Abstract:This study aimed to improve the thermodynamic performance of nanoliposomes (NLs) using fucoidan (F) as the second-layer coating biopolymer along with chitosan (CS), to control the delivery and bioavailability of catechin (C) and juglone (J). The stabilized liposomal carrier of F/CS-conjugated JC-NL (F-CS-JC-NL) was developed with optimum concentrations of CS (0.09 wt%) and F (0.10 wt%), with the highest encapsulation efficiency of juglone (95.47%) and catechin (90.88%). Physicochemical characterization revealed that F-CS-JC-NL disclosed improved stability under different pH and ionic strengths, with the maximum juglone/catechin retention under thermal, oxidative and storage conditions. In vitro digestion revealed that NL double-coating (F-CS-JC-NL) significantly reduced compound leakage in the gastrointestinal tract, providing a controlled release and better bioavailability of juglone/catechin compared to CS-JC-NL and JC-NL. Conclusively, this study provides a novel NL-based de-livery carrier with enhanced physicochemical stability and controlled release that might have promising use in delivering functional ingredients.

Abstract:A novel biocompatible shell-crosslinked nanocapsule system was developed based on nanoemulsion templates stabilized by a class of food proteins. The nanoemulsion templates were prepared using a combination of mechanical mixing and high-pressure homogenization, while the nanocapsule shell formed simultaneously through calcium ions-induced crosslinking of the food proteins. These core–shell structured nanocapsules with a particle size of about 200nm showed high drug-loading capacity and well improved stability in comparison with their nanoemulsion counterpart. The nanocapsule suspension can be freeze-fried directly; and the solidified nanocapsules can be well reconstituted in water, retaining their original particle size. It is concluded that the nanoemulsion-templated core–shell structured nanocapsules can be used as novel drug delivery systems with high loading capacity for poorly water-soluble drugs as well as well improved long-term and storage stability. Furthermore, the presence of surface food proteins introduces carboxyl and amine moieties, which enables the nanocapsules to anchor ligands, suggesting its potential application in targeting drug delivery, bioimaging and therapeutics.
Resveratrol Nanoparticles Coated by Metal-Polyphenols Supramolecular Enhance Antioxidant Activity and Long-Term Stability of Dietary Gel

Abstract:Resveratrol (RES) is an important functional substance with multiple active properties. However, RES is susceptible to natural environmental conditions that reduce its bioactivity. To improve the bioavailability of RES, in this study, Catechin and Fe3+/Ca2+ were selected to form supramolecules, which were then coated on the surface of hydrophobic RES nanoparticles (RES NPs) to create composite RES NPs. The obtained composite RES NPs demonstrated higher antioxidant capacity and better photo-thermal stability than RES NPs. Additionally, a pectin (PE) dietary gel was designed as a delivery carrier for RES. The results showed that the incorporation of composite RES NPs not only endowed the gels with significant dietary activity but also enhanced the texture, water retention capacity and hydrophobicity. After 28 days of storage, the retention rate of RES could be maintained above 90 % in the dietary gels. Meanwhile, the controlled release of RES was achieved in in vitro simulated digestion.

Abstract:The purpose of the study was to explore the effect of different type of crosslinked cyclodextrins for the delivery of poorly soluble, photosensitive drug, resveratrol. Crosslinkers, diphenyl carbonate and pyromellitic dianhydride were used to prepare carbonyl (NS-I) and carboxylate (NS-II) crosslinked cyclodextrin respectively. The solubility and in silico molecular interaction of resveratrol with these NS at different crosslinker ratio were studied. The results showed enhanced solubility and better interaction of resveratrol with nanosponges prepared with 1:4 Cyclodextrin: crosslinker ratio. The drug-loaded nanosponges (RES-NS-I and II) prepared using 1:4 crosslinked NS-I and II were characterized using DSC, PXRD, SEM, FTIR and evaluated for particle size, zeta potential, photodegradation, in vitro drug release, in vitro cytotoxicity and in vivo oral bioavailability in rats. Physical characterization confirmed the molecular inclusion of drug with NS. The release of the drug was increased to 2.5–3 folds in the dissolution medium, with initial drug release faster with RES-NS-II. Photostability was enhanced to 2.3 fold with RES-NS-II. The cytotoxicity test exhibited 1.5 fold reduction in IC50 with drug-loaded NS. RES-NS-II exhibited 2.5 fold increase in Cmax and fourfold decrease in Tmax. Carboxylate crosslinked Cyclodextrin using pyromellitic dianhydride proves to be an effective nanocarrier for resveratrol.

