Abstract:Excessive oxidative stress has been recognized as an important cause of the adverse health effects associated with exposure to ambient particulate matter (PM). Transition metals (TMs) (e.g., iron (Fe) and copper (Cu)) are known catalysts in the formation of reactive oxygen species (ROS) in surrogate lung fluid containing antioxidants. Humic-like substances (HULIS), extracted from atmospheric aerosols, retain the compositional complexity of real-world samples. It contains mixtures of organics that chelate TMs and was used in this work to examine the roles of atmospheric organics in affecting ROS formation and antioxidant depletion by TMs. Two types of metal-binding organics known to be present in HULIS, oxygen-containing (i.e., carboxylic acids) and reduced-nitrogen-containing organics (i.e., imidazoles), were first investigated for their effects on the ascorbic acid depletion (denoted as OPAA) and hydroxyl radical formation (denoted as OP•OH) from both Fe(II) and Cu(II) in phosphate buffered saline (pH 7.40) containing ascorbic acid. Our results show that carboxylic acids enhance the OPAA and OP•OH by TMs while imidazoles suppress them. Similar experiments using three HULIS samples with distinctly different chemical compositions revealed complexity in metal-organics interactions. While ambient HULIS showed negligible impacts, two biomass burning source HULIS samples from rice straw and sugar cane leaf burning displayed unambiguous suppression or enhancement effects on OPAA and OP•OH by TMs. The effect was metal-specific and source HULIS-specific. The distinct behaviors of the three HULIS types can be explained by their different chemical compositions, for example, outstanding higher level of alkaloid compounds (e.g., imidazoles) in rice straw burning HULIS was consistent with the suppression effect exerted by this source of HULIS. In addition, we found OPAA and OP•OH are well-correlated while the proportion of OP•OH/OPAA by Cu is noticeably lower than that by Fe, indicating varying sensitivity of the metals to different OP end points. Our work highlights the importance and complexity of metal-organics interactions and the advantages of comeasurements of ROS generation and antioxidant depletion when assessing oxidative stress elicited by atmospheric PM.
Synergistic and antagonistic interactions among organic and metallic components of the ambient particulate matter (PM) for the cytotoxicity measured by Chinese hamster ovary cells

Abstract:Although PM2.5 toxicity is known to be related to its chemical composition, the effect of interactions among various particles' components on the toxicity is not well explored. To understand these interactions, especially metals and organic compounds on PM2.5 cytotoxicity, we chose several redox-active substances known to be present in the ambient particles such as metals (Cu, Fe, and Mn) and quinones [9,10-phenanthraquinone (PQ), 1,2-naphthoquinone (1,2-NQ), 1,4-naphthoquinone (1,4-NQ), and 5-hydroxy-1,4-naphthoquinone (5,H-1,4-NQ)]. Cytotoxicity was assessed through a Chinese hamster ovary (CHO) cells assay and expressed by a median lethal concentration (LC50). Two methods were employed to assess the interactions. In the first method, we tested the impact of nontoxic level of a component on the LC50 of other components. In the second method, we mixed two components in different concentration ratios to expose the cells and calculated a mixture toxicity index (MTI). MTI is a composite value to quantify the nature of interactions such that the interactions are considered synergistic when MTI > 1, additive when 0 < MTI ≤ 1 and antagonistic when MTI < 0. The interactions between quinones and metals were largely synergistic by both methods. To further assess the environmental relevance of these mixtures, we extracted organic compounds termed as water-soluble Humic-like substances (HULIS) from real ambient PM samples and mixed them with individual metals. A similar pattern, as observed from the interaction of quinones and metals, was found. Moreover, the interactions became more synergistic as the relative concentration of metals with respect to water-soluble HULIS was decreased in these mixtures. With environmentally relevant mass concentration ratios of organics to metals (75-7500), the interactions were strongly synergistic (MTI = 1-115). These results indicate the importance of incorporating the interaction among various PM components for estimating the net toxicity of ambient PM2.5.

