Abstract:Early infection and peri-implantitis after implant restoration are major reasons for dental implant failure. Implant-associated infections are majorly attributed to biofilm formation. In this study, co-incorporated zinc- (Zn-) and strontium- (Sr-) nanorod coating on sandblasted and acid-etched (SLA) titanium (SLA-Zn/Sr) was fabricated by hydrothermal synthesis. It was aimed at promoting osteogenesis while inhibiting biofilm formation. The nanorod-like particles (phi 30-50 nm) were found to be evenly formed on SLA-Zn/Sr (Zn: 1.49 +/- 0.16 wt%; Sr: 21.69 +/- 2.74 wt%) that was composed of well-crystallized ZnTiO3 and SrTiO3 phases. With a sufficient interface bonding strength (42.00 +/- 3.00 MPa), SLA-Zn/Sr enhanced the corrosion resistance property of titanium. Besides, SLA-Zn/Sr promoted the cellular initial adhesion, proliferation and osteogenic differentiation of rBMSCs in vitro while inhibiting the adhesion of Staphylococcus aureus and Porphyromonas gingivalis . In addition, through down-regulating icaA gene expression, this novel surface reduced the secretion of polysaccharide intercellular adhesion (reduced by 87.9% compared to SLActive) to suppress the S. aureus biofilm formation. We, therefore, propose a new chemical modification on titanium for multifunctional implant material development. Due to the Zn/Sr co-doping in coating, material properties, early osteogenic effect and antibacterial ability of titanium can be simultaneously enhanced, which has the potential to be applied in dental implantation in the future.

Abstract:Background For dental implants, early infection and peri-implantitis after implant restoration are major reasons for implant failure. Plaque biofilm formation is the initiator for implant-related infection. Therefore, it is urgent to develop a new antibacterial implant materials or treatment methods. Aim/Hypothesis In this study, Sr- and Zn- incorporated micro nano-structured titanium surface with nanorods (MNT-Sr Zn) was fabricated by a two-step hydrothermal process, aiming to improve osseointegration and reduce implant failure caused by implant-related infection and broaden new ideas for the future research Material and Methods The Mod Ti surfaces were used as a control. The characterization of different surfaces was detected by a field emission scanning electron microscope (FE-SEM) equipped with energy dispersive spectra (EDS) detector, x-ray diffractometer (XRD) and inductively coupled plasma mass spectroscopy (ICP-MS). Meanwhile, Staphylococcus aureus (S. aureus) was selected to evaluate antibacterial properties through Live Dead Staining and watching the adhesive bacterial morphology under FE-SEM. What's more, rat bone marrow-derived mesenchymal stem cells (rBMSCs) were cultured on the surfaces to detect their early osteogenic properties, like the alkaline phosphatase(ALP) activity and the expressions of osteogenesis related genes. Results The results confirmed that nanorod-like particles with a diameter of about 30-50 nm of MNT-Sr Zn composed of well-crystallized ZnO and SrTiO3 phases. In the EDS spectra, the elements of Zn and Sr were evenly distributed, and approximately 1.49 ± 0.16 wt% and 21.69 ± 2.74 wt% respectively. Besides, the release of Sr2+ and Zn2+ was a controlled and long-term process. More importantly, Zn-containing nanorod-like particles and the morphology could work together, slightly decreasing adhesion of S. aureus but increasing the proportion of dead cells, thus inhibiting subsequent biofilm formation, compared with the controlled group. In addition to this, MNT-Sr Zn could enhance the osteogenic differentiation via the upregulation of the transcription of osteogenic related genes and the expression of ALP in vitro. Conclusion and Clinical Implications This research provides a new surface modification method on titanium substrate for multi-functional implant material development. Due to the co-doping of Sr and Zn in the coating, biomechanical stability, antibacterial capability and early osteogenic inductive effect are enhanced, which has the potential to be applied in dental implantation in the future.
Surface modification of the laser powder bed-fused Ti-Zr-Nb scaffolds by dynamic chemical etching and Ag nanoparticles decoration

