Abstract:Developing a biocompatible and efficient photothermal coupling agent with appropriate size is a prerequisite for the development of near-infrared (NIR) light-induced photothermal therapy (PTT). In the present study, polyaniline nanoparticles (PANPs) with a size of 48.5 ± 1.5 nm were fabricated and exhibited excellent dispersibility in water by a hydrothermal method and further surface functionalization by capping with F127. The developed F127-modified PANPs (F-PANPs) had a high molar extinction coefficient of 8.95 × 10(8) m(-1) cm(-1), and high NIR photothermal conversion efficiency of 48.5%. Furthermore, combined with NIR irradiation at 808 nm and injection of F-PANP samples, in vivo photothermal ablation of tumor with excellent treatment efficacy was achieved. In vitro transmission electron microscopy (TEM) images of cells, methyl thiazolyl tetrazolium (MTT) assay, histology, and hematology studies revealed that the F-PANPs exhibit low toxicity to living systems. Therefore, F-PANPs could be used as PTT agents for ablating cancer, and the concept of developing polyaniline-based nanoparticles can serve as a platform technology for the next generation of in vivo PTT agents.

Abstract:The growing incidence of cancer is a problem for modern medicine, since the therapeutic efficacy of applied modalities is still not satisfactory in terms of patients' survival rates, especially in the case of patients with brain tumors. The destructive influence of chemotherapy and radiotherapy on healthy cells reduces the chances of full recovery. With the development of nanotechnology, new ideas on cancer therapy, including brain tumors, have emerged. Photothermal therapy (PTT) is one of these. It utilizes nanoparticles (NPs) that can convert the light, preferably in the near-infrared (NIR) region, into heat. In this paper, we report the use of nanodiamonds (NDs) conjugated with biomimetic polydopamine (PDA) and indocyanine green (ICG) for glioblastoma cancer PTT therapy. The obtained materials were thoroughly analyzed in terms of their PTT effectiveness, as well as their physicochemical properties. The performed research demonstrated that NDs@PDA@ICG can be successfully applied in the photothermal therapy of glioblastoma for PTT and exhibited high photothermal conversion efficiency η above 40%, which is almost 10 times higher than in case of bare NDs. In regard to our results, our material was found to lead to a better therapeutic outcome and higher eradication of glioblastoma cells, as demonstrated in vitro.

Abstract:P hotothermal therapy is a direct and non-invasive therapeutic strategy. Herein, we report a general strategy for the construction of polyaniline-coated DNA nanoparticles for efficient photothermal therapy. In our design, two G4/hemin-based DNAzymes are precisely organized in a DNA template to mimic the function of horseradish peroxidase (HRP) for the catalytic oxidation of aniline. After polymerization, the polyaniline-coated DNA nanoparticles (PANI/DNA NPs) can be efficiently obtained as the photothermal agent to elicit a pronounced photothermal effect. After cellular uptake, the noticeable photothermal effect from 808 nm laser irradiation can achieve an efficient proliferation inhibition of tumor cells. This rationally developed photothermal agent based on DNA template-assisted polymerization presents a new avenue for the development of photothermal therapy.

Abstract:<p>Photoacoustic imaging‐guided photothermal therapy in the second near‐infrared (NIR‐II) window shows promise for clinical deep‐penetrating tumor phototheranostics. However, ideal photothermal agents in the NIR‐II window are still rare. Here, the emeraldine salt of polyaniline (PANI‐ES), especially synthesized by a one‐pot enzymatic reaction on sodium bis(2‐ethylhexyl) sulfosuccinate (AOT) vesicle surface (PANI‐ES@AOT, λ<sub>max</sub> ≈ 1000 nm), exhibits excellent dispersion in physiological environment and remarkable photothermal ability at pH 6.5 (photothermal conversion efficiency of 43.9%). As a consequence of the enhanced permeability and retention effect of tumors and the doping‐induced photothermal effect of PANI‐ES@AOT, this pH‐sensitive NIR‐II photothermal agent allows tumor acidity phototheranostics with minimized pseudosignal readout and subdued normal tissue damage. Moreover, the enhanced fluidity of vesicle membrane triggered by heating is beneficial for drug release and allows precise synergistic therapy for an improved therapeutic effect. This study highlights the potential of template‐oriented (or interface‐confined) enzymatic polymerization reactions for the construction of conjugated polymers with desired biomedical applications.</p>

