Abstract:Lignin is the largest source of bio-based aromatic compounds in nature, and its valorization is essential to the sustainability of lignocellulosic biorefining. Characterizing lignin-derived compounds remains challenging due to the heterogeneity of this biopolymer. Tandem mass spectrometry is a promising tool for lignin structural analytics, as fragmentation patterns of model compounds can be extrapolated to identify characteristic moieties in complex samples. This work extended previous resonance excitation-type collision-induced dissociation (CID) methods that identified lignin oligomers containing β-O-4, β-5, and β-β bonds, to also identify characteristics of 5-5, β-1, and 4-O-5 dimers, enabled by quadrupole time-of-flight (QTOF) CID with energy-resolved mass spectrometry (ERMS). Overall, QTOF-ERMS offers in-depth structural information and could ultimately contribute to tools for high-throughput lignin dimer identification.

Abstract:Hydrogenolysis has emerged as one of the most effective means of converting polymeric lignin into monoaromatic fragments of value. Reported yields may be higher than for other methods and can exceed the theoretical yields estimated from measures of the content of lignin's most readily cleaved alkyl-aryl ether bonds in beta-ether units. The high yields suggest that other units in lignin are being cleaved. Diaryl ether units are important units in lignin, and their cleavage has been examined previously using simple model compounds, such as diphenyl ether. Herein, the hydrogenolysis of model compounds that closely resemble the native lignin 4-O-5 diaryl ether units was analyzed. The results provided unexpected insights into the reactivity and partial cleavage of these compounds. The models and lignin polymer produced not only monomers, but also unusual 1,3,5-meta-substituted aromatics that appear to be diagnostic for the presence and the cleavage of the 4-O-5 diaryl ether unit in lignin.

Abstract:The effective cleavage of C-O bonds in linkages of lignin was one of the significant strategies promoting lignin valorization. Herein, the strategy of C-O bonds cleavage of lignin using metal triflate as the catalyst was developed. The carboxylic acid or alcohol could be used as the nucleophile to stabilize the reactive intermediates formed during the depolymerization of lignin, and the corresponding ester/ether compounds could be obtained. This catalytic system was suitable for the C-O bond cleavage in α-O-4 and β-O-4 linkages with excellent efficiency. Additionally, reaction conditions were optimized. The reaction mixture was detected by 1 H NMR, and no other byproducts were found. As for treated lignin samples, the cleavage of C-O bonds in linkages was determined by 2D HSQC NMR, the increased content of the phenol hydroxyl group was proved by FT-IR, and the reduced molecular weight was investigated by GPC. Furthermore, multiple phenolic compounds were detected by GC-MS in the reaction mixtures.

Abstract:The efficient conversion of lignin contributes to reducing human reliance on fossil energy. As a complicated biopolymer, studies on the mechanism of lignin depolymerization is limited by inadequate structural identification of high molecular weight (MW) products like heavy phenolics. Up to now, no individual method can generate both MW and structural information in operando conditions. As a promising approach, tandem mass spectrometry (MS/MS) techniques can provide structural information via the dissociation of target ions. In this study, MS/MS technique was performed both in offline and in‐situ mode during lignin depolymerization. The fundamental guidelines based on MS/MS dissociation principles for typical inter‐unit linkages like β‐O‐4, 5‐5, β‐β, β‐5, and β‐1 were well established. Based on that, major phenolic dimers are successfully identified, including chemical formula and types of inter‐unit linkages. More significantly, real‐time monitoring of structural evolution was achieved by applying in‐situ MS/MS analysis during lignin depolymerization. The results show the different evolution pathways of isomers with same chemical formula, confirming that structural changes during lignin depolymerization is common and obvious. Overall, this study develops an advanced strategy for the full‐view analysis of lignin depolymerization, achieving the static analysis of composition and structure, both monitoring the dynamic evolution of structures.

