Abstract:MgGa alloys are considered highly potential biodegradable materials, owing to its good mechanical properties and appropriate corrosion resistance. However, it is still far from application due to the lack of biological evaluation. In the present study, biocompatibility, osteogenesis and antibacterial activity of extruded Mg-xGa (x = 1 and 5 wt%) alloys were investigated by in vitro cell culture experiments and in vivo implantation. The cell adhesion and proliferation of osteoblast precursor cells (MC3T3-E1) showed the excellent cytocompatibility of Mg-1Ga and poor cytocompatibility of Mg-5Ga. The osteogenic activity was evaluated and revealed that Ga3+ in the Mg-1Ga extract had the ability to enhance osteogenic differentiation through the facilitation of its early stages. In vivo studies in a rat femoral condyle model revealed that both Mg-1Ga and Mg-5Ga significantly promoted new bone formation without causing any adverse effects. Mg-5Ga exhibited a much higher corrosion rate in vivo than Mg-1Ga. Its osteogenic activity was better due to the rapid release of Mg2+ and Ga3+, but this caused premature structural integrity loss. Mg-1Ga and Mg-5Ga released Ga3+ to inhibit E. coli and S. aureus, with antibacterial rate increasing with Ga content. Our studies demonstrate that Mg-Ga alloys have the potential to be used as osteogenic and antibacterial implant materials. STATEMENT OF SIGNIFICANCE: This study evaluates the biocompatibility, osteogenesis, and antibacterial activity of Mg-Ga alloys, which are promising biodegradable materials for medical applications. The study finds that Mg-1Ga exhibits excellent cytocompatibility and promotes osteogenic differentiation, facilitating the early stages of osteoblast precursor cell development. In vivo studies in a rat femoral condyle model reveal that Mg-1Ga and Mg-5Ga significantly promote new bone formation without causing any adverse effects. The antibacterial activity of both alloys is evaluated against E. coli and S. aureus, with the inhibition rate increasing with Ga content. These findings suggest that Mg-Ga alloys have the potential to serve as osteogenic and antibacterial implant materials, providing significant insights into the development of novel biomedical implants.
Addition of Zn to the Ternary Mg-Ca-Sr Alloys Significantly Improves Their Antibacterial Property.

Abstract:Most of the magnesium (Mg) alloys possess excellent biocompatibility, mechanical property and biodegradability in orthopedic applications. However, these alloys may suffer from bacterial infections due to their insufficient antibacterial capability. In order to reduce the post-surgical infections, a series of biocompatible Mg-1Ca-0.5Sr-xZn (x=0, 2, 4, 6) alloys were fabricated with the addition of antibacterial Zn with variable content and evaluated in terms of their biocompatibility and antibacterial property. The in vitro corrosion study showed that Mg-1Ca-0.5Sr-6Zn alloys exhibited a higher hydrogen evolution volume after 100 h immersion and resulted in a higher pH value of the immersion solution. Our work indicated that Zn-containing Mg alloys exhibited good biocompatibility with high cell viability. The antibacterial studies reveal that the number of bacteria adhered on all of these Mg alloy samples diminished remarkably compared to the Ti-6Al-4V control group. We also found that the proliferation of the bacteria was inhibited by these Mg alloys extracts. Among the prepared alloys, Mg-1Ca-0.5Sr-6Zn alloy not only exhibited a strong antibacterial effect, but also promoted the proliferation of MC3T3-E1 osteoblasts, suggesting that it is a promising alloy with both good antibacterial property and good biocompatibility for use as an orthopedic implant.
Improving in vitro and in vivo corrosion resistance and biocompatibility of Mg-1Zn-1Sn alloys by microalloying with Sr

