Abstract:Currently major part of the world’s energy requirements is fulfilled by fossil resources. Severe economic and environmental problems along with their limited resources are still a major issue. Long-term environmental and commercial concern has focussed into huge research in the past decades to replace fossil fuels with renewable sources of energy. The main focus of energy research is lignocellulosic substrates, which seems to be the best for energy application due to easily availability, cost-effective and environment favourable production process. Therefore, lignocellulosic materials provide economical, environmental and energetic benefits with an alternative to traditional or high cost substrates. However, the close association of major constituents of lignocellulosic biomass forms different barriers, which hinders the enzymatic hydrolysis of cellulose and hemicellulose to different fermentable sugars. To overcome this problem, different pretreatment approaches including physical, chemical, biological and combination of them have been used to improve the enzymatic accessibility or digestibility of lignocellulosic biomass. Various pretreatment methods have specific effect on different lignocellulosic components. Among all the methods, biological methods have performed better with respect to yield, cost and environmental related problems. Pretreated substrates from various pretreatments have been saccharified for the production of sustainable and renewable biofuels like ethanol, butanol, hydrogen, acetoin and 2,3-butanediol.

Abstract:With the draining of petroleum derivatives, expanding natural contamination issues, there has been rising enthusiasm for the examination of lignocellulosic biomass for an alternative fsource of energy. Characterization of various biomass, its intermediate, and products is a need for conversion of any biomass to biofuels. Chemical composition of lignocellulosic biomass is an essential point for developing potent pretreatment technologies to break its rigid structure, conversion of sugar by different enzymes mainly cellulose to glucose and even various microorganisms which can ferment sugars into bioethanol and other value-added green chemicals. In this present review work, the main focus is on the proximate and ultimate analysis of different feedstocks, and altered pretreatment techniques such as physical, chemical, physicochemical, and biological methods for bioethanol production have been addressed, which ultimately will help in overcoming the recalcitrance of lignocellulosic biomass by degrading the lignin fraction, breaking down of lignocellulose components, hydrolysis, and fermentation process. Recently, combined pretreatment is gaining popularity as it is more favorable and profitable for improving chemical yield and process of enzymatic hydrolysis of LBs, but it increases the cost of operation. Acid pretreatment, steam explosion, and hydrothermal processes all together show a comparatively high effect on degrading hemicelluloses fraction. Alkali, oxidative, and organosolv pretreatment are more efficient in removing and degrading of lignin portion. This present study will empower a better idea and knowledge of the available process with the upcoming advanced processes which would help to overcome the limitations and establish technology to facilitate the pretreatment methods to make an authentic concept of biorefinery.

Abstract:Lignocellulosic biomass (LCB) is plentifully and naturally available carbon source produced mostly from agro, food and food processing industries with a global estimate of 1.3 billion tonnes per year. Since LCB is inexpensive and considered as waste, it opens an avenue for cost-effective alternate source of energy. Lignocellulosic biomass can be effectively and efficiently converted to biofuels (e.g. bioethanol, biodiesel and biogas) and value-added products like organic acids, enzymes, biopolymers, biochar etc. However, the bottleneck in using lignocellulosic biomass on industrial scale is its structural complexity and recalcitrance nature. Thus, pretreatment of biomass is an essential step for efficient delignification of biomass. This process separates cellulose and hemicellulose from lignin of the complex polymer matrix. Thereby, reduces the size of the matrix and increases the surface area of cellulose and hemicelluloses to be accessible for enzymes and microbes for hydrolysis and fermentation respectively. The pretreatment process includes physical, chemical, physicochemical or biological. Mechanical milling, ultrasound and microwave radiation as physical; Acid/alkaline hydrolysis, organosolv, ionic liquids and ozonolysis as chemical; ammonia fiber explosion, CO2 explosion steam explosion, liquid hot water treatment as physicochemical methods are established. The use of certain species of bacteria, fungus and yeast in biological methods of pretreatment is yet to establish on large scale.

