Abstract:The indica rice(Oryza sativa L.) 9311 was treated by 350 Gy 60Co gamma irradiation to induce agronomical and developmental mutants for genetic and functional genomics researches. The M1 plants was transplanted and harvested by single plant. A total of 5 000 families of M2 generation were grown, the diverse morphology mutations were occurred on 4 136 single plants in 1 665 families, which were preliminary screened and identified according to their morphological performance during different growing stages, and 2 001 morphological mutations was screened out. Then, the mutants were planted and confirmed again in M3 generations, a total of 1 996 mutants with defects occurring in leaf, stalk, panicle, fertility and mature time were identified. The calculated mutation frequency of morphological character was 33.3% according to the percentage of mutational families in total families of M2 generations,which indicated that 9311 is sensitive to gamma irradiation. The results also indicated that both single gene mutation and multi-gene mutations were occurred simultaneously due to the gamma irradiation in the present experiment. Furthermore, some mutants with good agronomic traits were identified, which can be used directly in the current rice breeding programs.
In vitro selection of gamma irradiated shoots of ginger (Zingiber officinale Rosc.) against Fusarium oxysporum f.sp. zingiberi and molecular analysis of the resistant plants

Abstract:In the present investigation, an attempt was made to develop Fusarium yellows resistant plants of ginger (Zingiber officinale Rosc.) var. Himgiri using in vitro mutagenesis and selection technique. Axenic in vitro cultures were subjected to gamma irradiation (10–100 Gy) for mutation induction. LD50 was calculated after every 4 weeks, and was observed to be 15 Gy after 16 weeks of irradiation. Surviving 10 Gy irradiated shoots were cultured on selective medium containing different concentrations (0–20%) of fungal culture filtrate (FCF) obtained from Fusarium oxysporum f.sp. zingiberi for in vitro selection. FCF concentration of 17.5% in the selective medium was found to be the highest on which 5% shoots survived after first selection cycle, which further reduced to 1.1% after third continuous cycle of selection. It was followed by 4.3% shoot survival after third selection cycle on 15% FCF concentration. Surviving, selected shoots from 15 and 17.5% FCF were multiplied on multiplication medium and rooted plantlets were hardened with 100% survival. On in vivo evaluation 46.4 and 52% plants selected at 15 and 17.5% FCF were found to be highly resistant. Molecular analysis with disease specific SSR primers differentiated between FCF selected, tissue culture propagated and gamma irradiated plants. Two unique bands were obtained in 15 and 17.5% FCF selected plantlets with GIN 6 and GIN 9 primers, sequencing, of which showed 98 and 97% homology with disease resistance protein-like gene CC-NBS-LRR from clones ZwP627 and ZoP620 of Z. officinale. After in vivo bioassay, SSR analysis of selected highly resistant plants again confirmed the unique bands with both the primers. The DNA sequences obtained from these primers have been published in GenBank under accession number MN497252 and MN497253.
EFFECT OF ~(60)Co γ-rays IRRADIATION ON GROWTH AND ANTIOXIDANT ENZYME ACTIVITIES OF RHIZOMES IN Cymbidium goeringii

Abstract:The rhizomes of Cymbidium goeringii were cut into tip,middle and basal segments with a length of about 1 cm,and the segments were irradiated with()~(60)Co γ-rays at doses of 0,5,10,20,30 and 50 Gy,respectively. The growth characters,the activities of antioxidant enzymes,including superoxide dismutase(SOD),catalase(CAT) and guaiacol peroxidase(G-POD),and peroxidase isozyme of the were measured.The results showed that the segments irradiated at the dose of 30 and 50 Gy could survive,but couldn't form new rhizomes and the growth was almost stagnated.With the increase of the doses,the SOD and CAT activities of the rhizome were increased first,then significantly decreased,but the reverse effect was observed in G-POD activities,which was decreased first,then increased.Combining both the effect results on the growth characters,on activity changes of peroxidase isozyme and the changes of the anti-oxidant enzymes after irradiation,it was concluded that the appropriate doses of()~(60)Co γ-rays irradiation for mutation breeding of Cymbidium goeringii should be in the rage of 20 to 30 Gy,and the optimal part of the rhizome seems to be the tip segment.

