Abstract:The discovery of neuronal intrinsic plasticity (IP) processes which persistently modify a neuron's excitability necessitates a new concept of the neuronal plasticity mechanism and may profoundly influence our ideas on learning and memory. In this paper, we propose a spike-based IP model/adaptation rule for an integrate-and-fire (IF) neuron to model this biological phenomenon. By utilizing spikes denoted by Dirac delta functions rather than computing instantaneous firing rates for the time-dependent stimulus, this simple adaptation rule adjusts two parameters of an individual IF neuron to modify its excitability. As a result, this adaptation rule helps an IF neuron to keep its firing activity in a relatively “low but not too low” level and makes the spike-count distributions computed with adjusted window sizes similar to the experimental results.

Abstract:The phenomenon in which the response of a neuronal network to a weak signal is significantly enhanced in moderate noise is known as stochastic resonance (SR). Most of the previous studies on the transmission of signals by networks have been based on static synaptic connections, whereas dynamic synaptic connections modified by spike-time-dependent plasticity (STDP) are the basis of learning and memory in the nervous system. In this paper, we explore the phenomenon of SR in a neuronal network consisting of different ratios of excitatory vertebral neurons and inhibitory interneurons. The equivalent circuit method was employed to assess the average energy efficiency of the network. The differences in signal response before and after the introduction of STDP were compared for purely excitatory, purely inhibitory and excitatory-inhibitory networks, respectively. It was found that excitatory STDP promotes the network's response to weak signals, while inhibitory STDP has the opposite effect. The introduction of the inhibitory STDP makes the inhibitory network insensitive to the modulation of the coupling strength and increases its robustness. Furthermore, in the excitatory-inhibitory network, we found that STDP had little effect on the overall signalling of the network, and that the network's response to weak signals was more stable. Our findings contribute to the understanding of the importance of excitatory-inhibitory balance in ensuring accurate transmission and processing of information and provide new insights into the role of STDP in neuronal information processing.

Abstract:Learning and memory in the brain are implemented by complex, time-varying changes in neural circuitry. The computational rules according to which synaptic weights change over time are the subject of much research, and are not precisely understood. Until recently, limitations in experimental methods have made it challenging to test hypotheses about synaptic plasticity on a large scale. However, as such data become available and these barriers are lifted, it becomes necessary to develop analysis techniques to validate plasticity models. Here, we present a highly extensible framework for modeling arbitrary synaptic plasticity rules on spike train data in populations of interconnected neurons. We treat synaptic weights as a (potentially nonlinear) dynamical system embedded in a fully-Bayesian generalized linear model (GLM). In addition, we provide an algorithm for inferring synaptic weight trajectories alongside the parameters of the GLM and of the learning rules. Using this method, we perform model comparison of two proposed variants of the well-known spike-timing-dependent plasticity (STDP) rule, where nonlinear effects play a substantial role. On synthetic data generated from the biophysical simulator NEURON, we show that we can recover the weight trajectories, the pattern of connectivity, and the underlying learning rules.

Abstract:Spike-timing-dependent plasticity (STDP) has attracted considerable experimental and theoretical attention over the last decade. In the most basic formulation, STDP provides a fundamental unit - a spike pair - for quantifying the induction of long-term changes in synaptic strength. However, many factors, both pre- and postsynaptic, can affect synaptic transmission and integration, especially when multiple spikes are considered. Here we review the experimental evidence for multiple types of nonlinear temporal interactions in STDP, focusing on the contributions of individual spike pairs, overall spike rate, and precise spike timing for modification of cortical and hippocampal excitatory synapses. We discuss the underlying processes that determine the specific learning rules at different synapses, such as postsynaptic excitability and short-term depression. Finally, we describe the success of efforts toward building predictive, quantitative models of how complex and natural spike trains induce long-term synaptic modifications.