Abstract:Oxidative potential (OP) has been proposed as a useful descriptor for the ability of particulate matter (PM) to generate reactive oxygen species (ROS) and consequently induce oxidative stress in biological systems, which has been recognized as one of the most important mechanisms responsible for PM toxicity. The dithiothreitol (DTT) assay is one of the most frequently used techniques to quantify OP because it is low-cost, easy-to-operate, and has high repeatability. With two thiol groups, DTT has been used as a surrogate of biological sulfurs that can be oxidized when exposed to ROS. Within the DTT measurement matrix, OP is defined as the DTT consumption rate. Often, the DTT consumption can be attributed to the presence of transition metals and quinones in PM as they can catalyze the oxidation of DTT through catalytic redox reactions. However, the DTT consumption by non-catalytic PM components has not been fully investigated. In addition, weak correlations between DTT consumption, ROS generation, and cellular responses have been observed in several studies, which also reveal the knowledge gaps between DTT-based OP measurements and their implication on health effects. In this review, we critically assessed the current challenges and limitations of DTT measurement, highlighted the understudied DTT consumption mechanisms, elaborated the necessity to understand both PM-bound and PM-induced ROS, and concluded with research needs to bridge the existing knowledge gaps.

Insights into reactive oxygen species formation induced by water-soluble organic compounds and transition metals using spectroscopic method

Abstract:Ambient particulate matter (PM) can cause adverse health effects via their ability to produce Reactive Oxygen Species (ROS). Water-Soluble Organic Compounds (WSOCs), a complex mixture of organic compounds which usually coexist with Transition Metals (TMs) in PM, have been found to contribute to ROS formation. However, the interaction between WSOCs and TMs and its effect on ROS generation are still unknown. In this study, we examined the ROS concentrations of V, Zn, Suwannee River Fulvic Acid (SRFA), Suwannee River Humic Acid (SRHA) and the mixtures of V/Zn and SRFA/SRHA by using a cell-free 2',7'-Dichlorodihydrofluorescein (DCFH) assay. The results showed that V or Zn synergistically promoted ROS generated by SRFA, but had an antagonistic effect on ROS generated by SRHA. Fluorescence quenching experiments indicated that V and Zn were more prone to form stable complexes with aromatic humic acid-like component (C1) and fulvic acid-like component (C3) in SRFA and SRHA. Results suggested that the underlying mechanism involving the fulvic acid-like component in SRFA more tending to complex with TMs to facilitate ROS generation through π electron transfer. Our work showed that the complexing ability and complexing stability of atmospheric PM organics with metals could significantly affect ROS generation. It is recommended that the research deploying multiple analytical methods to quantify the impact of PM components on public health and environment is needed in the future.

Singlet Oxygen Triggered by Superoxide Radicals in a Molybdenum Cocatalytic Fenton Reaction with Enhanced REDOX Activity in the Environment.

Abstract:As an important reactive oxygen species (ROS) with selective oxidation, singlet oxygen (1O2) has wide application prospects in biology and the environment. However, the mechanism of 1O2 formation, especially the conversion of superoxide radicals (·O2-) to 1O2, has been a great controversy. This process is often disturbed by hydroxyl radicals (·OH). Here, we develop a molybdenum cocatalytic Fenton system, which can realize the transformation from ·O2- to 1O2 on the premise of minimizing ·OH. The Mo0 exposed on the surface of molybdenum powder can significantly improve the Fe3+/Fe2+ cycling efficiency and weaken the production of ·OH, leading to the generation of ·O2-. Meanwhile, the exposed Mo6+ can realize the transformation of ·O2- to 1O2. The molybdenum cocatalytic effect makes the conventional Fenton reaction have high oxidation activity for the remediation of organic pollutants and prompts the inactivation of Staphylococcus aureus, as well as the adsorption and reduction of heavy metal ions (Cu2+, Ni2+, and Cr6+). Compared with iron powder, molybdenum powder is more likely to promote the conversion from Fe3+ to Fe2+ during the Fenton reaction, resulting in a higher Fe2+/Fe3+ ratio and better activity regarding the remediation of organics. Our findings clarify the transformation mechanism from ·O2- to 1O2 during the Fenton-like reaction and provide a promising REDOX Fenton-like system for water treatment.
Particle Size Distributions of Oxidative Potential of Lung-Deposited Particles: Assessing Contributions from Quinones and Water-Soluble Metals.

