Abstract:models have now become a realistic alternative to animal models for cardiotoxicity assessment. However, the cost and expertise required to implement electrophysiology systems to study cardiac cells poses a strong obstacle to their widespread use. This study presents a novel, cost-effective approach for cardiac electrophysiology using fully-printed graphene-based microelectrode arrays (pGMEAs) coupled with an open-source signal acquisition system. We characterized the pGMEAs’ electrical properties and biocompatibility, observing low impedance values and cell viability. We demonstrated the platform’s capability to record spontaneous electrophysiological activity from HL-1 cell cultures, and we monitored and quantified their responses to chemical stimulation with noradrenaline. This study demonstrates the feasibility of producing fully-printed, graphene-based devices for electrophysiology. The accessible and versatile platform we present here represents a step further in the development of alternative methods for cardiac safety screening.

Abstract:This study introduces a new methodology for simultaneous multi‐material fabrication using a commercial 3D printer, enabling miniaturized bio‐shaped PEDOT:PSS structures on flexible substrates. Conducting bio‐shapes are combined with polymerizable deep eutectic solvents which enhance adhesion, ionic conductivity, and long‐term stability. The multi‐material electrode presents good skin adhesion, low skin impedance, and high signal‐to‐noise ratio, with excellent performance on forearm electromyography. In this work, a new method of multi‐material printing in one‐go using a commercially available 3D printer is presented. The approach is simple and versatile, allowing the manufacturing of multi‐material layered or multi‐material printing in the same layer. To the best of the knowledge, it is the first time that 3D printed Poly(3,4‐ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS) micro‐patterns combining different materials are reported, overcoming mechanical stability issues. Moreover, the conducting ink is engineered to obtain stable in‐time materials while retaining sub‐100 μm resolution. Micro‐structured bio‐shaped protuberances are designed and 3D printed as electrodes for electrophysiology. Moreover, these microstructures are combined with polymerizable deep eutectic solvents (polyDES) as functional additives, gaining adhesion and ionic conductivity. As a result of the novel electrodes, low skin impedance values showed suitable performance for electromyography recording on the forearm. Finally, this concluded that the use of polyDES conferred stability over time, allowing the usability of the electrode 90 days after fabrication without losing its performance. All in all, this demonstrated a very easy‐to‐make procedure that allows printing PEDOT:PSS on soft, hard, and/or flexible functional substrates, opening up a new paradigm in the manufacturing of conducting multi‐functional materials for the field of bioelectronics and wearables.

Electropolymerization processing of side-chain engineered EDOT for high performance microelectrode arrays

Abstract:Microelectrode Arrays (MEAs) are popular tools for in vitro extracellular recording. They are often optimized by surface engineering to improve affinity with neurons and guarantee higher recording quality and stability. Recently, PEDOT:PSS has been used to coat microelectrodes due to its good biocompatibility and low impedance, which enhances neural coupling. Herein, we investigate on electro-co-polymerization of EDOT with its triglymated derivative to control valence between monomer units and hydrophilic functions on a conducting polymer. Molecular packing, cation complexation, dopant stoichiometry are governed by the glycolation degree of the electro-active coating of the microelectrodes. Optimal monomer ratio allows fine-tuning the material hydrophilicity and biocompatibility without compromising the electrochemical impedance of microelectrodes nor their stability while interfaced with a neural cell culture. After incubation, sensing readout on the modified electrodes shows higher performances with respect to unmodified electropolymerized PEDOT, with higher signal-to-noise ratio (SNR) and higher spike counts on the same neural culture. Reported SNR values are superior to that of state-of-the-art PEDOT microelectrodes and close to that of state-of-the-art 3D microelectrodes, with a reduced fabrication complexity. Thanks to this versatile technique and its impact on the surface chemistry of the microelectrode, we show that electro-co-polymerization trades with many-compound properties to easily gather them into single macromolecular structures. Applied on sensor arrays, it holds great potential for the customization of neurosensors to adapt to environmental boundaries and to optimize extracted sensing features.

