Abstract:Microelectrode arrays (MEAs) for stimulation and signal recording of in vitro cultured neurons are presented. Each MEA is composed of 60 independent electrodes with 59 working ones and one reference one. These electrodes are divided into 30 pairs. Through each pair of electrodes, four independent states can be realized to define the accessing modes of neurons cultured on the surface of the electrodes. A total MEA covers an area of 10 mm×10 mm. MEAs are fabricated in a silicon-based semiconductor process. An implemented MEA is bonded on a specially designed printed-circuit-board (PCB) and surrounded by a culture chamber. An impedance measurement has been made to verify the electrical characteristics of MEAs. The surface was modified to enhance the biocompatibility. A series of PC12 cells culture experiments validates the effectiveness of the modification. An extracellular signal recording experiment with acetylcholine (Ach) as a stimulant has been carried out, and the results show the feasibility of MEAs for extracellular action potential recording. Extracellular electrical stimulation and recording experiments have been carried out too. They indicate that MEAs can be used for extracellular stimulation, recording, simultaneous stimulation and recording, and isolation of PC12 cells network cultured in vitro.

Abstract:Bioelectronic interfaces have the potential to revolutionize the treatment of medical disorders and augment physiology. Implantable devices such as pacemakers and deep brain stimulators have already been deployed to control activity in diseases including Parkinson’s disease and epilepsy. These devices typically operate by delivering electrical stimulation at pre-programmed intervals (known as open-loop stimulation). Recent advances in machine learning and low-power integrated circuits have led to the emergence of personalized medical devices that monitor the user’s state and stimulate in response to measured biological activity (known as closed-loop stimulation). There are two key questions that require fundamental research to achieve breakthroughs in personalized devices: 1) What biomarkers and algorithms are best suited to detecting biological states (e.g. seizures in epilepsy)? and 2) What types of electrical stimuli are optimal for controlling these states? The answer to these questions can be explored in vitro using multielectrode array (MEA) systems that interface with biological tissue with reduced experimental complexity, better reproducibility, and fewer confounding variables present in whole organisms. However, existing MEA systems have functional limitations and closed-source designs that prevent researchers from developing improvements. This paper introduces OpenMEA, an open-source platform for closed-loop bioelectronics research. OpenMEA includes designs for the components necessary to build a benchtop in vitro laboratory, including electrophysiological recording and stimulation electronics, a microfluidic perfusion system, and physical designs for multielectrode arrays. The system is demonstrated with the electrical recording and stimulation of epileptogenic human and rodent brain slices. The aim of OpenMEA is to democratize bioelectronic research tools to accelerate the deployment of devices for the treatment of disorders and beyond.

Abstract:Microelectrode arrays (MEAs) play a crucial role in investigating the electrophysiological activities of neuronal populations. Although two-dimensional (2D) neuronal cell cultures have predominated in neurophysiology in monitoring in-vitro the electrophysiological activity, recent research shifted toward culture using three-dimensional (3D) neuronal network structures for developing more sophisticated and realistic neuronal models. Nevertheless, many challenges remain in the electrophysiological analysis of 3D neuron cultures, among them the development of robust platforms for investigating the electrophysiological signal at multiple depths of the 3D neurons' networks. While various three-dimensional (3D) MEAs have been developed to probe specific depths within the layered nervous system, the fabrication of microelectrodes with different heights, capable of probing neural activity from the surface as well as from the different layers within the neural construct, remains challenging. This study presents a novel 3D MEA with microelectrodes of different heights, realized through a multi-stage mold-assisted electrodeposition process. Our pioneering platform allows meticulous control over the height of individual microelectrodes as well as the array topology, paving the way for the fabrication of 3D MEAs consisting of electrodes with multiple heights that could be tailored for specific applications and experiments. The device performance was characterized by measuring electrochemical impedance, and noise, and capturing spontaneous electrophysiological activity from neurospheroids derived from human induced Pluripotent Stem Cells (h-iPSCs). These evaluations unequivocally validated the significant potential of our innovative multi-height 3D MEA (MH-MEA) as an avant-garde platform for in vitro 3D neuronal studies.

