Abstract:A simple, sensitive, accurate and precise liquid chromatography mass spectrometry (LC-MS) method was developed for determination of pyrimethamine and sulfadoxine in pharmaceutical formulation. The chromatographic separation was performed on Phenomenex C18 column (I.D. 250 mm x 4.6mm, 5μm) and the column oven temperature was set at 30 ºC, with an injection volume of 5 μl, acetonitrile and Phosphate buffer in the ratio 75: 25 (v/v) at pH 2.5 as mobile phase. The Electrospray Ionization (ESI) was performed using nitrogen gas to assist nebulization (flow rate was set at 1.0 mL/min), the mass spectrometer was operated in the multiple reaction monitoring (MRM) mode. As part of the method validation, specificity, limit of detection (LOD), limit of quantification (LOQ), linearity, accuracy, precision and robustness were determined. The limit of detection and limit of quantification for pyrimethamine and sulfadoxine were 0.69 and 0.82 ng/mL, and limit of quantification for pyrimethamine and sulfadoxine were 2.29 and 2.73 ng/mL respectively. The intra- and inter-day precisions were lower than 0.53% while the accuracy ranged from 99.99% to 100.11%. The admissible robustness indicates that the method remains unaffected by small but deliberate variations.

Abstract:Abstract Background In the dapagliflozin (DPF) synthesis, 5-Bromo-2-chlorobenzoic acid (5-BC impurity) and 4-Bromo-1-chloro-2-(4-ethoxybenzyl) benzene (4-BC impurity) are used as starting and reagent sources, respectively. The presence of 5-BC and 4-BC impurities in DPF could potentially affect the effectiveness of the final DPF product. The purpose of this investigation was to develop a stability indicating HPLC methodology for the separation of DPF, its process-related impurities and degradants. The method of analysis was developed on Xbridge Phenyl C18 column of dimensions, 250 × 4.6 mm, 5 μm with gradient elution using mobile phase made up of 0.05% aqueous trifluoroacetic acid and acetonitrile (AcN). Results The method proposed indicates a good linearity ( R 2 = 0.9996 and 0.9993), good system precision (RSD ≤ 2%), good method precision (RSD ≤ 2%), accuracy (50–150%), LOD (0.000053 ppm and 0.0000165 ppm) and LOQ (0.00016 ppm and 0.00005 ppm) for 4-BC impurity and 5-BC impurity, respectively. LC–MS was used to detect and characterise degradants that were obtained in acid and base condition were identified and characterised. A comparison of the fragmentation pattern of the [M + H] + ions of DPF and its degradation products revealed the most likely processes for the generation of degradation products. Conclusion DPF sample quality can be evaluated using the suggested method for the presence of 4-BC impurity, 5-BC impurity and 2-(3-(4-ethoxy benzyl)-4-chloro phenyl)-tetrahydro-6-(hydroxy methyl)-2H-pyran-3,4,5-triol.

Abstract:A selective reversed-phase high-performance liquid chromatography (RP-HPLC) technique was developed for 1α-hydroxy vitamin D2 (Doxercalciferol) related substances analysis. The technique is proficient of eluting various impurities related to process and degradation. To separate both the vitamin D2 Analogs i.e., doxercalciferol and ergocalciferol. HPLC of Shimadzu, provided with autosampler, column oven, quaternary pump and UV detector, were used for development studies. The mobile phase used water and acetonitrile. The diluent used was methanol and detection was examined at wavelength 265 nm. The method is sensitive for doxercalciferol, ergocalciferol, impurity-A and impurity-B; the limit of detection (LoD), value is 0.01% and limit of quantitation (LoQ) value is 0.03%. Linearity of doxercalciferol and related impurities at various concentrations was performed and the obtained correlation coefficient for doxercalciferol is 0.995, ergocalciferol is 0.997, impurity-A is 0.990 and impurity-B is 0.993. The optimized method was validated and demonstrated that the technique is selective, precise, linear, sensitive and accurate. The method is simple, inexpensive and useful for routine testing of doxercalciferol API. In addition, the technique was effectively applied for the elution of both the vitamin D2 Analogs i.e., doxercalciferol and ergocalciferol. Further, the same method is applicable for the determination of assay for these two API's and the same assay method was validated.

