Abstract:Tephritidae is a large family that includes several fruit and vegetable pests. These organisms usually harbor a variegated bacterial community in their digestive systems. Symbiotic associations of bacteria and fruit flies have been well-studied in the genera Anastrepha, Bactrocera, Ceratitis, and Rhagoletis. Molecular and culture-based techniques indicate that many genera of the Enterobacteriaceae family, especially the genera of Klebsiella, Enterobacter, Pectobacterium, Citrobacter, Erwinia, and Providencia constitute the most prevalent populations in the gut of fruit flies. The function of symbiotic bacteria provides a promising strategy for the biological control of insect pests. Gut bacteria can be used for controlling fruit fly through many ways, including attracting as odors, enhancing the success of sterile insect technique, declining the pesticide resistance, mass rearing of parasitoids and so on. New technology and recent research improved our knowledge of the gut bacteria diversity and function, which increased their potential for pest management. In this review, we discussed the diversity of bacteria in the economically important fruit fly and the use of these bacteria for controlling fruit fly populations. All the information is important for strengthening the future research of new strategies developed for insect pest control by the understanding of symbiotic relationships and multitrophic interactions between host plant and insects.

Abstract:Abstract Background The Sterile Insect Technique (SIT) is being applied for the management of economically important pest fruit flies (Diptera: Tephritidae) in a number of countries worldwide. The success and cost effectiveness of SIT depends upon the ability of mass-reared sterilized male insects to successfully copulate with conspecific wild fertile females when released in the field. Methods We conducted a critical analysis of the literature about the tephritid gut microbiome including the advancement of methods for the identification and characterization of microbiota, particularly next generation sequencing, the impacts of irradiation (to induce sterility of flies) and fruit fly rearing, and the use of probiotics to manipulate the fruit fly gut microbiota. Results Domestication, mass-rearing, irradiation and handling, as required in SIT, may change the structure of the fruit flies’ gut microbial community compared to that of wild flies under field conditions. Gut microbiota of tephritids are important in their hosts’ development, performance and physiology. Knowledge of how mass-rearing and associated changes of the microbial community impact the functional role of the bacteria and host biology is limited. Probiotics offer potential to encourage a gut microbial community that limits pathogens, and improves the quality of fruit flies. Conclusions Advances in technologies used to identify and characterize the gut microbiota will continue to expand our understanding of tephritid gut microbial diversity and community composition. Knowledge about the functions of gut microbes will increase through the use of gnotobiotic models, genome sequencing, metagenomics, metatranscriptomics, metabolomics and metaproteomics. The use of probiotics, or manipulation of the gut microbiota, offers significant opportunities to enhance the production of high quality, performing fruit flies in operational SIT programs.

Abstract:Bactrocera dorsalis infests a wide range of host fruits, some of which are suitable hosts, while others are less suitable ones. Although reports highlighted the bacteria-mediated adaptations to different host-fruits, the phenotypic cost and/or gain of the association between insect’s gut bacteria and fruit substrates has not been fully studied. In this study, we hypothesized that, when the larvae are confronted with host fruits with different suitability, its community structure changes over time as a result of its adaptation. To support this, a single female fruit fly was allowed to lay eggs on five different host fruits, then we monitored the larval development parameters and analyzed the gut bacterial communities of larvae fed apple and banana at first (F1) and fifth (F5) generations. The larval developmental parameters (length and dry weight) did not vary significantly across generations, but the developmental duration was greatly affected by fruit types and larval stages. The larval development time was extended considerably in apple and tomato, but shortened in banana and mango. Insects grown in apple and banana have similar bacterial phylotypes to those of the parental female at F1 (predominance of Proteobacteria). However, at F5, the proportions of Firmicutes, Bacteroidetes, Actinobacteria, Verrucomicrobia and Cyanobacteria increased substantially in larvae grown in apple, compared to banana. These findings confirm our hypothesis and suggest that, gut bacteria and host fruits are intimately tied in predicting B. dorsalis larval development, important data for bacteria-based control of this pest.

