Abstract:Lignin, a major component of lignocellulosic biomass, is crucial to plant growth and development but is a major impediment to efficient biomass utilization in various processes. Valorizing lignin is increasingly realized as being essential. However, rapid condensation of lignin during acidic extraction leads to the formation of recalcitrant condensed units that, along with similar units and structural heterogeneity in native lignin, drastically limits product yield and selectivity. Catechyl lignin (C-lignin), which is essentially a benzodioxane homopolymer without condensed units, might represent an ideal lignin for valorization, as it circumvents these issues. We discovered that C-lignin is highly acid-resistant. Hydrogenolysis of C-lignin resulted in the cleavage of all benzodioxane structures to produce catechyl-type monomers in near-quantitative yield with a selectivity of 90% to a single monomer.

Abstract:Industrial biorefineries remain limited due to inefficient valorization of their lignin streams. Rarely can a lignin depolymerization method be combined with a current polysaccharide-centric biorefinery process. Hydrogenolysis is among the more promising methods for depolymerizing lignin on an industrial scale. We performed reaction kinetics and mechanistic studies on lignin model compounds to understand lignin hydrogenolysis pathways, demonstrating that lignin end-units and internal units react significantly differently. Understanding the reaction mechanism and its sensitivity to variables helped us establish a continuous lignin upgrading process from various fractionated lignins. Near-theoretical yields of lignin platform monomers with > 80% overall product selectivity were obtained in a continuous hydrogenolysis process using a Pd/C catalyst.

Abstract:The utilization of biomass in the production of renewable bioenergy and biomaterials has been a popular topic since the past decades as they are rich in carbohydrates. Most biomasses, such as wood, monocotyledons, and agriculture residues, need to be pretreated before the conversion of carbohydrates in order to break down the recalcitrant cell wall structure and increase the fiber accessibility. To date, a variety of pretreatment methods have been developed that vary from physical to chemical and biological methods. Pretreatment processes affect the cell wall physical structure as well as the chemical structure of the cell wall constituents. Comparing to the studies of the cellulose and hemicelluloses structural changes during pretreatment, such studies on lignin are relatively limited. On the other hand, in order to utilize the part of lignin from biorefinery processes, the understanding of the lignin structural changes during the refining process becomes important. In this study, typical pretreatment methods such as hydrothermal pretreatment, alkaline pretreatment, biodegradation, and oxidative pretreatment are introduced and their corresponding impacts on the lignin structures are reviewed.

Abstract:Lignocellulose is mainly composed of hydrophobic lignin and hydrophilic polysaccharide polymers, contributing to an indispensable carbon resource for green biorefineries 1,2 . When chemically treated, lignin is compromised owing to detrimental intra- and intermolecular crosslinking that hampers downstream process 3,4 . The current valorization paradigms aim to avoid the formation of new C–C bonds, referred to as condensation, by blocking or stabilizing the vulnerable moieties of lignin 5,6,7 . Although there have been efforts to enhance biomass utilization through the incorporation of phenolic additives 8,9 , exploiting lignin's proclivity towards condensation remains unproven for valorizing both lignin and carbohydrates to high-value products. Here we leverage the proclivity by directing the C–C bond formation in a catalytic arylation pathway using lignin-derived phenols with high nucleophilicity. The selectively condensed lignin, isolated in near-quantitative yields while preserving its prominent cleavable β-ether units, can be unlocked in a tandem catalytic process involving aryl migration and transfer hydrogenation. Lignin in wood is thereby converted to benign bisphenols (34–48 wt%) that represent performance-advantaged replacements for their fossil-based counterparts. Delignified pulp from cellulose and xylose from xylan are co-produced for textile fibres and renewable chemicals. This condensation-driven strategy represents a key advancement complementary to other promising monophenol-oriented approaches targeting valuable platform chemicals and materials, thereby contributing to holistic biomass valorization.