Abstract:Metallic lattice scaffolds are designed to mimic the architecture and mechanical properties of bone tissue and their surface compatibility is of primary importance. This study presents a novel surface modification protocol for metallic lattice scaffolds printed from a superelastic Ti-Zr-Nb alloy. This protocol consists of dynamic chemical etching (DCE) followed by silver nanoparticles (AgNP) decoration. DCE, using an 1HF + 3HNO3 + 12H2O23% based solution, was used to remove partially-fused particles from the surfaces of different as-built lattice structures (rhombic dodecahedron, sheet gyroid, and Voronoi polyhedra). Subsequently, an antibacterial coating was synthesized on the surface of the scaffolds by a controlled (20 min at a fixed volume flowrate of 500 mL/min) pumping of the functionalization solutions (NaBH4 (2 mg/mL) and AgNO3 (1 mg/mL)) through the porous structures. Following these treatments, the scaffolds' surfaces were found to be densely populated with Ag nanoparticles and their agglomerates, and manifested an excellent antibacterial effect (Ag ion release rate of 4-8 ppm) suppressing the growth of both E. coli and B. subtilis bacteria up to 99 %. The scaffold extracts showed no cytotoxicity and did not affect cell proliferation, indicating their safety for subsequent use as implants. A cytocompatibility assessment using MG-63 spheroids demonstrated good attachment, spreading, and active migration of cells on the scaffold surface (over 96 % of living cells), confirming their biotolerance. These findings suggest the promise of this surface modification approach for developing superelastic Ti-Zr-Nb scaffolds with superior antibacterial properties and biocompatibility, making them highly suitable for bone implant applications.

Abstract:Introduction Titanium (Ti) and its alloys are widely utilized in endosseous implants. However, their clinical efficacy is marred by complications arising from bacterial infections owing to their inadequate antibacterial properties. Consequently, enhancing the antibacterial attributes of implant surfaces stands as a pivotal objective in the realm of implantable materials research. Methods In this study, we employed sequential anodization and plasma immersion ion implantation (PIII) technology to fabricate a silver-embedded sparsely titania nanotube array (SNT) on the near-β titanium alloy Ti-5Zr-3Sn-5Mo-15Nb (TLM) implants. The surface characteristics, antimicrobial properties, biocompatibility, and osteogenic activity of the silver-nanomodified SNT implant (SNT Ag) surface, alongside peri-implant inflammatory responses, were meticulously assessed through a combination of in vitro and in vivo analyses. Results Compared with polished TLM and SNT, the silver-embedded SNT (SNT Ag) surface retained the basic shape of nanotubes and stably released Ag+ at the ppm level for a long time, which demonstrated an effective inhibition and bactericidal activity against Staphylococcus aureus (SA) while maintaining ideal cytocompatibility. Additionally, the subtle modifications in nanotubular topography induced by silver implantation endowed SNT Ag with enhanced osteogenic activity and mitigated inflammatory capsulation in soft tissue peri-implants in a rat model. Conclusion Incorporating a silver-embedded SNT array onto the implant surface demonstrated robust antibacterial properties, impeccable cytocompatibility, exceptional osteogenic activity, and the potential to prevent inflammatory encapsulation around the implant site. The Silver-PIII modification strategy emerges as a highly promising approach for surface applications in endosseous implants and trans-gingival implant abutments.

Laser-Assisted Nanotexturing and Silver Immobilization on Titanium Implant Surfaces to Enhance Bone Cell Mineralization and Antimicrobial Properties

