Abstract:Flies escape danger by jumping into the air and flying away. The giant fibre system (GFS) is the neural circuit that mediates this simple behavioural response to visual stimuli. The sensory signal is received by the giant fibre and relayed to the leg and wing muscle motorneurons. Many of the neurons in the Drosophila GFS are uniquely identifiable and amenable to cell biological, electrophysiological and genetic studies. Here we review the anatomy and development of this system and highlight its utility for studying many aspects of nervous system biology ranging from neural development and synaptic plasticity to the aetiology of neural disorder.

Abstract:The term “animal welfare” is widely used to refer to an animal’s quality of life. It encompasses the following: animals should be healthy, well fed, and housed in an environment that they might themselves choose; animals should be relatively free from negative states, such as pain, fear and distress, and capable of enjoying life; and animals should be able to carry out behaviors and activities that they are strongly motivated to do (1). The public has long been concerned about the welfare of animals as illustrated by our laws to prevent animal cruelty, the existence of humane organizations funded by charitable donations, and the use of public money to fund animal use oversight bodies. Public concern for animal welfare extends to animals used for science: although studies show that the public supports animal-based research, people also want animal pain and distress to be minimized (2). In Canada, continuing improvements to the welfare of animals used in science have occurred over the past few decades partly because of the explicit adoption of a set of principles to guide the ethical evaluation of animal use. This is the “Three Rs” tenet — Replacement, Reduction and Refinement (3). The tenet is grounded in the premise that animals should be used only if a scientist’s best efforts to find a nonanimal alternative have failed, and that when animals are needed, only the most humane methods should be used on the smallest number of animals required to obtain valid information (4). Specifically, “Replacement alternatives” refers to methods that avoid or replace the use of animals in an area where animals would otherwise have been used, including both absolute replacements (replacing animals with inanimate systems, such as computer programs) and relative replacements (replacing more sentient animals, such as vertebrates, with animals that current scientific evidence indicates have a significantly lower potential for pain perception, such as some invertebrates). “Reduction alternatives” refers to any strategy that will result in fewer animals being used to obtain sufficient data to answer the research question, or in maximizing the information obtained per animal and thus potentially limiting or avoiding the subsequent use of additional animals, without compromising animal welfare. “Refinement alternatives” refers to the modification of husbandry or experimental procedures to minimize pain and distress, and to enhance the welfare of an animal used in science from the time it is born until its death (5). Today in Canada and internationally, the Three Rs ethic of animal use is an accepted part of the culture of animal-based science. In Canada, the Canadian Council on Animal Care (CCAC) is the national organization that has the responsibility for overseeing the care and use of animals in science. Through the overarching CCAC policy statement on: Ethics of animal investigation (4), the CCAC has incorporated adherence to the Three Rs tenet as the fundamental basis for the ethical oversight of scientific animal care and use. Briefly, the oversight process requires that use of animals for a scientific purpose (research, teaching, testing) is subject to review and approval by an animal care committee located at the institution where the animal use is proposed. To reflect the broader public, animal care committees are composed of not only scientists and veterinary professionals, but also non-users of animals from the local institution and community representatives. The CCAC also has a strong mandate to communicate with the Canadian public and to provide information to scientists to enable compliance with accepted standards. The CCAC has developed a new internet resource to communicate how the Three Rs tenet is implemented and to distribute related science-based resources: the “Three Rs Microsite” (5). The Microsite offers information on a wide range of topics related to Three Rs-Alternatives, for example Experimental Design, Telemetry, and Humane Killing; information on special topics such as Agricultural Research and Three Rs and Genetically Engineered Animals; plus a section with Additional Resources, including Species Specific Resources and a list of Three-Rs related journals.

Abstract:Two new acylamide metal-organic frameworks (MOFs), based on mixed N- and O-donor ligands, with 4-connected topologies have been obtained, namely poly[[μ2-N(1),N(4)-bis(pyridin-3-yl)terephthalamide]bis(μ3-4,4'-oxydibenzoato)dizinc(II)], [Zn2(C14H8O5)2(C18H14N4O2)]n, (1), and poly[[(μ2-benzene-1,4-dicarboxylato)[μ2-N(4),N(4')-bis(pyridin-3-yl)-[1,1'-biphenyl]-4,4'-dicarboxamide]dicadmium(II)] dihydrate], {[Cd(C8H4O4)(C24H18N4O2)]·2H2O}n, (2). Complex (1) is a 4-connected CdSO4 net with no interpenetration, where the Zn(II) cation is regarded as a 4-connecting node with square geometry. Complex (2) is a 4-connected dia net with threefold interpenetration, where the Cd(II) cation acts as a 4-connecting node with tetrahedral geometry. A survival-critical role for giant interneurons during predation

