Abstract:To better understand the digestive capacity in domestic pigeons (Columba livia), this study was conducted to evaluate nutrient transporters and digestive enzymes gene expression in small intestine and yolk sac membrane (YSM) during pre- and posthatch development. We investigated the oligopeptide transporter Pept1, sodium glucose transporter SGLT1, glucose transporter GLUT2, aminopeptidase-N (APN), and sucrase-isomaltase (SI). Intestine was collected at embryo d 12, 14, and 16, day of hatch, and d 1, 3, 5, 8, and 14 posthatch. The YSM was collected at embryo d 12, 14, 16, and day of hatch. The cDNA fragments for Pept1, SGLT1, GLUT2, APN, and SI were isolated and cloned using reverse-transcription PCR. The sequences data showed that these genes were highly identical to the gene of chicken. The mRNA expression of each gene was assayed using real-time PCR. Expression of intestinal nutrient transporters increased linearly (P<0.001) with age whereas that of APN increased quadratically (P<0.001) and SI increased cubically (P=0.001). Levels of PepT1 mRNA were greatest in the duodenum (P=0.001), GLUT2 and SGLT1 mRNA were greatest in the jejunum (P=0.002) and APN were greatest in the ileum (P=0.004). The YSM expressed all the examined genes. The YSM-expressed genes decreased between embryo d 16 and day of hatch, whereas intestine-expressed genes increased. Our findings provide a comprehensive profile of gene expression patterns of nutrient transporter and digestive enzyme in the small intestine and YSM of pigeons and establish a foundation for future research on the nutrients requirements for young pigeons.
Metabolomics analysis of the yolk of Zhijin white goose during the embryogenesis based on LC-MS/MS

Abstract:The egg yolk of the goose is rich in lipids, proteins and minerals, which is the main source of nutrition during the goose embryogenesis. Actually, the magnitude and variety of nutrients in yolk are dynamically changed to satisfy the nutritional requirements of different growth and development periods. The yolk sac membrane (YSM) plays a role in metabolizing and absorbing nutrients from the yolk, which are then consumed by the embryo or extra-fetal tissues. Therefore, identification of metabolites in egg yolk can help to reveal nutrient requirement in goose embryo. In this research, to explore the metabolite changes in egg yolk at embryonic day (E) 7, E12, E18, E23, and E28, we performed the assay using ultra-high performance liquid chromatography/tandem mass spectrometry (UHPLC-MS/MS). The findings showed that E7 and E12, E23 and E28 were grouped together, while E18 was significantly separated from other groups, indicating the changes of egg yolk development and metabolism. In total, 1472 metabolites were identified in the egg yolk of Zhijin white goose, and 636 differential metabolites (DMs) were screened, among which 264 were upregulated and 372 were downregulated. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that the DMs were enriched in the biosynthesis and metabolism of amino acids, digestion and absorption of protein, citrate cycle (TCA cycle), aminoacyl-tRNA biosynthesis, phosphotransferase system (PTS), mineral absorption, cholesterol metabolism and pyrimidine metabolism. Our study may provide new ideas for improving prehatch embryonic health and nutrition.

Goose Embryonic Development, Glucose and Thyroid Hormone Concentrations, and Eggshell Features Depend on Female Age and Laying Period

Abstract:This study aimed to evaluate embryonic development; analyze the glucose, triiodothyronine (T3), and thyroxine (T4) concentrations in the blood of embryos and goslings; and assess the structure and temperature (EST) of the eggshell. The eggs that were analyzed were from four laying seasons of White Kołuda® geese at three periods (90 eggs × 4 groups × 3 periods). The different embryo proportions, fetal membranes in the egg, and sizes of internal organs indicate a different growth rate and degree of embryo development depending on the laying age and laying period. The goose age influenced the hormone concentrations in the embryos' blood on the 28th day of incubation, which supports a relationship between the females' age and development. The eggshell thickness and density change depending on the laying age and the laying period. A decrease in eggshell thickness in the eggs up to the third season was found after the 16th day of incubation (simultaneously, the density showed an increasing trend). A lower EST distinguished the eggs from the oldest geese in the first half of the hatch. The formation of the chorioallantois membrane was associated with an increase in EST in the oldest geese.

