Abstract:Objective: To observe the modulatory effect of subthalamic nucleus (STN) on activity of motor cortex during exhausting exercise. Methods: Electrocorticogram (ECoG) and local field potentials (LFPs) recording techniques were applied simultaneously to observe the dynamic changes of oscillations in sensorimotor area and STN of rat during exhausting exercise. Results: Rats ran well initiatively with treadmill at the beginning of the exercise, about 45 min (45 +/- 11.5) later, movement capacity reduced. Corresponding electrical property showed that STN activity increased significantly while activity of cortex decreased significantly. Subsequently rats continued exercise with minor external stimulation utill exhaustion. Activity of ECoG reached the minimum under exhausting stations (P < 0.01), while the activity of LFPs changed insignificantly (P > 0.05). Conclusion: During the exhausting exercise, the cortex activity was extensively depressed with the development of fatigue, while the activity of STN increased significantly at the early stage of fatigue, STN took part in the modulation of central fatigue through negative induction. And the increase of STN activity may be one of the key measures accounting for protective inhibition.
The Effect of Anti-Fatigue of mGluR4, GABA-ARα1 and GABA-BR1 on the Substantia Nigra Pars Reticular During Exercise

Abstract:Objective: The substantia nigra pars reticular (SNR) is the vital nuclei sending out information from basal ganglia and it plays an important part in motor behavior maintenance and motor regulation. It aims to observe the changes of mGluR4,GABA-ARα1 and GABA-BR1 in substantia nigra pars reticular of rats induced by one single bout of exhaustive exercise, and to discuss the relationship among the expression of mGluR4,GABA-ARα1, GABA-BR1 and exercise fatigue. Methods: Thirty-six male wistar rats were divided into three groups at random including CG, 0EG and 90EG,which having 12 rats. Meanwhile, peripheral blood samples were drawn from the vein of tail at different time points (pre- exercise, exercise for 30 min, exercise for 90 min and exhaustion immediately during exercise, and 30 min and 90 min post exercise) ,and blood concentrations of lactic acid (LD) ,blood urea nitrogen (BUN) and activities of creatine kinase (CK) were detected by automatic analyser. Furthermore, the technique of Immunohistochemistry has been used to indicate the protein expression of metabotropic glutamate receptors like mGluR4 and gamma-aminobutyric acid a receptor like GABAAR1, also gamma-aminobutyric acid B receptor like GABAB1. Results: The expression level of mGluR4 in SNR in immediately post-exercise group (0EG) and 90 min post-exercise group (90EG) increase significantly than control group (CG), and the expression level of GABA-ARα1 in SNR at 0EG and 90EG were also significantly higher than control group, and GABA-BR1 in SNR in 0EG and 90EG were both significantly lower than CG. Moreover, it presented significant increases in the levels of LD, BUN and CK in peripheral blood compared with those at rest. Conclusion: Exercise fatigue could result in up-regulation of mGluR4 expression,GABA-ARα1 and down-regulation of GABA-BR1 expression in substantia nigra pars reticular of rats, and it also induce the muscle injury by increase the level of LD, CK and BUN in peripheral blood, which suggested that substantia nigra pars reticular was an important brain rigion to modulate the motor function, and mGluR4,GABA-ARα1 and GABA-BR1 were three important receptors related with the muscle injury induced by exercise fatigue. So the mGluR4, GABA-ARα1 and GABA-BR1 could be the new novel substance produced to prevent from exercise fatigue.
Neuroplasticity of dopamine circuits after exercise: implications for central fatigue

