Abstract:Objectives: Soil salinity affects the growth of crop plants, leading to reduced productivity, and is a major challenge for wheat production worldwide. Various adaptations and mitigation approaches in combination with tolerant wheat genotypes can be useful for the sustainability of crop production in saline environments. However, the development of salt-tolerant wheat genotypes is one of the best and most efficient solutions for obtaining desirable yields. Considering these issues, an investigation was carried out under hydroponic nutrient culture conditions to assess the genetic variability and selection of salt-tolerant wheat genotypes by categorizing inequitable morphophysiological and genetic variability as well as multivariate analysis. Methods: To meet the objectives of this study, 100 wheat genotypes were tested hydroponically in 0 (control) and 15 dS m-1 salt solutions. Conclusion: For all the wheat genotypes grown under saline conditions, the shoot length (SL), root length (RL), shoot fresh weight (SFW), root fresh weight (RFW), total fresh weight (TFW), shoot dry weight (SDW), root dry weight (RDW), and total dry weight (TDW) decreased significantly. Furthermore, significant variation was observed among the genotypes in terms of their characteristics only under saline conditions. In the case of genetic diversity analysis, a high genotypic coefficient of variation (GCV), phenotypic coefficient of variation (PCV), genetic advance in the percentage of the mean (GAM) and high heritability (h2b) were recorded for all tested wheat genotypes based on the SDW, RDW and TDW. Correlation analysis for both genotypic and phenotypic relationships revealed strong positive correlations for TDW, SDW, TFW and SFW. Principal component analysis (PCA) revealed that TDW, TFW, SDW, and SFW were the most discriminative variables for the wheat genotypes, which was confirmed by discriminant function analysis (DFA). PCA-biplot analysis also revealed significant positive correlations between SDW and SFW and between TDW and TFW. Hierarchical cluster analysis was performed for ten clusters based on the relative performance of the genotypes, where the genotypes were characterized into salt-tolerant, medium-salt-tolerant, medium-salt-susceptible and salt-susceptible groups. Among the genotypes, G11, G25 and G29 under cluster VII were categorized as salt tolerant based on their outstanding performance in terms of characteristics only under saline conditions. D2 analysis proved that the wheat genotypes of this cluster were highly divergent from the other cluster genotypes; as a result, these genotypes might be utilized as parents in the development of salt-tolerant wheat genotypes. The current study concluded that SDW and TDW could be employed as criteria for selecting and defining salt-tolerant genotypes during the early growth stage of wheat.

Abstract:The narrow genetic variation for drought adaptive traits and biomass allocation in wheat ( Triticum aestivum L.) presents a major bottleneck for breeding. Induced mutagenesis creates genetic variation and complements conventional breeding for drought tolerance improvement. The aims of this study were to induce mutations in wheat genotype LM43 using three ethyl methanesulphonate (EMS) treatments, and to develop mutant populations for improving drought tolerance, biomass allocation and agronomic performance. Experiments were conducted under controlled and field conditions at the University of KwaZulu-Natal. Data on percentage germination (%G), days to 90% maturity (DTM), plant height (PH), shoot biomass (SB), root biomass (RB), root-shoot ratio (RSR), spike length (SL), spikelet count (SPS), thousand seed weight (TSW), and grain yield (GY) were collected from M 1 to M 4 generations. Significant ( p < 0.001) differences among individuals and generations were observed for all the assessed traits and the generation × population interaction effects were significant ( p < 0.01) for SB, TSW, and GY due to EMS treatments. The differences among the generations showed that the mutagenic effects were cumulative and exhibited clear segregations in subsequent generations. The new selections with unique biomass allocation, drought response and agronomic performance will be useful for wheat improvement programs.
Identification and Characterization of Wheat Germplasm for Salt Tolerance

Abstract:Salinity is one of the limiting factors of wheat production worldwide. A total of 334 internationally derived wheat genotypes were employed to identify new germplasm resources for salt tolerance breeding. Salt stress caused 39, 49, 58, 55, 21 and 39% reductions in shoot dry weight (SDW), root dry weight (RDW), shoot fresh weight (SFW), root fresh weight (RFW), shoot height (SH) and root length (RL) of wheat, respectively, compared with the control condition at the seedling stage. The wheat genotypes showed a wide genetic and tissue diversity for the determined characteristics in response to salt stress. Finally, 12 wheat genotypes were identified as salt-tolerant through a combination of one-factor (more emphasis on the biomass yield) and multifactor analysis. In general, greater accumulation of osmotic substances, efficient use of soluble sugars, lower Na+/K+ and a higher-efficiency antioxidative system contribute to better growth in the tolerant genotypes under salt stress. In other words, the tolerant genotypes are capable of maintaining stable osmotic potential and ion and redox homeostasis and providing more energy and materials for root growth. The identified genotypes with higher salt tolerance could be useful for developing new salt-tolerant wheat cultivars as well as in further studies to underline the genetic mechanisms of salt tolerance in wheat.

