Abstract:Numerous photothermal agents (PTAs) require high-intensity and long-duration laser excitation for photothermal therapy (PTT), resulting in light damage to healthy skin and tissue as well as limiting their biomedical applications. Integrating desirable near-infrared (NIR) absorption and high photothermal conversion efficiency (PCE) into a single small-molecule PTA is an important prerequisite for realizing efficient PTT, but is a serious challenge. Herein, through molecular engineering strategy, an acceptor-donor-acceptor (A-D-A) type PTA (ADA3) was readily developed for 808 nm laser-driving photothermal imaging and PTT of tumor. Theoretical calculations and experiment results show molecular engineering strategy is significant in regulating the structure and energy gap of PTAs, so as to effectively induce a narrow band gap for NIR absorption and further optimize photothermal properties. ADA3 possesses molar extinction coefficient of 3.1 × 104 M-1 cm-1 at 808 nm, followed being assembled into nanoparticles, ADA3-NPs show high PCE of 80.3%. In vivo experiments indicate that ADA3-NPs have excellent antitumor capability under one-time, low-intensity and short-duration (808 nm, 330 mW/cm2, 3 min) laser irradiation. Therefore, this work definitely exemplifies the enormous potential of molecular engineering strategy and provides an effective method for developing small-molecule PTAs.

Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.

Abstract:Breast cancer has become a huge burden with continued rise of incidence and death rate worldwide. Various methods for diagnosis and therapy of breast cancer have met the challenges of lack of complete information about the tumor location and limited therapy efficacy. Although aggregation-induced emission luminogens (AIEgens) have shown great promise for various cancer treatment applications, they may be incompetent for deep-seated tumor diagnosis due to the limited penetration depth. Herein, we designed and prepared a radiolabeled AIEgen-based organic photothermal agent for bimodal PET/fluorescence imaging-guided breast tumor photothermal therapy. The prepared multifunctional nanoparticles (Ga-68-TPA-TTINC NPs) with NIR-II fluorescence, gamma irradiation and photothermal conversion property could be efficiently taken up by tumor cells and induce reactive oxygen species burst in vitro, further boosting the photothermal treatment of tumor in vivo. More importantly, the nanoprobe could target and clearly visualize 4T1 tumor xenografts through PET and NIR-II fluorescence imaging with high tumor/muscle ratio up to 4.8, which provides a promising tool and solution for breast tumor theranostics.

Abstract:Phototheranostic nanoagent with NIR-II fluorescence imaging and photothermal therapy (PTT) may allow high-quality imaging guided efficient ablation of deep-tissue tumors. Non-specific distribution in vivo may lead to poor imaging results and a risk of damage to normal tissues. However, developing an activable NIR-II nanoagent for photothermal therapy is challenging. Herein, a new polarity-sensitive phototheranostic nanoagent ( DC@PEG ) is developed for combinational tumor-specific NIR-II fluorescence imaging and photothermal therapy. DC@PEG has a maximum emission exceed 1200 nm at 808 nm excitation. In vivo fluorescence imaging indicated the nanoparticles could accurately distinguish normal tissue (high polarity environment) and tumor tissue (low polarity environment). Furthermore, DC@PEG effectively inhibited the growth of 4T1 tumor under 808 nm laser irradiation with a high photothermal conversion efficiency of 48.1%. This work provides a promising strategy for development of excellent NIR-II nanoagent with good biocompatibility and deep tissue penetration. This novel nanoagent can served as a robust tool for photothermal therapy of deep tumors.

Abstract:Given their easy structural modification and good biocompatibility advantages, near-infrared (NIR) organic dyes with a large molar extinction coefficient, while a superlow fluorescence quantum yield shows considerable potential application in photothermal therapy (PTT). Herein, a new NIR-absorbing asymmetric cyanine dye, namely, RC, is designed and synthesized via the hybrid of rhodamine and hemicyanine derivatives. RC-BSA nanoparticles (NPs) are fabricated by using the bovine serum albumin (BSA) matrix. The NPs exhibit a strong NIR absorption peak at ∼868 nm and 28.7% photothermal conversion efficiency. Based on these features, RC-BSA NPs exhibit excellent performance in ablating tumor under a 915 nm laser radiation through a PTT mechanism. These NPs show no obvious toxicity to the treated mice. Thus, RC-BSA NPs can used as a new NIR laser-triggered PTT agent in cancer treatment.