Abstract:Habitual exercise increases plasticity in a variety of neurotransmitter systems. The current review focuses on the effects of habitual physical activity on monoamine dopamine (DA) neurotransmission and the potential implication of these changes to exercise-induced fatigue. Although it is clear that peripheral adaptations in muscle and energy substrate utilization contribute to this effect, more recently it has been suggested that central nervous system pathways "upstream" of the motor cortex, which initiate activation of skeletal muscles, are also important. The contribution of the brain to exercise-induced fatigue has been termed "central fatigue." Given the well-defined role of DA in the initiation of movement, it is likely that adaptations in DA systems influence exercise capacity. A reduction in DA neurotransmission in the substantia nigra pars compacta (SNpc), for example, could impair activation of the basal ganglia and reduce stimulation of the motor cortex leading to central fatigue. Here we present evidence that habitual wheel running produces changes in DA systems. Using in situ hybridization techniques, we report that 6 weeks of wheel running was sufficient to increase tyrosine hydroxylase mRNA expression and reduce D2 autoreceptor mRNA in the SNpc. Additionally, 6 weeks of wheel running increased D2 postsynaptic receptor mRNA in the caudate putamen, a major projection site of the SNpc. These results are consistent with prior data suggesting that habitually physically active animals may have an enhanced ability to increase DA synthesis and reduce D2 autoreceptor-mediated inhibition of DA neurons in the SNpc compared to sedentary animals. Furthermore, habitually physically active animals, compared to sedentary controls, may be better able to increase D2 receptor-mediated inhibition of the indirect pathway of the basal ganglia. Results from these studies are discussed in light of our understanding of the role of DA in the neurobiological mechanisms of central fatigue.
Screening People in the Waiting Room for Vestibular Impairments

Abstract:Objective Primary care physicians need good screening tests of the vestibular system to help them determine whether patients who complain of dizziness should be evaluated for vestibular disorders. The goal of this study was to determine whether current, widely used screening tests of the vestibular system predict subsequent performance on objective diagnostic tests of the vestibular system (ENG). Methods Of 300 subjects who were recruited from the waiting room of a primary care clinic and were screened there, 69 subjects subsequently volunteered for ENGs in the otolaryngology department. The screening study included age, history of vertigo, head impulse tests, Dix-Hallpike maneuvers, and the Clinical Test of Sensory Integration and Balance with the head still and the head pitching at 0.33 Hz. The ENG included Dix-Hallpike maneuvers, vestibular-evoked myogenic potentials, bithermal water caloric tests, and low-frequency sinusoids in the rotatory chair in darkness. Results The scores on the screening were related to the total ENG, but odds ratios were not significant for some variables, probably because of the small sample size. Conclusions A larger sample may have yielded stronger results, but in general the high odds ratios suggest a relation between the ENG score and Dix-Hallpike responses and between the ENG scores and some Clinical Test of Sensory Integration and Balance responses.

Abstract:Exercise fatigue is a common physiological phenomenon in sports. The central nervous system (CNS) has important regulating effect on exercise fatigue. The basal ganglia are involved in motor function regulation through direct and indirect pathways. Striatum is the main input nucleus of basal ganglia and plays an important role in movement control. PURPOSE: Explore the role of dopaminergic synapse and D2 dopamine receptors (D2DR) in movement control by investigating synaptic ultrastructural change of the dorsolateral striatum (DLS) and D2DR antagonist/agonist effect on autonomic activity in exercise-induced fatigue rats. METHODS: Male Wistar rats were randomly divided into either the sham control group (SCG), 1-day fatigue group (1FG), 3-day fatigue group (3FG), 7-day fatigue group (7FG), 24-hour recovery group (24RG) or 48-hour recovery group (48RG). The synaptic ultrastructure was observed by transmission electron microscopy. Further a D2DR antagonist and agonist were used to interfere with the autonomic exercise of rats with Open Field Test. RESULTS: We found that: (1) The proportion of asymmetrical synapse of DLS (SCG: 41±2(%), 7FG: 28±4(%), p<0.05), number of presynaptic vesicles (SCG: 13.35±4.58 (number/μm2), 7FG: 5.92±2.7 (number/μm2), p<0.05), number of synaptic terminal mitochondria (SCG: 0.19±0.18 (number/μm2), 7FG: 0.15±0.1 (number/μm2), p<0.05), and the area of synaptic terminals(SCG: 123925.64±54773.42 (μm2), 7FG: 84447.16±29495.42 (μm2), p<0.05), decreased with the increase of exercise days. (2) The total exercise distance of each group decreased gradually with the increase of exercise intensity (p<0.05). Additional rat’s exhaustion time was also significantly shortened after injection of the D2DR antagonist(before: 147.33±5.63, after: 103±4.14, p<0.01). CONCLUSIONS: The activity of synapses in the dorsolateral striatum of rats decreased with the increase of exercise fatigue. Exercise fatigue reduced the autonomic activity of rats, which was further enhanced by D2DR antagonist intervention. This indicates that the regulatory role of D2DR in motion may be related to synaptic plasticity of dopaminergic in substantia nigra and striatum, suggesting that D2DR can be used as an important target to address exercise fatigue.
Endurance training effects on neurotransmitter release in rat striatum: an in vivo microdialysis study