Abstract:Salinity poses a significant challenge to global wheat production. The screening of wheat cultivars for salt tolerance is essential for developing new varieties resilient to salinity and advancing breeding strategies. Identifying key traits from a myriad of morphological, physiological, and molecular markers in screening studies can enhance selection efficiency and expedite the process. In this study, 16 common wheat cultivars were subjected to 0 and 150 mM NaCl stress for ten days during the seedling stage. To assess the salinity tolerance of wheat cultivars comprehensively, we employed a diverse array of morpho-physiological, biochemical, and molecular markers, coupled with multivariate analysis. Salt stress resulted in reduced germination and seedling growth across most wheat cultivars. Notably, the 'Ikizce-96' and 'Demir-2000' genotypes exhibited a higher salinity tolerance index, with the least reduction in morpho-physiological parameters. These cultivars demonstrated elevated osmoregulator proline content and enhanced activity of reactive oxygen species-scavenging antioxidant enzymes. Under 150 mM NaCl, tolerant cultivars exhibited up-regulated transcript expression levels of TaHKT1;4 , TaP5CS , and TaDHN marker genes compared to salt-sensitive cultivars. The accuracy of our findings was validated through principal component analysis, hierarchical clustering analysis, and a correlation network. Proline content, glutathione reductase, catalase, and superoxide dismutase activity emerged as the most reliable indicators of salt tolerance in wheat under saline conditions. The identified salt-tolerant cultivars hold promise as donor parents in salinity tolerance breeding programs.

Abstract:Soil salinity adversely affects plant growth and development, reducing the yield of most crops, including wheat. The highly salt-tolerant wheat germplasm lines W4909 and W4910 were derived from a cross between two moderately salt-tolerant lines, the Chinese Spring (CS)/Thinopyrum junceum disomic addition line AJDAj5 (AJ) and the Ph-inhibitor line (Ph-I) derived from CS/Aegilops speltoides. Molecular markers for gene introgressions in W4909 and W4910 were not reported. Four sequence-tagged site (STS) molecular markers of Ph-I were developed and tested in the above-mentioned lines and the F2 progenies of the two crosses, Anza (AZ) × 4740 (sib of W4910) and Yecora Rojo (YR) × 4728 (sib of W4909). Additionally, homogeneity was assessed in several derivatives of W4909, 4728, W4910, and 4740 using the four markers. The four STS markers are not associated with salt tolerance, but they provide an indication of the transfer of chromatin in 3B chromosome of Ae. speltoides via Ph-I. Moreover, salt tolerance and leaf sodium concentration were determined in CS, AJ, Ph-I, 7151 (progeny of W4909), 7157 (progeny of W4910), AZ, and YR under salt treatment and control. Surprisingly, AJ had the lowest leaf sodium concentration under the control and salt treatment, indicating greater sodium exclusion than that in CS, AZ, and YR. This low level of leaf sodium concentration was heritable from 4740 to its hybrid progenies. On the other hand, the higher leaf sodium concentration, indicative of the tissue tolerance to salinity in Ph-I, had been inherited by both W4909 and W4910 and then transmitted to their hybrid progenies. One offspring line each in both W4909 and W4910 (7762 and 7159, respectively) were homozygous for the three molecular markers and lacked the marker psr1205 of Su1-Ph1 gene, making them better materials than the original lines for future research on, for example, whole-genome sequencing and gene mining. The implications of these findings for the utilization of W4909 and W4910 in breeding salt-tolerant wheat cultivars are discussed.