Abstract:Phototherapy has garnered considerable interest for its potential to revolutionize conventional cancer treatment. Organic materials with near-infrared II (NIR-II, 1000-1700 nm) fluorescence and photothermal effects are key for precise tumor diagnosis and treatment, yet optimizing their output for higher resolution and reduced photodamage remains a challenge. Herein, a multifunctional NIR-II photosensitizer (LSC) has been developed using the aggregation-induced emission (AIE) technology. The utilization of Thieno[3,2-b]thiophene as an electron-rich and bulky donor/acceptor bridge has allowed for the elongation of conjugation length and distortion of the AIE main chain. This strategic modification effectively enhances the electron push-pull effect, endowing the LSC with a Stokes shift of over 400 nm and AIE characteristics. We have successfully built-up stable nanoparticles called FA-LSC NPs using a nano-precipitation method. These nanoparticles exhibit high NIR-II fluorescent brightness (ε × QY = 1064 M-1 cm-1) and photothermal conversion efficiency (41%). Furthermore, the biocompatible FA-LSC NPs demonstrate effective tumor accumulation and exceptional photothermal therapeutic efficacy both in vitro and in vivo. These nanoparticles were applied to fluorescence-photothermal dual-mode imaging-guided photothermal ablation in a Hela tumor xenograft mouse model, resulting in favorable photothermal clearance outcomes.

Abstract:Near infrared (NIR) fluorescent organic molecules as fluorescent probes accurately guide photothermal therapy as a potential antitumor method. However, the aggregation and quenching of organic fluorescent molecules and poor tissue permeability greatly limit their therapeutic effect and clinical transformation. In this paper, with a D-A-D structure as the molecular skeleton, cyclopentadithiophene (CPDT) as the donor (D), diketopyrrolopyrrole (DPP) as the acceptor (A), and long-chain isooctane as the shielding unit, organic fluorescent small molecules with a strong absorption band and bright NIR-II emission were synthesized. Then, tetraphenylethylene (TPE) molecules with typical AIE structure characteristics were introduced on both sides of the organic fluorescent small molecules, and an organic small molecular fluorophore (TDA) with AIE characteristics and the photothermal effect was designed. Through a series of experimental characterization techniques, it is proved that TDA NPs have good biocompatibility and tissue permeability, and can accurately locate the tumor location through NIR-II fluorescence imaging to achieve local photothermal treatment of tumors.

Abstract:A novel low‐molecular‐weight dibenzofulvene‐based D‐π‐A phototheranostic agent (PTA) with NIR‐II emission and high PCE is developed by the strategies of enhancing D‐A strength, introducing molecular rotors/vibrators, and facilitating intermolecular π–π stacking. The PTA is successfully applied in the multi‐modal imaging (NIR‐II fluorescence/photoacoustic/photothermal imaging)‐guided photothermal therapy. Organic molecules with second near‐infrared (NIR‐II, 1000–1700 nm) emission have significant advantages over NIR‐I (600–900 nm) dyes, and have been widely used as phototheranostic agents (PTAs) and fluorescent probes. However, the two biggest challenges for such reagents are improving photothermal conversion efficiency (PCE), and simplifying tedious synthetic procedures. Herein, different from the traditional NIR skeleton, for the first time, it develops two novel dibenzofulvene‐based NIR‐II emission PTAs via D‐π‐A concept with high PCEs through two simple synthetic steps. The increased intramolecular charge transfer effect and extended π‐conjugation effectively red‐shift their emissions to the NIR‐II region. In addition, the introduction of molecular rotors/vibrators and the formation of π–π stacking prompt the two PTAs to exhibit high PCEs. Notably, FE‐IDMN nanoparticles (FE‐IDMN NPs) achieve an exciting PCE of 82.6%, higher than most reported photothermal agents with NIR‐II emission. FE‐IDMN NPs also possess good colloidal, pH, and photothermal stabilities, as well as excellent photoacoustic response, which is further successfully applied for NIR‐II fluorescence/photoacoustic/photothermal imaging‐guided photothermal therapy. This work provides a simple strategy for constructing new D‐π‐A PTAs with low molecular weight, NIR‐II emission, and high PCE for cancer therapy.

