Abstract:In this paper we present an efficient method to detect hardware Trojans under high levels of process variations, by measuring delays for vectors generated using a minimally delay-invasive Trojan model. Our method focuses on various sources of process variations and significantly reduces the effects of variations on delays by calibrating delays measured on each fabricated chip. Using test structures and additional measurements on these, our approach significantly reduces the number of chips that we need to test to detect minimally-invasive Trojans with a desired level of confidence. Our approach for tackling high levels of process variations can be used in conjunction with any other type of parametric Trojan detection strategy to significantly improve its efficiency.

Abstract:Oxidative potential (OP) has been proposed as a useful descriptor for the ability of particulate matter (PM) to generate reactive oxygen species (ROS) and consequently induce oxidative stress in biological systems, which has been recognized as one of the most important mechanisms responsible for PM toxicity. The dithiothreitol (DTT) assay is one of the most frequently used techniques to quantify OP because it is low-cost, easy-to-operate, and has high repeatability. With two thiol groups, DTT has been used as a surrogate of biological sulfurs that can be oxidized when exposed to ROS. Within the DTT measurement matrix, OP is defined as the DTT consumption rate. Often, the DTT consumption can be attributed to the presence of transition metals and quinones in PM as they can catalyze the oxidation of DTT through catalytic redox reactions. However, the DTT consumption by non-catalytic PM components has not been fully investigated. In addition, weak correlations between DTT consumption, ROS generation, and cellular responses have been observed in several studies, which also reveal the knowledge gaps between DTT-based OP measurements and their implication on health effects. In this review, we critically assessed the current challenges and limitations of DTT measurement, highlighted the understudied DTT consumption mechanisms, elaborated the necessity to understand both PM-bound and PM-induced ROS, and concluded with research needs to bridge the existing knowledge gaps.

Abstract:The exposure to ambient fine particulate matter (PM2.5) can induce oxidative stress, contributing to global burden of diseases. The evaluation of the oxidative potential (OP) of PM2.5 is thus critical for the health risk assessment. We collected ambient PM2.5 samples in Hangzhou city, China for four consecutive quarters in the year 2017 and investigated the oxidation property of PM2.5 components by the dithiothreitol (DTT) assay. The annual mean of ambient PM2.5 mass concentrations in 2017 was 63.05 μg m-3 (median: 57.34, range: 6.67-214.33 μg m-3) with the significant seasonal variations ranking as winter > spring > summer > autumn. Secondary inorganic aerosol (SIA) species including SO42-, NO3- and NH4+ totally account for >50% of PM2.5 mass. The annual mean volume-normalized DTT activity (DTTv) showed a relatively high value of 0.62 nmol/min/m3 (median: 0.62, range: 0.11-1.66 nmol/min/m3) and DTTv of four seasons was roughly at the same level, indicating a high annual exposure level of ambient PM2.5. SIA species were correlated well with the corresponding DTTv and showed significant diurnal variations with strong or moderate correlations at day and weak correlations at night, suggesting strong secondary formation in daytime with contribution to the particulate OP. The annual mean mass-normalized DTT activity (DTTm) had a relatively low value of 6.39 pmol/min/μg (median: 5.63, range: 1.99-22.70 pmol/min/μg), indicating low intrinsic oxidative toxicity. The DTTm of four seasons ranked as autumn > winter > spring > summer, indicating seasonal variations of the DTT-active components. The PM2.5 mass concentration is more related to exposure levels than intrinsic properties of components, while OP is determined by the components rather than PM2.5 mass concentration. Our results provide an insight into reactive oxygen species-induced health risk of PM2.5 exposure and decision for subsequent emission control.