Abstract:Electrophysiological biosensors embedded in planar devices represent a state of the art approach to measure and evaluate the electrical activity of biological systems. This measurement method allows for the testing of drugs and their influences on cells or tissues, cytotoxicity, as well as the direct implementation into biological systems in vivo for signal transduction. Multi-electrode arrays (MEAs) with metal or metal-like electrodes on glass substrates are one of the most common, well-established platforms for this purpose. In recent years organic electrochemical transistors (OECTs) made of poly(2,3-dihydrothieno-1,4-dioxin)-poly(styrenesulfonate) (PEDOT:PSS) have as well shown their value in transducing and amplifying the ionic signals in biological systems. We developed OECT devices in a wafer-scale process and used them as electrophysiological biosensors measuring electrophysiological activity of the cardiac cell line HL-1. Our optimized devices show very promising properties such as good signal-to-noise ratio as well as the ability to record fast components of extracellular signals. Combined with an easy, cost effective fabrication and the transparency of the polymer, this platform offers a valuable alternative to traditional MEA systems for future cell sensing applications.

Abstract:The proliferation of medical wearables necessitates the development of novel electrodes for cutaneous electrophysiology. In this work, poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS) is combined with a deep eutectic solvent (DES) and polyethylene glycol diacrylate (PEGDA) to develop printable and biocompatible electrodes for long-term cutaneous electrophysiology recordings. The impact of printing parameters on the conducting properties, morphological characteristics, mechanical stability and biocompatibility of the material were investigated. The optimised eutectogel formulations were fabricated in four different patterns -flat, pyramidal, striped and wavy- to explore the influence of electrode geometry on skin conformability and mechanical contact. These electrodes were employed for impedance and forearm EMG measurements. Furthermore, arrays of twenty electrodes were embedded into a textile and used to generate body surface potential maps (BSPMs) of the forearm, where different finger movements were recorded and analysed. Finally, BSPMs for three different letters (B, I, O) in sign-language were recorded and used to train a logistic regressor classifier able to reliably identify each letter. This novel cutaneous electrode fabrication approach offers new opportunities for long-term electrophysiological recordings, online sign-language translation and brain-machine interfaces.

Abstract:Planar microelectrode arrays (MEAs) are limited in tracking signals in complex 3D cell cultures. To overcome this, a novel approach integrates rapid prototyping, additive manufacturing, and wet etching. Laser‐patterned MEAs now feature high‐aspect ratio ink‐jet printed 3D electrode structures, enabling precise recordings within organoids. The 3D MEAs allow capturing electrophysiological activity in cortical organoids for in vitro neural research. Monitoring electrical activity across multiple planes in 3D cell cultures and organoids is imperative to comprehensively understand their functional connections and behavior. However, traditional planar microelectrode arrays (MEAs) are intended for surface recordings and are inadequate in addressing this aspect. The limitations, such as longer production times and limited adaptability imposed by standard clean‐room techniques, constrain the design possibilities for 3D electrode arrays and potentially hinder effective cell‐electrode coupling. To tackle this challenge, a novel approach is presented that leverages rapid prototyping processes and additive manufacturing in combination with wet etching and electrodeposition to enhance electrode fabrication and performance. The laser‐patterned MEAs on glass, polyimide (PI) foil, or polyethylene terephthalate (PET) foil substrates incorporate high‐aspect ratio (up to 44:1) ink‐jet printed 3D electrode structures with heights up to 1 mm at a pitch of 200 μm, enabling precise recording within cell tissues. The specific shapes of the electrode tips and customizable 3D structures provide great flexibility in electrode placement. The versatility of the 3D MEAs is demonstrated by recording the electrophysiological activity of cortical organoids in situ, paving the way for investigating neural activity under regular or various pathologically altered conditions in vitro in a high throughput manner.

Abstract:Microelectrode arrays (MEAs) have become key components in retinal implants for recording of neural signals or electrical stimulation of nerves. In this paper, implantable flexible MEAs based on a novel polymer, parylene, were developed by designing MEMS process. And electrical performance of prepared microelectrodes was characterized electrochemically. Results showed that impedance and phase delay of testing MEAs were 2000 Omega similar to 100 Omega and -20 degrees from 10(3)Hz to 10(5) Hz, respectively, which would supply a better application with neural interface for retinal prosthesis.

