Abstract:Lignocellulose waste was served as promising raw material for bioethanol production. Bioethanol was considered to be a potential alternative energy to take the place of fossil fuels. Lignocellulosic biomass synthesized by plants is regenerative, sufficient and cheap source for bioethanol production. The biotransformation of lignocellulose could exhibit dual significance-reduction of pollution and obtaining of energy. Some strategies are being developing and increasing the utilization of lignocellulose waste to produce ethanol. New technology of bioethanol production from natural lignocellulosic biomass is required. In this paper, the progress in genetic manipulation strategies including gene editing and synthetic genomics for the transformation from lignocellulose to ethanol was reviewed. At last, the application prospect of bioethanol was introduced.

Abstract:This review paper examines acid and alkaline pretreatments on perennial grasses for second-generation (2G) bioethanol production, a relatively unexplored area in this field. It compares the efficiency of these pretreatments in producing fermentable sugar and bioethanol yield. This study finds that alkaline pretreatment is more effective than acidic pretreatment in removing lignin and increasing sugar yield, leading to higher ethanol yields. However, it is costlier and requires longer reaction times than acidic pretreatment, while acidic pretreatment often leads to the formation of inhibitory compounds at higher temperatures, which is undesirable. The economic and environmental impacts of lignocellulosic biomass (LCB) are also assessed. It is revealed that LCB has a lower carbon but higher water footprint and significant costs due to pretreatment compared to first-generation biofuels. This review further explores artificial intelligence (AI) and advanced technologies in optimizing bioethanol production and identified the gap in literature regarding their application to pretreatment of perennial grasses. This review concludes that although perennial grasses hold promise for 2G bioethanol, the high costs and environmental challenges associated with LCB necessitate further research. This research should focus on integrating AI to optimize the pretreatment of LCB, thereby improving efficiency and sustainability in 2G biofuel production.

Abstract:Pennisetum purpureum is an easily cultivated fast-growing perennial grass with great potential as a bioenergy crop due to its high biomass production. Recent advancements in the processing of P. purpureum for the generation of bioethanol are highlighted in this review paper. Various pretreatment methods, namely, enzymatic, acid, and alkaline pretreatment have been investigated to enhance the efficiency of cellulose hydrolysis, which is a crucial step in bioethanol production. Moreover, genetic engineering approaches for enhancing the biomass yield and reducing the lignin are being discussed in this review. The production of bioethanol can be increased by several fermentation processes, including saccharification, separated hydrolysis, simultaneous saccharification, and fermentation. This comprehensive review explores recent advancements in bioethanol production, encompassing technological innovations, diversification of feedstock sources, genetic engineering methodologies, inventive pretreatment techniques, refinements in enzymatic hydrolysis, and enhancements in fermentation, ultimately highlighting the potential of P. purpureum as a sustainable bioethanol source while addressing the challenges and opportunities in its commercial-scale production.

Abstract:Grass straw represents an attractive and renewable resource for the production of value-added products like bioethanol. However, due to its recalcitrant structure and heterogeneity, efficient biodegradation of the grass straw remains a challenge for biorefinery. In view of this objective, the efficiency of the bacterial consortium was evaluated on grass straw pretreated with mild alkaline acid solutions. The results revealed that the pretreatment of the grass straw significantly enhanced the saccharification efficiency of the bacterial consortium. The Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) analyses revealed that chemical pretreatment altered the structure of the grass straw, removing the majority of the lignin and hydrogen bonding between cellulose and hemicellulose. The field emission scanning electron microscopy (FESEM) showed an altered surface of the pretreated grass straw with pores and tunnels. During saccharification, the highest reducing sugars (224.33 ± 1.15 μg/mL) were released for 3% grass straw loadings using 2.5% of the bacterial consortium after 14 days of incubation, signifying the synergism of bacterial strains for lignocellulose bioconversion. Subsequently, the highest ethanol yield of 3.59 ± 0.85% (v/v) was attained at pH 5.5 on the 4th day of fermentation by Saccharomyces cerevisiae using 8% of yeast culture/inoculum. The overall obtained results highlight the significance of the cellulolytic bacterial consortium towards lignocellulosic biorefinery by secreting grass straw hydrolyzing cellulases and hemicellulases, signifying its valorization as feedstock as well as ethanol fermentation.