Abstract:Short-term plasticity (STP) is important for understanding how neuronal circuits can perform different computations. The STP of a neuron pair can be measured directly using paired whole-cell recordings. Besides, the cross-correlation between the presynaptic and postsynaptic neuronal firing is usually used as a proxy for estimating the synaptic properties. However, the relationships between the synaptic inputs and the spiking properties of the postsynaptic neurons during the STP still remain unclear. Here, we characterized the STP of both synaptic input, measured by the postsynaptic field potential (PFP), and spike transmission at the retinocollicular pathway of mice. We found that the STP of the retinocollicular pathway is mainly facilitating, where the second presynaptic spike induces a larger PFP and higher postsynaptic firing rate than the first presynaptic spike. The facilitation in the postsynaptic firing rate is generally larger than the PFP facilitation. Interestingly, the last postsynaptic spike timing also has a large facilitating effect on the postsynaptic spiking upon receiving a presynaptic input spike. However, the PFP does not depend on the last postsynaptic spike timing, suggesting that there is an input-independent component of spike transmission in STP. Overall, our results indicate that the STP of the retinocollicular pathway is likely a two-stage process, where the spiking plasticity of the postsynaptic neuron could be independent of its inputs.

Abstract:Transcranial brain stimulation and evidence of ephaptic coupling have recently sparked strong interests in understanding the effects of weak electric fields on the dynamics of brain networks and of coupled populations of neurons. The collective dynamics of large neuronal populations can be efficiently studied using single-compartment (point) model neurons of the integrate-and-fire (IF) type as their elements. These models, however, lack the dendritic morphology required to biophysically describe the effect of an extracellular electric field on the neuronal membrane voltage. Here, we extend the IF point neuron models to accurately reflect morphology dependent electric field effects extracted from a canonical spatial "ball-and-stick" (BS) neuron model. Even in the absence of an extracellular field, neuronal morphology by itself strongly affects the cellular response properties. We, therefore, derive additional components for leaky and nonlinear IF neuron models to reproduce the subthreshold voltage and spiking dynamics of the BS model exposed to both fluctuating somatic and dendritic inputs and an extracellular electric field. We show that an oscillatory electric field causes spike rate resonance, or equivalently, pronounced spike to field coherence. Its resonance frequency depends on the location of the synaptic background inputs. For somatic inputs the resonance appears in the beta and gamma frequency range, whereas for distal dendritic inputs it is shifted to even higher frequencies. Irrespective of an external electric field, the presence of a dendritic cable attenuates the subthreshold response at the soma to slowly-varying somatic inputs while implementing a low-pass filter for distal dendritic inputs. Our point neuron model extension is straightforward to implement and is computationally much more efficient compared to the original BS model. It is well suited for studying the dynamics of large populations of neurons with heterogeneous dendritic morphology with (and without) the influence of weak external electric fields.

Abstract:The precise timing of neuronal spikes may lead to changes in synaptic connectivity and is thought to be crucial for learning and memory. However, the effect of spike timing on neuronal connectivity in the intact brain remains unknown. Using closed-loop optogenetic stimulation in CA1 of freely moving mice, we generated unique spike patterns between presynaptic pyramidal cells (PYRs) and postsynaptic parvalbumin (PV)–immunoreactive cells. The stimulation led to spike transmission changes that occurred together across all presynaptic PYRs connected to the same postsynaptic PV cell. The precise timing of all presynaptic and postsynaptic cell spikes affected transmission changes. These findings reveal an unexpected plasticity mechanism, in which the spike timing of an entire cell assembly has a more substantial impact on effective connectivity than that of individual cell pairs.

Abstract:Dendrites on neurons support nonlinear electrical excitations, but the computational significance of these events is not well understood. We developed molecular, optical, and analytical tools to map sub-millisecond voltage dynamics throughout the dendritic trees of CA1 pyramidal neurons under diverse optogenetic and synaptic stimulus patterns, in acute brain slices. We observed history-dependent spike back-propagation in distal dendrites, driven by locally generated Na+ spikes (dSpikes). Dendritic depolarization created a transient window for dSpike propagation, opened by A-type KV channel inactivation, and closed by slow NaV inactivation. Collisions of dSpikes with synaptic inputs triggered calcium channel and N-methyl-D-aspartate receptor (NMDAR)-dependent plateau potentials, with accompanying complex spikes at the soma. This hierarchical ion channel network acts as a spike-rate accelerometer, providing an intuitive picture of how dendritic excitations shape associative plasticity rules.

