Abstract:Oligomer of styrene and maleic anhydride synthesized by bulk RAFT polymerization (SMA-RAFT) was used to construct a novel strategy for robust nanoencapsulation via interfacially confined controlled/living radical miniemulsion polymerization. After ammonolysis, SMA-RAFT becomes amphiphilic and can be used as a surfactant to prepare miniemulsion. The ammonolyzed SMA-RAFT molecules would self-assemble on the interface of water/droplets. This self-assembly property combining with the RAFT living polymerization chemistry demands the polymer chains to grow inwards gradually in particles, leading to the formation of a polymer shell. The hydrophilicity of ammonolyzed SMA-RAFT agent tuned by the ammonolyzed degree or structures of SMA-RAFT agent was found to play a key role in the final morphology. The well-defined nanocapsules with little solid particles can be obtained by using partially ammonolyzed SMA-RAFT with 0.5wt% SDS as a co-surfactant.

Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.

Abstract:The crystallization-driven self-assembly of polymers based on semicrystalline poly(epsilon-caprolactone) cores is currently an area of high interest on account of their well-known biocompatibility and biodegradability, yet a comprehensive understanding of coil-crystalline-coil type triblock copolymer assembly behavior with respect to this core chemistry is yet to be realized. Herein, we demonstrate the simple preparation of well-defined tuneable 1D and 2D structures based on poly(epsilon-caprolactone) (PCL) triblock copolymers of different block ratios synthesized by ring-opening polymerization (ROP) and reversible addition-fragmentation chain transfer (RAFT) polymerization. In this report, the assembly of PCL-based amphiphiles in various solvents was investigated to tune the morphology and size of the assemblies with well-defined 2D platelets and long cylinders produced when using long soluble coronal blocks or under good solvent conditions. By contrast, truncated short fibers were obtained for less soluble PCL-containing block copolymers or under poor solubility conditions for the core block as a consequence of the increasing amount of nuclei formed in the crystallization process. Furthermore, the length of PCL-based 1D nanostructures could be controlled by tuning self-assembly conditions where the micelles' lengths varied from 93 to 1200 nm with narrow dispersities. This easy assembly methodology greatly simplifies the lengthy procedure required to prepare biodegradable 1D and 2D nanostructures from PCL with tuneable sizes, which demonstrate great potential as drug-delivery vehicles in the realm of biomedicine.

Abstract:The synthesis of temperature responsive nanoencapsulation of inorganic salts through reversible addition-fragmentation transfer (RAFT) interfacial confined inverse miniemulsion polymerization was reported by using a poly(ethylene oxide)-MacroRAFT (PEO-RAFT) agent. It was found that the nanocapsules (NC) could be applied in medical diagnose or pharmaceuticals therapy because of the thermo-sensitive polymer shell that can release the core contents of the NCs around the body temperature. It was also found that as the polymerization was a radical living/controlled polymerization confined in the interface, the polymer shell could be functionally modified via post reaction. The results show that the strength of the polymer shell can be increased by cross-linking polymerization to obtain rigid hollow NCs.
Transforming the Stability, Encapsulation, and Sustained Release Properties of Calcium Alginate Beads through Gel-Confined Coacervation

Abstract:Calcium alginate (Ca2+/alginate) gel beads find use in diverse applications, ranging from drug delivery and tissue engineering to bioprocessing, food formulation, and agriculture. Unless modified, however, these gels have limited stability in alkaline media (including phosphate buffers), and their high solute permeability limits their ability to efficiently encapsulate and slowly release water-soluble small molecules. Here, we show how these limitations can be addressed by mixing the alginate solutions used in the bead preparation with the nontoxic anionic polymer polyphosphate (PP). Upon complexing Ca2+ ions, PP undergoes complex coacervation (i.e., liquid/liquid phase separation into a Ca2+/PP-rich coacervate phase and a dilute supernatant phase). At lower PP concentrations, the Ca2+/PP coacervate appears to simply remain dispersed within the beads. Though its presence makes the beads more stable in alkaline media (phosphate-buffered saline and seawater), it has little impact on the bead stiffness, morphology, and (at least in the absence of substantial payload/coacervate association) encapsulation and release properties. When the PP concentrations exceed a critical value, however, Ca2+/PP coacervation within the gelling Ca2+/alginate beads collapses the resulting beads into more compact, interpenetrating polymer networks. Besides their enhanced stability to alkaline environments, these hybrid beads exhibit irregular morphologies with wrinkled and dimpled surface structures and macroscopic (closed) internal pores, and their collapse into these polymer-rich networks also makes them significantly stiffer than their PP-free counterparts. Crucially, these beads also exhibit a much lower solute permeability, which enables highly efficient encapsulation and multiday release of water-soluble small molecules (with the beads encapsulating >90% of the added model payload and sustaining its release over 3-5 d). Collectively, these findings provide a mild and simple (single-step) pathway to generating ionically cross-linked alginate beads with significantly enhanced stability, encapsulation efficiency, and sustained release.

