Abstract:Optimal embryonic development and growth of meat-type chickens (broilers) rely on incubation conditions (oxygen, heat, and humidity), on nutrients and on energy resources within the egg. Throughout incubation and according to the embryo's energy balance, the main energy storage molecules (creatine and glycogen) are continuously utilized and synthesized, mainly in the embryonic liver, breast muscle, and the extraembryonic yolk sac (YS) tissue. During the last phase of incubation, as the embryo nears hatching, dynamic changes in energy metabolism occur. These changes may affect embryonic survival, hatchlings' uniformity, quality and post hatch performance of broilers, hence, being of great importance to poultry production. Here, we followed the dynamics of creatine and glycogen from embryonic day (E) 11 until hatch and up to chick placement at the farm. We showed that creatine is stored mainly in the breast muscle while glycogen is stored mainly in the YS tissue. Analysis of creatine synthesis genes revealed their expression in the liver, kidney, YS tissue and in the breast muscle, suggesting a full synthesis capacity in these tissues. Expression analysis of genes involved in gluconeogenesis, glycogenesis, and glycogenolysis, revealed that glycogen metabolism is most active in the liver. Nevertheless, due to the relatively large size of the breast muscle and YS tissue, their contribution to glycogen metabolism in embryos is valuable. Towards hatch, post E19, creatine levels in all tissues increased while glycogen levels dramatically decreased and reached low levels at hatch and at chick placement. This proves the utmost importance of creatine in energy supply to late-term embryos and hatchlings.

Effects of Posthatch Feed Deprivation on Residual Yolk Absorption, Macronutrients Synthesis, and Organ Development in Broiler Chicks

Abstract:The aim of the research was to evaluate the dynamic changes of early posthatch starvation on residual yolk absorption, synthesis of macronutrients (protein, lipid, and glycogen), and organ development in broiler chicks. A total of 720 1-day-old chicks (Lingnan Yellow) were randomly assigned to 3 treatments: group A (nonfasted), group B (fasting for 24 h after placement), and group C (fasting for 48 h after placement). The trial lasted for 168 h, and water was provided ad libitum all the time. Sampling was performed at 0, 24, 48, 72, 120, and 168 h. Nonfasting (group A) promoted (P < 0.05) the absorption of amino acids, fatty acids, mineral elements, protein, and maternal antibody in the residual yolk of broiler chicks. The concentration of insulin-like growth factor 1 in plasma and the liver was higher (P < 0.05) in group A. Nonfasting enhanced (P < 0.05) the synthesis of protein and glycogen in the breast muscle and liver; the relative weights of the liver, pancreas, and spleen; and body weight, but retarded (P < 0.05) the synthesis of triglyceride in the liver. The results indicated that nonfasting (group A) after placement promoted the absorption of residual yolk and synthesis of protein and glycogen in the breast muscle and liver, whereas early feed deprivation promoted the synthesis of lipid in the liver. Thereby, nonfasting after placement promoted organ development and body growth of broiler chicks.

Abstract:To better understand the digestive capacity in domestic pigeons (Columba livia), this study was conducted to evaluate nutrient transporters and digestive enzymes gene expression in small intestine and yolk sac membrane (YSM) during pre- and posthatch development. We investigated the oligopeptide transporter Pept1, sodium glucose transporter SGLT1, glucose transporter GLUT2, aminopeptidase-N (APN), and sucrase-isomaltase (SI). Intestine was collected at embryo d 12, 14, and 16, day of hatch, and d 1, 3, 5, 8, and 14 posthatch. The YSM was collected at embryo d 12, 14, 16, and day of hatch. The cDNA fragments for Pept1, SGLT1, GLUT2, APN, and SI were isolated and cloned using reverse-transcription PCR. The sequences data showed that these genes were highly identical to the gene of chicken. The mRNA expression of each gene was assayed using real-time PCR. Expression of intestinal nutrient transporters increased linearly (P<0.001) with age whereas that of APN increased quadratically (P<0.001) and SI increased cubically (P=0.001). Levels of PepT1 mRNA were greatest in the duodenum (P=0.001), GLUT2 and SGLT1 mRNA were greatest in the jejunum (P=0.002) and APN were greatest in the ileum (P=0.004). The YSM expressed all the examined genes. The YSM-expressed genes decreased between embryo d 16 and day of hatch, whereas intestine-expressed genes increased. Our findings provide a comprehensive profile of gene expression patterns of nutrient transporter and digestive enzyme in the small intestine and YSM of pigeons and establish a foundation for future research on the nutrients requirements for young pigeons.
Effects of Dietary Supplementation of Gegf on the Growth Performance and Immunity of Broilers