Abstract:In the present study we use the in vivo microdialysis sampling technique to register extracellular levels of neurotransmitters in the striatum of trained and untrained rats. We further evaluate the influence of 1 h of exercise on the striatal release of dopamine (DA), noradrenaline (NA), glutamate (GLU) and gamma-aminobutyric acid (GABA) in trained and untrained rats. Male Wistars were randomly assigned to a training or control group. The exercise training consisted of running on a treadmill for 6 weeks, 5 days week-1, with running time and speed gradually increased from 30 min at 19 m min-1 during the first week to 80 min at 26 m min-1 during the final training week. The animals of the control group were placed on the treadmill twice a week, and received a total of four 'adaptation sessions', in which they exercised 15-45 min at 26 m min-1. Brain dialysates were analysed with microbore liquid chromatography (LC), with electrochemical detection (monoamines and GABA) and fluorescence detection (GLU). Soleus citrate synthase and basal striatal concentrations of DA, NA and GLU were significantly different between the trained and control animals. Sixty minutes of exercise significantly increased extracellular DA, NA and GLU levels in both groups, but there was no statistically significant difference in the exercise-induced increase between trained and control animals. There was no statistical difference in basal or exercise-induced GABA levels between trained and control animals. The results indicate that exercise training appears to result in diminished basal activity of striatal neurotransmitters, while maintaining the necessary sensitivity for responses to acute exercise.

Analysis of trace amino acid neurotransmitters in hypothalamus of rats after exhausting exercise using microdialysis

Abstract:A simple but effective coupling of microdialysis and capillary electrophoresis with laser induced fluorescence detection technique was applied to analysis of amino acid neurotransmitters in the hypothalamus of rats after acute exhausting exercise. The separation of amino acids was achieved using an uncoated fused-silica capillary (57 cm x 75 microm I.D.) with a buffer of 10 mM disodium tetraborate at pH 10 and an applied voltage of 12.5 kV. The detection limit was 10(-10) M for each amino acid. It is sufficiently sensitive and rapid for the determination of amino acids in a 5-microl Microdialysate. In comparison to pre-exercise, a significant increase in the levels of six hypothalamic amino acids (arginine, glycine, lysine, glutamic acid, alanine, gamma-amino-n-butyric acid) was found after exercise. These results demonstrate that the increase of metabolic amino acids in the hypothalamus of rats can be induced by exhausting exercise and suggests that amino acid neurotransmitters may play functional roles in the central effects of exercise.

Abstract:Introduction Dopaminergic, opiod and endocannabinoid neurotransmission are thought to play an important role in the neurobiology of acute exercise and, in particular, in mediating positive affective responses and reward processes. Recent evidence indicates that changes in fractional amplitude of low-frequency fluctuations (zfALFF) in resting-state functional MRI (rs-fMRI) may reflect changes in specific neurotransmitter systems as tested by means of spatial correlation analyses. Methods Here, we investigated this relationship at different exercise intensities in twenty young healthy trained athletes performing low-intensity (LIIE), high-intensity (HIIE) interval exercises, and a control condition on three separate days. Positive And Negative Affect Schedule (PANAS) scores and rs-fMRI were acquired before and after each of the three experimental conditions. Respective zfALFF changes were analyzed using repeated measures ANOVAs. We examined the spatial correspondence of changes in zfALFF before and after training with the available neurotransmitter maps across all voxels and additionally, hypothesis-driven, for neurotransmitter maps implicated in the neurobiology of exercise (dopaminergic, opiodic and endocannabinoid) in specific brain networks associated with “reward” and “emotion.” Results Elevated PANAS Positive Affect was observed after LIIE and HIIE but not after the control condition. HIIE compared to the control condition resulted in differential zfALFF decreases in precuneus, temporo-occipital, midcingulate and frontal regions, thalamus, and cerebellum, whereas differential zfALFF increases were identified in hypothalamus, pituitary, and periaqueductal gray. The spatial alteration patterns in zfALFF during HIIE were positively associated with dopaminergic and μ-opioidergic receptor distributions within the ‘reward' network. Discussion These findings provide new insight into the neurobiology of exercise supporting the importance of reward-related neurotransmission at least during high-intensity physical activity.