Co-encapsulation of quercetin and resveratrol: Comparison in different layers of zein-carboxymethyl cellulose nanoparticles

Abstract:Three nanoparticles were fabricated for the co-delivery of quercetin and resveratrol. Nanoparticles consisted of a zein and carboxymethyl cellulose assembled using antisolvent precipitation/layer-by-layer deposition method. Nanoparticles contained quercetin in the core and resveratrol in the shell, resveratrol in the core and quercetin in the shell or both quercetin and resveratrol in the core. The particle sizes of nanoparticles were 280.4, 214.8, and 181.8 nm, respectively. Zeta-potential was about -50 mV and PDI was about 0.3. The different positions of polyphenol distribution nanoparticles could reduce the competition between the two polyphenols, the encapsulation rate, loading rate and storage stability reached up to 91.7 %, 5.37 % and 97.1 %, respectively. FT-IR showed that hydrophobic and electrostatic interactions were the main driving forces of nanoparticle assembly. XRD showed that two polyphenols were successfully encapsulated in nanoparticles. TGA showed that distributing the nanoparticles in different layers would enhance thermal stability. TEM and SEM showed that polysaccharides attached to the surface of nanoparticles formed a core-shell structure with uniform particle size. All three nanoparticles could release two polyphenols slowly in simulated gastrointestinal digestion, Korsmeyer-Peppas was the most suitable kinetic release model. Therefore, biopolymer-based nanocarriers can be created to enhance the loading, stability, and bioaccessibility of co-encapsulated nutraceuticals.

Abstract:Resveratrol (Res), a non-flavonoid polyphenolic compound extracted from natural plants, has many pharmacological effects such as anti-tumor, organ protection, improvement of cardiovascular disease, and anti-inflammation. However, due to its poor water solubility and light-degradable property, its bioavailability is restricted. In this study, the hyaluronic acid (Ha)-coated poloxamer 188 (F68) and poloxamer 407 (F127) and Ha-coated Res nanomicelles (Ha-F68/F127-Res-Mc) were prepared by film dispersion method. The nanomicelles were evaluated by indicators of encapsulation efficiency ( EE ), drug loading ( DL ), particle size, polydispersity index and ζ-potential. Besides, using the Box–Behnken design (BBD) based response surface methodology (RSM), obtained the optimum prescription process to obtain the Ha-F68/F127-Res-Mc. Compared with the Res active pharmaceutical ingredient (API), Ha-F68/F127-Res-Mc had higher water solubility and photostability, and could achieve sustained release. Compared with the Res active pharmaceutical ingredient (API), Ha-F68/F127-Res-Mc had higher water solubility and photostability, and could achieve sustained release. In conclusion, the prepared nanomicelles have stronger cytotoxic effect on MCF-7 than doxorubicin (DOX). This will provide a basis for the development of Res antitumor injection, and provide a reference for the research on nano-delivery systems of other BCS II drugs in the future.

Abstract:Resveratrol loaded nanoparticles (nano-resveratrol) containing a zein core surrounded by surfactant (Tween-NPs) or carboxymethyl chitosan (CMCS-NPs) shell were fabricated with different particle sizes, surface charges and colloidal stabilities. Changes of physicochemical properties for the two nano-resveratrols, as well as their antioxidant potentials and cytotoxicity were investigated during a static in vitro gastrointestinal tract (GIT) digestion. Results showed that the Tween-NPs had a much higher bioaccessibility (84.1 ± 19.2%) than that of CMCS-NPs (36.6 ± 4.2%) after the GIT digestion, which was expected due to the steric barrier of the CMCS coating. Both nano-resveratrols could sustained their antioxidant activities after digestion. However, the Tween-NPs had a significantly higher cytotoxicity against MCF-7 cells than CMCS-NPs and free resveratrol, while a reduction in cytotoxicity of Tween-NPs was observed after the digestion. The bioactivities results were well correlated with the physicochemical properties and dissolution of resveratrol under environmental stress.