Abstract:Redox-active species in ambient particulate matter (PM) cause adverse health effects through the production of reactive oxygen species (ROS) in the human respiratory tract. However, respiratory deposition of these species and their relative contributions to oxidative potential (OP) have not been described. Size-segregated aerosols were collected during haze and nonhaze periods using a micro-orifice uniform deposit impactor sampler at an urban site in Shanghai to address this issue. Samples were analyzed for redox-active species content and PM OP. The average dithiothreitol (DTT) activity of haze samples was approximately 2.4-fold higher than that of nonhaze samples and significantly correlated with quinone and water-soluble metal concentrations. The size-specific distribution data revealed that both water-soluble OPvDTT (volume-normalized OP quantified by DTT assay) and OPmDTT (mass-normalized OP) were unimodal, peaking at 0.56-1 and 0.1-0.32 μm, respectively, due to contributions from accumulation-mode quinones and water-soluble metals. We further estimated that transition metals (mainly copper and manganese) contributed 55 ± 13% of the DTT activity while quinones accounted for only 8 ± 3%. Multiple-path particle dosimetry calculations estimated that OP deposition in the pulmonary region was mainly from accumulation-mode transition metals despite quinones having the highest DTT activity. This behavior is primarily attributed to the efficiency of deposition of transition metals in the pulmonary region being approximately 1.2-fold greater than that of quinones. These results reveal that accumulation-mode transition metals are significant contributors to the OP of deposited water-soluble particles in the pulmonary region of the lung.
Pollutants from primary sources dominate the oxidative potential of water-soluble PM2.5 in Hong Kong in terms of dithiothreitol (DTT) consumption and hydroxyl radical production.

Abstract:Increasing scientific findings show that the adverse health effects of PM2.5 are related not only to its mass but also PM2.5 sources and chemical compositions. Here, we conducted a comprehensive characterization and source apportionment of oxidative potential (OP) of water-soluble PM2.5 collected in Hong Kong for one year. Two OP indicators, namely dithiothreitol (DTT) consumption and ∙OH formation, were quantified. Six PM2.5 sources, i.e. secondary sulfate, biomass burning, secondary organic aerosol (SOA), vehicle emissions, marine vessels, and a metal-related factor, were apportioned and identified to be DTT active. The four primary sources accounted for 83.5% of DTT activity of water-soluble PM2.5, with the metal-related factor and marine vessels as the leading contributors. However, only three sources, i.e. metal-related factor, vehicle emissions, and SOA, showed ∙OH generation ability, with a predominant contribution of 96.2% from the two primary sources, especially the metal-related factor (84.5%). Based on the source apportionment results, we further evaluate the intrinsic OP of water-soluble PM2.5 from each source. Marine vessels exhibited the highest intrinsic DTT activity; while metal-related factor was most effective in ∙OH generation.
Quantification of synergistic, additive and antagonistic effects of aerosol components on total oxidative potential

Abstract:The interaction-based oxidative potential (OPint) represents the prediction of binary mixture effects distinguishing from linear additivity by including information on binary mixtures among PM components. The objective of this work is to develope a reliable estimate on the possible synergistic or antagonistic possibility of binary PM components and to quantify the combined effect. We firstly assessed the interactions among PM components in generating the OP based on DTT consumption rate and AA depletion. We started with the standard solution sequence (from 0.005 to 10 μM), including quinones (PQ, 1,2-NQ, and 1,4-NQ) and metals (Cu, Mn, and Fe). The interactions between metals were antagonistic interactions in DTT consumption. Cu showed antagonistic interaction with PQ, but additive with 1,2-NQ and 1,4-NQ. Mn interacted synergistically with 1,4-NQ in DTT consumption but antagonistically with PQ (where CPQ < 2.5 μM) and 1,4-NQ (where CPQ < 2 μM). Fe showed synergistic with quinones in investigated concentration range (from 0.01 to 5 μM). Finally, applying a moderate approach, on the basis of interaction magnitude (M = 5), resulted in 1-17% higher environmental risks, compared with the classical calculation method using simple addition. This work highlights a new approach to quantify the interaction effects between metals and quinones in PM components, and apportioning the components' contributions for PM OP.
The Interplay of Brown Carbon (brc) Surrogates and Copper: Implications for the Oxidative Potential of Ambient Particles