Abstract:In this work, we propose time-integrated spike-timing-dependent plasticity (TI-STDP), a mathematical model of synaptic plasticity that allows spiking neural networks to continuously adapt to sensory input streams in an unsupervised fashion. Notably, we theoretically establish and formally prove key properties related to the synaptic adjustment mechanics that underwrite TI-STDP. Empirically, we demonstrate the efficacy of TI-STDP in simulations of jointly learning deeper spiking neural networks that process input digit pixel patterns, at both full image and patch-levels, comparing to two powerful historical instantations of STDP; trace-based STDP (TR-STDP) and event-based post-synaptic STDP (EV-STDP). Usefully, we demonstrate that not only are all forms of STDP capable of meaningfully adapting the synaptic efficacies of a multi-layer biophysical architecture, but that TI-STDP is notably able to do so without requiring the tracking of a large window of pre- and post-synaptic spike timings, the maintenance of additional parameterized traces, or the restriction of synaptic plasticity changes to occur within very narrow windows of time. This means that our findings show that TI-STDP can efficiently embody the benefits of models such as canonical STDP, TR-STDP, and EV-STDP without their costs or drawbacks. Usefully, our results further demonstrate the promise of using a spike-correlation scheme such as TI-STDP in conducting credit assignment in discrete pulse-based neuromorphic models, particularly those than acquire a lower-level distributed representation jointly with an upper-level, more abstract representation that self-organizes to cluster based on inherent cross-pattern similarities. We further demonstrate TI-STDP's effectiveness in adapting a simple neuronal circuit that learns a simple bi-level, part-whole hierarchy from sensory input patterns.

Abstract:Neural variability plays a central role in neural coding and neuronal network dynamics. Unreliability of synaptic transmission is a major source of neural variability: synaptic neurotransmitter vesicles are released probabilistically in response to presynaptic spikes and are recovered stochastically in time. The stochastic dynamics of this process interacts with variability in the arrival times of presynaptic spikes to shape the variability of the postsynaptic response. We use continuous time Markov chain methods to analyze a model of short term synaptic depression with stochastic vesicle dynamics coupled with three different models of presynaptic spiking: one model in which the timing of presynaptic spikes are modeled as a Poisson process, one in which spikes occur more regularly than a Poisson process and one in which spikes occur more irregularly. We use this analysis to investigate how variability in a presynaptic spike train is transformed by short term synaptic depression and stochastic vesicle dynamics to determine the variability of the postsynaptic response to a presynaptic spike train. We find that regular presynaptic spiking increases the average rate at which vesicles are released, that the number of vesicles released over a time window is more variable for smaller time windows than for larger time windows, and that fast presynaptic spiking gives rise to Poisson-like variability of the postsynaptic response even when presynaptic spike times are highly non-Poisson. Our results complement and extend previously reported theoretical results and provide possible explanations for some trends observed in recorded data.
Dynamics of spike transmission and suppression between principal cells and interneurons in the hippocampus and entorhinal cortex

Abstract:Synaptic excitation and inhibition are essential for neuronal communication. However, the variables that regulate synaptic excitation and inhibition in the intact brain remain largely unknown. Here, we examined how spike transmission and suppression between principal cells (PCs) and interneurons (INTs) are modulated by activity history, brain state, cell type, and somatic distance between presynaptic and postsynaptic neurons by applying cross‐correlogram analyses to datasets recorded from the dorsal hippocampus and medial entorhinal cortex (MEC) of 11 male behaving and sleeping Long Evans rats. The strength, temporal delay, and brain‐state dependency of the spike transmission and suppression depended on the subregions/layers. The spike transmission probability of PC–INT excitatory pairs that showed short‐term depression versus short‐term facilitation was higher in CA1 and lower in CA3. Likewise, the intersomatic distance affected the proportion of PC–INT excitatory pairs that showed short‐term depression and facilitation in the opposite manner in CA1 compared with CA3. The time constant of depression was longer, while that of facilitation was shorter in MEC than in CA1 and CA3. During sharp‐wave ripples, spike transmission showed a larger gain in the MEC than in CA1 and CA3. The intersomatic distance affected the spike transmission gain during sharp‐wave ripples differently in CA1 versus CA3. A subgroup of MEC layer 3 (EC3) INTs preferentially received excitatory inputs from and inhibited MEC layer 2 (EC2) PCs. The EC2 PC–EC3 INT excitatory pairs, most of which showed short‐term depression, exhibited higher spike transmission probabilities than the EC2 PC–EC2 INT and EC3 PC–EC3 INT excitatory pairs. EC2 putative stellate cells exhibited stronger spike transmission to and received weaker spike suppression from EC3 INTs than EC2 putative pyramidal cells. This study provides detailed comparisons of monosynaptic interaction dynamics in the hippocampal–entorhinal loop, which may help to elucidate circuit operations.

