Abstract:Acute and long-term exercise differentially affect brain functions. It has been suggested that neuronal activation is one of the mechanisms for exercise-induced enhancement of brain functions. However, the differential effects of acute and long-term exercise on the spatial and temporal profiles of neuronal activation in the brain have been scarcely explored. In this study, we profiled the expression of c-Fos, a marker of neuronal activation, in selected 26 brain regions of 2-month-old male C57/B6 mice that received either a single bout of treadmill running (acute exercise) or a 4-week treadmill training (long-term exercise) at the same duration (1 h/day) and intensity (10 m/min). The c-Fos expression was determined before, immediately after, and 2 h after the run. The results showed that acute exercise increased the densities of c-Fos+ cells in the ventral hippocampal CA1 region, followed by (in a high to low order) the primary somatosensory cortex, other hippocampal subregions, and striatum immediately after the run; significant changes remained evident in the hippocampal subregions after a 2-h rest. Long-term exercise increased the densities of c-Fos+ cells in the striatum, followed by the primary somatosensory, primary and secondary motor cortices, hippocampal subregions, hypothalamic nuclei, and lateral periaqueductal gray; significant changes remained evident in the striatum, hippocampal subregions, hypothalamic nuclei, and lateral periaqueductal gray after a 2-h rest. Interestingly, the densities of c-Fos+ cells in the substantia nigra and ventral tegmental area only increased after a 2-h rest after the run in the long-term exercise group. The densities of c-Fos+ cells were positively correlated with the expression of brain-derived neurotrophic factor in the selected brain regions. In conclusion, both acute and long-term treadmill running at mild intensity induce c-Fos expression in the limbic system and movement-associated cortical and subcortical regions, with long-term exercise involving more brain regions (i.e., hypothalamus and periaqueductal gray) and longer lasting effects.

Long-Term Exercise Is a Potent Trigger for ΔFosB Induction in the Hippocampus along the dorso–ventral Axis

Abstract:Physical exercise improves multiple aspects of hippocampal function. In line with the notion that neuronal activity is key to promoting neuronal functions, previous literature has consistently demonstrated that acute bouts of exercise evoke neuronal activation in the hippocampus. Repeated activating stimuli lead to an accumulation of the transcription factor ΔFosB, which mediates long-term neural plasticity. In this study, we tested the hypothesis that long-term voluntary wheel running induces ΔFosB expression in the hippocampus, and examined any potential region-specific effects within the hippocampal subfields along the dorso-ventral axis. Male C57BL/6 mice were housed with or without a running wheel for 4 weeks. Long-term wheel running significantly increased FosB/ΔFosB immunoreactivity in all hippocampal regions measured (i.e., in the DG, CA1, and CA3 subfields of both the dorsal and ventral hippocampus). Results confirmed that wheel running induced region-specific expression of FosB/ΔFosB immunoreactivity in the cortex, suggesting that the uniform increase in FosB/ΔFosB within the hippocampus is not a non-specific consequence of running. Western blot data indicated that the increased hippocampal FosB/ΔFosB immunoreactivity was primarily due to increased ΔFosB. These results suggest that long-term physical exercise is a potent trigger for ΔFosB induction throughout the entire hippocampus, which would explain why exercise can improve both dorsal and ventral hippocampus-dependent functions. Interestingly, we found that FosB/ΔFosB expression in the DG was positively correlated with the number of doublecortin-immunoreactive (i.e., immature) neurons. Although the mechanisms by which ΔFosB mediates exercise-induced neurogenesis are still uncertain, these data imply that exercise-induced neurogenesis is at least activity dependent. Taken together, our current results suggest that ΔFosB is a new molecular target involved in regulating exercise-induced hippocampal plasticity.