Abstract:Magnesium (Mg) and its alloys have attracted attention as potential biodegradable materials in orthopedics due to their mechanical and physical properties, which are compatible with those of human bone. However, the effect of the mismatch between the rapid material degradation and fracture healing caused by the adverse effect of hydrogen (H2), which is generated during degradation, on surrounding bone tissue has severely restricted the application of Mg and its alloys. Thus, the development of new Mg alloys to achieve ideal degradation rates, H2 evolution and mechanical properties is necessary. Herein, a novel Mg-1Zn-1Sn-xSr (x = 0, 0.2, 0.4, and 0.6 wt%) quaternary alloy was developed, and the microstructure, mechanical properties, corrosion behavior and biocompatibility in vitro/vivo were investigated. The results demonstrated that a minor amount of strontium (Sr) (0.2 wt %) enhanced the corrosion resistance and mechanical properties of Mg-1Zn-1Sn alloy through grain refinement and second phase strengthening. Simultaneously, due to the high hydrogen overpotential of tin (Sn), the H2 release of the alloys was significantly reduced. Furthermore, Sr-containing Mg-1Zn-1Sn-based alloys significantly enhanced the viability, adhesion and spreading of MC3T3-E1 cells in vitro due to their unique biological activity and the ability to spontaneously form a network structure layer with micro/nanotopography. A low corrosion rate and improved biocompatibility were also maintained in a rat subcutaneous implantation model. However, excessive Sr (>0.2 wt %) led to a microgalvanic reaction and accelerated corrosion and H2 evolution. Considering the corrosion resistance, H2 evolution, mechanical properties and biocompatibility in vitro and in vivo, Mg-1Zn-1Sn-0.2Sr alloy has tremendous potential for clinical applications.
Evaluation on the corrosion resistance, antibacterial property and osteogenic activity of biodegradable Mg-Ca and Mg-Ca-Zn-Ag alloys

Abstract:The rapid degradation of magnesium (Mg)-based implants in physiological environment limits its clinical applications, and alloying treatment is an effective way to regulate the degradation rate of Mg-based materials. In the present study, three Mg alloys, including Mg-0.8Ca (denoted as ZQ), Mg-0.8Ca-5Zn-1.5Ag (denoted as ZQ71) and Mg-0.8Ca-5Zn-2.5Ag (denoted as ZQ63), were fabricated by alloying with calcium (Ca), zinc (Zn) and silver (Ag). The results obtained from electrochemical corrosion tests and in vitro degradation evaluation demonstrated that the three Mg alloys exhibited distinct corrosion resistance, and ZQ71 exhibited the lowest degradation rate in vitro among them. After addition of Zn and Ag, the antibacterial potential of Mg alloys was also enhanced. The in vitro cell experiments showed that all the three Mg alloys had good biocompatibility. After implantation in a rat femoral defect, ZQ71 showed significantly higher osteogenic activity and bone substitution rate than ZQ63 and ZQ, due to its higher corrosion resistance as well as the stimulatory effects of the released metallic ions. In addition, the average daily degradation rate of each Mg alloy in vivo was significantly higher than that in vitro, as could be due to the implantation site located in the highly vascularized trabecular region. Importantly, the correlations between the in vitro and in vivo degradation parameters of the Mg alloys were systematically analyzed to find out the potential predictors of the in vivo degradation performance of the materials. The current work not only evaluated the clinical potential of the three biodegradable Mg alloys as bone grafts but also provided a feasible approach for predicting the in vivo degradation behavior of biodegradable materials.

Abstract:Implant-associate infection (IAI) is a major cause of failure of bone implant materials, and one of the significant challenges in clinical managements. A synergistic coating strategy combining montmorillonite (MMT) sustained release, adsorption of bacteria and gentamicin (GS) bactericidal is proposed herein to tackle infection issues. Surface morphology, microstructure and chemical composition of the samples were investigated using SEM, XRD, FT-IR and XPS. Electrochemical experiments and immersion experiments reveal that corrosion resistance of Mg samples with GS/MMT coatings was higher than that of bare Mg alloy substrate in DMEM solution. In vitro studies demonstrated that the GS/MMT coating had a significant inhibitory effect on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The viability of MC3T3-E1 cells was 92.7% after a co-culturing for 72 h. After a subcutaneous transplantation of 90 days, the survival rate was 100% for GS/MMT-coated Mg alloy specimens with no infection at the implantation sites and no toxic damage to liver, kidney and local muscles pathological sections. This study provides a novel method for the preparation of sustained-release antimicrobial coatings on biodegradable Mg alloys as promising candidates for orthopedic implant materials.
In vitro evaluation of MgSr and MgCaSr alloys via direct culture with bone marrow derived mesenchymal stem cells