Abstract:Oxidative potential (OP) assays are feasible methods to comprehensively understand how exposure to atmospheric chemical components can influence the formation of reactive oxygen species (ROS) in the human body. The increase of ROS concentration can enhance the oxidation of numerous components, such as of the DNA, proteins, and lipids, which cause mutations and cell damage, leading to respiratory illness. According to available studies, the mechanisms of PM-related health effects are not totally understood. The aim of the present study is to assess which available assays are suitable for evaluating the OP of different Mineral dust (MD) samples and what limitations may apply to either method. Cell-free assays are assessed including methods based on the interaction of ascorbic acid (AA) and dithiothreitol (DTT) with soluble aerosol chemical components.Oxidative potential experiments were carried out on both commercially available standard solutions, ultrapure water (DTT) and buffer solution (AA), as well as on soluble extracts of one-quarter filter samples obtained using both shaking and ultrasound procedures. Most limitations were related to high concentrations of transition metals in the buffer solution composition, which was treated using Chelex 100 sodium resin. Transition metals were strongly correlated to both methods, such as Ti, Cr, V, and Mn for DTT assays, and Fe, and Sr for AA assay. The mineral dust OP values were lower than the OP of particulate matter samples from urban metropolitan centers. Such results could be related to the fewer metal and quinone concentrations in the MD samples in comparison to the urban sample. DTT assay has shown more sensibility to the MD content compared with AA chemical procedure. These assays contribute to building an impact-evaluation model for assessing the variation of MD OP based on its different chemical composition.

Abstract:Acellular assays of oxidative potential (OP) induced by ambient particulate matters (PMs) are of great significance in screening for toxicity in PMs. In this review, several typical OP measurement techniques, including the respiratory tract lining fluid assay (RTLF), ascorbate depletion assay (AA), dithiothreitol assay (DTT), chemiluminescent reductive acridinium triggering (CRAT), dichlorofluorescin assay (DCFH) and electron paramagnetic/spin resonance assay (EPR/ESR) are discussed and their sensitivity to different PMs species composition, PMs size distribution and seasonality is compared. By comparison, the DTT assay tends to be the preferred method providing a more comprehensive measurement with transition metals and quinones accumulated in the fine PMs fraction. Specific transition metals (i.e., Mn, Cu, Fe) and quinones are found to contribute OPDTT directly whereas the redox properties of PMs species may be changed by the interactions between themselves. The selection of the appropriate OP measurement methods and the accurate analysis of the relationship between the methods and PM components is conducive to epidemiological researches which are related with oxidative stress induced by PMs exposure.

Abstract:Various methods have been applied to quantify oxidative stress induced by atmospheric particle matter (PM), which are often employing different principles, dissimilar positive controls, normalization techniques and reporting units, as well as displaying quite diverse sensitivities. To date, no universal method has been accepted and the need remains to identify and validate a method that could easily be utilized across varied study designs, different types of samples, and be reproducible between different laboratories. In this study we propose and demonstrate the utility of a measure of biological ROS response utilizing a broad ROS probe (DCFH-DA) in an in vitro rat alveolar macrophage system. A method comparison and validation experiments were conducted in two independent laboratories. Method performance was characterized using extracts of ambient PM as well as a variety of positive control materials. We demonstrate that our method can yield very good reproducibility, sensitivity and accuracy. The coefficient of variation (CV) of the untreated control group and positive controls ranged from 8% to 11% and 7%-13%, respectively, while the average signal to noise ratio (SNR) of untreated (negative) control group and positive controls for both labs was larger than 3. Comparing the dose response relationship for multiple positive controls between the two labs, the NIST reference material (Urban Dust) extract appeared to be the most consistent reference material for the ROS measurement (R2 = 0.996). The relative percent difference (RPD) of ROS activities as measured by two different labs and expressed as equivalents of Urban Dust was only 14%, which suggests that as the optimal normalization method. Our results indicate the macrophage-DCFH-DA method is effective in assessing the biological oxidative potential of PM exposure in different laboratories and should be utilized for cross-comparison of biological ROS response between different studies.