Abstract:EGF has been shown to stimulate the growth of animals. In this study, the content of EGF in chicken embryos (gallus EGF, gEGF) aged from 1 to 20 days of incubation were determined by ELISA kit, and the 5-day-old chicken embryos with the highest content of 5593 pg/g were selected to make gEGF crude extracts. A total of 1500 1-day-old Xianju chickens were randomly divided into five groups with six replicates of 50 chickens each. The control group was fed a basal diet, and other treatment diets were supplemented with 4, 8, 16 and 32 ng/kg gEGF crude extract, respectively. The experiment lasted for 30 days. Chicks were harvested at the end of the experiment, and liver, spleen, thymus, bursa and serum samples were collected. Results showed that average daily gain (ADG) and average daily feed intake (ADFI) of 16 ng/kg group were higher than those in the control group (p < 0.05). The serum uric acid (UA) of the 16 ng/kg group was reduced (p < 0.01), and the serum alkaline phosphatase (AKP) of the 16 ng/kg group increased (p < 0.01). The gEGF extract also increased chick’s antioxidant capacity, decreased malondialdehyde (MDA) and increased catalase (CAT) in the liver and serum of 16 ng/kg groups in compared to the control group (p < 0.01). Furthermore, immunity was improved by the addition of gEGF to broiler diets. The serum immunoglobin A (IgA) content of 8 and 16 ng/kg groups and the serum immunoglobin M (IgM) content of 4 and 8 ng/kg groups were increased (p < 0.05) compared to the control group. The bursa index of each experimental group was higher than the control group (p < 0.01). These findings demonstrate that the crude extract of gEGF prepared in this experiment could improve the growth performance, antioxidant capacity and immunity of broilers.

Abstract:The objectives of the experiment were to investigate the changes of body weight ( BW) , digestive organ weight, serum protein metabolism indices, digestive enzymes activity of pancreas and intestine mucosa, and to determine the developmental pattern of digestive tract in domestic pigeons ( Columbia livia ) .Thirty squabs ( White Carneau) were euthanized by cervical dislocation for samples collection at 2, 4, 6, 8 and 10 days posthatch ( six squabs each day) .The results showed as follows:1) day of age had significant effect on postnatal BW ( yolk involved) , organ weight of proventriculus, gizzard, liver, pancreas, intestine, and con-tents of total protein, albumin, urea acid, usea nitiogen in serum, and activity of pancreatic amylase, lipase, trypsin, specific activity of lipase, and activity and specific activity of intestinal disaccharidase ( maltase and sucrase) as well as phosphatase, respectively ( P<0.05) .In specific, the increased indicators with age includ-ed the relative weight of liver, pancreas, intestine together with the absolute weight of BW, proventriculus, gizzard, pancreas, liver, intestine (P<0.01), and contents of total protein, albumin in serum (P<0.01), and the activities of pancreatic amylase, lipase, trypsin followed with the specific activity of lipase ( P<0.05) , and activity and specific activity of intestinal disaccharidase ( maltase and sucrase ) as well as phosphatase ( P<0.01) .Contents of urea acid, urea nitrogen in serum and specific activity of trypsin declined linearly with day of age ( P<0.05 ) .Whereas the relative weight of proventriculus and gizzard went up, then dropped ( P<0.05).2) Segment had significant effect on activity of maltase and sucrose of squabs (P<0.05).3) Interac-tion between day of age and segment had significant effect on activity of maltase and sucrose and specific activ-ity of sucrose ( P<0.05) .It is concluded that the developmental capacity of protein metabolism, digestive sys-tem, pancreatic and intestinal enzyme activity elevated significantly improved with days of age.Hence, it is a vital stage of 2 to 10 days of age posthatch to promote the digestion capacity for squabs.