Abstract:PURPOSE: Irisin, Bdnf And Il-6 Are Exercise-Induced Myokines Which Exert Their Effects In Driving Brown-Fat-Like Development (Irisin) And Regulating Fat Oxidation. However, Their Dynamic Expression In Response To Exercise Is Not Fully Known. Here We Examined The Level Of Irisin, Bdnf And Il-6 As Well As The Activation Of Ampk And Akt At Different Time Points Following Exercise, In Order To Explore The Pattern Of Their Expression And Biological Functions. METHODS: Sprague-Dawley Rats Were Subjected To Downhill Running At 17m/Min For 90 Minutes. Serum Creatine Kinase Activity Was Tested At Day 1, 3, 5, 7, And 14 Following Exercise To Evaluate Skeletal Muscle Injury. The Serum Level And Skeletal Expression Of Bdnf, Il-6 And Irisin Were Measured By Elisa And Real-Time Rt-Pcr, Respectively. The Activation Of Ampk And Akt Signaling Pathway Was Confirmed By Western Blot. To Compare The Difference Between Two Groups, T-Test Was Employed Using Spss Software And P < 0.05 Was Considered Statistically Significant. RESULTS: An Increase Of Ck Activity Was Observed At Day 3 And Day 5 Following Exercise, Confirming Exercise-Related Skeletal Muscle Injury. We Didn’T Find Any Increase Of Il-6 In The Rats Following Exercise Compared To Sedentary Group, Although It Is Reported That Il-6 Level Is Elevated Dramatically During Exercise. The Level Of Irisin And Bdnf Was Increased Both In The Serum And In The Skeletal Muscle In Day 1 And Day 3 Following Downhill Running; Then, While Irisin Level Returned To The Baseline, Serum Bdnf Level Was Continuously Elevated. Besides, Ampk Activation Was Found At Day 1 And Day 3, But Akt Was Kept Activated Over All The Time Points. CONCLUSIONS: Although Il-6, Irisin And Bdnf Are All Exercise-Induced Myokines, Their Expression Is Regulated Differently. Irisin And Bdnf Are Both Reported To Be Involved In The Activation Of Ampk Signaling Pathway. However, We Found That The Increase Of Irisin Was In Accordance With Ampk Activation, While Bdnf Increase Is In Accordance With Akt Activation. Our Results Suggest That Bdnf May Participate In Skeletal Muscle Regeneration Via Akt-Mediated Pathway In Addition To Fat Oxidation. Therefore, Exercise-Induced Myokines Are Regulated Precisely And Sequentially, Which In Turn Exert Different Functions At Different Time Following Exercise.

Abstract:Rats submitted to acute incremental exercises following the model established by Bedford,neuron-endocrine system,lipid peroxidation and cytokine were detected.Results showed that most of the changes of hypothalamus-pituitary-adrenal axis and lipid peroxidation induced by exercise lasted at least 5 hours after the acute exercise in this model.

Effect of Plasma Endogenous Opioids on Cardiovascular Regulation under Different Central Neuro-Command During Isometric Exercise

Abstract:Objective To explore the effect of plasma endogenous opioids (EOPs) on cardiovascular regulation under different central neuro command during isometric exercise.Methods 15 polio patients, as the central neuro command model (Polio) and 10 hemiplegia patients, as the spinal central command model (Hemi),were studied. Exercise with unaffected limbs was used as the normal model of integration of neuro control. The subjects performed sustained maximal isometric exercise with the affected extremity (AIE) first, after 10 minute rest, then performed sustained maximal isometric exercise with the unaffected extremity (NIE). Heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP) and plasma concentration of Ler Enkephalin (L EK),β Endorphin (β END) and Dynorphin A (DYN) were measured pre exercise, during AIE, NIE and after exercise, respectively.Results ① HR, SBP and DBP increased significantly during AIE and NIE comparing with pre exercise in the Polio group (P 0.01 ). The responses to NIE were stronger than that to AIE. ②HR, SBP and DBP increased significantly during AIE and NIE comparing with pre exercise in the Hemi group (P 0.01 ). The responses to NIE were weaker than that to AIE. ③ The responses of plasma L EK, β END and DYN in AIE and NIE presented the same trend as the cardiovascular responses.Conclusion ①Central neuro command has an independent role in regulation of plasma EOPs release and cardiovascular response during isometric exercise.② When the function of cerebral cortex is deficit, the abnormal sub cortex command may induce extortionate plasma EOPs release and cardiovascular response.

