Abstract:Metal-organic frameworks (MOFs), as drug delivery carriers, with high loading capacity and controllable release behavior can provide a more efficacious therapy in cancer treatments. In our work, a novel biocompatible zinc MOF Zn-cpon-1 with the (3,6)-connected rtl 3D topological network was designed and synthesized via employing ClO4- anion as template. The optically and chemically stable Zn-cpon-1 could be verified as a pH-responsive dual-emission platform and excellent drug delivery carrier with higher 5-fluorouracil (5-FU) (44.75 wt %) loading behavior than 6-mercaptopurine (6-MP) (4.79 wt %) ascribed to the influence of size and shape matching. The multiple interactions between Zn-cpon-1 and 5-FU drug molecules have been discussed and evidenced, which could be quantitatively estimated via the rate constant related to the topological structure. Specially, the gust release behavior of 5-FU@Zn-cpon-1 microcrystal was described and programmed via the Weibull distribution model and could be dual-triggered by the temperature and pH stimulus. This study illustrates that the Zn-cpon-1 without any postmodification performs a favorable potential of being used as biomedical drug delivery alternative carriers in effective drug payload, flexible release administration, and superior dual-stimuli responsiveness.

Abstract:Metal-organic frameworks (MOFs) are coordination compounds that possess an adjustable structure and controllable function. Despite their wide applications in various industries, the use of MOFs in the fields of food and biomedicine is limited mainly due to their potential biological toxicity. Researchers have thus focused on developing biocompatible MOFs to address this issue. Among them, cyclodextrin-based metal-organic frameworks (CD-MOFs) have emerged as a promising alternative. CD-MOFs are novel MOFs synthesized using naturally carbohydrate cyclodextrin and alkali metal cations, and possess renewable, non-toxic, and edible characteristics. Due to their high specific surface area, controllable porosity, great biocompatibility, CD-MOFs have been widely used in various delivery systems, such as encapsulation of nutraceuticals, flavors, and antibacterial agents. Although the field of CD-MOF materials is still in its early stages, they provide a promising direction for the development of MOF materials in the delivery field. This review describes classification and structural characteristics, followed by an introduction to formation mechanism and commonly used synthetic methods for CD-MOFs. Additionally, we discuss the status of the application of various delivery systems based on CD-MOFs. Finally, we address the challenges and prospects of CD-MOF materials, with the aim of providing new insights and ideas for their future development.

Abstract:Cyclodextrin-based metal-organic frameworks (CD-MOFs) are gaining traction in the realm of drug delivery due to their inherent versatility and potential to amplify drug efficacy, specificity, and safety. This article explores the predominant preparation techniques for CD-MOFs, encompassing methods like vapor diffusion, microwave-assisted, and ultrasound hydrothermal approaches. Native CD-MOFs present compelling advantages in drug delivery applications. They can enhance drug loading capacity, stability, solubility, and bioavailability by engaging in diverse interactions with drugs, including host-guest, hydrogen bonding, and electrostatic interactions. Beyond their inherent properties, CD-MOFs can be customized as drug carriers through two primary strategies: co-crystallization with functional components and surface post-modifications. These tailored modifications pave the way for controlled release manners. They allow for slow and sustained drug release, as well as responsive releases triggered by various factors such as pH levels, glutathione concentrations, or specific cations. Furthermore, CD-MOFs facilitate targeted delivery strategies, like pulmonary or laryngeal delivery, enhancing drug delivery precision. Overall, the adaptability and modifiability of CD-MOFs underscore their potential as a versatile platform for drug delivery, presenting tailored solutions that cater to diverse biomedical and industrial needs.

Abstract:Cyclodextrin metal-organic frameworks (CD-MOFs) show considerable advantages of edibility, degradability, low toxicity, and high drug loading, which have attracted enormous interest, especially in drug delivery. This review summarizes the typical synthesis approaches of CD-MOFs, the drug loading methods, and the mechanism of encapsulation and release. The influence of the structure of CD-MOFs on their drug encapsulation and release is highlighted. Finally, the challenges CD-MOFs face are discussed regarding biosafety assessment systems, stability in aqueous solution, and metal ion effect.

Abstract:In many applications the chip-embedded core of miniature devices is a micro-structured planar spiral power transformer (MPST), and operational and electromagnetic compatibility are key issues to its sizing. Numerical simulation aided design may then be used to solve them. This paper presents mathematical models and numerical simulation results related to sizing the ferrite core, the lumped parameters of the MPST, and their sensitivity with respect to the core thickness (the spacing between the planar windings) and the magnetic permeability of the ferrite core. These outcomes may be of interest to the electrical design of the device that uses the MPST and in analyzing its electromagnetic compatibility with the interfering working environment. Finally, we address the high-frequency noise reduction in DC/DC flyback power sources with MPST.