Abstract:Oxidative stress is one of the key mechanisms linking ambient particulate matter (PM) exposure with various adverse health effects. The oxidative potential of PM has been used to characterize the ability of PM induced oxidative stress. Hydroxyl radical (•OH) is the most destructive radical produced by PM. However, there is currently no high-throughput approach which can rapidly measure PM-induced •OH for a large number of samples with an automated system. This study evaluated four existing molecular probes (disodium terephthalate, 3'-p-(aminophenyl)fluorescein, coumarin-3-carboxylic acid, and sodium benzoate) for their applicability to measure •OH induced by PM in a high-throughput cell-free system using fluorescence techniques, based on both our experiments and on an assessment of the physicochemical properties of the probes reported in the literature. Disodium terephthalate (TPT) was the most applicable molecular probe to measure •OH induced by PM, due to its high solubility, high stability of the corresponding fluorescent product (i.e., 2-hydroxyterephthalic acid), high yield compared with the other molecular probes, and stable fluorescence intensity in a wide range of pH environments. TPT was applied in a high-throughput format to measure PM (NIST 1648a)-induced •OH, in phosphate buffered saline. The formed fluorescent product was measured at designated time points up to 2 h. The fluorescent product of TPT had a detection limit of 17.59 nM. The soluble fraction of PM contributed approximately 76.9% of the •OH induced by total PM, and the soluble metal ions of PM contributed 57.4% of the overall •OH formation. This study provides a promising cost-effective high-throughput method to measure •OH induced by PM on a routine basis.

Abstract:Exposure to ambient air pollution has an adverse influence on human health. There is increasing evidence that oxidative potential (OP), the capacity of airborne pollutants to oxidize target molecules by generating redox oxidizing species, is a plausible metric for particulate matter (PM) toxicity. Here we describe the commonly used acellular techniques for measuring OP (respiratory tract lining fluid, dithiothreitol, ascorbic acid, and electron paramagnetic resonance assays) and review the PM chemical constituents that have been identified to drive the OP response. We further perform a review of the epidemiologic literature to identify studies that reported an association between exposure to ambient PM and a health outcome in a human population, and in which exposure was measured by both PM mass concentration and OP. Laboratory studies have shown that specific redox-active metals and quinones are able to contribute OP directly. However, interactions among PM species may alter the redox properties of PM components. In ambient PM measurements, all OP assays were found to be correlated with metals (Fe, Cu) and organic species (photochemically aged organics). Across the epidemiological studies reviewed, associations between fine PM (PM2.5) mass and cardio-respiratory outcomes were found to be stronger at elevated OP levels but findings varied across the different OP measurement techniques. Future work should aim to identify specific situations in which PM OP can improve air pollution exposure assessment and/or risk management. This may be particularly useful in countries with low PM2.5 mass concentrations over broad spatial scales where such information may greatly improve the efficiency of risk management activities.

Associations between particle physicochemical characteristics and oxidative capacity: An indoor PM10 study in Beijing, China

Abstract:A study was undertaken to determine the use of a plasmid DNA scission assay to evaluate the causal relationships between particle oxidative capacity and physico-chemistry. Field emission scanning electron microscopy (FESEM), image analysis (IA) and inductively coupled plasma-mass spectrometry (ICP-MS) were employed to investigate the physico-chemical characteristics of indoor PM10 (particulate matter with an aerodynamic diameter of 10μm or less) in Beijing, China. Six PM10 samples (indoor smoker's living room; indoor non-smoker's living room and kitchen; and outdoor Beijing city; winter versus summer) were selected to represent typical indoor Beijing PM10 environments that contain high particle mass. The PM10 collected from a kitchen and two smoker's homes had the lowest TD50 (toxic dosage of PM10 causing 50% plasmid DNA damage), being as low as 45μgml−1 (kitchen) and 100μgml−1 (living room), which suggests a high oxidative capacity, with the PM10 generated in kitchens appearing to be the most toxic. The indoor PM10 from the non-smoker's home and outdoor PM10 samples demonstrated high TD50 values and were deemed less bioreactive (i.e. caused limited DNA damage). FESEM observations revealed that four types of particle species were prevalent in Beijing indoor PM10; soot aggregates, minerals, coal fly ash and unknown fine particles. IA showed that higher percentages of soot and unknown fine particles were associated with the lower TD50 values, suggesting that soot and the unknown fine particles may be important components responsible for the observed plasmid DNA damage. The water-soluble trace elements were negatively correlated with the TD50 values, implying that the DNA damage may be attributed to the water-soluble fraction of the PM10. Water-soluble zinc revealed the best relationship with the TD50 values than other analyzed elements, signifying it may play a role in driving the oxidative damage.
Development of a Physiologically Relevant Online Chemical Assay To Quantify Aerosol Oxidative Potential