Abstract:Magnesium (Mg) and its alloys have been widely investigated as the most promising biodegradable metals to replace conventional non-degradable metals for temporary medical implant applications. New Mg alloys have been developed for medical applications in recent years; and the concept of alloying Mg with less-toxic elements have aroused tremendous interests due to the promise to address the problems associated with rapid degradation of Mg without compromising its cytocompatibility and biocompatibility. Of particular interests for orthopedic/spinal implant applications are the additions of calcium (Ca) and strontium (Sr) into Mg matrix because of their beneficial properties for bone regeneration. In this study, degradation and cytocompatibility of four binary MgSr alloys (Mg-xSr, x = 0.2, 0.5, 1 and 2 wt%) and four ternary MgCaSr alloys (Mg-1Ca-xSr, x = 0.2, 0.5, 1 and 2 wt%) were investigated and compared via direct culture with bone marrow-derived mesenchymal stem cells (BMSCs). The influence of the alloy composition on the degradation rates were studied and compared. Moreover, the cellular responses to the binary MgSr alloys and the ternary MgCaSr alloys were comparatively evaluated; and the critical factors influencing BMSC behaviors were discussed. This study screened the degradability and in vitro cytocompatibility of the binary MgSr alloys and the ternary MgCaSr alloys. Mg-1Sr, Mg-1Ca-0.5Sr and Mg-1Ca-1Sr alloys are recommended for further in vivo studies toward clinical translation due to their best overall performances in terms of degradation and cytocompatibility among all the alloys studied in the present work. Statement of significance: Traditional Mg alloys with slower degradation often contain aluminum or rare earth elements as alloying components, which raised safety and regulatory concerns. To circumvent unsafe elements, nutrient elements such as calcium (Ca) and strontium (Sr) were selected to create Mg-Sr binary alloys and Mg-Ca-Sr ternary alloys to improve the safety and biocompatibility of bioresorbable Mg alloys for medical implant applications. In this study, in vitro degradation and cellular responses to four binary Mg-xSr alloys and four ternary Mg-1Ca-xSr alloys with increasing Sr content (up to 2 wt%) were evaluated in direct culture with bone marrow derived mesenchymal stem cells (BMSCs). The roles of Sr and Ca in tuning the alloy microstructure, degradation behaviors, and BMSC responses were collectively compared in the BMSC direct culture system for the first time. The most promising alloys were identified and recommended for further in vivo studies toward clinical translation.
In vitro and in vivo studies on a Mg-Sr binary alloy system developed as a new kind of biodegradable metal

Abstract:Magnesium alloys have shown potential as biodegradable metallic materials for orthopedic applications due to their degradability, resemblance to cortical bone and biocompatible degradation/corrosion products. However, the fast corrosion rate and the potential toxicity of their alloying element limit the clinical application of Mg alloys. From the viewpoint of both metallurgy and biocompatibility, strontium (Sr) was selected to prepare hot rolled Mg-Sr binary alloys (with a Sr content ranging from 1 to 4 wt.%) in the present study. The optimal Sr content was screened with respect to the mechanical and corrosion properties of Mg-Sr binary alloys and the feasibility of the use of Mg-Sr alloys as orthopedic biodegradable metals was investigated by in vitro cell experiments and intramedullary implantation tests. The mechanical properties and corrosion rates of Mg-Sr alloys were dose dependent with respect to the added Sr content. The as-rolled Mg-2Sr alloy exhibited the highest strength and slowest corrosion rate, suggesting that the optimal Sr content was 2 wt.%. The as-rolled Mg-2Sr alloy showed Grade I cytotoxicity and induced higher alkaline phosphatase activity than the other alloys. During the 4 weeks implantation period we saw gradual degradation of the as-rolled Mg-2Sr alloy within a bone tunnel. Micro-computer tomography and histological analysis showed an enhanced mineral density and thicker cortical bone around the experimental implants. Higher levels of Sr were observed in newly formed peri-implant bone compared with the control. In summary, this study shows that the optimal content of added Sr is 2 wt.% for binary Mg-Sr alloys in the rolled state and that the as-rolled Mg-2Sr alloy in vivo produces an acceptable host response.
In Vitro and in Vivo Studies on Biomedical Magnesium Low-Alloying with Elements Gadolinium and Zinc for Orthopedic Implant Applications