Abstract:Second near-infrared (NIR-II,1000 ∼ 1700 nm) therapeutic window presents an increased tissue penetration and elevated maximal permissible exposure in the application of photothermal therapy (PTT). However, the lack of NIR-II photothermal conversion agents (PCAs) limit their further development. In this work, we rationally designed and successfully developed three novel indolium-like heptamethine cyanine dyes (NFs) by installing N,N- diethylamino on the terminal ends of a conjugated polyene backbone and replacing the middle chlorine atom with o- mercapto benzoic acid and p- mercapto benzoic acid. Notably, NF2 with stronger rotating group encapsulated in organic nanoparticles (NF2 NPs) exhibited high photothermal conversion efficiency (PCE), which could come up to (61.3 %). Then we conducted serial experiments to further investigate PTT capability of NF2 NPs 4 T1 cell line and nude mice bearing 4 T1 tumor. As expected, the resulting NF2 NPs presented the excellent photothermal conversion ability and superb PTT effect both in vivo and in vitro. This study will inspire more work for future design and clinical applications of NIR-II therapeutic agents.

Abstract:Although acceptor-donor-acceptor (A-D-A)-type molecules offer advantages in constructing NIR absorbing photothermal agents (PTAs) due to their strong intramolecular charge transfer and molecular planarity, their applications in photothermal therapy (PTT) of tumors remain insufficiently explored. In particular, the influence of ESP distribution on the optical properties of A-D-A photosensitizers has not been investigated. Herein, we analyze and compare the difference in ESP distribution between A-D-A-type small molecules and polymers to construct NIR absorbing PTAs with a high extinction coefficient (epsilon) and high photothermal conversion efficiency (PCE). The calculation results of density functional theory (DFT) indicate that the large ESP difference makes A-D-A-type small molecules superior to their polymer counterparts in realizing tight molecular packing and strong NIR absorbance. Among the as-prepared nanoparticles (NPs), Y6 NPs exhibited an obvious bathochromic shift of absorption peak from 711 nm to 822 nm, with the NIR-II emission extended to 1400 nm. Moreover, a high epsilon value of 5.69 L g-1 cm-1 and a PCE of 66.3% were attained, making Y6 NPs suitable for PTT. With a concentration of 100 mu g mL-1, Y6 NPs in aqueous dispersion yielded a death rate of 93.4% for 4T1 cells upon 808 nm laser irradiation (1 W cm-2) for 10 min, which is comparable with the best results of recently reported PTT agents. We explored a new way of utilizing the ESP difference to improve the molecular packing of (A-D-A) type molecules, and obtained a series of NIR PTAs with good PTT performance. The in vivo antitumor study demonstrates effective suppression of tumors.

Abstract:Photothermal therapy (PTT) has received constant attention as an efficient cancer therapy method due to locally selective treatment, which is not affected by the tumor microenvironment. In this study, a novel 880 nm near-infrared (NIR) laser-triggered photothermal agent (PTA), 3TT-IC-4Cl, was used for PTT of a tumor in deep tissue. Folic acid (FA) conjugated amphiphilic block copolymer (folic acid-polyethylene glycol-poly (β-benzyl-L-aspartate)10, FA-PEG-PBLA10) was employed to encapsulate 3TT-IC-4Cl by nano-precipitation to form stable nanoparticles (TNPs), and TNPs exhibit excellent photothermal stability and photothermal conversion efficiency. Furthermore, the in vitro results showed TNPs display excellent biocompatibility and significant phototoxicity. These results suggest that 880 nm triggered TNPs have great potential as effective PTAs for photothermal therapy of tumors in deep tissue.

Abstract:Recently, extensive efforts have been devoted to the development of the second near-infrared bio-window (NIR-II, 1000-1700 nm) theranostic agents owing to the excellent tissue-penetration capability of NIR-II light. The exploration of organic NIR-II photothermal therapy materials, especially those with absorption peak over 1000 nm, is an appealing yet significantly challenging task. Herein, we have designed conjugated polymer nanoparticles (PIS NPs) with NIR-II absorption peak at 1026 nm through a combined strategy of introducing quinoid donor-acceptor (D-A) structures, constructing intramolecular "conformational locks" and extending the conjugation area to narrow the band gap. Irradiated at 1064 nm, PIS NPs showed remarkable photothermal conversion performance for efficient photothermal ablation of tumor cells in vitro and in vivo. This study provides useful insights into the rational design of organic NIR-II photothermal agents based on multiple strategies.
A perylene diimide probe for NIR-II fluorescence imaging guided photothermal and type I/type II photodynamic synergistic therapy