Abstract:Despite the great advancement and wide use of titanium (Ti) and Ti-based alloys in different orthopedic implants, device-related infections remain the major complication in modern orthopedic and trauma surgery. Most of these infections are often caused by both poor antibacterial and osteoinductive properties of the implant surface. Here, we have demonstrated a facile two-step laser nanotexturing and immobilization of silver onto the titanium implants to improve both cellular integration and antibacterial properties of Ti surfaces. The required threshold laser processing power for effective nanotexturing and osseointegration was systematically determined by the level of osteoblast cells mineralized on the laser nanotextured Ti (LN–Ti) surfaces using a neodymium-doped yttrium aluminum garnet laser (Nd:YAG, wavelength of 1.06 μm). Laser processing powers above 24 W resulted in the formation of hierarchical nanoporous structures (average pore 190 nm) on the Ti surface with a 2.5-fold increase in osseointegration as compared to the pristine Ti surface. Immobilization of silver nanoparticles onto the LN–Ti surface was conducted by dip coating in an aqueous silver ionic solution and subsequently converted to silver nanoparticles (AgNPs) by using a low power laser-assisted photocatalytic reduction process. Structural and surface morphology analysis via XRD and SEM revealed a uniform distribution of Ag and the formation of an AgTi-alloy interface on the Ti surface. The antibacterial efficacy of the LN–Ti with laser immobilized silver (LN–Ti/LI–Ag) was tested against both Gram-positive (Staphylococcus aureus) and Gram-negative (Escherichia coli) bacteria. The LN–Ti/LI–Ag surface was observed to have efficient and stable antimicrobial properties for over 6 days. In addition, it was found that the LN–Ti/LI–Ag maintained a cytocompatibility and bone cell mineralization property similar to the LN–Ti surface. Alcohol and Female Puberty The Role of Intraovarian Systems

Abstract:Alcohol consumption during early adolescence may suppress the secretion of specific female reproductive hormones, thereby delaying puberty and adversely affecting the maturation of the reproductive system. These effects occur through several mechanisms, including altered production and secretion by the ovaries of estradiol, a key steroid hormone involved in the timing and regulation of female reproductive events. Alcohol can affect estradiol production by interfering with the normal function of regulatory hormones produced by the brain and the pituitary gland. Recent research has demonstrated additional potential mechanisms for alcohol’s effects on female reproductive capability, including interference with specific regulatory systems located entirely within the ovary. Such “intraovarian” systems include the insulin-like growth factor-1 (IGF-1) and nitric oxide (NO) systems. Alcohol can dampen the stimulatory effects of the ovarian IGF-1 system and can increase the inhibitory effects of the ovarian NO system. These effects combine to decrease estradiol secretion. Thus, alcohol impairs ovarian function not only by interfering with hormonal communication between the brain, pituitary gland, and ovaries but also by directly altering the function of regulatory systems within the ovaries themselves. These results provide further evidence of the risks of underage drinking and the importance of its prevention.
Modern methods of surface modification for new-generation titanium alloys

Abstract:The constantly growing need for the use of implants in osteotomy is mainly due to the aging population and the need for long-term use of this type of biomaterials. Improving implant materials requires the selection of appropriate functional properties. Currently used titanium (Ti) alloys, such as Ti6Al4V and Ti6Al7Nb, are being replaced by materials with better biocompatibility, such as vanadium (V) or niobium (Nb), allowing for creation of the so-called new generation alloys. These new alloys, with the incorporation of zirconium (Zr), iron, and tantalum, possess Young's modulus close to that of a bone, which further improves the improves the biomaterial's. biocompatibility. This article describes the atomic layer deposition (ALD) method and its possible applications in the new generation of titanium alloys for biomedical applications. Also, the exemplary results of tin oxide (SnO2) thin coatings deposited by ALD and physical vapor deposition (PVD) methods are presented. This study aimed to evaluate the physicochemical properties of a Ti13Nb13Zr alloy used for elements in the skeletal system. As the temperature and the number of cycles vary, the results demonstrate that the surface area of the samples changes. The uncoated Ti13Nb13Zr alloy exhibits hydrophilic properties. However, all coated specimens improve in this respect and provide improved clinical results. after the applied modification, the samples have a smaller contact angle, but still remain in the range of 0-90°, which makes it possible to conclude that their nature remains hydrophilic. Coating the specimens decreased the mineralization risk of postoperative complications. As a result, the biomaterials demonstrated improved effectiveness, decreased complication indicators, and improved patient well-being.
Antibacterial Surfaces Bearing Silver, Copper and Zinc Nanoparticles on Additively Manufactured Titanium Implants