Abstract:The complex and heterogeneous nature of the lignin macromolecule has presented a lasting barrier to its utilization. To achieve high lignin yield, the technical lignin extraction process usually severely modifies and condenses the native structure of lignin, which is a critical drawback for its utilization in conversion processes. In addition, there is no method capable of separating lignin from plant biomass with controlled structural properties. Here, we developed an N-heterocycle-based deep eutectic solvent formed between lactic acid and pyrazole (La–Py DES) with a binary hydrogen bonding functionality resulting in a high affinity toward lignin. Up to 93.7% of lignin was extracted from wheat straw biomass at varying conditions from 90 °C to 145 °C. Through careful selection of treatment conditions as well as lactic acid to pyrazole ratios, lignin with controlled levels of ether linkage content, hydroxyl group content, and average molecular weight can be generated. Under mild extraction conditions (90 °C to 120 °C), light-colored native-like lignin can be produced with up to 80% yield, whereas ether linkage–free lignin with low polydispersity can be obtained at 145 °C. Overall, this study offers a new strategy for native lignin extraction and generating lignin with controlled structural properties.

Abstract:Lignin, an aromatic polymer, is a crucial component of lignocellulosic biomass and is a significant source of renewable aromatics found in nature. Lignin constitutes up to 30 wt % of lignocellulosic biomass and is generated in excess of 100 million tons per year from global paper and bio-ethanol industries. Unfortunately, most technical lignin produced in this manner is burned as a heat source, without any value-added valorization. The conversion of technical lignin into valuable compounds can significantly increase the profitability of biorefineries. However, the productivity of this process is constrained by the severe lignin condensation that occurs during extraction procedures, resulting in the formation of robust C–C bonds, such as methylene linkages, which lead to monomer yields of only 8–15%. Herein, we present an approach to overcome this limitation by producing well-defined aromatic monomers from condensed lignin fractions. The approach employs commercial zeolite-catalyzed cleavage of methylene linkages with water as both the reacting medium and the reactant. Up to 7–10% of additional aromatic monomers can be obtained from various types of technical lignin, thus increasing the total monomer yields by 59–102%. The findings of this study present a promising avenue for the valorization of technical lignin and offer potential for the development of circular economy.

Abstract:This article explores the important, and yet often overlooked, solid-state structures of selected bioaromatic compounds commonly found in lignin hydrogenolysis oil, a renewable bio-oil that holds great promise to substitute fossil-based aromatic molecules in a wide range of chemical and material industrial applications. At first, single-crystal X-ray diffraction (SCXRD) was applied to the lignin model compounds, dihydroconiferyl alcohol, propyl guaiacol, and eugenol dimers, in order to elucidate the fundamental molecular interactions present in such small lignin-derived polyols. Then, considering the potential use of these lignin-derived molecules as building blocks for polymer applications, structural analysis was also performed for two chemically modified model compounds, i.e., the methylene-bridging propyl-guaiacol dimer and propyl guaiacol and eugenol glycidyl ethers, which can be used as precursors in phenolic and epoxy resins, respectively, thus providing additional information on how the molecular packing is altered following chemical modifications. In addition to the expected H-bonding interactions, other interactions such as π–π stacking and C–H∙∙∙π were observed. This resulted in unexpected trends in the tendencies towards the crystallization of lignin compounds. This was further explored with the aid of DSC analysis and CLP intermolecular energy calculations, where the relationship between the major interactions observed in all the SCXRD solid-state structures and their physico-chemical properties were evaluated alongside other non-crystallizable lignin model compounds. Beyond lignin model compounds, our findings could also provide important insights into the solid-state structure and the molecular organization of more complex lignin fragments, paving the way to the more efficient design of lignin-based materials with improved properties for industrial applications or improving downstream processing of lignin oils in biorefining processes, such as in enhancing the separation and isolation of specific bioaromatic compounds).

