Abstract:The objective of the present study was to improve the stability of (water-in-oil-in-water) double emulsions by dual gelation, including the gelation of lipid phase through oleogels and the gelation of continuous aqueous phase through hydrogels. Glycerol monostearate (GMS) could be used to form the oleogels and improve the stability of primary emulsions and double emulsions through reinforcing oil film. However, the redundant GMS crystals in both the oil phase and the continuous phase could cause the destabilization of double emulsions. The double emulsions reinforced by oleogels were successfully encapsulated in Ca-alginate hydrogel beads, and this encapsulation would not influence the integrity of double emulsions. The redundant GMS crystals might cause the leakage of the inner aqueous phase in the encapsulation process, and this leakage was more likely to happen during the mixing step, rather than the crosslinking step. Ca-alginate hydrogel beads were conducive to the confinement of the encapsulated oil globules.

Abstract:Formation of water-in-diesel oil (w/o) nano-emulsion has been achieved by a low-energy emulsification method by stabilizing a new combination of nonionic sorbitan esters surfactants, that is PEG20-sorbitan monostearate and sorbitan monooleate in mixed proportions. Different combinations of the surfactants (T6 + S8) have been tested and the best possible combination of mixed surfactants is found at a surfactants ratio of 35:65 (wt/wt) for T6:S8 at hydrophile–lipophile balance (HLB) = 8.01, which resulted in smaller droplet size of 44.87 nm. A phase diagram study is performed to identify the zones of formation of transparent, translucent, and opaque emulsions (44 nm < droplet size < 700 nm). The mechanism responsible for the instability of emulsion is explained by Ostwald ripening, with inference describing a decrease in particle size with the Ostwald ripening rate. In case of nano-emulsion of droplet size 64.28 nm, the Oswald ripening rate is found as 0.0874 × 10−27 m3 · s−1. Comparison of Ostwald ripening rate with other sets of surfactants obtained by different authors showed the lowest rate among them, indicative of enhanced stability. A rheological study of the tested set of nano-emulsions depicts the Newtonian behavior (1.0371 ≤ n ≤ 1.0826) over a wider range of shear rates (10–1000 s−1) at different temperatures (25–40°C).

Abstract:Nano-emulsions consist of fine oil-in-water dispersions, having droplets covering the size range of 100-600 nm. In the present work, nano-emulsions were prepared using the spontaneous emulsification mechanism which occurs when an organic phase and an aqueous phase are mixed. The organic phase is an homogeneous solution of oil, lipophilic surfactant and water-miscible solvent, the aqueous phase consists on hydrophilic surfactant and water. An experimental study of nano-emulsion process optimisation based on the required size distribution was performed in relation with the type of oil, surfactant and the water-miscible solvent. The results showed that the composition of the initial organic phase was of great importance for the spontaneous emulsification process, and so, for the physico-chemical properties of the obtained emulsions. First, oil viscosity and HLB surfactants were changed, alpha-tocopherol, the most viscous oil, gave the smallest droplets size (171 +/- 2 nm), HLB required for the resulting oil-in-water emulsion was superior to 8. Second, the effect of water-solvent miscibility on the emulsification process was studied by decreasing acetone proportion in the organic phase. The solvent-acetone proportion leading to a fine nano-emulsion was fixed at 15/85% (v/v) with EtAc-acetone and 30/70% (v/v) with MEK-acetone mixture. To strength the choice of solvents, physical characteristics were compared, in particular, the auto-inflammation temperature and the flash point. This phase of emulsion optimisation represents an important step in the process of polymeric nanocapsules preparation using nanoprecipitation or interfacial polycondensation combined with spontaneous emulsification technique.