Abstract:Large-scale epidemiological studies have shown a close correlation between adverse human health effects and exposure to ambient particulate matter (PM). The oxidative potential (OP) of ambient PM has been implicated in inducing toxic effects associated with PM exposure. In particular, reactive oxygen species (ROS), either bound to PM or generated by particulate components in vivo, substantially contribute to the OP and therefore toxicity of PM by lowering antioxidant concentrations in the lung, which can subsequently lead to oxidative stress, inflammation, and disease. Traditional methods for measuring aerosol OP are labor intensive and have poor time resolution, with significant delays between aerosol collection and ROS analysis. These methods may underestimate ROS concentrations in PM because of the potentially short lifetime of some ROS species; therefore, continuous online, highly time-resolved measurement of ROS components in PM is highly advantageous. In this work, we develop a novel online method for measuring aerosol OP based on ascorbic acid chemistry, an antioxidant prevalent in the lung, thus combining the advantages of continuous online measurement with a physiologically relevant assay. The method limit of detection is estimated for a range of atmospherically important chemical components such as Cu(II) 0.22 ± 0.03 μg m-3, Fe(II) 47.8 ± 5.5 μg m-3, Fe(III) 0.63 ± 0.05 μg m-3, and secondary organic aerosol 41.2 ± 6.9 μg m-3, demonstrating that even at this early stage of development, the online method is capable of measuring the OP of PM in polluted urban environments and smog chamber studies.
Oxidative potential of particulate matter collected at sites with different source characteristics

Abstract:BACKGROUND: The oxidative potential (OP) of particulate matter (PM) has been proposed as a more health relevant metric than PM mass. Different assays exist for measuring OP and little is known about how the different assays compare.AIM: To assess the OP of PM collected at different site types and to evaluate differences between locations, size fractions and correlation with PM mass and PM composition for different measurement methods for OP.METHODS: PM2.5 and PM10 was sampled at 5 sites: an underground station, a farm, 2 traffic sites and an urban background site. Three a-cellular assays; dithiothreitol (OP(DTT)), electron spin resonance (OP(ESR)) and ascorbate depletion (OP(AA)) were used to characterize the OP of PM.RESULTS: The highest OP was observed at the underground, where OP of PM10 was 30 (OP(DTT)) to >600 (OP(ESR)) times higher compared to the urban background when expressed as OP/m(3) and 2-40 times when expressed as OP/μg. For the outdoor sites, samples from the farm showed significantly lower OP(ESR) and OP(AA), whereas samples from the continuous traffic site showed the highest OP for all assays. Contrasts in OP between sites were generally larger than for PM mass and were lower for OP(DTT) compared to OP(ESR) and OP(AA). Furthermore, OP(DTT)/μg was significantly higher in PM2.5 compared to PM10, whereas the reverse was the case for OP(ESR). OP(ESR) and OP(AA) were highly correlated with traffic-related PM components (i.e. EC, Fe, Cu, PAHs), whereas OP(DTT) showed the highest correlation with PM mass and OC.CONCLUSIONS: Contrasts in OP between sites, differences in size fractions and correlation with PM composition depended on the specific OP assay used, with OP(ESR) and OP(AA) showing the most similar results. This suggests that either OP(ESR) or OP(AA) and OP(DTT) can complement each other in providing information regarding the oxidative properties of PM, which can subsequently be used to study its health effects.

Abstract:Atmospheric particulate matter (PM) has been related to numerous adverse health effects in humans. Nowadays, it is believed that one of the possible mechanisms of toxicity could be the oxidative stress, which involves the development of reactive oxygen species (ROS). Different assays have been proposed to characterize oxidative stress, such as dithiothreitol (DTT) and ascorbic acid (AA) acellular assays (OP DTT and OP AA ), as a metric more relevant than PM mass measurement for PM toxicity. This study evaluates the OP of the bioaccessible fraction of 65 PM 10 samples collected at an Atlantic Coastal European urban site using DTT and AA assays. A physiologically based extraction (PBET) using Gamble's solution (GS) as a simulated lung fluid (SLF) was used for the assessment of the bioaccessible fraction of PM 10 . The use of the bioaccessible fraction, instead of the fraction assessed using conventional phosphate buffer and ultrasounds assisted extraction (UAE), was compared for OP assessment. Correlations between OP DTT and OP AA , as well as total and bioaccessible concentrations of polycyclic aromatic hydrocarbons (PAHs) and metal(oid)s, were investigated to explore the association between those compounds and OP. A correlation was found between both OP (OP DTT and OP AA ) and total and bioaccessible concentrations of PAHs and several metal(oid)s such as As, Bi, Cd, Cu, Ni, and V. Additionally, OP DTT was found to be related to the level of K + .