Abstract:Lignocellulosic biomass generated from different sectors (agriculture, forestry, industrial) act as biorefinery precursor for production of second-generation (2G) bioethanol and other biochemicals. The integration of various conversion techniques on a single platform under biorefinery approach for production of biofuel and industrially important chemicals from LCB is gaining interest worldwide. The waste generated on utilization of bio-resources is almost negligible or zero in a biorefinery along with reduced greenhouse gas emissions, which supports the circular bioeconomy concept. The economic viability of a lignocellulosic biorefinery depends upon the efficient utilization of three major components of LCB—cellulose, hemicellulose and lignin. The heterogeneous structure and recalcitrant nature of LCB is main obstacle in its valorization into bioethanol and other value-added products. The success of bioconversion process depends upon methods used during pre-treatment, hydrolysis and fermentation processes. The cost involved in each step of the bioconversion process affects the viability of cellulosic ethanol. The lignocellulose biorefinery has ample scope, but much-focused research is required to fully utilize major parts of lignocellulosic biomass with zero wastage. The present review entails lignocellulosic biomass valorization for ethanol production, along with different steps involved in its production. Various value-added products produced from LCB components were also discussed. Recent technological advances and significant challenges in bioethanol production are also highlighted in addition to future perspectives.Graphical abstract

Abstract:The sustained increase in global energy needs and environmental impacts from greenhouse gases emitted by burning non-renewable fuels has led to a focus on developing renewable energy sources. Bioethanol can meet future energy demands of nations. Lignocellulosic feedstock and algal biomass are considered effective sources of substrates for converting fermentable sugars into ethanol. The structural configuration of lignin-based cellulosic biomass is modified using integrated pretreatment technologies, including physiochemical and biological methods. The hydrolysis of cellulose with cellulase enzymes consists of cellulase adsorption to the cellulosic surface, disintegration of cellulose polymers into simple sugar compounds, and desorption of cellulase. Soybean straw fermented in Thermoanaerobacterium aotearoense at 55 °C and 15 g/L total sugars provided 0.34 g/g ethanol yield. Rice straw feedstock hydrolyzed using cellulase enzymes at 30 °C, 74 h, and pH 4.5 yielded 0.44 g/g ethanol. The macroalgae species Ulva rigida in solid-state fermentation using Saccharomyces cerevisiae at 37 °C, pH 5, and 3 h yielded 330 mg/g glucose. The microalga Chlorella vulgaris hydrolyzed at 45 °C, pH 6, and 48 h using amylases provided a 3.8 g/L ethanol yield. Different fermentation processes for converting reduced sugars into ethanol include submerged, solid-state, and very high-gravity fermentation.

Abstract:<p>Plant cell walls contain the most abundant sustainable biomass resource for biofuels and biomaterials on the earth. However, lignocellulose recalcitrance generally requires a costly biomass process unacceptable for large-scale bioethanol production with the potential formation of secondary wastes. To address this bottleneck-like biomass recalcitrance issue, this review attempts to connect recent innovation progress regrading up-stream lignocellulose modification, middle-stream cellulases production and down-stream biomass processing. Particularly, the site-specific gene editing is demonstrated for precise and mild modification of plant cell walls to generate recalcitrance-much-reduced cellulose nanofibers, which not only leads to little impact on plant strength and biomass yield, but also causes remarkably enhanced enzymatic saccharification in major bioenergy crops. By selecting the size-reduced cellulose nanofibers of engineered bioenergy crops as enzyme-inducing substrate, fungal strains are then engineered to secret the optimal cellulases enzymes cocktails enabled for complete enzymatic saccharification of diverse lignocellulose residues from cost-effective biomass pretreatments. Consequently, engineered yeast strains could use both hexoses and xylose released from complete saccharification as carbon sources for maximum bioethanol production by an efficient co-fermentation. Finally, the green-like processing technology is introduced to generate biomaterials and biochemicals by using the remaining lignin-rich residues. Therefore, this work has originally proposed a novel strategy that dynamically cascades the engineered bioenergy crops and fungal strains with the advanced biomass process technology, which should be considered as next generation of integrated biotechnology for both cost-effective biofuels production and value-added bioproducts with minimum waste releases into the environment.</p>

Abstract:Switchgrass is a promising feedstock for value-added applications due to its high productivity, potentially low requirements for agricultural inputs and positive environmental impacts. The objective of this paper is to review published research on the conversion of switchgrass into bioethanol and other value-added products. Environmental benefits associated with switchgrass include the potential for carbon sequestration, nutrient recovery from runoff, soil remediation and provision of habitats for grassland birds. Pretreatment of switchgrass is required to improve the yields of fermentable sugars. Based on the type of pretreatment, glucose yields range from 70% to 90% and xylose yields range from 70% to 100% after hydrolysis. Following pretreatment and hydrolysis, ethanol yields range from 72% to 92% of the theoretical maximum. Other value-added uses of switchgrass include gasification, bio-oil production, newsprint production and fiber reinforcement in thermoplastic composites. Future prospects for research include increased biomass yields, optimization of feedstock composition for bioenergy applications, and efficient pentose fermentation to improve ethanol yields.