Abstract:Biological cells are characterized by highly complex phenomena and processes that are, to a great extent, interdependent. To gain detailed insights, devices designed to study cellular phenomena need to enable tracking and manipulation of multiple cell parameters in parallel; they have to provide high signal quality and high-spatiotemporal resolution. To this end, we have developed a CMOS-based microelectrode array system for in vitro applications that integrates six measurement and stimulation functions, the largest number to date. Moreover, the system features the largest active electrode array area to date (<inline-formula> <tex-math notation="LaTeX">$4.48 \times 2.43$ </tex-math></inline-formula> mm<sup>2</sup>) to accommodate 59 760 electrodes, while its power consumption, noise characteristics, and spatial resolution (13.5-<inline-formula> <tex-math notation="LaTeX">$\mu$ </tex-math></inline-formula>m electrode pitch) are comparable to the best state-of-the-art devices. The system includes: 2048 action potential (AP, bandwidth: 300 Hz–10 kHz) recording units, 32 local-field-potential (LFP, bandwidth: 1 Hz–300 Hz) recording units, 32 current recording units, 32 impedance measurement units, and 28 neurotransmitter detection units, in addition to the 16 dual-mode voltage-only or current/voltage-controlled stimulation units. The electrode array architecture is based on a switch matrix, which allows for connecting any measurement/stimulation unit to any electrode in the array and for performing different measurement/stimulation functions in parallel.

Abstract:Background: The construction of multichannel micro-electrode arrays (MEA) generally requires complex and expensive procedures. Here we describe a simple, cheap, flexible method of linear MEA construction. New method: Our method allows manufacture of linear MEA (cross section ∼ 375 × 250 μm with 12 electrodes) of any desired length, with customized spacing of the electrode tips (down to a minimum spacing of 200 μm or less) to suit different needs and experiments. We describe the following steps: (1) set-up for MEA construction; (2) building of a construction jig; (3) building the reference, ground and optional electrical stimulation electrodes; (4) treatment of the main recording microwires; (5) soldering of the microwires to the main connector plug and arrangement of the microwires in a customizable array; and (6) testing of the MEA resistance and correct connections. Finally, we describe methods for quick surgical implantation of multiple MEAs and bipolar micro-stimulation electrodes for in vivo experiments in free-moving rats. Results: We provide examples of multi-site local field potentials from prolonged recordings in awake and free-moving rodents, with recordings viable for months, as well as samples of electrical stimulation effects on cortical and hippocampal recordings. Hippocampal recordings showed clear phase reversal and amplitude changes across its layers. Conclusions: We briefly discuss how the arrays can support other forms of stimulation such as optogenetic probes.
MEA-ToolBox: an Open Source Toolbox for Standardized Analysis of Multi-Electrode Array Data

Abstract:Functional assessment of in vitro neuronal networks-of relevance for disease modelling and drug testing-can be performed using multi-electrode array (MEA) technology. However, the handling and processing of the large amount of data typically generated in MEA experiments remains a huge hurdle for researchers. Various software packages have been developed to tackle this issue, but to date, most are either not accessible through the links provided by the authors or only tackle parts of the analysis. Here, we present ''MEA-ToolBox'', a free open-source general MEA analytical toolbox that uses a variety of literature-based algorithms to process the data, detect spikes from raw recordings, and extract information at both the single-channel and array-wide network level. MEA-ToolBox extracts information about spike trains, burst-related analysis and connectivity metrics without the need of manual intervention. MEA-ToolBox is tailored for comparing different sets of measurements and will analyze data from multiple recorded files placed in the same folder sequentially, thus considerably streamlining the analysis pipeline. MEA-ToolBox is available with a graphic user interface (GUI) thus eliminating the need for any coding expertise while offering functionality to inspect, explore and post-process the data. As proof-of-concept, MEA-ToolBox was tested on earlier-published MEA recordings from neuronal networks derived from human induced pluripotent stem cells (hiPSCs) obtained from healthy subjects and patients with neurodevelopmental disorders. Neuronal networks derived from patient's hiPSCs showed a clear phenotype compared to those from healthy subjects, demonstrating that the toolbox could extract useful parameters and assess differences between normal and diseased profiles.