Abstract:BACKGROUND.Implant-associated infection (IAI) is a rising complication in bone-related medical treatments using metal implants. Preventive measures, such as implant surfaces with integrated antibacterial properties, are hence required. The emergence of antibiotic-resistant pathogens has led to a growing interest in inorganic nanoparticles as antibacterial agents. In addition, implants should simultaneously stimulate bone tissue regeneration and integration to improve implant longevity. Additive manufacturing (AM) of orthopedic implants has attracted interest as improved bone tissue regeneration and integration was demonstrated by AM porous implants. AM porous implants, however, are at enhanced risks of IAIs; these porous implants require integrated antibacterial properties. This study focused on a systematic comparison of the antibacterial properties of AM porous Ti6Al4V implant surfaces with silver nanoparticles (Ag NPs), copper nanoparticles (Cu NPs) and/or zinc nanoparticles (Zn NPs) against methicillin resistant Staphylococcus aureus (MRSA). METHODS.Medical grade porous Ti6Al4V implants were additively manufactured using selective laser melting (SLM). Subsequently, the SLM Ti6Al4V implants were biofunctionalized by plasma electrolytic oxidation (PEO) using an electrolyte that consisted of Ca/P-based species and ratios of Ag NPs, Cu NPs and/or Zn NPs. Implants with 0, 25, 75 and 100% of Ag NPs, Cu NPs and/or Zn NPs were synthesized. After biofunctionalization, the surface morphology, structure and chemistry of the implants were investigated using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS). The antibacterial properties of the implants against MRSA were studied in vitro as well as ex vivo. The leachable antibacterial activity was studied in an agar diffusion assay, the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of Ag+, Cu2+ and Zn2+ ions was determined by a micro-dilution assay and the bactericidal activity of the implants was quantified by a colony-forming unit (CFU) count in vitro against surface-adherent and non-adherent MRSA. Furthermore, ex vivo antibacterial properties were determined using a femoral murine infection model mimicking the in vivo environment. RESULTS.Biofunctionalization of the porous SLM Ti6Al4V implants by PEO resulted in a uniform and homogeneous micro-/nano-porous oxide layer that covered the entire surface of the implants. SEM and EDS analysis demonstrated the presence of Ag NPs, Cu NPs and Zn NPs in the TiO2 surface layer. The agar diffusion assay demonstrated a strong leachable antibacterial activity for the implants with Ag NPs and Cu NPs or Zn NPs, while no leachable antibacterial activity was observed for implants without Ag NPs. Antibacterial testing showed that Ag+, Cu2+ and Zn2+ ions, as well the combinations of the ions, were growth inhibitory and bactericidal to MRSA. In addition, implants with ≥ 50% of Ag NPs and combinations of ≥ 75% of Ag NPs and Cu NPs or Zn NPs, fully eradicated surface-adherent and non-adherent MRSA within 24 h and prevented biofilm formation up to 24 h. Moreover, strong ex vivo bactericidal properties against MRSA were demonstrated for these implants within 24 h. CONCLUSION. Biofunctionalized surfaces with Ag NPs and Cu NPs or Zn NPs on porous SLM Ti6Al4V implants showed strong in vitro and ex vivo antibacterial properties against MRSA. The implants with 50% of Ag NPs and combinations of 75% of Ag NPs and 25% of Cu NPs or Zn NPs are promising for further development, to prevent IAIs and improve the longevity of the implants in clinical applications.