Abstract:Soil salinity is one of the major constraints to crop production worldwide. The multifaceted nature of salinity tolerance traits complicates plant screening and the identification of salt‐tolerant germplasm to be used for the genetic advancement of corps. Many screening criteria have been suggested to distinguish between genotypes. Most of these were applied under controlled environmental conditions and limited to one developmental stage of plants. As a result, most of the reported tolerance could not be validated under field conditions. This study employed a membership function value (MFV) to assess NaCl tolerance of eight wheat (Triticum aestivum) genotypes measured at germination, vegetative and reproductive stages, as an integrative tool for the overall plant performance. Salt stresses had an adverse effect on plant physiological (residual transpiration, stomatal density, chlorophyll fluorescence characteristics; leaf N, Na+ and K+ content) and agronomical (plant height; biomass; root and tiller number; grain) characteristics. Based on this assessment, plants were divided into three contrasting groups: salt tolerant, moderately salt tolerant and salt sensitive. Although genotypes did not show the same degree of tolerance in germination, glasshouse and field experiments, the variety Yu‐07 showed consistently better performance in all trials, whilst Aus19720 was most sensitive in glasshouse and field experiments. These contrasting genotypes could be of a potential value for further studies to uncover the genetic mechanisms governing salt stress response in wheat.

Abstract:Bottlenecks, including limited genetic variation and the ongoing loss of genetic diversity, have hindered the development of modern wheat cultivars., making it crucial to use genetic diversity from wild relatives to improve wheat's adaptation to abiotic stress, such as salinity. This study assessed the phenotypic and epigenetic variation of introgressed wheat lines (BC4F2) derived from hybridizing two wheat cultivars with Aegilops cylindrica (AC). This study assessed the phenotypic and epigenetic variation of 156 introgressed wheat lines (BC4F2) derived from hybridization between wheat cultivars "Chinese Spring" (CS) and "Roshan" (R) and Aegilops cylindrica (AC). These lines and their recurrent parents (total of 158) were evaluated under normal and saline field conditions for the agronomic traits and stress tolerance indices. The data were used to select the most tolerant and most sensitive lines. Then, the selected BC4F2 lines and their parents (AC, CS, and R) were subjected to physiological, DNA cytosine methylation, and expression analysis of HKT1;5, NHX1, and SOS1 genes under control and salt stress conditions. Agro-physiological, epigenetic, and gene expression analyses showed the significant effects of salt stress and genetic background, as well as the differential response of the BC4F2 lines to salt stress. The variations in leaf and root K, Na, and K/Na ratios, and leaf Chla, Chlb, Car, and MDA levels, unlike DPPH radical scavenging levels, between salt-tolerant and salt-sensitive BC4F2 lines under saline conditions indicated a substantial distinction in salinity tolerance responses. RT-qPCR indicated higher expression levels of NHX1 and SOS1 genes in the leaf and root tissues of tolerant lines than those of sensitive lines. Global leaf and root DNA methylation analysis revealed the significant effects of salinity on the methylation modifications and confirmed the successful introgression of the salt-tolerance epigenome from Ae. cylindrica into wheat. Exploiting the genetic diversity of wild wheat relatives is a crucial goal for increasing genetic and epigenetic variation to enhance plant adaptation to salt stress.

Abstract:The successful screening and evaluation of salt-tolerant germplasm at the germination and seedling stages is of great importance for promoting the breeding of wheat varieties with salt tolerance. In this study, 70 wheat varieties bred in different regions were evaluated for salt tolerance through hydroponic exposure to different concentrations of salt. The relative water absorption, water absorption rate, dehiscence rate, germination rate, and germination index of seeds, and plant height, root length, stem diameter, and biomass of seedlings were determined at the germination and seedling stages of wheat, and the salt tolerance was identified and evaluated using multivariate statistical analysis. The germination ability and seedling growth potential of wheat germplasms decreased with the aggravation of salt stress. Based on the comprehensive salt tolerance index at the germination stage, our study identified 35 varieties to be salt-tolerant. There were nine varieties further screened for having strong salt tolerance according to the comprehensive salt tolerance index at the germination and seedling stages. SN41, Emam, YN301, and JM262 were superior in salt-tolerance, and YM39, LM30, JM60, YN999, and SD29 were salt-tolerant. Our study suggests that the biomass of seedlings can be used as a key parameter for assessing wheat germplasm's ability to withstand salt. Our results can provide some basic materials for cultivating new germplasm with salt tolerance and excavating the related genes of wheat.