Abstract:Ternary magnesium alloys with low combined addition of elements gadolinium and zinc were developed in the present work, with their microstructures, mechanical properties, in vitro degradation behaviors, and cytotoxicity being systematically studied. Furthermore, the Mg-1.8Zn-0.2Gd alloy, with the best in vitro performance, was implanted into Sprague Dawley rats to examine its in vivo degradation performance for up to 6 months. It was found that Mg-1.8Zn-0.2Gd, composed of a single α-Mg phase, owned excellent strength and toughness that were comparable to the CE marked MAGNEZIX, the mischmetal added Mg alloy. Owing to the uniform single-phased microstructure, the degradation rate of this alloy was around 0.12 mm/y measured by electrochemical testing, which was comparable to high purity magnesium. Moreover, the Mg-1.8Zn-0.2Gd alloy exhibited no cytotoxicity to L929, MG63, and VSMC cells. In vivo degradation characterized by micro-computed tomography revealed that the Mg-1.8Zn-0.2Gd implant could maintain structural integrity in the first 2 months, and serious degradation could be observed after 6 months. A remarkable 100% survival rate of experimental animals was observed with no negative effects on bone tissues. The implant and the surrounding bone were well integrated within 2 months, implying good biocompatibility and osteoconductivity of the experimental alloy. On the basis of the above findings, the feasibility of Mg-Zn-Gd alloys for use as orthopedic implants was systematically discussed. This study provides a new strategy for development of high-performance Mg-rare earth (RE)-based alloys with superior mechanical properties and corrosion resistance while effectively avoiding the possible standing toxic effect of RE elements.

Abstract:To meet the growing demand for antibacterial implants for bone-implant-associated infection therapy and avoid the adverse effects of secondary surgery, a degradable platform with pH responsiveness and ion-associated antibacterial properties was constructed. A small amount of Sn added to Mg-1Zn alloy reduces the biocorrosion rate, which can be attributed to Sn participation in outer-layer film formation, significantly reducing the biocorrosion rate and hydrogen evolution rate after implantation in vivo. These Mg alloys, which are susceptible to degradation in the acidic bacterial microenvironment, degrade by releasing Mg, Zn and Sn, producing favorably alkaline and antibacterial conditions. Samples with the composition of Mg-1Zn-0.5Sn were found to be beneficial for promoting initial cell adhesion and proliferation, resulting in improved biocompatibility and biosafety. The biocompatibility of this alloy was confirmed by the healthy behavior of animals and the absence of acute or chronic toxicity in the liver, spleen, and kidneys. Our results demonstrate that Mg-1Zn-0.5Sn is safe for biological systems, enabling its efficacious use in biomedical applications.

Abstract:Objective: To review the pharmacology, pharmacokinetics, efficacy, and safety of fingolimod, a novel immune modulator. Data Sources: Information was obtained through a MEDLINE search (1966–February 2007) and from published abstracts. Search terms included fingolimod, FTY720, FTY-720, and sphlngosine-1-phosphate receptor agonist. Study Selection and Data Extraction: All English-language studies and abstracts pertaining to fingolimod were considered for inclusion. Preference was given to human data. Data Synthesis: Fingolimod Is the first in a new class of immune modulators known as the sphingosine-1-phosphate receptor agonists. It is administered orally once dairy and causes a dose-related reduction in the number ot circulating lymphocytes by preventing their egress from secondary lymph organs, but it does not alter T-cell activation or proliferation. Bradycardia and lymphopenia are the most common adverse effects. Clinical trials have evaluated the efficacy of fingolimod in renal transplantation and multiple sclerosis (MS). Further research for renal transplantation will not take place, but Phase 3 studies in MS are underway, as Phase 2 study results are favorable. Conclusions: Due to its distinct mechanism of action and its oral administration, fingolimod may be a useful therapeutic option for patients with relapsing forms of MS. More data are needed to assess the safety and clinical utility of fingolimod.
Effects of Ga/Ag addition on corrosion behavior, cytotoxicity, and antibacterial activity of biodegradable Mg–2Zn alloy