Abstract:The effects of in ovo feeding of carbohydrates on hatchability, BW, yolk sac weights (YSW), pectoral muscle weights (PMW), liver and pectoral muscle glycogen concentration, serum glucose level, and hepatic glucose-6-phosphatase activity of domestic pigeons, hatched from eggs laid by a 40-wk-old breeder flock, were investigated. At 14.5 of incubation, fertile eggs were injected with 200 μL of 1.5% maltose (M) + 1.5% sucrose (S), 2.5% M + 2.5% S, 3.5% M + 3.5% S, or 4.5% M + 4.5% S in 0.75% saline, with controls not injected. Results showed that in ovo injection with 1.5% M + 1.5% S or 2.5% M + 2.5% S increased the hatchability compared with the control, whereas injection of 4.5% M + 4.5% S decreased the hatchability. The BW at hatch was quadratic, and BW was maximized by injecting 2.5% M + 2.5% S. The YSW at hatch decreased linearly by the injection with 3.5% M + 3.5% S compared with the control group. In ovo injection of 2.5% M + 2.5% S increased the PMW at hatch. There were no significant differences between any of the treatment groups for liver glycogen reserves. Serum glucose level at hatch was quadratic, and the glucose level was maximized between supplemental 2.5% M + 2.5% S and supplemental 3.5% M + 3.5% S. The pectoral muscle glycogen reserves increased quadratically as supplemental carbohydrates increased, and the response was maximized by injecting 2.5% M + 2.5% S. In conclusion, the present results demonstrate that the injected carbohydrates are available for use and storage. In ovo feeding of carbohydrates, especially at the level of 2.5% M + 2.5% S, on 14.5 d of incubation can improve the hatchability, BW, and PMW by elevating the pectoral muscle glycogen reserves in domestic pigeons at hatch. Results also suggested that in ovo injection of carbohydrates could increase the yolk sac nutrient utilization and hence might enhance the pigeon enteric development.

Abstract:This study aimed to investigate the effects of time access to post-hatch feeding on the growth performance, hormone secretion, intestinal morphology, and intestinal microbiota structure of broilers. A total of 900 broilers were randomly allocated to 3 treatment groups, with 6 replicates of 50 broilers each. The 3 treat-ments were: immediate feeding (Group 2 h), delayed access to feed for 24 h (Group 24 h), and delayed access to feed for 48 h (Group 48 h). The experiment lasted for 50 d. Results revealed that Group 2 h had a higher aver-age daily gain (ADG) and average daily feed intake (ADFI) as well as a lower feed-to-gain ratio (F/G) than Group 48 h during the starter period (P < 0.05). Compared with Group 48 h, broilers in Group 2 h exhib-ited significantly elevated villus height (VH) and villus height to crypt depth ratio (VH: CD) in the duodenum, increased Occludin, and Claudin-1 mRNA expression in the jejunum but decreased crypt depth (CD) in the duo-denum at 50 d (P < 0.05). Meanwhile, broilers in Groups 2 h and 24 h had increased glycogen (Gn) and protein (Pro) levels in breast muscle and TG levels in the liver, as well as a higher concentration of serum T3, T4, and IGF-1 compared with Group 48 h at 21 d (P < 0.05). Besides, intestinal microbiota consisted primarily of Fir-micutes, Bacteroidetes, and Proteobacteria at the phy-lum level at 21 d and 50 d; at the genus level, broilers in Group 2 h displayed significantly reduced abundance of Escherichia at 21 d and Bacteroides at 50 d compared with Group 48 h (P < 0.05). Collectively, these findings signal that early post-hatch feeding measures, especially at 21 d, improve hormone secretion, intestinal morphol-ogy, and the growth performance of broilers by enhanc-ing intestinal health and modulating the intestinal microbiota.