Abstract:Phototherapy has garnered significant attention in the past decade. Photothermal and photodynamic synergistic therapy combined with NIR fluorescence imaging has been one of the most attractive treatment options because of the deep tissue penetration, high selectivity and excellent therapeutic effect. Benefiting from the superb photometrics and ease of modification, perylene diimide (PDI) and its derivatives have been employed as sensing probes and therapeutic agents in the biological and biomedical research fields, and exhibiting excellent potential. Herein, we reported the development of a novel organic small-molecule phototherapeutic agent, PDI-TN. The absorption of PDI-TN extends into the NIR region, which provides feasibility for NIR phototherapy. PDI-TN overcomes the traditional Aggregation-Caused Quenching (ACQ) effect and exhibits typical characteristics of Aggregation-Induced Emission (AIE). Subsequently, PDI-TN NPs were obtained by using an amphiphilic triblock copolymer F127 to encapsulate PDI-TN. Interestingly, the PDI-TN NPs not only exhibit satisfactory photothermal effects, but also can generate O2•- and 1O2 through type I and type II pathways, respectively. Additionally, the PDI-TN NPs emit strong fluorescence in the NIR-II region, and show outstanding therapeutic potential for in vivo NIR-II fluorescence imaging. To our knowledge, PDI-TN is the first PDI derivative used for NIR-II fluorescence imaging-guided photodynamic and photothermal synergistic therapy, which suggests excellent potential for future biological/biomedical applications.

Abstract:Photodynamic therapy (PDT), a noninvasive therapeutic strategy for cancer treatment, which always suffers from the low reactive oxygen species (ROS) yield of traditional organic dyes. Herein, we present lipid-encapsulated aggregation-induced emission nanoparticles (AIE NPs) that have a high quantum yield (23%) and a maximum two-photon absorption (TPA) cross-section of 560 GM irradiated by near-infrared light (800 nm). The AIE NPs can serve as imaging agents for spatiotemporal imaging of tumor tissues with a penetration depth up to 505 μm on mice melanoma model. Importantly, the AIE NPs can simultaneously generate singlet oxygen (1O2) and highly toxic hydroxyl radicals (•OH) upon irradiation with 800 nm irradiation for photodynamic tumor ablation. In addition, the AIE NPs can be effectively cleared from the mouse body after the imaging and therapy. Polycyclic naphthalenediimide-based nanoparticles for NIR-II fluorescence imaging guided phototherapy

Abstract:Optical imaging and phototherapy in the second near-infrared window (NIR-II, 900-1700 nm) can reduce tissue auto-fluorescence and photon scattering, which facilitates higher spatial resolution and deeper tissue penetration depth for solid tumor theranostics. Herein, a polycyclic naphthalenediimide (NDI) based chromophore 13-amino-4,5-dibromo-2,7- di(dodecan-6-yl)-1H-isoquinolino[4,5,6-fgh]naphtho[1,8-bc][1,9]phenanthroline-1,3,6,8(2H,7H,9H)-tetraone (NDI-NA) was designed and synthesized. With large polycyclic π-systems, NDI-NA molecule possesses broad near-infrared (NIR) absorption (maximum at 777 nm) and emission (maximum at 921 nm). By nanoprecipitation, NDI-NA nanoparticles (NPs) were formed in aqueous solution with J-aggregative state, which showed huge red-shift in both absorption spectrum (maximum at 904 nm) and emission spectrum (maximum at 1,020 nm), endowing NDI-NA NPs efficient NIR-II fluorescence imaging capability. Besides, the NPs present effective tumor-targeting capability in vivo based on the enhanced permeation and retention (EPR) effect. More importantly, NDI-NA NPs simultaneously have high photothermal conversion efficiency (30.8%) and efficient reactive oxygen species generation ability, making them remarkably phototoxic to cancer cells. The polycyclic chromophore based multifunctional NDI-NA NPs as NIR-II phototheranostic agents possess bright future for clinical NIR-II imaging-guided cancer phototherapy.

Semiconducting Polymer Nanoparticles for NIR-II Fluorescence Imaging-Guided Photothermal/thermodynamic Combination Therapy.

