between phase and amplitude dynamics with the possibility of defining connectivity as a stationary and dynamic variable. The turbulent flow of phase dynamics which occurs in networks of Kuramoto oscillators would indicate turbulent changes in dynamic connectivity for coupled cortical columns which is something that has been recorded in epileptic seizures. We used the results we derived to estimate a seizure propagation model which allowed for inversions using the Laplace assumption (Dynamic Causal Modelling). The seizure propagation model was trialed on simulated data, and future work will investigate the estimation of the connectivity matrix from empirical data. This model can be used to predict changes in seizure evolution after virtual changes in the connectivity network, something that could be of clinical use when applied to epilepsy surgical cases.
Synchronization transitions in adaptive simplicial complexes with cooperative and competitive dynamics

Abstract:Adaptive network is a powerful presentation to describe different real-world phenomena. However, current models often neglect higher-order interactions (beyond pairwise interactions) and diverse adaptation types (cooperative and competitive) commonly observed in systems like the human brain and social networks. This work addresses this gap by incorporating these factors into a model that explores their impact on collective properties like synchronization. Through simplified network representations, we investigate how the simultaneous presence of cooperative and competitive adaptations influences phase transitions. Our findings reveal a transition from first-order to second-order synchronization as the strength of higher-order interactions increases under competitive adaptation. We also demonstrate the possibility of synchronization even without pairwise interactions, provided there is strong enough higher-order coupling. When only competitive adaptations are present, the system exhibits second-order-like phase transitions and clustering. Conversely, with a combination of cooperative and competitive adaptations, the system undergoes a first-order-like phase transition, characterized by a sharp transition to the synchronized state without reverting to an incoherent state during backward transitions. The specific nature of these second-order-like transitions varies depending on the coupling strengths and mean degrees. With our model, we can control not only when the system synchronizes but also the way the system goes to synchronization.
