Abstract:Recent oil spill responses such as the Deepwater Horizon event have underscored the need for crude oil ecotoxicological threshold data for shallow water corals to assist in natural resource damage assessments. We determined the toxicity of a mechanically agitated oil-seawater mixture (high-energy water-accommodated fraction, HEWAF) of a sweet crude oil on a branched stony coral, Pocillopora damicornis. We report the results of two experiments: a 96 h static renewal exposure experiment and a "pulse-chase" experiment of three short-term exposure durations followed by a recovery period in artificial seawater. Five endpoints were used to determine ecotoxicological values: 1) algal symbiont chlorophyll fluorescence, 2) a tissue regeneration assay and a visual health metric with three endpoints: 3) tissue integrity, 4) tissue color, and 5) polyp behavior. The sum of 50 entrained polycyclic aromatic hydrocarbons (tPAH50) was used as a proxy for oil exposure. For the 96 h exposure dose response experiment, dark-adapted maximum quantum yield (Fv/Fm) of the dinoflagellate symbionts was least affected by crude oil (EC50 = 913 μg/L tPAH50); light-adapted effective quantum yield (EQY) was more sensitive (EC50 = 428 μg/L tPAH50). In the health assessment, polyp behavior (EC50 = 27 μg/L tPAH50) was more sensitive than tissue integrity (EC50 = 806 μg/L tPAH50) or tissue color (EC50 = 926 μg/L tPAH50). Tissue regeneration proved to be a particularly sensitive measurement for toxicity effects (EC50 = 10 μg/L tPAH50). Short duration (6-24 h) exposures using 503 μg/L tPAH50 (average concentration) resulted in negative impacts to P. damicornis and its symbionts. Recovery of chlorophyll a fluorescence levels for 6-24 h oil exposures was observed in a few hours (Fv/Fm) to several days (EQY) following recovery in fresh seawater. The coral health assessments for tissue integrity and tissue color were not affected following short-term oil exposure durations, but the 96 h treatment duration resulted in significant decreases for both. A reduction in polyp behavior (extension) was observed for all treatment durations, with recovery observed for the short-term (6-24 h) exposures within 1-2 days following placement in fresh seawater. Wounded and intact fragments exposed to oil treatments were particularly sensitive, with significant delays observed in tissue regeneration. Estimating ecotoxicological values for P. damicornis exposed to crude oil HEWAFs provides a basis for natural resource damage assessments for oil spills in reef ecosystems. These data, when combined with ecotoxicological values for other coral reef species, will contribute to the development of species sensitivity models.
Metatranscriptomic response of deep ocean microbial populations to infusions of oil and/or synthetic chemical dispersant

Abstract:Oil spills are a frequent perturbation to the marine environment that has rapid and significant impacts on the local microbiome. Previous studies have shown that exposure to synthetic dispersant alone did not enhance heterotrophic microbial activity or oxidation rates of specific hydrocarbon components but increased the abundance of some taxa (e.g., Colwellia). In contrast, exposure to oil, but not dispersants, increased the abundance of other taxa (e.g., Marinobacter) and stimulated hydrocarbon oxidation rates. Here, we advance these findings by interpreting metatranscriptomic data from this experiment to explore how and why specific components of the microbial community responded to distinct organic carbon exposure regimes. Dispersant alone was selected for a unique community and for dominant organisms that reflected treatment- and time-dependent responses. Dispersant amendment also led to diverging functional profiles among the different treatments. Similarly, oil alone was selected for a community that was distinct from treatments amended with dispersants. The presence of oil and dispersants with added nutrients led to substantial differences in microbial responses, likely suggesting increased fitness driven by the presence of additional inorganic nutrients. The oil-only additions led to a marked increase in the expression of phages, prophages, transposable elements, and plasmids (PPTEPs), suggesting that aspects of microbial community response to oil are driven by the "mobilome," potentially through viral-associated regulation of metabolic pathways in ciliates and flagellates that would otherwise throttle the microbial community through grazing.IMPORTANCEMicrocosm experiments simulated the April 2010 Deepwater Horizon oil spill by applying oil and synthetic dispersants (Corexit EC9500A and EC9527A) to deep ocean water samples. The exposure regime revealed severe negative alterations in the treatments' heterotrophic microbial activity and hydrocarbon oxidation rates. We expanded these findings by exploring metatranscriptomic signatures of the microbial communities during the chemical amendments in the microcosm experiments. Here we report how dominant organisms were uniquely associated with treatment- and time-dependent trajectories during the exposure regimes; nutrient availability was a significant factor in driving changes in metatranscriptomic responses. Remarkable signals associated with PPTEPs showed the potential role of mobilome and viral-associated survival responses. These insights underscore the time-dependent environmental perturbations of fragile marine environments under oil and anthropogenic stress.
Effects of crude oil and gas condensate spill on coastal benthic microbial populations

