Abstract:Mold growth on body donations remains an underreported yet serious issue in anatomical teaching. Bacterial and fungal growth pose health risks to lecturers and students, alongside with ethical and aesthetic concerns. However, limited information exists on the presence of bacteria and fungi on body donations and their underlying causes. To investigate the potential impact of airborne germs on body donation contamination, we conducted indoor air measurements before, during, and after our anatomical dissection course, with outdoor measurements serving as a control. Tissue samples from the dissected body donations were collected to assess the germ load, with qualitative and quantitative microbiological analyses. Air samples from the dissection hall contained no fungi, but various fungal species were identified in the adjacent stairways and outdoors which implies that fungal occurrence in the dissection hall air was independent of lecturers' and students' presence. Moreover, our results indicate that adequate ventilation filters can effectively reduce indoor fungal germs during courses, while the bacterial load in room air appears to increase, likely due to the presence of lecturers and students. Additionally, the tissue samples revealed no bacterial or fungal germs which implies that our ethanol-formalin-based embalming solution demonstrates an effective long-term antimicrobial preservation of corpses.

[Formaldehyde exposure levels and exposure control measures during an anatomy dissecting course]

Abstract:The evaporation of formaldehyde from cadavers can produce high exposures among students and instructors. A possible causal role for formaldehyde has been considered likely for tumor of the nasopharynx and the nasal cavities in human beings. Due to this reason, Japan Ministry of Education, Culture, Sports, Science and Technology (MEXT) has set a guideline, which includes--decrease in gaseous formaldehyde in gross anatomy dissection laboratories and a guide to medical students about the toxicity of formaldehyde and protective method to avoid damages to skin, mucous, membrane, etc, in 2002. To understand what effective plans should be regarding the awareness of students about this notification, this study measured the gaseous formaldehyde concentrations in the anatomy dissection room and also analyzed the formaldehyde-related symptoms, and frequency of using protective measures. The study was conducted over a period of 3 months during the anatomy dissection exercise. We found that immediately after removing the cadavers' plastic covering, formaldehyde concentrations in the dissection room increased sharply. The concentration reached a peak point of 0.62 ppm after 10 minutes of starting of the class. This was much above the recommended level of 0.5 ppm set by Japan Society for Occupational Health. After 30 minutes of achieving the peak the formaldehyde level started decreasing gradually to a level of 0.11 ppm. Formaldehyde-related symptoms were observed in 59% of students. They had experienced symptoms of irritation of eyes, nose, throat, airways, skin, and headache during the course. Ocular discomfort was found significantly higher in the contact lenses users compared to the spectacle users or the normal eye sight group. Although, the guidelines about toxicity of formaldehyde and its protective measures to prevent damages to skin, mucous membrane etc. were informed to every student, only 52% of the students used both the mask containing activated carbon and the rubber gloves in every practical class without fail. Environmental Health Criteria 89 of International Program of Chemical Safety states, "It must be regarded that formaldehyde fluid is not absorbed directly into tissues through the skin". So the students may be allowed in some cases to touch the cadaver, treated by formaldehyde content fixative, by bare hands to understand the feel of certain organs and tissues. These results support that the rules of health supervision including necessity to use of protective measures, monitoring of indoor air formaldehyde etc. should be adhered by students and instructors in anatomy dissection room during the practical class.

Abstract:Hands-on courses utilizing preserved human tissues for educational training offer an important pathway to acquire basic anatomical knowledge. Owing to the reevaluation of formaldehyde limits by the European Commission, a joint approach was chosen by the German-speaking anatomies in Europe (Germany, Austria, Switzerland) to find commonalities among embalming protocols and infrastructure. A survey comprising 537 items was circulated to all anatomies in German-speaking Europe. Clusters were established for "ethanol"-, formaldehyde-based ("FA"), and "other" embalming procedures, depending on the chemicals considered the most relevant for each protocol. The logistical framework, volumes of chemicals, and infrastructure were found to be highly diverse between the groups and protocols. Formaldehyde quantities deployed per annum were three-fold higher in the "FA" (223 L/a) compared to the "ethanol" (71.0 L/a) group, but not for "other" (97.8 L/a), though the volumes injected per body were similar. "FA" was strongly related to table-borne air ventilation and total fixative volumes ≤1000 L. "Ethanol" was strongly related to total fixative volumes >1000 L, ceiling- and floor-borne air ventilation, and explosion-proof facilities. Air ventilation was found to be installed symmetrically in the mortuary and dissection facilities. Certain predictors exist for the interplay between the embalming used in a given infrastructure and technical measures. The here-established cluster analysis may serve as decision supportive tool when considering altering embalming protocols or establishing joint protocols between institutions, following a best practice approach to cater toward best-suited tissue characteristics for educational purposes, while simultaneously addressing future demands on exposure limits.
Formaldehyde exposure in gross anatomy laboratory of Suranaree University of Technology: a comparison of area and personal sampling