Abstract:Background Lignin, nature’s dominant aromatic polymer, is found in most terrestrial plants in the approximate range of 15 to 40% dry weight and provides structural integrity. Traditionally, most large-scale industrial processes that use plant polysaccharides have burned lignin to generate the power needed to productively transform biomass. The advent of biorefineries that convert cellulosic biomass into liquid transportation fuels will generate substantially more lignin than necessary to power the operation, and therefore efforts are underway to transform it to value-added products. Advances Bioengineering to modify lignin structure and/or incorporate atypical components has shown promise toward facilitating recovery and chemical transformation of lignin under biorefinery conditions. The flexibility in lignin monomer composition has proven useful for enhancing extraction efficiency. Both the mining of genetic variants in native populations of bioenergy crops and direct genetic manipulation of biosynthesis pathways have produced lignin feedstocks with unique properties for coproduct development. Advances in analytical chemistry and computational modeling detail the structure of the modified lignin and direct bioengineering strategies for targeted properties. Refinement of biomass pretreatment technologies has further facilitated lignin recovery and enables catalytic modifications for desired chemical and physical properties. Outlook Potential high-value products from isolated lignin include low-cost carbon fiber, engineering plastics and thermoplastic elastomers, polymeric foams and membranes, and a variety of fuels and chemicals all currently sourced from petroleum. These lignin coproducts must be low cost and perform as well as petroleum-derived counterparts. Each product stream has its own distinct challenges. Development of renewable lignin-based polymers requires improved processing technologies coupled to tailored bioenergy crops incorporating lignin with the desired chemical and physical properties. For fuels and chemicals, multiple strategies have emerged for lignin depolymerization and upgrading, including thermochemical treatments and homogeneous and heterogeneous catalysis. The multifunctional nature of lignin has historically yielded multiple product streams, which require extensive separation and purification procedures, but engineering plant feedstocks for greater structural homogeneity and tailored functionality reduces this challenge.

Abstract:Practical, high-yield lignin depolymerization methods could greatly increase biorefinery productivity and profitability. However, development of these methods is limited by the presence of interunit carbon-carbon bonds within native lignin, and further by formation of such linkages during lignin extraction. We report that adding formaldehyde during biomass pretreatment produces a soluble lignin fraction that can be converted to guaiacyl and syringyl monomers at near theoretical yields during subsequent hydrogenolysis (47 mole % of Klason lignin for beech and 78 mole % for a high-syringyl transgenic poplar). These yields were three to seven times those obtained without formaldehyde, which prevented lignin condensation by forming 1,3-dioxane structures with lignin side-chain hydroxyl groups. By depolymerizing cellulose, hemicelluloses, and lignin separately, monomer yields were between 76 and 90 mole % for these three major biomass fractions.

Abstract:Lignin is a cross linked aromatic polymer and it is abundant in nature. It is regarded as a potential substitute of fossil resource and industrial raw materials due to its highly aromatic character via lignin valorization and commercialization. Lignin depolymerization is an effective solution to achieve lignin valorization by yielding value-added fine chemicals (such as monophenols and biofuels). Due to their stable structure, complex spatial configurations and electronic effect, lignin depolymerization often suffered from serious condensation and complexity of products. Commonly, this results in low yield and selectivity that limit large scale of lignin depolymerization. A challenge exists, however, on how to develop high selective catalytic strategies/reactors, which would enable efficient and selective catalysis by controlling the bond breaking mode and inhibiting the condensation of active intermedia. In this review, we begin by introducing the potential of lignin as an emerging star for lignin commercialization. And then catalytic depolymerization strategy (e.g., catalytic pyrolysis, catalytic hydrolysis, catalytic oxidation, catalytic reduction and two-step redox) and also their selective bond breaking mechanism are reviewed. Particularly, we report latest vision about lignin highly efficient valorization targeting at reaching market, by controlling of reaction condensation and using continuous-flow reactor depolymerization. Most importantly, the limitations and potential solutions for lignin depolymerization are proposed to present opportunities for future industrialization applications.
Structural studies of an enzymatically produced Norway spruce lignin-carbohydrate complex