Abstract:Female Mediterranean fruit flies (Ceratitis capitata) oviposit in fruits, within which the larvae develop. This development is associated with rapid deterioration of the fruit, and frequently with invasion by secondary pests. Most research on the associations between medflies and microorganisms has focused on the bacteria inhabiting the digestive system of the adult fly, while the role of the fruit in mediating, amplifying or regulating the fruit fly microflora has been largely neglected. In this study, we examine the hypothesis that the host fruit plays a role in perpetuating the fly-associated bacterial community. Using direct and cultured-based approaches, we show that this community is composed in its very large majority of diazotrophic and pectinolytic Enterobacteriaceae. Our data suggest that this fly-associated enterobacterial community is vertically transmitted from the female parent to its offspring. During oviposition, bacteria are transferred to the fruit, establish and proliferate within it, causing its decay. These results show that the host fruit is indeed a central partner in the fruit fly-bacterial interaction as these transmitted bacteria are amplified by the fruit, and subsequently maintained throughout the fly's life. This enterobacterial community may contribute to the fly's nitrogen and carbon metabolism, affecting its development and ultimately, fitness.

Abstract:The fruit fly Anastrepha obliqua is an economically important pest. The sterile insect technique to control it involves mass production and release of sterile flies to reduce the reproduction of the wild population. As noted in different Tephritidae, the performance of sterile males may be affected by the assimilation of nutrients under mass-rearing conditions. In the wild, the fly's life cycle suggests the acquisition of different organisms that could modulate its fitness and physiology. For A . obliqua , there is no information regarding microorganisms other than bacteria. This study analyzed bacteria, fungal, and archaea communities in the A . obliqua gut through denaturing gradient gel electrophoresis (DGGE) profiles of 16S (using a different set of primers for bacteria and archaea) and 18S ribosomal DNA markers. We found that wild flies presented higher microbial diversity related to fructose assimilation than laboratory species, suggesting that microorganisms have led to a specialized metabolism to process nutrients associated with an artificial diet. We identified species that have not been previously described in this fruit fly, especially actinobacteria and archaea, by employing different primer sets aimed at the same molecular marker but targeting diverse hypervariable regions of 16S rDNA. The possibility that Archaea affect fly fitness should not be ignored. This report on the intestinal microbial (bacteria, archaea, and fungi) composition of A . obliqua contributes to our understanding of the role of microorganisms in the development and physiology of the flies.

Abstract:The gut microbiome of insects directly or indirectly affects the metabolism, immune status, sensory perception and feeding behavior of its host. Here, we examine the hypothesis that in the oriental fruit fly (Bactrocera dorsalis, Diptera: Tephritidae), the presence or absence of gut symbionts affects foraging behavior and nutrient ingestion. We offered protein-starved flies, symbiotic or aposymbiotic, a choice between diets containing all amino acids or only the non-essential ones. The different diets were presented in a foraging arena as drops that varied in their size and density, creating an imbalanced foraging environment. Suppressing the microbiome resulted in significant changes of the foraging behavior of both male and female flies. Aposymbiotic flies responded faster to the diets offered in experimental arenas, spent more time feeding, ingested more drops of food, and were constrained to feed on time-consuming patches (containing small drops of food), when these offered the full complement of amino acids. We discuss these results in the context of previous studies on the effect of the gut microbiome on host behavior, and suggest that these be extended to the life history dimension.

Abstract:BACKGROUND:The gut of most insects harbours nonpathogenic microorganisms. Recent work suggests that gut microbiota not only provide nutrients, but also involve in the development and maintenance of the host immune system. However, the complexity, dynamics and types of interactions between the insect hosts and their gut microbiota are far from being well understood.METHODS/PRINCIPAL FINDINGS:To determine the composition of the gut microbiota of two lepidopteran pests, Spodoptera littoralis and Helicoverpa armigera, we applied cultivation-independent techniques based on 16S rRNA gene sequencing and microarray. The two insect species were very similar regarding high abundant bacterial families. Different bacteria colonize different niches within the gut. A core community, consisting of Enterococci, Lactobacilli, Clostridia, etc. was revealed in the insect larvae. These bacteria are constantly present in the digestion tract at relatively high frequency despite that developmental stage and diet had a great impact on shaping the bacterial communities. Some low-abundant species might become dominant upon loading external disturbances; the core community, however, did not change significantly. Clearly the insect gut selects for particular bacterial phylotypes.CONCLUSIONS:Because of their importance as agricultural pests, phytophagous Lepidopterans are widely used as experimental models in ecological and physiological studies. Our results demonstrated that a core microbial community exists in the insect gut, which may contribute to the host physiology. Host physiology and food, nevertheless, significantly influence some fringe bacterial species in the gut. The gut microbiota might also serve as a reservoir of microorganisms for ever-changing environments. Understanding these interactions might pave the way for developing novel pest control strategies.