Abstract:Cadavers are usually preserved by embalming solution which is composed of formaldehyde (FA), phenol, and glycerol. Therefore, medical students and instructors have a higher risk of exposure to FA inhalation from cadavers during dissection. Therefore, the objective of this study was to evaluate the FA exposure in indoor air and breathing zone of medical students and instructors during dissection classes in order to investigate the relationship between them. The indoor air and personal air samples in breathing zone were collected three times during anatomy dissection classes (in January, August, and October of 2014) with sorbent tubes, which were analyzed by high-performance liquid chromatography (HPLC). The air cleaner machines were determined by weight measurement. Pulmonary function tests and irritation effects were also investigated. The mean of FA concentrations ranged from 0.117 to 0.415 ppm in the indoor air and from 0.126 to 1.176 ppm in the breathing zone of students and instructors. All the personal exposure data obtained exceeded the threshold limit of NIOSH and WHO agencies. The air cleaner machines were not significant difference. The pulmonary function of instructors showed a decrease during attention of classes and statistically significant decreasing in the instructors more than those of the students. Clinical symptoms that were observed in nose and eyes were irritations with general fatigue. We suggested that the modified exhaust ventilation and a locally ventilated dissection work table were considered for reducing FA levels in the gross anatomy dissection room.

Abstract:The application of most chemical fixatives, such as formalin, in the anatomic pathology laboratory requires safety training and hazardous chemical monitoring due to the toxicity and health risks associated with their use. Consequently, the use of formalin has been banned in most applications in Europe; the primary exception is its use in the histology laboratory in lieu of a suitable and safer alternative. Glyoxal based solutions, several of which are available commercially, are the most promising alternative fixatives, because they are based on a mechanism of fixation similar to that of formalin. Unlike formalin, however, glyoxal based solutions do not dissociate from water and therefore do not require ventilation measures such as a fume hood. A primary barrier to the adoption of commercially available glyoxal based solutions is their low pH, which can produce undesirable morphological and antigenic tissue alterations; however, a recently available neutral pH glyoxal product (glyoxal acid free) (GAF) has been developed to mitigate the challenges of low pH. We compared the morphology and histochemistry among tissues fixed in 10% neutral buffered formalin, a commercially available acidic glyoxal product (Prefer), and GAF. Tissues fixed in formalin and Prefer exhibited similar morphology and staining properties; tissues fixed with 2% GAF exhibited deleterious effects.

Abstract:The professional use of formaldehyde is a major occupational health concern in medical laboratory operations. Quantification of various risks associated with chronic exposure to formaldehyde may help in understanding the related hazards. This study designed to assess the health risks associated with inhalation exposure to formaldehyde involving biological, cancer, and non-cancer risks in medical laboratories. This study was performed in the hospital laboratories at Semnan Medical Sciences University. Risk assessment was performed in pathology, bacteriology, hematology, biochemistry, and serology laboratories with 30 employees, using formaldehyde in their daily routine operations. We determined area and personal exposures to airborne contaminant, applied standard air sampling, and analytical method recommended by National Institute of Occupational Safety and Health (NIOSH). We addressed formaldehyde-specific hazard by estimating peak blood level, life cancer risk, and the hazard quotient of non-cancer risks, adapted from Environmental Protection Agency (EPA) assessment method. The airborne formaldehyde concentrations in the laboratory personal samples ranged 0.0156–0.5940 ppm (mean = 0.195 ppm, SD = 0.048) and area exposure ranged 0.0285–1.0810 ppm (mean = 0.462 ppm, SD = 0.087). Based on workplace exposure, peak blood levels of formaldehyde were estimated at minimum 0.0026 mg/l to maximum 0.152 mg/l (mean = 0.015 mg/l, SD = 0.016). The mean cancer risk levels in terms of area and personal exposures were estimated respectively at 3.93 E − 8 μg/m 3 and 1.84 E − 4 μg/m 3 , and for the non-cancer risk levels of the same exposures measured respectively at 0.03 μg/m 3 and 0.07 μg/m 3 . Formaldehyde levels were significantly higher among laboratory employees, especially bacteriology workers. Exposure and risk could be minimized by strengthening control measures including the use of management controls, engineering controls, and respiratory protection equipment to reduce exposure levels of all workers to less than the allowed exposure limits and improve indoor air quality in the workplace.