Effects of in Ovo Injection of Carbohydrate Solution on Small Intestine Development in Domestic Pigeons (columba Livia)

Abstract:The objective of this study was to test the hypothesis that in ovo injection of carbohydrates into pigeon (Columba livia) amnion may improve the small intestine development. At d 14.5 of incubation, 80 fertile eggs were injected with 200 μL of carbohydrate solution, and 80 control eggs were not injected. The carbohydrate solution (wt/vol) contained 2.5% maltose + 2.5% sucrose, all dissolved in 0.75% saline. Twelve eggs from each treatment were randomly sampled at d 16 of incubation and the day of hatch, embryos or young pigeons were euthanized, and the jejunum samples were collected. Jejunal villus surface area, activity of the brush border enzymes, sucrase, maltase, aminopeptidase-N, and alkaline phosphatase, and mRNA expression of the digestion-absorption related genes oligopeptide transporter 1, sodium glucose transporter 1, glucose transporter 2, aminopeptidase-N, and sucrase-isomaltase were examined. Results showed that in ovo injection of carbohydrate solution caused a villus surface area increase of 38% on d 16 of incubation and 23% on day of hatch relative to controls (P < 0.05). The in ovo injected pigeons exhibited greater (P < 0.05) activities of jejunal sucrase, maltase, and alkaline phosphatase from d 16 of incubation to day of hatch compared with the controls. At day of hatch, aminopeptidase-N activity in embryos injected in ovo was approximately 27% greater (P < 0.05) than control embryos. Enhanced expressions of the jejunal sodium glucose transporter 1, glucose transporter 2, and aminopeptidase-N mRNA were found at d 16 of incubation in embryos that received carbohydrate solution into the amniotic fluid in comparison with the control group (P < 0.05). These results indicate that the in ovo injected pigeon may hatch with more mature enterocytes and greater intestinal digestive and absorptive capacity than the conventional hatchling. Therefore, the in ovo injected pigeons may become more precocial at hatch and easier to hand-rear during the immediate posthatch period.

Abstract:This study aimed to evaluate the hepatic glucose and lipid metabolism-related parameters of adult male and female White King pigeons (Columba livia) during incubation and chick rearing. At day 4 (I4), 10 (I10) and 17 (I17) of incubation and day 1 (R1), 7 (R7), 15 (R15) and 25 (R25) of chick rearing, livers were sampled from six pigeons for each sex. Glycogen and fat contents, activities of glycolytic enzymes (hexokinase, HK; 6-phosphofructokinase, 6-PFK), and genes expressions of key enzymes involved in glycolysis (pyruvate kinase, PK; glucokinase, GK), gluconeogenesis (phosphoenolpyruvate carboxykinase cytosolic, PCK1; fructose-1,6-bisphosphatase, FBP1; glucose-6-phosphatase, G6Pase), fatty acid synthesis (fatty acid synthase, FAS; acetyl-CoA carboxylase, ACC) and fatty acid β-oxidation (carnitine palmitoyltransferase 1, CPT1; acyl-CoA 1, ACO) were measured. In male and female pigeon livers, glycogen content and HK activity dramatically increased after I17 and after R1, respectively; expressions of FBP1 and G6Pase genes were maximized at R15; activity of 6-PFK and expressions of PK and CPT1 genes were highest at R7; fat content and expressions of FAS and ACC genes steeply increased from I10 to R1. In females, hepatic expressions of GK and PCK1 genes were greatest at R7 and I17, respectively; however, in males, both of them were maximized at R15. Hepatic expression of ACO gene was significantly enhanced at R1 compared to I17 and R7 in males, whereas it was notably up-regulated at I17 and R7 in females. Furthermore, expressions of PCK1, GK, FAS and ACC genes were in significant relation to fat content in the livers of female pigeons, while fat content in male pigeons was highly correlated with expression of PCK1, ACC, CPT1 and ACO genes. In conclusion, regulations of glucose and lipid metabolic processes were enhanced in parent pigeon livers from terminal phases of incubation to mid phase of chick rearing with sexual effects.