Abstract:We want to thank this group of scholars and practitioners1 for their comments on our recent paper,2 and thank the editor for giving us the opportunity to respond to those comments. We completely agree with the points being made in the letter. We agree that gathering perspectives only from health professionals is inherently limited. The paper we published in CMEJ 6(1) drew on data from a larger qualitative study, which – as we noted in the paper – included interviews with nurses and with LGBTQ patients. Most good qualitative research produces far more data than can be analyzed in a single paper. Accordingly, we have published several other papers from the study. Some of those focus on the experiences of nurses and physicians,3,4 some on the experiences of the LGBTQ participants,5–8 and some on both.9 Some specifically teased out the experiences of and with transgender participants for group specific analyses. We also completely agree that physicians (and other health professionals) need training regarding working effectively with LGBTQ populations, not only in entry-to-practice programs, but also through continuing education for practitioners. We want to emphasize that at either level training in communication skills and in patient-centered care – while invaluable – is not sufficient. Skillfully treating each patient as an individual may still ignore social group differences that continue to have predictable (though not inevitable) effects on health, health care and everyday experiences. Developing the skills to recognize and explore important social differences, as well as to recognize the biases, assumptions and limitations connected to one’s own social and cultural background are essential first steps toward more equitable health care.

Abstract:The effects of()~(60)Co γ-rays irradiation on germination rate of corms of saffron(Crocus sativus L.)were studied and the results were as follows:(1)The corm germination rate raised with the corm fresh weight M(g) increasing,and the corms of 3<M≤5 were suitable for mutation breeding,the single sprout per corm was over 96%;(2)The corms of 3<M≤4 and 4<M≤5 were irradiated by 5～35 Gy()~(60)Co γ-rays,the typical radiation effect of dose on germination rate were obtained,and the semi-lethal dose(D_(50)) for the corms of 3<M≤4 and 4<M≤5 were 16 and 20 Gy,respectively,indicating that the corms of 3<M≤4 was more sensitive than that of 4<M≤5 ones;(3)A higher temperature-tolerance mutant named ZF893 were obtained from the offspring of 4<M≤5 corms which were irradiated by 25 Gy()~(60)Co γ-rays and the stigma productivity of ZF893 was 1.3 times as that of the parent,for the field growing period,total weight of daughter-corms and flower numbers of ZF893 were 22%,27% and 30% higher than that of the parent,respectively.(4)The soluble protein SDS-PAGE patterns between ZF893 and it's parent were very similar,but the 54.8 kD bands were much stronger in ZF893 and the 20.9 kD bands which were clear in ZF893,but were nearly absent in it's parent.
Gamma irradiation-induced variability in morpho-agronomic and oil quality traits of Mentha piperita L

Abstract:Purpose: To develop elite mutant in Mentha piperita having morphotype and oil content of Mentha arvensis, and oil quality of M. piperita. Materials and methods: The dormant runner of menthol rich genotype MPK-5 were subjected to different doses of γ- irradiation (10, 20, 30, 50, 70, 90, and 110 Gy) at a dose rate of 55 Gy/min to induce the genetic variability for herb and oil yield as well as oil quality. A wide spectrum of variability for agro-morphological traits, herb yield, and quality profile was observed among the mutants. Results: The developed and selected superior mutants viz. MPK-5(1) and MPK-5(3) have a mean herb yield potential of 23.923 and 21.503 Kg/9 m2; mean oil yield of 92.953 and 80.047 ml/plot; mean menthol content of 69.012% and 69.160% with mean menthofuran content of 1.554% and 0.531%, respectively. Conclusions: Mutational breeding through γ- irradiation is considered complementary to the conventional breeding method, to broaden the spectrum of genetic variability. The developed and selected mutants namely viz. MPK-5(1) and MPK-5(3) identified as promising mutants, based on herb yield, oil yield, and essential oil quality, could be used as a parental line for exploitation in hybridization program/recombinant breeding.
Biological effects of gamma-ray radiation on tulip ( Tulipa gesneriana L.)