Abstract:Atmospheric particulate matter (PM) poses adverse effects on human via producing reactive oxygen species (ROS). Oxidative potential (OP, ability to generate ROS) can be induced by various chemicals in PM, while their interplay remains poorly characterized. Here, we systematically assessed influences of Cu2+ on OP of Brown carbon (BrC) (e.g., imidazoles) using dithiothreitol (DTT) assay. Results showed DTT consumption rate exhibited an initial rise and later decline (0.25 -0.56 mu M/min) along with increase of BrC concentration (0.1 - 2 mu M), while no general trend was observed for center dot OH formation. Although Cu2+ showed either antagonism or synergism with BrC against DTT consumption, Cu2+ displayed antagonism with most BrC against center dot OH generation. Fluorescence quenching experiments provided evidence of complexation between Cu2+ and water-soluble organic compounds (WSOCs, from ambient PM2.5), which was influenced by Cu2+ concentration. Further parallel factor analysis of spectra showed that polycarboxylate-type humic acid-like substances (complexation site number (n): 0.46, complexation equilibrium constant (k): 0.51) and fulvic acid-like compounds (n: 0.42, k: 0.62) were the main components in WSOCs that complexed with Cu2+. Our results suggest the interactions of BrC and copper play a crucial role in PM OP and highlight complexation in evaluation of PM OP.
A Novel High-Throughput Approach to Measure Hydroxyl Radicals Induced by Airborne Particulate Matter

Abstract:Oxidative stress is one of the key mechanisms linking ambient particulate matter (PM) exposure with various adverse health effects. The oxidative potential of PM has been used to characterize the ability of PM induced oxidative stress. Hydroxyl radical (•OH) is the most destructive radical produced by PM. However, there is currently no high-throughput approach which can rapidly measure PM-induced •OH for a large number of samples with an automated system. This study evaluated four existing molecular probes (disodium terephthalate, 3'-p-(aminophenyl)fluorescein, coumarin-3-carboxylic acid, and sodium benzoate) for their applicability to measure •OH induced by PM in a high-throughput cell-free system using fluorescence techniques, based on both our experiments and on an assessment of the physicochemical properties of the probes reported in the literature. Disodium terephthalate (TPT) was the most applicable molecular probe to measure •OH induced by PM, due to its high solubility, high stability of the corresponding fluorescent product (i.e., 2-hydroxyterephthalic acid), high yield compared with the other molecular probes, and stable fluorescence intensity in a wide range of pH environments. TPT was applied in a high-throughput format to measure PM (NIST 1648a)-induced •OH, in phosphate buffered saline. The formed fluorescent product was measured at designated time points up to 2 h. The fluorescent product of TPT had a detection limit of 17.59 nM. The soluble fraction of PM contributed approximately 76.9% of the •OH induced by total PM, and the soluble metal ions of PM contributed 57.4% of the overall •OH formation. This study provides a promising cost-effective high-throughput method to measure •OH induced by PM on a routine basis.
Interactive Enhancements Of Ascorbic Acid And Iron In Hydroxyl Radical Generation In Quinone Redox Cycling