Abstract:Metal-organic frameworks (MOFs) show promising application in biomedicine and pharmaceutics owing to their extraordinarily high surface area, tunable pore size, and adjustable internal surface properties. However, MOFs are prepared from non-renewable or toxic materials, which limit their real-world applications. Cyclodextrins (CDs) are a typical natural and biodegradable cyclic oligosaccharide and are primarily used to enhance the aqueous solubility, safety, and bioavailability of drugs by virtue of its low toxicity and highly flexible structure, offering a peculiar ability to form CD/drug inclusions. A sophisticated strategy where CD is deployed as a ligand to form an assembly of cyclodextrin-based MOFs (CD-MOFs) may overcome real-world application drawbacks of MOFs. CD-MOFs incorporate the porous features of MOFs and the encapsulation capability of CD for drug molecules, leading to outstanding properties when compared with traditional hybrid materials. This review focuses on the inclusion technology and drug delivery properties associated with CD-MOFs. In addition, synthetic strategies and currently developed uses of CD-MOFs are highlighted as well. Also, perspectives and future challenges in this rapidly developing research area are discussed.

Abstract:γ-cyclodextrin-based metal-organic frameworks (γ-CD-MOFs) have considerable potential as a novel porous material with biocompatible properties for use as carrier-based drug delivery. Oregano essential oil (OEO) has been remarkably applied in the food and medical field. Herein, γ-CD-MOFs were prepared in 88% yield by a modified vapor diffusion method for encapsulating liquid OEO. At optimal encapsulation conditions (120 °C for 1 h), encapsulation efficiency and loading capacity reached 27.32% and 8.43%, respectively. Moreover, crystal structure characterization revealed that encapsulation of OEO did not change the crystal state of γ-CD-MOFs. Thus, based on a thorough characterization of prepared inclusion complexes, it was demonstrated that OEO was successfully encapsulated with increased thermal stability. Moreover, γ-CD-MOFs exhibited outstanding release control in vitro experiments. Collectively, the results indicated that γ-CD-MOFs can be used as an effective carrier to deliver OEO.

Abstract:Nanocomposite Hydrogels are versatile and adaptable in drug delivery, suitable for various administrations. They offer prolonged and regulated drug release which makes them valuable in targeting cancer treatments. This study used the water/oil/water approach to construct a drug nanocarrier. It encapsulated hazelnut oil, serving as a protective and organic membrane that enveloped the nanocarrier and regulated drug release. Quercetin (QC), a potent flavonoid with potential therapeutic applications, faces challenges due to its hydrophobic nature and poor solubility. This approach presents a novel approach to enhance QC's solubility and stability by integrating it with copper carbon quantum dots (Cu-CQDs) into a pH-sensitive carboxymethyl cellulose (CMC) and starch-based hydrogel nanocomposite. This system aims to improve QC delivery while minimizing adverse effects. The nanocomposite exhibited exceptional drug loading and encapsulation efficiencies of 47.00 % ± 0.45 and 86.25 % ± 0.75, respectively, ranking among the top-reported values for similar systems. Fourier-transform infrared spectroscopy (FTIR) was used to examine molecular interactions. X-ray diffraction (XRD) assessment demonstrated the presence of components beneficial for drug solubility enhancement at 2Θ = 17–24°, where QC's crystalline peaks are absent from the CMC/Starch/Cu-CQDs@QC XRD pattern. A particle size of 151.57 ± 37 nm is optimized for selectively targeting cancer cells, minimizing interaction with healthy cells, as revealed through DLS assessment. Morphological characteristics analyzed by FE-SEM support this observation as well. Zeta potential measurement (+38.8 mV) highlights nanocarrier stability. CMC/Starch/Cu-CQDs@QC displayed remarkable cytotoxicity against cancerous cells, reducing U87-MG cell viability to 48 % while maintaining high cell viability (92 %) for normal L929 cells. These results emphasize the promise of this nanocomposite as a QC drug delivery system by indicating improved anticancer efficacy with fewer adverse effects.

Abstract:Green tea catechins have received significant attention due to their potent antioxidant activity as well as diverse biological properties. Stabilizing and protecting catechins from degradation is very important but challenging. In this work, a nanosized edible γ-cyclodextrin-based metal–organic framework (CD-MOF) was designed and fabricated by a facile vapor diffusion route and applied for the encapsulation of a model green tea catechin, (-)-epigallocatechin gallate (EGCG) for the first time. Downsizing CD-MOF to the nanoscale is a suitable method to tackle down new applications. Significantly, the antioxidant activity results show that the CD-MOF-EGCG can remarkably enhance the antioxidant activity in alkaline solutions, as compared to that of free EGCG. Furthermore, the prepared CD-MOF-EGCG showed strong cancer cell growth inhibitory effects on C6 cells as confirmed by the cell viability assay. This work demonstrates that such safe and nontoxic nanoscale CD-MOF-based porous materials hold great promise for applications in the field of stabilization of catechins for biomedical applications.