Abstract:Tulip, being an important ornamental plant, generally requires lengthy and laborious procedures to develop new varieties using traditional breeding methods requires. But ionizing radiation potentially accelerates the breeding process of ornamental plant species. The biological effects of γ-ray irradiation on tulip, therefore, were investigated through establishing an irradiation-mediated mutation breeding protocol to accelerate its breeding process. ISSR-PCR molecular marker technique was further used to identify the mutants of phenotypic variation plants. This study showed that low irradiation doses (5 Gy) stimulated bulb germination to improve the survival rate of tulip, while high irradiation doses (20 to 100 Gy) significantly ( P < 0.05) inhibited its seed germination and growth, and decreased the flowering rate, petal number, flower stem length and flower diameter. More than 40 Gy significantly ( P < 0.05) decreased the total chlorophyll content and increased the malondialdehyde (MDA) content in tulips. Interestingly, three types of both stigma variations and flower pattern variations, and four types of flower colour variations were observed. With increasing the irradiation dose from 5 to 100 Gy, the anthocyanin and flavonoid contents continuously decreased. Scanning electron microscopy (SEM) analysis evidenced that high irradiation doses altered the micromorphology of leaf stomata. Microscopic observations of tulip root apical mitosis further showed the abnormal chromosomal division behaviour occurring at different mitotic phases under irradiation treatment (80 Gy). Increasing the irradiation dose from 20 to 100 Gy enhanced the micronucleus rate. Moreover, the suspected genetic variation in tulips was evaluated by inter-simple sequence repeat (ISSR) analysis, and the percentage of polymorphic bands was 68%. Finally, this study concludes that that 80 Gy may be an appropriate radiation does to better enhance the efficiency of mutagenic breeds in tulip plants. Using γ-ray irradiation, therefore, is expected to offer a theoretical basis for mutation breeding in tulips.

Abstract:SUMMARY Ginger is cultivated in tropical and subtropical regions and is one of the most crucial spices worldwide owing to its special taste and scent. Here, we present a high‐quality genome assembly for 'Small Laiwu Ginger', a famous cultivated ginger in northern China. The ginger genome was phased into two haplotypes, haplotype A (1.55Gb), and haplotype B (1.44Gb). Analysis of Ty1/Copia and Ty3/Gypsy LTR retrotransposon families revealed that both have undergone multiple retrotransposon bursts about 0–1 million years ago. In addition to a recent whole‐genome duplication event, there has been a lineage‐specific expansion of genes involved in stilbenoid, diarylheptanoid, and gingerol biosynthesis, thereby enhancing 6‐gingerol biosynthesis. Furthermore, we focused on the biosynthesis of 6‐gingerol, the most important gingerol, and screened key transcription factors ZoMYB106 and ZobHLH148 that regulate 6‐gingerol synthesis by transcriptomic and metabolomic analysis in the ginger rhizome at four growth stages. The results of yeast one‐hybrid, electrophoretic mobility shift, and dual‐luciferase reporter gene assays showed that both ZoMYB106 and ZobHLH148 bind to the promoters of the key rate‐limiting enzyme genes ZoCCOMT1 and ZoCCOMT2 in the 6‐gingerol synthesis pathway and promote their transcriptional activities. The reference genome, transcriptome, and metabolome data pave the way for further research on the molecular mechanism underlying the biosynthesis of 6‐gingerol. Furthermore, it provides precious new resources for the study on the biology and molecular breeding of ginger.

Abstract:Ginger (Zingiber officinale Roscoe, Zingeberaceae) is a medicinal plant widely used as food, spice, or flavoring agent worldwide. 6-Shogaol is a compound of prime interest in exhibiting anti-inflammatory, antioxidant and chemopreventive effects. The objective of the study is to investigate the effect of microwave-assisted drying (MAD) followed by microwave-assisted extraction (MAE) so as to produce 6-Shogaol enriched Ginger with improved therapeutic benefits. Various drying techniques viz. shade drying, tray drying, microwave-assisted drying and osmotic dehydration as a pretreatment were used for drying Ginger rhizomes. The dried rhizomes were extracted by conventional solvent extraction and microwave-assisted extraction techniques and tested for content of 6-Shogaol using the newly developed HPLC method whereas total flavonoid and polyphenol content were determined using the UV spectrophotometric method. Subjecting the microwave dried Ginger to microwave-assisted extraction for 45 min at constant power level of 284 W resulted in a significant rise in the extractability of 6-Shogaol (1.660 ± 0.018), total polyphenols (855.46 ± 5.33) and flavonoids (617.97 ± 6.40) compared to the conventional method of extraction. The proposed Ginger processing method of microwave drying followed by microwave extraction outperforms traditional methods in terms of speed, convenience, and performance thus can be scaled up to industrial levels.