Abstract:Quinones are toxicological substances in inhalable particulate matter (PM). The mechanisms by which quinones cause hazardous effects can be complex. Quinones are highly active redox molecules that can go through a redox cycle with their semiquinone radicals, leading to formation of reactive oxygen species. Electron spin resonance spectra have been reported for semiquinone radicals in PM, indicating the importance of ascorbic acid and iron in quinone redox cycling. However, these findings are insufficient for understanding the toxicity associated with quinone exposure. Herein, we investigated the interactions among anthraquinone (AQ), ascorbic acid, and iron in hydroxyl radical (.OH) generation through the AQ redox cycling process in a physiological buffer. We measured .OH concentration and analyzed the free radical process. Our results showed that AQ ascorbic acid, and iron have synergistic effects on .OH generation in quinone redox cycling; i.e., ascorbyl radical oxidized AQ to semiquinone radical and started the redox cycling, iron accelerated this oxidation and enhanced .OH generation through Fenton reactions, while ascorbic acid and AQ could help iron to release from quartz surface and enhance its bioavailability. Our findings provide direct evidence for the redox cycling hypothesis about airborne particle surface quinone in lung fluid.
Assessment of oxidative potential by hydrophilic and hydrophobic fractions of water-soluble PM2.5 and their mixture effects

Abstract:Transition metals (TMs) (e.g. copper (Cu) and iron (Fe)) and certain organic compounds are known active constituents causing oxidative potential (OP) by inhaled ambient fine particulate matter (PM2.5) in lung fluid. Humic-like substances (HULIS), isolated from atmospheric PM2.5, are largely metal-free and contain mixtures of organics that are capable of complexing TMs. TMs and HULIS co-exist in the water-extractable part of PM2.5. In this work, we used a solid phase extraction procedure to isolate the water-soluble TMs in the hydrophilic fraction (HPI) and HULIS in the hydrophobic fraction (HPO) and carried out this isolation procedure to a set of 32 real-world PM2.5 samples collected in Beijing and Hong Kong, China. We quantified two OP endpoints, namely hydroxyl radical formation (denoted as OP•OH) and ascorbic acid depletion (denoted as OPAA), by the two fractions separately and combined, as well as by the bulk water-soluble aerosols. OP•OH and OPAA were well-correlated in both separate fractions and their combined mixtures or bulk water-soluble aerosols. OP by HPI far exceeded that by HPO. On a per unit PM2.5 mass basis, the Hong Kong samples on average had a higher OPAA and OP•OH than the Beijing samples due to more water-soluble Cu. For HPI, Cu was a dominant OP•OH and OPAA contributor (>80%), although water-soluble Fe was present at a concentration approximately one order of magnitude higher. Suppression effects on OP•OH were observed through comparing the OP of the bulk water-soluble aerosol with that of HPI. Our work reveals the importance of monitoring PM2.5 chemical compositions (especially water-soluble redox active metals). Furthermore, we demonstrate the need to consider metal-organic interactions when evaluating the aggregate OP by PM2.5 from individual components or apportioning OP by PM2.5 to specific chemical components.

Associations between particle physicochemical characteristics and oxidative capacity: An indoor PM10 study in Beijing, China

Abstract:A study was undertaken to determine the use of a plasmid DNA scission assay to evaluate the causal relationships between particle oxidative capacity and physico-chemistry. Field emission scanning electron microscopy (FESEM), image analysis (IA) and inductively coupled plasma-mass spectrometry (ICP-MS) were employed to investigate the physico-chemical characteristics of indoor PM10 (particulate matter with an aerodynamic diameter of 10μm or less) in Beijing, China. Six PM10 samples (indoor smoker's living room; indoor non-smoker's living room and kitchen; and outdoor Beijing city; winter versus summer) were selected to represent typical indoor Beijing PM10 environments that contain high particle mass. The PM10 collected from a kitchen and two smoker's homes had the lowest TD50 (toxic dosage of PM10 causing 50% plasmid DNA damage), being as low as 45μgml−1 (kitchen) and 100μgml−1 (living room), which suggests a high oxidative capacity, with the PM10 generated in kitchens appearing to be the most toxic. The indoor PM10 from the non-smoker's home and outdoor PM10 samples demonstrated high TD50 values and were deemed less bioreactive (i.e. caused limited DNA damage). FESEM observations revealed that four types of particle species were prevalent in Beijing indoor PM10; soot aggregates, minerals, coal fly ash and unknown fine particles. IA showed that higher percentages of soot and unknown fine particles were associated with the lower TD50 values, suggesting that soot and the unknown fine particles may be important components responsible for the observed plasmid DNA damage. The water-soluble trace elements were negatively correlated with the TD50 values, implying that the DNA damage may be attributed to the water-soluble fraction of the PM10. Water-soluble zinc revealed the best relationship with the TD50 values than other analyzed elements, signifying it may play a role in driving the oxidative damage.