Health care organizations that hope to survive and prosper will need to develop new skills such as performance standards, risk-sharing arrangements, and selection and pricing criteria for this changed environment. They will also need to become involved in all aspects of the provision of health care, with responsibility for management, marketing, and maintaining quality while controlling money, services, and promises. Buyers will increasingly demand certain features from health care organizations, including flexibility in benefits and financial arrangements; controls on price and utilization of services; monitoring the quality of care; and financial stability. Abstract:As an alternative or supplement to conventional tumor treatment, photothermal therapy (PTT) is a rapid development technique for tumor elimination with the advantages of remote manipulation and minimal invasiveness. Although many different nanomaterials used in PTT have achieved excellent tumor treatment effects, poor tumor accumulation and high tumor heat resistance might lead to low PTT efficacy and frequent tumor recurrence. Herein, a kind of tumor-targeting nanoparticle (PDA-HA/STA9090 NPs) was rationally designed through conjugating hyaluronic acid (HA) on the surface of polyamine (PDA), along with loading the heat shock protein 90 (HSP90) inhibitor Ganetespib (STA9090). The PDA-HA/ST9090 NPs not only presented a high photothermal conversion efficiency (35.2%) but also enhanced the accumulation in tumoral sites for efficient downregulation of HSP90 in vitro. Accordingly, the mice treated with PDA-HA/STA9090 NPs under 808 nm laser irradiation showed the most effective tumor inhibition. Taken together, this work could not only provide a potential strategy to reduce hyperthermia side effects but also prompt the therapeutic effect of PTT with the assistance of HSP inhibition.

Abstract:Due to the strong and tunable photothermal effect, metallic nanoparticles are of enormous interest in light-activated biomedical applications, such as photoacoustic imaging (PAI) and photothermal therapy (PTT). However, the photothermal conversion efficiency (PCE) of existing metallic photothermal agents is still unsatisfactory. Herein, we develop an efficient photothermal theranostic agent based on a gold nanostar@polyaniline core-shell nanocomposite with high PCE for PAI-guided PTT at a low dosage. After optimizing the relative composition of polyaniline (PANI) and gold nanostars (AuNSs), this nanocomposite eventually empowers an outstanding PCE of up to 78.6%, which is much better than AuNSs or PANI alone and most of the existing photothermal theranostic agents. Besides, the nanocomposite can act as a targeted probe for tumors by hyaluronic acid (HA) modification without compromising the photothermal performance. The obtained nanoprobes named AuNSPHs exhibit promising biocompatibility and great performance of PAI-guided PTT to treat triple-negative breast cancer both in vitro and in vivo. More importantly, a single injection of AuNSPHs significantly suppresses tumor growth with a low dosage of Au (0.095 mg/kg), which is attributed to the high PCE of AuNSPHs. Taking advantage of the exhilarating photothermal conversion ability, this theranostic agent can safely potentiate the antitumor therapeutic efficacy of laser-induced ablation and holds great potential for future medical applications.
Organic–inorganic hybrid photothermal nanomaterials for combined photothermal and chemotherapy therapy of tumors under the dual biological window

Abstract:In view of the advantages of non-invasive photothermal therapy (PTT) in the clinical treatment of tumors, such as high efficiency of tumor ablation and less side effects, combination therapy based on PTT has attracted widespread attention. In this study, organic–inorganic hybrid nanoparticles (NPs) of PANI@W18O49 with uniform morphology and PANI@W18O49@Fe3O4 with magnetic targeting were synthesized by hydrothermal method. It was found that the hybrid NPs not only did not affect the photothermal properties of W18O49, but also improve its antioxidant properties, which made the synthesized NPs more suitable for PTT as a photothermal agent than W18O49 itself. The results of absorbance and photothermal measurement of NPs under different wavelengths (808 and 1064 nm) show that the photothermal performance of hybrid NPs is stable. The photothermal conversion efficiency of PANI@W18O49 under 808 and 1064 nm laser irradiation can reach to 50.43 and 30.69%, respectively, and that of PANI@W18O49@Fe3O4 can also reach to 63.9 and 32.55%; the photothermal temperature can induce apoptosis of cancer cells. In drug release experiments, it was demonstrated that the hybrid NPs could control the release of DOX under the dual stimulus response (pH and temperature) to reduce the side effects caused by the uncontrolled release of drug under physiological conditions. In addition, the survival rate of cancer cells decreased to 20.05 and 22.33% by NIR irradiation after co-incubation of drug-loaded NPs, indicating that the prepared hybrid NPs have the potential to be applied in the combined treatment of photothermal and chemotherapy for cancer.Graphical Preparation methods of PW and PWF NPs and schematic diagram of their application in chemotherapy/photothermal combined therapy for tumors.