Abstract:The use of the appropriate emulsifier is essential for forming a stable nanoemulsion delivery system that can maintain the sustained release of its contents. Health concerns have prompted the search for natural biopolymers to replace traditional synthetic substances as emulsifiers. In this study, an oregano essential oil (OEO) nanoemulsion-embedding system was created using soybean protein isolate (SPI), tea saponin (TS), and soy lecithin (SL) as natural emulsifiers and then compared to a system created using a synthetic emulsifier (Tween 80). The results showed that 4% Tween 80, 1% SPI, 2% TS, and 4% SL were the optimal conditions. Subsequently, the influence of emulsifier type on nanoemulsion stability was evaluated. The results revealed that among all the nanoemulsions, the TS nanoemulsion exhibited excellent centrifugal stability, storage stability, and oxidative stability and maintained high stability and encapsulation efficiency, even under relatively extreme environmental conditions. The good stability of the TS nanoemulsion may be due to the strong electrostatic repulsion generated by TS molecules, which contain hydroxyl groups, sapogenins, and saccharides in their structures. Overall, the natural emulsifiers used in our study can form homogeneous nanoemulsions, but their effectiveness and stability differ considerably.

Abstract:Micrometer-sized double emulsions and antibubbles were produced and stabilized via the Pickering mechanism by colloidal interfacial layers of polymeric nanoparticles (NPs). Two types of nanoparticles, consisting either of polylactic acid (PLA) or polylactic-co-glycolic acid (PLGA), were synthesized by the antisolvent technique without requiring any surfactant. PLA nanoparticles were able to stabilize water-in-oil (W/O) emulsions only after tuning the hydrophobicity by means of a thermal treatment. A water-in-oil-in-water (W/O/W) emulsion was realized by emulsifying the previous W/O emulsion in a continuous water phase containing hydrophilic PLGA nanoparticles. Both inner and outer water phases contained a sugar capable of forming a glassy phase, while the oil was crystallizable upon freezing. Freeze drying the double emulsion allowed removing the oil and water and replacing them with air without losing the three-dimensional (3D) structure of the original emulsion owing to the sugar glassy phase. Reconstitution of the freeze-dried double emulsion in water yielded a dispersion of antibubbles, i.e., micrometric bubbles containing aqueous droplets, with the interfaces of the antibubbles being stabilized by a layer of adsorbed polymeric nanoparticles. Remarkably, it was possible to achieve controlled release of a flourescent probe (calcein) from the antibubbles through heating to 37 °C leading to bursting of the antibubbles.

Abstract:Exploring the natural, safe, and effective antimicrobial is one of the preferable ways to control foodborne bacteria. In this work, novel oil-in-water nanoemulsions were formulated with sophorolipids and eugenol without any co-surfactant using a self-assembling strategy. These nanoemulsions showed high stability with sizes less than 200 nm when exposure to low concentrations of salt ions, various pH values (5.0, 7.0, 10.0), storage temperature and time. The synergistic antibacterial effects against both Gram-negative Escherichia coli and Gram-positive Bacillus cereus were determined with a minimum inhibitory concentration (MIC) value of 0.5 mg/mL and 0.125 mg/mL, respectively. Further microscopy (SEM, TEM, LCSM) examination and ATP/Na + -K + -ATPase assay results showed that the morphological changes, intensive cell membrane permeability, leakage of ATP, and decreased Na + -K + -ATPase contributed to the antibacterial effects. Moreover, the bonding mechanism between nanoemulsions and cell membranes were further evaluated by FTIR and ITC using a DPPC vesicle model, which demonstrated that the nanoemulsions adsorbed on the surface of bilayer, interacted with the hydrophobic chains of DPPC membrane mainly through the hydrophobic interaction, and altered the structural integrity of the lipid bilayer. These results not only provide a facile green strategy for fabricating stable nanoemulsions, but also highlight a new perspective for stabilizing essential oils for their widely application in food industry.

