Abstract:Bacterial infections have become major threats to public health all over the world. With the emergence of antibiotic resistance, it is urgent to develop novel antimicrobial materials to efficiently overcome drug resistance with high bactericidal activity. In this work, UiO-66-NH-CO-MoS2 nanocomposites (UNMS NCs) are constructed through the amidation reaction. The UNMS NCs are positively charged which is beneficial for capturing and restricting bacteria. Significantly, UNMS NCs possess a synergistic bactericidal efficiency based on near-infrared irradiation (808 nm) regulated combination of photothermal, photodynamic, and peroxidase-like enzymatic activities. Both the photodynamic property and nanozymatic activity of UNMS NCs can lead to the generation of reactive oxygen species. The UNMS NCs show high catalytic activity in a wide pH range and exhibit excellent antibacterial ability against ampicillin-resistant Escherichia coli and methicillin-resistant Staphylococcus aureus with negligible cytotoxicity. Interestingly, due to the 808 nm irradiation-induced hyperthermia in the presence of UNMS NCs, the glutathione oxidation process can be accelerated, resulting in bacterial death more easily. Mice wound models are established to further manifest that UNMS NCs can promote wound healing with good biosafety in living systems.

Abstract:The rise and spread of antimicrobial resistance is creating an ever greater challenge in wound management. Nanofibrous membranes (NFMs) incorporated with antibiotics have been widely used to remedy bacterial wound infections owing to their versatile features. However, misuse of antibiotics has resulted in drug resistance, and it remains a significant challenge to achieve both high antibacterial efficiency and without causing bacterial resistance. Here, the ‘MOF-first’ strategy was adopted, the porphyrinic metal-organic frameworks nanoparticles (PCN−224 NPs) were pre-synthesized first, and then the composite antibacterial PCN−224 NPs @ poly (ε-caprolactone) (PM) NFMs were fabricated via a facile co-electrospinning technology. This strategy allows large amounts of effective MOFs to be integrated into nanofibers to effectively eliminate bacteria without bacterial resistance and to realize a relatively fast production rate. Upon visible light (630 nm) irradiation for 30 min, the PM−25 NFMs have the best 1O2 generation performance, triggering remarkable photodynamic antibacterial effects against both S. aureus, MRSA, and E. coli bacteria with survival rates of 0.13%, 1.91%, and 2.06% respectively. Considering the photodynamic antibacterial performance of the composite nanofibrous membranes functionalized by porphyrinic MOFs, this simple approach may provide a feasible way to use MOF materials and biological materials to construct wound dressing with the versatility to serve as an antibacterial strategy in order to prevent bacterial resistance.

Abstract:Phototherapy is a low-risk alternative to traditional antibiotics against drug-resistant bacterial infections. However, optimizing phototherapy agents, refining treatment conditions, and addressing misuse of agents, remain a formidable challenge. This study introduces a novel concept leveraging the unique customizability of metal-organic frameworks (MOFs) to house size-matched dye molecules in “single rooms”. The mesoporous iron(III) carboxylate nanoMOF, MIL-100(Fe), and the hydrophobic heptamethine cyanine photothermal dye (Cy7), IR775, are selected as model systems. Their combination is predicted to minimize dye-dye interactions, leading to exceptional photostability and efficient light-to-heat conversion. Furthermore, MIL-100(Fe) preserves the antimicrobial nature of hydrophobic IR775, enabling it to disrupt bacterial cell envelopes. Through electrospinning, MIL-100(Fe)@IR775 nanoparticles are shaped into a gelatin-based film dressing for the treatment of skin wounds infected by Methicillin-resistant Staphylococcus aureus (MRSA). Activation of the dressing requires only a portable near-infrared light-emitting diode (NIR LED) and induces both low-dose photodynamic therapy (LPDT) and mild-temperature photothermal therapy (MPTT). Combined with the antimicrobial properties of IR775 and ferroptosis-like lipid peroxidation induced by MIL-100(Fe), the photoactive dressing eradicates MRSA and the healing is as quick as the uninfected wounds. This safe, cost-effective, and multifunctional therapeutic wound dressing offers a promising solution to overcome the current bottleneck in phototherapy.

