Abstract:Bacterial infections have become major threats to public health all over the world. With the emergence of antibiotic resistance, it is urgent to develop novel antimicrobial materials to efficiently overcome drug resistance with high bactericidal activity. In this work, UiO-66-NH-CO-MoS2 nanocomposites (UNMS NCs) are constructed through the amidation reaction. The UNMS NCs are positively charged which is beneficial for capturing and restricting bacteria. Significantly, UNMS NCs possess a synergistic bactericidal efficiency based on near-infrared irradiation (808 nm) regulated combination of photothermal, photodynamic, and peroxidase-like enzymatic activities. Both the photodynamic property and nanozymatic activity of UNMS NCs can lead to the generation of reactive oxygen species. The UNMS NCs show high catalytic activity in a wide pH range and exhibit excellent antibacterial ability against ampicillin-resistant Escherichia coli and methicillin-resistant Staphylococcus aureus with negligible cytotoxicity. Interestingly, due to the 808 nm irradiation-induced hyperthermia in the presence of UNMS NCs, the glutathione oxidation process can be accelerated, resulting in bacterial death more easily. Mice wound models are established to further manifest that UNMS NCs can promote wound healing with good biosafety in living systems.
Cuprous Oxide–demethyleneberberine Nanospheres for Single Near-Infrared Light-Triggered Photoresponsive-Enhanced Enzymatic Synergistic Antibacterial Therapy

Abstract:In this work, novel cuprous oxide-demethyleneberberine (Cu2O-DMB) nanomaterials are successfully synthesized for photoresponsive-enhanced enzymatic synergistic antibacterial therapy under near-infrared (NIR) irradiation (808 nm). Cu2O-DMB has a spherical morphology with a smaller nanosize and positive ζ potential, can trap bacteria through electrostatic interactions resulting in a targeting function. Cu2O-DMB nanospheres show both oxidase-like and peroxidase-like activities, and serve as a self-cascade platform, which can deplete high concentrations of GSH to produce O2˙- and H2O2, then H2O2 is transformed into ˙OH, without introducing exogenous H2O2. At the same time, Cu2O-DMB nanospheres become photoresponsive, producing 1O2 and having an efficient photothermal conversion effect upon NIR irradiation. The proposed mechanism is that the generated ROS (O2˙-, ˙OH and 1O2) and hyperthermia can have synergetic effects for killing bacteria. Moreover, hyperthermia is not only beneficial for destroying bacteria, but also effectively enhances the efficiency of ˙OH production and accelerates GSH oxidation. Upon NIR irradiation, Cu2O-DMB nanospheres exhibit excellent antibacterial ability against methicillin-resistant Staphylococcus aureus (MRSA) and ampicillin-resistant Escherichia coli (AREC) with low cytotoxicity and bare bacterial resistance, destroy the bacterial membrane causing an efflux of proteins and disrupt the bacterial biofilm formation. Animal experiments show that the Cu2O-DMB + NIR group can efficiently treat MRSA infection and promote wound healing. These results suggest that Cu2O-DMB nanospheres are effective materials for combating bacterial infections highly efficiently and to aid the development of photoresponsive enzymatic synergistic antibacterial therapy.
Metal-Organic Framework/Ag-Based Hybrid Nanoagents for Rapid and Synergistic Bacterial Eradication