Abstract:Marine sediments are ecologically-important environments that act as a long-term depository for different contaminants from natural and anthropogenic sources. We investigated the response of crude-oil and gas-condensate spills on benthic microbial populations in the oligotrophic southeastern Mediterranean Sea using costume-design benthocosms. Additions of crude oil and gas condensate significantly changed the sediment's chemical properties, with 2-fold elevated levels of total organic carbon (TOC) and up to ∼ 6-fold higher concentration of total polycyclic aromatic hydrocarbons (∑PAHs) relative to unamended sediments. Naphthalene and benzo(a)pyrene were the dominant species comprising the PAHs in both the crude-oil and gas-condensate treatments (29–43% and 26–35%, respectively). Porewater PO 4 3+ drastically declined throughout the experiment, whereas NO 2 − + NO 3 − decreased ∼100 days post hydrocarbons addition and then increased in the remaining ∼100 days till the conclusion of the experiment. This temporal variability in NO 2 − + NO 3 − hints that hydrocarbon pollution may affect the interplay between benthic denitrification and N 2 fixation, thus affecting nutrient limitation for benthic heterotrophic bacteria and phytoplankton. Moreover, our results show that crude oil and gas-condensate usually lead to a decline in benthic autotrophic microbial biomass (50–80%), while heterotrophic bacterial abundances remained unchanged, and bacterial production rapidly increased (maximal 1,600%, crude-oil > gas-cindensate). These effects were prolonged and lasted several months post hydrocarbons addition, highlighting the sediments as a repository for oil contaminants. Amplicon sequencing of the 16S rRNA gene revealed hydrocarbonoclastic bacteria including Methylophaga , Ponticaulis and Alcanivorax genera post crude-oil addition and Actinobacterota 67–14 lineage following gas-condensate amendments. Our results may enable applying a better science-based environmental policy for the benthic marine environment.

Abstract:The oil sands industry in Canada, produces heavy unconventional oils, diluted for transport and called diluted bitumen. However, despite advances in our knowledge of the ecotoxicological risk that these products represent, their effects on benthic organisms following a spill are still largely unknown. In order to fill these gaps, this study aims to determine the lethal and sublethal effects of two diluted bitumens (Bluesky and Cold Lake) and one conventional oil (Lloydminster) for two freshwater benthic invertebrates: Chironomus riparius and Hyalella azteca. The objective of this study is to assess the toxicity of dissolved hydrocarbons, resulting from the physical dispersion of oil, immediately after a spill on the benthic invertebrates. To this end, organisms were exposed for 7 days for chironomids and 14 days for amphipods to a fraction containing soluble hydrocarbons (WAF: water accommodated fraction; 10 g/L, 18 h of agitation, followed by 6 h of sedimentation) with natural or artificial sediment. After exposure, the effects of hydrocarbons were determined using size, mortality, and antioxidant capacities. Dissolved hydrocarbons induced mortality for both species, but these hydrocarbons disappeared very quickly from the water column, regardless of the oil type. The amphipods were sensitive to both types of oil while the chironomids were only sensitive to diluted bitumens. The presence of a natural sediment seems to provide a protective role against dissolved hydrocarbons. The antioxidant enzymes measured (CAT, SOD and GPx) do not appear to be relevant biomarkers for the exposure of these organisms to diluted bitumen.
Hydrocarbons occurrence and transcriptomic response of oyster Crassostrea virginica from lagoons of the Southern Gulf of Mexico