Abstract:Biodegradable tri-component graft copolymers, chitosan-poly(epsilon-caprolactone)-poly(ethylene glycol) (CPP), were synthesized via a mild route, using sodium dodecyl sulfate-chitosan complex (SCC) as a precursor. Both PCL and PEG could be conveniently conjugated to the hydroxyl sites of chitosan without the need of tedious chemical protection/deprotection processes, thereby leaving the amino groups of chitosan intact. The self-assembly and release behavior of the copolymer micelles were investigated. Paclitaxel and rutin were used as model drugs. Spherical micelles could be formed through self-assembly of CPP in aqueous media. The micelle diameter increased with PEGylation degree and ranged from 30 to 45 nm. The incorporation of drugs into the micelles significantly raised the micelle diameter and diversified the micelle morphologies. The micelles were further subjected to glutaraldehyde treatment to prolong the release of the incorporated drugs. It was found that the crosslinking process shrunk the drug-loaded micelles. In addition, the micelles were endowed with self-luminescent properties after crosslinked with glutaraldehyde. Abstract:To improve the drug-loading capacity and control the initial release of amphiphilic drug carriers, a series of N-phthaloyl-chitosan-g-(PEO-PLA-PEO) compounds were synthesized with well-defined structures. The self-assembly behavior of copolymers in aqueous solutions was confirmed by various techniques such as fluorescence spectrometry, dynamic light scattering, and transmission electron microscopy. The results demonstrated that the micellization behavior of graft copolymers was different from that of their linear counterparts. The micelle sizes of the graft copolymers could be tuned with chemical composition as well as temperature. Furthermore, when hydrophobic indomethacin was loaded into the micelles, the graft copolymer micelles trapped more indomethacin than PEO-PLA-PEO micelles, facilitating the in vitro control of the initial burst release of the drug. Drug release could be controlled in vitro by simply altering the EO/LA ratio of the grafting chains. The graft copolymer showed low cytotoxicity to 293T cells, indicating its great potential application for drug delivery.

Abstract:The hydrophobic modification of sodium alginate was realized by direct grafting with glycolic acid. The products, polyglycolic acid grafting from sodium alginate (Alg-g-PGA), self-assembled into nanoparticles of 200–250 nm in diameter through the interaction of hydrophobic PGA segments in the aqueous solution. Alg-g-PGA nanoparticles were cross-linked into hydrogel microspheres by calcium chloride, which was used as drug delivery vehicles for controlled release. The results of drug (ibuprofen) release model exhibited high drug loading rate and prolonged drug release speed of the Alg-g-PGA gel microspheres. The one-step method of synthesizing hydrophobically modified sodium alginate processes simple operation, mild condition, and low-cost raw material, resulting in its wide application of biological medicine in the future.

Abstract:Alginate is a natural biopolymer widely studied for pharmaceutical applications due to its biocompatibility, low toxicity, and mild gelation abilities. This review summarizes recent advances in alginate-based encapsulation systems for targeted drug delivery. Alginate formulations like microparticles, nanoparticles, microgels, and composites fabricated by methods including ionic gelation, emulsification, spray drying, and freeze drying enable tailored drug loading, enhanced stability, and sustained release kinetics. Alginate microspheres prepared by spray drying or ionic gelation provide gastric protection and colon-targeted release of orally delivered drugs. Alginate nanoparticles exhibit enhanced cellular uptake and tumor-targeting capabilities through the enhanced permeation and retention effect. Crosslinked alginate microgels allow high drug loading and controlled release profiles. Composite alginate gels with cellulose, chitosan, or inorganic nanomaterials display improved mechanical properties, mucoadhesion, and tunable release kinetics. Alginate-based wound dressings containing antimicrobial nanoparticles promote healing of burns and chronic wounds through sustained topical delivery. Although alginate is well-established as a pharmaceutical excipient, more extensive in vivo testing is needed to assess clinical safety and efficacy of emerging formulations prior to human trials. Future opportunities include engineered systems combining stimuli-responsiveness, active targeting, and diagnostic capabilities. In summary, this review discusses recent advances in alginate encapsulation techniques for oral, transdermal, and intravenous delivery, with an emphasis on approaches enabling targeted and sustained drug release for enhanced therapeutic outcomes.