Abstract:Recent emerged metal-organic frameworks (MOFs), as superior drug carriers, provide novel strategies to combat pathogenic bacterial infections. Although various antibacterial metal ions can be easily introduced in MOFs for chemical bacterial ablation, such a single-model bactericidal method suffers from high-dose use, limited antibacterial efficiency, and slow sterilization rate. Hence, developing a dual bactericidal system is urgently required. Herein, we report an MOF/Ag-derived nanocomposite with efficient metal-ion-releasing capability and robust photo-to-thermal conversion effect for synergistic sterilization. The MOF-derived nanocarbon consisting of metallic zinc and a graphitic-like carbon framework is first synthesized, and then Ag nanoparticles (AgNPs) are evenly introduced via the displacement reaction between Zn and Ag+. Upon near-infrared irradiation, the fabricated nanoagents can generate massive heat to destroy bacterial membranes. Meanwhile, abundant Zn2+ and Ag+ ions are released to make chemical damage to bacterial intracellular substances. Systematic antibacterial experiments reveal that such dual-antibacterial effort can endow the nanoagents with nearly 100% bactericidal ratio for highly concentrated bacteria at a very low dosage (0.16 mg/mL). Furthermore, the nanoagents exhibit less cytotoxicity, which provides potential possibilities for the applications in the biological field. In vivo assessment indicates that the nanocomposites can realize rapid and safe wound sterilization and are expected to be an alternative to antibiotics. Overall, we present an easily fabricated structure-engineered nanocomposite with chemical and photothermal effects for broad-spectrum bacterial sterilization.
Ag-Activated Metal−Organic Framework with Peroxidase-like Activity Synergistic Ag+ Release for Safe Bacterial Eradication and Wound Healing

Abstract:Silver nanoparticles (Ag NPs), a commonly used antibacterial nanomaterial, exhibit broad-spectrum antibacterial activity to combat drug-resistant bacteria. However, the Ag NPs often causes a low availability and high toxicity to living bodies due to their easy aggregation and uncontrolled release of Ag+ in the bacterial microenvironment. Here, we report a porous metal−organic framework (MOF)-based Zr-2-amin-1,4-NH2-benzenedicarboxylate@Ag (denoted as UiO-66-NH2-Ag) nanocomposite using an in-situ immobilization strategy where Ag NPs were fixed on the UiO-66-NH2 for improving the dispersion and utilization of Ag NPs. As a result, the reduced use dose of Ag NPs largely improves the biosafety of the UiO-66-NH2-Ag. Meanwhile, after activation by the Ag NPs, the UiO-66-NH2-Ag can act as nanozyme with high peroxidase (POD)-like activity to efficiently catalyze the decomposition of H2O2 to extremely toxic hydroxyl radicals (·OH) in the bacterial microenvironment. Simultaneously, the high POD-like activity synergies with the controllable Ag+ release leads to enhanced reactive oxygen species (ROS) generation, facilitating the death of resistant bacteria. This synergistic antibacterial strategy enables the low concentration (12 μg/mL) of UiO-66-NH2-Ag to achieve highly efficient inactivation of ampicillin-resistant Escherichia coli (AmprE. coli) and endospore-forming Bacillus subtilis (B. subtilis). In vivo results illustrate that the UiO-66-NH2-Ag nanozyme has a safe and accelerated bacteria-infected wound healing.

Abstract:Bacterial infections caused by multidrug-resistant bacteria pose an extremely serious treat to human health However, nanoscale antibacterial materials can effectively inhibit bacterial growth without inducing bacterial resistance. Herein, we have developed a highly effective antibacterial agent by immobilizing silver nanoparticles (AgNPs) within a novel covalent organic framework (COF). The COF BDP/CD-S-3 -Ag system was synthesized in situ using BODIPY phoyosensitizer (BDP) and carbon quantum dots (CD-S-3) as precursors. The optimized COF BDP/CD-S-3 -Ag system exhibited excellent photodynamic therapy (PDT) and photothermal therapy (PTT). Importantly, the COF BDP/CD-S-3 -Ag system, with many active groups, can capture and kill bacteria effectively by interacting with them, thereby contributing to improved antibacterial efficacy. As expected, the designed COF BDP/CD-S-3 -Ag system combines the advantages of COF BDP/CD-S-3 (preventing the aggregation of AgNPs and having the ability to capture bacteria) and AgNPs (antibacterial ability), demonstrating excellent antibacterial efficacy. This study developed a new class of antibacterial materials and took an important step forward in promoting the application of COF-based antibacterial materials.