Abstract:Epidemiological studies have suggested that exposure to excessive amounts of particle-bound reactive oxygen species (ROS) pollution can lead to a range of health problems. To identify the chemical composition and sources of particulate matter of diameter <2.5 μm (PM2.5)-bound ROS, high-time-resolution PM2.5 samples were collected in Xi'an, China in the periods July 24 to August 5, 2017 (summer), and January 1 to 6, 2017 (winter). Dithiothreitol (DTT) assay was used to measure the oxidative potential of PM2.5-bound ROS. The associations of six types of molecular compositions with ROS were evaluated, and source-specific contributions to ROS were quantified. The results indicated greater activity and diurnal fluctuations in PM2.5-bound ROS in winter than in summer. The mass-normalised DTT consumption rate (DTTm) reached a peak at 8:00 a.m. in summer for polluted and nonpolluted days. In winter, levels of ROS were higher during the day than night, regardless of PM2.5 pollution level. In summer, CHNO and CHOS had the greatest effect on DTTm on polluted days and CHO and CHNOS had the greatest effect on DTTm on nonpolluted days. On polluted days in winter, all molecules (CHO, CHNO, CHOS, CHNOS, CHN and CHNS) investigated in this study had limited effects on DTTm. DTTm on nonpolluted days was mainly affected by CHNO and CHNS. CHNO and CHNOS molecules from secondary reactions favoured ROS production. Results from source apportionment based on positive matrix factorization showed that in summer, vehicle emission was the primary contributor to ROS generation (41.3%), followed by secondary formation (33.7%), fugitive dust (18.6%), and coal combustion (6.3%), whereas in winter, biomass burning and secondary formation were the primary contributors (>58%) to ROS. Our work helps clarify the mechanisms of ROS formation and contributes to theoretical research on PM2.5 toxicity.

Abstract:Oxidation potential (OP) is an emerging index to measure the toxicity of atmospheric particulate matter (PM), but little research data on indoor PM OP are available. This study focuses on the size-resolved particle OP and source apportionment of PM from indoor environments. Different-size PM samples were gathered from March 6, 2022, to April 18, 2022, from offices, canteens, dormitories, and laboratories using Anderson samplers. Particles from these indoor environments were separated into nine size fractions, and the chemical components (including transition metals, ions, and water-soluble organic carbon (WSOC)) were measured. The water-soluble OP (OPws) of the investigated PM was highest in the dormitory (61.02 nmol min−1 μg−1) and lowest in the canteen (25.58 nmol min−1 μg−1), varying by factors of up to 2.38. Additionally, the particulate Mn (r2 = 0.94), Fe (r2 = 0.93), and Zn (r2 = 0.92) concentrations were closely related to OPws. The OPws distribution in indoor PM samples featured a single peak from 0.7 to 3.2 μm. ROS (·OH) showed the same trend (1.1–3.2 μm) as PM OP, which shows that PM3.2 is the main contributor to adverse health effects among all size fractions of PM due to the strengthened oxidative ability of PM3.2 compared to PM > 3.2. The source apportionment results showed that secondary sources (52.3%), construction sources (28%), vehicle emissions (18.5%), and dust sources (1.2%) were the main sources of indoor PM OP. Multiple path particle dosimetry (MPPD) model tests evidenced that PM3.2 control the lung OPws and ROS (accounting for >71.49% and 68.07%, respectively) deposition because of the high deposition efficiency and metal concentration in the alveolar region. The experimental trend of OP was closely related to that of ROS. Overall, since OPws is directly associated with diverse health effects for PM OP, special attention to PM3.2 impact.

