Abstract:This paper presents two validated chromatographic methods, namely thin-layer chromatography (TLC)–densitometry and high-performance liquid chromatography (HPLC), for the simultaneous determination of caffeine (CAF), codeine (COD), paracetamol (PAR) in the presence of p-aminophenol (PAP) in their quaternary mixture. Good separation was achieved by using silica gel as the stationary phase and chloroform‒methanol‒acetone‒ammonia (8:1:2:0.1, V/V) as the mobile phase in the case of TLC–densitometry with retardation factor (RF) values of 0.38 ± 0.02, 0.24 ± 0.02, 0.61 ± 0.02 and 0.46 ± 0.02 for PAR, COD, CAF and PAP, respectively. Zorbax ODS column with mobile phase consisting of 0.1% orthophosphoric acid and acetonitrile (92:8, V/V) was used as stationary and mobile phase for HPLC, respectively, with retention time (tR) values of 1.1, 3.5, 4.6 and 8.5 min for CAF, PAP, COD and PAR, respectively. The two proposed methods were validated as per the International Council for Harmonisation guidelines. Finally, they were successfully applied for determination of the cited drugs in their quaternary mixture and marketed formulation.

Abstract:Objective:To establish a method for simultaneous determination of three consitituents in AEC Tablet containing as- pirin,ethenzamide and caffeine.Method:Inertsil-ODS column is used.The mobile phase is composed of methanol -0.1% di- ethylamine-acetic acid(40:56:4)with flow rate of 1.0mL/min.The detection wavelength is 280nm.Results:The linear re- sponse range were 120-480μg/mL for aspirin(r=0.9998),40-160μg/mL for ethenzamide(r=0.9998),20-80μg/mL for caf- feine(r=0.9997).The mean recoveries of aspirin,ethenzamide,caffeine were 100.3%,100.7%,99.7% respectively.Conclu- sion:The method is simple,quick and suitable for determination of these three constituents in compound preparation simulta- neously.

Abstract:Abstract A stability-indicating hybrid micelle liquid chromatography accompanied by UV detection was developed for the simultaneous analysis of either paracetamol (PCA) or pseudoephedrine hydrochloride (PSU) with their synthetic impurities. Mixture I contains PCA with p -amino phenol and p -nitro phenol, while mixture II involves the estimation of PSU with benzaldehyde and benzoic acid. Both mixtures were separated using a C18 column that was thermostatically maintained at 40°C and operating under a flow rate of 1.5 mL/min, applying UV detection at 240 nm for mixture I and 220 nm for mixture II. In both cases, the mobile phase consisted of 0.1 M sodium dodecyl sulfate, acetonitrile, and triethylamine (90:10:0.3, v/v/v) and adjusted to pH 4 (mixture I) or pH 3.7 (mixture II) using 2.0 M O -phosphoric acid. The proposed method was validated and successfully applied to assay different pharmaceuticals containing PCA or PSU. Moreover, the stability-indicating nature of the proposed method was proved through applying photolytic degradation procedures for PCA.

Abstract:Multicomponent drugs are medications that combine two or more active pharmaceutical ingredients in a single dosage form. These dosage forms improve the patient compliance, reduce the risk of drug interactions, and simplify dosing regimens. However, quality control of these multicomponent dosage forms can be challenging, especially if the final product contains four or more ingredients that are active (comprise stabilizers, preservatives, excipients, and other components). This problem can be more pronounced if the excipients can interfere with the analysis. In this work, a stability indicating assay method was developed and validated (according to the ICH International Guidelines) for the simultaneous determination of hydroquinone (HQ), tretinoin (TRT), hydrocortisone (HCA), butylated hydroxytoluene (BHT), methyl paraben (MP) and propyl paraben (PP) in commercially available pharmaceutical creams. The proposed method is based on gradient elution using X-Bridge C18 (150 × 4.6 mm, 5μm) column with a flow rate of 1 mL/min. The linear ranges (μg/mL) were 240-560 for HQ, 24-56 for MP, 132-308 for HCA, 6-14 for PP, 12-28 for BHT, 6.6-15 for TRT. During the validation process, the intra- and interday precision and trueness (evaluated as recovery) were found to be below 2.0% and between 100–102%, respectively. System suitability tests (SST) allow validating the herein proposed procedure specifically for pharmaceutical and industrial applications. SST test shows that the reported procedure fulfill with the Guidelines, allowing excellent separation of the analytes with very sensitive, accurate (precise and true) and reproducible quantitation of each analytes. The method was successfully applied in forced degradation studies of the six analytes. Specifically, acid degradation slightly affected HCA and BHT (91% recovery), while alkaline degradation drastically reduced HCA recovery (5.5%) and moderately affected BHT (85%). Photodegradation primarily influenced TRT quantity, and oxidative degradation intensified the BHT peak (130%).