Abstract:The effects of in ovo feeding of carbohydrates on hatchability, BW, yolk sac weights (YSW), pectoral muscle weights (PMW), liver and pectoral muscle glycogen concentration, serum glucose level, and hepatic glucose-6-phosphatase activity of domestic pigeons, hatched from eggs laid by a 40-wk-old breeder flock, were investigated. At 14.5 of incubation, fertile eggs were injected with 200 μL of 1.5% maltose (M) + 1.5% sucrose (S), 2.5% M + 2.5% S, 3.5% M + 3.5% S, or 4.5% M + 4.5% S in 0.75% saline, with controls not injected. Results showed that in ovo injection with 1.5% M + 1.5% S or 2.5% M + 2.5% S increased the hatchability compared with the control, whereas injection of 4.5% M + 4.5% S decreased the hatchability. The BW at hatch was quadratic, and BW was maximized by injecting 2.5% M + 2.5% S. The YSW at hatch decreased linearly by the injection with 3.5% M + 3.5% S compared with the control group. In ovo injection of 2.5% M + 2.5% S increased the PMW at hatch. There were no significant differences between any of the treatment groups for liver glycogen reserves. Serum glucose level at hatch was quadratic, and the glucose level was maximized between supplemental 2.5% M + 2.5% S and supplemental 3.5% M + 3.5% S. The pectoral muscle glycogen reserves increased quadratically as supplemental carbohydrates increased, and the response was maximized by injecting 2.5% M + 2.5% S. In conclusion, the present results demonstrate that the injected carbohydrates are available for use and storage. In ovo feeding of carbohydrates, especially at the level of 2.5% M + 2.5% S, on 14.5 d of incubation can improve the hatchability, BW, and PMW by elevating the pectoral muscle glycogen reserves in domestic pigeons at hatch. Results also suggested that in ovo injection of carbohydrates could increase the yolk sac nutrient utilization and hence might enhance the pigeon enteric development.

Abstract:The present study was conducted to determine the changes in concentrations of hormones and growth factors and their related receptor gene expressions in crop tissue, relative organ weight, and serum biochemical parameters in male and female pigeons during incubation and chick-rearing periods under artificial farming conditions. Seventy-eight pairs of 60-week-old White King pigeons with 2 fertile eggs per pair were randomly divided into 13 groups by different breeding stages. Serum prolactin and insulin-like growth factor-1 (IGF-1) concentrations in crop tissue homogenates were the highest in both male and female pigeons at 1 d of chick-rearing (R1), while epidermal growth factor (EGF) in female pigeons peaked at d 17 of incubation (I17) (P < 0.05). mRNA expression of the prolactin and EGF receptors in the crop tissue increased at the end of incubation and the early chick-rearing stage in both sexes. However, estrogen, progesterone, and growth hormone receptor expression each decreased during the early chick-rearing stage (P < 0.05). In male pigeons, IGF-1 receptor gene expression reached its peak at R7, while in female pigeons, it increased at the end of incubation. The relative weight of breast and abdominal fat in both sexes and thighs in the males was lowest at R7, and then gradually increased to the incubation period level. Serum total protein, albumin, and globulin concentrations increased to the highest levels at I17 (P < 0.05). Total cholesterol, triglyceride, and low-density lipoprotein reached their highest values at I17 in male pigeons and R25 in female pigeons (P < 0.05). In conclusion, hormones, growth factors, and their receptors potentially underlie pigeon crop tissue development. Changes in organs and serum biochemical profiles suggested their different breeding-cycle patterns with sexual effects.