Abstract:The continuous emergence of drug-resistant bacteria has become a serious threat to human and animal health. Developing new non-antibiotic therapeutic drugs with high efficiency and low resistance is a promising strategy for combating drug-resistant bacteria. Here, a novel composite structure based on a hollow metal-organic framework (MOF) wrapped Au nanorod and loaded Pt nanoparticles (Au NR@H-ZIF-8@Pt) is designed for combating drug-resistant bacteria. The Au NR@H-ZIF-8@Pt composite structures serve a twofold purpose: (1) showing superior photothermal conversion efficiency under near-infrared light irradiation, thus achieving an antibacterial effect based on photothermal effect, and (2) exhibiting high-efficiency peroxidase-like activity, and effectively killing bacteria by reactive oxygen species (ROS) generated in situ . By combining photothermal and catalytic effects for in vitro antibacterial experiments, Au NR@H-ZIF-8@Pt exhibits efficient, rapid, broad-spectrum antibacterial activity (99.7% for Pseudomonas aeruginosa and 98.3% for Staphylococcus aureus). In addition, in vivo experiments further demonstrate the advantages of the material in promoting bacterial infection wound healing. This study not only facilitates the design of novel biological nanomaterials, but also provides new ideas for combating drug-resistant bacteria.

A Triple-Enhanced Chemodynamic Approach Based on Glucose-Powered Hybrid Nanoreactors for Effective Bacteria Killing

Abstract:Rapid evolution of multidrug resistance in bacterial pathogens is outpacing the development of new antibiotics, and chemodynamic therapy (CDT) provides an excellent alternative. However, achieving highly efficient CDT is still a great challenge, since the pH in the infection site is close to neutral and the supply of H2O2 is inadequate. We herein constructed the antibacterial nanoreactors. Indocyanine green (ICG) and glucose oxidase (GOx) were incorporated into homologous zeolitic imidazolate framework-8 (ZIF-8) nanoparticles coating with metal polyphenol network (MPN) composed by Fe3+ and tannic acid (TA). The well-designed nanoreactors could simultaneously break the pH and H2O2 limitations, and generate hyperthermia under irradiation, thus realizing a triple-enhanced CDT for high-efficiency sterilization. Furthermore, the nanoreactors could combine CDT with photothermal therapy (PTT) and photodynamic therapy (PDT), which not only improved the bactericidal efficiency and broadened the antibacterial spectrum, but also alleviated the antibiotics resistance issues. Remarkably, the proposed nanoreactors achieved a robust in vitro bacterial killing against Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) and Gram-negative Pseudomonas aeruginosa. The nanoreactors achieved an 99.7% MRSA reduction in an MRSA-induced murine abscess model accompanied with negligible toxicity. Overall, this study provides a promising strategy for multiple-enhanced CDT and multimodal combined therapy for pathogenic infections.
Silver‐Infused Porphyrinic Metal–Organic Framework: Surface‐Adaptive, On‐Demand Nanoplatform for Synergistic Bacteria Killing and Wound Disinfection

Abstract:The fabrication of functional nanoplatforms for combating multidrug‐resistant bacteria is of vital importance. Among them, silver nanoparticles (Ag NPs) have shown an antibacterial effect; however, the remainder cores of Ag NPs after use might have a toxic effect on humans. Thus, Ag ions based materials have been fabricated to substitute Ag NPs for antibacterial applications. Nevertheless, the always‐on release state leads to the low biocompatibility, which limits their biomedical applications. In addition, the single effect also restricts their antibacterial ability. Herein, a powerful surface‐adaptive, on‐demand antimicrobial nanoplatform is fabricated by coating hyaluronic acid (HA) on Ag ions loaded photosensitive metal‐organic frameworks to exhibit a strong synergistic effect. The nanoplatform shows good biocompatibility with nontargeted cells, as negatively charged HA can prevent the release of Ag ions. While in the presence of targeted bacteria, the secreted hyaluronidase can degrade HA on the nanoplatform and produce positively charged nanoparticles, which display increased affinity to bacteria and show a strong synergistic antibacterial effect owing to the released Ag ions and generated reactive oxygen species under visible light irradiation. Importantly, due to the outstanding on‐demand antimicrobial performance, the nanoplatform also shows great effects on treating multidrug‐resistant bacteria infected wounds in mice models.