Abstract:Human organic anion transporting polypeptides (OATP) 1B1 and 1B3 are multispecific transporters that mediate uptake of amphipathic organic compounds into hepatocytes. The two OATPs contain 12 transmembrane domains (TMs) and share 80% amino acid sequence identity. Besides common substrates with OATP1B1, OATP1B3 specifically transports cholecystokinin octapeptide (CCK-8). To determine which structural domains and/or residues are important for the substrate selectivity of OATP1B3, we constructed a series of chimeric proteins between OATP1B3 and 1B1, expressed them in HEK293 cells, and determined rates of uptake of CCK-8 along with surface expression of the proteins. Replacing TM10 in OATP1B3 with TM10 of OATP1B1 resulted in a dramatically reduced degree of CCK-8 transport, indicating that TM10 is crucial for recognition and/or translocation of CCK-8. Using site-directed mutagenesis, we identified three key residues within TM10, namely, Y537, S545, and T550. When we replaced these residues with the corresponding amino acid residues found in OATP1B1, the level of CCK-8 transport was similarly low as for the replacement of the whole TM10. Kinetic experiments showed that the K m values for CCK-8 transport in the TM10 replacement and triple mutant were only 1.3 and 1.1 microM, respectively, as compared to 16.3 microM for wild-type OATP1B3. Similarly, the V max values dropped from 495.5 pmol (normalized mg) (-1) min (-1) for wild-type OATP1B3 to 13.3 and 19.0 pmol (normalized mg) (-1) min (-1) for the TM10 replacement and triple mutant, respectively. Molecular modeling indicated that two of the three identified residues might form hydrogen bonds with CCK-8. In conclusion, we have identified three amino acid residues (Y537, S545, and T550) in TM10 of OATP1B3 that are important for CCK-8 transport.

Abstract:This research aims to optimize the parameters for the production of a stable nanoemulsion containing a mixture of essential oils (EOs) with a surfactant blend (Tween and Span 80) and assess its antimicrobial potency against spoilage bacteria and foodborne pathogens. The impact of a surfactant blend's hydrophilic-lipophilic balance (HLB) as well as surfactant:oil ratio was investigated based on the mean particle size (z-average) and polydispersity index (PDI). Results showed HLB value of 12 and a ratio of surfactant to oil at 0.75:1 exhibited an emulsion with the smallest z-average and PDI. Following this, a response surface methodology (RSM) employing central composite design (CCD) was utilized to formulate an optimal nanoemulsion using the microfluidization (MF) technique. The RSM revealed that the microfluidizer pressure of 15,000 psi and 5 cycles produces a z-average of 38.11 nm, PDI of 0.27, ζ-potential of 37.8 mV, and EE of 83.8%. The comparison between the experimental and anticipated results of the optimized nanoemulsion revealed no significant differences (p > 0.05). The optimal nanoemulsion displayed excellent stability during 30 days of storage at both 4 and 30 °C compared to 5 days for coarse emulsion. The nanoemulsion exhibited potent antimicrobial activity, showing minimum inhibitory concentration (MIC) values in the range of 0.019 to 0.156 μL/mL, surpassing the antimicrobial efficacy of the coarse emulsion, which had MIC values between 0.039 to 0.312 μL/mL. Thus, this study emphasized the efficacy of RSM design in developing an optimal nanoemulsion with enhanced antimicrobial properties using EOs, making it a promising natural food preservative.

Abstract:Different types and ratios of surfactant, co-surfactant, and oil phase, have a greater impact on nanoemulsion preparation. The presence of surfactants in the nanoemulsion can reduce surface tension and characteristic stability. In this study, four groups of oil-in-water (O/W) nanoemulsions (NEs) with different ratios of surfactant and co-surfactant, and two oils were formulated as carriers of Rhodiola rosea. The variable optimization was investigated and then indicated as optimization group A (Opt A) with the formula of 10% of transcutol, 16.63% of tween 80, Opt B with 10% of tween 80, 29.87% of span 80, Opt C with 28.42% of transcutol, 30% of labrasol, and Opt D with 30% of transcutol, 30% of tween 80. Labrafac and soybean oil were used as the oil phase. The optimized formula using the response surface method (RSM) by design expert software showed the ideal conditions with a higher desirability score. Desirability score are 0.72% (Opt A), 0.81% (Opt B), 0.76% (Opt C) and 0.98% (Opt D), the desirability rating close to 1 indicates a high possibility that the projected values would closely match the experimental results for the optimum formula. All of the optimized formulation were also checked for the characteristics of nanoemulsion including particle size, polydispersity index (PDI), zeta potential, viscosity, encapsulation efficiency, transmission electron microscope (TEM), antioxidant activity, skin irritation test and stability studies. Our study provides a promising combination of surfactant-co-surfactant and oil phases to produce a stable nanoemulsion that can be used in pharmaceuticals and cosmetics in the future.