Abstract:The objective of the study was to investigate the mechanism by which dietary energy concentration regulates laying performance in geese. Eighty 558-day-old female Sichuan White geese were randomly allotted to two dietary treatments, each treatment was fed 1 of 2 experimental diets containing 10.00 (deficient) or 11.80MJ/kg metabolizable energy (sufficient) for 30 days. Laying performance, hormone concentration and gene expressions in hypothalamus-pituitary-gonadal axis were examined in geese. Birds fed the sufficient-energy diet had significantly higher average egg weight, daily laying rate, and lower feed to egg ratio than those fed the deficient-energy (p<0.05). The birds fed sufficient-energy diet had higher concentration of serum insulin like growth factor 1 (IGF-1), gonadotropin-releasing hormone (GnRH), follicle-stimulating hormone (FSH) and estradiol (E-2) than those in deficient-energy diet (p<0.05). The mRNA expression levels of GnRH in the hypothalamus, FSH in the pituitary and E-2 in the ovary of birds fed sufficient-energy diet were higher than the corresponding counterpart in deficient-energy diet (p<0.05), respectively. In conclusion, the study implied that dietary energy modifies laying possibly through regulating reproductive hormone secretion and gene expression in hypothalamus-pituitary-gonad axis in laying geese.

Abstract:We present analytical and numerical results on the heat conduction in a linear mixing system. In particular we consider a quasi-one-dimensional channel with triangular scatterers with internal angles which are irrational multiples of pi, and we show that the system obeys the Fourier law of heat conduction. Therefore, deterministic diffusion and normal heat transport which are usually associated with full hyperbolicity, actually take place in systems without exponential instability.
Comparative Transcriptome Analysis Provides Novel Insights into the Effect of Lipid Metabolism on Laying of Geese

Abstract:The lower egg production of geese (20~60 eggs per year) compared with chicken and duck limits the development of the industry, while the yolk weight and fatty liver susceptibility of geese was higher than that of other poultry. Therefore, the relationship between lipid metabolism and the laying performance of geese remains to be explored. Phenotypically, we observed that the liver fat content of the high-, low-, and no-egg production groups decreased in turn, while the abdominal fat weight increased in turn. For transcriptional regulation, the KEGG pathways related to lipid metabolism were enriched in all pairwise comparisons of abdominal fat and liver through functional analysis. However, some KEGG pathways related to inflammation and the circadian rhythm pathway were enriched by DEGs only in abdominal fat and the liver, respectively. The DEGs in ovarian stroma among different groups enriched some KEGG pathways related to ovarian steroidogenesis and cell adhesion. Our research reveals that lipid metabolism regulated by the circadian rhythm of the liver may directly or indirectly affect ovarian function through the inflammation and hormone secretion of abdominal fat. These results offer new insights into the regulation mechanisms of goose reproductive traits.

The developmental pattern related to fatty acid uptake and oxidation in the yolk sac membrane and jejunum during embryogenesis in Muscovy duck