Abstract:Large axon-diameter descending neurons are metabolically costly but transmit information rapidly from sensory neurons in the brain to motor neurons in the nerve cord. They have thus endured as a common feature of escape circuits in many animal species where speed is paramount. Though often considered isolated command neurons triggering fast-reaction-time, all-or-none escape responses, giant neurons are just one of multiple parallel pathways enabling selection between behavioral alternatives. Such degeneracy among escape circuits makes it unclear if and how giant neurons benefit prey fitness. Here we competed flies with genetically-silenced Giant Fibers (GFs) against flies with functional GFs in an arena with wild-caught damselfly predators and find that GF silencing decreases prey survival. Kinematic analysis of damselfly attack trajectories shows that decreased prey survival fitness results from GF-silenced flies failing to escape during predator attack speeds and approach distances that would normally elicit successful escapes. When challenged with a virtual looming predator, fly GFs promote survival by enforcing selection of a short-duration takeoff sequence as opposed to reducing reaction time. Our findings support a role for the GFs in promoting prey survival by influencing action selection as a means to enhance escape performance during realistically complex predation scenarios.

Age-related changes in climbing behavior and neural circuit physiology in Drosophila

Abstract:Identifying the cellular and molecular basis for functional decline remains key to understanding aging. To this end, we have characterized age-dependent changes in climbing and the electrophysiology of the giant fiber circuitry in wild type (Wt) and mutant flies with altered lifespan (methuselah and fragile-X). Our data demonstrate a gradual decline in climbing in Wt and methuselah flies aged 5-45 days. In contrast, fragile-X flies climbed poorly even at 5 days and failed completely at 45 days. We then examined whether synaptic transmission to indirect flight muscles along the giant fiber circuit was altered with aging. At 5 days, the dorsal longitudinal muscle (DLM) in Wt flies followed high frequency stimulation well (at 130 Hz or above). At 35 and 45 days, these flies only followed 60-80 Hz. Methuselah flies did not follow stimuli as well as the Wt flies did at 5 and 25 days, but they were similar to Wt flies at older ages. Fragile-X flies responded poorly even at 5 days (40 Hz) and worsened at 35 days (30 Hz). Unlike DLMs, the tergotrochanteral muscle followed high frequency stimuli relatively well in all genotypes, suggesting that the peripheral interneuron along the DLM pathway or the DLM muscular synapse is prone to age-dependent functional decline. These studies reveal subcellular structures as potential targets of aging, indicating that the giant fiber pathway can be used as a model circuit for quantitative studies of aging in flies as well as fly models of age-related human neurological disorders.
Behavioral Metabolution - Metabolism Based Behavior Enables New Forms of Adaptation and Evolution

Abstract:Both metabolism and behavior play a key role in biological theory and artificial life modelling. Yet, despite their centrality there has been very little exploration of the relationship between these concepts and almost no exploration of how the interaction between the two could impact on evolution or instantiate alternative mechanisms for evolutionary processes. We present a simulation model of bacteria capable of metabolism-based chemotaxis: a minimal metabolic system capable of modulating behavior by influencing the probability of flagellar rotation (like in E. coli chemotaxis). We perform two illustrative experiments. In the first, the incorporation of a chemical compound into metabolism qualitatively improves the chemotactic strategy. In the second, an encounter with a specific chemical compound leads to a reaction that opens up a new metabolic pathway while automatically regulating chemotaxis towards that same compound. Both experiments illustrate the adaptive potential of metabolism-based behavior and can be used to explore the idea of “Behavioral Metabolution,” a co-evolutionary synergy between behavior and metabolism. We abstract some principles of behavioral metabolution and discuss its application to early prebiotic evolution.
Neuronal ON/OFF Motion Detection Circuits Underlying Looming-Evoked Escape Behavior in Drosophila

Abstract:Abstract In Drosophila , early visual processing of motion information segregates in separate ON and OFF pathways. These pathways have been studied in the context of local directional motion detection leading to the encoding of optic flow that provides visual information for flight stabilization. Less is known about their role in detecting impending collision and generating escape behaviors. ‘Looming’, the simulated approach of an object at constant speed towards an animal, provides a powerful stimulus eliciting jump escape behaviors in stationary flies. We presented looming stimuli mimicking the approach of either a dark object on a bright background or a light object on a dark background, while inactivating neurons belonging either to the ON- or the OFF-motion detection pathways by expressing the dominant Drosophila temperature-sensitive mutant shibire ts in different cells of the ON/OFF pathway. Inactivation of ON, respectively OFF, neurons led to selective decreases in escape behavior to light, resp. dark, looming stimuli. Quantitative analysis showed a nearly perfect splitting of these effects according to the ON/OFF type of the targeted neural populations. Our results suggest that Drosophila ON/OFF motion detection pathways play an important role in controlling jump escape responses according to looming stimulus polarity. They further imply that the biophysical circuits triggering Drosophila jump escape behaviors likely differ substantially from those characterized in other arthropods. Summary Inactivating fly neurons of the ON or OFF directional motion detection pathways during escape behavior selectively reduced jump responses to light and dark looming stimuli, respectively.