Abstract:Lignocellulose can become the best opportunity for the society to reduce its dependency on the harmful petroleum based products as well as to produce clean energy. In each part of the production cycle, biomass based products have a better environmental profiles than their petroleum based counterparts. Woody biomass has a vast availability, but it suffers from recalcitrance that is mostly caused by lignin that is functioning as a matrix, surrounding and binding the carbohydrates that are currently the most valuable of the wood components.Lignin-carbohydrate (LC) bonds are believed to be a key element in this recalcitrance and research has shown that these types of bonds are common in wood. These bonds are important in an economical point of view as well, as e.g. residual lignin structures in pulp (lignins bonded to the cellulose and hemicelluloses) require expensive bleaching sequences for their removal.The LC-structures can also be exploited technically as we now have demonstrated. We developed a method that utilizes phenolic end groups that are bonded to different hemicelluloses for cross-linking. The enzyme laccase was used for the cross-linking to create a cost-efficient processing scheme to both isolate and increase the molecular weight of the hemicelluloses. Membrane filtration was used as the key separation technique, which enables the establishment of industrial scale production. The final product had improved mechanical and thermal properties and could be used e.g. as barrier film component in renewable packaging. Nanocomposite formation with nanofibrillated cellulose was also studied. This improved the film properties further. The complexes are also possible to use as model compounds for lignin-carbohydrate complexes in wood.This technique can also be seen to mimick the lignification and lignin-carbohydrate network formation phenomena in plants enabling the formation of entire networks of wood components. Our results suggests that the side chains of hemicellulose might play an important role in network formation and that hemicellulose molecules can carry more than one lignin phenolic end group to fulfill this capability.

Abstract:The conversion of lignin to value-added platform biochemicals and functional biomaterials paves the way towards a carbon-negative bio-based economy. However, the heterogeneous molecular structure, molecular weight, and functional groups of lignin significantly influence its processibility during valorization, modification, and polymerization. This study provides a detailed structural and property characterization of lignin components fractionated from the waste stream of a lignocellulosic biorefinery process via a simple gradient acid method. The lignin structures from different fractionated pH were identified using advanced non-destructive techniques such as gel permeation chromatography (GPC), quantitative 13C, 31P, and 2-D nuclear magnetic resonance (NMR) in combination with chemo/thermal property measurements. GPC results indicated a gradually decreased molecular weight of lignin with the fractionated pH. The lignin precipitated at pH 9.0 had the highest β-O-4 ether bonds and those at pH 5.0 and 7.0 owned the highest yield and relatively more aliphatic and phenolic hydroxyl groups. The lignin fractions after pH 3.0 were mainly monomers and dimers. It is concluded that the gradient acid method is an easy-to-operate method to fractionalize lignin, which provides a potential path to enhance its processability towards value-added and carbon-negative bioproducts.

Abstract:Breaking down lignin into smaller units is the key to generate high value-added products. Nevertheless, dissolving this complex plant polyphenol in an environment-friendly way is often a challenge. Levulinic acid, which is formed during the hydrothermal processing of lignocellulosic biomass, has been shown to efficiently dissolve lignin. Herein, levulinic acid was evaluated as a medium for the reductive electrochemical depolymerization of the lignin macromolecule. Copper was chosen as the electrocatalyst due to the economic feasibility and low activity towards the hydrogen evolution reaction. After depolymerization, high-resolution mass spectrometry and nuclear magnetic resonance spectroscopy revealed lignin-derived monomers and dimers. A predominance of aryl ether and phenolic groups was observed. Depolymerized lignin was further evaluated as an anti-corrosion coating, revealing enhancements on the electrochemical stability of the metal. Via a simple depolymerization process of biomass waste in a biomass-based solvent, a straightforward approach to produce high value-added compounds or tailored biobased materials was demonstrated.