Abstract:This study aimed to investigate the developmental change of body growth and gene expression related to fatty acid uptake and oxidation in the yolk sac membrane (YSM) and jejunum during embryogenesis in Muscovy ducks. The weights of embryos and yolk sac (YS) (5 embryos per replicate, n = 6) were recorded on embryonic days (E)16, E19, E22, E25, E28, E31, and the day of hatch (DOH). The fat and fatty acid contents in YSM, jejunal histology, and gene expression related to fatty acid metabolism in YSM and jejunum were determined in each sampling time. Among the nonlinear models, the maximum growth is estimated at 2.83 (E22.5), 2.67 (E22.1), and 2.60 (E21.3) g/d using logistic, Gompertz, and Von Bertalanffy models, respectively. The weight of YS, and ether extract-free YS as well as the amounts of fat and fatty acids in YS decreased (P < 0.05) linearly, whereas the villus height, crypt depth, villus height/crypt depth, and musculature thickness in jejunum increased (P < 0.05) linearly during embryogenesis. The mRNA expression of CD36, SLC27A4, and FABP1 related to fatty acid uptake as well as the mRNA and protein expressions of PPARα and CPT1 related to fatty acid oxidation increased in a quadratic manner (P < 0.05) in both YS and jejunum, and the maximum values were achieved during E25 to E28. In conclusion, the maximum growth rate of Muscovy duck embryos was estimated at 2.60 to 2.83 g/d on E21.3 to E23.5, while the accumulations of lipid and fatty acid in YS were decreased in association with the increased absorptive area of morphological structures in jejunum. The gene and protein expression involved in fatty acid metabolism displayed a similar enhancement pattern between YSM and jejunum during E25 to E28, suggesting that fatty acid utilization could be strengthened to meet the energy demand for embryonic development.
Goose Hepatic IGFBP2 Is Regulated by Nutritional Status and Participates in Energy Metabolism Mainly through the Cytokine-Cytokine Receptor Pathway

Abstract:Changes in the nutritional status of animals significantly affect their health and production performance. However, it is unclear whether insulin-like growth factor-binding protein 2 (IGFBP2) mediates these effects. This study aimed to investigate the impact of changes in nutritional and energy statuses on hepatic IGFBP2 expression and the mechanism through which IGFBP2 plays a mediating role. Therefore, the expression of IGFBP2 was first determined in the livers of fasting/refeeding and overfeeding geese. The data showed that overfeeding inhibited IGFBP2 expression in the liver compared with the control (normal feeding) group, whereas the expression of IGFBP2 in the liver was induced by fasting. Interestingly, the data indicated that insulin inhibited the expression of IGFBP2 in goose primary hepatocytes, suggesting that the changes in IGFBP2 expression in the liver in the abovementioned models may be partially attributed to the blood insulin levels. Furthermore, transcriptome sequencing analysis showed that the overexpression of IGFBP2 in geese primary hepatocytes significantly altered the expression of 337 genes (including 111 up-regulated and 226 down-regulated genes), and these differentially expressed genes were mainly enriched in cytokine-cytokine receptor, immune, and lipid metabolism-related pathways. We selected the most significant pathway, the cytokine-cytokine receptor pathway, and found that the relationship between the expression of these genes and IGFBP2 in goose liver was in line with the findings from the IGFBP2 overexpression assay, i.e., the decreased expression of IGFBP2 was accompanied by the increased expression of LOC106041919, CCL20, LOC106042256, LOC106041041, and IL22RA1 in the overfed versus normally fed geese, and the increased expression of IGFBP2 was accompanied by the decreased expression of these genes in fasting versus normally fed geese, and refeeding prevented or attenuated the effects of fasting. The association between the expression of these genes and IGFBP2 was verified by IGFBP2-siRNA treatment of goose primary hepatocytes, in which IGFBP2 expression was induced by low serum concentrations. In conclusion, this study suggests that IGFBP2 mediates the biological effects induced by changes in nutritional or energy levels, mainly through the cytokine-cytokine receptor pathway.
A Serum Metabolic Profiling Analysis During the Formation of Fatty Liver in Landes Geese Via GC-TOF/MS