Abstract:Short-term changes in efficacy have been postulated to enhance the ability of synapses to transmit information between neurons, and within neuronal networks. Even at the level of connections between single neurons, direct confirmation of this simple conjecture has proven elusive. By combining paired-cell recordings, realistic synaptic modeling, and information theory, we provide evidence that short-term plasticity can not only improve, but also reduce information transfer between neurons. We focus on a concrete example in rat neocortex, but our results may generalize to other systems. When information is contained in the timings of individual spikes, we find that facilitation, depression, and recovery affect information transmission in proportion to their impacts upon the probability of neurotransmitter release. When information is instead conveyed by mean spike rate only, the influences of short-term plasticity critically depend on the range of spike frequencies that the target network can distinguish (its effective dynamic range). Our results suggest that to efficiently transmit information, the brain must match synaptic type, coding strategy, and network connectivity during development and behavior.

A causal inference approach of monosynapses from spike trains

Abstract:Neuroscientists have worked on the problem of estimating synaptic properties, such as connectivity and strength, from simultaneously recorded spike trains since the 1960s. Recent years have seen renewed interest in the problem, coinciding with rapid advances in the technology of high-density neural recordings and optogenetics, which can be used to calibrate causal hypotheses about functional connectivity. Here, a rigorous causal inference framework for pairwise excitatory and inhibitory monosynaptic effects between spike trains is developed. Causal interactions are identified by separating spike interactions in pairwise spike trains by their timescales. Fast algorithms for computing accurate estimates of associated quantities are also developed. Through the lens of this framework, the link between biophysical parameters and statistical definitions of causality between spike trains is examined across a spectrum of dynamical systems simulations. In an idealized setting, we demonstrate a correspondence between the synaptic causal metric developed here and the probabilities of causation developed by Tian and Pearl. Since the probabilities of causation are derived under distinct assumptions and include data from experimental randomization, this opens up the possibility of testing the synaptic inference framework's assumptions with juxtacellular or optogenetic stimulation. We simulate such an experiment with a biophysically detailed channelrhodopsin model and show that randomization is not achieved; strong confounding persists even with strong stimulations. A principal goal is to ask how carefully articulated causal assumptions might better inform the design of neural stimulation experiments and, in turn, support experimental tests of those assumptions.

Abstract:Abstract Coarse-graining microscopic models of biological neural networks to obtain mesoscopic models of neural activities is an essential step towards multi-scale models of the brain. Here, we extend a recent theory for mesoscopic population dynamics with static synapses to the case of dynamic synapses exhibiting short-term plasticity (STP). The extended theory offers an approximate mean-field dynamics for the synaptic input currents arising from populations of spiking neurons and synapses undergoing Tsodyks–Markram STP. The approximate mean-field dynamics accounts for both finite number of synapses and correlation between the two synaptic variables of the model (utilization and available resources) and its numerical implementation is simple. Comparisons with Monte Carlo simulations of the microscopic model show that in both feedforward and recurrent networks, the mesoscopic mean-field model accurately reproduces the first- and second-order statistics of the total synaptic input into a postsynaptic neuron and accounts for stochastic switches between Up and Down states and for population spikes. The extended mesoscopic population theory of spiking neural networks with STP may be useful for a systematic reduction of detailed biophysical models of cortical microcircuits to numerically efficient and mathematically tractable mean-field models.