Abstract:The chemical complexity of lignin remains a major challenge for lignin valorization into commodity and fine chemicals. A knowledge of the lignin features that favor its valorization and which plants produce such lignins can be used in plant selection or to engineer them to produce lignins that are more ideally suited for conversion. Sixteen biomass samples were compositionally surveyed by NMR and analytical degradative methods, and the yields of phenolic monomers following hydrogenolytic depolymerization were assessed to elucidate the key determinants controlling the depolymerization. Hardwoods, including those incorporating monolignol p-hydroxybenzoates into their syringyl/guaiacyl copolymeric lignins, produced high monomer yields by hydrogenolysis, whereas grasses incorporating monolignol p-coumarates and ferulates gave lower yields, on a lignin basis. Softwoods, with their more condensed guaiacyl lignins, gave the lowest yields. Lignins with a high syringyl unit content released elevated monomer levels, with a high-syringyl polar transgenic being particularly striking. Herein, we distinguish phenolic monomers resulting from the core lignin vs those from pendent phenolate esters associated with the biomass cell wall, acylating either polysaccharides or lignins. The basis for these observations is rationalized as a means to select or engineer biomass for optimal conversion to worthy phenolic monomers.

Abstract:Lignin valorization via thermochemical approaches has the potential to produce renewable fuels and value-added chemicals, which are of great significance to the sustainable development of human beings. During the thermochemical depolymerization which involves acid-catalyzed, alkali-catalyzed, oxidative, reductive, pyrolytic, and other reactions, the lignin structure will undergo a series of bond cleavage, condensation, and functional group changes, while the mechanism is still unclear. To improve the efficiency, the analysis of the evolution of intermediates during depolymerization is very important, among which soft ionization mass spectrometry plays a vital role. This review aims to summarize the research progress of process analysis of lignin depolymerization in both gas-phase, typically thermal and catalytic pyrolysis, and liquid-phase via online mass spectrometry. The challenges and our insights into the future development of the lignin valorization as well as soft ionization mass spectrometry methods are also discussed.

Abstract:Lignin has long been recognized as a potential feedstock for aromatic molecules; however, most lignin depolymerization methods create a complex mixture of products. The present study describes an alkaline aerobic oxidation method that converts lignin extracted from poplar into a collection of oxygenated aromatics, including valuable commercial compounds such as vanillin and p-hydroxybenzoic acid. Centrifugal partition chromatography (CPC) is shown to be an effective method to isolate the individual compounds from the complex product mixture. The liquid-liquid extraction method proceeds in two stages. The crude depolymerization mixture is first subjected to ascending-mode extraction with the Arizona solvent system L (pentane/ethyl acetate/methanol/water 2:3:2:3), enabling isolation of vanillin, syringic acid, and oligomers. The remaining components, syringaldehyde, vanillic acid, and p-hydroxybenzoic acid (pHBA), were resolved by using ascending-mode extraction with solvent mixture comprising dichloromethane/methanol/water (10:6:4) separation. These results showcase CPC as an effective technology that could provide scalable access to valuable chemicals from lignin and other biomass-derived feedstocks.

Extraction Of Lignin From Wood Biomass Using A New Ionic Liquid At Open Atmosphere

