Abstract:BACKGROUND:Butanol is an industrial commodity and also considered to be a more promising gasoline substitute compared to ethanol. Renewed attention has been paid to solvents (acetone, butanol and ethanol) production from the renewable and inexpensive substrates, for example, lignocellulose, on account of the depletion of oil resources, increasing gasoline prices and deteriorating environment. Limited to current tools for genetic manipulation, it is difficult to develop a genetically engineered microorganism with combined ability of lignocellulose utilization and solvents production. Mixed culture of cellulolytic microorganisms and solventogenic bacteria provides a more convenient and feasible approach for ABE fermentation due to the potential for synergistic utilization of the metabolic pathways of two organisms. But few bacteria pairs succeeded in producing biobutanol of high titer or high productivity without adding butyrate. The aim of this work was to use Clostridium cellulovorans 743B to saccharify lignocellulose and produce butyric acid, instead of adding cellulase and butyric acid to the medium, so that the soluble sugars and butyric acid generated can be subsequently utilized by Clostridium beijerinckii NCIMB 8052 to produce butanol in one pot reaction.RESULTS:A stable artificial symbiotic system was constructed by co-culturing a celluloytic, anaerobic, butyrate-producing mesophile (C. cellulovorans 743B) and a non-celluloytic, solventogenic bacterium (C. beijerinckii NCIMB 8052) to produce solvents by consolidated bioprocessing (CBP) with alkali extracted deshelled corn cobs (AECC), a low-cost renewable feedstock, as the sole carbon source. Under optimized conditions, the co-culture degraded 68.6 g/L AECC and produced 11.8 g/L solvents (2.64 g/L acetone, 8.30 g/L butanol and 0.87 g/L ethanol) in less than 80 h. Besides, a real-time PCR assay based on the 16S rRNA gene sequence was performed to study the dynamics of the abundance of each strain during the co-culturing process, which figured out the roles of each strain at different periods in the symbiosis.CONCLUSION:Our work illustrated the great potential of artificial symbiosis in biofuel production from lignocellulosic biomass by CBP. The dynamics of the abundance of C. beijerinckii and C. cellulovorans revealed mechanisms of cooperation and competition between the two strains during the co-culture process.

Abstract:2,3-butanediol is a valuable chemical feedstock and liquid fuel.Biological production of 2,3-butanediol from low-cost cellulosic material is of both economic and social benefits.Fermentation of 2,3-butanediol from corn stover hydrolysate by Klebsiella oxytoca ZU-03 was studied,during which both hexose and pentose in the hydrolysate were fully used as substrate.The results show that sodium sulfate concentration under 20 g/L in corn stover hydrolysate has no significant effects on fermentation;acetic acid and ethanol,as the main byproducts,affected the fermentation when their concentrations were higher than 20 and 5 g/L respectively.The optimal initial pH is 6.0-6.5 and the initial total sugar concentration is 80-100 g/L.When the total sugar concentration is 80 g/L(glucose 47.25 g/L,xylose 32.75 g/L,the cultures were shaking at 30 ℃ for 60 h with initial pH 6.0,a 2,3-butanediol concentration of 37.20 g/L was achieved and 99.36 % of the total sugar was consumed with the yield of 0.468 g/g(total sugar),which reached 94 % of the theoretical maximum.This result may be very useful in enhancing the efficient usage of cellulosic biomass.

Abstract:Producing biofuels directly from cellulose, known as consolidated bioprocessing, is believed to reduce costs substantially compared to a process in which cellulose degradation and fermentation to fuel are accomplished in separate steps. Here we present a metabolic engineering example for the development of a Clostridium cellulolyticum strain for isobutanol synthesis directly from cellulose. This strategy exploits the host's natural cellulolytic activity and the amino acid biosynthesis pathway and diverts its 2-keto acid intermediates toward alcohol synthesis. Specifically, we have demonstrated the first production of isobutanol to approximately 660 mg/liter from crystalline cellulose by using this microorganism.
Modulation of the Acetone/Butanol Ratio during Fermentation of Corn Stover-Derived Hydrolysate by Clostridium beijerinckii Strain NCIMB 8052