Abstract:During the process of fatty liver production by overfeeding, the levels of endogenous metabolites in the serum of geese would change dramatically. This study investigated the effects of overfeeding on serum metabolism of Landes geese and the underlying mechanisms using a metabolomics approach. Sixty Landes geese of the same age were randomly divided into the following three groups with 20 replicates in each group: D0 group (free from gavage); D7 group (overfeeding for 7 days); D25 group (overfeeding for 25 days). At the end of the experiment, 10 geese of similar weight from each group were selected for slaughter and sampling. The results showed that overfeeding significantly increased the body weight and the liver weight of geese. Serum enzymatic activities and serum lipid levels were significantly enhanced following overfeeding. Gas chromatography time-of-flight/mass spectrometry (GC-TOF/MS) was employed to explore the serum metabolic patterns, and to identify potential contributors to the formation of fatty liver and the correlated metabolic pathways. Relative to overfeeding for 7 days, a large number of endogenous molecules in serum of geese overfed for 25 days were altered. Continuous elevated levels of pyruvic acid, alanine, proline and beta-glycerophosphoric acid and reduced lactic acid level were observed in the serum of overfed geese. Pathway exploration found that the most of significantly different metabolites were involved in amino acids, carbohydrate and lipid metabolism. The present study exhibited the efficient capability of Landes geese to produce fatty liver, identified potential biomarkers and disturbed metabolic pathways in liver steatosis. These findings might reveal the underlying mechanisms of fatty liver formation and provide some theoretical basis for the diagnosis and treatment of liver diseases.

Abstract:Summary Parotid saliva was collected from normocalcemic and hypercalcemia subjects, and it was found that the latter had higher parotid fluid calcium and phosphorus levels than the controls. The observations did not permit discrimination as to the etiology of the hypercalcemia. The reasons for the elevated levels of phosphorus have not been established.
Gene Expression of Amino Acid Transporter in Pigeon (columbia Livia) Intestine During Post-Hatch Development and Its Correlation with Amino Acid in Pigeon Milk.

Abstract:This study was conducted to evaluate gene expression of the amino acid transporter in post-hatch pigeon small intestine and the association of pigeon milk amino acid with the above transporter's gene expression. A total of 48 pigeon breeding families were randomly allocated to 8 groups of 6 replicates of one parental pigeon pair and 2 squabs. Samples of pigeon milk and duodenum, jejunum, and ileum were collected on d 1, 2, 3, 4, 6, 8, 10, and 14 post hatch. The results showed that levels of crude protein (8.93 to 15.56%) were highest in pigeon milk on an air-dry basis. Amino acid content in pigeon milk remained constant in the first 4 d, declined abruptly at d 6, then increased dramatically from d 8 to 14. There was a significant effect of interaction between age and intestinal segments on those amino acid transporters gene expression. mRNA abundance of ATB0'+, SNAT-2, LAT-4, rBAT, b0'+AT, EAAT-3 and PAT-1 was highest in the ileum; B0AT1, asc-1, and IMINO were predominate in the jejunum; and CAT-1 and y+LAT2 were greatest in the duodenum. Age-related changes of amino acid transporter mRNA was inconsistent. mRNA levels of SNAT-2, rBAT, y+LAT2, b0'+AT, and EAAT-3 ascended with age, whereas that of asc-1, CAT-1, and IMINO diminished significantly. Levels of B0AT1 and PAT-1 mRNA abundance were minimized at d 6. However, few correlations were found between pigeon milk amino acid and the amino acid transporter gene expressions in squab small intestine. Our findings provide a comprehensive elaboration on ontogeny of the amino acid transporter in post-hatch pigeon intestine.

Abstract:The purpose of this study was to assess the relationship between the isometric and isokinetic peak torque (PT) (speed of movement 0, 60, and 180 degrees.s-1) and the peak torque acceleration energy (PTAE) and average power (AP) of multiple contractions (180 degrees.s-1) of the quadriceps and hamstrings in 38 patients having a chronic, complete anterior cruciate ligament (ACL) insufficiency in the knee. At every test speed the Pearson product moment (r) and the Spearman rank (rs) correlation coefficients between the PT and PTAE were highly significant for both muscle groups in the uninjured (r = 0.79-0.94, rs = 0.78-0.94) as well as in the ACL-insufficient (r = 0.87-0.95, rs = 0.83-0.94) knees. The results were the same between the PT and AP (uninjured: r = 0.70-0.90, rs = 0.68-0.90; ACL insufficient: r = 0.86-0.93, rs = 0.82-0.93). In both knees and in both muscle groups the correlation coefficients increased simultaneously with the speed of movement, obviously since the PTAE and AP were registered at the highest test speed (180 degrees.s-1). In conclusion, concerning ACL-insufficient or uninjured knees, the PTAE or AP analysis may offer little additional information about thigh muscle function to that attained by more simple measurement, the PT analysis.