Abstract:Gut microbial communities are critical in determining the evolutive success of fruit fly phytophagous pests (Diptera, Tephritidae), facilitating their adaptation to suboptimal environmental conditions and to plant allelochemical defences. An important source of variation for the microbial diversity of fruit flies is represented by the crop on which larvae are feeding. However, a "crop effect" is not always the main driver of microbial patterns, and it is often observed in combination with other and less obvious processes. In this work, we aim at verifying if environmental stress and, by extension, changing environmental conditions, can promote microbial diversity in Zeugodacus cucurbitae (Coquillett), a cosmopolitan pest of cucurbit crops. With this objective, 16S rRNA metabarcoding was used to test differences in the microbial profiles of wild fly populations in a large experimental setup in Eastern Central Tanzania. The analysis of 2,973 unique ASV, which were assigned to 22 bacterial phyla, 221 families and 590 putative genera, show that microbial α diversity (as estimated by Abundance Coverage Estimator, Faith's Phylogenetic Diversity, Shannon-Weiner and the Inverse Simpson indexes) as well as β microbial diversity (as estimated by Compositional Data analysis of ASVs and of aggregated genera) significantly change as the species gets closer to its altitudinal limits, in farms where pesticides and agrochemicals are used. Most importantly, the multivariate dispersion of microbial patterns is significantly higher in these stressful environmental conditions thus indicating that Anna Karenina effects contribute to the microbial diversity of Z . cucurbitae . The crop effect was comparably weaker and detected as non-consistent changes across the experimental sites. We speculate that the impressive adaptive potential of polyphagous fruit flies is, at least in part, related to the Anna Karenina principle, which promotes stochastic changes in the microbial diversity of fly populations exposed to suboptimal environmental conditions.

Phylogenomic analysis and molecular identification of true fruit flies

Abstract:The family Tephritidae in the order Diptera, known as true fruit flies, are agriculturally important insect pests. However, the phylogenetic relationships of true fruit flies, remain controversial. Moreover, rapid identification of important invasive true fruit flies is essential for plant quarantine but is still challenging. To this end, we sequenced the genome of 16 true fruit fly species at coverage of 47–228×. Together with the previously reported genomes of nine species, we reconstructed phylogenetic trees of the Tephritidae using benchmarking universal single-copy ortholog (BUSCO), ultraconserved element (UCE) and anchored hybrid enrichment (AHE) gene sets, respectively. The resulting trees of 50% taxon-occupancy dataset for each marker type were generally congruent at 88% nodes for both concatenation and coalescent analyses. At the subfamily level, both Dacinae and Trypetinae are monophyletic. At the species level, Bactrocera dorsalis is more closely related to Bactrocera latifrons than Bactrocera tryoni . This is inconsistent with previous conclusions based on mitochondrial genes but consistent with recent studies based on nuclear data. By analyzing these genome data, we screened ten pairs of species-specific primers for molecular identification of ten invasive fruit flies, which PCR validated. In summary, our work provides draft genome data of 16 true fruit fly species, addressing the long-standing taxonomic controversies and providing species-specific primers for molecular identification of invasive fruit flies.