Abstract:ABSTRACT Producing biobutanol from lignocellulosic biomass has shown promise to ultimately reduce greenhouse gases and alleviate the global energy crisis. However, because of the recalcitrance of a lignocellulosic biomass, a pretreatment of the substrate is needed which in many cases releases soluble lignin compounds (SLCs), which inhibit growth of butanol-producing clostridia. In this study, we found that SLCs changed the acetone/butanol ratio (A/B ratio) during butanol fermentation. The typical A/B molar ratio during Clostridium beijerinckii NCIMB 8052 batch fermentation with glucose as the carbon source is about 0.5. In the present study, the A/B molar ratio during batch fermentation with a lignocellulosic hydrolysate as the carbon source was 0.95 at the end of fermentation. Structural and redox potential changes of the SLCs were characterized before and after fermentation by using gas chromatography/mass spectrometry and electrochemical analyses, which indicated that some exogenous SLCs were involved in distributing electron flow to C. beijerinckii , leading to modulation of the redox balance. This was further demonstrated by the NADH/NAD + ratio and trxB gene expression profile assays at the onset of solventogenic growth. As a result, the A/B ratio of end products changed significantly during C. beijerinckii fermentation using corn stover-derived hydrolysate as the carbon source compared to glucose as the carbon source. These results revealed that SLCs not only inhibited cell growth but also modulated the A/B ratio during C. beijerinckii butanol fermentation. IMPORTANCE Bioconversion of lignocellulosic feedstocks to butanol involves pretreatment, during which hundreds of soluble lignin compounds (SLCs) form. Most of these SLCs inhibit growth of solvent-producing clostridia. However, the mechanism by which these compounds modulate electron flow in clostridia remains elusive. In this study, the results revealed that SLCs changed redox balance by producing oxidative stress and modulating electron flow as electron donors. Production of H 2 and acetone was stimulated, while butanol production remained unchanged, which led to a high A/B ratio during C. beijerinckii fermentation using corn stover-derived hydrolysate as the carbon source. These observations provide insight into utilizing C. beijerinckii to produce butanol from a lignocellulosic biomass.

Abstract:Clostridium butyricum SCUT 620, a promising biorefinery chassis, has been demonstrated to efficiently utilize monosaccharides, disaccharides, and polysaccharides for butyric acid production. However, the absence of genetic manipulation tools has restricted its further development and application. For the first time, an efficient electroporation method for C. butyricum SCUT 620 was developed based on the analysis of the restriction-modification system and in vivo methylation. The highest electrotransformation efficiency reached 785 transformants/μg DNA after optimizing electroporation parameters. As a proof of concept, C. butyricum SCUT 620 was genetically modified to improve starch hydrolysis and butyrate tolerance, and to enable the synthesis of a new product, butanol, using non-grain biomass cassava starch. This study paves the way for the metabolic engineering of C. butyricum SCUT 620, with the objective of achieving various substrates utilization and chemicals production.

Abstract:It is attractive to co-produce n-butanol and isobutanol from lignocellulosic biomass in engineered strains or consortia, because they can be coseparated and combined for use as biofuel. However, very few attempts have been made to date. To address that, a heterologous isobutanol pathway was first introduced to enable Clostridium beijerinckii to co-produce n-butanol and isobutanol. Following medium optimization, the recombinant C. beijerinckii produced 194 mg/L isobutanol and 7.16 g/L n-butanol from glucose. C. cellulovorans was then genetically engineered by overexpressing adhE1, kivD, and yqhD to synthesize n-butanol and isobutanol with alkali-extracted, deshelled corn cobs (AECC) as the sole carbon source. It produced 156 mg/L isobutanol and 1.81 g/L n-butanol from 39.5 g/L AECC within 120 h. Finally, the above two recombinant strains were mix-cultured and optimized. The optimal consortium degraded 55.1 g/L AECC and produced 1.05 g/L isobutanol and 6.22 g/L n-butanol, which were 6.73- and 3.44-fold higher than those obtained from single culture of recombinant C. cellulovorans. Besides, the titer and yield of total butanol are 15.6% and 11.9%, respectively, higher than those of recombinant consortia that only produce n-butanol. Synthetic biology empowers artificial consortia with expanded functions, as well as improved performance, showing great potential in consolidated bioprocessing.