Abstract:Introduction: The adverse effects of high glucose on embryos can be traced to the preimplantation stage. This study aimed to observe the effect of high glucose on early-stage embryos. Methods and results: Seven-week-old ICR female mice were superovulated and mated, and the zygotes were collected. The zygotes were randomly cultured in 5 different glucose concentrations (control, 20mM, 40mM, 60mM and 80mM glucose). The cleavage rate, blastocyst rate and total cell number of blastocyst were used to assess the embryo quality. 40 mM glucose was selected to model high glucose levels in this study. 40mM glucose arrested early embryonic development, and the blastocyst rate and total cell number of the blastocyst decreased significantly as glucose concentration was increased. The reduction in the total cell number of blastocysts in the high glucose group was attributed to decreased proliferation and increased cell apoptosis, which is associated with the diminished expression of GLUTs (GLUT1, GLUT2, GLUT3). Furthermore, the metabolic characterization of blastocyst culture was observed in the high-glucose environment. Discussion: The balance of glycolysis and oxidative phosphorylation at the blastocyst stage was disrupted. And embryo development arrest due to high glucose is associated with changes in glycolysis and oxidative phosphorylation, as well as abnormalities in the TCA cycle and amino acid metabolism.

Abstract:Egg performance significantly impacts the development of the local goose industry. The hypothalamus plays an essential role in the egg production of birds. However, few potential candidate genes and biological functions related to egg production in geese have been identified in hypothalamus tissue. In this study, 115 geese were raised and observed for 5 months during the laying period. To understand the regulation mechanism of egg production, the hypothalamus transcriptome profiles of these geese were sequenced using RNA-seq. The hypothalamus samples of four high egg production (HEP) and four low egg production (LEP) geese were selected and collected, respectively. A total of 14,679 genes were identified in the samples. After multiple bioinformatics analyses, Gene Ontology (GO) annotations indicated that genes related to egg production were mainly enriched in biological processes of "response to light stimulus," "sensory system development," and "visual perception." Six potential candidate genes (PDE6C, RHO, MFRP, F2, APOB, and IL6) based on their corresponding GO terms and interaction networks were identified. These identified candidate genes can be used as selection markers to improve the egg production of Wanxi white geese. Our study highlights how visual perception may affect the regulation of geese egg production.

Abstract:Gestational diabetes mellitus (GDM) is a worldwide pregnancy complication. Gestational diabetes can significantly impact fetus development. However, the effects of high glucose on embryological development post-fertilization are yet to be researched. Danio rerio embryos are a great model for studying embryonic development. In this study, the effects on embryological (morphological and genetic) development were examined in the presence of a high-glucose environment that mimics the developing fetus in pregnant women with GDM. Fertilized zebrafish embryos were treated with normal media and high glucose for 5 days from 3 h post-fertilization (hpf) to 96 hpf, respectively, as control and experimental groups. Morphological changes are recorded with microscope images. Hatch rate and heart rate are compared between groups at set time points. RNA-Seq is performed to examine the gene changes in the experimental group. Glucose delayed the zebrafish embryo development by slowing the hatch rate by about 24 h. The brain, heart, and tail started showing smaller morphology in the glucose group compared to the control group at 24 hpf. Heart rate was faster in the glucose group compared to the control group on days 2 and 3 with a statistically significant difference. Among the zebrafish whole genome, the significantly changed genes were 556 upregulated genes and 1118 downregulated genes, respectively, in the high-glucose group. The metabolic and Wnt pathways are altered under high-glucose conditions. These conditions contribute to significant physiological differences that may provide insight into the functionality of post-embryological development.