Abstract:Lignocellulosic biomass (LB) is a promising sugar feedstock for biofuels and other high-value chemical commodities. The recalcitrance of LB, however, impedes carbohydrate accessibility and its conversion into commercially significant products. Two important factors for the overall economization of biofuel production is LB pretreatment to liberate fermentable sugars followed by conversion into ethanol. Sustainable biofuel production must overcome issues such as minimizing water and energy usage, reducing chemical usage and process intensification. Amongst available pretreatment methods, microorganism-mediated pretreatments are the safest, green, and sustainable. Native biodelignifying agents such as Phanerochaete chrysosporium, Pycnoporous cinnabarinus, Ceriporiopsis subvermispora and Cyathus stercoreus can remove lignin, making the remaining substrates amenable for saccharification. The development of a robust, integrated bioprocessing (IBP) approach for economic ethanol production would incorporate all essential steps including pretreatment, cellulase production, enzyme hydrolysis and fermentation of the released sugars into ethanol. IBP represents an inexpensive, environmentally friendly, low energy and low capital approach for second-generation ethanol production. This paper reviews the advancements in microbial-assisted pretreatment for the delignification of lignocellulosic substrates, system metabolic engineering for biorefineries and highlights the possibilities of process integration for sustainable and economic ethanol production.

Abstract:<p>Biomass as a resource is present in large quantities and offers environmental benefits from sequestration of carbon to bioenergy production. Production of renewable and sustainable biofuel (bioethanol) from biomass is gaining overwhelming interest globally. Biofuels are important and beneficial from environmental and economic points of view. The gasohol, ethanol blended gasoline, utilization for the transportation sector significantly reduces greenhouse gas (GHG) emissions as well as consumption of fossil fuels. Bioethanol produced from renewable biomass especially lignocellulosic biomass (LCB) provides opportunities for a sustainable, cleaner, environmentally safe, carbon-neutral fuel, and green alternatives for fossil fuel. Although LCB is present in the massive amounts on planet earth, it is not utilized properly for bioethanol production due to many hurdles in the bioconversion process like pretreatment, high cost of enzymes, mixed sugar conversion, fermentation etc. Great efforts have been devoted in technological improvement in the bioconversion process including biotechnological and metabolic engineering fields. This review focuses on the current status, technological advances, and new interventions in the bioconversion process of LCB into bioethanol with special focus on pre-treatment methods, fermentation approaches, and detoxification processes.</p>

Abstract:The most abundant natural renewable resource in the world, lignocellulosic biomass (LCB), has the potential to be exploited as a substitute green feedstock for the synthesis of various chemicals, materials, and biofuels. The annual global production of 13 billion tonnes of LCB offers an opportunity to cater to the increasing energy and materials requirement of process industries and also restricts the discharge of greenhouse gases. Although LCB is enriched with valuable ingredients such as cellulose, lignin, and hemicellulose, its recalcitrant nature limits its efficient utilisation. These components of LCB are strongly interlinked with each other, which resists their isolation and conversion valorisation into useful products. To disrupt the complicated structure of LCB and to isolate the lignocellulosic components in pure form, pretreatment is a crucial process in the bio-refinery, ensuring the economic feasibility of downstream processes. This review provides an outline of the structure, composition, and various sources of LCB; and the necessity of the pretreatment. Moreover, this article provides an in-depth analysis of the underlying mechanisms, advantages, and limitations of various pretreatment methods, such as physical, chemical, biological, and physicochemical. Further, the impact of chemical pretreatment techniques on the physicochemical characteristics of the material that is extracted from the biomass is also covered in detail through the rigorous evaluation of performance metrics, including substrate digestibility, sugar yield, inhibitor production, and energy requirements. This review provides a balanced and comprehensive overview of the state-of-the-art pretreatment strategies and their impact on biomass valorisation that will be useful to the scientists, engineers, and policy makers interested in biomass conversion technologies.