Abstract:Photothermal therapy is a promising phototherapeutic modality that has been widely studied in cancer therapy. However, because of the influence of heat shock protein (HSP), the therapeutic efficacy of photothermal therapy (PTT) is significantly suppressed. To improve the therapeutic efficacy, different tumor-specific therapeutic modalities have been chosen to combine with PTT. However, most of them rely on endogenous stimuli to trigger combination therapy, which may suffer from the issue of incomplete activation. Herein, we develop a PTT/thermodynamic combination therapeutic nanosystem whose therapeutic process is controlled by an external stimulus, near-infrared (NIR) light. The nanosystem (ADPPTN) is composed of a second NIR (NIR-II) fluorescent semiconducting polymer (SP) (DPPT) as the core, and a carboxyl group-decorated amphiphilic copolymer (PSMA-PEG) as the shell with an azo-containing compound (AIPH) loaded via electrostatic interaction. Under 808 nm laser irradiation, DPPT can generate heat to conduct PTT, while the elevated temperature may further trigger the release of AIPH radicals, conducting thermodynamic therapy (TDT). In addition, the NIR-II fluorescence signal emitted from DPPT can light the tumor. Compared with the nanoparticles without AIPH (DPPTN), ADPPTN has better anticancer efficacy under laser irradiation both in vitro and in vivo. Thus, our study provides an NIR-II fluorescence imaging-guided PTT/TDT combination therapeutic nanosystem for efficient cancer theranostics.
An Activatable Phototheranostic Nanoplatform for Tumor Specific NIR-II Fluorescence Imaging and Synergistic NIR-II Photothermal-Chemodynamic Therapy.

Abstract:The phototheranostics in the second near-infrared window (NIR-II) have proven to be promising for the precise cancer theranostics. However, the non-responsive and "always on" imaging mode lacks the selectivity, leading to the poor diagnosis specificity. Herein, a tumor microenvironment (TME) activated NIR-II phototheranostic nanoplatform (Ag2 S-Fe(III)-DBZ Pdots, AFD NPs) is designed based on the principle of Förster resonance energy transfer (FRET). The AFD NPs are fabricated through self-assembly of Ag2 S QDs (NIR-II fluorescence probe) and ultra-small semiconductor polymer dots (DBZ Pdots, NIR-II fluorescence quencher) utilizing Fe(III) as coordination nodes. In normal tissues, the AFD NPs maintain in "off" state, due to the FRET between Ag2 S QDs and DBZ Pdots. However, the NIR-II fluorescence signal of AFD NPs can be rapidly "turn on" by the overexpressed GSH in tumor tissues, achieving a superior tumor-to-normal tissue (T/NT) signal ratio. Moreover, the released Pdots and reduced Fe(II) ions provide NIR-II photothermal therapy (PTT) and chemodynamic therapy (CDT), respectively. The GSH depletion and NIR-II PTT effect further aggravate CDT mediated oxidative damage toward tumors, achieving the synergistic anti-tumor therapeutic effect. The work provides a promising strategy for the development of TME activated NIR-II phototheranostic nanoprobes.
Gd2O3/b‐TiO2 composite nanoprobes with ultra‐high photoconversion efficiency for MR image‐guided NIR‐II photothermal therapy