Abstract:The rhizome is an economically important part of ginger ( Zingiber officinale Rosc.). However, the mechanism of ginger rhizome enlargement remains unclear. In this study, we performed an integrated analysis of the hormone content and transcriptome of ginger at three rhizome enlargement stages: initial enlargement (S1), middle enlargement (S2), and peak enlargement (S3). With rhizome enlargement, the levels of the hormones zeatin (ZT), gibberellic acid (GA), indole acetic acid (IAA), and jasmonic acid (JA) were significantly increased, and this increase was positively correlated with rhizome diameter. Transcriptomic analysis identified a large number of differentially expressed genes (DEGs); the number of DEGs were 2,206 in the transition from S1 to S2, and 1,151 in the transition from S2 to S3. The expression of several genes related to hormone biosynthesis and signalling and cell division or expansion, and transcription factors was significantly altered, which suggests that these genes play essential roles in rhizome enlargement. The results of correlation analysis suggested that the process of ginger rhizome enlargement may be primarily related to the regulation of endogenous cytokinin, GA 3 , auxin, and JA biosynthesis pathways and signal transduction; GRAS , HB , MYB , MYB122 , bZIP60 , ARF1 , ARF2 , E2FB1 , and E2FB2 , which may regulate the expression of rhizome formation-related genes; and CYC2 , CDKB1 , CDKB2 , EXPA1 , and XTH7 , which may mediate cell division and expansion. These results provide gene resources and information that will be useful for the molecular breeding in ginger.

Abstract:Polyploids exhibit significant breeding value because of their better vegetative yields, higher agronomic characteristics, and greater tolerance to stress. The present study reports the effective in vitro induction of tetraploidy in ginger ( Zingiber officinale Rosc.) cv. Himgiri using different concentrations of colchicine (0.1%, 0.2%, 0.3%, and 0.4%) for different durations (6 to 48 h). The 0.1% colchicine treatment for 6 h had the best outcomes, with a maximum bud survival percentage of 73.33%, which decreased with the increase in colchicine concentration and treatment periods. Shoots regenerated from buds treated with 0.2% colchicine for 24 h showed good multiplication and elongation. The lethal time (LT 50 ) values declined as colchicine concentration increased, emphasizing its influence on the survival of treated buds over time. The maximum induction of polyploidy (50%) was achieved with 0.2% colchicine treatment for 6 and 24 h, whereas no polyploidy was induced with 0.4% colchicine treatment, which was reported to be lethal to the cells. After repeated subculturing, flow cytometric investigation indicated varied ploidy levels in regenerated shoots, with stable tetraploids. The morphological features, physiological characteristics, and biochemical compositions were evaluated, and substantial increases in plant height, leaf dimensions, tiller numbers, photosynthetic rates, and oleoresin content were observed. HPLC analysis depicted maximum gingerol content (498.36 μg g −1 ) in harvested rhizomes of tetraploid induced with 0.2% colchicine treatment for 24 h. Molecular analysis employing SCoT and CBDP revealed significant genomic alteration with polymorphism of 1.087% and 35.67%, respectively. The stable tetraploids developed in the present study will provide excellent resources for future ginger production and genetic improvement.