Abstract:Formaldehyde, which is recognized as a harmful indoor air pollutant for human health, is emitted mainly from urea formaldehyde resin in wood products. Chamber tests are used to evaluate formaldehyde emission rates from these products. However, there is no available formaldehyde standard reference emission source to assess the performance of chamber testing systems. In this work, a LIFE (liquid-inner tube diffusion-film-emission) formaldehyde reference is described. The formaldehyde source consists of a polytetra-fluoroethene (PTFE) tube that holds a formaldehyde-water solution with a concentration of 16 g formaldehyde per 100 mL water, with a thin polydimethylsiloxane (PDMS) film cover. Formaldehyde emission parameters for the PDMS film (diffusion coefficient and partition coefficient) were determined experimentally, thereby enabling the prediction of the formaldehyde emissions from the source for use as a reference value in a chamber. Chamber tests were conducted in a 51 L stainless steel ventilated chamber. The impacts of temperature and relative humidity on the emissions were investigated Results show the LIFE's chamber test results match those predicted by a mass transfer model. As a result, this formaldehyde source may be used to generate a reference concentration in product emission testing chambers, thereby providing a powerful tool to evaluate the performance of the chamber testing systems.

FA is released from several sources, both temporary (e.g., combustion processes) and permanent (e.g., building products). This work proposes an innovative fully-automated application of headspace solid-phase microextraction (SPME) with on-fiber derivatization for the analysis of airborne FA emitted from liquid solutions or solid manufacts, in static mode, via gas chromatography-mass spectrometry. The method was tested in a wide range of airborne FA concentrations, using SPME and SPME Arrow fibers: the inter-day LOD and LOQ for SPME and SPME Arrow were evaluated, resulting in 0.072 and 0.215 ppm and 0.014 and 0.042 ppm, respectively. Moreover, other conventional detectors, such as Electron Capture Detector (ECD), Thermoionic Specific Detector (TSD), Photoionization Detector (PID), and Flame Ionization Detector (FID), were tested to set an analytical method to meet different requirements. The sensitivity and linearity of PID, FID and MS were comparable, while TSD and ECD were not suitable for the developed method, due to issues of response or linearity. MS results to be the most suitable and perfoming detector, however PID and FID result to be cheaper valid alternatives.

Abstract:This research focuses on the evaluation of diverse approaches for removing formaldehyde from indoor environments, which is a significant concern for indoor air quality. The study systematically examines physical, chemical, and biological methods to ascertain their effectiveness in formaldehyde mitigation. Physical methods, including air circulation and adsorption, particularly with activated carbon and molecular sieves, are assessed for their efficiency in various concentration scenarios. Chemical methods, such as photocatalytic oxidation using titanium dioxide and plasma technology, are analyzed for their ability to decompose formaldehyde into non-toxic substances. Additionally, biological methods involving plant purification and microbial transformation are explored for their eco-friendly and sustainable removal capabilities. Abstract:Background: Exposure to formaldehyde from a preserved cadaver can cause several health problems in students, lecturers, and laboratory technicians working in medical anatomy laboratories. These conditions include respiratory tract irritation and cancer due to acute and chronic exposure, respectively. Lowering the frequency of contact with formaldehyde from preserved organs helps to ensure the safety of students, lecturers, and laboratory technicians. Therefore, this study aims to determine the effectiveness of low concentration formalin as a preservative solution for goat kidneys. The organ was selected due to its anatomical structure similarity to humans. Method : Goat kidneys with 50-60 ml of volume were collected, washed, immersed in preservative solution, and stored for two months. The samples were then divided into four groups, which were preserved with different formalin concentrations, namely 30%, 20%, 10%, and 5%. The preservation parameters, namely organ structure and integrity, color, springiness, odor, and mold growth were qualitatively described, while the kidney volume was quantitatively measured. Results : After the samples were preserved for two months, they were observed, and all groups showed similar characteristics. The kidneys preserved with 5% or 10% formalin showed equal volume before and after two months of preservation. Conclusion : Low concentration formalin has the same effectiveness in preserving goat kidneys as the high concentration. It also has the potential to be applied in human anatomy learning.