Abstract:Grass raw materials collected from grasslands cover more than 30% of Europe's agricultural area. They are considered very attractive for the production of different biochemicals and biofuels due to their high availability and renewability. In this study, a perennial ryegrass ( Lolium perenne ) was exploited for second-generation bioethanol production. Grass press–cake and grass press-juice were separated using mechanical pretreatment, and the obtained juice was used as a fermentation medium. In this work, Saccharomyces cerevisiae was utilized for bioethanol production using the grass press-juice as the sole fermentation medium. The yeast was able to release about 11 g/L of ethanol in 72 h, with a total production yield of 0.38 ± 0.2 g Ethanol /g sugars . It was assessed to improve the fermentation ability of Saccharomyces cerevisiae by using the short-term adaptation. For this purpose, the yeast was initially propagated in increasing the concentration of press-juice. Then, the yeast cells were re-cultivated in 100%(v/v) fresh juice to verify if it had improved the fermentation efficiency. The fructose conversion increased from 79 to 90%, and the ethanol titers reached 18 g/L resulting in a final yield of 0.50 ± 0.06 g Ethanol /g sugars with a volumetric productivity of 0.44 ± 0.00 g/Lh. The overall results proved that short-term adaptation was successfully used to improve bioethanol production with S. cerevisiae using grass press-juice as fermentation medium.

Abstract:With the draining of petroleum derivatives, expanding natural contamination issues, there has been rising enthusiasm for the examination of lignocellulosic biomass for an alternative fsource of energy. Characterization of various biomass, its intermediate, and products is a need for conversion of any biomass to biofuels. Chemical composition of lignocellulosic biomass is an essential point for developing potent pretreatment technologies to break its rigid structure, conversion of sugar by different enzymes mainly cellulose to glucose and even various microorganisms which can ferment sugars into bioethanol and other value-added green chemicals. In this present review work, the main focus is on the proximate and ultimate analysis of different feedstocks, and altered pretreatment techniques such as physical, chemical, physicochemical, and biological methods for bioethanol production have been addressed, which ultimately will help in overcoming the recalcitrance of lignocellulosic biomass by degrading the lignin fraction, breaking down of lignocellulose components, hydrolysis, and fermentation process. Recently, combined pretreatment is gaining popularity as it is more favorable and profitable for improving chemical yield and process of enzymatic hydrolysis of LBs, but it increases the cost of operation. Acid pretreatment, steam explosion, and hydrothermal processes all together show a comparatively high effect on degrading hemicelluloses fraction. Alkali, oxidative, and organosolv pretreatment are more efficient in removing and degrading of lignin portion. This present study will empower a better idea and knowledge of the available process with the upcoming advanced processes which would help to overcome the limitations and establish technology to facilitate the pretreatment methods to make an authentic concept of biorefinery.

Abstract:This study presents a molecular inhibitory mechanism by Fas-associated factor 1 (FAF1) on IκB kinase (IKK) activation, where divergent NF-κB-activating stimuli converge. FAF1 interacts with IKKβ in response to proinflammatory stimuli (such as tumor necrosis factor-α, interleukin-1β, and lipopolysaccharide) and suppresses IKK activation. Interaction of the leucine-zipper domain of IKKβ with FAF1 affected the IKK heterocomplex (IKKα/β) and homocomplex (IKKα/α, IKKβ/β) formations and attenuated IKKγ recruitment to IKKβ. Overexpression of FAF1 reduced the level of IKKβ activity, whereas FAF1 depletion increased the activity. These results indicate that FAF1 inhibits IKK activation and its downstream signaling by interrupting the IKK complex assembly through physical interaction with IKKβ. Taken together, FAF1 robustly suppresses NF-κB activation through the inhibition of IKK activation in combination with previously reported cytoplasmic retention of NF-κB p65 (Park, M. Y., Jang, H. D., Lee, S. Y., Lee, K. J., and Kim, E. (2004) J. Biol. Chem. 279, 2544–2549). Such redundant suppression would prevent inadvertent activation of the NF-κB pathway.