Abstract:Abstract Parkinson’s disease (PD) is characterized by a progressive loss of dopaminergic neurons. Exercise has been reported to slow the clinical progression of PD. We evaluated the dopaminergic system of patients with mild and early PD before and after a six-month program of intense exercise. Using 18 F-FE-PE2I PET imaging, we measured dopamine transporter (DAT) availability in the striatum and substantia nigra. Using NM-MRI, we evaluated the neuromelanin content in the substantia nigra. Exercise reversed the expected decrease in DAT availability into a significant increase in both the substantia nigra and putamen. Exercise also reversed the expected decrease in neuromelanin concentration in the substantia nigra into a significant increase. These findings suggest improved functionality in the remaining dopaminergic neurons after exercise. Further research is needed to validate our findings and to pinpoint the source of any true neuromodulatory and neuroprotective effects of exercise in PD in large clinical trials.

Abstract:Physical exercise influences the central dopaminergic, noradrenergic and serotonergic systems. A number of studies have examined brain noradrenaline (norepinephrine), serotonin (5-hydroxytryptamine; 5-HT) and dopamine with exercise. Although there are great discrepancies in experimental protocols, the results indicate that there is evidence in favour of changes in synthesis and metabolism of monoamines during exercise.There is a possibility that the interactions between brain neurotransmitters and their specific receptors could play a role in the onset of fatigue during prolonged exercise. The data on the effects of branched chain amino acid (BCAA) supplementation and ‘central fatigue’ seem to be conflicting, although recent studies suggest that BCAA supplementation has no influence on endurance performance.There are numerous levels at which central neurotransmitters can affect motor behaviour; from sensory perception, and sensory-motor integration, to motor effector mechanisms. However, the crucial point is whether or not the changes in neurotransmitter levels trigger or reflect changes in monoamine release. Until recently most studies were done on homogenised tissue, which gives no indication of the dynamic release of neurotransmitters in the extracellular space of living organisms.Recently, new techniques such as microdialysis and voltammetry were introduced to measure in vivo release of neurotransmitters. Microdialysis can collect virtually any substance from the brain of a freely moving animal with a limited amount of tissue trauma. This method allows measurement of local neurotransmitter release during on-going behavioural changes such as exercise.The results of the first studies using these methods indicate that the release of most neurotransmitters is influenced by exercise. Although the few studies that have been published to date show some discrepancies, we feel that these recently developed and more sophisticated in vivo methods will improve our insight into the relationship between the monoamine and other transmitters during exercise. Continued quantitative and qualitative research needs to be conducted so that a further understanding of the effects of exercise on brain neurotransmission can be gained.

Exercise Fatigue Induce the Oxidative Stress and the Expression of mGluR 4 and mGluR 5 on the Ventrolateral Thalamus in Rats

Abstract:Objective: The motor thalamus plays an important role during exercise. It aims to observe the changes of mGluR4 and mGluR5 in ventrolateral thalamus of rats induced by repeated exhaustive exercise, and to discuss the relationship of mGluR4, mGluR5 and oxidative damage occured during exhaustive exercise. Methods: There were 48 male wistar rats, which were randomly divided into four groups including CG, 0EG, 24EG and 48 EG, each group having 12 rats. Meanwhile, immunohistochemistry (IMM) technique was used to investigate the expression of positive cell and integrated optical density (IOD) of metabotropic glutamate receptor 4 (mGluR4) and metabotropic glutamate receptor 5 (mGluR5), and the impact of exhaustive exercise played on oxidative stress indexes such as malondialdehyde (MDA), glutathione peroxidase (GSH-PX) and superoxide dismutase (SOD) in ventrolateral thalamus of rats were also investigated in different groups. Results: Compared with CG, the expression of mGluR4 and mGluR5 protein of ventrolateral thalamus in 0EG and 24EG both significantly increased, and the value of mGluR4 in 48EG were still higher than control group, however the the value of mGluR5 in 48EG reduced to the rest level of control group. Meanwhile, the SOD activities of 0EG and 24EG group were significantly higher than the control group, and it was the same with GSH content in ventrolateral thalamus of rats. Meanwhile, MDA has been investigated that it increased significantly in 0EG and 24EG compared with control group, and the MDA level of 48EG was still significant higher than CG. Moreover, the indexes of muscle injury such as LD, CK and BUN all increased significantly post-exercise immediately and post-exercise 24 hours .Conclusion: Exercise fatigue could result in up-regulation of mGluR4 and mGluR5 and increace activity of SOD , GSH-PX and MDA in ventrolateral thalamus of rats, and it also induce the muscle injury by increase the level of LD, CK and BUN in serum, which suggested that ventrolateral thalamus was an important brain rigion to modulate the motor function, and mGluR4 and mGluR5 maybe two important receptors prevent from the increase of free radicals and muscle injury induced by exercise fatigue.