Abstract:The Gulf of Mexico is an important crude oil reserve worldwide, and the oyster Crassostrea virginica is an excellent candidate to study the oil spill impacts on marine invertebrates. In this work, the concentrations of polycyclic aromatic hydrocarbons (PAHs) and aliphatic hydrocarbons (AHs) from eight productive oyster areas in the Gulf of Mexico were measured on sediment, water, and tissues from C. virginica . In water, the highest AHs concentration was detected in Tamiahua (0.50 ng/mL), while for PAHs, the highest concentration was > 0.10 ng/mL in Tampamachoco. In sediment, Tamiahua and Tampamachoco lagoons had the highest AHs concentrations with values near 2.5 μg/g dry weight. Considering the PAHs, Tamiahua, Carmen, and Tampamachoco lagoons registered the highest levels, with values > 60 ng/g dry weight. In tissues from C. virginica , La Pesca, Cármen and Mecoacán presented the highest PAHs concentrations with values between 0.20 and 0.25 μg/g dry weight. Furthermore, from the molecular analysis of genes related with different phases of the xenobiotic detoxification process such as hypoxia inducible factor ( hif-1a ), cytochrome P450 10 ( cyp10 ), flavin mono-oxygenase ( fmo ), glutathione S-transferase ( gstΩ1 ), multidrug resistant protein ( mdrd1 ), catalase ( cat ), among others, the differences between lagoons were significant ( P < 0.05) and generally with Las Enramadas showing the highest expression levels. From genes evaluated in this study, hif-1a , cyp10 , fmo , mdrd1 and cat presented the highest expression differentials between lagoons. The above allowed us to validate the utility of molecular markers in the assessment of the hydrocarbons effect on oyster under the conditions from the Southern Gulf of Mexico.

Abstract:ABSTRACT As part of the American Petroleum Institute sponsored tidal area dispersant project involving two test spills of Murban crude oil in Long Cove, Searsport, Maine in August, 1981, water samples were collected. This paper deals with the analytical results for the analyses of water samples collected for analysis of non-volatile hydrocarbons by: infrared spectrophotometric quantitation of total CCl4 extractables, and gravimetric analysis of aliphatic and aromatic hydrocarbon fractions followed by capillary gas chromatography. In the dispersant-treated oil discharge area, there were two primary water sampling locations during the discharge phase of the experiment: an upper intertidal area (maximum depth = 2 meters) and a lower intertidal area (maximum depth = 3.5 meters). The gas chromatographic data for the water samples were treated numerically to obtain parameters whose values reflect the extent of dispersed oil weathering. For the aliphatics, the peak area ratio for n C14/n C18 was calculated for each sample. For the aromatics, the ratio for the peak area sum of the mono, di, and trimethyl naphthalenes to that for the mono, di, tri, and tetramethyl dibenzothiophenes was determined for each sample. At both sampling locations, dispersed oil in water sampled 10 cm off the bottom consistently had a smaller fraction of lower boiling aliphatic and aromatic hydrocarbons than water sampled at the same place and the same time 1⁄2 meter below the surface. In addition, the data show that there is a 12–50 fold decrease in hydrocarbon concentration on going from near surface to near bottom at any given time, even in water as shallow as 2 meters. The data indicate that the primary mechanism for hydrocarbon loss involves volatilization of hydrocarbon fractions. Analyses of water samples taken from submerged plumes of dispersed oil outside the sampling areas demonstrated slower loss of low boiling components consistent with the importance of atmospheric exchange in the weathering process. In the chemical dispersal of an oil spill, it may be most advantageous to use mixing methods that minimize vertical mixing in order to maintain a high concentration of emulsified oil in the upper 1⁄2 meter water layer. This will maximize the extent of loss of lower boiling hydrocarbon components into the atmosphere and thus minimize the toxicity of any dispersed oil fractions that diffuse downward and interact with benthic communities.

Abstract:Environmental control in the Arctic ecosystems needs to be changed to hi-tech monitoring technologies with IT systems for actual and ecologically effective 'arctic safeguard'. In circumstances of the fragile arctic ecosystems, priority bioassay of possible toxic hazards in natural seawater must be undertaken. It was considered useful to examine toxic responses of some indicator species. Bivalves are well-known indicators of pollution stress arising from the activities of man, such as off-shore drilling and oil transportation, dredging or the release of pollutants. Toxic effects of some drilling fluids on energy balance, behavior and survival of the common mussel Mytilus edulis L, were estimated in long-duration bioassays of about 30 days each. The highest sensitivity of behavioral responses to any changes in chemical water composition was found. Mussels of the Barents Sea had significant differences from the control by four behavioral parameters even at 0.01 g/l of the ferrochrome-lignosulfonate drilling fluid, while the significant differences from the control in oxygen consumption and filtration rates were observed only at the 10 g/l. Even small concentration of another standard water-based drilling fluid with barite (of about 0.05 g/l) had a lethal effect on M. edulis.
Clarifying the murk: unveiling bacterial dynamics in response to crude oil pollution, Corexit-dispersant, and natural sunlight in the Gulf of Mexico