Abstract:Vitamin B6 is an indispensable micronutrient in organisms and is widely distributed in blood, tissues, and organs. Changes in the content and ratio of vitamin B6 can affect the entire physiological condition of the body, so it becomes particularly important to reveal the relationship between changes in its content and disease by monitoring vitamin B6 levels in the organism. In this study, a two-dimensional liquid chromatography-UV detector (2D-LC-UV) was used to establish a method for the simultaneous detection of PLP, PA, and PL for the first time. First, PLP, PA, and PL were extracted with plasma: 0.6 M TCA: ultrapure water = 1:2:3 (v/v/v) and then derivatized. Enrichment and preliminary separation were performed on a one-dimensional column and automatically entered into a two-dimensional column for further separation. This method exhibited good selectivity, and the correlation coefficients for the analyte calibration curves were >0.99. The detection limits for PLP, PA, and PL were 0.1, 0.2, and 4 nmol/L, respectively. The results showed that the system has high loading capacity, excellent resolution, and a good peak shape. This method is expected to provide applicability for the determination of PLP, PA, and PL in pharmacological, pharmaceutical, and clinical research.

Abstract:High performance liquid chromatography is one of the techniques of choice for the separation and quantitative determination of drugs in mixture form. Ipriflavone, ascorbic acid, pyridoxine, vitamin D3, and lysine are formulated together as an adjuvant combination in osteoporosis In this work, we developed and validated two complementary high performance liquid chromatographic methods to determine the five compounds in their pharmaceutical dosage form. The first method (method A) was capable of determining ipriflavone, ascorbic acid, pyridoxine, and vitamin D3 in their bulk and combined pharmaceutical formulation. The method is based on Liquid Chromatographic separation with UV detection at 254 nm using Agilent Eclipse XDB-C18 column with a mobile phase consisting of 25 mM ammonium acetate buffer (pH 4.2): methanol in gradient mode. Due to the high polarity of lysine, it was difficult to achieve satisfactory retention on reversed phase columns. So, we separated it on a strong cation exchange column (Exsil 100 SCX) without derivatization with a mobile phase consisting of 10 mM sodium dihydrogen phosphate and 200 mM sodium chloride (pH 6) with UV detection at 210 nm (method B). Validation of the proposed methods was performed according to ICH guidelines Q2(R1). The proposed methods proved to be valid for selective analysis of the stated drugs in their bulk and combined pharmaceutical formulation. Greenness assessment of the developed methods was evaluated using three assessment tools: ESA, GAPI and the most recently developed tool AGREE, showing a satisfactory comprehensive guide of the greenness of the developed methods.

Abstract:A simple and simultaneous reverse phase high-performance liquid chromatographic method was developed for the quantification of aripiprazole (ARI) and two preservatives, namely, methyl paraben and propyl paraben in ARI oral solution. The method was developed on ACE C18 (4.6 × 250 mm, 5 µm) column using gradient elution of 0.1% v/v trifluoroacetic acid in water and acetonitrile as mobile phase components. Flow rate of 1.0 mL/min and 30°C column temperature were used for the method at quantification wavelength of 254 nm. The developed method was validated in accordance with International Conference on Harmonization guideline for various parameters. Forced degradation study was conducted in acid, base, peroxide, heat, and light stress conditions. ARI was found to degrade in oxidation, acid hydrolysis, and heat while it was stable under the remaining conditions. Specificity of the method was verified using Photo Diode Array (PDA) detector by evaluating purity of peaks from degradation samples. Major degradation impurities formed during stress study were identified using liquid chromatography–mass spectrometry method. The present method was useful for determining the content of all the three main analytes present in the oral solution without interference from degradation impurities. The method was robust under the deliberately modified conditions.

Abstract:The purpose of this research is to design and validate a unique, simple, and effective RP-HPLC analytical method using the QbD methodology. The QbD methodology not only confirms the method's robustness but also aids in the development of repeatable and reliable data. The goal of this work is to develop and validate a simple analytical RP-HPLC method for determining Cefpodoxime Impurity (Cefpodoxime Acid) in pharmaceutical formulations using the Analytical Quality by Design methodology (AQbd) in accordance with ICH Q8 guidelines. The Analytical Quality by Design technique was used in the proposed study. Cefpodoxime impurity (Cefpodoxime Acid) was chromatographically evaluated using an Inertsil C18 (5 m) column. The mobile phase was phosphate buffer and methanol pH 4.0 in the ratio of 60:40, which was driven onto the column at 0.8 ml/min using an isocratic elution protocol. 222 nm was discovered to be the detection wavelength for impurity estimation. Design expert 11 (Trial edition) software was used to investigate the effects of the mobile phase composition, flow rate, and pH. The analysis of cefpodoxime acid took 20 minutes to complete & at 15.6 minutes, the impurity was eluted.