Abstract:In this study, multifunctional nanoparticles were fabricated for tumor diagnosis and therapy. The gold nanoparticles, which were prepared from HAuCl4 and aspartate via in situ UV irradiation, were coated with polydopamine (PDA) by self-polymerization of dopamine. Subsequently, the Au@PDA nanoparticles were coated with the BSA–dextran conjugate (BD). The photothermal transduction efficiency of the Au@PDA@BD nanoparticles was 17.0%. During 808 nm laser irradiation, the temperature rise and photothermal cytotoxicity of the Au@PDA@BD nanoparticle solution depended on the polydopamine concentration. Because of the Au core, the Au@PDA@BD nanoparticles improved the brightness and resolution of tumor CT imaging after intratumoral injection of the nanoparticles into H22 tumor-bearing mice. Due to the dextran brush surface, Au@PDA@BD nanoparticles could prolong the circulation time in blood and accumulate at the tumor site after intravenous injection, and thus NIR laser irradiation could cause the tumor temperature to rise to ablate the tumor completely. Only once treatment by intravenous injection of Au@PDA@BD nanoparticles into the tumor-bearing mice followed by 808 nm laser irradiation for 10 min could completely cure the tumors even if the original tumor volume was about 250 mm3. At the same conditions, Au@PDA nanoparticles without the dextran brush surface had no treatment efficacy. Furthermore, Au@PDA@BD nanoparticles plus the laser irradiation had no histological toxicity on the main organs of the treated mice. The Au@PDA@BD nanoparticles show a great potential for highly efficient photothermal therapy of tumors.
Functionalized Magnetic Nanoparticles for NIR-Induced Photothermal Therapy of Potential Application in Cervical Cancer

Abstract:Photothermal therapy (PTT) holds great promise for cancer treatment with its effective ablation of solid tumors. As the essential core point, photothermal agents (PTAs) with excellent photothermal properties and good biocompatibility could help to fulfill highly efficient PTT. Herein, a novel type of nanoplatform Fe3O4@PDA/ICG (FPI) nanoparticle (NP) was designed and synthesized, which was composed of magnetic Fe3O4 and near-infrared excitable indocyanine green via encapsulation of polydopamine. The FPI NPs showed spherical structures in shape with uniform distribution and good chemical stability. Under 793 nm laser irradiation, FPI NPs could generate hyperthermia of 54.1 °C and photothermal conversion efficiency of 35.21%. The low cytotoxicity of FPI NPs was further evaluated and confirmed on HeLa cells with a high survival rate (90%). Moreover, under laser irradiation (793 nm), FPI NPs showed effective photothermal therapeutic characteristics for HeLa cells. Therefore, FPI NPs, as one of the promising PTAs, have great potential in the field of PTT for tumor treatment.

Abstract:Photothermal therapy (PTT) has attracted increasing attention as an effective anticancer strategy, which has minimal side effects and could suppress the tumor growth effectively. Herein, poly (ethylene glycol)-stabilized polydopamine-modified gold nanostars (AuNSs@PDA-PEG) nanoparticles (NPs) were fabricated for PTT of cancer. The AuNSs@PDA-PEG NPs exhibited strong near-infrared (NIR) absorbance and excellent photostability for highly efficient PTT. Cytotoxicity assay indicated that the NPs possess high biocompatibility. Moreover, the AuNSs@PDA-PEG NPs exhibited excellent photothermal killing performance in vitro, in which the cell viability decreased to 10.02% under the 808 nm laser irradiation. The apoptosis assay indicated that HeLa cells incubated with AuNSs@PDA-PEG NPs can be seriously destroyed by the NIR laser irradiation. The mechanisms of NIR PTT-induced cancer cell death involved mitochondrial dysfunction, increase in lysosomal membrane permeability, and autophagy. Western blot analysis confirmed AuNSs@PDA-PEG NPs-induced decrease in Bcl-2 expression and elevation of Bax expression. Thus, the biocompatible AuNSs@PDA-PEG NPs exhibit great potential in PTT of cancer.