Abstract:PEDOT:PSS hydrogel-based bioelectronic interfaces have gained significant attention in various fields including biomedical devices, wearable devices, and epidermal electronics. However, the development of high-performance bioelectronic interfaces that integrate excellent conductivity, strong adhesion, and advanced processing compatibility remains a challenge. Herein, we develop a high-performance bioelectronic interface by 3D printing of a novel poly(vinyl alcohol-formaldehyde) (PVAF)-PEDOT:PSS composite ink. Such a PEDOT:PSS-PVAF ink exhibits favorable rheological properties for direct-ink-writing 3D printing, enabling the fabrication of high-resolution patterns and three-dimensional structures with high aspect ratios. Hydrogel bioelectronic interface printed by such PEDOT:PSS-PVAF ink simultaneously achieves high conductivity (over 100 S m −1 ), strong adhesion (31.44 ± 7.07 kPa), as well as stable electrochemical performance (charge injection capacity of 13.72 mC cm −2 and charge storage capacity of 18.80 mC cm −2 ). We further integrate PEDOT:PSS-PVAF hydrogel bioelectronic interface to fabricate adhesive skin electrodes for electromyography (EMG) signal recording. The resultant EMG skin electrodes demonstrate superior performance and stability compared to commercial products, maintaining high signal-to-noise ratio of > 10 dB under varying weights and repetitive motions. These advantageous performance of PEDOT:PSS-PVAF based hydrogel bioelectronic interfaces may be helpful for diverse bioelectronic applications like healthcare monitoring and epidermal bioelectronics.

Abstract:The drift or fall of stretchable neural microelectrodes from the surface of wet and dynamic tissues severely hampers the adoption of microelectrodes for electrophysiological signal monitoring. Endowing the stretchable electrodes with adhesive ability is an effective way to overcome these problems. Current adhesives form tough adhesion to tissues by covalent interaction, which decreases the biocompatibility of the adhesives. Here, we fabricate a strong electrostatic adhesive (noncovalent interaction), highly conformal, stretchable microelectrode arrays (MEAs) for the electrophysiological interface. This MEA was composed of polypyrrole (PPy) as the electrode material and hydrogel as the stretchable substrate [the cross-linked and copolymerized hydrogel of 2-acrylamido-2-methylpropane sulfonic acid (AMPS), gelatin, chitosan, 2-methoxyethyl acrylate, and acrylic acid is named PAGMA]. Strong and stable electrostatic adhesion (85 kPa) and high stretchability (100%) allow for the integration of PPy MEAs based on the PAGMA hydrogel substrate (PPy-PAGMA MEAs) on diverse wet dynamic tissues. Additionally, by adjusting the concentration of AMPS in PAGMA, the hydrogel (PAGMA-1) can produce tough adhesion to many inorganic and elastomer materials. Finally, the PPy-PAGMA MEAs were toughly and conformally adhered on the rat's subcutaneous muscle and beating heart, and the rat's electrophysiological signals were successfully recorded. The development of these adhesive MEAs offers a promising strategy to establish stable and compliant electrode-tissue interfaces.

Abstract:Bioelectronics has been developed for recording the electrophysiological activity of diagnostic and therapeutic devices. However, current bioelectrodes still imperfectly comply with tissues, which results in high interfacial impedance and even mechanical detachment. Herein, we report a simple yet effective approach to overcome such hurdles by designing a highly conductive, adhesive hydrogel composite based on freeze-dried poly(3,4-ethylenedioxythiophene):poly (styrene sulfonate) (PEDOT:PSS), polyurethane (PU), and poly(acrylic acid) (PAA). With the continuous phase-separation of PEDOT:PSS, PU, and PAA, the resultant composite hydrogels can simultaneously achieve high adhesion (lap-shear strength > 8 kPa), stretchability (fracture strain > 1100%), and electrical conductivity (conductivity > 2 S/m) by overcoming the traditional trade-off between mechanical and electrical properties in conducting polymer hydrogels. Moreover, such hydrogels are readily applicable to advanced manufacturing techniques such as 3D printing. We further fabricated skin electrodes and achieved high quality and high signal-to-noise ratio EMG signal recording of the forearm.
Fabrication and Characterization of a Parylene-Based Three-Dimensional Microelectrode Array for Use in Retinal Prosthesis