Abstract:Enhancing the antimicrobial properties of Metal‐Organic Framework and imparting antioxidant properties through the strategy of "leveraging strengths and compensating weaknesses", which maximizes the potential of ammonium glycyrrhiza to improve the sterilization capacity of porphyrins in light and low oxygen environments. This strategy helps the development of integrated functional materials and offers nanoplatform for bacteriostatic preservation. Porphyrins, known for generating toxic singlet oxygen (1O2) to combat bacteria, face challenges such as hydrophilicity and limited lifespan and 1O2 yield. Conversely, triterpenoid compounds like ammonium glycyrrhizinate (AG) offer antioxidative and antibacterial properties but lack efficacy and stability. Combining them in Metal‐Organic Frameworks (MOFs) yields dual‐ligand zirconium (Zr)‐basedMOFs (M‐TG), capitalizing on porphyrins' membrane‐disrupting ability and AG's inhibition of bacterial membrane synthesis for a synergistic antibacterial effect. M‐TG resolves activity loss, enhances reactive oxygen species (ROS) yield, and extends stability, achieving a remarkable 99.999% sterilization rate. This innovative approach maximizes ligand properties through synergistic effects, promising significant advancements in antibacterial material design.

Abstract:The rising dangers of bacterial infections have created an urgent need for the development of a new generation of antibacterial technologies and therapeutics. Antibacterial photodynamic therapy (PDT), considered as a non‐invasive treatment with no drug resistance, has become a new promising photochemistry‐involved treatment strategy. Titanium oxide (TiO2) is proved to be a very efficient PDT agent among the photosensitive materials, while the property of a large band‐gap of TiO2 make it only be excited by ultraviolet light, which is harmful to organisms. In this work, a novel ligand‐to‐metal charge transfer (LMCT) mediated TiO2 PDT strategy was proposed via the harmless near‐infrared light irradiation. By choosing a mussel‐inspired material, polydopamine (PDA) was involved in forming mesoporous TiO2@PDA nanoparticles (mTiO2@PDA NPs). The catechol groups of PDA can attach the TiO2 tightly even in colloidal environments, and can also form the LMCT bridge, exciting TiO2 to exert PDT function via 808 nm irradiation. Combining the sonodynamic therapy (SDT) of TiO2 and the photothermal therapy properties of PDA, this simple structure mTiO2@PDA enables synergistic antibacterial applications with multiple functions under the dual excitation of NIR and ultrasound. Abstract:This manuscript highlights a novel application of metal–organic frameworks (MOFs) in enhancing hydrophobic cyanine dye‐based phototherapy. Abstract:Herein,a metal-organic framework（MOF） was modified using polydopamine（PDA） to develop the MOFPDA as a photoresponsive bacteria-killing agent under 660 nm light irradiation.The modification using PDA led to the production of not only more heat,but also much more ~1O 2 .This is because the PDA could interact with the porphyrin ring of the MOF through π-π interaction and the charge transfer between PDA and the MOFs decreases the ene rgy of the band of hybrid nanoparticles.In addition,greater levels of hyperthermia induced by PDA modification accelerated the charge trans fe r,which significantly strengthened the photocatalytic perfo rmance of MO F-PDA.Furthermore,after modification,the light abso rbance and water dispersibility of nanoparticles were both enhanced;both are important for the improvement of photocatalytic and photothermal properties.Consequently,MOF-PDA exhibited the highly effective antibacterial efficacy of 99.62 % and 99.97 % against Staphylococcus aureus and Escherichia coli,respectively,under 20 min 660 nm light irradiation.

Abstract:Bacterial infections pose a huge threat to human health due to the inevitable emergency of drug resistance. Metal-organic frameworks (MOFs) consisting of metal ions and organic linkers, as emerging efficient antibacterial material, have the merits of structural flexibility and adjustable physicochemical property. With assistance of photosensitive agents as organic linkers, MOFs have great potential in antibacterial application through photocatalytic therapy by the generation of reactive oxygen species (ROS). However, the limited light use efficiency and short lifespan of ROS are two obstacles for their applications. Inspired by the semiconductor heterostructure in photocatalysis, we rationally design and precisely synthesize MOFs based heterostructures, in which the TiO 2 nanoclusters are filled into the pores of Cu-TCPP nanosheets (i.e. TiO 2 NCs@Cu-TCPP HSs). And the composite materials possess three-dimensional (3D) hierarchical architectures, which have advantages of large surface area, excellent light-absorbing ability and photocatalytic efficiency. Significantly, this novel material displays >99.99% antibacterial efficiency against E. coli and S. aureus within 30 min and preserves the excellent antibacterial ability during reusing three times, which is superior to recently reported photocatalystic-based antibacterial materials. Our study provides new insights into the energy band engineering for enhanced antibacterial performance, paving a way for designing advanced clinical wound dressings.