Abstract:Purpose: To develop and validate RP-HPLC method for simultaneous determination of four active ingredients: picroside I (PSI), picroside II (PSII), phyllanthin (PHY) and boeravinone-B (BVB) in a polyherbal hepatoprotective tablet formulation.Methods: The study was carried out using Waters X-Bridge, C18 (250 mm x 4.6 mm, 5 μm) column with mobile phase consisting of 5 mM ammonium acetate in 10 % methanol and acetonitrile, with gradient programme at dual wavelengths of 220 nm and 274 nm and flow speed of 1 mL.min-1. The procedure was validated with respect to specificity, linearity, precision, accuracy, system suitability, limit of detection (LOD) and of quantification (LOQ), and robustness in line with ICH specifications.Results: The method was linear within the concentration range of 25 to 200 %, and the values of correlation coefficients (R2) were > 0.999. Intra-day and inter-day RSDs of PAs and RTs were < 5.0 %, with recovery in the range of 100.0 - 106.0 %.Conclusion: The four active ingredients have with good resolution with regard to the method used. The method is rapid, simple, highly selective, sensitive and cost effective, which make it an efficient method for quality assurance.

Abstract:Abstract A fast, sensitive, and green reverse-phase “high-performance thin-layer chromatography” approach for the simultaneous estimation of ibuprofen (IBF), caffeine (CAF), and paracetamol (PCM) in marketed formulations was established and verified in this study. The binary combination of acetone and water (80:20 v/v) was used as the green eluent system. The current method’s greenness was predicted using four different approaches, namely National Environmental Method Index, Analytical Eco-Score (89), ChlorTox (1.08 g), and the Analytical GREENness (83) approaches, which demonstrated an outstanding greener profile. The present approach was linear in the range of 25–800 ng·band −1 for the simultaneous estimation of IBF, CAF, and PCM. In addition, the current method was accurate (% recoveries = 100 ± 2), precise (%CV < 2%), robust (%CV < 2), sensitive (LOD = 1.13–2.71 ng·band −1 and LOQ = 3.39–8.10 ng·band −1 ), and green. The amount of IBF, CAF, and PCM in commercial tablets was determined to be 99.51%, 98.25%, and 100.64%, respectively. The present method for the simultaneous determination of IBF, CAF, and PCM in marketed tablets is supported by these data. The findings of this study suggested that the current approach may be consistently applied to analyze IBF, CAF, and PCM in marketed tablets.

Simultaneous Determination of Paracetamol and Amantadine in Human Plasma and Bioequivalence of Its Preparations in Healthy Volunteers by LC-MS

Abstract:OBJECTIVE: To develop an LC-MS method for the determination of paracetamol and amantadine in human plasma, and to evaluate their bioequivalence in healthy volunteers after given Pediatric paracetamol and amantadine hydrochloride effervescent tables. METHODS: After protein-precipitated with methanol, blood sample was separated on Shim-pack VP-ODS column using the mobile phase consisted of methanol-5 mmol·L-1 ammonium acetate (containing 0.5% formic acid)(45∶55) at a flow rate of 0.3 mL·min-1. Detection was carried out by API 3200 electrospray positive ionization mass spectrometry in the multiple reaction monitoring (MRM) mode. The MRM transitions of m/z 152.2→110.1, m/z 152.3→135.2, m/z 166.1→148.1 were used to quantify paracetamol, amantadine and pseudoephedrine(internal standard), respectively. RESULTS: The linear ranges were 0.080 0～12.0 mg·L-1 for paracetamol (r=0.996 4) and 0.008 0～1.20 mg·L-1 for amantadine (r=0.999 5), respectively. The RSDs ofintra-day and inter-day were both less than 6%, and the average method recoveries were 99.23% and 101.70%. The mean extraction recoveries were (97.0±2.0)% and (92.4±2.7)% for paracetamol and amantadine, respectively. CONCLUSION: The method is effective, sensitive, accurate, specific, rapid, and it is suitable for the determination of paracetamol and amantadine in human plasma. The method can be used for bioequivalence study of Pediatric paracetamol and amantadine hydrochloride effervescent tables.