Abstract:Exposure to ambient air pollution has an adverse influence on human health. There is increasing evidence that oxidative potential (OP), the capacity of airborne pollutants to oxidize target molecules by generating redox oxidizing species, is a plausible metric for particulate matter (PM) toxicity. Here we describe the commonly used acellular techniques for measuring OP (respiratory tract lining fluid, dithiothreitol, ascorbic acid, and electron paramagnetic resonance assays) and review the PM chemical constituents that have been identified to drive the OP response. We further perform a review of the epidemiologic literature to identify studies that reported an association between exposure to ambient PM and a health outcome in a human population, and in which exposure was measured by both PM mass concentration and OP. Laboratory studies have shown that specific redox-active metals and quinones are able to contribute OP directly. However, interactions among PM species may alter the redox properties of PM components. In ambient PM measurements, all OP assays were found to be correlated with metals (Fe, Cu) and organic species (photochemically aged organics). Across the epidemiological studies reviewed, associations between fine PM (PM2.5) mass and cardio-respiratory outcomes were found to be stronger at elevated OP levels but findings varied across the different OP measurement techniques. Future work should aim to identify specific situations in which PM OP can improve air pollution exposure assessment and/or risk management. This may be particularly useful in countries with low PM2.5 mass concentrations over broad spatial scales where such information may greatly improve the efficiency of risk management activities.

Connecting the Oxidative Potential of Fractionated Particulate Matter with Chromophoric Substances

Abstract:Particulate matter (PM) air pollution is recognized as a major cause of adverse health outcomes, yet much remains unknown about which fractions and compositions are the most harmful. Efforts in this direction can be improved by employing a cell‐free approach, dithiothreitol (DTT) assay, the so‐called oxidative potential (OP). Here, we provide a detailed examination of OP from fractionated PM by a combination extraction method of solvent and solid‐phase extractions (SPE). We estimate that the average OP of water‐soluble matter (WSM; 4.27 nmol/min/m3) is up to 1.2 times higher than methanol‐soluble matter (3.66 nmol/min/m3) in most seasons apart from summer. Of these WSM, OP is distributed almost equally between hydrophobic and hydrophilic fractions. OP of PM is mainly related to transition metals (Cu: 12.1%, Mn: 2.5%); apart from these, more than 80% of the total OP remains unidentified. Identification of this unexplained OP by a novel approach, integrating the chromophore characteristics with parallel factor analysis, has shown that chromophore1 (C1) accounted for 98% of OP, although this factor only made a small contribution to the total fluorescence. Our paper highlights the importance of a holistic approach by evaluating the chemical structure‐activity relationship between optical characteristics and OP. Information presented here draws a connection between light‐absorbing organic aerosols and a possible toxicological mechanism on the basis of their optical properties.

Abstract:An alternative metric to account for particulate matter (PM) composition-based toxicity is the ability of PM-species to generate reactive oxygen species (ROS) and deplete antioxidants, the so-called oxidative potential (OP). Acellular OP assays are the most used worldwide, mainly those based on ascorbic acid (AA) and dithiothreitol (DTT) depletion; OP values are calculated from AA/DTT concentration over time kinetic curves. Since a great variability in OP-DTT and OP-AA values can be found in the literature, the understanding of those factors affecting the kinetic rate of AA and DTT oxidation in the presence of PM-bound species will improve the interpretation of OP values. In this work, a kinetic study of the oxidation rate of AA and DTT driven by species usually found in PM (transition metals and naphthoquinone (NQ)) was carried out. In particular, the influence of the concentration of Cu(II), Fe(II), Fe(III), Mn(II), Mn(III), and 1,4-NQ, and the type of fluid used in the assay (phosphate buffer (PB), phosphate buffer saline (PBS) and artificial lysosomal fluid (ALF)) is analysed and discussed. The reaction orders with respect to the AA/DTT and the active compound, and the kinetic rate constants were also determined. The results show great variability in OP values among the studied species depending on the fluid used; the OP values were mostly higher in PB0.05M, followed by PBS1x and ALF. Moreover, different species concentration-responses for OP-DTT/OP-AA were obtained. These differences were explained by the different reaction orders and kinetic rate constants obtained for each active compound in each fluid.