Abstract:The study of children and childhood has historically been accorded a marginal place in the health, human, and social sciences. In part, this is due to prevailing Western ideology that constructs children as passive, presocial, innocent, and vulnerable. The dominant discourse is further characterized by the treatment of children as a homogeneous group, devoid of race, class, or gender. While many investigators have described strategies for the conduct of research that is situated in the interpretive paradigm, there has been no comparable articulation of ideas regarding the conduct of critically grounded research when our participants are children. The purpose of this article is to put forth a historical and contextual analysis of childhood, including a discussion of evolving perspectives about childhood. The manner by which changing social, political, and environmental landscapes have contributed to the marginalization and disenfranchisement of children is examined. Finally, strategies for conducting nursing research that is grounded in the critical paradigmatic perspective, with the simultaneous aims of action, change, and empowerment, are proposed.

Abstract:Aim: Oat protein-shellac nanoparticles (NPs) were developed as a delivery system for resveratrol to improve bioavailability. Materials & methods: The NPs were prepared from w/w emulsion followed by cold-gelation. In vitro release and cell uptake mechanism of NPs were estimated by HPLC and confocal laser scanning microscopy. In vivo bioavailability and hepatoprotective activity of encapsulated resveratrol were studied using rat models. Results & conclusion: NPs (90–300 nm) protected resveratrol in gastric fluid, while allowing controlled release into small intestine in vitro. The optimized NPs showed improvement in resveratrol cell uptake and transport when compared with free resveratrol. NP-100S increased resveratrol bioavailability up to 72.4%, and the absorbed resveratrol effectively prevented CCl4-induced hepatotoxicity by attenuating oxidative stress.

Abstract:Interest in measuring the quality of life (QL) in relation to health care has increased enormously in recent years. This is also true for end-stage renal failure where it is important not only to provide a better survival but also the quality of that survival. The aim of this study was to assess the relative influence of different kinds of treatment on end-stage renal disease after the patients' evaluation of their overall QL. We studied 167 patients receiving conservative treatment (45), haemodialysis (44), haemodialysis and erythropoieth (36), and continuous ambulatory peritoneal dialysis (42). The patients completed an original questionnaire consisting of 37 questions divided in five groups and generating 15 QL variables: personal data (name, gender, age, basic kidney disease); sociodemographic data influenced by the illness (family history, working ability, employment status); general health characteristics (fatigue, appetite, wound healing, sleep, resistance to cold); aspects of private life that are mostly influenced by the disease (social interaction, traveling, mood, sports, sexual life), and patients subjective assessment of their condition (self care and happiness). Patients on haemodialysis showed lower levels of QL than that on peritoneal dialysis related to fatigue (p < 0.01), working ability (p < 0.05), wound healing (p < 0.05), and appetite (p < 0.01) compared to the conservative treatment. Peritoneal dialysis had also a statistically significant positive influence on fatigue (p < 0.05) compared to conservative treatment. However, erythropoletin treatment showed better results with regard to traveling (p < 0.05), resistance to cold (p < 0.01), self care (p < 0.05) and mood (p < 0.05) compared to peritoneal dialysis, and working ability (p < 0.05), fatigue (p < 0.05) and mood (p < 0.05) compared to conservative treatment and haemodialysis.