Abstract:Salinity affects plant growth, development, yield, and is a big challenge for wheat growth across the globe. Possible feasible solution is creation of salt-tolerant material, genetic variation is a criterion to developing genetically superior individuals. To assess the genetic variation for salt tolerance, nationally and internationally-derived 81 wheat genotypes were selected and evaluated in 0- and 150-mM salt in nutritional culture at seedling stage. Results indicate that salinity levels reveal significant (p ≤ 0.01) differences for fresh root weight (RW), shoot length (SL), fresh shoot weight (SW), total plant length (TL), total fresh weight (TW), root/shoot weight ratio (RSWR), root/shoot length ratio (RSLR), and relative growth rate for weight (RGR-Wt). While, there was no difference for root length (RL). Hierarchical Clustering and Pairwise correlation analysis showed TW, RGR-Wt, SL, SW, and RW were positively correlated among themselves, whereas RL had poor correlations with all the traits except TL and RSLR. Hence, selection of SL can improve the performance of other parameters. Based on PCA analysis, SW and RGR-Wt were the major discriminative components for wheat genotypes. Present study explained that shoot related parameters could be used as a selection criterion to categorize salt-tolerant genotypes. Outperforming genotypes 1104 and 1106 in saline conditions could be used as parents in creation of salt-tolerant wheat genotypes, and parameters such as SL, SW, TW, and RGR-Wt for early screening will be important for creating salt-tolerant and high yielding wheat genotypes.
Physiological responses, ion accumulation and yield performance of wheat ( Triticum aestivum L.) to salt stress

Abstract:Wheat is affected by various biotic and abiotic stresses, especially salinity, which reduces the growth and yield drastically. High salinity is a major constraint for wheat productivity in many countries, including Bangladesh. With this view, here, an experiment was conducted to observe genotypic differences in physiological, ion accumulation, agro-morphological and yield performance of wheat against different levels of salinity. Experimental variables consisted of five salt tolerant genotypes (G 20–2–2, G 20–1–2, G 13–2, G 22–2, G 9–2), one susceptible genotype (G 24–2) and one standard check variety (BARI ghom 25), which assigned to four levels of salinity with electrical conductivities control (0.3), 4, 8 and 12 dS m −1 . Irrespective of genotypes, salinity stress significantly decreased the yield and yield attributes. Results of analyses based on salt tolerance indices of plant growth related and yield contributing parameters, ionic balance (Na + , K + and Na + / K + ratio), and stress indicators such as chlorophyll content, photosynthetic rate (Pn), stomatal conductance (gs) and transpiration rate (Tr) revealed genotypes G 20–2–2, G 13–2 and G 20–1–2 as salt tolerant, genotype G 9–2 as moderately salt-tolerant and G 24–2 and G 22–2 as salt-sensitive genotypes. Additionally, lower accumulations of hydrogen peroxide and malondialdehyde, and higher activities of antioxidant enzymes in the salt-tolerant genotypes G 20–2–2 and G 13–2 than in the salt-sensitive genotype G 24–2 indicated reduced oxidative damage in genotypes G 20–2–2 and G 13–2 relative to that in genotype G 24–2. Collectively, our findings suggest that the optimum growth and yield of salt tolerant genotypes are associated with decreased Na + / K + ratio, increased antioxidant enzymes activity and reduced oxidative stress.

Genetic networks underlying salinity tolerance in wheat uncovered with genome-wide analyses and selective sweeps