Abstract:Introduction Dopaminergic, opiod and endocannabinoid neurotransmission are thought to play an important role in the neurobiology of acute exercise and, in particular, in mediating positive affective responses and reward processes. Recent evidence indicates that changes in fractional amplitude of low-frequency fluctuations (zfALFF) in resting-state functional MRI (rs-fMRI) may reflect changes in specific neurotransmitter systems as tested by means of spatial correlation analyses. Methods Here, we investigated this relationship at different exercise intensities in twenty young healthy trained athletes performing low-intensity (LIIE), high-intensity (HIIE) interval exercises, and a control condition on three separate days. Positive And Negative Affect Schedule (PANAS) scores and rs-fMRI were acquired before and after each of the three experimental conditions. Respective zfALFF changes were analyzed using repeated measures ANOVAs. We examined the spatial correspondence of changes in zfALFF before and after training with the available neurotransmitter maps across all voxels and additionally, hypothesis-driven, for neurotransmitter maps implicated in the neurobiology of exercise (dopaminergic, opiodic and endocannabinoid) in specific brain networks associated with “reward” and “emotion.” Results Elevated PANAS Positive Affect was observed after LIIE and HIIE but not after the control condition. HIIE compared to the control condition resulted in differential zfALFF decreases in precuneus, temporo-occipital, midcingulate and frontal regions, thalamus, and cerebellum, whereas differential zfALFF increases were identified in hypothalamus, pituitary, and periaqueductal gray. The spatial alteration patterns in zfALFF during HIIE were positively associated with dopaminergic and μ-opioidergic receptor distributions within the ‘reward' network. Discussion These findings provide new insight into the neurobiology of exercise supporting the importance of reward-related neurotransmission at least during high-intensity physical activity.

Abstract:In this paper we discuss recent experimental and clinical findings which lead us to propose that prolonged rhythmic exercise can activate central opioid systems by triggering increased discharge from mechanosensitive afferent nerve fibers (Group III or A-delta) arising from contracting skeletal muscle. We review evidence that supports the concept that many of the cardiovascular, analgesic, and behavioral effects of exercise are mediated by this mechanism and that the same or similar mechanisms are responsible for the central and peripheral effects of acupuncture. Based on this hypothesis, and supporting evidence from human and animal studies, we suggest a mechanism and a potential therapeutic role for exercise in the treatment of selected patients with disorders as diverse as hypertension, addiction, depression, and anorexia nervosa.

Abstract:Exercise preconditioning (EP) has a good myocardial protective effect. This study explored whether EP improves electrocardiographic (ECG) signs of myocardial ischemic/hypoxic injury and the occurrence of malignant arrhythmia after exhaustive exercise. A total of 120 male SD rats were randomly divided into the control group (group C), early exercise preconditioning group (group EEP), late exercise preconditioning group (group LEP), exhaustive exercise group (group EE), early exercise preconditioning + exhaustive exercise group (group EEP + EE) and late exercise preconditioning + exhaustive exercise group (group LEP + EE). Changes in heart rate (HR), ST segment, T wave and QT corrected (QTc) intervals on ECG; hematoxylin-basic fuchsin-picric acid (HBFP) staining; and cTnI levels were used to study myocardial injury and the protective effect of EP. Compared with those in group C, the levels of plasma markers of myocardial injury, HBFP staining and ECG in group EE were significantly increased ( P < 0.05). Compared with those in group EE, the levels of plasma markers of myocardial injury, HBFP staining and ECG in group EEP + EE and group LEP + EE were significantly decreased ( P < 0.05). The results suggested that EP improved ECG signs of myocardial ischemic/hypoxic injury and malignant arrhythmias that occur after exhaustive exercise. The ST segment and T wave could also serve as indexes for evaluating exhaustive exercise-induced myocardial ischemia/hypoxia.