Abstract:A controllable antibacterial Ag@MOF@PDA is constructed by functionalizing cyclodextrin metal–organic frameworks with polydopamine (PDA) and ultrafine Ag NPs. The PDA coating enables its long‐term bactericidal capability by gradual releasing of Ag+, and NIR‐mediated hyperthermia can accelerate its release rate, which realizes a synergistic chemical‐photothermal capacity to combat bacteria and biofilm infections, representing a potential antibiotic‐free alternative in the "post‐antibiotic era." Bacterial infections pose a significant threat to global public health; therefore, the development of novel therapeutics is urgently needed. Herein, a controllable antibacterial nanoplatform utilizing cyclodextrin metal–organic frameworks (CD‐MOFs) as a template to synthesize ultrafine silver nanoparticles (Ag NPs) in their porous structure is constructed. Subsequently, polydopamine (PDA) is encapsulated on the CD‐MOFs' surface via dopamine polymerization to enhance the water stability and enable hyperthermia capacity. The resulting Ag@MOF@PDA generates localized hyperthermia and gradually releases Ag+ to achieve long‐term photothermal‐chemical bactericidal capability. The release rate of Ag+ can be accelerated by NIR‐mediated heating in a controllable manner, quickly reaching the effective concentration and reducing the frequency of medication to avoid potential toxicity. In vitro experiments demonstrate that the combined antibacterial strategy can not only effectively kill both gram‐negative and gram‐positive bacteria, but also directly eradicate mature biofilms. In vivo results confirm that both bacterial‐ and biofilm‐infected wounds treated with a combination of Ag@MOF@PDA and laser exhibit satisfactory recovery with minimal toxicity, displaying a superior therapeutic effect compared to other groups. Together, the results warrant that the Ag@MOF@PDA realizes synergistic antibacterial capacity and controllable release of Ag+ to combat bacterial and biofilm infections, providing a potential antibiotic‐free alternative in the "post‐antibiotic era."

Abstract:Numerous nanomedicines currently emerge to reduce the dramatic threat in antibiotics resistance for antibacterial application against severe bacterial infections, while it is restricted by over-reacted immune response to pathogenic bacteria. Herein, enzymatic activity is introduced into the zeolitic imidazolate framework-8 (ZIF-8) to achieve sterilization by releasing Zn ions, as well as inflammation regulation through the variable valence of Mn ions that are uniformly doped into its framework. Within this simple metal organic framework (MOF) structure design, Mn-ZIF-8 possesses the co-existence of Mn2+/Mn4+ to endow the nanocomposite with the anti-inflammatory capabilities, which can be adjusted through the redox environment. The enzymatic activity of Mn ions and superiority of pore structure of ZIF-8 are effectively combined to realize the substrate selection via reactant molecular size and high-efficiency internal catalytic performance. By such design, this nanocomposite would not only exhibit an excellent antibacterial performance against pathogenic bacteria, but also reshape the inflammatory immunity by regulating macrophage polarization to suppress over-reacted inflammation, leading to a favorably therapeutic efficiency on bacteria-infected wound healing in animal models. Taken together, this nanoplatform provides effective approach for accelerating infected wound healing via bacteria killing and inflammation modulation, and may be extended for the therapy of other severe bacteria-induced infections.