Abstract:Lignocellulosic materials, such as forest, agriculture, and agroindustrial residues, are among the most important resources for biorefineries to provide fuels, chemicals, and materials in such a way to substitute for, at least in part, the role of petrochemistry in modern society. Most of these sustainable biorefinery products can be produced from plant polysaccharides (glucans, hemicelluloses, starch, and pectic materials) and lignin. In this scenario, cellulosic ethanol has been considered for decades as one of the most promising alternatives to mitigate fossil fuel dependence and carbon dioxide accumulation in the atmosphere. However, a pretreatment method is required to overcome the physical and chemical barriers that exist in the lignin-carbohydrate composite and to render most, if not all, of the plant cell wall components easily available for conversion into valuable products, including the fuel ethanol. Hence, pretreatment is a key step for an economically viable biorefinery. Successful pretreatment method must lead to partial or total separation of the lignocellulosic components, increasing the accessibility of holocellulose to enzymatic hydrolysis with the least inhibitory compounds being released for subsequent steps of enzymatic hydrolysis and fermentation. Each pretreatment technology has a different specificity against both carbohydrates and lignin and may or may not be efficient for different types of biomasses. Furthermore, it is also desirable to develop pretreatment methods with chemicals that are greener and effluent streams that have a lower impact on the environment. This paper provides an overview of the most important pretreatment methods available, including those that are based on the use of green solvents (supercritical fluids and ionic liquids).

Abstract:An upsurge in global population and rapid urbanization has accelerated huge dependence on petroleum-derived fuels and consequent environmental concerns owing to greenhouse gas emissions in the atmosphere. An integrated biorefinery uses lignocellulosic feedstock as raw material for the production of renewable biofuels, and other fine chemicals. The sustainable bio-economy and the biorefinery industry would benefit greatly from the effective use of lignocellulosic biomass obtained from agricultural feedstocks to replace petrochemical products. Lignin, cellulose, hemicellulose, and other extractives, which are essential components of lignocellulosic biomass, must be separated or upgraded into useful forms in order to fully realize the potential of biorefinery. The development of low-cost and green pretreatment technologies with effective biomass deconstruction potential is imperative for an efficient bioprocess. The abundance of microorganisms along with their continuous production of various degradative enzymes makes them suited for the environmentally friendly bioconversion of agro-industrial wastes into viable bioproducts. The present review highlights the concept of biorefinery, lignocellulosic biomass, and its valorization by green pretreatment strategies into biofuels and other biochemicals. The major barriers and challenges in bioconversion technologies, environmental sustainability of the bioproducts, and promising solutions to alleviate those bottlenecks are also summarized.

Abstract:As the global demand for sustainable energy increases, lignocellulosic (such as agricultural residues, forest biomass, municipal waste, and dedicated energy crops) and algal (including macroalgae and microalgae) biomass have attracted considerable attention, because of their high availability of carbohydrates. This is a potential feedstock to produce biochemical and bioenergy. Pretreatment of biomass can disrupt their complex structure, increasing conversion efficiency and product yield. Therefore, this review comprehensively discusses recent advances in different pretreatments (physical, chemical, physicochemical, and biological pretreatments) for lignocellulosic and algal biomass and their biorefining methods. Life cycle assessment (LCA) which enables the quantification of the environmental impact assessment of a biorefinery also be introduced. Biorefinery processes such as raw material acquisition, extraction, production, waste accumulation, and waste conversion are all monitored under this concept. Nevertheless, there still exist some techno-economic barriers during biorefinery and extensive research is still needed to develop cost-effective processes.