Abstract:The gastrointestinal tract plays an important role in studies of avian ontogenies because of its key function in nutrient intake. The pattern of intestinal development during postnatal growth in domestic pigeons (Columba livia), as an altricial bird, would be different from the pattern observed in precocial poultry. In this study, the intestinal morphology, mucosal enzyme activities, and pancreatic enzyme activities were determined from hatch to 14 d of age in pigeons. Rapid morphological development was observed immediately after hatching. Villus area and crypt depth increased in parallel in the duodenum and jejunum and more slowly in the ileum. Mucosal enzyme activities per gram of duodenum, jejunum, and ileum varied significantly with segment and age. There were also intestinal segment × age interactions, in which enzyme activity increased most dramatically in the jejunum as compared with the duodenum and ileum for sucrase and maltase activity from day of hatch to 8 d, and for alkaline phosphatase activity through 14 d. However, aminopeptidase-N activity increased most dramatically from day of hatch to 3 d in the ileum as compared with the duodenum and jejunum. Changes in carbohydrate-digesting enzymes activity in duodenum, jejunum, ileum, and pancreas are more pronounced than for protein-digesting enzymes activity from 3 d after hatching. Intestinal total enzyme activities exhibited a steady increase, which was highly correlated with BW. Pancreatic enzymes indicated patterns somewhat similar to those for intestinal enzymes. It was concluded that development of the absorptive area and the capacity for hydrolysis in the duodenum, jejunum, and ileum was rapid immediately after hatching, although rates of development were different in the 3 segments. Young pigeons differ from poultry, in whom intestinal and pancreatic digestive enzyme activities per gram of tissue are generally constant or decline during postnatal development. Also, the data seem consistent with the idea that intestinal hydrolysis may be a determining step in digestion and hence growth.
Expression of Messenger Ribonucleic Acids of Luteinizing Hormone and Follicle-Stimulating Hormone Receptors in the Embryonic and Posthatch Gonads of the Chicken

Abstract:The expression of mRNA encoding luteinizing hormone receptor (LHR) and follicle-stimulating hormone receptor (FSHR) was studied in chicken gonads from day 10 of incubation to day 14 of posthatch by Northern hybridization and expression of each mRNA was compared between sexes. LHR mRNA levels in the left ovary increased from day 12 of incubation, reached a peak two days later and remained high thereafter. In contrast, those in the testes remained extremely low until day 18 of incubation but started increasing sharply from day 20 of incubation and reached peak levels at the posthatch day 7. FSHR mRNA levels of the gonads displayed similar changes between both sexes. The changes resembled those of LHR mRNA levels of the testis during the examined period. These results suggest that the earlier expression of LHR mRNA in the ovary is involved in earlier estrogen production and the later expression of LHR mRNA in the testis is involved in androgen production at the late stage of development in the chick. The later expression of FSHR mRNA may be associated with some gonadal functions in common to both sexes.
Effects of First Feed Administration on Small Intestinal Development and Plasma Hormones in Broiler Chicks

Abstract:(1) Background: Under practical conditions, newly hatched chicks were usually withheld feed and water for 48 to 72 h. It was shown that early feeding after hatch promoted gastrointestinal development of broiler chicks. However, the mechanism of early feeding affecting intestinal development in chicks needs further research. The present study was conducted to investigate the effects of first feed administration on intestinal morphology, barrier function, and plasma hormones in broilers during the initial 168 h posthatch. (2) Methods: A total of 720 one-day-old chicks (newborn chick, Lingnan Yellow) were placed 2 h after hatch and randomly assigned to three treatments: Group A (feed immediately after placement), Group B (fasting for 24 h after placement), and Group C (fasting for 48 h after placement). The trial lasted for 168 h and water ad libitum all the time. Sampling was performed at 0, 24, 48, 72, 120, and 168 h. (3) Results: Higher (p < 0.05) absolute weight and relative weight of the small intestine were observed in Group A. Moreover, the villus height, crypt depth, and ratio of the jejunum and ileum were significantly higher (p < 0.05) in Groups A and B than those in Group C. Microvilli of the duodenum were closely packed in Group A but sparse and disorganized in Groups B and C. The expression levels of mRNA and protein of tight junction genes (occludin and claudin-1) were upregulated (p < 0.05) in Group A. The levels of gastrin and insulin in plasma were decreased (p < 0.05) significantly in the Groups B and C. However, chicks in Groups B and C had higher (p < 0.05) plasma glucagon levels at 24 and 48 h after placement. (4) Conclusions: These results suggested that early feeding posthatch had a positive effect on small intestinal growth increasing weight and improving intestinal morphology and barrier function.
Effects of in Ovo Injection of Carbohydrate Solution on Small Intestine Development in Domestic Pigeons (columba Livia)