Abstract:Two-dimensional (2D) metal organic framework (MOF) nanosheets have been demonstrated to be promising templates for preparing functional 2D composites. Herein, we report the preparation of silver nanoparticle (Ag NP)-decorated 2D porphyrinic MOF nanosheets (denoted as Ag/Co-TCPP NSs) with a high efficiency for combined antibacterial therapy. The as-prepared Ag/Co-TCPP NSs feature both the advantages of Co-TCPP NSs and Ag NPs, which exhibit excellent antibacterial properties against both Gram-positive ( S. aureus ) and Gram-negative ( E. coli ) bacteria. Under a 660 nm laser irradiation, Ag/Co-TCPP NSs locally generated singlet oxygen ( 1 O 2 ) and promoted Ag NPs to be partly degraded for sustained release of toxic Ag + , achieving a synergistic sterilization effect. This effective strategy for achieving 2D MOF nanosheets with in situ loaded metal nanoparticles provides a guideline to broaden the diverse applications of MOFs and contributes to the development of new intelligent metal-based antibacterial nanomaterials.

Abstract:As one of the common reactive oxygen species, H2 O2 has been widely used for combating pathogenic bacterial infections. However, the high dosage of H2 O2 can induce undesired damages to normal tissues and delay wound healing. In this regard, peroxidase-like nanomaterials serve as promising nanozymes, thanks to their positive promotion toward the antibacterial performance of H2 O2 , while avoiding the toxicity caused by the high concentrations of H2 O2 . In this work, ultrasmall Au nanoparticles (UsAuNPs) are grown on ultrathin 2D metal-organic frameworks (MOFs) via in situ reduction. The formed UsAuNPs/MOFs hybrid features both the advantages of UsAuNPs and ultrathin 2D MOFs, displaying a remarkable peroxidase-like activity toward H2 O2 decomposition into toxic hydroxyl radicals (·OH). Results show that the as-prepared UsAuNPs/MOFs nanozyme exhibits excellent antibacterial properties against both Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria with the assistance of a low dosage of H2 O2 . Animal experiments indicate that this hybrid material can effectively facilitate wound healing with good biocompatibility. This study reveals the promising potential of a hybrid nanozyme for antibacterial therapy and holds great promise for future clinical applications.

Enzyme-triggered on-demand release of a H2O2-self-supplying CuO2@Fe3O4 nanoagent for enhanced chemodyamic antimicrobial therapy and wound healing

Abstract:Nanoagents for chemodynamic therapy (CDT) hold a promising future in the field of antimicrobials, especially copper peroxide (CuO2) (CP) nanomaterials which have garnered significant attention due to their ability to self-supply H2O2. Nevertheless, the poor stability of CuO2 remains a critical challenge which restricts its practical application in the antibacterial field. In this study, an advanced nano-antimicrobial system HA-CP@Fe3O4 with enzyme-responsive properties is developed by coating hyaluronic acid (HA) on CuO2-loaded iron tetraoxide nanoparticles. The coating of HA not only stabilizes the CuO2 nanomaterials but also provides responsiveness towards the enzyme hyaluronidase, which is typically secreted by some bacteria. The outer layer of HA in HA-CP@Fe3O4 undergoes decomposition in the presence of hyaluronidase-secreting bacteria, resulting in the release of CuO2@Fe3O4. The released CuO2@Fe3O4 then self-supplies H2O2 and generates reactive oxygen species (ROS) within the infected microenvironment through Fenton and Russell effects, to ultimately achieve effective and precise antimicrobial activity. Simultaneously, the magnetic property provided by Fe3O4 allows the substance to be directed towards the infection site. Both in vitro and in vivo tests demonstrated that HA-CP@Fe3O4 exhibited excellent antimicrobial capabilities at low concentration (30 μg mL-1), exceptional biocompatibility and the ability to accelerate wound healing. The findings of this work offer a new and promising approach for targeted and precise CDT.