Abstract:Abstract A stability-indicating hybrid micelle liquid chromatography accompanied by UV detection was developed for the simultaneous analysis of either paracetamol (PCA) or pseudoephedrine hydrochloride (PSU) with their synthetic impurities. Mixture I contains PCA with p -amino phenol and p -nitro phenol, while mixture II involves the estimation of PSU with benzaldehyde and benzoic acid. Both mixtures were separated using a C18 column that was thermostatically maintained at 40°C and operating under a flow rate of 1.5 mL/min, applying UV detection at 240 nm for mixture I and 220 nm for mixture II. In both cases, the mobile phase consisted of 0.1 M sodium dodecyl sulfate, acetonitrile, and triethylamine (90:10:0.3, v/v/v) and adjusted to pH 4 (mixture I) or pH 3.7 (mixture II) using 2.0 M O -phosphoric acid. The proposed method was validated and successfully applied to assay different pharmaceuticals containing PCA or PSU. Moreover, the stability-indicating nature of the proposed method was proved through applying photolytic degradation procedures for PCA.

Abstract:A novel, quick and precise RP-UHPLC analytical method for the simultaneous determination of moxifloxacin (MFX), voriconazole (VCZ) and pirfenidone (PIR) was developed and validated according to the International Conference on Harmonization guidelines using a QbD-driven response surface Box-Behnken design. The developed method was validated considering the selectivity, sensitivity, linearity, accuracy-precision, robustness, stability, limit of detection and limit of quantification, respectively. Resolution between MFX, VCZ and PIR was achieved using a gradient elution protocol against a Waters Symmetry Shield C18 column (150 × 4.6 mm2 , 5 μm) using an Agilent 1290, Infinity II series LC system. The method was applied to quantitatively estimate proprietary and in-house prepared pharmaceutical topical ophthalmic formulations containing MFX, VCZ and PIR at wavelength (λmax ) of 296, 260 and 316 nm. The method is sensitive enough to detect up to 0.1 ppm of analytes in the formulation. The method was further exploited to study and identify the possible degradation products of the analytes. The proposed chromatographic method is simple, economical, reliable and reproducible. In conclusion, the developed method could be applicable for routine quality control analysis of single or combined MFX, VCZ and PIR-containing units or bulk dosage forms in pharmaceutical industries and research organizations working on drug discovery and development.

Abstract:A selective, rapid, accurate and precise stability-indicating RP-HPLC method was developed for monitoring vilazodone in the presence of its degradation products (DPs) in API and pharmaceutical dosages. Chromatographic separation was achieved using a Grace phenyl C18 column (250 × 4.6 mm, 5 µm particle size) at a flow rate of 1.0 mL min−1 with a linear gradient elution of ammonium acetate buffer (10 mM, pH 5.0 adjusted by acetic acid) with acetonitrile as the mobile phase. Detection was performed at 240 nm. The chromatographic conditions were optimized by the design of experiments to obtain the best possible separation with a minimum number of trials. A face-centered central composite design (three levels for each factor) was employed for optimization, and an interaction of the independent variables (pH of mobile phase, column temperature and % of organic modifier) on the resolution of critical pairs was studied. The drug was subjected to the stress conditions of hydrolytic (acid, base and neutral), oxidative, thermal and photolytic degradation. It was found to be degraded significantly in hydrolytic (basic and acid) and oxidative conditions, while it was stable in neutral hydrolytic, thermal and photolytic conditions. LC-QTOF/MS/MS studies were carried out to characterize the major DPs. The method was fully validated for selectivity, linearity, accuracy, precision and robustness in compliance with ICH guideline Q2 (R1).

Abstract:Two drug substances namely pentoxifylline and triprolidine were subjected to various stress conditions and their degradation profile was studied with conventional LC-MS. Interestingly, these compounds under went distinct transformation giving rise to degradation products. These degradation products from alkali and oxidative degradation were isolated and purified with preparative HPLC. The structures of these products was elucidated using 1D, 2D NMR spectroscopy and high resolution mass spectrometry (Q-TOF LC/MS). A novel gem-dihydroperoxide, namely 1-(5,5-Bis-hydroperoxy-hexyl)-3,7-dimethyl-3,7-dihydro-purine-2,6-dione was identified as an oxidative degradation product of pentoxifylline. Two major base hydrolysis products which were reported earlier were also isolated and identified. Two oxidative degradation products of triprolidine were identified to be Triprolidine N-Oxide and Pyridin-2-yl-p-tolyl-methanone. An efficient stability indicating UHPLC separation methods were developed for pentoxifylline and triprolidine along with their degradation products, using 1.8 and#956;m, C18 reverse phase column. Baseline separation was achieved with a run time of four minutes. This study focuses on the validation of a high-performance liquid chromatography (HPLC) method for quantifying Atorvastatin and its impurities, addressing a critical aspect of pharmaceutical analysis. Objective: The primary objective is to conduct a comprehensive validation study for the HPLC method, covering specificity assessment, response function establishment, and a detailed analysis of precision, trueness, and tolerance intervals. The emphasis is on demonstrating the method's precision, accuracy, and stability-indicating capabilities across various concentrations and compounds. Methods: The HPLC method is validated through rigorous assessments, including specificity, response function establishment, and analyses of precision, trueness, and tolerance intervals. Induced degradation experiments are conducted to explore Atorvastatin's behavior under extreme conditions. Application of a multivariate optimization strategy to validate an RP-HPLC analytical method for determining hydroxychloroquine sulphate in pharmaceutical products and in forced degradation studies