Abstract:One of the most common causes of implant failure is peri-implantitis, which is caused by bacterial biofilm formation on the surfaces of dental implants. Modification of the surface nanotopography has been suggested to affect bacterial adherence to implants. Silver nanoparticles are also known for their antibacterial properties. In this study, titanium alloy implants were surface modified following silver plating, anodisation and sintering techniques to create a combination of silver, titanium dioxide and hydroxyapatite (HA) nanocoatings. Their antibacterial performance was quantitatively assessed by measuring the growth of Streptococcus sanguinis, proportion of live/dead cells and lactate production by the microbes over 24 h. Application of a dual layered silver-HA nanocoating to the surface of implants successfully inhibited bacterial growth in the surrounding media (100% mortality), whereas the formation of bacterial biofilm on the implant surfaces was reduced by 97.5%. Uncoated controls and titanium dioxide nanocoatings showed no antibacterial effect. Both silver and HA nanocoatings were found to be very stable in biological fluids with material loss, as a result of dissolution, to be less than 0.07% for the silver nanocoatings after 24 h in a modified Krebs-Ringer bicarbonate buffer. No dissolution was detected for the HA nanocoatings. Thus, application of a dual layered silver-HA nanocoating to titanium alloy implants creates a surface with antibiofilm properties without compromising the HA biocompatibility required for successful osseointegration and accelerated bone healing.

Silver Nanoparticles Enhance the Antibacterial Effect of Antibiotic-Loaded Bone Cement

Abstract:Purpose The goal of this study was to determine the antibacterial activity of bone cement in polymethyl methacrylate (PMMA) structures with varying amounts of silver nanoparticles (AgNPs) included. Additionally, we aimed to evaluate whether AgNPs affect the biomechanical properties of PMMA cement in our study. Materials and methods Between April 2020 and June 2020, we conducted a series of experiments to demonstrate the antibacterial characteristics by adding silver nanoparticles to PMMA bone cement. PMMA bone cement (Cemex, Tecres Company, Verona, Italy) was used as the base material. Seven different samples were prepared in order to evaluate the amount and presence of AgNPs. Cement samples containing AgNPs and teicoplanin at different concentrations and empty cement (control, without teicoplanin and AgNPs) were placed on Petri plates. The agar diffusion method was used to determine the antibacterial effect (Kirby-Bauer). Results Kirby-Bauer assays demonstrated that AgNPs added to bone cement increased the antimicrobial activity compared to antibiotic-free or only teicoplanin-loaded cement. It was observed that increasing the AgNPs ratio further increased the antimicrobial activity. Conclusion AgNPs in various combinations enhance antimicrobial activity synergistically while maintaining the mechanical strength of bone cement. Increasing the amount of AgNPs results in a significant increase in antimicrobial activity.
Antibacterial activity and biocompatibility of titanium nickelide augments with the addition of silver nanoparticles for bone grafting: an experimental study

Abstract:BACKGROUND: The relevance of this study was supported by the increasing number of infectious complications of bone augmentation in children and adults. Currently, porous titanium nickelide alloys are among the most preferred materials used in bone plasty. Despite the observable advantages of porous nickelide titanium alloys in terms of biochemical and biomechanical compatibility with the body, research on the antibacterial activity of alloys is ongoing to counter the development of infections at the implant–biological tissue border. AIM: To perform an experimental study of the biocompatible antibacterial surface in porous titanium nickelide alloys with the addition of silver nanoparticles. MATERIALS AND METHODS: Titanium nickelide alloys with 62% porosity were obtained using the self-propagating high-temperature synthesis method from nickel, titanium, and nanosilver powders at concentrations of 0.2 at.% Ag, 0.5 at.% Ag, and 1.0 at.% Ag, respectively. The experiment was conducted on nine sexually mature female white laboratory rats. They were divided into three groups, with three rats each. All animals were implanted with titanium nickelide along with porous granules of silver additives. The first group was the control, the second received 0.2 at.% silver, and the third received 0.5% silver. The standard method of incubating Staphylococcus epidermidis in liquid broth in the presence of the studied images was used to determine bactericidal activity, followed by seeding on solid media and counting colonies. RESULTS: The antibacterial effect of the samples on S. epidermidis gradually increased with increasing silver concentration. The significance of the differences between the experiment and control was confirmed by Student’s criterion p 0.005, whereas the sample without silver nanoparticles and the control do not differ significantly. Thus, these alloys may have bioactive properties because they contain silver nanoparticles. An alloy with a silver concentration of 0.5 at.% Ag showed the best antibacterial activity to S. epidermidis. In the clinical evaluation of the results of the experimental study, purulent inflammatory complications were not observed in all animals at all times. On day 75, the animals underwent computed tomography, which showed good occupancy of the bone defect and absence of a dystrophic effect on the area where the bone and soft tissue are in contact with the material. CONCLUSIONS: If the concentration of silver nanoparticles is increased up to 0.5 at%, the antibacterial activity and cytocompatibility of the implant also increase. Clinical experimental evaluation in all groups of animals showed that osteointegration of alloys with 0.5 at.% Ag begins immediately after implantation and is completed 2 weeks earlier than that in the remaining groups.