Abstract:Regular exercise prevents lipid abnormalities and conditions such as diabetes mellitus, hypertension, and obesity; it considerably benefits sedentary individuals. However, individuals exhibit highly variable responses to exercise, probably due to genetic variations. Animal models are typically used to investigate the relationship of intrinsic exercise capacity with physiological, pathological, psychological, behavioral, and metabolic disorders. In the present study, we investigated differential physiological adaptations caused by intrinsic exercise capacity and explored the regulatory molecules or mechanisms through multiomics approaches. Outbred ICR mice (n = 100) performed an exhaustive swimming test and were ranked based on the exhaustive swimming time to distinguish intrinsically high- and low-capacity groups. Exercise performance, exercise fatigue indexes, glucose tolerance, and body compositions were assessed during the experimental processes. Furthermore, the gut microbiota, transcriptome, and proteome of soleus muscle with intrinsically high exercise capacity (HEC) and low exercise capacity (LEC) were further analyzed to reveal the most influential factors associated with differential exercise capacities. HEC mice outperformed LEC mice in physical activities (exhaustive swimming and forelimb grip strength tests) and exhibited higher glucose tolerance than LEC mice. Exercise-induced peripheral fatigue and the level of injury biomarkers (lactate, ammonia, creatine kinase, and aspartate aminotransferase) were also significantly lower in HEC mice than in LEC mice. Furthermore, the gut of the HEC mice contained significantly more Butyricicoccus than that of the LEC mice. In addition, transcriptome data of the soleus muscle revealed that the expression of microRNAs that are strongly associated with exercise performance-related physiological and metabolic functions (i.e., miR-383, miR-107, miR-30b, miR-669m, miR-191, miR-218, and miR-224) was higher in HEC mice than in LEC mice. The functional proteome data of soleus muscle indicated that the levels of key proteins related to muscle function and carbohydrate metabolism were also significantly higher in HEC mice than in LEC mice. Our study demonstrated that the mice with various intrinsic exercise capacities have different gut microbiome as well as transcriptome and proteome of soleus muscle by using multiomics approaches. The specific bacteria and regulatory factors, including miRNA and functional proteins, may be highly correlated with the adaptation of physiological functions and exercise capacity.

Effects of high-intensity cycle exercise on sympathoadrenal-medullary response patterns

Abstract:Plasma proenkephalin peptide F immunoreactivity and catecholamines were examined on separate days in nine healthy males before and after maximal exercise to exhaustion at four intensities [36, 55, 73, and 100% of maximal leg power (MLP)] by use of a computerized cycle ergometer. The mean duration of 36, 55, 73, and 100% MLP was 3.31, 0.781, 0.270, and 0.1 min, respectively. All intensities were greater than those eliciting peak O2 uptake for the individual subjects. Blood samples were obtained before, immediately after exercise, and 5 and 15 min after exercise. Significant (P less than 0.05) increases in plasma peptide F immunoreactivity (i.e., from mean resting value of 0.18 to 0.43 pmol/ml) were observed immediately after exercise at 36% MLP. Significant increases in plasma epinephrine were observed immediately after exercise at 36% MLP (i.e., from mean resting value of 2.22 to 3.11 pmol/ml) and 55% MLP (i.e., from mean resting value of 1.67 to 2.98 pmol/ml) and 15 min after exercise at 100% MLP (i.e., from mean resting value of 1.92 to 3.88 pmol/ml). Significant increases for plasma norepinephrine were observed immediately after exercise (36, 55, 73, and 100% MLP), 5 min after exercise (36, 55, and 73% MLP), and 15 min after exercise (36% MLP). Increases in whole blood lactate were observed at all points after exercise for 36, 55, and 73% MLP and 5 min after exercise for 100% MLP. These data show that brief high-intensity exercise results in differential response patterns of catecholamines and proenkephalin peptide F immunoreactivity.
Influence of experimental navigation and violent exercise on neuropeptide Y and hypothalamic-pituitary-adrenal axis in rats