Abstract:Engineered 3D neural tissues made of neurons and glial cells derived from human induced pluripotent stem cells (hiPSC) are among the most promising tools in drug discovery and neurotoxicology. They represent a cheaper, faster, and more ethical alternative to in vivo animal testing that will likely close the gap between in vitro animal models and human clinical trials. Micro-Electrode Array (MEA) technology is known to provide an assessment of compound effects on neural 2D cell cultures and acute tissue preparations by real-time, non-invasive, and long-lasting electrophysiological monitoring of spontaneous and evoked neuronal activity. Nevertheless, the use of engineered 3D neural tissues in combination with MEA biochips still involves series of constraints, such as drastically limited diffusion of oxygen and nutrients within tissues mainly due to the lack of vascularization. Therefore, 3D neural tissues are extremely sensitive to experimental conditions and require an adequately designed interface that provides optimal tissue survival conditions. A well-suited technique to overcome this issue is the combination of the Air-Liquid Interface (ALI) tissue culture method with the MEA technology. We have developed a full 3D neural tissue culture process and a data acquisition system composed of high-end electronics and novel MEA biochips based on porous, flexible, thin-film membranes integrating recording electrodes, named as "Strip-MEA," to allow the maintenance of an ALI around the 3D neural tissues. The main motivation of the porous MEA biochips development was the possibility to monitor and to study the electrical activity of 3D neural tissues under different recording configurations, (i) the Strip-MEA can be placed below a tissue, (ii) or by taking advantage of the ALI, be directly placed on top of the tissue, or finally, (iii) it can be embedded into a larger neural tissue generated by the fusion of two (or more) tissues placed on both sides of the Strip-MEA allowing the recording from its inner part. This paper presents the recording and analyses of spontaneous activity from the three positioning configurations of the Strip-MEAs. Obtained results are discussed with the perspective of developing in vitro models of brain diseases and/or impairment of neural network functioning.

Flexible EMG arrays with integrated electronics for scalable electrode density

Abstract:Recent developments in electrode technology have demonstrated the power of flexible microelectrode arrays (FMEAs) for measuring muscle activity at high resolution. We recently introduced the Myomatrix array, a FMEA optimized for measuring the activity of individual motor units (the collection of muscle fibers innervated by a single motor neuron) [ ] in freely behaving animals. Although FMEAs are fundamentally changing the way EMG is acquired, the number of recording channels is limited by the size of the plug that interfaces with the digital amplifier hardware and the density of electrode connections on the array. Increasing EMG channel count and supporting electrophysiological studies in smaller animals depends on two seemingly incompatible goals: reducing device size while increasing the number of recording channels. The solution to this is to increase channel density, which is currently limited by requiring that separate headstage and FMEA components be used simultaneously. In our prior devices [ ], each FMEA had a dedicated wire output for every electrode input, creating a channel density is 1 : 1. To improve this channel density, we have developed a novel device integrating a digital amplifier (bare-die RHD2216 chip, Intan, Inc. [ ]) directly onto an FMEA. This new design reduces the device’s backend footprint by 74% and relocates the intan bare die from the headstage to the FMEA itself, creating a channel density of 1 : 3.2. Our methodology combines standard FMEA microfabrication with wire-bonding and surface-mounted components, enabling direct integration into a Serial Peripheral Interface (SPI) connection into the device itself, without any separate headstage. With this initial device we see a 1 : 3.2 channel density, but our method allows for using other bare die amplifiers (Intan, Inc., USA) for a channel density of 1 : 12.8. Our findings present a robust technique for chip embedding in custom FMEAs, applicable to in-vivo electrophysiology

Novel high-dense microelectrode array based multimodal bioelectronic monitoring system for cardiac arrhythmia re-entry analysis

Abstract:In recent decades, significant progress has been made in the treatment of heart diseases, particularly in the field of personalized medicine. Despite the development of genetic tests, phenotyping and risk stratification are performed based on clinical findings and invasive in vivo techniques, such as stimulation conduction mapping techniques and programmed ventricular pacing. Consequently, label-free non-invasive in vitro functional analysis systems are urgently needed for more accurate and effective in vitro risk stratification, model-based therapy planning, and clinical safety profile evaluation of drugs. To overcome these limitations, a novel multilayer high-density microelectrode array (HD-MEA), with an optimized configuration of 512 sensing and 4 pacing electrodes on a sensor area of 100 mm2, was developed for the bioelectronic detection of re-entry arrhythmia patterns. Together with a co-developed front-end, we monitored label-free and in parallel cardiac electrophysiology based on field potential monitoring and mechanical contraction using impedance spectroscopy at the same microelectrode. In proof of principle experiments, human induced pluripotent stem cell (hiPS)-derived cardiomyocytes were cultured on HD-MEAs and used to demonstrate the sensitive quantification of contraction strength modulation by cardioactive drugs such as blebbistatin (IC50 = 4.2 μM), omecamtiv and levosimendan. Strikingly, arrhythmia-typical rotor patterns (re-entry) can be induced by optimized electrical stimulation sequences and detected with high spatial resolution. Therefore, we provide a novel cardiac re-entry analysis system as a promising reference point for diagnostic approaches based on in vitro assays using patient-specific hiPS-derived cardiomyocytes.
A Novel Dual Mode Microelectrode Array For Neuroelectrical And Neurochemical Recording In Vitro