Abstract:Evidence has been presented indicating that a sufficient intake of a carboxylic cation exchanger in the potassium cycle increases the stool output of sodium in absolute as well as in relative terms (1-4). Two other findings accompanying the use of this form of the resin are noteworthy: a) a significant proportion of the potassium taken with or as part of the resin can be retained in the body and b) ingestion of this particular resin does not produce acidosis. Its efficiency in removing sodium is probably not as great, however, as that of either the hydrogen or the ammonium forms of the resin, each of which interferes with potassium absorption and induces a metabolic acidosis. The first half of this paper deals with findings observed during the simultaneous administration of the two types of resin in equal proportions, i.e., potassium cycle mixed with either the hydrogen or the ammonium cycle, as well as in mixtures in which the acidifying resin predominated. The second half of this report describes results obtained during the use of resin mixtures with ACTHor cortisone or both. The expanding recourse to adrenal cortical effects in the therapy of a wide variety of disease states (5-7) has further emphasized that prolonged therapy is frequently not possible because of two particularly undesirable concomitants, i.e., sodium retention and potassium depletion. It seems probable that these side effects by necessitating interruption or intermittency of treatment deprive patients of benefits which might be otherwise obtainable. The combined resin and steroid or hormone studies herein presented describe attempts at prevention or deferment of such deviations in cation metabolism.

Abstract:The present study examines for the first time the emission patterns and olfactory signatures of 9 complete human corpses of different stages of decomposition. Air sampling was performed inside the body bags with solid sorbents and analysed by coupled gas chromatography-mass spectrometry after thermal desorption (TD-GC-MS). Furthermore, odour-related substances were detected by gas chromatography-olfactometry (GC-O). Sulfurous compounds (mainly dimethyl di- and trisulfide) were identified as most important to the odour perception. Around 350 individual organic substances were detected by TD-GC-MS, notably sulfurous and nitrogenous substances as well as branched alkanes, aldehydes, ketones, alcohols, carboxylic acids, carboxylic acid esters and ethers. A range of terpenes was detected for the first time in a characteristic emission pattern over all decomposition stages. Concentrations of the substances varied greatly, and no correlation between the emission patterns, the stage of decomposition and the cause of death could be found. While previous studies often analysed pig cadavers or only parts of human tissue, the present study shows the importance of analysing complete human corpses over a range of decomposition stages. Moreover, it is shown that using body bags as a kind of "emission test chamber" is a very promising approach, also because it is a realistic application considering the usual transport and store of a body before autopsy.

Formaldehyde quantification using gas chromatography-mass spectrometry reveals high background environmental formaldehyde levels

Abstract:Formaldehyde (HCHO) is a human toxin that is both a pollutant and endogenous metabolite. HCHO concentrations in human biological samples are reported in the micromolar range; however, accurate quantification is compromised by a paucity of sensitive analysis methods. To address this issue, we previously reported a novel SPME-GC-MS-based HCHO detection method using cysteamine as an HCHO scavenger. This method showed cysteamine to be a more efficient scavenger than the widely used O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine, and enabled detection of aqueous HCHO in the nanomolar range and quantification in the micromolar range. However, quantification in this range required immersive extraction of the HCHO-derived thiazolidine, while a high background signal was also observed. Following on from these studies, we now report an optimised head-space extraction SPME-GC-MS method using cysteamine, which provides similarly sensitive HCHO quantification to the immersive method but avoids extensive wash steps and is therefore more amenable to screening applications. However, high background HCHO levels were still observed A Complementary GC-MS analyses using a 2-aza-Cope-based HCHO scavenger also revealed high background HCHO levels; therefore, the combined results suggest that HCHO exists in high (i.e. micromolar) concentration in aqueous samples that precludes accurate quantification below the micromolar range. This observation has important implications for ongoing HCHO quantification studies in water, including in biological samples.

Abstract:Formaldehyde is a harmful substance that can cause sick building syndrome and other diseases, such as contact allergy, asthma, leukemia, cancer, and brain/neuron disorders. Formaldehyde is a ubiquitous chemical owing to its use in many common products, including as a preservative in household and personal care products. To prevent overexposure to formaldehyde, a simple method for determining and controlling the formaldehyde content in commercial products is required. In this study, 3-aminoquinoline (3-AQ) was used to derivatize formaldehyde under mild conditions (2 min at 30°C) without the use of catalysts or activators. The derivatized sample solutions were separated using narrow-bore liquid chromatography with an ultraviolet (UV) detector in a run time of only 5 min. All sample extraction and derivatization protocols were performed on the microliter scale to reduce the use of organic solvents. The linear range for the determination was 5–1000 μg/mL, with a detection limit of approximately 1 μg/mL ( 2 ng per 2 μL injection). The proposed microscale method was successfully applied to the analysis of formaldehyde in commercial household products.