Abstract:A novel Parylene-based shape-transferring technique was developed to fabricate 3-D microelectrodes, which fulfilled the requirements of the high-density microelectrode array (MEA) used in retinal prosthesis. A process combining anisotropic and isotropic etching was utilized to form the 3-D silicon-tip array whose shape was transferred to the Parylene C film as a flexible and biocompatible substrate. Platinum was chosen as the electrode material owing to its high charge delivery capacity and chemical inertness in the physiological environment. An aluminum-photoresist dual-layer lift-off process was employed for platinum patterning on the substrate with high tips. To avoid the cracking problem existing in the platinum-Parylene C structure, a gold intermediate layer was introduced to retard the tendency. To test the superiority of the present 3-D flexible MEA compared with the planar one, electrical properties of MEAs were characterized both in vitro and in vivo. The experimental results showed that the impedance of the 3-D electrode was about 72.7% of the planar one at 1 kHz with the same footprint in vitro, and 71.5% in vivo. The preliminary surgical experiment validated the feasibility of the 3-D MEA in retinal prosthesis.
PDMS based multielectrode arrays for superior in-vitro retinal stimulation and recording

Abstract:Understanding of the neural response to electrical stimulation requires simultaneous recording from the various neurons of retina. Electrodes form the physical interface with the neural or retinal tissue. Successful retinal stimulation and recording demands conformal integration of these electrodes with the soft tissue to ensure establishment of proper electrical connection with the excitable tissue. Mechanical impedance of polydimethylsiloxane (PDMS) being compliant with that of retinal tissue, offers excellent potential as a substrate for metal electrodes. In this paper, Cr/Au micro electrodes with 200 μm diameter were fabricated on rigid and flexible PDMS substrates under crack free condition. Spontaneous buckling of thin films over PDMS substrates improved electrode performance circumventing the fabrication issues faced over a buckled surface. Individual electrodes from the multielectrode arrays (MEAs) were examined with electrochemical impedance spectroscopy and cyclic voltammetry. Controlled fabrication process as described here generates buckles in the metal films leading to increased electrode surface area that increases the charge storage capacity and decreases the interface impedance of the metal electrodes. At 1 kHz, impedance was reduced from 490 ± 27 kΩ to 246 ± 19 kΩ and charge storage capacity was increased from 0.40 ± 0.87 mC/cm2 to 2.1 ± 0.87 mC/cm2. Neural spikes recorded with PDMS based electrodes from isolated retina also contained less noise as indicated by signal to noise ratio analysis. The present study established that the use of PDMS as a substrate for MEAs can enhance the performance of any thin film metal electrodes without incorporation of any coating layers or nanomaterials.

Abstract:Poly(3,4‐ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) is a promising material because of its favorable electrical and mechanical properties, stability in ambient environments, and biocompatibility. It finds broad application in energy storage, flexible electronics, and bioelectronics. Additive manufacturing opens a plethora of new avenues to form and shape PEDOT:PSS, allowing for the rapid construction of customized geometries. However, there are difficulties in printing PEDOT:PSS while maintaining its attractive properties. A 3D printing method for PEDOT:PSS using a room‐temperature coagulation bath‐based direct ink writing technique is reported. This technique enables fabrication of PEDOT:PSS into parts that are of high resolution and high conductivity, while maintaining stable electrochemical properties. The coagulation bath can be further modified to improve the mechanical properties of the resultant printed part via a one‐step reaction. Furthermore, it is demonstrated that a simple post‐processing step allows the printed electrodes to strongly adhere to several substrates under aqueous conditions, broadening their use in bioelectronics. Employing 3D printing of PEDOT:PSS, a cortex‐wide neural interface is fabricated, and intracranial electrical stimulation and simultaneous optical monitoring of mice brain activity with wide field calcium imaging are demonstrated. This reported 3D‐printing technique eliminates the need for cumbersome experimental setups while offering desired material properties. Poly(3,4‐ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) has received increasing attention in the research community due to favorable properties for developing bioelectronic devices. This work reports a simple 3D printing method (direct ink writing) to directly pattern PEDOT:PSS into desired structures. The printed parts have excellent mechanical and electrochemical properties, which allows for the fabrication of a precise cortex‐wide neural interface for brain stimulation.