Abstract:Ga and Ag elements with good biocompatibility are introduced into a Mg–2Zn alloy to design an antibacterial alloy able to inhibit the bacterial infection of bone implants. The effects of Ga/Ag element on the corrosion behavior, cytotoxicity, and antibacterial activity of the as-extruded Mg–2Zn alloy are investigated in vitro . The immersion measurement results show that the mass loss rate of the Mg–2Zn alloy increases from 0.09 ± 0.006 mm/y to 0.24 ± 0.03 mm/y after the addition of 0.5 wt% Ga and 0.5 wt% Ag. This is due to that Ga and Ag elements can increase the cathodic current density and reduce the charge transfer resistance ( R ct ). Furthermore, Ag is enriched in the Mg 7 (ZnAg) 3 phase, resulting in an increase in the micro-galvanic corrosion and the maximum local corrosion depth. The cytotoxicity tests reveal that Ag and Ga do not induce toxicity to osteoblastic cells (MC3T3-E1) after incubation for 72 h. Moreover, the addition of Ag and Ga increases prominently the antibacterial activity of Mg–2Zn alloys, which is principally associated with the release of Ga and Ag ions and the alkaline antibacterial environment.

Abstract:Reasonable optimization of degradation rate, antibacterial performance and biocompatibility is crucial for the development of biodegradable zinc alloy medical implant devices with antibacterial properties. In this study, various amounts of Mg elements were incorporated into Zn5Cu alloy to modulate the degradation rate, antibacterial properties and biocompatibility. The effects of Mg contents on the microstructure, corrosion behavior, antibacterial properties and biocompatibility of Zn-5Cu-xMg alloy were extensively investigated. The results revealed that with an increase of Mg content, the amount of Mg2Zn11 phase increased and its galvanic effect with the Zn matrix was enhanced, which accelerated the corrosion process and led to higher corrosion rate and high degradation rate of the alloy. Additionally, there was an increased release of Mg2+ and Zn2+ ions from the alloy which imparted excellent resistance against Escherichia coli and Staphylococcus aureus bacteria and improved biocompatibility, subcutaneous antibacterial and immune microenvironment regulation properties. Zn-5Cu-2 Mg exhibited superior antibacterial ability, cell compatibility, proliferation effect, subcutaneous antibacterial and immune microenvironment regulation performances, which can work as a promising candidate of biodegradable antibacterial medical implants.

Abstract:Magnesium alloys have shown prospective applications as a new biodegradable metal within bone. To garantee the longterm biocompatibility, a Mg-Zn-Ca alloy, composing of essential elements for human, was prepared and its feasibility for orthopedic applications was investigated. The in vitro and in vivo corrosion of Mg-Zn-Ca alloy as well as the biocompatibility were studied. The in vitro corrosion tests in five kinds of physiological solutions showed that the corrosion rates and corrosion morphologies of the alloy were strongly influenced by the solution used. The addition of serum in Hank’s and MEM significantly slowed down the corrosion rate and improved the corrosion uniformity of the alloy. The corrosion rate decreased with increasing serum concentration. The alloy showed the slowest corrosion rate as well as homogeneous corrosion morphology in MEM+10%FBS. Both the indirect and direct cell experiments indicated good cytocompatibility of the extruded Mg-Zn-Ca alloy. In vivo , we observed a gradual degradation process from the surface of extruded Mg-Zn-Ca alloy and only 40% in volume of implant was left after 4 weeks implantation in medullary cavities of mice. The micro-CT and histological analyses revealed its good biocompatibility with peri-implant new bone formation and increasing cortical bone thickness with increasing implantation period. This study showed that the extruded Mg-Zn-Ca alloy provided sufficient biocompatibility for orthopedic application, though the in vivo corrosion rate should be further reduced for clinical use.
Antibacterial Pure Magnesium and Magnesium Alloys for Biomedical Materials—A Review