Abstract:The use of phospholipids (PL) as surfactants in micellar systems and microemulsions offers many advantages as drug delivery vehicles. PL are commonly used in combination with other non-food surfactants with cosolvents and cosurfactants to form a cascade of delivery structures. In this work, we incorporated phosphatidylcholine (PC) in our unique U-type water-dilutable phase diagrams exhibiting large isotropic regions of nanostructures. The nanometric liquid structures were prepared from food-grade emulsifiers. We formed water-free concentrates with PC that are fully diluted with water to form a variety of unusually structured nanodroplets. Due to the uniqueness of their composition, the designed concentrates derived from the nature of the oil phase, type of surfactants, and cosurfactants were characterized and found to be direct micelles (rather than inverse micelles), with the surfactant headgroups convexed toward the hydrophilic phase away from the micelle's core, even in the absence of water. The concentrates tend to self-assemble upon adding water to form O/W microemulsions even with small amounts of water (water-poor compositions of 0-20 wt% water). Upon further dilution with water the swollen micelles retain the oil as the inner phase. Multi-component compositions with two types of hydrophilic surfactants were investigated. The most significant enhancement in the total isotropic region was obtained by decreasing the triacetin and PC content in the system. We explored, by varying the nature of the headgroups and the nature and length of the lipidic (lipophilic) tails of the PL, the dilution capabilities of each of the systems.
Preparation of Nanoemulgels Containing Lemon Essential Oil and Pectin: Physical Stability and Rheological Properties

Abstract:Nanoemulgels are novel formulations of great interest for their use as dual-release systems and as fat substitutes in foods. Lemon essential oil, with a large number of benefits due to its antimicrobial, antifungal, and medicinal properties, and low methoxyl pectin, a natural polysaccharide capable of gelling by adding divalent ions such as calcium, are very appropriate ingredients to produce nanoemulgels with potential applications in industries such as cosmetics, agrochemistry, pharmaceuticals, or food. In this work, lemon-essential-oil-in-water nanoemulgels containing low methoxyl pectin derived from citrus peels were prepared following a three-step process that involves the preparation of a nanoemulsion, a pectin gel, and the mixture of both. In the first stage, the stirring time and the rotational rate employed during the mixing step were assessed. Once the preparation protocol was established, the pectin gel/nanoemulsion mass ratio was investigated. Different techniques were combined to evaluate the influence of the processing and the composition variables on the particle size distribution, mean diameters, flow curves, and physical stability of different emulgels obtained. It was found that the processing variables studied, stirring time, and rotational rate, do not influence the mean particle size of the emulgel, with values matching those of the starting nanoemulsion. However, 3 min and 200 rpm were selected for exhibiting the lowest TSI values. Regarding the composition, a higher content of pectin gel caused a higher viscosity, and therefore a higher physical stability, with the 75P/25E emulgel being the most stable. Aggregation of gel particles, because the pectin gel was really a sheared gel, was the main responsible contributor to the results obtained. This work highlights the importance of the preparation and formulation variables to develop stable, innovative formulations based on nanoemulgels.