Abstract:House flies (Musca domestica L.) are cosmopolitan, ubiquitous, synanthropic insects that serve as mechanical or biological vectors for various microorganisms. To fully assess the role of house flies in the epidemiology of human diseases, it is essential to understand the diversity of microbiota harbored by natural fly populations. This study aimed to identify the diversity of house fly gut bacteria by both culture-dependent and culture-independent approaches. A total of 102 bacterial strains were isolated from the gut of 65 house flies collected from various public places including a garden, public park, garbage/dump area, public toilet, hospital, restaurant/canteen, mutton shop/market, and house/human habitation. Molecular phylogenetic analyses placed these isolates into 22 different genera. The majority of bacteria identified were known potential pathogens of the genera Klebsiella, Aeromonas, Shigella, Morganella, Providencia, and Staphylococcus. Culture-independent methods involved the construction of a 16S rRNA gene clone library, and sequence analyses supported culture recovery results. However, additional bacterial taxa not determined via culture recovery were revealed using this methodology and included members of the classes Alphaproteobacteria, Deltaproteobacteria, and the phylum Bacteroidetes. Here, we show that the house fly gut is an environmental reservoir for a vast number of bacterial species, which may have impacts on vector potential and pathogen transmission.

Abstract:Abstract Gut bacteria play important roles in insect life cycle, and various routes can be used by insects to effectively transmit their gut bacteria. However, it is unclear if the gut bacteria can spread by actively attracting their insect hosts, and the recognition mechanisms of host insects are poorly understood. Here, we explore chemical interactions between Bactrocera dorsalis and its gut bacterium Citrobacter sp. (CF-BD). We found that CF-BD could affect the development of host ovaries and could be vertically transmitted via host oviposition. CF-BD could attract B. dorsalis to lay eggs by producing 3-hexenyl acetate (3-HA) in fruits that were hosts of B. dorsalis . Furthermore, we found that B. dorsalis could directly recognize CF-BD in fruits with their ovipositors in which olfactory genes were expressed to bind 3-HA. This work reports an important mechanism concerning the active spread of gut bacteria in their host insects.

Abstract:Insects,the largest group of muhicellular organisms on the Earth,are notable for their numerous species and varied morphology,generally harboring abundant microbes in their digestive tract.These microorganisms help the host in digestion,nutrition absorption,pheromone synthesis,as well as playing a role in the defense of harmful invaders,which strengthen the host immune activity.Lepidoptera is the second largest order in Insecta,with many common pest and beneficial insect species.Many methods and techniques have been developed for the research on the gut microbiota,which becomes a hot research topic in recent years.This review outlines the research of gut microbiota in lepidopteran insects (e.g.,Bombyx mori,Spodoptera littoralis and Plutella xylostella),including gut environment,microbial diversity and research methods.Furthermore,the community structure of several lepidopteran model insects and the roles of gut microbes in host's detoxification and immune system are summarized.The findings we review here should be valuable for the further research on the gut microbiota in lepidopteran insects,and pave the way for developing novel strategies to control pests and protect beneficial insects.

Abstract:Many species of the Tephritidae family are invasive and cause huge damage to agriculture and horticulture, owing to their reproductive characteristics. In this review, we have summarized the existing studies on the reproductive behavior of Tephritidae, particularly those regarding the genes and external factors that are associated with courtship, mating, and oviposition. Furthermore, we outline the issues that still need to be addressed in fruit fly reproduction research. The review highlights the implications for understanding the reproductive behavior of fruit flies and discusses methods for their integrated management and biological control. Abstract:Bactrocera dorsalis (Diptera: Tephritidae) is a serious menace to agricultural production worldwide. In order to prevent further damage, it is of paramount important that cost-effective strategies should be developed for their management. Gut bacteria has established diverse relationships with their insect hosts, which could be exploited in pest management programs to improve on control efficiency. In this study, gut bacteria isolates identified by culture dependent technique were incorporated into larval diets in an attempt to understand the roles they play in the development and survival of oriental fruit fly. From our results, the isolated bacteria belonged to four different phyla including the Firmicutes, Proteobacteria, Bacteroidetes and Actinobacteria. The response of the fly to different gut isolates varied greatly. Diets enriched with Enterococcus phoeniculicola had lower larval developmental duration, higher pupal weight, and an increased percentage survival. On the other hand, diets supplemented with Lactobacillus lactis had negative effects on B. dorsalis development. This study provides clues on how symbiotic bacteria could be exploited in mass rearing for an efficient implementation of the Sterile Insect Technique (SIT) in pest management programs.