Abstract:Using wood biomass as opposed to petroleum/fossil feedstock to synthesize chemicals and monomers offers the value proposition of a reduced carbon foot print. A critical step in biomass utilization is to obtain the fermentable monomeric pentose and hexose sugar subunits from various lig-nocellulosic polymers. However, lignin is a three-dimensional amorphous polymer based on methoxylated phenylpropanoid units, and is crosslinks to different polysaccharides with covalent bond. The presence of lignin is a barrier to enzymatic hydrosis of cellulose by cellulases, as well as inhibiting fermentation to form products such as ethanol. Usually, there are three approaches to fractionate biomass into useful chemicals. The first one is extract lignin at high temperature and pressure. The second approach involves more severe conditions such as the temperatures and pressures used in gasification, or use the mineral acids at elevated temperatures to liberate sugars for fermentation. The third way is dissolution of biomass using ionic liquid to extract lignin, and then re-precipitation the cellulosic residue. In this paper, we investigate the lignin extraction uses a purpose designed ionic liquid (IL) 1 -butyl-3-methylimidazolium acesulfamate [BMIM]Ace at room temperature and open atmosphere. It was synthesized from the sugar acesulfamate potassium (Ace K), and with good property of greater thermal stability, increased viscosities, and inability to dissolve crystalline cellulose. In addition, it is of low cost and nontoxic, and is expected to interact with lignin owing to its aromatic character of anion. Organic solvents were firstly used in the extraction process, which reduce the viscosity of IL and increase the lignin yield. The lignin can be obtained by using acetone as the reconstitution solvent (IL/acetone 1:3, v/v), and approximately 11.5% and 96.5% of lignin and the reconstitution cellulose-rich material have been achieved in one dissolution/reconstitution cycle by fresh IL, respectively. It was found that the presence of some organic solvents mix with IL can increase the extraction efficiency. The regenerated ionic liquid showed good retention of structure and properties. The regenerated lignin fractions exhibited large molecular weights (M-w, 3700-7810 g mol(-1)). The physicochemical properties of the extracted lignin fractions were characterized by Fourier transform infrared (FT-IR), and high-performance anion exchange chromatography (HPAEC). The reconstitution cellulose-rich material can be serving as a valuable source for enzymatic saccharification for next step. These results indicate a potential approach in further optimization of the choice of low-cost Its combined with organic solvents for the efficient utilization of the wood biomass.

Abstract:Converting biomass to produce renewable chemicals is one of the significant ways to realize the development of green chemistry and sustainable chemical industry. As the main component of lignocellulosic biomass, lignin is the most abundant natural polyphenol. Its unique phenolic properties and high carbon content make it potentially exploited. Biomass lignin depolymerization can be divided into thermochemical, chemical catalysis, electrocatalysis, and biological depolymerization methods. In this review, the catalytic reaction systems involved in different methods of lignin depolymerization were extensively described from various aspects, and the degradation products and reaction mechanisms were discussed and analyzed. The effects of novel green-deep eutectic solvents on biomass lignin depolymerization were reviewed in particular. Each method of biomass lignin depolymerization has its own advantages and disadvantages. The different sources of biomass lignin should be selected objectively according to its compositional structure, so as to achieve efficient biomass lignin depolymerization. Abstract:Electrochemical oxidation provides a green process for lignin depolymerization. In this work, depolymerization mechanisms of alkali and organosolv lignin on a PbO2 anode have been explored in details through 2D‐HSQC NMR, in‐situ IR spectroelectrochemistry and depolymerized product analysis. It not only clarifies lignin structure changes, but also reveals the selectivity in bond cleavage and product distribution in the oxidation process. To better understand the process and mechanism of lignin structural changes and relative product formation during PbO2 anode electrolysis, a Ti/Sb−SnO2/PbO2 composite electrode has been prepared and utilized for the alkaline electrolytic oxidation of both alkali lignin (AL) and bagasse organosolv lignin (OL). By combining electrochemical measurements, in‐situ FTIR, 2D‐HSQC NMR, and product distribution analysis, we not only investigate the impact of influence factors on the process of lignin depolymerization, but also reveal the depolymerization mechanism from the whole process of lignin structural changes in details. Under certain electrolysis conditions, the highest vanillin yield, 10.1 mg g−1, has been obtained through the electrolysis of alkali lignin, whereas the highest syringaldehyde yield of 4.0 mg g−1 has been achieved during the electrolysis of organic solvent lignin. In particular, AL mainly performs depolymerization through the selective cleavage C−C bonds to produce a series of G unit monomer products of vanillin, acetovanillone, and vanillic acid. By contrast, the depolymerization of OL shows significant selectivity in H units. Particularly, the production of the major H unit monomer product of 4‐vinylphenol has been confirmed by IR spectroelectrochemical studies from the oxidation and decarboxylation of p‐coumaric acid unit.