Abstract:Multiple surface treatment methods for titanium alloy prostheses, widely used in orthopedics, are available; however, these can affect bone integration and regeneration efficiency. In this study, through cell and animal experiments, we devised seven bone implant categories of Ti6Al4V based on surface preparation and post-processing technology (polishing, grit-blasting, fine titanium spraying, coarse titanium spraying, electron beam melting [EBM] printing, selective laser melting [SLM] printing, and post-processed SLM printing) and imaged each microscopic surface structure with a scanning electron microscope (SEM). Mechanical testing revealed excessive post-processing damaged the mechanical properties of the implants. In vitro, human bone marrow mesenchymal stem cells (hBMSCs) were cultured with implants, and the morphology of the cells adhering to the implant surface was observed using SEM and confocal laser scanning microscopy. Cell Counting Kit-8 (CCK-8) semi-quantitatively determined cell activity, indirectly reflecting the proliferation of hBMSCs. Alizarin red and alkaline phosphatase experiments assessed osteogenic differentiation. In vivo, experiments utilized the New Zealand rabbit femoral condyle bone defect model to assess bone regeneration and integration using micro-computed tomography, Van Giesen staining, and Masson staining. We found that 3D-printed implants with regular pore structures were more conducive to hBMSC osteogenic differentiation, while the presence of metal powder on NPT-SLM-printed implants hindered such differentiation. The post-treatment SLM scaffold surface may have some residual semi-melted powder; however, these powder residues have no significant effect on cell activity and differentiation. Surface treatment (grit-blasting and titanium spraying) of planar structures can enhance hBMSC adhesion but does not necessarily promote their differentiation. The framework structure of 3D printing may affect the osteogenic differentiation of hBMSCs, and for SLM-printed implants, excessive pursuit of a "powderless" state will damage the mechanical properties of the implant.