Abstract:ABSTRACT We isolated the LIP2 gene from the lipolytic yeastYarrowia lipolytica. It was found to encode a 334-amino-acid precursor protein. The secreted lipase is a 301-amino-acid glycosylated polypeptide which is a member of the triacylglycerol hydrolase family (EC 3.1.1.3 ). The Lip2p precursor protein is processed by the KEX2-like endoprotease encoded by XPR6. Deletion of the XPR6 gene resulted in the secretion of an active but less stable proenzyme. Thus, the pro region does not inhibit lipase secretion and activity. However, it does play an essential role in the production of a stable enzyme. Processing was found to be correct in LIP2A (multipleLIP2 copy integrant)-overexpressing strains, which secreted 100 times more activity than the wild type, demonstrating thatXPR6 maturation was not limiting. No extracellular lipase activity was detected with the lip2 knockout (KO) strain, strongly suggesting that extracellular lipase activity results from expression of the LIP2 gene. Nevertheless, thelip2 KO strain is still able to grow on triglycerides, suggesting an alternative pathway for triglyceride utilization inY. lipolytica.

Abstract:Peripheral physiological adaptations to exercise have been the subject of several studies. To adjust to the disturbance in resting homeostasis induced by the exercise stimulus, a number of regulatory systems are called upon to return the body to a new level of homeostasis. In contrast with our knowledge about the peripheral adaptations to exercise, studies relating exercise to brain neurotransmitter levels are scarce. It is of interest to examine the effect of acute and chronic exercise on neurotransmitter release, since movement initiation and control of locomotion have been shown to be related to striatal neurotransmitter function. One of the possible therapeutic modalities in movement and mental disorders is exercise therapy. But also chronic adaptations to training and disturbances caused by overtraining might be reflected in brain functioning. Physical exercise influences the central dopaminergic, noradrenergic, and serotonergic systems. A number of studies have examined brain noradrenaline (NA), serotonin (5-hydroxytryptamine, or 5-HT), and dopamine (DA) with exercise. Although there are great discrepancies in experimental protocols, the results indicate that there is evidence in favor of changes in synthesis and metabolism of monoamines during exercise. In previous studies, we used the microdialysis technique to monitor the release of neurotransmitters in different brain nuclei during exercise. We have shown that DA, NA, and 5-HT release is increased during exercise. We also showed that acute exercise and/or exercise training influences extracellular DA, NA, GLU, and GABA levels. Taken together this could indicate that chronic disturbance of homeostasis could also influence transmitter release and that this paradigm could serve as a tool to explore the central effects of overtraining.

Abstract:Treadmill exercise and neuromuscular electrical stimulation are common clinical approaches for stroke rehabilitation. Both animal and clinical studies have shown the functional improvements after these interventions. However, the neurochemical effects on the ischemic brain had not been well studied. This study aimed at evaluating the effects of treadmill exercise and neuromuscular electrical stimulation (NMES), and studying their effects during a 2-week training, on the levels of common neurotransmitters (aspartate, glutamate, taurine and gamma-aminobutyric acid (GABA)) in the hippocampus following transient focal cerebral ischemia. Either treadmill exercise or neuromuscular electrical stimulation was prescribed to the rats 24h after cerebral ischemia whereas Control group remained in cages for 2 weeks. Microdialysis technique was used to collect dialysates from ipsilesional hippocampus in vivo. It was found that the glutamate level was increased significantly during treadmill exercise and then returned to baseline level. Both interventions did not trigger significant effects on aspartate and glutamate basal levels during the 2 weeks. The relatively high taurine level in Control groups may suggest that the interventions might suppress the taurine release in hippocampus. GABA and aspartate levels did not showed significant changes over the 2 weeks in all groups. Abstract:The asymmetric synthesis of functionalized nitrocyclopropanes has been achieved by a one-pot, four-step method catalyzed by (S)-diphenylprolinol TMS ether, which joins two sequential domino reactions, namely a domino sulfa-Michael/aldol condensation of α,β-unsaturated aldehydes with 1,4-dithiane-2,5-diol, and a domino Michael/α-alkylation reaction of the derived chiral dihydrothiophenes with bromonitromethane. The title compounds were obtained in 27-45% yields, with high levels of diastereoselectivity (93:7 to 100:0 dr) and generally good enantioselectivities (up to 95:5 er).