Abstract:The 2010 Deepwater Horizon (DwH) Oil spill released an enormous volume of oil into the Gulf of Mexico (GoM), prompting the widespread use of chemical dispersants like Corexit ® EC9500A. The ecological consequences of this treatment, especially when combined with natural factors such as sunlight, remain unexplored in the context of marine bacterial communities' dynamics. To address this knowledge gap, our study employed a unique metaproteomic approach, investigating the combined effects of sunlight, crude Macondo surrogate oil, and Corexit on GoM microbiome across different mesocosms. Exposure to oil and/or Corexit caused a marked change in community composition, with a decrease in taxonomic diversity relative to controls in only 24 hours. Hydrocarbon (HC) degraders, particularly those more tolerant to Corexit and phototoxic properties of crude oil and/or Corexit, proliferated at the expense of more sensitive taxa. Solar radiation exacerbated these effects in most taxa. We demonstrated that sunlight increased the dispersant's toxicity, impacting on community structure and functioning. These functional changes were primarily directed by oxidative stress with upregulated proteins and enzymes involved in protein turnover, general stress response, DNA replication and repair, chromosome condensation, and cell division. These factors were more abundant in chemically treated conditions, especially in the presence of Corexit compared to controls. Oil treatment significantly enhanced the relative abundance of Alteromonas , an oil-degrading Gammaproteobacteria . In combined oil-Corexit treatments, the majority of identified protein functions were assigned to Alteromonas , with strongly expressed proteins involved in membrane transport, motility, carbon and amino acid metabolism and cellular defense mechanisms. Marinomonas , one of the most active genera in dark conditions, was absent from the light treatment. Numerous metabolic pathways and HC-degrading genes provided insights into bacterial community adaptation to oil spills. Key enzymes of the glyoxylate bypass, enriched in contaminant-containing treatments, were predominantly associated with Rhodobacterales and Alteromonadales . Several proteins related to outer membrane transport, photosynthesis, and nutrient metabolisms were characterized, allowing predictions of the various treatments on biogeochemical cycles. The study also presents novel perspectives for future oil spill clean-up processes.

Abstract:This work examined effects of model oil dispersants on dispersion, sorption and photodegradation of petroleum hydrocarbons in simulated marine systems. Three dispersants (Corexit 9500A, Corexit 9527A and SPC 1000) were used to prepare dispersed water accommodated oil (DWAO). While higher doses of dispersants dispersed more n-alkanes and PAHs, Corexit 9500A preferentially dispersed C11-C20 n-alkanes, whereas Corexit 9527A was more favorable for smaller alkanes (C10-C16), and SPC 1000 for C12-C28 n-alkanes. Sorption of petroleum hydrocarbons on sediment was proportional to TPH types/fractions in the DWAOs. Addition of 18mg/L of Corexit 9500A increased sediment uptake of 2-3 ring PAHs, while higher dispersant doses reduced the uptake, due to micelle-enhanced solubilization effects. Both dispersed n-alkanes and PAHs were susceptible to photodegradation under simulated sunlight. For PAHs, both photodegradation and photo-facilitated alkylation were concurrently taking place. The information can facilitate sounder assessment of fate and distribution of dispersed oil hydrocarbons in marine systems.
Sublethal effects of microplastic and oil co-exposure on biological rates and lipid profiles of keystone Arctic copepods