Iron/Molybdenum Sulfide Nanozyme Cocatalytic Fenton Reaction for Photothermal/Chemodynamic Efficient Wound Healing

Abstract:The issue of bacterial infectious diseases remains a significant concern worldwide, particularly due to the misuse of antibiotics, which has caused the emergence of antibiotic-resistant strains. Fortunately, the rapid development of nanomaterials has propelled significant progress in antimicrobial therapy, offering promising solutions. Among them, the utilization of nanoenzyme-based chemodynamic therapy (CDT) has become a highly hopeful approach to combating bacterial infectious diseases. Nevertheless, the application of CDT appears to be facing certain constraints for its low efficiency in the Fenton reaction at the infected site. In this study, we have successfully synthesized a versatile nanozyme, which was a composite of molybdenum sulfide (MoS2) and iron sulfide (FeS2), through the hydrothermal method. The results showed that iron/molybdenum sulfide nanozymes (Fe/Mo SNZs) with desirable peroxidase (POD) mimic activity can generate cytotoxic reactive oxygen species (ROS) by successfully triggering the Fenton reaction. The presence of MoS2 significantly accelerates the conversion of Fe2+/Fe3+ through a cocatalytic reaction that involves the participation of redox pairs of Mo4+/Mo6+, thereby enhancing the efficiency of CDT. Additionally, based on the excellent photothermal performance of Fe/Mo SNZs, a near-infrared (NIR) laser was used to induce localized temperature elevation for photothermal therapy (PTT) and enhance the POD-like nanoenzymatic activity. Notably, both in vitro and in vivo results demonstrated that Fe/Mo SNZs with good broad-spectrum antibacterial properties can help eradicate Gram-negative bacteria like Escherichia coli and Gram-positive bacteria like Staphylococcus aureus. The most exciting thing is that the synergistic PTT/CDT exhibited astonishing antibacterial ability and can achieve complete elimination of bacteria, which promoted wound healing after infection. Overall, this study presents a synergistic PTT/CDT strategy to address antibiotic resistance, providing avenues and directions for enhancing the efficacy of wound healing treatments and offering promising prospects for further clinical use in the near future.

Abstract:Enhancing the antimicrobial properties of Metal‐Organic Framework and imparting antioxidant properties through the strategy of "leveraging strengths and compensating weaknesses", which maximizes the potential of ammonium glycyrrhiza to improve the sterilization capacity of porphyrins in light and low oxygen environments. This strategy helps the development of integrated functional materials and offers nanoplatform for bacteriostatic preservation. Porphyrins, known for generating toxic singlet oxygen (1O2) to combat bacteria, face challenges such as hydrophilicity and limited lifespan and 1O2 yield. Conversely, triterpenoid compounds like ammonium glycyrrhizinate (AG) offer antioxidative and antibacterial properties but lack efficacy and stability. Combining them in Metal‐Organic Frameworks (MOFs) yields dual‐ligand zirconium (Zr)‐basedMOFs (M‐TG), capitalizing on porphyrins' membrane‐disrupting ability and AG's inhibition of bacterial membrane synthesis for a synergistic antibacterial effect. M‐TG resolves activity loss, enhances reactive oxygen species (ROS) yield, and extends stability, achieving a remarkable 99.999% sterilization rate. This innovative approach maximizes ligand properties through synergistic effects, promising significant advancements in antibacterial material design.

Abstract:This work reports a nanocatalytic antimicrobial agent (Si@Cu NSs) with enhanced CDT via p‐d orbital hybridization strategy and synergistic photothermal effect. This strategy enhances the Fenton‐like reaction of copper atoms and addresses the limited application of nanoparticles in pathophysiological environments. Excellent therapeutic effects demonstrate the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs. Recurrent bacterial infections, impenetrable microbial biofilms, and irremediable antibiotic resistance are the most perilous threats in orthopedic implant‐associated infections (IAIs). Recently, chemodynamic therapy (CDT) has been considered a promising therapy, while the clinical practices for existing CDT agents are limited by the low therapeutic efficiency in physiological circumstances. Herein, it is demonstrated that silica‐copper nanosheets (Si@Cu NSs) exhibit a combined therapeutic photothermal‐chemodynamic strategy for IAIs treatment with superior capacity and biocompatibility. This unique design endows the nanostructure with enhanced copper‐based Fenton‐like properties via p‐d orbital hybridization by incorporating copper nanoclusters (Cu NCs) on silicene nanosheets (Si NSs) matrix, integrating inherent photothermal performance of Si NSs with specific catalytic anti‐infection of Cu NCs against planktonic bacteria and biofilms both in vitro and in vivo. This study not only reveals the promising bio‐application prospects of 2D nanocatalytic biomaterials but also demonstrates the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs.