Abstract:A quick, simple, sensitive, efficient and stability‐indicating reverse‐phase ultraperformance liquid chromatographic method for the estimation of propylparaben, methylparaben and sodium benzoate in a pharmaceutical liquid oral formulation was developed. A Waters Acquity UPLC BEH C18, 50 × 2.1 mm, 1.7 μm i.d. column was used to perform chromatographic separation with a 0.1% perchloric acid mobile phase used as solvent A and a mixture of 0.1 % perchloric acid and methanol in the ratio 20:80 (v/v), respectively, as solvent B. The experiments were carried out at a flow rate of 0.4 ml/min and the detection wavelength was 240 nm. The compartment temperature of the column was set at 40°C and the injection volume was set at 2 μl. The main aim of the research was to develop a single UPLC assay method for promethazine (active ingredient) and preservatives in the oral solution of promethazine HCl and dextromethorphan HBr that contains promethazine (active ingredient) and methylparaben, propylparaben and sodium benzoate (preservatives). An assay of dextromethorphan HBr was developed and validated by another HPLC method. The drug and preservatives were eluted at retention times of 19.3 min for promethazine HCl, 9.3 min for methylparaben, 18.9 min for propylparaben and 8.9 min for sodium benzoate. Validation of the developed method was carried out as stated by the International Conference on Harmonization guidelines ICH Q2B and under USP . The analytical parameters verified specificity/selectivity, linearity, accuracy, ruggedness and robustness. The linearity ranges of promethazine HCL, methylparaben, propylparaben and sodium benzoate were 10–100, 10–80, 1.0–8.0 and 10–80 μg/ml, respectively, with a correlation coefficient of active ingredients and preservatives of 1.00. Percentage recoveries of promethazine, propylparaben, methylparaben, and sodium benzoate were 100.0–100.2, 99.0–100.3, 99.5–98.0 and 99.0–100.0%. The validated analytical method proves that the method is specific, precise, linear, accurate, sensitive, rugged and stable, indicating the quantification of the active ingredient and all preservatives in liquid oral formulations.

Abstract:A simple, precise, and accurate high-performance thin-layer chromatography (HPTLC) method was developed and authenticated to perform simultaneous estimation of dapagliflozin propanediol monohydrate (DAPA), vildagliptin (VILDA), and metformin (MET) in active pharmaceutical ingredient (API) and tablet dosage form. It was found that there is not any reported stability-indicating HPTLC method for the simultaneous determination of DAPA, VILDA and MET; hence, this method was developed. Merck HPTLC silica gel 60 F 254 plates were selected as the stationary phase. Acetonitrile, formic acid, and water (9:1:0.5, V/V ) were the chromatographic solvents used. A densitometric study was conducted at the wavelength of 215 nm. For DAPA, VILDA, and MET, the concentrations used to conduct the study were 0.2‒0.6 μg/band, 2‒6 μg/band, and 10‒30 μg/band, respectively. The peaks of DAPA, VILDA, and MET have retardation factors ( R F ) of 0.70, 0.45, and 0.34, respectively. The regression coefficients ( R 2 ) of DAPA, VILDA, and MET were 0.9955, 0.9958, and 0.9972, respectively. The limit of detection values of DAPA, VILDA, and MET were 0.0018 μg/band, 0.0094 μg/band, and 0.1434 μg/band. The limit of quantification of DAPA, VILDA, and MET were 0.0056 μg/band, 0.0285 μg/band, and 0.4346 μg/band, respectively. With respect to International Council for Harmonisation (ICH) prerequisites, the accuracy, precision, and robustness outcomes were within suitable limits. The outcomes of stability data revealed that the degradation by acid, base, hydrogen peroxide, and water produced distinct peaks of DAPA, VILDA, and MET, along with a few other peaks, having different R F values. The forced degradation study for DAPA, VILDA, and MET was performed. DAPA was found to be stable in acidic, oxidative, and neutral conditions. Under acidic, basic, and oxidative conditions, VILDA was observed to be stable. The proposed technique passes all the ICH criteria; hence, it is an accurate, easy, suitable, and stable method for the analysis of DAPA, VILDA, and MET in combined dosage form.