Abstract:Today air pollution caused by particulate matter (PM) is a global issue, especially in densely populated and high-traffic cities. The formation of reactive oxygen species (ROS) by various toxicological studies is considered as one of the important effects caused by airborne particles that can lead to adverse effects on human health. In this study, to answer the question of whether particle size affects oxidative potential (OP), we searched the main databases, including PubMed, Scopus, Embase, and Web of Science, and defined search strategy based on the MESH terms for the above-mentioned search engines. All articles published until 2021 were searched. An ANOVA was run using R software to show the correlation between the size distributions of particulate matter and oxidative potential (base on mass and volumetric units) in ambient air. As expected, the regression results showed that the relationship between particle size and OP values for the studies based on mass-logarithm has a significant difference in the different distribution size categories, which was related to the difference between the <2.5 and < 1 categories. However, ANOVA analysis did not show a significant difference in the volumetric OP logarithm in the different distribution size categories. In this study, it was found that sizes higher than 2.5 μm did not have much effect on human health, and it is recommended that future research focus on PM2.5.

Abstract:Oxidative potential (OP) has been identified as an important factor underlying the health effects of airborne particulate matter (PM). OP denotes the ability of PM to deplete antioxidants and form reactive oxygen species (ROS) in the lung. OP can be quantified using a variety of chemical assays and analytical techniques. Despite the widespread use of various OP metrics, there is no consensus or comprehensive inter-laboratory assessment on how these assays compare. In this work for the first time, we compared 11 OP indicators from acellular assays using standard reference material of urban PM. The OP indicators included ascorbic acid, glutathione, glutathione disulfide, cysteine, cystine, dithiothreitol, H 2 O 2 , • OH, O 2 •- , and empirical and theoretical redox potential of simulated lung lining fluid (SLF). The indicators showed first-order kinetics at low PM concentration (25 μg mL −1 ), whereas the kinetics were non-linear at higher PM concentrations. The indicators demonstrated mainly linear dose-response relationships at PM concentrations 25–100 μg mL −1 , following similar trends with water-soluble transition metals, but they were rarely proportional to PM concentrations, and demonstrated substantial differences in their sensitivities to PM. The results indicate the importance of using reduced reaction time for reliable OP quantification due to non-linearity in assay responses at high PM concentrations. This work shows that the choice of molecular probes and measurement techniques must be carefully considered when planning studies on OP of ambient air and should importantly include the association of OP metrics with health outcomes.

Abstract:An alternative metric to account for particulate matter (PM) composition-based toxicity is the ability of PM-species to generate reactive oxygen species (ROS) and deplete antioxidants, the so-called oxidative potential (OP). Acellular OP assays are the most used worldwide, mainly those based on ascorbic acid (AA) and dithiothreitol (DTT) depletion; OP values are calculated from AA/DTT concentration over time kinetic curves. Since a great variability in OP-DTT and OP-AA values can be found in the literature, the understanding of those factors affecting the kinetic rate of AA and DTT oxidation in the presence of PM-bound species will improve the interpretation of OP values. In this work, a kinetic study of the oxidation rate of AA and DTT driven by species usually found in PM (transition metals and naphthoquinone (NQ)) was carried out. In particular, the influence of the concentration of Cu(II), Fe(II), Fe(III), Mn(II), Mn(III), and 1,4-NQ, and the type of fluid used in the assay (phosphate buffer (PB), phosphate buffer saline (PBS) and artificial lysosomal fluid (ALF)) is analysed and discussed. The reaction orders with respect to the AA/DTT and the active compound, and the kinetic rate constants were also determined. The results show great variability in OP values among the studied species depending on the fluid used; the OP values were mostly higher in PB0.05M, followed by PBS1x and ALF. Moreover, different species concentration-responses for OP-DTT/OP-AA were obtained. These differences were explained by the different reaction orders and kinetic rate constants obtained for each active compound in each fluid.