Abstract:Wide variability does not exist in okra [Abelmoschus esculentus (L.) Moench], requiring induction of genetic variability. Mutagenesis creates desired variability among genotypes without altering the agronomic base which is sometimes difficult to induce using other breeding methods. Seed germination/seedling emergence and seedling growth were evaluated under laboratory and field conditions by imposing 300 to 1000 Gy (Gray, the SI unit of radiation) gamma radiation to determine median lethal (LD50) and median growth reduction (GR50) doses with Co60gamma rays. The LD50 and GR50 doses varied for both growing conditions. More than 80% of seed exposed to the LD50 dose of gamma radiation in the laboratory did not emerge under field condition. The LD50 and GR50 doses of gamma radiation determined under field condition exhibited close correspondence and presented a reliable picture regarding lethality and injury in the M1 generation (the first generation from the irradiated seed). The R2 for the linear model for seedling emergence, shoot and root length, fresh and dry weight of seedlings under field condition were 0.908, 0.902, 0.911, 0.983 and 0.972, respectively. Determination of the LD50/GR50 is important to optimize the radiation dose to isolate desired utilizable mutants. Dose optimization should be determined through use of LD50 and GR50 under field condition. The gamma radiation dose between 350 Gy and 450 Gy was optimum to create desirable mutants in okra. The optimal dose could induce a higher rate of mutation with little biological damage in succeeding generations of okra.
In vitro polyploid induction using colchicine for Zingiber Officinale Roscoe cv. ‘Fengtou’ ginger

Abstract:'Fengtou' ginger is a high quality ginger (Zingiber Officinale Roscoe) cultivar only planted in Laifeng County, Enshi, Huhei, China, and is a National Product of Geographical Indication originated. 'Fengtou' ginger yield is low compared with most ginger. Therefore, it is imperative to apply breeding methods to improve yield. One strategy to accelerate breeding is polyploid induction, which could improve yield and maintain quality. In this study, different concentrations of colchicine (50, 100, 150, 200 mg/L) and different durations of exposure (3, 5, 7 days) were applied to stem segments of adventitious buds in ginger. Among obtained polyploidy plantlets were identified by flow cytometric analysis. The optimal induction condition of tetraploids was that the concentrations of colchicine was 150 mg/L, induced for 7 days, the chromosome doubling rate reached 18%. The tetraploid plants achieved were larger than their diploid plants for leaf length, leaf width, leaf thickness, stem diameter and guard cell. The soluble sugar, soluble protein, proline and other substances were also significant difference, especially carotenoids concentration was 1.375 times in contrast with diploid plants, which showed that tetraploids ginger had potentiality to increase yield and better adaptability.

Frequency and type of inheritable mutations induced by γ rays in rice as revealed by whole genome sequencing

Abstract:Mutation breeding is based on the induction of genetic variations; hence knowledge of the frequency and type of induced mutations is of paramount importance for the design and implementation of a mutation breeding program. Although γ ray irradiation has been widely used since the 1960s in the breeding of about 200 economically important plant species, molecular elucidation of its genetic effects has so far been achieved largely by analysis of target genes or genomic regions. In the present study, the whole genomes of six γ-irradiated M 2 rice plants were sequenced; a total of 144–188 million high-quality ( Q >20) reads were generated for each M 2 plant, resulting in genome coverage of >45 times for each plant. Single base substitution (SBS) and short insertion/deletion (Indel) mutations were detected at the average frequency of 7.5×10 −6 –9.8×10 −6 in the six M 2 rice plants (SBS being about 4 times more frequent than Indels). Structural and copy number variations, though less frequent than SBS and Indel, were also identified and validated. The mutations were scattered in all genomic regions across 12 rice chromosomes without apparent hotspots. The present study is the first genome-wide single-nucleotide resolution study on the feature and frequency of γ irradiation-induced mutations in a seed propagated crop; the findings are of practical importance for mutation breeding of rice and other crop species.