Abstract:Cellulosic n‐butanol from renewable lignocellulosic biomass has gained increased interest. Previously, we have engineered Clostridium cellulovorans, a cellulolytic acidogen, to overexpress the bifunctional butyraldehyde/butanol dehydrogenase gene adhE2 from Clostridium acetobutylicum for n‐butanol production from crystalline cellulose. However, butanol production by this engineered strain had a relatively low yield of ~0.22 g/g cellulose due to the co‐production of ethanol and acids. We hypothesized that strengthening the carbon flux through the central butyryl‐CoA biosynthesis pathway and increasing intracellular NADH availability in C. cellulovorans adhE2 would enhance n‐butanol production. In this study, thiolase (thlACA) from Clostridium acetobutylicum and 3‐hydroxybutyryl‐CoA dehydrogenase (hbdCT) from Clostridium tyrobutyricum were overexpressed in C. cellulovorans adhE2 to increase the flux from acetyl‐CoA to butyryl‐CoA. In addition, ferredoxin‐NAD(P)+ oxidoreductase (fnr), which can regenerate the intracellular NAD(P)H and thus increase butanol biosynthesis, was also overexpressed. Metabolic flux analyses showed that mutants overexpressing these genes had a significantly increased carbon flux toward butyryl‐CoA, which resulted in increased production of butyrate and butanol. The addition of methyl viologen as an electron carrier in batch fermentation further directed more carbon flux towards n‐butanol biosynthesis due to increased reducing equivalent or NADH. The engineered strain C. cellulovorans adhE2‐fnrCA‐thlACA‐hbdCT produced n‐butanol from cellulose at a 50% higher yield (0.34 g/g), the highest ever obtained in batch fermentation by any known bacterial strain. The engineered C. cellulovorans is thus a promising host for n‐butanol production from cellulosic biomass in consolidated bioprocessing.This article is protected by copyright. All rights reserved.

Abstract:Bio-refining lignocellulosic resource offers a renewable and sustainable approach for producing biofuels and biochemicals. However, the conversion efficiency of lignocellulosic resource is still challenging due to the intrinsic inefficiency in co-utilization of xylose and glucose. In this study, the industrial bacterium Bacillus licheniformis was engineered for biorefining lignocellulosic resource to produce acetoin. First, adaptive evolution was conducted to improve acetoin tolerance, leading to a 19.6 % increase in acetoin production. Then, ARTP mutagenesis and 60 Co-γ irradiation was carried out to enhance the production of acetoin, obtaining 73.0 g/L acetoin from glucose. Further, xylose uptake and xylose utilization pathway were rewired to facilitate the co-utilization of xylose and glucose, enabling the production of 60.6 g/L acetoin from glucose and xylose mixtures. Finally, this efficient cell factory was utilized for acetoin production from lignocellulosic hydrolysates with the highest titer of 68.3 g/L in fed-batch fermentation. This strategy described here holds great applied potential in the biorefinery of lignocellulose for the efficient synthesis of high-value chemicals.

Metabolic and evolutionary responses of Clostridium thermocellum to genetic interventions aimed at improving ethanol production

Abstract:Background: Engineering efforts targeted at increasing ethanol by modifying the central fermentative metabolism of Clostridium thermocellum have been variably successful. Here, we aim to understand this variation by a multifaceted approach including genomic and transcriptomic analysis combined with chemostat cultivation and high solids cellulose fermentation. Three strain lineages comprising 16 strains total were examined. Two strain lineages in which genes involved in pathways leading to organic acids and/or sporulation had been knocked out resulted in four end-strains after adaptive laboratory evolution (ALE). A third strain lineage recapitulated mutations involving adhE that occurred spontaneously in some of the engineered strains. Results: Contrary to lactate dehydrogenase, deleting phosphotransacetylase (pta, acetate) negatively affected steady-state biomass concentration and caused increased extracellular levels of free amino acids and pyruvate, while no increase in ethanol was detected. Adaptive laboratory evolution (ALE) improved growth and shifted elevated levels of amino acids and pyruvate towards ethanol, but not for all strain lineages. Three out of four end-strains produced ethanol at higher yield, and one did not. The occurrence of a mutation in the adhE gene, expanding its nicotinamide-cofactor compatibility, enabled two end-strains to produce more ethanol. A disruption in the hfsB hydrogenase is likely the reason why a third end-strain was able to make more ethanol. RNAseq analysis showed that the distribution of fermentation products was generally not regulated at the transcript level. At 120 g/L cellulose loadings, deletions of spo0A, ldh and pta and adaptive evolution did not negatively influence cellulose solubilization and utilization capabilities. Strains with a disruption in hfsB or a mutation in adhE produced more ethanol, isobutanol and 2,3-butanediol under these conditions and the highest isobutanol and ethanol titers reached were 5.1 and 29.9 g/L, respectively. Conclusions: Modifications in the organic acid fermentative pathways in Clostridium thermocellum caused an increase in extracellular pyruvate and free amino acids. Adaptive laboratory evolution led to improved growth, and an increase in ethanol yield and production due a mutation in adhE or a disruption in hfsB. Strains with deletions in ldh and pta pathways and subjected to ALE demonstrated undiminished cellulolytic capabilities when cultured on high cellulose loadings.