Abstract:Implant-related infections are one of the major challenges faced by orthopedic surgeries. Developing implants with inherent antibacterial properties is an effective strategy to address this issue. Biodegradable magnesium and magnesium alloys have become a research hotspot due to their good bioactivity, mechanical properties, biocompatibility, and excellent antibacterial ability. However, magnesium and its alloys have rapid corrosion, and the difficulty in expelling harmful magnesium ions and hydrogen gas produced by degradation from the body. This review summarizes the mainstream surface modification techniques such as laser surface modification, friction stir processing, and micro-arc oxidation, along with their impact on the antimicrobial properties of magnesium-based materials. This paper reviews the latest research progress on improving the antibacterial properties of magnesium alloys through alloying and introduces the antibacterial effects of mainstream magnesium alloys and also elaborates on the antibacterial mechanism of magnesium alloy materials. It is expected to provide more basis and insights for the design of biodegradable magnesium alloys with antibacterial properties, thereby promoting their development and clinical application.

Abstract:Magnesium alloys are potential biodegradable materials and have received increasing attention due to their outstanding biological performance and mechanical properties. However, rapid degradation in the physiological environment and potential toxicity limit clinical applications. Recently, special magnesium–calcium (Mg–Ca) and magnesium–strontium (Mg–Sr) alloys with biocompatible chemical compositions have been reported, but the rapid degradation still does not meet clinical requirements. In order to improve the corrosion resistance, a rough, hydrophobic and ZrO2-containing surface film is fabricated on Mg–Ca and Mg–Sr alloys by dual zirconium and oxygen ion implantation. Weight loss measurements and electrochemical corrosion tests show that the corrosion rate of the Mg–Ca and Mg–Sr alloys is reduced appreciably after surface treatment. A systematic investigation of the in vitro cellular response and antibacterial capability of the modified binary magnesium alloys is performed. The amounts of adherent bacteria on the Zr–O-implanted and Zr-implanted samples diminish remarkably compared to the unimplanted control. In addition, significantly enhanced cell adhesion and proliferation are observed from the Zr–O-implanted sample. The results suggest that dual zirconium and oxygen ion implantation, which effectively enhances the corrosion resistance, in vitro biocompatibility and antimicrobial properties of Mg–Ca and Mg–Sr alloys, provides a simple and practical means to expedite clinical acceptance of biodegradable magnesium alloys.

Abstract:Magnesium (Mg) or its alloys are widely tested as potential orthopedic implants, particularly as biodegradable alloys for fracture fixation due to their mechanical properties are close to those of bone. Currently, available Mg or its alloys are confronted with challenges in passing regulatory biosafety tests prior to clinical trials due to its fast degradation and associated degradation products. The degradation of Mg is accompanied by the release of Mg ions, the rise of pH and osmolality in surrounding environments. According to the standard of ISO 10993 Part 13, the pH value shall be appropriate to the site of intended use maintaining in an appropriate range. Approaches to overcome these challenges include the selection of adequate alloying elements, proper surface treatment techniques and control of the degradation rate of Mg or its alloys developed as orthopedic implants. To date, Mg or its alloy-based bone implants have not yet been widely used in clinical applications as medical implants. This review critically summarized published methods to improve the corrosion resistance of Mg and its alloys. The current progress on in vitro cytotoxicity and in vivo biocompatibility properties of these metals was also reviewed. This review aimed to provide a reference for further research and development (R&D) of biodegradable Mg and its alloys with regard to the evaluation of their corrosion process and biocompatibility and facilitation of their translation to clinical applications.

Development of Novel Antibacterial Ti-Nb-Ga Alloys with Low Stiffness for Medical Implant Applications