Abstract:In the search for a replacement for fossil fuel and the valuable chemicals currently obtained from crude oil, lignocellulosic biomass has become a promising candidate as an alternative biorenewable source for crude oil. Hence, many research efforts focus on the extraction, degradation, and catalytic transformation of lignin, hemicellulose, and cellulose. Unfortunately, these processes result in the production of very complex mixtures. Further, while methods have been developed for the analysis of mixtures of oligosaccharides, this is not true for the complex mixtures generated upon degradation of lignin. For example, high-performance liquid chromatography/multiple stage tandem mass spectrometry (HPLC/MS(n)), a tool proven to be invaluable in the analysis of complex mixtures derived from many other biopolymers, such as proteins and DNA, has not been implemented for lignin degradation products. In this study, we have developed an HPLC separation method for lignin degradation products that is amenable to negative-ion-mode electrospray ionization (ESI doped with NaOH), the best method identified thus far for ionization of lignin-related model compounds without fragmentation. The separated and ionized compounds are then analyzed by MS(3) experiments to obtain detailed structural information while simultaneously performing high-resolution measurements to determine their elemental compositions in the two parts of a commercial linear quadrupole ion trap/Fourier-transform ion cyclotron resonance mass spectrometer. A lignin degradation product mixture was analyzed using this method, and molecular structures were proposed for some components. This methodology significantly improves the ability to analyze complex product mixtures that result from degraded lignin.

Abstract:Driven by the interest in metabolomic studies and the progress of imaging techniques, small molecule analysis is booming, while it remains challenging to be realized by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Herein, lignin, the second most abundant biomass in nature, was applied as a dual-ion-mode MALDI matrix for the first time to analyze small molecules. The low ionization efficiency and strong optical absorption properties make lignin a potential MALDI matrix in small molecule analysis. A total of 30 different small molecules were identified qualitatively and six kinds of representative molecules were detected quantitatively with a good linear response (R2 > 0.995). To verify the accuracy of our quantitative method in MALDI, myricitrin, a major bioactive component in Chinese bayberry, was analyzed in different cultivars and tissues. The myricitrin content in real samples detected by MALDI was highly consistent (R2 > 0.999) with that detected by high-performance liquid chromatography, thus indicating the applicability of the lignin matrix. Further characterization by ultraviolet and nuclear magnetic resonance spectroscopy was carried out to explain the possible mechanism of lignin as a matrix and provide more theories for a rational matrix design.

Abstract:There is need for well-defined lignin macromolecules for research related to their use in biomaterial and biochemical applications. Lignin biorefining efforts are therefore under investigation to meet these needs. The detailed knowledge of the molecular structure of the native lignin and of the biorefinery lignins is essential for understanding the extraction mechanisms as well as chemical properties of the molecules. The objective of this work was to study the reactivity of lignin during a cyclic organosolv extraction process adopting physical protection strategies. As references, synthetic lignins obtained by mimicking the chemistry of lignin polymerization were used. State-of-the-art nuclear magnetic resonance (NMR) analysis, a powerful tool for the elucidation of lignin inter-unit linkages and functionalities, is complemented with matrix-assisted laser desorption/ionization-time-of-flight-mass spectrometry (MALDI-TOF MS), to gain insights into linkage sequences and structural populations. The study unraveled interesting fundamental aspects on lignin polymerization processes, such as identifications of molecular populations with high degrees of structural homogeneity and the emergence of branching points in lignin structure. Furthermore, a previously proposed intramolecular condensation reaction is substantiated and new insights into the selectivity of this reaction are introduced and supported by density functional theory (DFT) calculations, where the important role of intramolecular π-π stacking is emphasized. The combined NMR and MALDI-TOF MS analytical approach, together with computational modeling, is important for deeper fundamental lignin studies and will be further exploited.