Abstract:AIMS: This study aimed to test the hypothesis that different neurotransmitters and hormones are presented at exercise fatigue in hot temperatures with differing relative humidities (RH).MAIN METHODS: Eight trained male athletes performed a graded maximum oxygen consumption (VO2max) test in five different environmental conditions, namely, 21°C/20% RH (Normal), 33°C/20% RH (Hot 20%), 33°C/40% RH (Hot 40%), 33°C/60% RH (Hot 60%), and 33°C/80% RH (Hot 80%). Blood samples were taken pre- and post-exercise and analyzed for noradrenaline (NA), adrenaline (ADR), dopamine (DA), serotonin (5-HT), 5-hydroxyindoleacetic acid (5-HIAA), and prolactin (PRL). Weight and oral and skin temperatures were recorded pre- and post-exercise. Heart rate was continuously monitored throughout the exercise.KEY FINDINGS: Hot 20%, Hot 40%, and Hot 80% had lower VO2max levels compared with Normal (P<0.05). The data pooled from all five conditions indicated that the NA (P<0.0001), PRL (P<0.0001), 5-HT (P=0.002), 5-HIAA (P=0.029), and DA (P=0.016) levels were affected by exercise. Accordingly, NA level was significantly associated with performance time. However, ADR did not show any significant effect between pre- and post-exercise (P=0.187).SIGNIFICANCE: The maximal aerobic capacity was impaired in high temperature and most humidity conditions. NA was strongly associated with exercise time, which suggested that exercise duration and intensity had an important influence on NA neurotransmitter level irrespective of the environmental conditions.

Abstract:The arbuscular mycorrhizal (AM) symbiosis, which forms between plant hosts and ubiquitous soil fungi of the phylum Glomeromycota, plays a key role for the nutrient uptake of the majority of land plants, including many economically important crop species. AM fungi take up nutrients from the soil and exchange them for photosynthetically fixed carbon from the host. While our understanding of the exact mechanisms controlling carbon and nutrient exchange is still limited, we recently demonstrated that (i) carbon acts as an important trigger for fungal N uptake and transport, (ii) the fungus changes its strategy in response to an exogenous supply of carbon, and that (iii) both plants and fungi reciprocally reward resources to those partners providing more benefit. Here, we summarize recent research findings and discuss the implications of these results for fungal and plant control of resource exchange in the AM symbiosis.
A test of the catecholamines hypothesis for an acute exercise-cognition interaction

Abstract:The purpose of the study was to examine the usage of norepinephrine (NE) and dopamine (DA) in the brain when exercising while simultaneously undertaking cognitive tests. Plasma concentrations of the NE metabolite 3-methoxy 4-hydroxyphenylglycol (MHPG) and the DA metabolite homovanillic acid (HVA) showed a linear increase from rest to exercising at 40% and 80% maximum power output (W.max) while simultaneously undertaking cognitive tasks (random number generation (RNG) and response time). Delta plasma concentrations of MHPG and HVA at each exercise intensity while undertaking cognitive tasks and while exercising without cognitive tasks did not differ. Taking blood samples at 0, 1, 3, and 5 min following cessation of exercise did not affect results. Regression correlations showed that delta MHPG and HVA plasma concentrations at the 1 and 3 min sampling times were strong predictors of delta RNG, response time and movement time. Reaction time at 80% W.max significantly increased, while movement time at 80% W.max significantly decreased. It was concluded that these results provide no support for a direct effect of increased catecholamines concentrations on cognitive performance during exercise. The regression data suggest that there is some relationship between exercise, catecholamines concentrations and cognition.