Abstract:Objective: To explore the effects of experimental navigation and violent exercise on neuropeptide Y and hypothalamic-pituitary-adrenal axis and the interaction between them in rats.Methods: Thirty-six male rats were randomly assigned to three groups: Group A,B and C.Group A received 180 minutes' experimental navigation as well as psychological stress induced by a closely placed cat.Group B received treatment as group A,and after that,received an intensive running on treadmill for 120min.Group C(the controls) was only treated with restriction of water and food intakes.The levels of NPY,ACTH,CORT were measured by RIA.Results:Serum NPY level decreased significantly in group A(P=0.026)and increased significantly in group B(P<0.01) compared with group C.The mean serum CORT level increased remarkably in group B compared with group C and A(both P<0.01).And the serum ACTH level decreased significantly in group B(P=0.033).Conclusion: Experimental navigation may cause significantly decrease of serum NPY level.Violent exercises after psychological stress may increase the NPY level and activate HPA axis.The increased secretion of NPY might have an effect on enhanced serum levels of CORT and CORT in rats.
The Effects of Voluntary Physical Exercise-Activated Neurotrophic Signaling in Rat Hippocampus on mRNA Levels of Downstream Signaling Molecules

Abstract:Physical exercise results in the increased expression of neurotrophic factors and the subsequent induction of signal transduction cascades with a positive impact on neuronal functions. In this study, we used a voluntary physical exercise rat model to determine correlations in hippocampus mRNA expression of the neurotropic factors Bdnf, VegfA, and Igf1; their receptors TrkB, Igf1R, VegfR1, and VegfrR2; and downstream signal transducers Creb, Syn1, and Vgf. In hippocampi of physically exercised rats, the mRNA expression levels of Bdnf transcript 4 (Bdnf-t4), VegfA, and Igf1, as well as VegfR1, TrkB, Creb, Vgf, and Syn1, were increased. Bdnf-t4 mRNA expression positively correlated with mRNA expression of Creb, Vgf, and Syn1 in hippocampi of exercised rats. A correlation between Bdnf-t4 and Syn1 mRNA expression was also observed in hippocampi of sedentary rats. Igf1 and VegfA mRNA expression was positively correlated in hippocampi of both exercised and sedentary rats. But, neither Igf1 nor VegfA mRNA expression was correlated with the expression of Bdnf-t4 or the expression of the signal transducers Creb, Syn1, and Vgf. In hippocampi of exercised rats, Creb mRNA expression was positively correlated with TrkB, Syn1, and Vgf mRNA expression and with the correlation between Creb and Vgf mRNA expression also observed in hippocampi of sedentary rats. To examine for causality of the in vivo observed correlated mRNA expression levels between Bdnf-t4 and the other examined transcripts, we used nuclease-deactivated CRISPR-Cas9 fused with VP64 to induce mRNA expression of endogenous Bdnf-t4 in rat PC12 cells. Following Bdnf-t4 mRNA induction, we observed increased Creb mRNA expression. This in vitro result is in accordance with the in vivo results and supports that under specified conditions, an increase in Creb mRNA expression can be a downstream signal transduction event due to induction of endogenous Bdnf mRNA expression.