Abstract:Butanol is widely used as an important bulk chemical and is a potential biofuel. The depletion of fossil fuels and advances in synthetic biotechnology have led to renewed interest in the biological production of butanol. Solventogenic Clostridium was commonly used to produce butanol through traditional acetone‐butanol‐ethanol (ABE) fermentation. However, its relatively slow growth rate, low butanol tolerance, and poor production efficiency have hindered the further application of this procedure. Recently, other promising industrial hosts, including Escherichia coli , Saccharomyces cerevisiae , and Clostridium tyrobutyricum , have been studied for potential use in the production of butanol. This review comprehensively summarizes the advantages and challenges of different non‐solvent strains for butanol production to identify better the ideal non‐solvent hosts for butanol production. Abstract:Abstract Background Butyl acetate is a versatile compound that is widely used in the chemical and food industry. The conventional butyl acetate synthesis via Fischer esterification of butanol and acetic acid using catalytic strong acids under high temperature is not environmentally benign. Alternative lipase-catalyzed ester formation requires a significant amount of organic solvent which also presents another environmental challenge. Therefore, a microbial cell factory capable of producing butyl acetate through fermentation of renewable resources would provide a greener approach to butyl acetate production. Result Here, we developed a metabolically engineered strain of Escherichia coli that efficiently converts glucose to butyl acetate. A modified Clostridium CoA-dependent butanol production pathway was used to synthesize butanol which was then condensed with acetyl-CoA through an alcohol acetyltransferase. Optimization of alcohol acetyltransferase expression and redox balance with auto-inducible fermentative controlled gene expression led to an effective titer of 22.8 ± 1.8 g/L butyl acetate produced in a bench-top bioreactor. Conclusion Building on the well-developed Clostridium CoA-dependent butanol biosynthetic pathway, expression of an alcohol acetyltransferase converts the butanol produced into butyl acetate. The results from this study provided a strain of E. coli capable of directly producing butyl acetate from renewable resources at ambient conditions.

Abstract:Bioproduction of renewable chemicals is considered as an urgent solution for fossil energy crisis. However, despite tremendous efforts, it is still challenging to generate microbial strains that can produce target biochemical to high levels. Here, we report an example of biosynthesis of high-value and easy-recoverable derivatives built upon natural microbial pathways, leading to improvement in bioproduction efficiency. By leveraging pathways in solventogenic clostridia for co-producing acyl-CoAs, acids and alcohols as precursors, through rational screening for host strains and enzymes, systematic metabolic engineering-including elimination of putative prophages, we develop strains that can produce 20.3g/L butyl acetate and 1.6g/L butyl butyrate. Techno-economic analysis results suggest the economic competitiveness of our developed bioprocess. Our principles of selecting the most appropriate host for specific bioproduction and engineering microbial chassis to produce high-value and easy-separable end products may be applicable to other bioprocesses. Esters can be used as fuels and specialty chemicals for food flavoring, cosmetic and pharmaceutical industries. Here, the authors systematically engineer clostridia, including discovery and deletion of prophages to increase strain stability, for the production of butyl acetate and butyl butyrate from corn stover at low cost.

Abstract:This study has addressed the issue of enhanced biobutanol production through Consolidated Bioprocessing, with the use of clostridial co-cultures combining C. acetobutylicum (Cac) and C. pasteurianum (Cpa). To begin with, the total Acetone-Butanol-Ethanol (ABE) solvent production and butanol selectivity of individual clostridial cultures grown on different carbon sources present in lignocellulosic hydrolysates. Thereafter, statistical optimization of ABE fermentation with co-culture system was carried out using Box-Behnken design. The optimization variables were Cac to Cpa inoculum ratio (X 1 in mL mL−1), sodium concentration in the medium (X 2 g L−1), and xylose/glucose ratio (X 3 g g−1 L L−1), whereas response variables were butanol titre (Y 1 in g L−1), biomass growth (Y 2 OD600), total ABE titre (Y 3 in g L−1). Maximum values of the response variables with the corresponding set of optimization parameters were as follows: Y 1, max = 12.1 ± 0.45 (for X 1 = 0.30, X 2 = 5.90, X 3 = 0), Y 2, max = 4.15 ± 0.03 (for X 1 = 0.3, X 2 = 2.42, X 3 = 0.14), Y 3, max = 23.1 ± 0.55 (for X 1 = 0.46, X 2 = 6.36, X 3 = 0). The study demonstrated a substantial ≈ 22% and 61% increment in butanol titre in co-culture system compared to individual fermentations with Cac and Cpa grown on pure glucose, respectively.