Abstract:Microbial colonization and biofilm formation on implanted devices represent an important complication in orthopaedic and dental surgery and may result in implant failure. Controlled release of antibacterial agents directly at the implant site may represent an effective approach to treat these chronic complications. Resistance to conventional antibiotics by pathogenic bacteria has emerged in recent years as a major problem of public health. In order to overcome this problem, non-conventional antimicrobial agents have been under investigation. In this study, polyacrylate-based hydrogel thin coatings have been electrosynthesised on titanium substrates starting from poly(ethylene glycol diacrylate)–co–acrylic acid. Silver nanoparticles (AgNPs) with a narrow size distribution have been synthesized using a “green” procedure and immobilized on Ti implant surfaces exploiting hydrogel coatings’ swelling capabilities. The coatings have been characterized by XPS and SEM/EDX, while their silver release performances have been monitored by ICP–MS. The antibacterial activity of these AgNP-modified hydrogel coatings was tested evaluating in vitro inhibition growth of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli, among the most common pathogens in orthopaedic infections. Moreover, a preliminary investigation of the biocompatibility of silver-loaded coatings versus MG63 human osteoblast-like cells has been performed. An important point of strength of this paper, in fact, is the concern about the effect of silver species on the surrounding cell system in implanted medical devices. Abstract:Micro-arc oxidation (MAO) is a versatile surface-modification method that promotes higher wear and corrosion resistance, osseointegration, and biological activity to titanium alloys' surfaces. This study aimed to modify the surface of a recently developed metastable β Ti alloy, which exhibits more favorable mechanical properties for implant applications compared to some commercial Ti alloys, by incorporating Ag into the coatings to introduce a bactericidal function to the surface. The Ti-30Nb-5Mo alloy, with lower elastic modulus, was treated by the MAO method using electrolyte solutions containing calcium acetate, magnesium acetate, β-glycerol phosphate, and varied concentrations of silver nitrate (1.5 mM, 2.5 mM, and 3.5 mM). With an increase in the concentration of silver ions in the electrolyte, the galvanostatic period during the MAO process decreased from 1.7 s to 0.5 s. The Ca/P ratio increased from 0.72 up to 1.36. X-ray diffraction showed that the MAO coatings were formed by rutile and anatase TiO2 main phases and calcium phosphates. X-ray photoelectron spectroscopy analysis detected the presence of amorphous Nb2O5, CaCO3, and MgCO3, and metallic and oxide forms of Ag. The increase in Ag in the electrolyte decreased the coating thickness (from 14.2 μm down to 10.0 μm), increased the contact angle (from 37.6° up to 57.4°), and slightly increased roughness (from 0.64 μm up to 0.79 μm). The maximum inhibition of Enterococcus faecalis, Pseudomonas aeruginosa, and Candida albicans strains growth was of 43%, 43%, and 61%, respectively. The Ag did not negatively affect the differentiation of adipose-tissue-derived mesenchymal stem cells. Therefore, the treatment of the surface of the innovative Ti-30Nb-5Mo alloy by the MAO method was effective in producing a noncytotoxic porous coating with bactericidal properties and improved osseointegration capabilities.

Abstract:Prosthetic joint infection is one of the most serious complications that can lead to failure of a total joint replacement. Recently, the rise of multidrug resistant bacteria has substantially reduced the efficacy of antibiotics that are typically incorporated into acrylic bone cement. Silver nanoparticles (AgNPs) are an attractive alternative to traditional antibiotics resulting from their broad-spectrum antimicrobial activity and low bacterial resistance. The purpose of this study, therefore, was to incorporate metallic silver nanoparticles into acrylic bone cement and quantify the effects on the cement's mechanical, material and antimicrobial properties. AgNPs at three loading ratios (0.25, 0.5, and 1.0% wt/wt) were incorporated into a commercial bone cement using a probe sonication technique. The resulting cements demonstrated mechanical and material properties that were not substantially different from the standard cement. Testing against Staphylococcus aureus and Staphylococcus epidermidis using Kirby-Bauer and time-kill assays demonstrated no antimicrobial activity against planktonic bacteria. In contrast, cements modified with AgNPs significantly reduced biofilm formation on the surface of the cement. These results indicate that AgNP-loaded cement is of high potential for use in primary arthroplasty where prevention of bacterial surface colonization is vital.

Abstract:OBJECTIVE To determine associations between in vitro minimum inhibitory concentrations (MICs) of tilmicosin against Mannheimia haemolytica and Pasteurella multocida and in vivo tilmicosin treatment outcome among calves with clinical signs of bovine respiratory disease (BRD). DESIGN Observational, retrospective, cohort study. ANIMALS 976 feeder calves with clinical signs of BRD enrolled in 16 randomized clinical trials. PROCEDURES Records of clinical trials from October 26, 1996, to November 15, 2004, were searched to identify calves with BRD from which a single isolate of M haemolytica or P multocida was identified via culture of deep nasal swab samples prior to treatment with tilmicosin (10 mg/kg [4.5 mg/lb], SC) and for which MICs of tilmicosin against the isolate were determined. The MICs of tilmicosin against recovered isolates and response to tilmicosin treatment were evaluated. RESULTS Tilmicosin resistance among M haemolytica and P multocida isolates was uncommon (6/745 [0.8%] and 16/231 [6.9%], respectively). Treatment outcome, defined as success or failure after tilmicosin treatment, did not vary with the MIC of tilmicosin against recovered isolates. The proportion of treatment failures attributed to M haemolytica isolates categorized as resistant (MIC of tilmicosin, ≥ 32 μg/mL) or not susceptible (MIC of tilmicosin, ≥ 16 μg/mL), was 0.2% and 0.5%, respectively. CONCLUSIONS AND CLINICAL RELEVANCE Recovery of tilmicosin-resistant M haemolytica or P multocida isolates was rare, and no association was detected between MIC of tilmicosin and treatment response.