Abstract:Abstract Bismuth metal–organic frameworks (MOFs) as catalysts are scarce. Herein, the first MOF comprising meso‐ tetra(4‐carboxyphenyl) porphine (TCPP) as an organic ligand and Bi ion as a metal node, denoted as Bi‐TCPP, is synthesized by a convenient one‐step hydrothermal method. Compared to other 2D (Cu‐TCPP) or 3D (PCN‐222) MOFs with TCPP as a ligand, Bi‐TCPP has ultrafast singlet oxygen generation ability and a high photothermal conversion efficiency of 49.5% under the irradiation of 660 nm light. This is due to Bi‐TCPP's strong ligand‐to‐metal charge transfer (LMCT) events that form singlet oxygen ( 1 O 2 ). Further, when the electrons and holes above the bandgap relax to the band edge, the excess energy is converted into heat. In vitro and in vivo experiments show excellent antibacterial and repair effects. Hence, the synthesis of Bi‐TCPP provides new ideas for the study of rare bismuth‐based MOFs, and its excellent biocompatibility provides a possibility for further application.
Enhanced photocatalytic activity and photothermal effects of cu-doped metal-organic frameworks for rapid treatment of bacteria-infected wounds

Abstract:<h2 class="section-title u-h3 u-margin-l-top u-margin-xs-bottom">Abatract</h2>Owing to the increasing drug-resistance, antibiotics-free strategy is urgent for the therapy of bacterial infections. Herein, Cu2+ is introduced into the porphyrin ring of PCN 224, which enhances the photocatalytic property of MOFs. The doped Cu2+ could trap electrons, thus suppresses electron-hole recombination and accelerates the transfer of carriers under 660 nm light irradiation, thereby enhancing the yields of reactive oxygen species (ROS). Furthermore, the Cu-MOFs' photothermal effect is strengthened due to the d–d transition. However, overdoping of Cu2+ weakens the photocatalytic and photothermal properties due to the trapping of more photo-electrons by Cu2+ and the decreased light absorption of porphyrin at 660 nm. Doping with 10% Cu2+ endows the MOFs with the best antibacterial efficacy of 99.71% against Staphylococcus aureus within 20 min because of the synergistic effects of ROS and heat. Furthermore, the in vivo results show that Cu2+-MOFs kill bacteria efficiently, thus accelerating wound healing.

Abstract:Injured tissues are susceptible to infections that suppress wound healing. Rapid and safe sterilization is therefore urgent for bacteria-infected wounds, especially under harsh conditions without antibiotic availability. In this work, we developed a photoresponsive metal-organic framework (MOF) heterojunction that responds to 660 nm light irradiation. It is composed of two kinds of MOFs (Prussian blue [PB] and PCN-224), which exhibited an enhanced photocatalytic performance due to the accelerated charge transfer of the heterojunction and the fast separation of photogenerated electron-hole pairs between PB and PCN-224. The combination of the enhanced photocatalytic performance and the intrinsic photothermal effect of PB endowed the MOFs heterojunction with highly effective sterilizing rates of 99.84% and 99.3% against Staphylococcus aureus and its biofilm after 660 nm light irradiation for 15 min, respectively. The iron and zirconium ions released from PB-PCN-224 composites were biocompatible and their cytotoxicity was negligible. More importantly, in vivo experiments showed that PBPCN-224 can expedite wound healing.

Abstract:The development of nanotheranostic agents is attracting attention as an innovative strategy to synergistically detect bacterial infection and eliminate bacteria. A novel MOF (ZIF-67)-armored zinc peroxide core–shell structure with a loaded organic NIR probe (ONP) is reported herein. These nanomaterials serve as “reactive oxygen species factories” that eradicate drug resistant bacteria with image guided and in situ activated photodynamic therapy (PDT). The up-regulated levels of ONOO− at infected sites can be detected by the ONP. ONP could react with ONOO− generating MB as an imaging agent and as a photosensitizer. The ZnO2@ZIF-67(ZZ) nanogenerator simultaneously generates O2 and H2O2 in an infected, weakened acidic microenvironment. The PDT can be applied on-demand to eliminate bacteria depending on illumination of the probe. The in situ activated PDT strategy can effectively exterminate drug-resistant bacteria, using small amounts of nanomaterials, with no obvious adverse effects in vitro and in vivo. The ONP@ZnO2@ZIF-67(ONP@ZZ) nanoagent simultaneously eliminates drug-resistant bacteria, reduces inflammation, and accelerates wound healing. This novel nanotheranostic agent can act as an effective antibacterial platform by integrating infection detection and bacterial eradication synergistically, and bring new light for the increasingly severe challenge of drug-resistant bacteria in clinic.