Abstract:Microplastics (MPs) and petroleum hydrocarbons are contaminants of emerging concern in the Arctic, but little is known about their co-exposure effects. In this study, we present the first assessment of the sublethal impacts resulting from combined exposure to microplastics and oil in three key Arctic copepod species. Specifically, we investigated the effects of a 5-day exposure to oil alone (1 μL L −1 ) and in combination with MPs (polyethylene microspheres, 20 μm, 20 MP mL −1 ) and dispersant (Corexit 9500, 0.05 μL L −1 ) on the biological functions and lipid profiles of the planktonic copepods Metridia longa , Calanus finmarchicus , and Calanus glacialis . Exposure to oil alone caused a significant reduction (34–58%) in fecal pellet production, but neither microplastics nor dispersant increased the negative effect of oil on fecal pellet production. C. glacialis and C. finmarchicus exposed to the studied pollutants for 5 days produced eggs with delayed hatching and lower hatching success. The highest hatching inhibition (50%) was observed in eggs of C. glacialis exposed to oil plus MPs and dispersant for five days. This indicates that maternal transfer of oil components to eggs negatively affects embryonic development and hatching. Lipid content and fatty acids profiles varied among the studied copepod species but were not affected by the tested pollutants after five days of exposure. By microscopical observation of fecal pellets, ingestion of small oil droplets and MPs was confirmed in all species, but the estimated ingestion of MPs was low (<25 MPs cop −1 d −1 , <0.2% of total offered MPs) suggesting avoidance of MP consumption in copepods. Our results indicate that virgin MPs did not increase the toxicity of oil to the studied Arctic copepods under co-exposure conditions, and dispersants can slightly increase certain adverse effects of oil (hatching). However, environmentally relevant concentrations of oil alone can negatively impact Arctic keystone copepods and potentially the biological carbon pump. These findings emphasize the need to reduce petrogenic pollution and the risk of oil spills in the sensitive Arctic ecosystem.

Comparative Toxicity of Oil Spill Herding Agents to Aquatic Species

Abstract:Chemical herding agents are surfactant mixtures used to coalesce spilled oil and increase slick thickness to facilitate mechanical recovery or in situ burning. Only two herders are currently listed on the United States' National Oil and Hazardous Substances Pollution Contingency Plan or National Contingency Plan product schedule for potential use in spill response: the surface collecting agents Siltech OP-40™ and ThickSlick 6535™. Toxicity data for spill response agents are frequently available only for two estuarine species, mysid shrimp (Americamysis bahia) and inland silversides (Menidia beryllina), and are particularly limited for herding agents. Toxicity can vary over several orders of magnitude across product type and species, even within specific categories of spill response agents. Seven aquatic species were tested with both Siltech OP-40™ and ThickSlick 6535™ to evaluate acute herder toxicity and relative species sensitivity. The toxicity assessment included: acute tests with A. bahia and M. beryllina, the freshwater crustacean Ceriodaphina dubia, and the freshwater fish Pimephales promelas; development of the echinoderm Arbacia unctulate; and growth of a freshwater alga Raphidocelis subcapitata and marine alga Dunaliella tertiolecta. Siltech acute toxicity values ranged from 1.1 to 32.8 ppm. ThickSlick acute toxicity values ranged from 2.2 to 126.4 ppm. The results of present study show greater toxicity of Siltech compared to ThickSlick with estimated acute hazard concentrations intended to provide 95% species protection of 1.1 and 3.6 ppm, respectively, on empirical data and 0.64 and 3.3 ppm, respectively, with the addition of interspecies correlation data. Abstract:Petroleum hydrocarbons can be discharged into the marine environment during offshore oil and gas production or as a result of oil spills, with potential impacts on marine organisms. Ecotoxcicological assay durations (typically 24 to 96 h) used to characterise risks to exposed organisms may not always reflect realistic environmental exposure durations in a high energy offshore environment where hydrocarbons are mixed and diluted rapidly in the water column. To investigate this, we adapted three sensitive toxicity tests to incorporate a short-term pulse exposure to three petroleum-based products: a produced water (PW), the water-accommodated fraction (WAF) of a condensate, and a crude oil WAF. We measured 48-h survival of the copepod Acartia sinjiensis, 72-h larval development of the sea urchin Heliocidaris tuberculata, and 48-h embryo survival and deformities of yellowtail kingfish Seriola lalandi, after exposure to a dilution series of each of the three products for 2 h, 4−12 h, 24 h and for the standard duration of each toxicity test (continuous exposure). Effects on copepod survival and sea urchin larval development were significantly reduced in short-term exposures to PW and WAFs compared to continuous exposures. Fish embryos, however, showed an increased frequency of deformities at elevated concentrations regardless of exposure duration, although there was a trend towards increased severity of deformities with continuous exposure. The results demonstrate how exposure duration alters toxic response and how incorporating relevant exposure duration to contaminants into toxicity testing may aid interpretation of more realistic effects (and hence an additional line of evidence in risk assessment) in the receiving environment.This article is protected by copyright. Concentrations in human blood of petroleum hydrocarbons associated with the BP/Deepwater Horizon oil spill, Gulf of Mexico