Abstract:Objective: The substantia nigra pars reticular (SNR) is the vital nuclei sending out information from basal ganglia and it plays an important part in motor behavior maintenance and motor regulation. It aims to observe the changes of mGluR4,GABA-ARα1 and GABA-BR1 in substantia nigra pars reticular of rats induced by one single bout of exhaustive exercise, and to discuss the relationship among the expression of mGluR4,GABA-ARα1, GABA-BR1 and exercise fatigue. Methods: Thirty-six male wistar rats were divided into three groups at random including CG, 0EG and 90EG,which having 12 rats. Meanwhile, peripheral blood samples were drawn from the vein of tail at different time points (pre- exercise, exercise for 30 min, exercise for 90 min and exhaustion immediately during exercise, and 30 min and 90 min post exercise) ,and blood concentrations of lactic acid (LD) ,blood urea nitrogen (BUN) and activities of creatine kinase (CK) were detected by automatic analyser. Furthermore, the technique of Immunohistochemistry has been used to indicate the protein expression of metabotropic glutamate receptors like mGluR4 and gamma-aminobutyric acid a receptor like GABAAR1, also gamma-aminobutyric acid B receptor like GABAB1. Results: The expression level of mGluR4 in SNR in immediately post-exercise group (0EG) and 90 min post-exercise group (90EG) increase significantly than control group (CG), and the expression level of GABA-ARα1 in SNR at 0EG and 90EG were also significantly higher than control group, and GABA-BR1 in SNR in 0EG and 90EG were both significantly lower than CG. Moreover, it presented significant increases in the levels of LD, BUN and CK in peripheral blood compared with those at rest. Conclusion: Exercise fatigue could result in up-regulation of mGluR4 expression,GABA-ARα1 and down-regulation of GABA-BR1 expression in substantia nigra pars reticular of rats, and it also induce the muscle injury by increase the level of LD, CK and BUN in peripheral blood, which suggested that substantia nigra pars reticular was an important brain rigion to modulate the motor function, and mGluR4,GABA-ARα1 and GABA-BR1 were three important receptors related with the muscle injury induced by exercise fatigue. So the mGluR4, GABA-ARα1 and GABA-BR1 could be the new novel substance produced to prevent from exercise fatigue.
Acute effect of exercise intensity on circulating FGF-21, FSTL-1, cathepsin B, and BDNF in young men

Abstract:Background/objectives: Exercise intensity is potentially an important regulator of various exerkines secretion, but the optimal exercise intensity to increase and sustain exerkines levels, including FGF-21, FSTL-1, cathepsin B, and BDNF in humans, has not yet been fully elucidated. This study aimed to examine the circulating levels of FGF-21, FSTL-1, cathepsin B, and BDNF according to the exercise intensity.Methods: Nine young men (24.0 ± 0.4 years old) performed 4 different experimental sessions at 1-week intervals: 1) a control session (CTRL; no exercise); 2) moderate-intensity continuous exercise (MICE, 55% HRR); 3) vigorous-intensity continuous exercise (VICE, 85% HRR); and 4) high-intensity interval exercise (HIIE, 4 repetitions of a 30-s of "all out" cycling workout followed by a 4-min recovery). Blood samples were collected at 4 different time points (pre-exercise, immediately post-exercise, 30 min post-exercise, and 90 min post-exercise).Results: Serum FGF-21, FSTL-1, cathepsin B, and BDNF were higher in HIIE than in CTRL immediately post-exercise, and FSTL-1, cathepsin B, and BDNF were higher in HIIE than in MICE immediately post-exercise (<i>P</i> < 0.05). The AUC for FGF-21, FSTL-1, and BDNF was higher in HIIE than in CTRL, and the AUC for FGF-21 and BDNF was higher in HIIE than in MICE (<i>P</i> < 0.05). Furthermore, the change in blood lactate was positively correlated with the changes in all exerkines.Conclusions: This study demonstrates that acute HIIE effectively increases serum FGF-21, FSTL-1, cathepsin B, and BDNF compared to MICE. Therefore, the secretion of exerkines, including FGF-21, FSTL-1, cathepsin B, and BDNF may be exercise intensity-dependent.

Abstract:This paper introduces principled methods for the reliability analysis of business processes that rely on web services. The input to the problem is the BPEL specification of a business process and the output is the prediction of the process's reliability. The first step to this end involves a method for the translation of the BPEL specification to its corresponding UML model. The second step of the reliability analysis involves a principled way for the annotation of the UML model with the necessary extensions for the specification of reliability properties that characterise the behaviour of the elements that constitute the process. The third step of the analysis comprises the systematic mapping of the extended UML model to block diagrams and Markov models which are subsequently used to compute the reliability of the process.