Abstract:Deep insight of reaction mechanism in lignin model compounds is helpful to achieve the directed depolymerization of lignin or biomass to chemicals or fuels. In this study, the density functional theory (DFT) calculation was employed to investigate the cleavage mechanism of the C–O bonds in lignin dimers. Additionally, the intrinsic chemical reactivity of molecular in term of the Fukui function was applied to predict the most probable sites which react with hydrogen free radicals (H·). It was found that the O atoms in lignin dimers are the most reaction site involving H· because of the large f (0). By this method, the most rational path from a series of reaction paths was screen out. Apart from the Fukui function, the average local ionization energy (ALIE) was analyzed to prove the reliability of Fukui function. The kinetic analysis of the reaction path was performed to further understand the impact of temperature on the reaction rate constant (KTST). It is observed that benzyl phenyl ether (BPE) with higher KTST could be easily cleaved because of the relatively low energy barrier.

Abstract:RATIONALE Two structurally similar bioactive regioisomers, 3-O-tetradecanoylingenol and 20-O-tetradecanoylingenol, from Euphorbia wallichii presented quite different fragmentation behaviors. Revealing the related fragmentation pathways may improve the efficiency of characterization and identification of such type of compounds. METHODS The two regioisomers were subjected to collision-induced dissociation (CID) on Finnigan LCQDECA and LTQ Orbitrap XL instruments. Based on the CID results, the possible fragmentation pathways were proposed and investigated with density functional theory (DFT) calculations. RESULTS Elimination of C14H28O2 (tetradecanoic acid) for 3-O-tetradecanoylingenol and the sequential losses of C4H8 (butylene) for 20-O-tetradecanoylingenol were observed in ESI-MS/MS, witnessed also by HR-ESI-MS/MS. The fragmentation pathways were proposed and verified by calculating the activation energy of their transition states by DFT calculations. CONCLUSIONS Based on the observations, fragmentation pathways for the two regioisomers were proposed and verified by calculating the energy of the reactants, products and the corresponding transition states using DFT. This report should have value in rapid identification of the derivatives of ingenol and other regioisomers in natural products. Copyright (C) 2012 John Wiley & Sons, Ltd.

Abstract:Two-dimensional gas chromatography (GC × GC) and two-dimensional liquid chromatography (LC × LC) are nowadays widely used in academia and industry due to their high separation power. However, as far as we know, the complementarity of these two techniques has not yet been thoroughly studied based on the analysis of the same sample. Therefore, this was undertaken here by analysing the liquid fraction obtained after depolymerising a natural waste - lignin - with GC × GC and off-line comprehensive LC × SFC (SFC: supercritical fluid chromatography). Using complementary techniques is also important for lignin valorisation, as thorough structural characterisation of the depolymerised product can aid with developing and improving valorisation processes. For the tentative identification, NIST library was used for GC × GC-MS results and MS-DIAL together with SIRIUS for LC × SFC-MS/MS data. This allowed to study which compounds are detectable with the different 2D methods but also to discuss the limitations of the data analysis processes. The previous knowledge that LC techniques are more suitable than GC × GC for the analysis of larger oligomers and other low volatility compounds was confirmed; however, it was seen that GC × GC enabled the analysis of smaller compounds, such as aliphatic alcohols and saturated compounds. Overall, the study demonstrates the complementarity of the two techniques but also draws attention to the different detectable compound groups and classifications that the two techniques can provide.
High-Resolution Tandem Mass Spectrometry-Based Analysis of Model Lignin-Iron Complexes: Novel Pipeline and Complex Structures

Abstract:Understanding the chemical nature of soil organic carbon (SOC) with great potential to bind iron (Fe) minerals is critical for predicting the stability of SOC. Organic ligands of Fe are among the top candidates for SOCs able to strongly sorb on Fe minerals, but most of them are still molecularly uncharacterized. To shed insights into the chemical nature of organic ligands in soil and their fate, this study developed a protocol for identifying organic ligands using ultrahigh-performance liquid chromatography-high-resolution tandem mass spectrometry (UHPLC-HRMS/MS) and metabolomic tools. The protocol was used for investigating the Fe complexes formed by model compounds of lignin-derived organic ligands, namely, caffeic acid (CA), p-coumaric acid (CMA), vanillin (VNL), and cinnamic acid (CNA). Isotopologue analysis of 54/56Fe was used to screen out the potential UHPLC-HRMS (m/z) features for complexes formed between organic ligands and Fe, with multiple features captured for CA, CMA, VNL, and CNA when 35/37Cl isotopologue analysis was used as supplementary evidence for the complexes with Cl. MS/MS spectra, fragment analysis, and structure prediction with SIRIUS were used to annotate the structures of mono/bidentate mono/biligand complexes. The analysis determined the structures of monodentate and bidentate complexes of FeLxCly (L: organic ligand, x = 1-4, y = 0-3) formed by model compounds. The protocol developed in this study can be used to identify unknown organic ligands occurring in complex environmental samples and shed light on the molecular-level processes governing the stability of the SOC.