Abstract:Formaldehyde is commonly used in detergents and cosmetic products as antibacterial agent and preservative. This substance is unfavorable for human health because it is known to be toxic for humans and causes irritation of eyes and skins. The toxicology studies of this compound indicate risk of detergents and cosmetic formulations with a minimum content of 0.05% free formaldehyde. Therefore, determination of formaldehyde as quality control parameter is very important. In this study, a photoluminescence method was achieved by using 2-methyl acetoacetanilide. Also, the Box-Behnken design was applied for optimization of Hantzsch reaction for formaldehyde derivatization. The investigated factors (variables) were temperature, % v/v ethanol, reaction time, ammonium acetate, and 2-methyl acetoacetanilide concentration. The linear range was obtained from 0.33–20 × 10 −7 M (1–60 μ g·kg −1 ) and the limit of detection (LOD) was 0.12 μ g·kg −1 . The proposed method was applied for the analysis of Iranian brands of liquid detergents and cosmetic products. The formaldehyde content of these products was found to be in the range of 0.03–3.88%. Some brands of these products had higher concentration than the maximum allowed concentration of 0.2%. High recoveries (96.15%–104.82%) for the spiked dishwashing liquid and hair shampoo indicate the proposed method is proper for the assessment of formaldehyde in detergents and cosmetic products. The proposed methodology has some advantages compared with the previous methods such as being rapid, without the necessity of applying separation, low cost, and the fact that the derivatization reaction is carried out at room temperature without any heating system.

Abstract:Formaldehyde is the smallest existing aldehyde, a highly reactive color less gas at room temperature and ubiquitously present in our atmosphere. Because of its reactivity leading to the crosslinking of macromolecules like proteins, it is widely used in industrial applications, but also in cell biology in order to preserve cells and tissues for further analysis. In this work, we show that formaldehyde releasing solutions commonly used for fixation of cells, can diffuse via the gas phase to the neighboring well and influence signaling processes in the therein cultured cells. To analyze this effect, we utilized a stable reporter cell line for YAP signaling or a gene expression-based reporter for activation of the NF-κB pathway. Especially the stable reporter cell line can also be used as sensor for bioavailable formaldehyde. The observed impact of formaldehyde on cellular signaling underscores the need for careful planning of experimental protocols and emphasizes the importance of implementing proper controls when utilizing this reagent in cellular signaling analyses.

Abstract:Formaldehyde (FA) is a highly toxic substance present in many matrices, including freshwater as well as found in natural mechanisms such as rainfall and combustion of organic matter. Consumption of water contaminated with high levels of FA can cause severe short-term or long-term health problems. Due to these health risks, procedures are necessary to determine and quantify FA in aqua sources This paper reports on a study of fluorimetric determination of FA using a nickel(2 + )-diketonate coordination compound immobilized as a solid precursor. The compound was characterized by electronic absorption, Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), thermogravimetry (TG), optical microscopy (OM), and scanner electron microscopy (SEM). The methodology was based on the reaction of the synthesized compound with an ammoniacal buffer generating a selective reagent for formaldehyde: fluoral-P. The product of the reaction generates 3,5-diacetyl-1,4-dihydrolutidine (DDL), which is responsible for the fluorescence of the system. Several parameters such as temperature, duration of heating time, and dilution effect with the best effects were studied to carry out FA determination. Under the optimum experimental conditions, a linear response ranging from 1.0 to 10.0 mg/L FA (R = 0.997 and n = 10), and a detection (3σ criterion) and quantification (10 σ criterion) limit estimated at 0.129 and 0.389 mg/L, respectively were achieved. The FA analysis was able to be conducted in 05 min with a relative standard deviation estimated at 1.10 %.

Abstract:Based on the gas–liquid phase chemiluminescence tester independently developed by our laboratory, a highly sensitive, fast and accurate on-line detection method of formaldehyde gas in ambient atmosphere is established. The chemiluminescence system and the trace formaldehyde gas in the air directly undergo an interface heterogeneous chemiluminescence reaction to obtain a strong chemiluminescence signal. Through the measurement of the chemiluminescence signal intensity, a highly sensitive, real-time and on-line method for the determination of formaldehyde in the air was established. Factors influencing the experimental results such as gallic acid, potassium dichromate, reaction medium concentration, surfactant type and concentration, pump speed, tube length, and interfering gas were discussed based on the single factor and orthogonal analysis results. Finally, the optimal detection conditions were collected, and the detection results were compared with the national standard phenol reagent method. The results show that when the concentration of the standard formaldehyde gas is in the range of 0–0.582 μg L −1 , the linear equation of this method is y = 208 x + 29.667, the linear coefficient is R 2 = 0.997, and the minimum detection concentration of formaldehyde is 2.327 × 10 −3 μg L −1 . Under the same external conditions, the comparison and analysis using the national standard phenol reagent method proved that this method has the advantages of fast detection speed, low detection limit, good sensitivity, and accurate results, which can be used for real-time and online determination of trace formaldehyde in ambient air.