Abstract:During/after the BP/Deepwater Horizon oil spill, cleanup workers, fisherpersons, SCUBA divers, and coastal residents were exposed to crude oil and dispersants. These people experienced acute physiological and behavioral symptoms and consulted a physician. They were diagnosed with petroleum hydrocarbon poisoning and had blood analyses analyzed for volatile organic compounds; samples were drawn 5-19 months after the spill had been capped. We examined the petroleum hydrocarbon concentrations in the blood. The aromatic compounds m,p-xylene, toluene, ethylbenzene, benzene, o-xylene, and styrene, and the alkanes hexane, 3-methylpentane, 2-methylpentane, and iso-octane were detected. Concentrations of the first four aromatics were not significantly different from US National Health and Nutritional Examination Survey/US National Institute of Standards and Technology 95th percentiles, indicating high concentrations of contaminants. The other two aromatics and the alkanes yielded equivocal results or significantly low concentrations. The data suggest that single-ring aromatic compounds are more persistent in the blood than alkanes and may be responsible for the observed symptoms. People should avoid exposure to crude oil through avoidance of the affected region, or utilizing hazardous materials suits if involved in cleanup, or wearing hazardous waste operations and emergency response suits if SCUBA diving. Concentrations of alkanes and PAHs in the blood of coastal residents and workers should be monitored through time well after the spill has been controlled.
Effect of oil spill on compound-specific stable carbon isotope composition of amino acid in Nitzschia closterium and Heterosigma akashiwo

Abstract:Marine microalgae, the most important primary producers in marine ecosystems, are susceptible to toxicants and usually selected as the promising organisms for chemical risk testing. The stable carbon isotope ratios (δ13C) of amino acids (AAs) were measured to determine if compound-specific stable isotope analysis could be used to provide insight into the toxic effects of oil spill on the marine microalgae. The water accommodated fractions (WAF) of #180 fuel oil were selected as the toxic targets with different concentrations in acute (96 h) toxic tests. Naphthalene, phenanthrene, anthracene, and fluoranthene were detected as the predominant congeners of polycyclic aromatic hydrocarbons in the WAF, and may be the main toxic components. At the same WAF concentration, the δ13C values of leucine in Nitzschia closterium increased with the culture time, whereas decreased in Heterosigma akashiwo. However, with the increasing WAF concentrations, the δ13C values of glutamic acid exhibited the increasing trend in both of the two kinds of microalgae at the same culture time. The similar phenomenon was also observed for histidine in H. akashiwo, while opposite in N. closterium, but both enriching in δ13C compared to the controlled experiments without the WAF. Under the different culture times and WAF concentrations, the δ13C values of alanine and methionine showed unexpected trend. These findings indicated that the synthesis pathways of AA in microalgae cells were affected by the WAF, thus leading to the carbon isotopic fractionation in AAs. The present study provide a novel approach to explore the toxicity mechanism of primary producers under oil spill stress, and monitor and evaluate the marine ecological risk of oil spill pollution.
Non-target analysis of photochemically produced hydrocarbon oxidation products of crude oil at high latitudes and their biological effects

Abstract:With new oil and gas lease sales in high-latitude regions, there exists a need to better understand the chemical fate of spilled oil and its effects on biological life. To address this need, laboratory simulations of crude oil spills under sub-Arctic conditions were conducted using artificial seawater and exposure to solar irradiation to create Hydrocarbon Oxidation Products (HOPs). HOPs characterization and their biological effects were assessed using ultra high-performance liquid chromatography (UHPLC) with high resolution mass Orbitrap spectrometry and the aryl hydrocarbon receptor (AhR) chemically activated luciferase gene expression (CALUX) assay. Non-target UHPLC-Orbitrap mass spectrometry analysis identified 251 HOPs that were in greater abundance in light-exposed samples than dark controls. Oxidized polycyclic aromatic hydrocarbons were also detected, including phenanthrene quinone, anthraquinone, hydroxyanthraquinone, and 9-fluoreneone. The composition of HOPs were consistent with photo-products of alkylated two to four ring PAHs, primarily compounds between 1 and 3 aromatic rings and 1–3 oxygens. The HOP mixture formed during photochemical weathering of Cook Inlet crude oil induced greater AhR activity than parent petroleum products solubilized in dark controls, indicating that HOPs, as a complex mixture, may contribute to petroleum toxicity more than the parent petroleum compounds. These non-targeted approaches provide the most comprehensive analysis of hydrocarbon oxidation products to date, highlighting the diversity of the complex mixture resulting from the photooxidation of crude oil and the limitations of targeted analyses for adequately monitoring HOPs in the environment. Taken together, these data identify a critical "blind spot" in environmental monitoring and spill clean-up strategies as there is a diverse pool of HOPs that may negatively impact human and ecosystem health.