Abstract:The communication between neurons is inherently electrical and chemical in nature. In situ, simultaneous acquisition for the dual mode signals is important for neuroscience research. In this paper, the concept of dual mode neural microelectrode array (MEA) sensor was proposed, and a low cost thin film MEA chip for in vitro test was fabricated using standard lithography technology. The sensor incorporates arrayed microelectrodes, a counter electrode and a reference electrode on one glass slide, which is suitable for electrophysiological and electrochemical recording in vitro. Electrophysiological recordings were carried out on acute hippocampus slice. Local field potentials and three different spike firing patterns with the amplitude ranging from +/- 20 mu V to +/- 60 mu V were acquired by the arrayed microelectrodes. Electrochemical current response of the microelectrodes to calibrated dopamine solution was tested. A good linear relationship between the current and dopamine concentration was observed, with the detection sensitivity of 4671 mu A mM(-1) cm(-2) and a correlation coefficient of 0.986. Abstract:Biological studies on in vitro cell cultures are of fundamental importance to investigate cells response to external stimuli, such as new drugs for treatment of specific pathologies, or to study communication between electrogenic cells. Although three-dimensional (3D) nanostructures brought tremendous improvements on biosensors used for various biological in vitro studies, including drug delivery and electrical recording, there is still a lack of multifunctional capabilities that could help gaining deeper insights in several bio-related research fields. In this work, the electrical recording of large cell ensembles and the intracellular delivery of few selected cells are combined on the same device by integrating microfluidics channels on the bottom of a multi-electrode array decorated with 3D hollow nanostructures. The novel platform allows to record intracellular-like action potentials from large ensembles of cardiomyocytes derived from human Induced Pluripotent Stem Cells (hiPSC) and from the HL-1 line, while different molecules are selectively delivered into single/few targeted cells. The proposed approach shows high potential for enabling new comprehensive studies that can relate drug effects to network level cell communication processes.

Abstract:Two‐photon polymerization (2PP) lithography and surface micromachining technology allow the rapid fabrication of polymer templates to guide the growth of electrodes for three‐dimensional microelectrode arrays (3D MEAs), enabling in turn, device customizability, high signal‐to‐noise ratio (SNR) recordings, low bending stiffness, and minimized cross‐sectional footprint for in vitro and in vivo neural applications: from 3D neuronal cell cultures to the cortex of living mice. Planar microelectrode arrays (MEAs) for – in vitro or in vivo – neuronal signal recordings lack the spatial resolution and sufficient signal‐to‐noise ratio (SNR) required for a detailed understanding of neural network function and synaptic plasticity. To overcome these limitations, a highly customizable three‐dimensional (3D) printing process is used in combination with thin film technology and a self‐aligned template‐assisted electrochemical deposition process to fabricate 3D‐printed‐based MEAs on stiff or flexible substrates. Devices with design flexibility and physical robustness are shown for recording neural activity in different in vitro and in vivo applications, achieving high‐aspect ratio 3D microelectrodes of up to 33:1. Here, MEAs successfully record neural activity in 3D neuronal cultures, retinal explants, and the cortex of living mice, thereby demonstrating the versatility of the 3D MEA while maintaining high‐quality neural recordings. Customizable 3D MEAs provide unique opportunities to study neural activity under regular or various pathological conditions, both in vitro and in vivo, and contribute to the development of drug screening and neuromodulation systems that can accurately monitor the activity of large neural networks over time.