Abstract:The objective of this study was to test the hypothesis that in ovo injection of carbohydrates into pigeon (Columba livia) amnion may improve the small intestine development. At d 14.5 of incubation, 80 fertile eggs were injected with 200 μL of carbohydrate solution, and 80 control eggs were not injected. The carbohydrate solution (wt/vol) contained 2.5% maltose + 2.5% sucrose, all dissolved in 0.75% saline. Twelve eggs from each treatment were randomly sampled at d 16 of incubation and the day of hatch, embryos or young pigeons were euthanized, and the jejunum samples were collected. Jejunal villus surface area, activity of the brush border enzymes, sucrase, maltase, aminopeptidase-N, and alkaline phosphatase, and mRNA expression of the digestion-absorption related genes oligopeptide transporter 1, sodium glucose transporter 1, glucose transporter 2, aminopeptidase-N, and sucrase-isomaltase were examined. Results showed that in ovo injection of carbohydrate solution caused a villus surface area increase of 38% on d 16 of incubation and 23% on day of hatch relative to controls (P < 0.05). The in ovo injected pigeons exhibited greater (P < 0.05) activities of jejunal sucrase, maltase, and alkaline phosphatase from d 16 of incubation to day of hatch compared with the controls. At day of hatch, aminopeptidase-N activity in embryos injected in ovo was approximately 27% greater (P < 0.05) than control embryos. Enhanced expressions of the jejunal sodium glucose transporter 1, glucose transporter 2, and aminopeptidase-N mRNA were found at d 16 of incubation in embryos that received carbohydrate solution into the amniotic fluid in comparison with the control group (P < 0.05). These results indicate that the in ovo injected pigeon may hatch with more mature enterocytes and greater intestinal digestive and absorptive capacity than the conventional hatchling. Therefore, the in ovo injected pigeons may become more precocial at hatch and easier to hand-rear during the immediate posthatch period.
Ontogeny of hepatic metabolism in two broiler lines divergently selected for the ultimate pH of the Pectoralis major muscle

Abstract:Background: Nutrient availability during early stages of development (embryogenesis and the first week post-hatch) can have long-term effects on physiological functions and bird metabolism. The embryo develops in a closed structure and depends entirely on the nutrients and energy available in the egg. The aim of this study was to describe the ontogeny of pathways governing hepatic metabolism that mediates many physiological functions in the pHu + and pHu- chicken lines, which are divergently selected for the ultimate pH of meat, a proxy for muscle glycogen stores, and which differ in the nutrient content and composition of eggs. Results: We identified eight clusters of genes showing a common pattern of expression between embryonic day 12 (E12) and day 8 (D8) post-hatch. These clusters were not representative of a specific metabolic pathway or function. On E12 and E14, the majority of genes differentially expressed between the pHu + and pHu- lines were overexpressed in the pHu + line. Conversely, the majority of genes differentially expressed from E18 were overexpressed in the pHu- line. During the metabolic shift at E18, there was a decrease in the expression of genes linked to several metabolic functions (e.g. protein synthesis, autophagy and mitochondrial activity). At hatching (D0), there were two distinct groups of pHu + chicks based on hierarchical clustering; these groups also differed in liver weight and serum parameters (e.g. triglyceride content and creatine kinase activity). At D0 and D8, there was a sex effect for several metabolic pathways. Metabolism appeared to be more active and oriented towards protein synthesis (RPS6) and fatty acid β-oxidation (ACAA2, ACOX1) in males than in females. In comparison, the genes overexpressed in females were related to carbohydrate metabolism (SLC2A1, SLC2A12, FoxO1, PHKA2, PHKB, PRKAB2 and GYS2). Conclusions: Our study provides the first detailed description of the evolution of different hepatic metabolic pathways during the early development of embryos and post-hatching chicks. We found a metabolic orientation for the pHu + line towards proteolysis, glycogen degradation, ATP synthesis and autophagy, likely in response to a higher energy requirement compared with pHu- embryos. The metabolic orientations specific to the pHu + and pHu- lines are established very early, probably in relation with their different genetic background and available nutrients.