Abstract:Inorganic-organic hybrid materials, especially metal-organic frameworks (MOFs) composed of metals and organic linkers, have emerged as a new class of versatile materials owing to their tunable structure and controllable functionality. As typical biocompatible MOFs, cyclic oligosaccharide cyclodextrin-based carbohydrate metal-organic frameworks (CD-MOFs) have recently attracted considerable attention due to their edible, renewable and biodegradable nature. Herein, we focus on the latest advances concerning these materials. First, the synthesis methods and structural diversity of CD-MOFs are introduced and summarized. Besides, the synthetic strategies of moisture-resistant CD-MOFs are also emphasized. Moreover, their applications, including gas adsorption, separation, sensing, memristor fabrication, as templates in nanoparticle synthesis, light emission and especially drug delivery, are systematically discussed and highlighted. Finally, to conclude the review, some insights and current challenges that need to be addressed for the further development of these materials are proposed. We anticipate that this review will result in a better understanding of CD-MOFs and will help maximize the potential functions of these materials.
Ethanol-Mediated Synthesis of γ-Cyclodextrin-Based Metal-Organic Framework as Edible Microcarrier: Performance and Mechanism

Abstract:Here, an ethanol-mediated method was introduced to fabricate γ-cyclodextrin-based metal-organic frameworks (γ-CD-MOFs) as microcarriers for epigallocatechin-3-gallate (EGCG). Through adjusting ethanol gas diffusion temperature and ethanol liquid feed speed, we achieved control of crystallization efficiency and crystals size without extra surfactants. Under the sequential regulatory by ethanol in two phases, the obtained γ-CD-MOFs with cubic shape exhibited excellent crystallinity, high surface area, and uniform size distribution. Through the interplay of hydrogen bonding, hydrophobic interactions and π stacking, EGCG molecules could be stored efficiently within cavities and tunnels of the γ-CD-MOFs with high load capability of 334 mg g -1 . More importantly, the incorporation of EGCG within frameworks wouldn't disintegrate the unique body-centered cubic structure of γ-CD-MOFs, in turn, would improve the thermostability and antioxidative activity of EGCG. Significantly, all food-grade materials ensured the γ-CD-MOFs high acceptance and applicability for food and biomedical applications.

Abstract:Cyclodextrin metal-organic framework by ultrasound-assisted rapid synthesis for caffeic acid (CA) loading and antibacterial application (U-CD-MOF) was successfully studied and this method shortened the preparation time to a few minutes. It was found that the ultrasonic power, reaction time and temperature would affect the morphology and size of the obtained crystal. Under the optimal conditions, U-CD-MOF had a cubic structure with uniform size of 8.60 ± 1.95 μm. U-CD-MOF was used to load the antibacterial natural product CA to form the composite () and the loading rate of to CA could reach 19.63 ± 2.53%, which was more than twice that of γ-CD. Various techniques were applied to characterize the synthesized crystal, including Powder X-ray diffraction (PXRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermogravimetric analysis (TGA), and N2 adsorption. In addition, antibacterial tests were performed on the obtained crystal. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of for Escherichia coli O157: H7 (E. coli O157: H7) were both 25 mg·mL−1, and the MIC for Staphylococcus aureus (S. aureus). was 25 mg·mL−1. The sustained release behavior of to CA in ethanol fitted well to Higuchi model and the loading of CA was supported by molecular docking results. In general, U-CD-MOF was successfully achieved by ultrasound-assisted rapid synthesis and the obtained crystal was further evaluated for potential antibacterial application.

Abstract:<p>Cyclodextrin-metal-organic frameworks (CD-MOFs) based on cyclodextrin (CD) have typical edible, renewable and biodegradable characteristics. CD-MOFs have gradually attracted the attention of researchers in the food industry due to their high specific surface area and good biocompatibility properties. Here, we focus on the latest research progress on CD-MOFs in the food industry. The strategy of CD-MOFs as carriers of bioactive compound to improve the solubility and stability of bioactive compound is emphasized. In addition, the potential applications of CD-MOFs in the food industry are reviewed, such as carriers of bioactive compound used as food additives, antibacterial compounds used in food packaging, etc. Finally, this review points out the problem that non-food grade reagents added during the preparation process of CD-MOFs which restrict the use of CD-MOFs in food applications.</p>