Abstract:Acidolysis in conjunction with stabilization of reactive intermediates has emerged as one of the most powerful methods of lignin depolymerization that leads to high aromatic monomer yields. In particular, stabilization of reactive aldehydes using ethylene glycol results in the selective formation of the corresponding cyclic acetals (1,3-dioxolane derivatives) from model compounds, lignin, and even from softwood lignocellulose. Given the high practical utility of this method for future biorefineries, a deeper understanding of the method is desired. Here, we aim to elucidate key mechanistic questions utilizing a combination of experimental and multilevel computational approaches. The multiscale computational protocol used, based on ReaxFF molecular dynamics, represents a realistic scenario, where a typical experimental setup can be reproduced confidently given the explicit molecules of the solute, catalyst, and reagent. The nudged elastic band (NEB) approach allowed us to characterize the key intermolecular interactions involved in the reaction paths leading to crucial intermediates and products. The high level of detail obtained clearly revealed for the first time the unique role of sulfuric acid as a proton donor and acceptor in lignin β-O-4 acidolysis as well as the reaction pathways for ethylene glycol stabilization, and the difference in reactivity between compounds with different methoxy substituents.Multiscale computational modeling reveal the role of sulfuric acid and ethylene glycol in the selective breakdown of lignin.This article has not yet been cited by other publications.

A Mechanistic Study of Fragmentation of Deprotonated N,2‐diphenyl‐acetamides in Electrospray Ionization Tandem Mass Spectrometry

Abstract:RATIONALE Exploring the fragmentation mechanism of amide ions in mass spectrometry has attracted great interest because of the desire to analyze the amino acid sequences of peptides and proteins. However, the collision‐induced dissociation (CID) mechanism of deprotonated small amides has been rarely studied in electrospray ionization mass spectrometry (ESI‐MS). The fragmentation of deprotonated N, 2‐diphenylacetamides exhibited some characteristic fragment ions, which are not derived from the conventional cleavage route. Therefore, clarification of their fragmentation mechanism is very important and useful for structural analysis of related amides and peptides. METHODS All CID experiments were carried out using an electrospray ionization ion trap mass spectrometer in negative ion mode. In addition, the accurate masses of fragments were measured on an ESI quadrupole time‐of‐flight (Q‐TOF) mass spectrometer in negative ion mode. Deuterium‐labeled 2‐phenyl‐ N ‐(4‐trifluoromethylphenyl)acetamide was synthesized and its ESI fragmentation spectrum had been obtained. Theoretical calculations were carried out by the density functional theory (DFT) method at the B3LYP level of theory with the 6‐31G++(d,p) basis set. RESULTS Deprotonated N, 2‐diphenylacetamides mainly generate four kinds of ions in CID: benzyl anion, aniline anion, phenyl‐ethenone anion and isocyanato‐benzene anion bearing respective substituent groups. The benzyl anion and the aniline anion can be generated by direct decomposition. The phenyl‐ethenone anion and the isocyanato‐benzene anion were proposed to be yielded from proton transfer within an ion‐neutral complex, and the intensities of two competitive product ions are well correlated with the substituent constants. The mechanism was also supported by theoretical calculations. CONCLUSIONS The characteristic fragment ions of deprotonated N ,2‐diphenylacetamides were proposed to be produced via an ion‐neutral complex mechanism, which was proved by deuterium‐labeling experiments, theoretical calculations and substituent effects. Abstract:Lignin is abundant in natural world, and it can be converted into value-added chemicals by thermo-chemical method. Since the insufficient understanding of the lignin pyrolysis mechanism limits practical application of lignin pyrolysis, it is quite important to deeply understand the mechanism of lignin pyrolysis from the molecular level. In this work, 1-(4-hydroxy-3-methoxyphenyl)-2-(2-methoxyphenoxy)-1-ethanol was chosen as a β-O-4 type dimer model compound of lignin. Combining the density functional theory (DFT) method with Py-GC/MS to analyze the pyrolysis behavior of lignin dimer model compound, 9 reasonable reaction paths were studied by DFT. The results showed that 2-methoxy-4-vinylphenol (P3) and 2-methoxyphenol (P4) are the main products of lignin dimer model compound pyrolysis. The kinetic and thermodynamics analysis indicates the homolytic cleavage of Cβ–O is the initial reaction step for forming P3 and P4. In the subsequent reactions, P3 is mainly formed by hydrogenation and then dehydration. P4 is mainly formed by hydrogenation. Increasing temperature can promote the spontaneous reaction of the main paths. The exploration for the pyrolysis mechanism of lignin dimer is helpful to directionally regulate lignin pyrolysis products in future studies.