Abstract:Vehicle emissions are an important source of particulate matter (PM) in urban areas and have well-known adverse health effects on human health. Oxidative potential (OP) is used as a quantification metric for indexing PM toxicity. In this study, by using a liquid spot sampler (LSS) and the dithiothreitol (DTT) assay, the diurnal OP variation was assessed at a ground-level urban monitoring station. Besides, since the monitoring station was adjacent to the main road, the correlation between OP and traffic volume was also evaluated. PM components, including metals, water-soluble inorganic aerosols (WSIAs), black carbon (BC), and polycyclic aromatic hydrocarbons (PAHs), were also simultaneously monitored. The daytime and evening mean ± std volume-normalized OP (OPv) were 0.46 ± 0.27 and 0.48 ± 0.26 nmol/min/m3, and exhibited good correlations with PM1.0 and BC; however, these concentrations were only weakly correlated with mass-normalized OP (OPm). The mean ± std OPm was higher in the daytime (41.3 ± 13.8 pmol/min/μg) than in the evening (36.1 ± 11.5 pmol/min/μg). According to the PMF analysis, traffic emissions dominated the diurnal OP contribution. Organic matter and individual metals associated with non-exhaust traffic emissions, such as Mn, Fe, and Cu, contributed substantially to OP. Diurnal variations of PAH concentrations suggest that photochemical reactions could enhance OP, highlighting the importance of atmospheric aging on PM toxicity.

Abstract:Particulate matter (PM) air pollution is recognized as a major cause of adverse health outcomes, yet much remains unknown about which fractions and compositions are the most harmful. Efforts in this direction can be improved by employing a cell‐free approach, dithiothreitol (DTT) assay, the so‐called oxidative potential (OP). Here, we provide a detailed examination of OP from fractionated PM by a combination extraction method of solvent and solid‐phase extractions (SPE). We estimate that the average OP of water‐soluble matter (WSM; 4.27 nmol/min/m3) is up to 1.2 times higher than methanol‐soluble matter (3.66 nmol/min/m3) in most seasons apart from summer. Of these WSM, OP is distributed almost equally between hydrophobic and hydrophilic fractions. OP of PM is mainly related to transition metals (Cu: 12.1%, Mn: 2.5%); apart from these, more than 80% of the total OP remains unidentified. Identification of this unexplained OP by a novel approach, integrating the chromophore characteristics with parallel factor analysis, has shown that chromophore1 (C1) accounted for 98% of OP, although this factor only made a small contribution to the total fluorescence. Our paper highlights the importance of a holistic approach by evaluating the chemical structure‐activity relationship between optical characteristics and OP. Information presented here draws a connection between light‐absorbing organic aerosols and a possible toxicological mechanism on the basis of their optical properties.

Abstract:Large-scale epidemiological studies have consistently shown that exposure to ambient particulate matter (PM) is responsible for a variety of adverse health effects. However, the specific physical and chemical properties of particles that are responsible for the observed health effects, as well as the underlying mechanisms of particle toxicity upon exposure, remain largely uncertain. Studies have widely suggested that the oxidative potential (OP) of aerosol particles is a key metric to quantify particle toxicity. OP is defined as the ability of aerosol particle components to produce reactive oxidative species (ROSs) and deplete antioxidants in vivo. Traditional methods for measuring OP using acellular assays largely rely on analyzing PM collected in filters offline. This is labor intensive and involves a substantial time delay between particle collection and OP analysis. It therefore likely underestimates particle OP because many reactive chemical components which contribute to OP are short-lived and therefore degrade prior to offline analysis. Thus, new techniques are required to provide a robust and rapid quantification of particle OP, capturing the chemistry of oxidizing and short-lived, highly reactive aerosol components and their concentration dynamics in the atmosphere. To address these measurement shortcomings, we developed a portable online instrument that directly samples particles into an ascorbic acid-based assay under physiologically relevant conditions of pH 6.8 and 37 ∘ C, providing continuous, accurate OP measurements with a high time resolution (5 min). The instrument runs autonomously for up to 3 d and has a detection limit of about 5 μg m −3 in an urban environment, which allows the characterization of particle OP even in low-pollution areas.