Abstract:Biogas production from agricultural residues represents an effective and sustainable option for handling vast quantities of lignocellulosic waste for meeting the global energy demand. However, the recalcitrance of lignocellulosic biomass due to the presence of cellulose and hemicellulose in a structurally complex lignocellulosic matrix, and the crystallinity of cellulose create a hindrance in biogas production by restricting the availability of fermentable sugars to microbial action. This paper assesses the different pretreatment strategies adopted for making the lignocellulosic biomass amenable to anaerobic digestion by altering the structure of lignocellulose and eliminating lignin, thus increasing the accessibility of microbes to the easily degradable components. The review highlights the advantages and limitations of each technology—physical for size reduction (chipping, milling, extrusion, cavitation), thermal for breaking down of hydrogen bonds (conventional heating, steam explosion, microwave irradiation, hydrothermal), chemical for decreasing the crystallinity and polymerization degree of cellulose (alkalis, acids, gases, oxidizing agents, various solvents), biological for enhancing the digestibility (microbial, enzymatic), and their effect on improving the degradation efficiency and biogas yield. Further, the review discusses the role of some emerging technologies and other less explored options which may require optimization at higher scale so that the confidence in the translation of this knowledge can be increased and this abundantly available raw material can be effectively utilized. With the goal of improving the applicability of pretreatment technologies, some recommendations are put forth so that the efficiency of anaerobic digestion can be increased and the development of pretreatment technologies can be promoted on a large scale.

Abstract:Agricultural residue is produced in large quantities during crop harvesting, and open burning of this waste causes environmental pollution and health risks. Due to the structural complexity of the lignocellulose and problems associated with physical and chemical methods of its pretreatments, there is an utmost need for an eco-friendly pretreatment strategy. Biological pretreatment involving microorganisms and their enzymes is an environment-benign and economic process due to lack of release or requirement of toxic chemicals during the process. Among microorganisms, filamentous fungi (mainly Basidiomycetes) with efficient enzymatic machinery have been used in efficient delignification and bioconversion of lignocellulosic biomass. Enzyme-mediated pretreatment has further improved the saccharification of plant biomass with no sugar loss as in case of microbial pretreatment. Composting, ensiling, solid state fermentation, and biogas production are based on biological pretreatment, which are used for the generation of value-added products. Biological pretreatment does not require/release toxic chemicals but, is highly useful in detoxification of such toxic compounds. Biological pretreatment is significantly affected by biotic and abiotic factors. Pretreated biomass is hydrolyzed by cellulases and xylanases into sugars that are fermented into biofuels, organic acids, enzymes, and other products. The slow and long incubation nature of biological pretreatment has been overcome by combining with milder physico-chemical methods. Furthermore, the consolidated bioprocessing-based biorefinery approach has enhanced the potential of biological pretreatment by involving microbial consortium for the production of biofuels and other value-added products in a single step. Therefore, biological pretreatment-based biorefinery approach would be quite beneficial for the large-scale production of value-added products from lignocellulose with concomitant reduction in environmental pollution and solid waste management.

Abstract:Lignocellulosic biomass is one of the potential feedstocks to produce second-generation cellulosic ethanol and biochemicals. To enhance the enzymatic digestibility of lignocellulosic biomass for efficient enzymatic saccharification, a variety of pretreatment methods have been studied. Among these, organosolv pretreatment using ethanol is a promising pretreatment method owing to its inherent advantages, such as low solvent cost, lack of toxicity, the ability to retain most cellulose fraction in substrates for enzymatic hydrolysis, coproduction of high-purity lignin and hemicellulosic sugars, easy solvent recovery, and reuse. In this review, the research progress regarding different types of ethanol organosolv pretreatment processes has been summarized in terms of methods, substrate properties, reaction mechanisms, delignification kinetic as well as the impact of pretreatment methods on the enzymatic digestibility. Attempts are also made to provide insights into the complete utilization of lignocellulosic biomass to achieve high potential revenues. Though some ethanol organosolv processes have been studied or are being developed towards commercialization, ethanol organosolv pretreatment is still facing some challenges. Finally, the direction for future work is given to develop a proper ethanol organosolv pretreatment for commercialization.