Abstract:Isobutanol is regarded as a renewable alternative to fossil fuels. In this study, Bacillus licheniformis DWc9n was able to grow in the presence of 16 g/L isobutanol, showing high resistance to isobutanol. To achieve high-level production of isobutanol, a push–pull–restrain strategy was employed. First, native alcohol dehydrogenase YugJ with the highest synthesis efficiency of isobutanol was identified by comparing different heterologous and endogenous alcohol dehydrogenases. Eliminating competing pathways resulted in a remarkable increase in isobutanol production. Moreover, gene bcd encoding leucine dehydrogenase was identified as the rate-limiting enzyme for isobutanol production, and overexpression of bcd led to a 3.29-fold increase in isobutanol titer, increasing to 5.54 g/L from 1.29 g/L. Subsequently, we conducted chromosome engineering to increase the supply of precursor α-ketoisovalerate and combined cofactor engineering to increase the availability of NADPH. The resultant strain IB13 produced 7.81 g/L isobutanol, increasing by 40.11-fold compared to its parental strain IB1. Finally, 10.80 g/L isobutanol with a yield of 44% (mol/mol glucose) in shake flasks was achieved under the optimal fermentation conditions, which was the highest obtained for Bacillus species. Our results demonstrated the potential to engineer B. licheniformis strain to produce 2-keto acid-derived chemicals such as isobutanol efficiently from renewable feedstocks.

Abstract:BackgroundThe shortage of food based feedstocks has been one of the stumbling blocks in industrial biomanufacturing. The acetone bioproduction from the traditional acetone–butanol–ethanol fermentation is limited by the non-specificity of products and competitive utilization of food-based substrates. Using genetically modified Escherichia coli to produce acetone as sole product from the cost-effective non-food based substrates showed great potential to overcome these problems.ResultsA novel acetone biosynthetic pathway were constructed based on genes from Clostridium acetobutylicum (thlA encoding for thiolase, adc encoding for acetoacetate decarboxylase, ctfAB encoding for coenzyme A transferase) and Escherichia coli MG1655 (atoB encoding acetyl-CoA acetyltransferase, atoDA encoding for acetyl-CoA: acetoacetyl-CoA transferase subunit α and β). Among these constructs, one recombinant MG1655 derivative containing the hybrid pathway consisting of thlA, atoDA, and adc, produced the highest level of acetone from acetate. Reducing the gluconeogenesis pathway had little effect on acetone production, while blocking the TCA cycle by knocking out the icdA gene enhanced the yield of acetone significantly. As a result, acetone concentration increased up to 113.18 mM in 24 h by the resting cell culture coupling with gas-stripping methods.ConclusionsAn engineered E. coli strain with optimized hybrid acetone biosynthetic pathway can utilize acetate as substrate efficiently to synthesize acetone without other non-gas byproducts. It provides a potential method for industrial biomanufacturing of acetone by engineered E. coli strains from non-food based substrate.

Abstract:Background: Clostridium acetobutylicum is one of the most important butanol producing strains. However, environmental stress in the fermentation process usually leads to a lower yield, seriously hampering its industrialization. In order to systematically investigate the key intracellular metabolites that influence the strain growth and butanol production, and find out the critical regulation nodes, an integrated analysis approach has been carried out in this study.Results: Based on the gas chromatography-mass spectrometry technology, the partial least square discriminant analysis and the pathway analysis, 40 metabolic pathways linked with 43 key metabolic nodes were identified. Indepth analysis showed that lots of amino acids metabolism promoted cell growth but exerted slight influence on butanol production, while sugar metabolism was favorable for cell growth but unfavorable for butanol synthesis. Besides, both lysine and succinic acid metabolism generated a complex effect on the whole metabolic network. Dicarboxylate metabolism exerted an indispensable role on cell growth and butanol production. Subsequently, rational feeding strategies were proposed to verify these conclusions and facilitate the butanol biosynthesis. Feeding amino acids, especially glycine and serine, could obviously improve cell growth while yeast extract, citric acid and ethylene glycol could significantly enhance both growth and butanol production.Conclusions: The feeding experiment confirmed that metabolic profiling combined with pathway analysis provided an accurate, reasonable and practical approach to explore the cellular metabolic activity and supplied a basis for improving butanol production. These strategies can also be extended for the production of other important biochemical compounds.