Abstract:β-cyclodextrin (CD) grafted N-maleoyl chitosan (CD-g-NMCS) with two different degrees of substitution (DS) of N-maleoyl (DS = 21.2% and 30.5%) were synthesized from maleic anhydride and chitosan bearing pendant cyclodextrin (CD-g-CS). CD-g-NMCS based nanoparticles were prepared via an ionic gelation method together with chitosan and CD-g-CS nanoparticles. The size and zeta potential of prepared CD-g-NMCS nanoparticles were 179.2~274.0 nm and 36.2~42.4 mV, respectively. In vitro stability test indicated that CD-g-NMCS nanoparticles were more stable in phosphate-buffered saline compared with chitosan nanoparticles. Moreover, a poorly water-soluble drug, ketoprofen (KTP), was selected as a model drug to study the obtained nanoparticle's potentials as drug delivery carriers. The drug loading efficiency of CD-g-NMCS20 nanoparticles were 14.8% for KTP. MTT assay showed that KTP loaded CD-g-NMCS nanoparticles were safe drug carriers. Notably, in vitro drug release studies showed that KTP was released in a sustained-release manner for the nanoparticles. The pharmacokinetic of drug loaded CD-g-NMCS20 nanoparticles were evaluated in rats after intravenous administration. The results of studies revealed that, compared with free KTP, KTP loaded CD-g-NMCS20 nanoparticles exhibited a significant increase in AUC0→24h and mean residence time by 6.6-fold and 2.9-fold, respectively. Therefore, CD-g-NMCS nanoparticles could be used as a novel promising nanoparticle-based drug delivery system for sustained release of poorly water-soluble drugs. The carboxylic acid groups of the CD-g-NMCS molecule provide convenient sites for further structural modifications including introduction of tissue- or disease- specific targeting groups.

Abstract:Resveratrol is a polyphenol with diverse pharmacological activities, but its clinical efficacy is limited due to low solubility/permeability, light-induced isomerization, auto-oxidation, and rapid metabolism. Nanodelivery systems, such as liposomes, polymeric nanoparticles, lipid nanocarriers, micelles, nanocrystals, inorganic nanoparticles, nanoemulsions, protein-based nanoparticles, exosomes, macrophages, and red blood cells (RBCs) have shown great potential for improving the solubility, biocompatibility, and therapeutic efficacy of resveratrol. This review comprehensively summarizes the recent advances in resveratrol nanoencapsulation and describes potential strategies to improve the pharmacokinetics of existing nanoformulations, enhance targeting, reduce toxicity, and increase drug release and encapsulation efficiency. The article also suggests that in order to avoid potential safety issues, resveratrol nanoformulations must be tested in vivo in a wide range of diseases.

Progesterone Concentrations in Maternal and Fetal Serum Are Lower During Functional Dystocia Than in Normal Labor

Abstract:Concentrations of progesterone and estradiol (E2) were measured in parturient serum and in fetal cord serum during normal labor and in women with functional dystocia. In the study group, there were no cases of cephalopelvic disproportion. In oxytocin-resistant dystocia, the course of labor could not be corrected with oxytocin. To ascertain the effect of oxytocin, we included a number of women whose labor had been induced with oxytocin, followed by normal cervical dilatation and descent of the fetus. All the parturients were grouped retrospectively into those with normal labor and those with dystocia, based on previous definitions. The serum concentrations of progesterone in both the fetal cord and maternal vein were found to be significantly lower in the oxytocin-resistant dystocia group than in women in spontaneous normal labor and those with oxytocin-induced labor and normal progression (P < .05-.005). Oxytocin had no evident effect on the serum concentration of either progesterone or E2, nor did concentrations vary following epidural blockade. Serum E2 concentrations in the maternal vein were similar in all delivery groups. Fetal cord E2 serum concentrations were similar in all vaginal deliveries. Women with the most severe oxytocin-resistant dystocia, delivered by cesarean, had significantly lower serum concentrations of E2 in fetal cord serum compared with the vaginally delivered women (P < .001).
Co-delivery of curcumin and piperine in zein-carrageenan core-shell nanoparticles: Formation, structure, stability and in vitro gastrointestinal digestion

Abstract:<p>Core-shell biopolymer nanoparticles were fabricated using a hydrophobic protein (zein) as the core and a hydrophilic anionic polysaccharide (ι-carrageenan) as the shell. The outer layer was physically cross-linked using cationic calcium ions to form salt bridges between the anionic carrageenan molecules. The potential of using these core-shell nanoparticles to co-deliver two important nutraceuticals (curcumin and piperine) was also investigated. A combination of electrostatic attraction, hydrogen bonding, and hydrophobic interactions was shown to be responsible for the assembly of the curcumin-piperine-loaded nanoparticles. Low levels of calcium ions (0–2 mM) led to the formation of small nanoparticles with compact structures, whereas higher levels (2–5 mM) promoted particle aggregation. The structural organization and morphology of the core-shell nanoparticles also depended on calcium levels, as seen by changes in circular dichroism, fluorescence spectroscopy, and electron microscopy. The core-shell nanoparticles were shown to be effective at retarding the photo- and thermal-degradation of the encapsulated curcumin and piperine. In addition, the core-shell nanoparticles could delay the release of the nutraceuticals under <em>in-vitro</em> gastrointestinal conditions, which may improve their oral bioavailability. In summary, core-shell nanoparticles assembled from zein and ι-carrageenan may be effective co-delivery systems for curcumin and piperine.</p>