Abstract:Abstract In this study, a multivariate optimization strategy was used to develop and validate a simple, rapid, accurate, cost-effective, and stability-indicative RP-HPLC analytical method for quantifying hydroxychloroquine sulphate (HCQ) in coated tablets. The validation conditions involved isocratic elution mode, using a mixture of buffer solution at pH 2.2 and methanol (74:26, v/v) as the mobile phase, an Agilent® reverse phase column, model Zorbax Eclipse Plus C18 (250 cm × 4.6 mm × 5 μm), a flow rate of 1.3 mL min −1 , column temperature 40 °C and detection at 343 nm. The method showed linearity in the range of 4–44 μg mL −1 , with a correlation coefficient (R) of 0.9998. Recovery obtained average values of between 99.71 and 100.84% and precision with average RSD values of <2%. The robustness demonstrated by assessing the effect of seven variables (pH of the mobile phase buffer; percentage of methanol; filter brand; mobile phase flow rate; wavelength; column temperature and sample agitation time), with effect values for each variable lower than the calculated value of s√2 (1.43), showed that none of these factors had a significant influence on the analytical response. The method was applied to samples of the reference medicine Plaquinol® 400 mg and similar Reuquinol 400 mg, nomenclature established by the National Health Surveillance Agency (Anvisa), law no. 978 of 10 February 1999, purchased from local pharmacies. Results showed advantages and benefits in relation to the official method and those reported in the literature. The application of the multivariate strategy, the choice of methanol, in a lower proportion in the organic phase, due to its low toxicity, economy and easier availability compared to acetonitrile, and the other organic solvents used was a promising and important alternative for the analytical method. Furthermore, the use of reversed stationary phase, common in quality control laboratories, provided an analyte retention time of 4.595 min, demonstrating good performance and speed in routine analyses.

Abstract:Ensuring the quality control of active pharmaceutical ingredients is crucial for drug products being introduced into the market. Even for established drugs, it is necessary to maintain a cutting-edge impurity control system. To analyze caffeine and chlorphenoxamine hydrochloride in their binary mixture, as well as theophylline and chlorphenoxamine N-oxide as related substances, a reversed phase-high performance liquid chromatography combined with a diode array detector system was created. The chromatographic separation was conducted using a C18 X-select Waters® column. The mobile phase consisted of 20.0 mM potassium dihydrogen phosphate modified to pH 3 with o-phosphoric acid and methanol. A gradient elution program was adopted at a flow rate of 1.3 mL/min and detected at a wavelength of 222 nm. The present methodology demonstrates a concentration ranging from 2–60, 1–80, 0.5–20 to 0.4–20 μg/mL for chlorphenoxamine hydrochloride, caffeine, chlorphenoxamine N-Oxide and theophylline, respectively. Chlorphenoxamine N-Oxide, being an impurity of chlorphenoxamine was prepared by refluxing intact drug with 5% H 2 O 2 for 24 h at 100 °C. One of the objectives of the analytical community is to promote the adoption of green analysis methods, which involve the development of environmentally friendly techniques. The levels of greenness and whiteness were evaluated using four specific tools: Eco-Scale System, GAPI, AGREE, and RGB tool. Furthermore, we have evaluated the greenness of the analytical method presented and compared its performance and greenness to that of the approach described in the literature. In this study, results from CPX and CAF analysis were compared to those obtained in a previous study. The result shows that there is no notable variation in precision and accuracy. The proposed method was validated in accordance with the requirements of ICH.