Esterification of Alginate with Alkyl Bromides of Different Carbon Chain Lengths via the Bimolecular Nucleophilic Substitution Reaction: Synthesis, Characterization, and Controlled Release Performance

Abstract:To extend the alginate applicability for the sustained release of hydrophobic medicine in drug delivery systems, the alkyl alginate ester derivative (AAD), including hexyl alginate ester derivative (HAD), octyl alginate ester derivative (OAD), decyl alginate ester derivative (DAD), and lauryl alginate ester derivative (LAD), were synthesized using the alkyl bromides with different lengths of carbon chain as the hydrophobic modifiers under homogeneous conditions via the bimolecular nucleophilic substitution (SN2) reaction. Experimental results revealed that the successful grafting of the hydrophobic alkyl groups onto the alginate molecular backbone via the SN2 reaction had weakened and destroyed the intramolecular hydrogen bonds, thus enhancing the molecular flexibility of the alginate, which endowed the AAD with a good amphiphilic property and a critical aggregation concentration (CAC) of 0.48~0.0068 g/L. Therefore, the resultant AAD could form stable spherical self-aggregated micelles with the average hydrodynamic diameter of 285.3~180.5 nm and zeta potential at approximately −44.8~−34.4 mV due to the intra or intermolecular hydrophobic associations. With the increase of the carbon chain length of the hydrophobic side groups, the AAD was more prone to self-aggregation, and therefore was able to achieve the loading and sustained release of hydrophobic ibuprofen. Additionally, the swelling and degradation of AAD microcapsules and the diffusion of the loaded drug jointly controlled the release rate of ibuprofen. Meanwhile, the AAD also displayed low cytotoxicity to the murine macrophage RAW264.7 cells. Thanks to the good amphiphilic property, colloidal interface activity, hydrophobic drug-loading performance, and cytocompatibility, the synthesized AAD exhibited a great potential for the development of hydrophobic pharmaceutical formulations.

Abstract:Novel calcium-alginate (Ca-alginate) capsules with aqueous core and thermo-responsive membrane are successfully prepared by introducing a co-extrusion minifluidic approach, and the thermo-responsive gating characteristics of Ca-alginate capsule membranes embedded with poly(N-isopropylacrylamide) (PNIPAM) microspheres are investigated systematically. The experimental results show that the prepared Ca-alginate capsules are highly monodisperse, and the average diameter and membrane thickness of Ca-alginate capsules are about 2.96 mm and 0.11 mm respectively. The Ca-alginate capsule membranes exhibit desired thermo-responsive gating property. With increasing the content of PNIPAM microspheres embedded in the Ca-alginate capsule membranes, the thermo-responsive gating coefficient of the capsule membranes increases simply. When solute molecules diffuse through the capsule membrane, the thermo-responsive gating coefficient is significantly affected by the molecular weight of solute molecules.

Abstract:In this investigation, we used a kind of polyelectrolyte, sodium alginate, as a model biomacromolecule to investigate the aggregation behaviors in aqueous solution after partial protonation of carboxylate groups in the alginate molecules. It is demonstrated that the alginate assemblies with core-shell structure can be generated by the partial protonation of carboxylate groups in sodium alginate chains using the protons released gradually from the reaction of K(2)S(2)O(8) with water at 70 degrees C in aqueous solution. The partial cross-linked alginate assemblies are pH sensitive and can change to hollow structure in the medium with relatively high pH value. This approach avoids use of block or grafted copolymers as the precursors or any other template to prepare assemblies and capsules, and provides a functional surface for subsequent chemical reaction at the surface (e.g., for binding biomolecules and for surface grafting). Such unique assemblies are also expected to be useful in biomedical fields.
The Molecular Structure and Self-Assembly Behavior of Reductive Amination of Oxidized Alginate Derivative for Hydrophobic Drug Delivery