Abstract:Studies on the yolk and albumen content in bird eggs, and the effects of variations in their relative loads in the phenotype of the birds, have revealed multiple consequences at different levels of biological organization, from biochemical traits to behavior. However, little is known about the effect of albumen variation on energetics performance during development and early ontogeny, despite the fact that variation in energy expenditure may have consequences in terms of fitness for both feral and domestic species. In this work, we evaluated experimentally whether variations in the content of albumen of Gallus gallus eggs could generate differences in metabolic rates during embryonic development. Additionally, we assessed changes in the activity of mitochondrial enzymes (cytochrome c oxidase and citrate synthase) in skeletal muscles and liver. Finally, we evaluated the success of hatching of these embryos and their metabolic rates (MR) post-hatching. The results revealed a significant reduction in MR in the last fifth of embryonic life, and reduced catabolic activities in the skeletal muscle of chicks hatched from albumen-removed eggs. However, the same group demonstrated an increase in catabolic activity in the liver, suggesting the existence of changes in energy allocation between tissues. Besides, we found a decrease in hatching success in the albumen-removed group, suggesting a negative effect of the lower albumen content on eggs, possibly due to lower catabolic activities in skeletal muscle. We also found a compensatory phenomenon in the first week after hatching, i.e., birds from albumen-removed eggs did not show a decrease in MR either at thermoneutral temperatures or at 10°C, compared to the control group. Collectively, our data suggest that a reduction in albumen may generate a trade-off between tissue metabolic activities, and may explain the differences in metabolic rates and hatching success, supporting the immediate adaptive response (IAR) hypothesis.

Abstract:The objective of this study was to evaluate the age-related changes in antioxidant status and the lipid metabolism of pigeon squabs (Columba livia), by determining the BW, antioxidant indices, serum lipid levels, lipid metabolism-related enzyme activities, lipid metabolism-related gene expression, and liver morphology in squabs. Ten squabs were randomly selected and sampled on the day of hatching (DOH), days 7 (D7), 14 (D14) and 21 (D21) post-hatch, respectively. The results showed that BW of squabs increased linearly from DOH to D21. The minimum fold of BW gain was observed in the phase from D14 to D21. Serum triglyceride and free fatty acid levels displayed linear and quadratic trends as age increased, with these maximum responses in D14. Serum low-density lipoprotein cholesterol level responded to age linearly and quadratically with the minimum in D14. Serum high-density lipoprotein cholesterol level and the ratio of high-density lipoprotein cholesterol to low-density lipoprotein cholesterol increased linearly with age, whereas the very low-density lipoprotein cholesterol level decreased linearly. The activities of glutathione peroxidase, catalase, and superoxide dismutase in liver displayed linear and quadratic trends as age increased, with these minimum responses in D14. Hepatic malondialdehyde concentration responded to age linearly and quadratically, with the maximum in D14. Activities of lipoprotein lipase, hepatic lipase, and 3-hydroxy-3-methyl glutaryl coenzyme A reductase in liver responded to age linearly and quadratically, with these minimum responses in D14. Hepatic hormone-sensitive lipase activity displayed linear and quadratic trends as age increased with the maximum in D14. Hepatic acetyl CoA carboxylase activity on D14 was significantly lower than squabs on DOH and D7. Hepatic carnitine palmitoyltransferase 1 mRNA expression responded to age linearly and quadratically, with minimum response in D14. Hepatic mRNA expression of acetyl CoA carboxylase and fatty acid synthetase increased linearly with age. Hepatic Oil-Red-O staining area displayed a quadratic trend as age increased, with the maximum response in D14. In conclusion, the phase from DOH to D14 was a crucial development stage for growth, antioxidant status and lipid metabolism in pigeon squabs. The results suggest it is better to take nutritional manipulation in squabs before D14.