Abstract:A genetic framework underpinning salinity tolerance at reproductive stage was revealed by genome-wide SNP markers and major adaptability genes in synthetic-derived wheats, and trait-associated loci were used to predict phenotypes. Using wild relatives of crops to identify genes related to improved productivity and resilience to climate extremes is a prioritized area of crop genetic improvement. High salinity is a widespread crop production constraint, and development of salt-tolerant cultivars is a sustainable solution. We evaluated a panel of 294 wheat accessions comprising synthetic-derived wheat lines (SYN-DERs) and modern bread wheat advanced lines under control and high salinity conditions at two locations. The GWAS analysis revealed a quantitative genetic framework of more than 200 loci with minor effect underlying salinity tolerance at reproductive stage. The significant trait-associated SNPs were used to predict phenotypes using a GBLUP model, and the prediction accuracy (r2) ranged between 0.57 and 0.74. The r2 values for flag leaf weight, days to flowering, biomass, and number of spikes per plant were all above 0.70, validating the phenotypic effects of the loci discovered in this study. Furthermore, the germplasm sets were compared to identify selection sweeps associated with salt tolerance loci in SYN-DERs. Six loci associated with salinity tolerance were found to be differentially selected in the SYN-DERs (12.4 Mb on chromosome (chr)1B, 7.1 Mb on chr2A, 11.2 Mb on chr2D, 200 Mb on chr3D, 600 Mb on chr6B, and 700.9 Mb on chr7B). A total of 228 reported markers and genes, including 17 well-characterized genes, were uncovered using GWAS and EigenGWAS. A linkage disequilibrium (LD) block on chr5A, including the Vrn-A1 gene at 575 Mb and its homeologs on chr5D, were strongly associated with multiple yield-related traits and flowering time under salinity stress conditions. The diversity panel was screened with more than 68 kompetitive allele-specific PCR (KASP) markers of functional genes in wheat, and the pleiotropic effects of superior alleles of Rht-1, TaGASR-A1, and TaCwi-A1 were revealed under salinity stress. To effectively utilize the extensive genetic information obtained from the GWAS analysis, a genetic interaction network was constructed to reveal correlations among the investigated traits. The genetic network data combined with GWAS, selective sweeps, and the functional gene survey provided a quantitative genetic framework for identifying differentially retained loci associated with salinity tolerance in wheat.
Mapping QTLs conferring salt tolerance and micronutrient concentrations at seedling stage in wheat

Abstract:Abstract Soil salinization and degradation is one of the consequences of climate change. Identification of major salt tolerance genes and marker assisted selection (MAS) can accelerate wheat breeding for this trait. We genotyped 154 wheat F 2 lines derived from a cross between salt tolerant and susceptible cultivars using the Axiom Wheat Breeder’s Genotyping Array. A high-density linkage map of 988 single nucleotide polymorphisms (SNPs) was constructed and utilized for quantitative trait loci (QTL) mapping for salt tolerance traits and mineral concentrations under salinity. Of 49 mapped QTLs, six were for Na + exclusion (NAX) and two QTLs (qSNAX.2 A.1, qSNAX.2 A.2) on chromosome 2 A coincided with a reported major NAX QTL ( Nax1 or HKT1;4 ). Two other major NAX QTLs were mapped on 7 A, which contributed 11.23 and 18.79% of the salt tolerance respectively. In addition to Ca +2 and Mg +2 QTLs, twenty-seven QTLs for tissue Phosphorus, Zinc, Iron, Manganese, Copper, Sulphur and Boron concentrations under salinity were also mapped. The 1293 segregating SNPs were annotated/located within genes for various ion channels, signalling pathways, transcription factors (TFs), metabolic pathways and 258 of them showed differential expression in silico under salinity. These findings will create new opportunities for salt tolerance breeding programs.

Abstract:Salt stress results in the severe decline of yield and quality in wheat. In the present study, salt-tolerant Tritipyrum (“Y1805”) and salt-sensitive wheat “Chinese Spring” (“CS”) were selected from 121 wheat germplasms to test their physiological, antioxidant enzyme, and transcriptomic responses and mechanisms against salt stress and recovery. 56 chromosomes were identified in “Y1805” that comprised A, B, and D chromosomes from wheat parent and E chromosomes from Thinopyrum elongatum , adding to salt-tolerant trait. Salt stress had a greater inhibitory effect on roots than on shoots, and “Y1805” demonstrated stronger salt tolerance than “CS.” Compared with “CS,” the activities of superoxide dismutase and catalase in “Y1805” significantly increased under salt stress. “Y1805” could synthesize more proline and soluble sugars than “CS.” Both the net photosynthetic rate and chlorophyll a/b were affected by salt stress, though the level of damage in “Y1805” was significantly less than in “CS.” Transcriptome analysis showed that the differences in the transcriptional regulatory networks of “Y1805” were not only in response to salt stress but also in recovery. The functions of many salt-responsive differentially expressed genes were correlated closely with the pathways “peroxisome,” “arginine and proline metabolism,” “starch and sucrose metabolism,” “chlorophyll and porphyrin metabolism,” and “photosynthesis.”