Abstract:Abstract The role of symbiotic microbes in insects, especially the beneficial character of this interaction for insects, has received much attention in recent years as it has been related to important aspects of the host insects' biology such as development, reproduction, survival, and fitness. Among insect hosts, tephritid fruit flies are well known to form beneficial associations with their symbionts. To control these destructive agricultural pests, environmentally friendly approaches, like the sterile insect technique as a component of integrated pest management strategies, remain most effective. In this study, changes in the bacterial profile of mass‐reared oriental fruit flies, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), were examined in both larval and adult stages and also after irradiation by employing a 16S rRNA gene‐based Illumina sequencing approach. Proteobacteria was the prevalent bacterial phylum in non‐irradiated adults and larvae. Alphaproteobacteria was the most abundant class in larvae but almost absent in adults, which was dominated by Gammaproteobacteria. Firmicutes were present in both developmental stages but at lower relative abundance. At genus level, Acetobacter prevailed in the larval stage and members of the Enterobacteriaceae family in adults. Irradiated samples exhibited higher diversity and richness indices compared to the non‐irradiated oriental fruit flies, whereas no significant changes were observed between the two developmental stages of the non‐irradiated samples. Lactobacillus , members of the Orbacecae family, and Morganella were detected but to a lesser degree upon irradiation, whereas the relative abundance of Lactococcus and Orbus increased. The bacterial profile of larvae appeared to be different compared to that of adult B. dorsalis flies. The subsequent application of irradiation at the pupal stage led to the development of different microbiota between treated and untreated samples, affecting diversity and operational taxonomic unit composition. Irradiated samples of oriental fruit flies were characterized by higher species diversity and richness.

Abstract:ABSTRACT Fruit flies are a familiar sight in many food service facilities. Although they have been long considered as “nuisance pests,” some of their typical daily activities suggest they may pose a potential public health threat. The aim of this study was to provide evidence of the ability of small flies to transfer bacteria from a contaminated source, food, or waste to surfaces or ready-to-eat food. Laboratory experiments were conducted by using purpose-built fly enclosures to assess the bacterial transfer capability of fruit flies. Drosophila repleta were capable of transferring Escherichia coli O157:H7, Salmonella Saint Paul, and Listeria innocua from an inoculated food source to the surface of laboratory enclosures. In addition, using an inoculated doughnut and noncontaminated lettuce and doughnut surfaces, fly-mediated cross-contamination of ready-to-eat food was demonstrated. Fruit flies were shown to be capable of accumulating approximately 2.9 × 103 log CFU of E. coli per fly within 2 h of exposure to a contaminated food source. These levels of bacteria did not decrease over an observation period of 48 h. Scanning electron micrographs were taken of bacteria associated with fly food and contact body parts and hairs during a selection of these experiments. These data, coupled with the feeding and breeding behavior of fruit flies in unsanitary areas of the kitchen and their propensity to land and rest on food preparation surfaces and equipment, indicate a possible role for fruit flies in the spread of foodborne pathogens.

Abstract:Houseflies, Musca domestica L., are an ubiquitous pest that can transmit numerous diseases and threaten human health. Increasing insecticide resistance shown by houseflies necessitates the develop new control alternatives. The housefly gut is densely colonized with microorganisms that interact with each other dynamically and benefit the host's health. However, the impact of multiple symbiotic bacteria on the composition of housefly gut microbiota and the host's activities remains unclear.

Abstract:The high invasiveness of phytophagous insects is related to their adaptability to various environments, that can be influenced by their associated microbial community. Microbial symbionts are known to play a key role in the biology, ecology, and evolution of phytophagous insects, but their abundance and diversity are suggested to be influenced by environmental stressors. In this work, using 16 S rRNA metabarcoding we aim to verify (1) if laboratory rearing affects microbial symbiont communities of Zeugodacus cucurbitae females, a cosmopolitan pest of cucurbitaceous crops (2) if temperature, diet quality, and antibiotic treatments affect microbial symbiont communities of both laboratory and wild populations, and (3) if changes in microbial symbiont communities due to temperature, diet and antibiotic affect longevity and fecundity of Z. cucurbitae .