Abstract:The objective of present review is to provide a scientific overview of sugarcane as a potential feedstock for biofuel and use of genome editing approach for improvement of industrial and agronomical traits in sugarcane. Sugarcane, a perennial tropical grass with a high biomass index, is a promising feedstock for bioethanol production, and its bagasse, rich in lignocellulosic material, serves as an ideal feedstock for producing second-generation bioethanol. To improve the conversion of sugarcane biomass into biofuels, developing varieties with improved biomass degradability and high biomass and sucrose content is essential. The complex genome architecture and earlier lack of sequence data hindered biotechnological advancements in sugarcane, but recent genome sequence updates offer new opportunities for sugarcane improvement. The first genetically modified sugarcane was developed in 1992 by Bower and Birch using microprojectile bombardment of embryogenic callus. Since then, transgenic techniques have rapidly evolved, leading to the advancement of genome editing technologies. Application of genome editing tools particularly CRISPR/Cas system has been successfully used in sugarcane for editing. Recently, multiple alleles of the magnesium chelatase and acetolactate synthase genes in sugarcane have been successfully edited through multiplexing. Additionally, CRISPR-edited sugarcane varieties with modified cell wall components and increased sucrose content for enhanced bioethanol production have been developed. At the end, the future of CRISPR-edited crops will depend on how well regulatory frameworks adapt to the rapidly evolving technology.

Cellulosic ethanol production: Progress, challenges and strategies for solutions

Abstract:Lignocellulosic biomass is a sustainable feedstock for fuel ethanol production, but it is characterized by low mass and energy densities, and distributed production with relatively small scales is more suitable for cellulosic ethanol, which can better balance cost for the feedstock logistics. Lignocellulosic biomass is recalcitrant to degradation, and pretreatment is needed, but more efficient pretreatment technologies should be developed based on an in-depth understanding of its biosynthesis and regulation for engineering plant cell walls with less recalcitrance. Simultaneous saccharification and co-fermentation has been developed for cellulosic ethanol production, but the concept has been mistakenly defined, since the saccharification and co-fermentation are by no means simultaneous. Lignin is unreactive, which not only occupies reactor spaces during the enzymatic hydrolysis of the cellulose component and ethanol fermentation thereafter, but also requires extra mixing, making high solid loading difficult for lignocellulosic biomass and ethanol titers substantially compromised, which consequently increases energy consumption for ethanol distillation and stillage discharge, presenting another challenge for cellulosic ethanol production. Pentose sugars released from the hydrolysis of hemicelluloses are not fermentable with Saccharomyces cerevisiae used for ethanol production from sugar- and starch-based feedstocks, and engineering the brewing yeast and other ethanologenic species such as Zymomonas mobilis with pentose metabolism has been performed within the past decades. However strategies for the simultaneous co-fermentation of pentose and hexose sugars that have been pursued overwhelmingly for strain development might be modified for robust ethanol production. Finally, unit integration and system optimization are needed to maximize economic and environmental benefits for cellulosic ethanol production. In this article, we critically reviewed updated progress, and highlighted challenges and strategies for solutions.

Abstract:Lignocellulosic biomass (LB) is a promising sugar feedstock for biofuels and other high-value chemical commodities. The recalcitrance of LB, however, impedes carbohydrate accessibility and its conversion into commercially significant products. Two important factors for the overall economization of biofuel production is LB pretreatment to liberate fermentable sugars followed by conversion into ethanol. Sustainable biofuel production must overcome issues such as minimizing water and energy usage, reducing chemical usage and process intensification. Amongst available pretreatment methods, microorganism-mediated pretreatments are the safest, green, and sustainable. Native biodelignifying agents such as Phanerochaete chrysosporium, Pycnoporous cinnabarinus, Ceriporiopsis subvermispora and Cyathus stercoreus can remove lignin, making the remaining substrates amenable for saccharification. The development of a robust, integrated bioprocessing (IBP) approach for economic ethanol production would incorporate all essential steps including pretreatment, cellulase production, enzyme hydrolysis and fermentation of the released sugars into ethanol. IBP represents an inexpensive, environmentally friendly, low energy and low capital approach for second-generation ethanol production. This paper reviews the advancements in microbial-assisted pretreatment for the delignification of lignocellulosic substrates, system metabolic engineering for biorefineries and highlights the possibilities of process integration for sustainable and economic ethanol production.

Abstract:This paper investigates bioethanol production from switchgrass, focusing on enhancement of efficiency through various pretreatment methods and comparing two bioethanol production processes: simultaneous saccharification and fermentation (SSF) and separate hydrolysis and fermentation (SHF). Physical, chemical, and biological pretreatment processes are applied to enhance the breakdown of switchgrass's lignocellulosic structure. Effects of pretreatments, enzymatic hydrolysis, and fermentation on ethanol yield are discussed in detail. The comparative analysis reveals that SSF yields higher ethanol outputs within shorter times by integrating hydrolysis and fermentation into a single process. In contrast, SHF offers more control by separating these stages. The comparative analysis highlights that SSF achieves higher ethanol yields more efficiently, although it might restrict SHF's operational flexibility. This study aims to provide a comprehensive overview of the current pretreatments, hydrolysis methods, and fermentation processes in bioethanol production from switchgrass, offering insights into their scalability, economic viability, and potential environmental benefits. The findings are expected to contribute to the ongoing discussions and developments in renewable bioenergy solutions, supporting advancing more sustainable and efficient bioethanol production techniques.