Abstract:Increased soil salinization, tightly related to global warming and drought and exacerbated by intensified irrigation supply, implies highly detrimental effects on staple food crops such as wheat. The situation is particularly alarming for durum wheat (DW), better adapted to arid/semi-arid environments yet more sensitive to salt stress than bread wheat (BW). To enhance DW salinity tolerance, we resorted to chromosomally engineered materials with introgressions from allied halophytic Thinopyrum species. "Primary" recombinant lines (RLs), having portions of their 7AL arms distally replaced by 7el 1 L Th. ponticum segments, and "secondary" RLs, harboring Th. elongatum 7EL insertions "nested" into 7el 1 L segments, in addition to near-isogenic lines lacking any alien segment (CLs), cv. Om Rabia (OR) as salt tolerant control, and BW introgression lines with either most of 7el 1 or the complete 7E chromosome substitution as additional CLs, were subjected to moderate (100 mM) and intense (200 mM) salt (NaCl) stress at early growth stages. The applied stress altered cell cycle progression, determining a general increase of cells in G1 and a reduction in S phase. Assessment of morpho-physiological and biochemical traits overall showed that the presence of Thinopyrum spp. segments was associated with considerably increased salinity tolerance versus its absence. For relative water content, Na + accumulation and K + retention in roots and leaves, oxidative stress indicators (malondialdehyde and hydrogen peroxide) and antioxidant enzyme activities, the observed differences between stressed and unstressed RLs versus CLs was of similar magnitude in "primary" and "secondary" types, suggesting that tolerance factors might reside in defined 7el 1 L shared portion(s). Nonetheless, the incremental contribution of 7EL segments emerged in various instances, greatly mitigating the effects of salt stress on root and leaf growth and on the quantity of photosynthetic pigments, boosting accumulation of compatible solutes and minimizing the decrease of a powerful antioxidant like ascorbate. The seemingly synergistic effect of 7el 1 L + 7EL segments/genes made "secondary" RLs able to often exceed cv. OR and equal or better perform than BW lines. Thus, transfer of a suite of genes from halophytic germplasm by use of fine chromosome engineering strategies may well be the way forward to enhance salinity tolerance of glycophytes, even the sensitive DW.

Abstract:Abiotic stresses like salinity and abnormally cold environments cause significant yield losses in many crops including wheat. Therefore, concerted efforts are being made by breeders to develop new varieties with salt and cold tolerance to ensure stable yields over varied environments. This study was undertaken to screen six hundred and seventy-seven accessions of international wheat genetic resources to identify lines with high level of tolerance to salinity and cold environments. Based on the results of two years study in different agroecological locations, 51 accessions were classified as salt tolerant and 115 accessions were classified as cold tolerant. Of these, 35 accessions had good agronomic characteristics. Also, there were 39 genotypes with combined tolerance to cold as well as salinity. Thus, there were good lines which can be used directly or as parents for breeding wheat varieties for wide adaptation and high yield. Further analysis of the data showed that early genotypes had good cold tolerance but a poor salt tolerance. It was also observed that small number of both test spikelet and spikelet, short spike length and good seed-plump were positively associated with cold tolerance. Therefore, maturity and spike traits should be taken into considered when selecting wheat lines for wide adaptation breeding.

Abstract:Background: Salinity stress is a major adverse environmental factor that can limit crop yield and restrict normal land use. The selection of salt-tolerant strains and elucidation of the underlying mechanisms by plant breeding scientists are urgently needed to increase agricultural production in arid and semi-arid regions. Results: In this study, we selected the salt-tolerant wheat ( Triticum aestivum ) strain ST9644 as a model to study differences in expression patterns between salt-tolerant and salt-sensitive strains. High-throughput RNA sequencing resulted in more than 359.10 Gb of clean data from 54 samples, with an average of 6.65 Gb per sample. Compared to the IWGSC reference annotation, we identified 50,096 new genes, 32,923 of which have functional annotations. Comparisons of abundances between salt-tolerant and salt-sensitive strains revealed 3,755, 5,504, and 4,344 genes that were differentially expressed at 0, 6, and 24 h, respectively, in root tissue under salt stress. KEGG pathway analysis of these genes showed that they were enriched for phenylpropanoid biosynthesis (ko00940), cysteine and methionine metabolism (ko00270), and glutathione metabolism (ko00480). We also applied weighted gene co-expression network analysis (WGCNA) analysis to determine the time course of root tissue response to salt stress and found that the acute response lasts >6 h and ends before 12 h. We also identified key alternative splicing factors showing different splicing patterns in salt-sensitive and salt-tolerant strains; however, only few of them were differentially expressed in the two groups. Conclusion: Our results offer a better understanding of wheat salt tolerance and improve wheat breeding.