Abstract:The release of neurotransmitters from synapses obeys complex and stochastic dynamics. Depending on the recent history of synaptic activation, many synapses depress the probability of releasing more neurotransmitter, which is known as synaptic depression. Our understanding of how synaptic depression affects the information efficacy, however, is limited. Here we propose a mathematically tractable model of both synchronous spike-evoked release and asynchronous release that permits us to quantify the information conveyed by a synapse. The model transits between discrete states of a communication channel, with the present state depending on many past time steps, emulating the gradual depression and exponential recovery of the synapse. Asynchronous and spontaneous releases play a critical role in shaping the information efficacy of the synapse. We prove that depression can enhance both the information rate and the information rate per unit energy expended, provided that synchronous spike-evoked release depresses less (or recovers faster) than asynchronous release. Furthermore, we explore the theoretical implications of short-term synaptic depression adapting on longer time scales, as part of the phenomenon of metaplasticity. In particular, we show that a synapse can adjust its energy expenditure by changing the dynamics of short-term synaptic depression without affecting the net information conveyed by each successful release. Moreover, the optimal input spike rate is independent of the amplitude or time constant of synaptic depression. We analyze the information efficacy of three types of synapses for which the short-term dynamics of both synchronous and asynchronous release have been experimentally measured. In hippocampal autaptic synapses, the persistence of asynchronous release during depression cannot compensate for the reduction of synchronous release, so that the rate of information transmission declines with synaptic depression. In the calyx of Held, the information rate per release remains constant despite large variations in the measured asynchronous release rate. Lastly, we show that dopamine, by controlling asynchronous release in corticostriatal synapses, increases the synaptic information efficacy in nucleus accumbens.

Abstract:Cortical neurons are subject to sustained and irregular synaptic activity which causes important fluctuations of the membrane potential (Vm). We review here different methods to characterize this activity and its impact on spike generation. The simplified, fluctuating point-conductance model of synaptic activity provides the starting point of a variety of methods for the analysis of intracellular Vm recordings. In this model, the synaptic excitatory and inhibitory conductances are described by Gaussian-distributed stochastic variables, or colored conductance noise. The matching of experimentally recorded Vm distributions to an invertible theoretical expression derived from the model allows the extraction of parameters characterizing the synaptic conductance distributions. This analysis can be complemented by the matching of experimental Vm power spectral densities (PSDs) to a theoretical template, even though the unexpected scaling properties of experimental PSDs limit the precision of this latter approach. Building on this stochastic characterization of synaptic activity, we also propose methods to qualitatively and quantitatively evaluate spike-triggered averages of synaptic time-courses preceding spikes. This analysis points to an essential role for synaptic conductance variance in determining spike times. The presented methods are evaluated using controlled conductance injection in cortical neurons in vitro with the dynamic-clamp technique. We review their applications to the analysis of in vivo intracellular recordings in cat association cortex, which suggest a predominant role for inhibition in determining both sub- and supra-threshold dynamics of cortical neurons embedded in active networks.

Abstract:In neural systems, synaptic plasticity is usually driven by spike trains. Due to the inherent noises of neurons, synapses and networks, spike trains typically exhibit externally uncontrollable variability such as spatial heterogeneity and temporal stochasticity, resulting in variability of synapses, which we call efficacy variability. Spike patterns with the same population rate but inducing different efficacy variability may result in neuronal networks with sharply different structures and functions. However, how the variability of spike trains influences the efficacy variability remains unclear. Here, we systematically study this influence when spike patterns possess four aspects of statistical features, i.e. synchronous firing, auto-temporal structure, heterogeneity of rates and heterogeneity of cross-correlations, under spike-timing dependent plasticity (STDP) after dynamically bounding the mean strength of plastic synapses into or out of a neuron (synaptic homeostasis). We then show the functional importance of efficacy variability on the encoding and maintenance of connection patterns and on the early development of primary visual systems driven by retinal waves. We anticipate our work brings a fresh perspective to the understanding of the interaction between synaptic plasticity and dynamical spike patterns in functional processes of neural systems.