Abstract:Abstract The synergistic effect of antibacterial and anti-inflammatory is needed to overcome the problem of wound healing difficulties. Based on the favorable antibacterial and anti-inflammatory effect of zinc ions (Zn2+) and the physicochemical properties of metal organic frameworks (MOFs), we prepared nanosized zinc-based MOF: Zn-BTC with the ability to slowly release Zn2+. In cellular levels, Zn-BTC possessed lower toxicity to fibroblasts and enhanced capacity of cell proliferation and migration. It also had good bactericidal effect on multiple drug-resistant bacteria by reducing 41.4% MRSA and 47.2% Escherichia coli. In addition, Zn-BTC also displayed the ability of lowering the expression of antioxidant genes: superoxide dismutase 1, superoxide dismutase 2 and interleukin 6, and enhancing the expression of wound healing genes: transforming growth factors-β and type I collagen. Finally, it also demonstrated that Zn-BTC could effectively improve the skin wound healing of SD rats and had no toxicity on major organs. The favorable biocompatibility, antibacterial and anti-inflammatory properties of Zn-BTC gave a new insight of designing novel MOFs for promoting skin wound healing.

Abstract:Skin is the first natural barrier of the human body. Bacterial infection seriously hinders the healing process of skin wounds and poses a great threat to human health. Therefore, it is particularly urgent to develop new antimicrobial strategies for bacterial pathogen clearance and wound healing. In this study, a metal-organic framework (MOF), Fe-MIL88B-NH2, was incorporated with photosensitizer indocyanine green (ICG) to construct composite nanoparticles (MOF@ICG NPs) with multiple antibacterial activities. Under mild near-infrared (NIR) irradiation, the photosensitizer ICG in the MOF@ICG NPs undergoes photothermal conversion (~45 °C) and photodynamic reactions to generate heat and singlet oxygen (1O2). In addition, the Fenton reaction of the NPs with Hydrogen peroxide (H2O2) in the bacterial infection microenvironment resulted in the generation of hydroxyl radicals (·OH), thus achieving the three-mode combination of low-temperature photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT). The experimental results in vitro showed that MOF@ICG MPs had excellent antibacterial property and good cytocompatibility, which presented some ability to promote the migration of L-929 fibroblasts. Furthermore, under NIR irradiation, MOF@ICG NPs could significantly kill bacteria and promote skin wound healing according to the results of animal experiments. The wound healing rate reached 87.1% after 7 days of treatment. The research results break through the limitations of single-mode antibacterial technology, and provide certain theoretical guidance and technical support for the research and development of new antibacterial materials.

Abstract:Open wounds are prone to infection and difficult to heal, which even threatens the life of patients because bacterial infections can induce other lethal complications without prompt treatment. The commonly used antibiotics treatment for bacterial infections has been reported to cause globally bacterial resistance and even the occurrence of superbacteria. The highly effective and antibiotic-independent therapeutic strategies are urgently needed for treating various kinds of bacteria-infected diseases. In this work, we synthesized an eco-friendly nanohybrid material (ZnDMZ) consisting of a kind of biodegradable metal organic framework (MOF, ZIF-8) combined with Zn-doped MoS2 (Zn-MoS2) nanosheets, which exhibited great ability to kill bacteria and promote the healing of bacteria-infected wounds under 660 nm light irradiation. The underlying mechanism is that besides the local hyperthermia, the nanohybrid material exhibits enhanced photocatalytic performance than single component in it, i.e., it can also be excited by 660 nm light to produce more oxygen radical species (ROS) due to the following factors. On one hand, the Zn doping can reduce the work function and the band gap of MoS2, which promotes the movement of photoexcited electrons to the surface of the material. On the other hand, the combination between Zn-MoS2 and MOF induces the formation of a built-in electric field due to their work function difference, thus accelerating the separation of photoexcited electron-hole pairs. Because of the synergy of photocatalytic effect, photothermal effect and the released Zn ions, the synthesized ZnDMZ possessed a highly effective antibacterial efficacy of 99.9% against Staphylococcus aureus under 660 nm light irradiation for 20 min without cytotoxicity. In vivo tests showed that this nanohybrid material promoted the wound healing due to the released Zn ions. This nanohybrid will be promising for rapid and portable treatment of bacteria-infected open wounds in pathogenic bacteria contaminated environments.