Abstract:Lignocellulosic biomass has been recognized as promising feedstock for biofuels production. However, the high cost of pretreatment is one of the major challenges hindering large-scale production of biofuels from these abundant, indigenously-available, and economic feedstock. In addition to high capital and operation cost, high water consumption is also regarded as a challenge unfavorably affecting the pretreatment performance. In the present review, advances in lignocellulose pretreatment technologies for biofuels production are reviewed and critically discussed. Moreover, the challenges faced and future research needs are addressed especially in optimization of operating parameters and assessment of total cost of biofuel production from lignocellulose biomass at large scale by using different pretreatment methods. Such information would pave the way for industrial-scale lignocellulosic biofuels production. Overall, it is important to ensure that throughout lignocellulosic bioethanol production processes, favorable features such as maximal energy saving, waste recycling, wastewater recycling, recovery of materials, and biorefinery approach are considered.

Abstract:Lignocellulosic biomass is an abundant and renewable resource that potentially contains large amounts of energy. It is an interesting alternative for fossil fuels, allowing the production of biofuels and other organic compounds. In this paper, a review devoted to the processing of lignocellulosic materials as substrates for fermentation processes is presented. The review focuses on physical, chemical, physicochemical, enzymatic, and microbiologic methods of biomass pretreatment. In addition to the evaluation of the mentioned methods, the aim of the paper is to understand the possibilities of the biomass pretreatment and their influence on the efficiency of biofuels and organic compounds production. The effects of different pretreatment methods on the lignocellulosic biomass structure are described along with a discussion of the benefits and drawbacks of each method, including the potential generation of inhibitory compounds for enzymatic hydrolysis, the effect on cellulose digestibility, the generation of compounds that are toxic for the environment, and energy and economic demand. The results of the investigations imply that only the stepwise pretreatment procedure may ensure effective fermentation of the lignocellulosic biomass. Pretreatment step is still a challenge for obtaining cost-effective and competitive technology for large-scale conversion of lignocellulosic biomass into fermentable sugars with low inhibitory concentration.

Abstract:To improve the competence of cellulose hydrolysis, pretreatment is required to efficiently break its recalcitrant structure. Pretreatment has been viewed as one of the most expensive processing steps in cellulosic biomass-to-fermentable sugars conversion, and it has great potential for improvement of efficiency and lowering of cost through research and development. Pretreatment is an important cost-driver of lignocellulose conversion to biofuel and an important step prior to enzyme hydrolysis. It disrupts the plant cell wall network and partially separates the major polymer components (lignin, cellulose, and hemicellulose). However, pretreatment of lignocellulosic materials may also result in the release of inhibitors and deactivators of the enzymatic hydrolysis of cellulose. Development of enzyme processes for hydrolysis of cellulose to glucose must reduce inhibition and deactivation effects in order to enhance hydrolysis and reduce enzyme usage. Therefore, great attentions have paid in designing pretreatment technologies to split recalcitrant characteristics of lignocellulose biomass.

Abstract:Pre-treatment of biomass is a necessary step for delignification as well as an unavoidable step in the journey of biomass to biofuel. Apart from delignification, defragmentation and partial depolymerization of biomass are two other additional useful steps in the overall process of pre-treatment. Ultimately these three steps together make the downstream enzymatic hydrolysis process more effective in producing optimum total reducing sugar. Several different pre-treatment technologies are available to delignify various types of biomasses with each of them being unique concerning specific advantages and disadvantages, characteristics of its own. However, different pre-treatment methods, in combination, have been found beneficial with higher delignification efficiency in comparison to a single pre-treatment method, implemented on its own. Apart from physical pre-treatment procedures like milling, grinding, steam explosion, etc., pre-treatment methods can be divided, roughly, into three groups: biological pre-treatment, physical pre-treatment and chemical pre-treatment. Chemical pre-treatments always arrest significantly higher attention due to their effectiveness and minimum treatment time required. In this review, a thorough discussion of various chemical and biological pre-treatment technologies, along with their efficacy and effective outcomes, have been discussed in order to understand their impacts on bioenergy industries.