Abstract:Flexible electrode array, a new‐generation neural microelectrode, is a crucial tool for information exchange between living tissues and external electronics. Till date, advances in flexible neural microelectrodes have been limited because of their high impedance and poor mechanical consistency at tissue interfaces. Here we introduce a highly sensitive and omnidirectionally stretchable polymeric electrode array (PEA). Micropyramid–nanowire composite structures are constructed to increase the effective surface area of PEA, achieving an exponential reduction in impedance compared with gold (Au) and flat polypyrrole (PPy) electrodes. Moreover, for the first time, we design a suspended umbrella structure to enable PEA with omnidirectional stretchability of up to ∼20%. The PEA can withstand 1000 cycles of mechanical loads without decrease in performance. As a proof of concept, PEA are conformally attached to a rat heart and tibialis anterior muscle, and electrophysiological signals (electrocardiogram and electromyogram) of the rat are successfully recorded. High-density Stretchable Microelectrode Array Based on Multilayer Serpentine Interconnections

Abstract:Microelectrode array (MEA) is a powerful tool for recording electrophysiological signals and studying the electrophysiological features of nerve, retina, and cardiomyocytes. Stretchable electrodes can form conformal contact with dynamic biological tissues/organs (e.g. heart) to enhance the coupling efficiency in electrophysiological measurements. However, MEAs based on conventional materials require serpentine interconnections to enable stretchability. A large number of serpentine interconnects take up spaces and hinder the improvement of spatial resolution. Here, we introduce a multilayer design strategy that yields a stretchable MEA with spatial density of 7.3 sites mm−2 and biaxial stretchability of 30%. The serpentine interconnects of each layer overlap vertically to reduce the area of the wires used for connection. In vitro validation in phosphate buffered saline and ex vivo test on perfused mouse heart show a 100% yield rate of the multilayer stretchable MEA, with capabilities in spatiotemporal mapping of electrophysiological signals at high spatial resolution.

Abstract:In the past ten years, wearable electronics underwent tremendous growth. Undoubtedly, one of the fields that led this trend is represented by biomedical applications. In this field, wearable technologies can provide unique features such as the unobtrusive monitoring of biopotentials. Polymerbased electrodes developed for this purpose can take advantage of their seamless integration in the garments. However, the available solutions exhibit fragility in relation with the stretchability of the fabric, causing significant performance degradation.In this work, this problem is tackled by a novel deposition approach based on screen-printing technology. The electrodes are deposited onto the pre-stretched fabric to ensure the full functionality during common operating conditions. To this aim, a novel PEDOT:PSS conductive ink formulation and printing procedure were conceived. In order to prove the electrode performance for surface electromyography, we printed the electrodes directly onto a commercial stretchable polyester sleeve for sport applications. The electrodes allowed to reliably record the muscular activity of the forearm with performance comparable to that of commercial gelled Ag/AgCl electrodes. The obtained results suggest that the proposed approach can be valuably used in health and fitness applications.

Abstract:The acquisition of biopotentials from the surface of the human body is an important and common procedure that is extremely useful both in the clinical practice and in non‐clinical applications such as sport monitoring and human‐machine interfaces. To avoid the employment of bulky recording systems and to allow optimal long‐term measurements, recently tattoo‐like electrodes were developed, aiming at obtaining high‐quality signals while at the same time maximizing the comfort of the subject. The ability to seamlessly contact epidermal electrodes is of paramount importance for ensuring both these objectives and represents one of the biggest challenges in this field. In this paper, we propose a simple yet efficient way to fabricate free‐standing conductive/ferrimagnetic PEDOT:PSS‐based epidermal electrodes by employing a convenient spray coating technique. This approach takes full advantage of the "bioelectronics bag of tricks" and offers a new methodology to tackle the problem of epidermal electrodes interfacing. The proposed electrodes are in fact fabricated using a conductive polymeric ink deposited through a large‐area technique and can be easily transferred onto the skin, where they can be conveniently contacted by means of magnetic connectors without disrupting their conformability. These electrodes have been employed with excellent results for the detection of different biopotentials, namely electrocardiogram, electromyogram and electro‐oculogram, and demonstrated very good performance for the detection of biosignals from body parts, such as the face, where standard pre‐gelled electrodes are not ideal, thus demonstrating the effectiveness of the approach for the development of a new generation of functional films suitable for critical biopotentials monitoring.