Abstract:BackgroundSoil salinity has been affecting wheat production worldwide over past few decades. Evaluation of wheat genotypes for salinity tolerance at germination and vegetative growth level is crucial. Marker assisted selection is a technique used extensively for choosing salt-tolerant genotypes from breeding populations to introduce novel genes.Methods and materialsThe current study's main goal was to discover salt-stress resistant genes; genetic divergence and genome-wide connection by using recently designed candidate gene-based simple-sequence-repeat markers (cg-SSRs). The phenotypic connection of morphological features during the germination growth stage i.e., germination period, root length/weight and shoot length/weight, and vegetative growth stages i.e., root length/weight and shoot length/weight were tested in a group of 50 wheat genotypes. Significant difference was observed in germination rate, root length and weight among control and saline treatments.ConclusionTotal 30 SSR markers were utilized to test salinity resistance genes in wheat genotypes. Three (10%) of which were monomorphic, one (3.34%) showed no result, and the other 26 (86%) were polymorphic. Using 30 polymorphic markers discovered total 37 alleles. The polymorphic information content (PIC), quantifies each SSR locus capacity to discriminate between wheat, varied from 0.00 to 0.38 with an average of 0.19. Association analysis revealed that 26 primers were associated with morphological features, 03 with root length and the remaining 23 with germination. Utilizing morphological data, stress tolerance index (STI) was designed concluding that Auqab-2000, Margala-99 and Ufaq showed better resistance against salinity among other wheat genotypes. Cluster analysis demonstrated that wheat genotypes have vast genetic variability.

Abstract:Drought response in wheat is considered a highly complex process, since it is a multigenic trait; nevertheless, breeding programs are continuously searching for new wheat varieties with characteristics for drought tolerance. In a previous study, we demonstrated the effectiveness of a mutant known as RYNO3936 that could survive 14 days without water. In this study, we reveal another mutant known as BIG8-1 that can endure severe water deficit stress (21 days without water) with superior drought response characteristics. Phenotypically, the mutant plants had broader leaves, including a densely packed fibrous root architecture that was not visible in the WT parent plants. During mild (day 7) drought stress, the mutant could maintain its relative water content, chlorophyll content, maximum quantum yield of PSII (Fv/Fm) and stomatal conductance, with no phenotypic symptoms such as wilting or senescence despite a decrease in soil moisture content. It was only during moderate (day 14) and severe (day 21) water deficit stress that a decline in those variables was evident. Furthermore, the mutant plants also displayed a unique preservation of metabolic activity, which was confirmed by assessing the accumulation of free amino acids and increase of antioxidative enzymes (peroxidases and glutathione S-transferase). Proteome reshuffling was also observed, allowing slow degradation of essential proteins such as RuBisCO during water deficit stress. The LC-MS/MS data revealed a high abundance of proteins involved in energy and photosynthesis under well-watered conditions, particularly Serpin-Z2A and Z2B, SGT1 and Calnexin-like protein. However, after 21 days of water stress, the mutants expressed ABC transporter permeases and xylanase inhibitor protein, which are involved in the transport of amino acids and protecting cells, respectively. This study characterizes a new mutant BIG8-1 with drought-tolerant characteristics suited for breeding programs.

Abstract:Salinity is a major abiotic stress that threatens crop yield and food supply in saline soil areas. Crops have evolved various strategies to facilitate survival and production of harvestable yield under salinity stress. Wheat (Triticum aestivum L.) is the main crop in arid and semiarid land areas, which are often affected by soil salinity. In this review, we summarize the conventional approaches to enhance wheat salt tolerance, including cross-breeding, exogenous application of chemical compounds, beneficial soil microorganisms, and transgenic engineering. We also propose several new breeding techniques for increasing salt tolerance in wheat, such as identifying new quantitative trait loci or genes related to salt tolerance, gene stacking and multiple genome editing, and wheat wild relatives and orphan crops domestication. The challenges and possible countermeasures in enhancing wheat salinity tolerance are also discussed.