Abstract:A new simple, rapid, and sensitive liquid chromatographic method has been developed and validated for the simultaneous determination of caffeine, paracetamol, pseudoephedrine, aspirin, dextromethorphan, and chlorpheniramine in pharmaceutical tablet formulations and was performed on conventional C18 and monolithic silica C18 columns. Chromatographic separation was achieved by using a mixture of methanol and a buffer containing 10 mM NaH2PO4, 3 mM heptane sulfonic acid sodium salt, 0.1% triethylamine, pH 2.5 (adjusted with hydrochloric acid) in a ratio 50:50 v/v, 40:60 v/v for conventional C18 and monolithic silica C18 columns, respectively. Quantitation was achieved with UV detection at 214 nm, based on peak area. The proposed method could be used in the presence of salicylic acid as impurity or decomposition product in dosage forms containing aspirin. The proposed method was validated and successfully applied for the analysis of pharmaceutical formulations and laboratory-prepared mixtures containing these multicomponent combinations.
HPTLC-Densitometric Analysis of Eperisone Hydrochloride and Paracetamol in Their Combined Tablet Dosage Form

Abstract:A simple, precise, accurate, and reliable HPTLC method has been developed and validated for the analysis of EPE-Eperisone hydrochloride and PCM-Paracetamol in their combined dosage form. Identification and analysis were performed on 100 mm × 100 mm layer thickness 0.2 mm, precoated silica gel G 60 -F 254 aluminum sheet, prewashed with methanol, and dried in an oven at 50°C for 5 min. Toluene : methanol : ethyl acetate : glacial acetic acid (4 : 3.5 : 2.5 : 0.05) (v/v/v/v) was used as mobile phase. Calibration plots were established showing the dependence of response (peak area) on the amount chromatographed. The validated calibration ranges were 200–700 ng/spot and 1300–4550 ng/spot for EPE and PCM with correlation coefficient ( R 2 ) 0.994 and 0.996, respectively. Average % recovery was between 98.61–100.94% and 99.18–100.57% for EPE and PCM, respectively. The spots were scanned at 248 nm in a reflectance mode. The proposed method was validated as per ICH guidelines and successfully applied to the estimation of EPE and PCM in their combined tablet dosage form.
Sustainable liquid chromatographic determination and purity assessment of a possible add-on triple-action over-the-counter pharmaceutical combination in COVID-19

Abstract:Nowadays, all researchers are focused on combating the pandemic COVID-19. According to recent statistics, most patients are managed at home. An over-the-counter (OTC) triple action formula containing paracetamol (PAR), aspirin (ASP), and diphenhydramine (DIPH) is widely prescribed for pain, fever and as night-time sleep aid. For COVID-19 patients, this combination is now suggested as part of symptomatic therapy and prophylaxis. In this work, two simple liquid chromatographic approaches were designed for simultaneous determination of PAR, ASP, and DIPH in Excedrin® PM caplets, beside three specified official toxic impurities, namely, p-aminophenol, p-nitrophenol, and salicylic acid. The first method comprised high-performance thin-layer chromatographic separation coupled with densitometric quantification, on silica gel HPTLC 60 F254 aluminium sheets as the stationary phase, ethyl acetate-methanol-aqueous ammonium hydroxide (10.0: 2.0: 0.1, by volume) as the developing system and scanning was performed at 210.0 nm. The second one is a high-performance liquid chromatography coupled with diode array detector. Successful separation of the six components was performed on XTerra C18 column with isocratic elution of mobile phase 0.1% triethylamine acidified water: methanol (70:30, v/v) adjusted with o-phosphoric acid to pH 3.0 and methanol (90:10, v/v) with flow rate programming and detection at 210.0 nm. Validation of the proposed methods was performed according to ICH guidelines. Both methods were successfully used for quality control of the cited drugs in their marketed formulation. Moreover, the in-vitro release study was monitored using the proposed HPLC-DAD method. The greenness profile of the proposed methods was assessed and comparatively evaluated through various assessment tools, specifically; the analytical eco-scale system, national environmental method index (NEMI), green analytical procedure index (GAPI) and analytical greenness (AGREE) metric.
Validation of HPLC-DAD method for analysis of paracetamol and potassium sorbate in liquid oral formulations and its application to forced degradation study