Abstract:Lignocellulose is a promising feedstock for butanol production owing to its abundance and renewability, however, most solventogenic clostridia could not directly convert these low-cost resources. Fermentable sugars could only be produced through pretreatment and hydrolysis processes, but furan and phenolic derivatives occurring in the lignocellulosic hydrolysate would inhibit cells' growth and final butanol production capability. Compared to reported solventogenic Clostridia, the newly isolated Clostridium sp. strain LJ4 possessed high tolerance towards furan derivatives. Genes of sdr4, sdr1, akr, fmn and ad2 in strain LJ4 may play key roles in the bioconversion of furan derivatives based on the transcriptional levels' analysis. As the phenolic derivatives containing in the undetoxified corncob hydrolysate were higher than the limited tolerance of strain LJ4, an electrochemical detoxification method was accordingly adopted to reduce the toxicity of phenolic derivatives without the loss of reducing sugars and generation of by-products. After 24 h detoxification of acid treated corncob hydrolysate, the phenolic compounds were decreased by nearly 40%, and 4.7 g/L of butanol was finally produced by strain LJ4 with 1.6-fold improvement than that from the un-detoxified corncob hydrolysate. Electrochemical technique could become a promising approach for biofuels or biochemicals production from undetoxified lignocellulosic hydrolysate owing to its high removal efficiency of phenolic derivatives without sugar loss.

Abstract:Saccharomyces cerevisiae has increasingly been engineered as a cell factory for efficient and economic production of fuels and chemicals from renewable resources. Notably, a wide variety of industrially important products are derived from the same precursor metabolite, acetyl-CoA. However, the limited supply of acetyl-CoA in the cytosol, where biosynthesis generally happens, often leads to low titer and yield of the desired products in yeast. In the present work, combined strategies of disrupting competing pathways and introducing heterologous biosynthetic pathways were carried out to increase acetyl-CoA levels by using the CoA-dependent n-butanol production as a reporter. By inactivating ADH1 and ADH4 for ethanol formation and GPD1 and GPD2 for glycerol production, the glycolytic flux was redirected towards acetyl-CoA, resulting in 4-fold improvement in n-butanol production. Subsequent introduction of heterologous acetyl-CoA biosynthetic pathways, including pyruvate dehydrogenase (PDH), ATP-dependent citrate lyase (ACL), and PDH-bypass, further increased n-butanol production. Recombinant PDHs localized in the cytosol (cytoPDHs) were found to be the most efficient, which increased n-butanol production by additional 3 fold. In total, n-butanol titer and acetyl-CoA concentration were increased more than 12 fold and 3 fold, respectively. By combining the most effective and complementary acetyl-CoA pathways, more than 100mg/L n-butanol could be produced using high cell density fermentation, which represents the highest titer ever reported in yeast using the clostridial CoA-dependent pathway.

Abstract:Current environmental challenges and energy crises highlight the urgent need for a transition in energy mix. In this study, an innovative semi-photosynthetic biohybrid system that combined light-activated cadmium selenide quantum dots (CdSe QDs) with engineered Gram-positive anaerobic bacteria, Clostridium acetobutylicumg (C. acetobutylicumg), was developed to enhance renewable butanol production. The results demonstrated that CdSe QDs could be biosynthesized intracellularly within C. acetobutylicumg through the introduction of glutathione pathway, without causing significant damage to bacteria. Furthermore, this system showed remarkable tolerance to butanol and weak acids. Under illumination, the biological synthesized CdSe QDs enabled C. acetobutylicumg to achieve a 45.5 % increase in NADH/NAD+ ratio compared to C. acetobutylicumg without CdSe QDs. When utilizing undetoxified rice straw hydrolysate in photo-fermentation, this system achieved a butanol titer of 14.82 g/L and a yield of 0.29 g/g. Overall, this work aims to effectively harness solar energy and biomass resources for sustainable clean biofuel production.