Abstract:Chamomile (Matricariarecutita L.) is one of the most important medicinal plants with various applications. The flowers and flower heads are the main organs inthe production of essential oil. The essential improvement goals of chamomile are considered to be high flower yield and oil content, as well asthe suitability for mechanical harvesting. The present study aimed to improve the flower yield, oil content and mechanical harvestability of German chamomile via chemical and physical mutagens. Three German chamomile populations (Fayum, Benysuif and Menia) were irradiated with 100, 200, 300 and 400 Gray doses of gamma rays, as well as chemically mutagenized using 0.001, 0.002 and 0.003 mol/mL of sodium azide for 4 h. The two mutagens produced a wide range of changes in the flowers' shape and size. At M3 generation, 18 mutants (11 from gamma irradiation and 7 from sodium azide mutagenization) were selected and morphologically characterized. Five out of eighteen mutants were selected for morphological and chemical characterization for oil content, oil composition and oil quality in M4 generation. Two promising mutants, F/LF5-2-1 and B/HNOF 8-4-2, were selected based on their performance in most studied traits during three generations, as well as the high percentage of cut efficiency and a homogenous flower horizon, which qualify them as suitable candidates for mechanical harvesting. The two mutants are late flowering elite mutants; the F/LF5-2-1 mutant possessed the highest oil content (1.77%) and number of flowers/plant (1595), while the second promising B/HNOF 8-4-2 mutant hada high oil content (1.29%) and chamazulene percentage (13.98%) compared to control plants. These results suggest that the B/HNOF 8-4-2 and F/LF5-2-1 mutants could be integrated as potential parents into breeding programs for a high number of flowers, high oil content, oil composition and oil color traits for German chamomile improvement.

Abstract:Ginger (Zingiber officinale Roscoe) is an important medicinal crop grown for its aromatic rhizome which is used as a spice, food, flavouring agent and medicine. It has been characterised for its hypoglycemic, hypotensive, antioxidant and antibiotic properties. This study was conducted to determine the impact of plant growth-promoting potential of bacterial strain Bacillus subtilis L2 on plant growth and physiological properties of ginger. The experiment was carried out in randomised block design with three replications in pot experiments. The plants were grown in greenhouse conditions for three months. The results showed that at 8 and 12 weeks after planting (WAP) bacterial inoculation increased plant height, leaf length, number of leaves per plant and leaf width. Inoculation with B. subtilis L2 significantly increased plant height by 16, 20 and 18% compared to control at 4, 8 and 12 WAP. At 8 and 12 WAP, leaf length significantly raised by B. subtilis L2 as compared to uninoculated control. B. subtilis L2 significantly increased the number of leaves per plant and leaf width by 30 and 21% respectively when comparing with non-inoculated plants at 8 WAP. The percentage increase in chlorophyll content resulted from the inoculation with B. subtilis L2 over the control was 10.5%, 15.5% and 18.4% at 4, 8 and 12 WAP respectively. It is concluded that there is a significant positive effect of inoculation with B. subtilis L2 on the growth of ginger. B. subtilis L2 strain can be used as a potential agent or bio-fertiliser for stimulation of ginger growth.

Abstract:The increasing demand for red ginger ( Zingiber officinale Roxb. var. rubrum Rosc.) both at the domestic and international levels has led to the need for gingerol production, a main compound of red ginger, which has various pharmacological activities. The urgency of this research is related to gingerol produced in cultivated red ginger, which often shows variability in quantity due to genetic variation and differences in geographical and environmental conditions where it is grown, so it requires gingerol standardization efforts. Through tissue culture techniques, it is possible to propagate plants in a controlled environment, ensuring genetic uniformity and minimizing variations caused by genetic factors. Red ginger raw materials that can be produced consistently, quickly, and land-efficiently with high gingerol content and pesticide-free have become an essential economic necessity. In this joint study with PT. Bintang Toedjoe, researchers intend to utilize root culture bioprocessing technology to increase gingerol production from red ginger. Root cultures have stabile genetics and growth faster; thus, these techniques imply the formation of organs or structures conducive to enhanced gingerol production. Our research has revealed successful protocols for inducing and multiplying suitable callus for organogenesis. Through the application of hormones, the best callus induction is using a combination of 3 ppm 2,4-D and 0.2 ppm BA with a callus production percentage of 67%. On the other hand, a satisfactory callus multiplication rate was used using 1 ppm 2,4-D with the most significant increase in explant area (79 mm 2) by ruler alignment. Meanwhile, the rooting response was prominent at 1 ppm 2,4-D + 3 ppm BA.