Abstract:This study was conducted to evaluate the effect of in ovo feeding of cationic amino acids on hatchability, hatch weights, and organ developments in pigeon squabs. Two experiments were conducted in this study. Eggs in Exp. 1 were subjected to modification of in ovo feeding in pigeons. Optimal time was determined by checking amniotic fluid volume, and suitable length was confirmed through ink injection. Results showed that the optimum time of in ovo feeding was on d 13 of embryonic development, and the suitable injected length was 20 mm to reach the amniotic cavity of the embryo. Eggs in Exp. 2 were transferred to access in ovo feeding of cationic amino acids. A total of 75 fertile pigeon eggs was randomly distributed into 5 treatments of 15 replicate eggs. Treatments in Exp. 2 consisted of non-injected controls (Control), a sterile buffered solution (0.75% saline), or a cationic amino acid mixture (> 98.5% purity crystalline L-arginine, > 98% purity crystalline L-lysine, and > 98.5% purity L-histidine) containing 0.1, 1, or 10% concentration (Conc.), which were relative to their total content in the eggs, respectively. The crystalline amino acids were dissolved in 200 μL buffered solution prior to in ovo feeding. After hatching, hatch weight (HW) and organ weight (OW) of the squabs were measured immediately. In ovo feeding of cationic amino acids increased the proportions of yolk-free hatch weight to hatch weight (YFHW/HW) (quadratic P = 0.01), and those of OW to YFHW including the heart (quadratic P = 0.01), kidney (quadratic P < 0.01), and liver (quadratic P = 0.02) compared to the control group, and the levels of those ratios were maximized in the 1% Conc group. Also, a proportion of small intestine weight to YFHW improved (linear P = 0.02, quadratic P = 0.05) after in ovo feeding. The organ weight of the head, leg, heart, lung, kidney, proventriculus, pancreas, liver, and small intestine correlated with YFHW positively (0.4 < correlation coefficient < 0.8, P < 0.05). In conclusion, cationic amino acids injection into amnion can improve the embryonic development, which may be mediated by the increment of relative organ weight.

Abstract:The present study was conducted to determine the changes in concentrations of hormones and growth factors and their related receptor gene expressions in crop tissue, relative organ weight, and serum biochemical parameters in male and female pigeons during incubation and chick-rearing periods under artificial farming conditions. Seventy-eight pairs of 60-week-old White King pigeons with 2 fertile eggs per pair were randomly divided into 13 groups by different breeding stages. Serum prolactin and insulin-like growth factor-1 (IGF-1) concentrations in crop tissue homogenates were the highest in both male and female pigeons at 1 d of chick-rearing (R1), while epidermal growth factor (EGF) in female pigeons peaked at d 17 of incubation (I17) (P < 0.05). mRNA expression of the prolactin and EGF receptors in the crop tissue increased at the end of incubation and the early chick-rearing stage in both sexes. However, estrogen, progesterone, and growth hormone receptor expression each decreased during the early chick-rearing stage (P < 0.05). In male pigeons, IGF-1 receptor gene expression reached its peak at R7, while in female pigeons, it increased at the end of incubation. The relative weight of breast and abdominal fat in both sexes and thighs in the males was lowest at R7, and then gradually increased to the incubation period level. Serum total protein, albumin, and globulin concentrations increased to the highest levels at I17 (P < 0.05). Total cholesterol, triglyceride, and low-density lipoprotein reached their highest values at I17 in male pigeons and R25 in female pigeons (P < 0.05). In conclusion, hormones, growth factors, and their receptors potentially underlie pigeon crop tissue development. Changes in organs and serum biochemical profiles suggested their different breeding-cycle patterns with sexual effects.

Abstract:This study was conducted to evaluate gene expression of the amino acid transporter in post-hatch pigeon small intestine and the association of pigeon milk amino acid with the above transporter's gene expression. A total of 48 pigeon breeding families were randomly allocated to 8 groups of 6 replicates of one parental pigeon pair and 2 squabs. Samples of pigeon milk and duodenum, jejunum, and ileum were collected on d 1, 2, 3, 4, 6, 8, 10, and 14 post hatch. The results showed that levels of crude protein (8.93 to 15.56%) were highest in pigeon milk on an air-dry basis. Amino acid content in pigeon milk remained constant in the first 4 d, declined abruptly at d 6, then increased dramatically from d 8 to 14. There was a significant effect of interaction between age and intestinal segments on those amino acid transporters gene expression. mRNA abundance of ATB0'+, SNAT-2, LAT-4, rBAT, b0'+AT, EAAT-3 and PAT-1 was highest in the ileum; B0AT1, asc-1, and IMINO were predominate in the jejunum; and CAT-1 and y+LAT2 were greatest in the duodenum. Age-related changes of amino acid transporter mRNA was inconsistent. mRNA levels of SNAT-2, rBAT, y+LAT2, b0'+AT, and EAAT-3 ascended with age, whereas that of asc-1, CAT-1, and IMINO diminished significantly. Levels of B0AT1 and PAT-1 mRNA abundance were minimized at d 6. However, few correlations were found between pigeon milk amino acid and the amino acid transporter gene expressions in squab small intestine. Our findings provide a comprehensive elaboration on ontogeny of the amino acid transporter in post-hatch pigeon intestine.
Changes in hepatic glucose and lipid metabolism-related parameters in domestic pigeon (Columba livia) during incubation and chick rearing