Abstract:During the Deepwater Horizon (DWH) oil spill, massive quantities of oil were deposited on the seafloor via a large-scale marine oil-snow sedimentation and flocculent accumulation (MOSSFA) event. The role of chemical dispersants (e.g., Corexit) applied during the DWH oil spill clean-up in helping or hindering the formation of this MOSSFA event are not well-understood. Here, we present the first experiment related to the DWH oil spill to specifically investigate the relationship between microbial community structure, oil and Corexit®, and marine oil-snow in coastal surface waters. We observed the formation of micron-scale aggregates of microbial cells around droplets of oil and dispersant and found that their rate of formation was directly related to the concentration of oil within the water column. These micro-aggregates are potentially important precursors to the formation of larger marine oil-snow particles. Therefore, our observation that Corexit® significantly enhanced their formation suggests dispersant application may play a role in the development of MOSSFA events. We also observed that microbial communities in marine surface waters respond to oil and oil plus Corexit® differently and much more rapidly than previously measured, with major shifts in community composition occurring within only a few hours of experiment initiation. In the oil-amended treatments without Corexit®, this manifested as an increase in community diversity due to the outgrowth of several putative aliphatic- and aromatic-hydrocarbon degrading genera, including phytoplankton-associated taxa. In contrast, microbial community diversity was reduced in mesocosms containing chemically dispersed oil. Importantly, different consortia of hydrocarbon degrading bacteria responded to oil and chemically dispersed oil, indicating that functional redundancy in the pre-spill community likely results in hydrocarbon consumption in both undispersed and dispersed oils, but by different bacterial taxa. Taken together, these data improve our understanding of how dispersants influence the degradation and transport of oil in marine surface waters following an oil spill and provide valuable insight into the early response of complex microbial communities to oil exposure.

Abstract:Abstract Background Biosurfactants are naturally derived products that play a similar role to synthetic dispersants in oil spill response but are easily biodegradable and less toxic. Using a combination of analytical chemistry, 16S rRNA amplicon sequencing and simulation-based approaches, this study investigated the microbial community dynamics, ecological drivers, functional diversity and robustness, and oil biodegradation potential of a northeast Atlantic marine microbial community to crude oil when exposed to rhamnolipid or synthetic dispersant Finasol OSR52. Results Psychrophilic Colwellia and Oleispira dominated the community in both the rhamnolipid and Finasol OSR52 treatments initially but later community structure across treatments diverged significantly: Rhodobacteraceae and Vibrio dominated the Finasol-amended treatment, whereas Colwellia , Oleispira , and later Cycloclasticus and Alcanivorax , dominated the rhamnolipid-amended treatment. Key aromatic hydrocarbon-degrading bacteria, like Cycloclasticus, was not observed in the Finasol treatment but it was abundant in the oil-only and rhamnolipid-amended treatments. Overall, Finasol had a significant negative impact on the community diversity, weakened the taxa-functional robustness of the community, and caused a stronger environmental filtering, more so than oil-only and rhamnolipid-amended oil treatments. Rhamnolipid-amended and oil-only treatments had the highest functional diversity, however, the overall oil biodegradation was greater in the Finasol treatment, but aromatic biodegradation was highest in the rhamnolipid treatment. Conclusion Overall, the natural marine microbial community in the northeast Atlantic responded differently to crude oil dispersed with either synthetic or biogenic surfactants over time, but oil degradation was more enhanced by the synthetic dispersant. Collectively, our results advance the understanding of how rhamnolipid biosurfactants and synthetic dispersant Finasol affect the natural marine microbial community in the FSC, supporting their potential application in oil spills.