Abstract:Peripheral physiological adaptations to exercise have been the subject of several studies. To adjust to the disturbance in resting homeostasis induced by the exercise stimulus, a number of regulatory systems are called upon to return the body to a new level of homeostasis. In contrast with our knowledge about the peripheral adaptations to exercise, studies relating exercise to brain neurotransmitter levels are scarce. It is of interest to examine the effect of acute and chronic exercise on neurotransmitter release, since movement initiation and control of locomotion have been shown to be related to striatal neurotransmitter function. One of the possible therapeutic modalities in movement and mental disorders is exercise therapy. But also chronic adaptations to training and disturbances caused by overtraining might be reflected in brain functioning. Physical exercise influences the central dopaminergic, noradrenergic, and serotonergic systems. A number of studies have examined brain noradrenaline (NA), serotonin (5-hydroxytryptamine, or 5-HT), and dopamine (DA) with exercise. Although there are great discrepancies in experimental protocols, the results indicate that there is evidence in favor of changes in synthesis and metabolism of monoamines during exercise. In previous studies, we used the microdialysis technique to monitor the release of neurotransmitters in different brain nuclei during exercise. We have shown that DA, NA, and 5-HT release is increased during exercise. We also showed that acute exercise and/or exercise training influences extracellular DA, NA, GLU, and GABA levels. Taken together this could indicate that chronic disturbance of homeostasis could also influence transmitter release and that this paradigm could serve as a tool to explore the central effects of overtraining.

Endurance training effects on neurotransmitter release in rat striatum: an in vivo microdialysis study

Abstract:In the present study we use the in vivo microdialysis sampling technique to register extracellular levels of neurotransmitters in the striatum of trained and untrained rats. We further evaluate the influence of 1 h of exercise on the striatal release of dopamine (DA), noradrenaline (NA), glutamate (GLU) and gamma-aminobutyric acid (GABA) in trained and untrained rats. Male Wistars were randomly assigned to a training or control group. The exercise training consisted of running on a treadmill for 6 weeks, 5 days week-1, with running time and speed gradually increased from 30 min at 19 m min-1 during the first week to 80 min at 26 m min-1 during the final training week. The animals of the control group were placed on the treadmill twice a week, and received a total of four 'adaptation sessions', in which they exercised 15-45 min at 26 m min-1. Brain dialysates were analysed with microbore liquid chromatography (LC), with electrochemical detection (monoamines and GABA) and fluorescence detection (GLU). Soleus citrate synthase and basal striatal concentrations of DA, NA and GLU were significantly different between the trained and control animals. Sixty minutes of exercise significantly increased extracellular DA, NA and GLU levels in both groups, but there was no statistically significant difference in the exercise-induced increase between trained and control animals. There was no statistical difference in basal or exercise-induced GABA levels between trained and control animals. The results indicate that exercise training appears to result in diminished basal activity of striatal neurotransmitters, while maintaining the necessary sensitivity for responses to acute exercise.

Abstract:Objective To observe the effects of a single bout of exhaustive exercise on the endoplasmic reticulum stress(ERS) in liver and skeletal muscle of mice,in order to explore the mechanisms underlying acute sports injury.Methods Sixteen C57/BL6 male mice were randomly divided into control group(n = 8) and exercise group(n = 8).After 4 hours of a single bout of exhaustive swimming,the protein expression of GRP78(glucose-regulated protein 78),p-PERK/PEPK(PKR-like ER kinase),p-IRE1/IRE1(inositol requiring enzyme 1) and mRNA of GRP78,XBP1s(X box binding protein 1 splicing),CHOP(C/EBP homologous protein),GADD34(growth arrest and DNA damage-inducible gene 34)were measured in liver and quadriceps;the contents of blood glucose,liver glycogen and muscle glycogen were assayed;the morphological changes in liver and quadriceps were observed under light microscope.Results Compared to the control group,the protein expression of GRP78,p-PERK/PERK,p-IRE1/IRE1 and the mRNA level of GRP78,XBP1s,CHOP,GADD34 increased markedly(P < 0.001) in liver and quadriceps in the exercise group;the concentrations of blood glucose,liver glycogen and muscle glycogen decreased significantly(P < 0.05);edema was observed in some hepatic cells.The negative correlations were found between glycogen contents and ERS biomarkers.Conclusion A single bout of exhaustive exercise could enhance the degree of ERS,which probably was associated with glycogen storage and consumption.