Designing Single-Atom Active Sites on sp2 -Carbon Linked Covalent Organic Frameworks to Induce Bacterial Ferroptosis-Like for Robust Anti-Infection Therapy

Abstract:With the threat posed by drug-resistant pathogenic bacteria, developing non-antibiotic strategies for eradicating clinically prevalent superbugs remains challenging. Ferroptosis is a newly discovered form of regulated cell death that can overcome drug resistance. Emerging evidence shows the potential of triggering ferroptosis-like for antibacterial therapy, but the direct delivery of iron species is inefficient and may cause detrimental effects. Herein, an effective strategy to induce bacterial nonferrous ferroptosis-like by coordinating single-atom metal sites (e.g., Ir and Ru) into the sp2 -carbon-linked covalent organic framework (sp2 c-COF-Ir-ppy2 and sp2 c-COF-Ru-bpy2 ) is reported. Upon activating by light irradiation or hydrogen peroxide, the as-constructed Ir and Ru single-atom catalysts (SACs) can significantly expedite intracellular reactive oxygen species burst, enhance glutathione depletion-related glutathione peroxidase 4 deactivation, and disturb the nitrogen and respiratory metabolisms, leading to lipid peroxidation-driven ferroptotic damage. Both SAC inducers show potent antibacterial activity against Gram-positive bacteria, Gram-negative bacteria, clinically isolated methicillin-resistant Staphylococcus aureus (MRSA), and biofilms, as well as excellent biocompatibility and strong therapeutic and preventive potential in MRSA-infected wounds and abscesses. This delicate nonferrous ferroptosis-like strategy may open up new insights into the therapy of drug-resistant pathogen infection.
Chemo-photothermal therapy of bacterial infections using metal-organic framework-integrated polymeric network coatings

Abstract:Surface modification of biomedical materials and devices using versatile nanocomposite coatings holds great promise for improving functionalities to defend against life-threatening bacterial infections. In this study, a one-step surface modification strategy was developed to deposit gold nanorods (AuNRs)- and curcumin (CUR)-encapsulated zeolitic imidazolate framework-8 (ZIF-8) nanoparticles (AuNRs-ZIF-CUR NPs or AZC) onto phytic acid (PA)-ε-polylysine (Ply) network coatings. In the solution mixture of PA, Ply and AZC, PA interacted with Ply via electrostatic interactions, and can also bind to AZC via metal chelation. The as-formed AZC-PA-Ply aggregates could be deposited onto various substrates via surface adhesion of PA and gravitational effects. The physicochemical and antibacterial properties of the AZC-PA-Ply network coatings on polydimethylsiloxane (PDMS) substrates were evaluated. The sustained release of zinc ions and CUR, as well as the contact-killing ability of Ply, endowed the AZC-PA-Ply network coatings with good antibacterial chemotherapeutic effects. In addition, the embedded AuNRs in the AZC-PA-Ply network coatings exhibited excellent photothermal conversion efficiency for the ablation of bacteria. Upon near-infrared (NIR) laser irradiation, the AZC-PA-Ply-coated PDMS surfaces exhibited strong antibacterial effects by disrupting the membrane integrity and cellular functions of the adhered bacteria. Thus, the AZC-PA-Ply network coatings displayed combined antibacterial chemotherapeutic and photothermal therapeutic effects. Furthermore, the AZC-PA-Ply-coated PDMS substrates exhibited effective bacterial infection prevention and good biocompatibility in an in vivo implant model. Hence, the versatile AZC-PA-Ply network coatings are potentially useful as a multi-modal antibacterial platform to eliminate infectious bacterial pathogens in biomedical applications.