Abstract:Abstract Lignocellulose is the most abundant biomass on earth with an annual production of about 2 × 10 11 tons. It is an inedible renewable carbonaceous resource that is very rich in pentose and hexose sugars. The ability of microorganisms to use lignocellulosic sugars can be exploited for the production of biofuels and chemicals, and their concurrent biotechnological processes could advantageously replace petrochemicals’ processes in a medium to long term, sustaining the emerging of a new economy based on bio-based products from renewable carbon sources. One of the major issues to reach this objective is to rewire the microbial metabolism to optimally configure conversion of these lignocellulosic-derived sugars into bio-based products in a sustainable and competitive manner. Systems’ metabolic engineering encompassing synthetic biology and evolutionary engineering appears to be the most promising scientific and technological approaches to meet this challenge. In this review, we examine the most recent advances and strategies to redesign natural and to implement non-natural pathways in microbial metabolic framework for the assimilation and conversion of pentose and hexose sugars derived from lignocellulosic material into industrial relevant chemical compounds leading to maximal yield, titer and productivity. These include glycolic, glutaric, mesaconic and 3,4-dihydroxybutyric acid as organic acids, monoethylene glycol, 1,4-butanediol and 1,2,4-butanetriol, as alcohols. We also discuss the big challenges that still remain to enable microbial processes to become industrially attractive and economically profitable.

Abstract:Bioenergy production from lignocellulosic biomass waste is among the most promising options to produce sustainable economic fuel in an eco-friendly way. This review is an attempt to use and explore Saccharum spontaneum grass as a potential lignocellulosic biomass waste to produce bioenergy. Many advantages, like high cellulose and hemicellulose content, renewable nature, huge availability, and non-participation in the food chain, make this biomass a waste of immense interest. Nevertheless, existing challenges and limitations need to be addressed in view of sustainable bioenergy production from this biomass. In this review, an overview of the utilization of waste Saccharum spontaneum grass as a potential feedstock for developing bioenergy has been provided in view of its physicochemical properties, which are fascinating from the perspective of renewable energy production. Furthermore, advanced and lower-cost strategies are being discussed, along with the possible challenges found in pretreatment, bioconversion, and bioenergy production using lignocellulosic biomass. In view of available research studies on the utilization of Saccharum spontaneum grass to produce bioenergy, numerous processes and key strategies have been discussed and evaluated with those of different LCBs for the production of renewable energies. Additionally, existing limitations and future scope, along with possible solutions in the process of producing green energy, have been discussed.

Abstract:tumor proliferation through the phosphoinositide 3-kinase-mammalian target of rapamycin (mTOR) pathway. Interestingly, the cultured HGG lines demonstrated increased expression of NLGN-3 when exposed to conditioned medium, implicating a feed-forward mechanism of expression. This further explains the robust proliferation seen in the experimental setting and allows greater rationale of targeting this pathway therapeutically. The clinical implications of this discovery were made clear with a query of The Cancer Genome Atlas.Although somaticmutations ofNLGN3 were infrequent in pediatric and adult brain tumors, mRNA expression was found to be inversely correlated with overall patient survival in 429 cases, with a hazard ratio for death with high vs low expression of 1.31 (95% confidence interval, 1.051.63). The specificity of the aforementioned results was strengthened by examination of the role of Nlgn-2, which did not demonstrate increased proliferation or survival differences. The implication of these findings, that the inherent physiological function of the host organ could be taken advantage of by a primary tumor, has far-reaching impact. Neuronal activity can induce tumor proliferation via the mTOR pathway, whereas the mTOR pathway has been implicated in the pathogenesis of a number of human epilepsies. Thus, brain activity may promote tumor growth, which in turn may induce pathological brain activity in a vicious cycle. However, this discovery also may provide a targetable checkpoint in glioma proliferation and pathogenesis. It adds a missing element to the glioma microenvironmental landscape and allows a new avenue for exploring diagnostic and therapeutic possibilities. Although a sobering finding regarding this devastating diagnosis, the neuronal contribution to the pathogenesis of HGG is critical to characterize and dissect so that it can be translated to the clinical treatment of glioma patients.