A stability indicating RP-HPLC-UV assay method for the simultaneous determination of hydroquinone, tretinoin, hydrocortisone, butylated hydroxytoluene and parabens in pharmaceutical creams

Abstract:Multicomponent drugs are medications that combine two or more active pharmaceutical ingredients in a single dosage form. These dosage forms improve the patient compliance, reduce the risk of drug interactions, and simplify dosing regimens. However, quality control of these multicomponent dosage forms can be challenging, especially if the final product contains four or more ingredients that are active (comprise stabilizers, preservatives, excipients, and other components). This problem can be more pronounced if the excipients can interfere with the analysis. In this work, a stability indicating assay method was developed and validated (according to the ICH International Guidelines) for the simultaneous determination of hydroquinone (HQ), tretinoin (TRT), hydrocortisone (HCA), butylated hydroxytoluene (BHT), methyl paraben (MP) and propyl paraben (PP) in commercially available pharmaceutical creams. The proposed method is based on gradient elution using X-Bridge C18 (150 × 4.6 mm, 5μm) column with a flow rate of 1 mL/min. The linear ranges (μg/mL) were 240-560 for HQ, 24-56 for MP, 132-308 for HCA, 6-14 for PP, 12-28 for BHT, 6.6-15 for TRT. During the validation process, the intra- and interday precision and trueness (evaluated as recovery) were found to be below 2.0% and between 100–102%, respectively. System suitability tests (SST) allow validating the herein proposed procedure specifically for pharmaceutical and industrial applications. SST test shows that the reported procedure fulfill with the Guidelines, allowing excellent separation of the analytes with very sensitive, accurate (precise and true) and reproducible quantitation of each analytes. The method was successfully applied in forced degradation studies of the six analytes. Specifically, acid degradation slightly affected HCA and BHT (91% recovery), while alkaline degradation drastically reduced HCA recovery (5.5%) and moderately affected BHT (85%). Photodegradation primarily influenced TRT quantity, and oxidative degradation intensified the BHT peak (130%).

Abstract:The present study discusses the development of simple, rapid, specific, precision, accuracy, stability indicating the HPLC method for the analysis of amlodipine besylate and valsartan tablet dosage form. The chromatographic separation was achieved using phosphate buffer with 1% triethyl amine (pH 3.0) as mobile phase‐A and mixed Methanol and buffer in the ratio of (65:35)(v/v) as mobile phase‐B. The detection of components was made at 237 nm for amlodipine besylate and valsartan. Analytical techniques should enrich sensitivity and specificity for the estimation of pharmaceutical drug products. Evaluated stress studies under different types of ICH conditions. The optimized HPLC method was validated as per the current ICH guidelines. The validated HPLC method was obtained highly specific with linearity ranging between 25 and 200 μgmL−1 of amlodipine besylate and 40–320 μgmL−1 of valsartan and both components correlation coefficient was > 0.999. The method showed high accuracy more than 97%. In stress studies, amlodipine besylate and valsartan were found to be sensitive to acid stress conditions and oxidation stress conditions. The method was found to be suitable for the quality control of amlodipine besylate and valsartan in the tablet as well as in stability‐indicating studies. The method was applied to the analysis of stability samples.

Abstract:The Current study aimed to develop and validate a reliable Reverse Phase High- Performance Liquid Chromatography (RP-HPLC) method for the quantification of Labetalol Hydrochloride and its related substances by using design of Experiments (DoE). The mobile phase A consists of Phosphate buffer (pH 3.7) (100 %) and the mobile phase B consist of Acetonitrile (100 %). The UV detector wavelength was 230 nm. The total run time was 60 minutes and flow rate was 0.8ml/min. Various parameters such as linearity, precision, accuracy, specificity, robustness, ruggedness, solution stability, Forced degradation, limit of detection, limit of quantification and system suitability were thoroughly evaluated according to USP guidelines. The method exhibited excellent linearity over the concentration range of 0.000258 to 0.003096 mg/ml with a correlation coefficient (r2) of 0.999, The Method precision ( % RSD) were found to be within acceptable limits. Accuracy studies revealed that the method was accurate with recovery values within the range of 99.6 % to 102.5 %. The method demonstrated high specificity, as indicated by the resolution of Labetalol Hydrochloride peak from potential impurities. Robustness and Ruggedness testing confirmed the method ability to produce consistent results under slight variations in chromatographic conditions. Furthermore, system suitability parameters such as resolution, tailing factor, and theoretical plates met the acceptance criteria, ensuring the method suitability for routine analysis. Overall, the developed RP-HPLC method proved to be accurate, precise, specific, and robust for the quantitative determination of Labetalol Hydrochloride in pharmaceutical formulations, thereby offering a valuable tool for quality control in pharmaceutical industries.