Abstract:On account of the rigid structure of alginate chains, the oxidation-reductive amination reaction was performed to synthesize the reductive amination of oxidized alginate derivative (RAOA) that was systematically characterized for the development of pharmaceutical formulations. The molecular structure and self-assembly behavior of the resultant RAOA was evaluated by an FT-IR spectrometer, a 1H NMR spectrometer, X-ray diffraction (XRD), thermal gravimetric analysis (TGA), a fluorescence spectrophotometer, rheology, a transmission electron microscope (TEM) and dynamic light scattering (DLS). In addition, the loading and in vitro release of ibuprofen for the RAOA microcapsules prepared by the high-speed shearing method, and the cytotoxicity of the RAOA microcapsules against the murine macrophage RAW264.7 cell were also studied. The experimental results indicated that the hydrophobic octylamine was successfully grafted onto the alginate backbone through the oxidation-reductive amination reaction, which destroyed the intramolecular hydrogen bond of the raw sodium alginate (SA), thereby enhancing its molecular flexibility to achieve the self-assembly performance of RAOA. Consequently, the synthesized RAOA displayed good amphiphilic properties with a critical aggregation concentration (CAC) of 0.43 g/L in NaCl solution, which was significantly lower than that of SA, and formed regular self-assembled micelles with an average hydrodynamic diameter of 277 nm (PDI = 0.19) and a zeta potential of about -69.8 mV. Meanwhile, the drug-loaded RAOA microcapsules had a relatively high encapsulation efficiency (EE) of 87.6 % and good sustained-release properties in comparison to the drug-loaded SA aggregates, indicating the good affinity of RAOA to hydrophobic ibuprofen. The swelling and degradation of RAOA microcapsules and the diffusion of the loaded drug jointly controlled the release rate of ibuprofen. Moreover, it also displayed low cytotoxicity against the RAW264.7 cell, similar to the SA aggregates. In view of the excellent advantages of RAOA, it is expected to become the ideal candidate for hydrophobic drug delivery in the biomedical field.

Abstract:With the aim to develop novel scaffolds for the sustained release of drugs, we initially developed an easy approach for the synthesis of α,ω-homobifunctional mercaptoacyl poly(alkyl oxide)s. This was based on the esterification of the terminal hydroxyl groups of poly(alkyl oxide)s with suitably S-4-methoxytrityl (Mmt)-protected mercapto acids, followed by the removal of the acid labile S-Mmt group. This method allowed for the efficient synthesis of the title compounds in high yield and purity, which were further used in the development of a thioether cross-linked liposome scaffold, by thia–Michael reaction of the terminal thiol groups with pre-formed nano-sized liposomes bearing maleimide groups on their surface. The reaction process was followed by 1H-NMR, using a Carr–Purcell–Meiboom–Gill (CPMG) relaxation dispersion NMR experiment (1H-NMR CPMG), which allowed for real-time monitoring and optimization of the reaction process. The thioether cross-linked liposomal scaffold that was synthesized was proven to preserve the nano-sized characteristics of the initial liposomes and allowed for the sustained release of calcein (which was used as a hydrophilic dye and a hydrophilic drug model), providing evidence for the efficient synthesis of a novel drug release scaffold consisting of nanoliposome building blocks.

Abstract:Current focus on particulate drug delivery entails the need for increased drug loading and sustained release of water soluble drugs. Commonly studied biodegradable polyesters, such as poly(lactide-co-glycolide) (PLGA) and poly(l-lactide) (PLLA), are lacking in terms of loading efficiency of these drugs and a stable encapsulation environment for proteins. While hydrogels could enable higher loading of hydrophilic drugs, they are limited in terms of controlled and sustained release. With this in mind, the aim was to develop microparticles with a hydrophilic drug-loaded hydrogel core encapsulated within a biodegradable polyester shell that can improve hydrophilic drug loading, while providing controlled and sustained release. Herein, we report a single step method of fabricating microparticles via a concurrent ionotropic gelation and solvent extraction. Microparticles fabricated possess a core-shell structure of alginate, encapsulated in a shell constructed of either PLGA or PLLA. The cross-sectional morphology of particles was evaluated via scanning electron microscopy, calcium alginate core dissolution, FT-IR microscopy and Raman mapping. The incorporation of alginate within PLGA or PLLA was shown to increase encapsulation efficiency of a model hydrophilic drug metoclopramide HCl (MCA). The findings showed that the shell served as a membrane in controlling the release of drugs. Such gel-core hydrophobic-shell microparticles thus allow for improved loading and release of water soluble drugs.