Abstract:There is still an urgent need to develop photothermal agents (PTAs) with stable photothermal stability and good biocompatibility, especially PTAs with functional groups for mild-temperature photothermal therapy (PTT). Herein, carboxylated nanodiamond (ND-COOH) was prepared by annealing of pristine nanodiamond (ND), and aminated nanodiamond (ND-PEI) was further synthesized by grafting ethylenediamine-branched polyethylenimine to ND-COOH. The mild-temperature photothermal effect of these NDs was certified. The photothermal conversion efficiency of the NDs was calculated to be ~20%, and the photothermal properties were stable for the NDs with the functional groups, which supplies possibility to further modify NDs as mild-temperature photothermal nanoplatforms. In vitro antibacterial experiments showed that almost all of the bacteria (>99%) co-cultured with NDs at concentration of 200 μg/mL and 150 μg/mL to S. aureus and to E. coli were killed after 808 nm irradiation for 10 min, respectively. 4T1 cells, mouse breast cancer cells, co-cultured with 200 μg/mL NDs under irradiation 10 min were ablated about 40%. Meanwhile, NDs showed no cytotoxicity and excellent biocompatibility. Therefore, NDs can be used as mild-temperature PTAs by near-infrared light-triggering, and NDs with functional groups will be new promising nanoplatforms which may inspire many new progresses in the field of mild-temperature PTT.

Abstract:Introduction The high concentration of glutathione (GSH) and hydrogen peroxide (H2O2) levels within the tumor microenvironment (TME) are the major obstacle to induce the unsatisfactory anticancer treatment efficiency. The synergistic cancer therapy strategies of the combination the GSH depletion enhanced chemodynamic therapy (CDT) with photothermal therapy (PTT) have been proved to be the promising method to significantly improve the therapeutic efficacy. Methods The copperphosphotungstate was incorporated into polyanilines to design copperphosphotungstate doped polyaniline nanorods (CuPW@PANI Nanorods) via chemical oxidant polymerization of aniline. The low long-term toxicity and biocompatibility were evaluated. Both in vitro and in vivo experiments were carried out to confirm the GSH depletion enhanced CDT/NIR-II PTT synergistic therapy. Results CuPW@PANI Nanorods feature biosafety and biocompatibility, strong NIR-II absorbance, and high photothermal-conversion efficiency (45.14%) in NIR-II bio-window, making them highly applicable for photoacoustic imaging and NIR-II PTT. Moreover, CuPW@PANI Nanorods could consume endogenous GSH to disrupt redox homeostasis and perform a Fenton-like reaction with H2O2 to produce cytotoxic •OH for the enhanced CDT. Furthermore, NIR-II photothermal-induced local hyperthermia accelerates •OH generation to enhance CDT, which realizes high therapeutic efficacy in vivo. Conclusion This study provides a proof of concept of GSH-depletion augmented chemodynamic/NIR-II photothermal therapy.

Abstract:In recent years, inorganic nanoparticles (NPs) have attracted increasing attention as potential theranostic agents in the field of oncology. Photothermal therapy (PTT) is a minimally invasive technique that uses nanoparticles to produce heat from light to kill cancer cells. Photothermal therapy (PTT) requires two essential elements: a photothermal agent (PTA) and near-infrared radiation (NIR). The PTAs role is to absorb NIR, which subsequently triggers hyperthermia within cancer cells. By raising the temperature in the tumor microenvironment (TME), PTT causes damage to the cancer cells. Nanoparticles (NPs) are instrumental in PTT as they facilitate the passive and active targeting of the PTA to the TME, making them crucial for the effectiveness of the treatment. In addition, specific targeting can be achieved through enhanced permeation and retention. Owing to their enormous advantages, such as altering the morphology and surface characteristics of nanocarriers comprising PTA, NPs have been exploited to facilitate tumor regression significantly. This review highlights the properties of PTAs, the mechanism of PTT, and the results obtained for the improved curative efficacy of PTT by utilizing the NPs platform.