Abstract:The 2010 Deepwater Horizon (DwH) oil spill contaminated ~1,773km of the Gulf of Mexico shorelines. Yet, few field data are available on the long-term fate and persistency of sediment-retained oil. While an unprecedented amount of oil dispersants was applied, the effects of oil dispersants on desorption of field aged oil remain unknown. This study aimed to investigate the abundance, distributions and physico-chemical availability of the oil retained in Bay Jimmy sediment, Louisiana, five years after the DwH oil spill, and to determine the effects of two model oil dispersants on the desorption potential of the residual oil. Total petroleum hydrocarbons (TPHs), n-alkanes and polycyclic aromatic hydrocarbons (PAHs) in the sediment were analyzed and compared with those in the crude oil and the pre-DwH levels, and batch desorption kinetic tests were carried out to quantify the dispersant effects on the desorption rate and extent. The biomarker hopanes profile and diagnostic ratio were analyzed, which confirmed the origin and persistence of the sediment-retained oil. After five-year natural weathering, the oil level in the sediment remained orders of magnitude higher than the pre-spill level. Nearly all low-molecular-weight n-alkanes and 2-ring PAHs had been degraded. Oil dispersants, SPC 1000 and Corexit EC9500A, were able to enhance solubilization of the sediment-retained oil upon resuspension of the sediment. Successive desorption experiments indicated that 71.6% of TPHs, 74.8% of n-alkanes, and 91.9% of PAHs in the sediment remained highly stable and hardly desorbable by seawater; yet, addition of 18mg/L of SPC 1000 enhanced the desorption and lowered these fractions to 57.3%, 68.1%, and 81.4%, respectively. The findings are important for understanding the natural weathering rate and persistence of oil residual and the effects of dispersants on the physical and biological availabilities of aged oil in coastal sediments.

Abstract:Oil production in Arctic ice-covered areas poses a risk for pollution of the ecosystem including that within the brine channel network of sea ice. Sea-ice autotrophs contribute substantially to Arctic primary production, but are inherently difficult to test for oil exposure responses in situ. This study had two objectives, first, we developed a suitable lab-based mesocosm system, second, we tested oil effects on sea-ice algae. Specifically, we investigated if Alaska North Slope crude oil exposure reduces ice algal abundance, biomass and concentration of extracellular polymeric substances (EPS) using indoor ice tanks over a 10-day exposure period. Six tanks in one cold room were used in pairs for the following treatments: (1) control, (2) oil release as a layer under ice and (3) release of dispersed oil. All tanks were inoculated with sea-ice microbial communities collected from Utqiaġvik, Alaska. After 10 days of exposure, the abundance of algae, dominated by the pennate diatom genus Nitzschia, and the concentrations of EPS and chlorophyll a were significantly lower in the oiled treatments compared to the control. We suggest light attenuation by the oil, reduced algal mobility, and oil toxicity as causes for this reduction. Observed changes in cell fluorescence characteristics based on DNA staining could be linked to the oil exposure and could provide a new tool for assessment of toxicity in microalgae.

Abstract:Determining the sensitivity of a diversity of species to spilled oil and chemically dispersed oil continues to be a significant challenge in spill response and impact assessment. We used standardized tests from the literature to develop species sensitivity distributions (SSDs) of acute aquatic toxicity values for several petroleum products and 2 Corexit oil dispersants. Fifth percentile hazard concentrations (HC5s) were computed from the SSDs and used to assess relative oil product toxicity and in evaluating the feasibility of establishing toxicity benchmarks for a community of species. The sensitivity of mysids (Americamysis bahia) and silversides (Menidia beryllina) were evaluated within the SSDs to determine if these common test species were appropriate surrogates for a broader range of species. In general, SSD development was limited by the availability of acute toxicity values that met standardization criteria for a diversity of species. Pooled SSDs were also developed for crude oil and Corexit dispersants because there was only small variability in the HC5s among the individual oil or dispersant products. The sensitivity of mysids and silversides varied across the oil and dispersant products, with the majority of toxicity values greater than the HC5. Application of SSDs appears to be a reasonable approach to developing oil product toxicity benchmarks, but additional toxicity data are needed for a larger range of species conducted under standardized test conditions.