Abstract:Objective.The micro-electrode array (MEA) is a cell-culture surface with integrated electrodes used for assays of electrically excitable cells and tissues. MEAs have been a workhorse in the study of neurons and myocytes, owing to the scalability and millisecond temporal resolution of the technology. However, traditional MEAs are opaque, precluding inverted microscope access to modern genetically encoded optical sensors and effectors.Approach. To address this gap, transparent MEAs have been developed. However, for many labs, transparent MEAs remain out of reach due to the cost of commercially available products, and the complexity of custom fabrication. Here, we describe an open-source transparent MEA based on the OpenEphys platform (Siegleet al2017J. Neural Eng.14045003).Main results.We demonstrate the performance of this transparent MEA in a multiplexed electrical and optogenetic assay of primary rat hippocampal neurons.Significance.This open-source transparent MEA and recording platform is designed to be accessible, requiring minimal microelectrode fabrication or circuit design experience. We include low-noise connectors for seamless integration with the Intan Technologies headstage, and a mechanically stable adaptor conforming to the 24-well plate footprint for compatibility with most inverted microscopes.

Abstract:The development of microelectrode arrays (MEAs) along with complementary advances in electronics, mechanics and software to connect with these arrays has led to the in vitro interfacing and benchtop electrophysiological models of several electrically active cells such as neurons and cardiomyocytes proving vital models and testing of human disease conditions in a dish/on a chip. This topical review deals with the micro/nanofabrication technology development of Microelectrodes Arrays from early silicon based developments to today's additive manufacturing technologies that have been employed to address bio-micro-electro-mechanical systems tool development in this space. Specifically 2D and 3D MEAs technologies have been reviewed in this paper along with a broad overview of some of the biological applications using these devices that are advancing the very state of biomedical research.

Abstract:Technological improvement of measurements for the electrical recordings from individual neurons within network is essential in neuroscience today. Here, we present a novel automatic positioning and sensing microelectrode array (APSMEA), which simultaneously positioned desired number of neurons onto 48 recording microelectrodes automatically and scathelessly by use of negative dielectrophoretic (DEP) forces, and facilitated the measurement of the electrophysiological activities of neuronal populations after functional synaptic connections formed between neurons. The results of multi-site electrophysiological recordings during drug administration also demonstrated the application of APSMEA in bioassay with cultured rat cortical neurons. Therefore, this device should benefit the investigation of neuronal networks in vitro with more comprehensive electrophysiological experiments, and also promise the possibility of a modular device for both cell manipulation and cell-based biosensor on microchip.

Abstract:This PhD thesis introduces the concept of meta-electrodes coupled with laser optoporation for high quality intracellular signals from hiPSCs-CM. These signals can be recorded on high-density commercial CMOS-MEAs from 3Brain characterized by thousands of electrode covered by a thin film of porous Platinum without any rework of the devices, 3D nanostructures or circuitry for electroporation. Subsequently, I attempted to translate these unique features of low invasiveness and reliability to other commercial MEA platforms, in order to develop a new tool for cardiac electrophysiological accurate recordings.

Interfacing Microfluidics with Microelectrode Arrays for Studying Neuronal Communication and Axonal Signal Propagation.

Abstract:Microelectrode arrays (MEAs) are widely used to study neuronal function in vitro. These devices allow concurrent non-invasive recording/stimulation of electrophysiological activity for long periods. However, the property of sensing signals from all sources around every microelectrode can become unfavorable when trying to understand communication and signal propagation in neuronal circuits. In a neuronal network, several neurons can be simultaneously activated and can generate overlapping action potentials, making it difficult to discriminate and track signal propagation. Considering this limitation, we have established an in vitro setup focused on assessing electrophysiological communication, which is able to isolate and amplify axonal signals with high spatial and temporal resolution. By interfacing microfluidic devices and MEAs, we are able to compartmentalize neuronal cultures with a well-controlled alignment of the axons and microelectrodes. This setup allows recordings of spike propagation with a high signal-to-noise ratio over the course of several weeks. Combined with specialized data analysis algorithms, it provides detailed quantification of several communication related properties such as propagation velocity, conduction failure, firing rate, anterograde spikes, and coding mechanisms. This protocol demonstrates how to create a compartmentalized neuronal culture setup over substrate-integrated MEAs, how to culture neurons in this setup, and how to successfully record, analyze and interpret the results from such experiments. Here, we show how the established setup simplifies the understanding of neuronal communication and axonal signal propagation. These platforms pave the way for new in vitro models with engineered and controllable neuronal network topographies. They can be used in the context of homogeneous neuronal cultures, or with co-culture configurations where, for example, communication between sensory neurons and other cell types is monitored and assessed. This setup provides very interesting conditions to study, for example, neurodevelopment, neuronal circuits, information coding, neurodegeneration and neuroregeneration approaches.