Abstract:Ibuprofen(IBU) is one of the most widely used non-steroidal anti-inflammatory drugs (NSAIDs), but its solubility is poor in both aqueous and physiological environments . To improve its oral bioavailability and absorption rate, utilizing material-loaded IBU to increase the half-life of IBU in the body is an effective method. Cyclodextrin metal-organic frameworks (CD-MOFs) possess several advantages, such as excellent biocompatibility, large surface area, and high porosity. They are considered highly promising drug carriers. However, CD-MOFs are unstable in humid environments, which can affect the preparation and storage of drugs. In this work, a novel cross-linked CL-CD-MOFs material was prepared, and on this basis, two drug-loaded materials, CL-CD-MOFs-IBU, were synthesized by changing the order of the cross-linking agent, isophorone diisocyanate (IPDI) and IBU in the reaction. The research indicates that the prepared CL-CD-MOFs exhibit stability in both water and humid environments, while CD-MOFs are easily dissolved under the same conditions. Therefore, the cross-linking of IPDI with CD-MOFs improves the poor water stability of CD-MOFs. Further analysis through drug loading performance and in vitro drug release tests reveals that the prepared CL-CD-MOFs exhibit high drug loading rate and encapsulation rate. Compared to the sequence of cross-linking followed by drug loading, the sequence of drug loading followed by cross-linking shows a more obvious improvement in IBU loading capacity and encapsulation rate. Additionally, it demonstrates prolonged drug release over time. The cytotoxicity of CL-CD-MOFs-IBU was evaluated by MTT method, and it was found that the material showed low cytotoxicity and good biocompatibility.

Abstract:Pulmonary drug delivery has attracted considerable attention in recent years. However, it is still a major challenge to deliver poorly water-soluble drugs to lungs with good solubility and fine aerodynamic performance. In this study, curcumin was loaded into cyclodextrin-based metal-organic frameworks (CD-MOFs) for pulmonary delivery. Compared with micronized curcumin prepared by jet milling, curcumin-loaded CD-MOFs (Cur-CD-MOFs) exhibited excellent aerodynamic performance, which was attributed to the unique porous structure and lower density of CD-MOFs. The dissolution test showed that the drug release rate of Cur-CD-MOFs was much faster than that of micronized curcumin. The all-atom molecular dynamic simulation showed that curcumin molecules were loaded into the hydrophobic cavities of CD-MOFs or entered into the large hydrophilic cavities to form nanoclusters. The elevated wettability of Cur-CD-MOFs and the unique spatial distribution feature of curcumin in porous interior of CD-MOFs might be favorable for the improved dissolution rate. The DPPH radical scavenging test showed that Cur-CD-MOFs had prominent antioxidant activities. Therefore, CD-MOFs were expected to be promising carriers for pulmonary delivery of poorly water-soluble drugs.

Abstract:The relatively new class of porous material known as metal-organic framework (MOF) exhibits unique features such as high specific surface area, controlled porosity and high chemical stability. Many green synthesis approaches for MOFs have been proposed using biocompatible metal ions and linkers to maximise their use in pharmaceutical fields. The involvement of biomolecules as an organic ligand can act promising because of their biocompatibility. Recently, cyclodextrin metal–organic frameworks (CD-MOFs) represent environmentally friendly and biocompatible characteristics that lead them to biomedical applications. They are regarded as a promising nanocarrier for drug delivery, due to their high specific surface area, high porosity, tuneable chemical structure, and easy fabrication. This review focuses on the unique properties of CD-MOF and the recent advances in methods for the synthesis of these porous structures with emphasis on particle size. Then, the state-of-the-art drug delivery systems with various drugs along with the performance of CD-MOFs as efficient drug delivery systems are presented. Particular emphasis is laid on researches investigating the drug delivery potential of γ-CD-MOF.

Abstract:Metal-organic frameworks (MOFs) offer diverse applications in the food industry, facilitating loading, protection, and controlled release of functional ingredients despite encountering loading capacity and functional activity limitations. This study focuses on curcumin‑zinc MOFs, harnessing curcumin's renowned health benefits and zinc to enhance pharmacological properties. We evaluated their synthesis efficiency, stability under varying conditions (pH, salt concentration, temperature), loading and antioxidant capacity. The results showed that microwave synthesis yielded MOFs with a 23.2 ± 4.5% yield, stable within pH 4-10, gradually decomposing in PBS. DPPH, ABTS, and H₂O₂ assays revealed varying free radical scavenging abilities. MOFs disintegrate in either acidic environments or contain H2O2 (at a concentration threshold of 10 μM). Post-disintegration, these MOFs significantly inhibiting the secretion of TNF-α by RAW264.7 cells induced by LPS. These findings highlight the potential of novel curcumin‑zinc MOF materials for nutrient delivery, addressing challenges in effectively delivering functional ingredients.