Abstract:Hypothesis Phase separation of heterogeneous liquids can occur in the presence of sufficient shear force between a solid interface and a liquid. We hypothesize that a continuous flow of oil through nanopores into water in combination with a given ultrasound energy can generate stable free standing oil emulsions in water in the absence of surfactants. Experiment An oil solution was pumped through a membrane with 100 nm pores at a controlled flow rate and introduced into an aqueous solution while being subjected to sonication. The resulting emulsion samples were collected at specific time intervals and characterized by dynamic light scattering and nanoparticle tracking analysis techniques. We evaluated the storage stability of the emulsions with and without surfactant at room temperature or 4 °C storage. Results Introducing oil into an aqueous phase using our Nanopore system in conjunction with ultrasound gave rise to surfactant-free milky oil nanodroplet solutions. Dynamic light scattering and nanoparticle tracking analysis showed that the produced surfactant-free oil nanodroplets were monodispersed in water at sizes less than 200 nm. Control experiments without the use of nanopores did not result in phase mixing. Nanoemulsions were also generated in a surfactant containing solution resulting in a similar size range of nanodroplets. This work demonstrates that our Nanopore system can generate stable nanoemulsions in the absence of a surfactant, persisting in phase separation for several days up to two weeks at room temperature and 4 °C storage. This work suggests that nanopores are effective in producing surfactant-free nanoemulsions, which offer a wide range of valuable applications varying from drug delivery and food engineering, to paints and pesticide development.

Abstract:Previously water-in-octane nano-emulsions were prepared by mixed surfactant systems. In this paper, a series of amphiphilic graft copolymers, namely poly (higher α-olefin-co-para-methylstyrene)-graft-poly(ethylene glycol) and poly (higher α-olefin-co-acrylic acid)-graft-poly(ethylene glycol), were used as additives to enhance the stability of these nano-emulsions. Although the amphiphilic graft copolymers did not impact the average diameter of the water droplets in these emulsions to a major extent, the stability of these emulsions were enhanced significantly by the interaction between surfactants and amphiphilic graft copolymers. Amphiphilic graft copolymers’ stabilizing capability depends on their chain structure and composition. In a proper range of poly(ethylene glycol) content, e.g. 6.0–34.0wt.%, the higher the poly(ethylene glycol) content, the higher the stabilizing capability of these amphiphilic graft copolymers. However, when amphiphilic graft copolymers contained too much poly(ethylene glycol), e.g. 64.8wt.%, their stabilizing capability dropped because of insufficient amphiphilic character. Furthermore, when amphiphilic graft copolymers had the same content of poly(ethylene glycol), their stabilizing capability increased as the carbon number of higher α-olefin increased. The most stable water-in-octane nano-emulsion could be stored for 86days at 25°C. At elevated temperature, e.g. 40°C, this emulsion could be stored for 8days. Under the influence of a centrifugal force, this emulsion could still exist for 210min at the rotational speed of 15,000rpm.
Physical and Oxidative Water-in-Oil Emulsion Stability by the Addition of Liposomes from Shrimp Waste Oil with Antioxidant and Anti-Inflammatory Properties