During the thermochemical depolymerization which involves acid-catalyzed, alkali-catalyzed, oxidative, reductive, pyrolytic, and other reactions, the lignin structure will undergo a series of bond cleavage, condensation, and functional group changes, while the mechanism is still unclear. To improve the efficiency, the analysis of the evolution of intermediates during depolymerization is very important, among which soft ionization mass spectrometry plays a vital role. This review aims to summarize the research progress of process analysis of lignin depolymerization in both gas-phase, typically thermal and catalytic pyrolysis, and liquid-phase via online mass spectrometry. The challenges and our insights into the future development of the lignin valorization as well as soft ionization mass spectrometry methods are also discussed.

Abstract:Our ability to study and valorize the lignin fraction of biomass is hampered by the fundamental and still unmet challenge of precisely quantifying native lignin's structural features. Here, we developed a rapid elevated-temperature 1H–13C Heteronuclear Single-Quantum Coherence Zero (HSQC0) NMR method that enables this precise quantification of native lignin structural characteristics even with whole plant cell wall (WPCW) NMR spectroscopy, overcoming fast spin relaxation in the gel phase. We also formulated a Gaussian fitting algorithm to perform automatic and reliable spectral integration. By combining HSQC0 measurements with yield measurements following depolymerization, we can confirm the combinatorial nature of radical coupling reactions during biosynthesis leading to a random sequential organization of linkages within a largely linear lignin chain. Such analyses illustrate how this analytical method can greatly facilitate the study of native lignin structure, which can then be used for fundamental studies or to understand lignin depolymerization methods like reductive catalytic fractionation or aldehyde-assisted fractionation.

Abstract:Lignin is an abundant polymer found in wood and grasses, but due to its heterogeneity and complex macromolecular structure it has been less utilized than cellulose. While the building blocks are known, the way they are linked is less understood. Here, Molecular Dynamics simulations were used to systematically characterize seven linkages found in native lignin. Their influence on lignin and water structure, and their interactions were analyzed. The study is limited to guaiacyl (G-G) dimers connected by the following common softwood linkages; 5-5′, 4-O-5′, α -O-4′, β -1′, β -5′, β -O-4′ and β - β ′. The simulations show that the linkage has a significant effect on conformational preference and lignin-water interaction. Especially, the behavior of the β -O-4′ shows unique properties, both in terms of conformational freedom and interaction with water. Within the β -O-4′ dimer, π – π stacking between the aromatic rings is possible. The molecule has two distinct common conformations, one compressed and one extended. These preferences also lead to a different effect of β -O-4′ dimer on the surrounding water, where water is found close to the linkage itself but expelled from the aromatic rings to a larger extent than the other linkages. These findings are important for lignin solubility as well as its depolymerization mechanisms.