Abstract:Photothermal therapy (PTT) has received constant attention as an efficient cancer therapy method due to locally selective treatment, which is not affected by the tumor microenvironment. In this study, a novel 880 nm near-infrared (NIR) laser-triggered photothermal agent (PTA), 3TT-IC-4Cl, was used for PTT of a tumor in deep tissue. Folic acid (FA) conjugated amphiphilic block copolymer (folic acid-polyethylene glycol-poly (β-benzyl-L-aspartate)10, FA-PEG-PBLA10) was employed to encapsulate 3TT-IC-4Cl by nano-precipitation to form stable nanoparticles (TNPs), and TNPs exhibit excellent photothermal stability and photothermal conversion efficiency. Furthermore, the in vitro results showed TNPs display excellent biocompatibility and significant phototoxicity. Abstract:Recently, the near-infrared-II (NIR-II, 1000–1350 nm) region has been extensively applied in deep-tissue photothermal therapy (PTT) on account of it having stronger tissue penetration and a higher maximum permissible exposure (MPE) than the near-infrared-I (NIR-I, 650–950 nm) region. In this study, we developed a rapid and convenient in situ polymerization strategy to fabricate polypyrrole nanosheets (PPy NSs) within a few minutes using manganese dioxide nanosheets (MnO2 NSs) as both the oxidant and the self-sacrificed template. The fabricated PPy NSs exhibited excellent NIR-II absorption, which conferred its high photothermal conversion efficiency (66.01%) at 1064 nm and its photoacoustic (PA) imaging capability. Both in vivo and in vitro studies have shown that that PPy NSs possess good biological safety and excellent PTT efficacy and PA imaging performances. Thus, the as-synthesized PPy NSs could effectively achieve PA imaging-guided photothermal tumor ablation under 1064 nm excitation. Our work provides a novel and promising method for the rapid preparation of PPy NSs without the addition of exogenous oxidants and subsequent template removal, which could be regarded as potential photothermal agents (PTAs) to integrate the diagnosis and treatment of cancer.

Abstract:Photothermal therapy has attracted significant attention as a minimally invasive therapy methodology. In this work, we report PEGylated nickel carbide nanocrystals (Ni3C NCs) as an efficient photothermal agent for the first time. The nanoparticles exhibit a broad absorption from the visible to the near-infrared regions and a rapid rise in temperature when irradiated by an 808 nm laser even at a concentration of 100 mu g mL(-1). In vitro and in vivo cytotoxicity assays demonstrate they have good biocompatibility, which lays an important foundation for their biological application. In vitro studies reveal the efficient damage of cancer cells by the exposure of 808 nm laser with a power density of 0.50 W cm(-2). Furthermore, hematoxylin and eosin (H & E) and terminal deoxynucleotidyl transferase biotin-dUTP nick-end labeling (TUNEL) staining of tumor slices confirmed the obvious destruction of cancer cells in vivo by an 808 nm laser (0.50 W cm(-2)) after only a 5 min application. Our work may open up a new application domain for transition metal carbides for biomedicine.

Abstract:Temperature is an essential factor in all biological processes and most of the biological activities are related to temperature. However, information about the temperature in living cells are limited. In photothermal therapy, an emerging cancer treatment due to its high spatial control and non-invasive nature, the local effect of intracellular temperature change is not well understood. Fluorescent nanodiamonds (ND) are a unique carbon material possessing optical defects inside the carbon lattice that enable sensing of temperature on a nanoscale completely independent of external conditions. Herein, we coat ND with polymers to form a nanogel shell around NDs that can absorb indocyanine green, a common photothermal agent, to obtain nanodiamond-nanogel-indocyanine green. Upon irradiation, we not only show successful killing of cancer cells with a high control in space but we are also able to sense the increase in temperature using single NDs as nanothermometers in cells.