Abstract:Liposomes made of partially purified phospholipids (PL) from Argentine red shrimp waste oil were loaded with two antioxidant lipid co-extracts (hexane-soluble, Hx and acetone-soluble, Ac) to provide a higher content of omega-3 fatty acids. The physical properties of the liposomes were characterized by Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS) and Differential Scanning Calorimetry (DSC). The antioxidant and anti-inflammatory activity of the lipid extracts and liposomal suspensions were evaluated in terms of Superoxide and ABTS radical scavenging capacities and TNF-α inhibition. Uni-lamellar spherical liposomes (z-average ≈ 145 nm) with strong negative ζ potential (≈ −67 mV) were obtained in all cases. The high content of neutral lipids in the Hx extract caused structural changes in the bilayer membrane and decreased entrapment efficiency regarding astaxanthin and EPA + DHA contents. The liposomes loaded with the Hx/Ac extracts showed higher antioxidant and anti-inflammatory activity compared with empty liposomes. The liposomal dispersions improved the physical and oxidative stability of water-in-oil emulsions as compared with the PL extract, inducing pronounced close packing of water droplets. The liposomes decreased hydroperoxide formation in freshly made emulsions and prevented thio-barbituric acid-reactive substances (TBARS) accumulation during chilled storage. Abstract:Nano-emulsions consist of stable suspensions of nano-scaled droplets that have huge loading capacities and are formulated with safe compounds. For these reasons, a large number of studies have described the potential uses of nano-emulsions, focusing on various aspects such as formulation processes, loading capabilities, and surface modifications. These studies typically concern direct nano-emulsions (i.e., oil-in-water), whereas studies on reverse nano-emulsions (i.e., water-in-oil) remain anecdotal. However, reverse nano-emulsion technology is very promising (e.g., as an alternative to liposome technology) for the development of drug delivery systems that encapsulate hydrophilic compounds within double droplets. The spontaneous emulsification process has the added advantages of optimization of the energetic yield, potential for industrial scale-up, improved loading capabilities, and preservation of fragile compounds targeted for encapsulation. In this study, we propose a detailed investigation of the processes and formulation parameters involved in the spontaneous nano-emulsification that produces water-in-oil nano-emulsions. The following details were addressed: (i) the order of mixing of the different compounds (method A and method B), (ii) mixing rates, (iii) amount of surfactants, (iv) type and mixture of surfactants, (v) amount of dispersed phase, and (vi) influence of the nature of the oil. The results emphasized the effects of the formulation parameters (e.g., the volume fraction of the dispersed phase, nature or concentration of surfactant, or nature of the oil) on the nature and properties of the nano-emulsions formed.

Abstract:Amongst the three fundamental components of a skin-care cosmetic emulsion, namely, oil, water, and emulsifiers (mainly surfactants), naturally derived alternatives to synthetic surfactants are yet to be utilized to their fullest potential, owing to their satisfactory properties like critical micelle concentration, hydrophilic-lipophilic balance, and critical packing parameter. Currently, there are no significant efforts towards green emulsions involving one or more biosurfactants, while significant combinations in varying proportions of their chemical contemporaries exist in the literature. The present study focuses on formulating a stable nanoemulsion system using a cocktail of a plant-derived surfactant ( Quillaja saponin) and a microbial-derived surfactant (surfactin), using coconut oil as a base oil. With a limited understanding of the behavior of mixed biosurfactant systems and their underlying interactions, most essentially the interactions between different hydrophilic moieties such as a cyclic peptide group of surfactin and oligosaccharides of Q . saponin, the primary focus of this study is to tune up the composition of biosurfactants that results in a stable nanoemulsion. A high-energy ultrasonication method is employed to formulate the emulsion, and hierarchical-based screening of various combinations has led us toward an optimal combination of composition and operating conditions. Six samples have been identified as better among the 20-sample set, and their stability has been tested for 45 days. However, based on the optimal energy consumption and oil-to-surfactant ratio, sample 6 of cluster 5 have been identified to be the best amongst the samples, with a mean droplet size of 173 nm, polydispersity index of 0.195 along with a zeta potential of –65 mV. Stability analyses involving heat, salt, pH, and centrifugation tests have proved the stability of the best-performing sample. This study can be a precursor to future formulation using mixed micellar systems.

Abstract:<p>To ensure emulsions to be continuously stable, the hydrolysates recovered from cod bones by papain acted as a natural surfactant to synthesize high-stability bilayer nano-emulsions. As assisted by Tween 20, the average diameter of synthesized nano-emulsion with enzymatic hydrolysate could exhibit stability between 300∼400 nm under a broad range of pH (4∼8), temperatures (30∼90 <sup>o</sup>C) and salt concentration (25∼250 mM). With the addition of the hydrolysates, the rate of increase of the TBARS value in the emulsion decreased. Moreover, the bilayer structure of the nano-emulsion was characterized under an atomic force microscopy and a cryo-scanning electron microscopy. Nano-LC-Q-TOF-MS was adopted to primarily identify peptides that contained hydrophobic and hydrophilic amino acids at the emulsion interface. Besides, the absorbed peptides on the interface of emulsion enhanced the stability of emulsion lipid oxidation.</p>