Abstract:Low photoconversion efficiency and limited tissue penetration are the major barriers for photothermal therapy (PTT) agent application. Here, an efficient organic/inorganic dual‐PTT agent guided by MRI was proposed. It is proved the oxygen vacancy band narrowed its band gap that led to NIR‐II region absorb. Furthermore, the defect energy level trapping carrier and conjugate synergistic heat generation result in high photoconversion efficiency. Photothermal therapy (PTT), as an important noninvasive and effective tumor treatment method, has been extensively developed into a powerful cancer therapeutic technique. Nevertheless, the low photothermal conversion efficiency and the limited tissue penetration of typical photothermal therapeutic agents in the first near‐infrared (NIR‐I) region (700–950 nm) are still the major barriers for further clinical application. Here, we proposed an organic/inorganic dual‐PTT agent of synergistic property driven by polydopamine‐modified black‐titanium dioxide (b‐TiO2@PDA) with excellent photoconversion efficiency in the second NIR (NIR‐II) region (1000–1500 nm). More specifically, the b‐TiO2 treated with sodium borohydride produced excessive oxygen vacancies resulting in oxygen vacancy band that narrowed the b‐TiO2 band gap, and the small band gap led to NIR‐II region wavelength (1064 nm) absorbance. Furthermore, the combination of defect energy level trapping carrier recombination heat generation and conjugate heat generation mechanism, significantly improved the photothermal performance of the PTT agent based on b‐TiO2. The photothermal properties characterization indicated that the proposed dual‐PTT agent possesses excellent photothermal performance and ultra‐high photoconversion efficiency of 64.9% under 1064 nm laser irradiation, which can completely kill esophageal squamous cells. Meanwhile, Gd2O3 nanoparticles, an excellent magnetic resonance imaging (MRI) agent, were introduced into the nanosystem with similar dotted core–shell structure to enable the nanosystem achieve real‐time MRI‐monitored cancer therapeutic performance. We believe that this integrated nanotherapeutic system can not only solve the application of PTT in the NIR‐II region, but also provide certain theoretical guidance for the clinical diagnosis and treatment of esophageal cancer.
D–A–D organic fluorescent probes for NIR-II fluorescence imaging and efficient photothermal therapy of breast cancer

Abstract:Near-infrared second region (NIR-II) fluorescent probes are used in the diagnosis of early cancer due to their high tissue penetration. However, there are still few reports on organic small molecule fluorescent probes with NIR-II fluorescence imaging (NIR-II FI) combined with efficient photothermal therapy (PTT). In this study, planar cyclopentadithiophene (CPDT) was incorporated into the twisted structural skeleton (D–A–D), and the strong acceptor TTQ molecule (A) and the donor triphenylamine (D) were introduced to synthesize an organic small molecule (TCT) with enhanced NIR-II fluorescence emission performance. To improve the hydrophilicity of TCT molecules, we used the nanoprecipitation method to coat DSPE-mPEG2000 on the TCT molecules and obtained nanoparticles (TCT-NPs) with a strong absorption band, good water dispersibility, and NIR-II FI ability, which realized NIR-II FI-guided PTT for breast cancer tumors. Due to their effective near-infrared absorption, TCT-NPs exhibit high photothermal conversion efficiency ( η = 40.1%) under 660 nm laser irradiation, making them a photothermal therapeutic agent with good performance. Therefore, TCT-NPs have the potential to diagnose, eliminate, and monitor the diffusion of cancer.

Small‐Molecule Phototheranostic Agent with Extended π‐Conjugation for Efficient NIR‐II Photoacoustic‐Imaging‐Guided Photothermal Therapy

Abstract:A small molecule photodiagnostic agent, CY‐1234 nanoparticles (NPs), is reported in this paper. Organic small molecules with near‐infrared‐II (NIR‐II) absorption are successfully designed by increasing the conjugation length and altering the intramolecular charge transfer. Efficient photothermal therapy guided by NIR‐II window photoacoustic imaging is realized by adjusting the aggregation shape of the molecules. Photoacoustic imaging (PAI) and photothermal therapy (PTT) conducted over the near‐infrared‐II (NIR‐II) window offer the benefits of noninvasiveness and deep tissue penetration. This necessitates the development of highly effective therapeutic agents with NIR‐II photoresponsivity. Currently, the predominant organic diagnostic agents used in NIR‐II PAI‐guided PTT are conjugated polymeric materials. However, they exhibit a low in vivo clearance rate and long‐term biotoxicity, limiting their clinical translation. In this study, an organic small molecule (CY‐1234) with NIR‐II absorption and nanoencapsulation (CY‐1234 nanoparticles (NPs)) for PAI‐guided PTT is reported. Extended π‐conjugation is achieved in the molecule by introducing donor–acceptor units at both ends of the molecule. Consequently, CY‐1234 exhibits a maximum absorption peak at 1234 nm in tetrahydrofuran. Nanoaggregates of CY‐1234 are synthesized via F‐127 encapsulation. They exhibit an excellent photothermal conversion efficiency of 76.01% upon NIR‐II light irradiation. After intravenous injection of CY‐1234 NPs into tumor‐bearing mice, strong PA signals and excellent tumor ablation are observed under 1064 nm laser irradiation. This preliminary study can pave the way for the development of small‐molecule organic nanoformulations for future clinical applications.