Abstract:Abstract Due to the frequent use of paracetamol formulations, it is useful to develop an analytical technique for the determination of intact paracetamol in presence of other drugs and excipients or the degradation products. In this study, a simple, isocratic, fast, specific, accurate and precise stability-indicating high performance liquid chromatography (HPLC) method has been developed and validated for simultaneous quantitative determination of paracetamol (PCM) and potassium sorbate (PS) in oral liquid formulations. The chromatographic separation was achieved on Zorbax SB C18 column (150 × 4.6 mm, 5 μm) with Zorbax SB C18 precolumn (12.5 × 4.6 mm, 5 μm) using distilled water pH 2 with ortho-phosphoric acid and acetonitrile (70:30, v/v) as a mobile phase, and UV detection at 235 nm. The temperature of the column was kept constant at 25 °C. The method was validated according to International Conference on harmonization (ICH) guidelines. The method demonstrated excellent linearity, with a correlation coefficient of 0.9996 and 0.9998 for PCM and PS, respectively, over the concentration ranges of 10–600 μg mL −1 (PCM) and 6–500 μg mL −1 (PS). The retention time was found to be 1.98 and 4.86 min for PCM and PS, respectively. Oral liquid formulation samples were subjected to various stress conditions (acidic and alkaline hydrolysis, as well as oxidative, heat and photolytic degradation) for the purpose of forced degradation study. The major degradation of paracetamol was achieved in acidic and basic stress conditions, while thermal and photolytic degradation generally had the least influence. On the other hand, potassium sorbate was highly susceptible to photolytic degradation. It was also shown that the formulation has strong influence on stability of tested compounds. Forced degradation studies demonstrated the stability-indicating power of the method and can be used to assess the stability of paracetamol and potassium sorbate in oral liquid formulations.

Diphenhydramine & Naproxen peaks in the chromatogram passed the system suitability criteria %RSD of peak areas of Diphenhydramine & Naproxen was not more than 2.0% for variation in mobile phase composition. The newly developed RP-HPLC method for determination of Diphenhydramine & Naproxen in tablet dosage forms is specific, precise, accurate and rapid. The proposed method can be conveniently adopted for routine quality control analysis.

Abstract:Aims: Metformin HCl, Dapagliflozin and Saxagliptin is a new drug combination for the treatment of Diabetes Mellitus which is one of the oldest and lethal diseases of mankind. Aim of the research work was to develop and validate novel, rapid, sensitive, specific, robust stability indicating analytical method for simultaneous estimation of: Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form as fixed dose formulation. Study Design: Method development and validation was performed as recommended in ICH guideline “Validation of analytical procedures: Test and Methodology Q2 (R1)”. Methodology: Method develop with chromatographic parameters as C18 column (250mm×4.6 mm, 5mm particle size), HPLC system with PDA detector and mobile phase contained a mixture of Phosphate Buffer pH 3.5 and Acetonitrile (80:20 v/v) + 1 ml triethylamine per 100 ml mobile phase. The flow rate was set to 1 ml/min with responses measured at 265 nm, injection volume was 20 μl, and run time of 15 mins. Results: The retention time of Metformin HCl, Dapagliflozin and Saxagliptin was 5.8 min, 6.8 mins and 8.4 min respectively with resolution of 3.5 between Metformin HCl and Dapagliflozin and 4.5 between Dapagliflozin and Saxagliptin. Linearity was established in the range of 250-1500 μg/ml for Metformin HCl, 1.25-7.5 μg/ml for Dapagliflozin and Saxagliptin with correlation coefficients more than 0.999. The percentage recoveries were between 98.39-101.66 for Metformin HCl, 99.01-101.77 for Dapagliflozin and 98.88-101.87 for Saxagliptin Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The forced degradation studies were performed by using HCl, NaOH, H2O2, thermal and UV Radiation. The developed method was successfully applied for the quantification and hyphenated instrumental analysis. Conclusion: Significance of developed method is that it can be utilize for routine or unknown sample analysis of assay of Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form developed by various Pharmaceutical Industry.