Abstract:Nanocarriers are essential for targeted and enhanced drug delivery. However, the use of toxic solvent and the complex of chemical functionalization have seriously limited their clinical translation. Here, a one-step strategy is developed to direct the self-assembly of biocompatible core-shell nanocapsules with tunable surface functionality by microfluidics. Upon rapid mixing and solvent exchange of ethanol with water in a microfluidic device, drug, oil, polymer, and polymer-PEG-function co-precipitate and self-assemble into core-shell nanocapsules with desired surface functionality as driven by energy minimization. The prepared PCL-PEG-FA core-shell nanocapsules demonstrate a high encapsulation efficiency and loading capacity and exhibit excellent tumor-targeting drug delivery performances both in vitro and in vivo via the binding of functional group on the nanocapsule surface with corresponding receptor on the tumor cell membrane. The immune activation in the tumor microenvironment is investigated in detail by flow cytometry and multiplex immunohistochemistry staining to reveal the underlying mechanism for enhanced anti-tumor performances. The developed strategy is green, facile, versatile, scalable and offers a promising platform for the design of advanced nanocarriers with hierarchical structure and desired function for enhanced drug delivery. Core-shell nanocapsules with tunable surface functionality are prepared by a one-step self-assembly method in a microfluidic device without any toxic solvent and additional chemical functionalization. Abstract:This research was aim to develop novel cyclodextrin/chitosan (CD/CS) nanocarriers for insoluble drug delivery through the mild ionic gelation method previously developed by our lab. A series of different beta-cyclodextrin (beta-CD) derivatives were incorporated into CS nanoparticles including hydroxypropyl-beta-cyclodextrin (HP-beta-CD), sulphobutylether-beta-cyclodextrin (SB-beta-CD), and 2,6-di-O-methy-beta-cyclodextrin (DM-beta-CD). Various process parameters for nanoparticle preparation and their effects on physicochemical properties of CD/CS nanoparticles were investigated, such as the type of CD derivatives, CD and CS concentrations, the mass ratio of CS to TPP (CS/TPP), and pH values. In the optimal condition, CD/CS nanoparticles were obtained in the size range of 215-276 nm and with the zeta potential from 30.22 mV to 35.79 mV. Moreover, the stability study showed that the incorporation of CD rendered the CD/CS nanocarriers more stable than CS nanoparticles in PBS buffer at pH 6.8. For their easy preparation and adjustable parameters in nanoparticle formation as well as the diversified hydrophobic core of CD derivatives, the novel CD/CS nanoparticles developed herein might represent an interesting and versatile drug delivery platform for a variety of poorly water-soluble drugs with different physicochemical properties. Abstract:Resveratrol (RES) is a common active factor in the functional food field, but poor water solubility and low bioavailability have limited its application. In the present study, the novel nanoparticles (RES-CBFMP NPs) using floral mushroom polysaccharide as the wall material have been developed for delivering RES, aiming to overcome its application shortcomings. After ratio optimization, RES-CBFMP NPs (RES-CBFMP,1:8 w/w), which combined through the hydrogen bonds between RES and CBFMP, showed the best overall performance, with the encapsulation efficiency (EE) of 49.74 ± 0.16%, loading efficiency (LE) of 5.53 ± 0.02%, particle size of 158.56 ± 1.97 nm and zeta-potential of -17.56 ± 0.24 mV. In addition, RES-CBFMP NPs exhibited good physicochemical stabilities, sustained gastrointestinal digestive release property, as well as improved in vitro antioxidant and anticancer activities. This study may contribute to the development of RES oral delivery systems and the application of hydrophobic active molecules in the functional food field.