Abstract:This study aimed to evaluate the hepatic glucose and lipid metabolism-related parameters of adult male and female White King pigeons (Columba livia) during incubation and chick rearing. At day 4 (I4), 10 (I10) and 17 (I17) of incubation and day 1 (R1), 7 (R7), 15 (R15) and 25 (R25) of chick rearing, livers were sampled from six pigeons for each sex. Glycogen and fat contents, activities of glycolytic enzymes (hexokinase, HK; 6-phosphofructokinase, 6-PFK), and genes expressions of key enzymes involved in glycolysis (pyruvate kinase, PK; glucokinase, GK), gluconeogenesis (phosphoenolpyruvate carboxykinase cytosolic, PCK1; fructose-1,6-bisphosphatase, FBP1; glucose-6-phosphatase, G6Pase), fatty acid synthesis (fatty acid synthase, FAS; acetyl-CoA carboxylase, ACC) and fatty acid β-oxidation (carnitine palmitoyltransferase 1, CPT1; acyl-CoA 1, ACO) were measured. In male and female pigeon livers, glycogen content and HK activity dramatically increased after I17 and after R1, respectively; expressions of FBP1 and G6Pase genes were maximized at R15; activity of 6-PFK and expressions of PK and CPT1 genes were highest at R7; fat content and expressions of FAS and ACC genes steeply increased from I10 to R1. In females, hepatic expressions of GK and PCK1 genes were greatest at R7 and I17, respectively; however, in males, both of them were maximized at R15. Hepatic expression of ACO gene was significantly enhanced at R1 compared to I17 and R7 in males, whereas it was notably up-regulated at I17 and R7 in females. Furthermore, expressions of PCK1, GK, FAS and ACC genes were in significant relation to fat content in the livers of female pigeons, while fat content in male pigeons was highly correlated with expression of PCK1, ACC, CPT1 and ACO genes. In conclusion, regulations of glucose and lipid metabolic processes were enhanced in parent pigeon livers from terminal phases of incubation to mid phase of chick rearing with sexual effects.
In-ovo feeding with creatine monohydrate: implications for chicken energy reserves and breast muscle development during the pre-post hatching period

Abstract:The most dynamic period throughout the lifespan of broiler chickens is the pre-post-hatching period, entailing profound effects on their energy status, survival rate, body weight, and muscle growth. Given the significance of this pivotal period, we evaluated the effect of in-ovo feeding (IOF) with creatine monohydrate on late-term embryos' and hatchlings' energy reserves and post-hatch breast muscle development. The results demonstrate that IOF with creatine elevates the levels of high-energy-value molecules (creatine and glycogen) in the liver, breast muscle and yolk sac tissues 48 h post IOF, on embryonic day 19 (p < 0.03). Despite this evidence, using a novel automated image analysis tool on day 14 post-hatch, we found a significantly higher number of myofibers with lower diameter and area in the IOF creatine group compared to the control and IOF NaCl groups (p < 0.004). Gene expression analysis, at hatch, revealed that IOF creatine group had significantly higher expression levels of myogenin (MYOG) and insulin-like growth factor 1 (IGF1), related to differentiation of myogenic cells (p < 0.01), and lower expression of myogenic differentiation protein 1 (MyoD), related to their proliferation (p < 0.04). These results imply a possible effect of IOF with creatine on breast muscle development through differential expression of genes involved in myogenic proliferation and differentiation. The findings provide valuable insights into the potential of pre-hatch enrichment with creatine in modulating post-hatch muscle growth and development.