Abstract:Metallic materials for long-term load-bearing implants still do not provide high antimicrobial activity while maintaining strong compatibility with bone cells. This study aimed to modify the surface of Ti13Nb13Zr alloy by electrophoretic deposition of a chitosan coating with a covalently attached Arg-Gly-Asp (RGD) peptide. The suspensions for coating deposition were prepared in two different ways either using hydroxyacetic acid or a carbon dioxide saturation process. The coatings were deposited using a voltage of 10 V for 1 min. The prepared coatings were examined using SEM, EDS, FTIR, and XPS techniques. In addition, the wettability of these surfaces, corrosion resistance, adhesion of the coatings to the metallic substrate, and their antimicrobial activity (E. coli, S. aureus) and cytocompatibility properties using the MTT and LDH assays were studied. The coatings produced tightly covered the metallic substrate. Spectroscopic studies confirmed that the peptide did not detach from the chitosan chain during electrophoretic deposition. All tested samples showed high corrosion resistance (corrosion current density measured in nA/cm2 ). The deposited coatings contributed to a significant increase in the antimicrobial activity of the samples against Gram-positive and Gram-negative bacteria (reduction in bacterial counts from 99% to, for CS-RGD-Acid and the S. aureus strain, total killing capacity). MTT and LDH results showed high compatibility with bone cells of the modified surfaces compared to the bare substrate (survival rates above 75% under indirect contact conditions and above 100% under direct contact conditions). However, the adhesion of the coatings was considered weak.
Biocompatibility analysis of titanium bone wedges coated by antibacterial ceramic-polymer layer

Abstract:This paper presents the surface treatment results of titanium, veterinary bone wedges. The functional coating is composed of a porous oxide layer (formed by a plasma electrolytic oxidation process) and a polymer poly(sebacic anhydride) (PSBA) layer loaded with amoxicillin (formed by dip coatings). The coatings were porous and composed of Ca (4.16%-6.54%) and P (7.64%-9.89% determined by scanning electron microscopy with EDX) in the upper part of the implant. The titanium bone wedges were hydrophilic (54° water contact angle) and rough (surface area (Sa):1.16 μm) The surface tension determined using diiodomethane was 68.6 ± 2.0° for the anodized implant and was similar for hybrid coatings: 60.7 ± 2.2°. 12.87 ± 0.91 μg/mL of amoxicillin was released from the implants during the first 30 min after immersion in the phosphate-buffered saline (PBS) solution. This concentration was enough to inhibit the Staphylococcus aureus ATCC 25923, and Staphylococcus epidermidis ATCC12228 growth. The obtained inhibition zones were between 27.3 ± 2.1 mm–30.7 ± 0.6 mm when implant extract after 1 h or 4 h immersion in PBS was collected. Various implant biocompatibility analyses were performed under in vivo conditions, including pyrogen test (3 rabbits), intracutaneous reactivity (3 rabbits, 5 places by side), acute systemic toxicity (20 house mice), and local lymph node assay (LLNA) (20 house mice). The extracts from implants were collected in polar and non-polar solutions, and the tests were conducted according to ISO 10993 standards. The results from the in vivo tests showed, that the implant's extracts are not toxic (mass body change below 5%), not sensitizing (SI < 1.6), and do not show the pyrogen effect (changes in the temperature 0.15oC). The biocompatibility tests were performed in a certificated laboratory with a good laboratory practice certificate after all the necessary permissions.