Abstract:Designing cell-compatible, bio-degradable, and stimuli-responsive hydrogels is very important for biomedical applications in cellular delivery and micro-scale tissue engineering. Here, we report achieving flexible control of cellular microencapsulation, permeability, and release by rationally designing a diblock copolymer, alginate-conjugated poly(N-isopropylacrylamide) (Alg-co-PNiPAM). We use the microfluidic technique to fabricate the bifunctional copolymers into thousands of mono-disperse droplet-templated hydrogel microparticles for controlled encapsulation and triggered release of mammalian cells. In particular, the grafting PNiPAM groups in the synthetic cell-laden microgels produce lots of nano-aggregates into hydrogel networks at elevated temperature, thereafter enhancing the permeability of microparticle scaffolds. Importantly, the hydrogel scaffolds are readily fabricated via on-chip quick gelation by triggered release of Ca2+ from the Ca-EDTA complex; it is also quite exciting that very mild release of microencapsulated cells is achieved via controlled degradation of hydrogel scaffolds through a simple strategy of competitive affinity of Ca2+ from the Ca-Alginate complex. This finding suggests that we are able to control cellular encapsulation and release through ion-induced gelation and degradation of the hydrogel scaffolds. Subsequently, we demonstrate a high viability of microencapsulated cells in the microgel scaffolds.

Abstract:In this paper, alginic acid-poly(2-(diethylamino)ethyl methacrylate) (ALG-PDEA) nanoparticles were successfully prepared in aqueous medium using a polymer-monomer pair reaction system consisting of the anionic alginic acid (ALG) and the cationic 2-(diethylamino)ethyl methacrylate (DEA), without any aid of surfactants or organic solvents. The ALG-PDEA nanoparticles were monodispersed and stable in aqueous solution. Nanoparticles with desired size could be obtained by varying the amount of initiator or changing the concentration of reactants in solution, which renders this system highly controllable. After the ALG moiety was gelled by Ca2+, the stability of the nanoparticles in basic or high salt concentration solutions could be notably enhanced. A pH-sensitive anticancer agent, hydroxycamptothecin (HCPT), was encapsulated in ALG-PDEA nanoparticles, and preliminary in vitro release as well as cytotoxicity experiments were carried out. It is found that this system seems to be a very promising carrier for the loading and delivery of labile drugs, taking into account that the preparation procedure is simple, mild, and organic solvent- and surfactant-free. Moreover, the abundant functional groups on the particle surface, such as carboxyls and hydroxyls, allow subsequent chemical modification, which may further unleash the potential of such a system in either biomedical applications or in the construction of other functional mesoscopic architectures.

One-Pot Synthesis of Amphiphilic Biopolymers from Oxidized Alginate and Self-Assembly as a Carrier for Sustained Release of Hydrophobic Drugs

Abstract:In this paper, we developed an organic solvent-free, eco-friendly, simple and efficient one-pot approach for the preparation of amphiphilic conjugates (Ugi-OSAOcT) by grafting octylamine (OCA) to oxidized sodium alginate (OSA). The optimum reaction parameters that were obtained based on the degree of substitution (DS) of Ugi-OSAOcT were a reaction time of 12 h, a reaction temperature of 25 °C and a molar ratio of 1:2.4:3:3.3 (OSA:OCA:HAc:TOSMIC), respectively. The chemical structure and composition were characterized by Fourier transform infrared spectroscopy (FTIR), 1H nuclear magnetic resonance (1H NMR), X-ray diffraction (XRD), thermogravimetry analyser (TGA), gel permeation chromatography (GPC) and elemental analysis (EA). It was found that the Ugi-OSAOcT conjugates with a CMC value in the range of 0.30-0.085 mg/mL could self-assemble into stable and spherical micelles with a particle size of 135.7 ± 2.4-196.5 ± 3.8 nm and negative surface potentials of -32.8 ± 0.4--38.2 ± 0.8 mV. Furthermore, ibuprofen (IBU), which served as a model poorly water-soluble drug, was successfully incorporated into the Ugi-OSAOcT micelles by dialysis method. The drug loading capacity (%DL) and encapsulation efficiency (%EE) of the IBU-loaded Ugi-OSAOcT micelles (IBU/Ugi-OSAOcT = 3:10) reached as much as 10.9 ± 0.4-14.6 ± 0.3% and 40.8 ± 1.6-57.2 ± 1.3%, respectively. The in vitro release study demonstrated that the IBU-loaded micelles had a sustained and pH-responsive drug release behavior. In addition, the DS of the hydrophobic segment on an OSA backbone was demonstrated to have an important effect on IBU loading and drug release behavior. Finally, the in vitro cytotoxicity assay demonstrated that the Ugi-OSAOcT conjugates exhibited no significant cytotoxicity against RAW 264.7 cells up to 1000 µg/mL. Therefore, the amphiphilic Ugi-OSAOcT conjugates synthesized by the green method exhibited great potential to load hydrophobic drugs, acting as a promising nanocarrier capable of responding to pH for sustained release of hydrophobic drugs.