Abstract:Abstract Bacterial infection poses a constant threat to human health. It is crucial to develop cost‐effective and multifunctional solutions to combat bacteria. In this study, we have taken inspiration from artificial photosynthesis and creatively formulated a hydrogel containing a photocatalytic metal‐organic cluster (MOC) for wound healing and antibacterial purposes. Complete photocatalytic cycles have been achieved by combining the oxidative Ti‐center and the reductive Cu‐center, in which reactive oxygen species ( 1 O 2 and ·OH) have been generated. The MOC has the capability to eliminate S. aureus and E. coli at a concentration of 40 μg·mL −1 . In addition, the hydrogel formulation (H‐MOC) has been applied to wounds infected with S. aureus , resulting in improved healing efficiency. Nanostructure‐Mediated Photothermal Effect for Reinforcing Physical Killing Activity of Nanorod Arrays

Abstract:A novel antibacterial strategy utilizes nanostructured "light traps" mediated PTT to enhance the physical puncture of bacteria by nanorod arrays, effectively eliminating biofilm infections without requiring additional photosensitivities. Combined with nitric oxide therapy, this method effectively prevents MRSA infection on the surface of titanium implants, providing a promising approach for sustainable antibacterial implant surfaces. The physical killing of bacteria based on surface topography has attracted much attention due to the sustainable and safe prevention of biofilm formation. However, the antibacterial efficiency of biomedical implants derived solely from nanostructures or microstructures is insufficient to combat bacteria against common infections, such as methicillin‐resistant Staphylococcus aureus with thick cell walls. Herein, photothermal therapy is carried out in the presence of nanorod arrays to mitigate infection of biomedical implants. Different from traditional photothermal therapy relying on a photosensitizer, the photothermal effect is mediated by light traps rendered by the nanorod arrays, and consequently, the photosensitizer is not needed. Finite element simulations and experiments are performed to elucidate the light‐to‐thermal conversion mechanism. This photothermal platform, in conjunction with thermosensitive nitric oxide therapy, is applied to treat titanium implant infection. The nanostructure‐mediated photothermal effect destroys bacterial cell walls by inhibiting peptidoglycan synthesis and increasing the membrane permeability by affecting fatty acid synthesis. Furthermore, the nanorods synergistically puncture the bacterial membrane easily as demonstrated by experiments and transcriptome analysis. The results provide insights into the development of efficient antibacterial treatment of implants by combining nanostructures and photothermal therapy.

Abstract:Surface modification of biomedical materials and devices using versatile nanocomposite coatings holds great promise for improving functionalities to defend against life-threatening bacterial infections. In this study, a one-step surface modification strategy was developed to deposit gold nanorods (AuNRs)- and curcumin (CUR)-encapsulated zeolitic imidazolate framework-8 (ZIF-8) nanoparticles (AuNRs-ZIF-CUR NPs or AZC) onto phytic acid (PA)-ε-polylysine (Ply) network coatings. In the solution mixture of PA, Ply and AZC, PA interacted with Ply via electrostatic interactions, and can also bind to AZC via metal chelation. The as-formed AZC-PA-Ply aggregates could be deposited onto various substrates via surface adhesion of PA and gravitational effects. The physicochemical and antibacterial properties of the AZC-PA-Ply network coatings on polydimethylsiloxane (PDMS) substrates were evaluated. The sustained release of zinc ions and CUR, as well as the contact-killing ability of Ply, endowed the AZC-PA-Ply network coatings with good antibacterial chemotherapeutic effects. In addition, the embedded AuNRs in the AZC-PA-Ply network coatings exhibited excellent photothermal conversion efficiency for the ablation of bacteria. Upon near-infrared (NIR) laser irradiation, the AZC-PA-Ply-coated PDMS surfaces exhibited strong antibacterial effects by disrupting the membrane integrity and cellular functions of the adhered bacteria. Thus, the AZC-PA-Ply network coatings displayed combined antibacterial chemotherapeutic and photothermal therapeutic effects. Furthermore, the AZC-PA-Ply-coated PDMS substrates exhibited effective bacterial infection prevention and good biocompatibility in an in vivo implant model. Hence, the versatile AZC-PA-Ply network coatings are potentially useful as a multi-modal antibacterial platform to eliminate infectious bacterial pathogens in biomedical applications.