Abstract:For the first time, a highly sensitive and simple LC–MS/MS method after one-step precipitation was developed and validated for the simultaneous determination of paracetamol (PA), pseudoephedrine (PE), dextrophan (DT) and chlorpheniramine (CP) in human plasma using diphenhydramine as internal standard (IS). The analytes and IS were separated on a YMC-ODS-AQ C18 Column (100mm×2.0mm, 3μm) by a gradient program with mobile phase consisting of 0.3% (v/v) acetic acid and methanol at a flow rate of 0.30mL/min. Detection was performed on a triple quadrupole tandem mass spectrometer via electrospray ionization in the positive ion mode. The method was validated and linear over the concentration range of 10–5000ng/mL for PA, 2–1000ng/mL for PE, 0.05–25ng/mL for DT and 0.1–50ng/mL for CP. The accuracies as determined from quality control samples were in range of −8.37% to 3.13% for all analytes. Intra-day and inter-day precision for all analytes were less than 11.54% and 14.35%, respectively. This validated method was successfully applied to a randomized, two-period cross-over bioequivalence study in 20 healthy Chinese volunteers receiving multicomponent formulations containing 325mg of paracetamol, 30mg of pseudoephedrine hydrochloride, 15mg of dextromethorphan hydrobromide and 2mg of chlorphenamine maleate.

Abstract:The present work aimed to develop and validate a simple, rapid, sensitive, accurate, and precise method for simultaneous determination of metformin hydrochloride and vildagliptin in tablet and biological samples. Isocratic elution of both the analytes was performed at 35 °C by injecting 20 μL into Thermo Hypersil ODS C18 column (5 μm, 4.6 mm× 250 mm), while the flow rate was set to 0.8 mL/min. The mobile phase comprised of methanol, acetonitrile, and phosphate buffer (5:30:65, v / v , pH 3.5), and wavelength was selected at 212 nm. The overall run time per sample was 7.0 min with a retention time of 3.36 and 5.41 min for metformin hydrochloride and vildagliptin, respectively. The calibration curve was linear from 10–140 μg/mL for metformin and 1–14 μg/mL for vildagliptin with a coefficient of determination ( R 2 ) ≤ 0.9919, while repeatability and reproducibility (expressed as relative standard deviation) were lower than 1.13 and 0.97%, respectively. Force degradation studies indicated a complete separation of the analytes in the presence of their degradation products providing a high degree of method specificity. The proposed reversed-phase high-performance liquid chromatography (RP-HPLC) method was demonstrated to be simple and rapid for the determination of metformin hydrochloride and vildagliptin in commercially available tablet and biological samples providing recoveries ranged between 100.13–100.29%.

Abstract:This study was aimed at developing and validating a UV spectrophotometric method for the estimation of paracetamol in chitosan coated alginate beads throughout their pharmaceutical development. Paracetamol content was assessed at 234 nm in various buffers to cover the physiological range of gastrointestinal fluid from pH 1.2 to pH 6.8. The linearity range was found to be 0.5–10 μg/mL. The proposed UV method was validated in accordance with main ICH guidelines. The findings showed that this UV spectrophotometric method is accurate, precise, and reproducible while also being uncomplicated, cost-effective, and not requiring a lot of time. It can be used to estimate paracetamol in routine quality control and drug dissolution studies.