Abstract:With the rising demand for medical implants and the dominance of implant-associated failures including infections, extensive research has been prompted into the development of novel biomaterials that can offer desirable characteristics. This study develops and evaluates new titanium-based alloys containing gallium additions with the aim of offering beneficial antibacterial properties while having a reduced stiffness level to minimise the effect of stress shielding when in contact with bone. The focus is on the microstructure, mechanical properties, antimicrobial activity, and cytocompatibility to inform the suitability of the designed alloys as biometals. Novel Ti-33Nb-xGa alloys (x = 3, 5 wt%) were produced via casting followed by homogenisation treatment, where all results were compared to the currently employed alloy Ti-6Al-4V. Optical microscopy, scanning electron microscopy (SEM), and energy dispersive spectroscopy (EDS) results depicted a single beta (β) phase microstructure in both Ga-containing alloys, where Ti-33Nb-5Ga was also dominated by dendritic alpha (α) phase grains in a β-phase matrix. EDS analysis indicated that the α-phase dendrites in Ti-33Nb-5Ga were enriched with titanium, while the β-phase was richer in niobium and gallium elements. Mechanical properties were measured using nanoindentation and microhardness methods, where the Young's modulus for Ti-33Nb-3Ga and Ti-33Nb-5Ga was found to be 75.4 ± 2.4 and 67.2 ± 1.6 GPa, respectively, a significant reduction of 37% and 44% with respect to Ti-6Al-4V. This reduction helps address the disproportionate Young's modulus between titanium implant components and cortical bone. Importantly, both alloys successfully achieved superior antimicrobial properties against Gram-negative P. aeruginosa and Gram-positive S. aureus bacteria. Antibacterial efficacy was noted at up to 90 ± 5% for the 3 wt% alloy and 95 ± 3% for the 5 wt% alloy. These findings signify a substantial enhancement of the antimicrobial performance when compared to Ti-6Al-4V which exhibited very small rates (up to 6.3 ± 1.5%). No cytotoxicity was observed in hGF cell lines over 24 h. Cell morphology and cytoskeleton distribution appeared to depict typical morphology with a prominent nucleus, elongated fibroblastic spindle-shaped morphology, and F-actin filamentous stress fibres in a well-defined structure of parallel bundles along the cellular axis. The developed alloys in this work have shown very promising results and are suggested to be further examined towards the use of orthopaedic implant components.
Microstructure, Mechanical Properties, in Vitro Degradation and Cytotoxicity Evaluations of Mg–1.5Y–1.2Zn–0.44Zr Alloys for Biodegradable Metallic Implants

Abstract:Mg-1.5Y-1.2Zn-0.44Zr alloys were newly developed as degradable metallic biomaterials. A comprehensive investigation of the microstructure, mechanical properties, in vitro degradation assessments and in vitro cytotoxicity evaluations of the as-cast state, as-heat treated state and as-extruded state alloys was done. The microstructure observations show that the Mg-1.5Y-1.2Zn-0.44Zr alloys are mainly composed of the matrix α-Mg phases and the Mg12ZnY secondary phases (LPS structure). The hot extrusion method significantly refined the grains and eliminated the defects of both as-cast and heat treated alloys and thereby contributed to the better mechanical properties and biodegradation resistance. The values of tensile strength and tensile yield strength of the alloy in the as-extruded condition are about 236 and 178 MPa respectively, with an excellent elongation of 28%. Meanwhile, the value of compressive strength is about 471 MPa and the value of bending strength is about 501 MPa. The superior bending strength further demonstrates the excellent ductility of the hot extruded alloys. The results of immersion tests and electrochemical measurements in the SBF indicate that a protective film precipitated on the alloy's surface with the extension of degradation. The protective film contains Mg(OH)2 and hydroxyapatite (HA) which can reinforce osteoblast activity and promote good biocompatibility. No significant cytotoxicity towards L-929 cells was detected and the immersion extracts of alloy samples could enhance the cell proliferation with time in the cytotoxicity evaluations, implying that the Mg-1.5Y-1.2Zn-0.44Zr alloys have the potential to be used for biomedical applications.

Abstract:Mg-Cu alloys are promising antibacterial implant materials. However, their clinical applications have been impeded by their high initial biodegradation rate, which can be alleviated using nanotechnology by for example surface nanomodification to obtain a gradient nanostructured surface layer. The present work (i) produced a gradient nanostructured surface layer with a ∼500 µm thickness on a Mg-0.2 Cu alloy by a surface mechanical grinding treatment (SMGT), and (ii) studied the biodegradation behavior in Hank's solution. The initial biodegradation rate of the SMGTed samples was significantly lower than that of the unSMGTed original counterparts, which was attributed to the surface nanocrystallization, and the fragmentation and re-dissolution of Mg2Cu particles in the surface of the SMGTed Mg-0.2 Cu alloy. Furthermore, the SMGTed Mg-0.2 Cu alloy had good antibacterial efficacy. This work creatively used SMGT technology to produce a high-performance Mg alloy implant material.