Abstract:Lignocelluloses are often a major or sometimes the sole components of different waste streams from various industries, forestry, agriculture and municipalities. Hydrolysis of these materials is the first step for either digestion to biogas (methane) or fermentation to ethanol. However, enzymatic hydrolysis of lignocelluloses with no pretreatment is usually not so effective because of high stability of the materials to enzymatic or bacterial attacks. The present work is dedicated to reviewing the methods that have been studied for pretreatment of lignocellulosic wastes for conversion to ethanol or biogas. Effective parameters in pretreatment of lignocelluloses, such as crystallinity, accessible surface area, and protection by lignin and hemicellulose are described first. Then, several pretreatment methods are discussed and their effects on improvement in ethanol and/or biogas production are described. They include milling, irradiation, microwave, steam explosion, ammonia fiber explosion (AFEX), supercritical CO(2) and its explosion, alkaline hydrolysis, liquid hot-water pretreatment, organosolv processes, wet oxidation, ozonolysis, dilute-and concentrated-acid hydrolyses, and biological pretreatments.

Abstract:Overcoming the recalcitrance (resistance of plant cell walls to deconstruction) of lignocellulosic biomass is a key step in the production of fuels and chemicals. The recalcitrance is due to the highly crystalline structure of cellulose which is embedded in a matrix of polymers-lignin and hemicellulose. The main goal of pretreatment is to overcome this recalcitrance, to separate the cellulose from the matrix polymers, and to make it more accessible for enzymatic hydrolysis. Reports have shown that pretreatment can improve sugar yields to higher than 90% theoretical yield for biomass such as wood, grasses, and corn. This paper reviews different leading pretreatment technologies along with their latest developments and highlights their advantages and disadvantages with respect to subsequent hydrolysis and fermentation. The effects of different technologies on the components of biomass (cellulose, hemicellulose, and lignin) are also reviewed with a focus on how the treatment greatly enhances enzymatic cellulose digestibility.

Abstract:Abstract Population increase and industrialization has resulted in high energy demand and consumptions, and presently, fossil fuels are the major source of staple energy, supplying 80% of the entire consumption. This has contributed immensely to the greenhouse gas emission and leading to global warming, and as a result of this, there is a tremendous urgency to investigate and improve fresh and renewable energy sources worldwide. One of such renewable energy sources is biogas that is generated by anaerobic fermentation that uses different wastes such as agricultural residues, animal manure, and other organic wastes. During anaerobic digestion, hydrolysis of substrates is regarded as the most crucial stage in the process of biogas generation. However, this process is not always efficient because of the domineering stableness of substrates to enzymatic or bacteria assaults, but substrates’ pretreatment before biogas production will enhance biogas production. The principal objective of pretreatments is to ease the accessibility of the enzymes to the lignin, cellulose, and hemicellulose which leads to degradation of the substrates. Hence, the use of pretreatment for catalysis of lignocellulose substrates is beneficial for the production of cost-efficient and eco-friendly process. In this review, we discussed different pretreatment technologies of hydrolysis and their restrictions. The review has shown that different pretreatments have varying effects on lignin, cellulose, and hemicellulose degradation and biogas yield of different substrate and the choice of pretreatment technique will devolve on the intending final products of the process.

Abstract:As a clean and renewable energy, bioenergy is one of the most promising alternatives to fossil fuels. Lignocellulose possesses great potential for bioenergy production, but the recalcitrant and heterogeneous structure limits its application. Pretreatment technology offers an effective solution to fractionate the main components of the lignocellulose and uncover the available cellulose. The obtained feedstock can be applied to bioconversion into energy, e.g., bioethanol, biogas, biohydrogen, etc. Here, the current state of lignocellulose pretreatment technologies was comprehensively reviewed, the advances in bioenergy production from pretreated lignocellulose was described, with particular attention to key challenges involved. Several new strategies for overcoming pretreatment barriers to realize highly efficient lignocellulose bioconversion were highlighted. The insights given in this review will facilitate further development on lignocellulosic bioenergy production, towards addressing the global energy crisis and climate change related to the use of fossil fuels.