Abstract:The exposure to ambient fine particulate matter (PM2.5) can induce oxidative stress, contributing to global burden of diseases. The evaluation of the oxidative potential (OP) of PM2.5 is thus critical for the health risk assessment. We collected ambient PM2.5 samples in Hangzhou city, China for four consecutive quarters in the year 2017 and investigated the oxidation property of PM2.5 components by the dithiothreitol (DTT) assay. The annual mean of ambient PM2.5 mass concentrations in 2017 was 63.05 μg m-3 (median: 57.34, range: 6.67-214.33 μg m-3) with the significant seasonal variations ranking as winter > spring > summer > autumn. Secondary inorganic aerosol (SIA) species including SO42-, NO3- and NH4+ totally account for >50% of PM2.5 mass. The annual mean volume-normalized DTT activity (DTTv) showed a relatively high value of 0.62 nmol/min/m3 (median: 0.62, range: 0.11-1.66 nmol/min/m3) and DTTv of four seasons was roughly at the same level, indicating a high annual exposure level of ambient PM2.5. SIA species were correlated well with the corresponding DTTv and showed significant diurnal variations with strong or moderate correlations at day and weak correlations at night, suggesting strong secondary formation in daytime with contribution to the particulate OP. The annual mean mass-normalized DTT activity (DTTm) had a relatively low value of 6.39 pmol/min/μg (median: 5.63, range: 1.99-22.70 pmol/min/μg), indicating low intrinsic oxidative toxicity. The DTTm of four seasons ranked as autumn > winter > spring > summer, indicating seasonal variations of the DTT-active components. The PM2.5 mass concentration is more related to exposure levels than intrinsic properties of components, while OP is determined by the components rather than PM2.5 mass concentration. Our results provide an insight into reactive oxygen species-induced health risk of PM2.5 exposure and decision for subsequent emission control.
The complexation between transition metals and water-soluble organic compounds (WSOC) and its effect on reactive oxygen species (ROS) formation

Abstract:Both Transition Metals (TMs) and Water–Soluble Organic Compounds (WSOC) in atmospheric particles can make a certain contribution to Reactive Oxygen Species (ROS), but the complexation between them and its subsequent effects on ROS formation are still unclear. Three typical TMs (Fe(II), Cu(II), Mn(II)) and two types of WSOCs (SRHA (Suwannee River Humic Acid) and SRFA (Suwannee River Fulvic Acid)) were chosen to study their complexation process and its effects on generation of ROS. The excitation-emission matrix (EEM) was used to obtain the three-dimensional fluorescence spectra of WSOC during complexation process, and the fluorescence regional integration method (FRI) and parallel factor analysis (PARAFAC) were used to quantitatively characterize the changes of different compositions of WSOC during complexation process. The results showed that the complexation between WSOC and Cu or Mn exhibit synergistic effects on ROS generation, while WSOC and Fe has a negative effect even though ROS produced by Fe is the highest among the three TMs. The hydrophobic fraction of WSOC and ROS generation rate decreased during the complexation process, despite ROS concentration increase. PARAFAC analysis results showed that the aromaticity of WSOC is greatly reduced by complexing with Cu, followed by Fe, and the least with Mn. Cu–WSOC exhibited the most significantly synergistic effect on ROS generation, suggesting aromatic and polar substituents (such as phenol and quinone groups) in WSOC dominated the complexation process. All of three TMs tend to form stable complexes with the polar groups in WSOC. Cu might be more capable of combining with the benzene ring by π-π electron transfer than Fe and Mn. The aromatic groups (π-π electron transfer) in WSOC could significantly enhance ROS generation, but polar groups not.