Abstract:Animals display selective escape behaviors when faced with environmental threats. Selection of the appropriate response by the underlying neuronal network is key to maximizing chances of survival, yet the underlying network mechanisms are so far not fully understood. Using synapse-level reconstruction of the Drosophila larval network paired with physiological and behavioral readouts, we uncovered a circuit that gates selective escape behavior for noxious light through acute and input-specific neuropeptide action. Sensory neurons required for avoidance of noxious light and escape in response to harsh touch, each converge on discrete domains of neuromodulatory hub neurons. We show that acute release of hub neuron-derived insulin-like peptide 7 (Ilp7) and cognate relaxin family receptor (Lgr4) signaling in downstream neurons are required for noxious light avoidance, but not harsh touch responses. Our work highlights a role for compartmentalized circuit organization and neuropeptide release from regulatory hubs, acting as central circuit elements gating escape responses.

Age-associated cardiac dysfunction in Drosophila melanogaster

Abstract:The fruit fly, Drosophila melanogaster, has served as a valuable model/organism for the study of aging and was the first organism possessing a circulatory system to have its genome completely sequenced. However, little is known about the function of the heartlike organ of flies during the aging process. We have developed methods for studying cardiac function in vivo in adult flies. Using 2 different cardiovascular stress methods (elevated ambient temperature and external electrical pacing), we found that maximal heart rate is significantly and reproducibly reduced with aging in Drosophila, analogous to observations in elderly humans. We also describe for the first time several other aspects of the cardiac physiology of young adult and aging Drosophila, including an age-associated increase in rhythm disturbances. These observations suggest that the study of declining cardiac function in aging flies may serve as a genetically tractable model for genome-wide mutational screening for genes that participate in or protect against cardiac aging and disease.
Fast deterministic construction of static dictionaries

Abstract:we consider the problem of constructing a static dictionary that supports access queries for a set of w-bit keys on a wbit RAM with an arithmetic instruction set consisting of addition, subtraction, multiplication, left and right shift, and bitwise boolean operations. We show that a static dictionary for n > 4 keys that occupies O(n) space and allows queries to be processed in O(loglog r-r) time can be constructed deterministically in O(n log n) time. No previous deterministic linear-space solution simultaneously achieved o(c) query time and n’+‘(‘) construction time. The actual result is more general and allows the query time to be lowered at the cost of increasing the construction time. We also apply known dynamization techniques ‘to derive a deterministic dynamic dictionary with a tradeoff between query time and update time.
The nicotinic acetylcholine receptor Dalpha7 is required for an escape behavior in Drosophila

Abstract:Acetylcholine is the major excitatory neurotransmitter in the central nervous system of insects. Mutant analysis of the Dalpha7 nicotinic acetylcholine receptor (nAChR) of Drosophila shows that it is required for the giant fiber-mediated escape behavior. The Dalpha7 protein is enriched in the dendrites of the giant fiber, and electrophysiological analysis of the giant fiber circuit showed that sensory input to the giant fiber is disrupted, as is transmission at an identified cholinergic synapse between the peripherally synapsing interneuron and the dorsal lateral muscle motor neuron. Moreover, we found that gfA1, a mutation identified in a screen for giant fiber defects more than twenty years ago, is an allele of Dalpha7. Therefore, a combination of behavioral, electrophysiological, anatomical, and genetic data indicate an essential role for the Dalpha7 nAChR in giant fiber-mediated escape in Drosophila.
A novel assay to evaluate action selection in escape behavior

Abstract:Background: How experience and individuality shape action selection remains a major question in neuroscience. Visually-evoked escape behavior within Drosophila melanogaster provides a robust model to study these mechanisms within neural circuits but requires novel assays to circumvent limitations of current behavior assays. Method: Here we describe and characterize a simple, low to moderate cost, and flexible assay for studying visually-evoked escape responses in tethered flies. This assay consists of a DLP projector, cylindrical rear projection screen, and an automated flight interruption motor all controlled within a MATLAB environment. Results: We find this assay effectively recapitulates fly behaviors previously observed in free behavior assays, and provides a novel opportunity to investigate the behavior of individual flies over the course of numerous stimulus presentations. Comparison to existing methods: Current Drosophila escape assays do not permit multiple stimulus presentations and can be highly complex and expensive to implement. Conclusions: This assay provides an effective system to further identify neural components and mechanisms underlying action selection within parallel sensorimotor pathways.
Age-Related Reduction of Recovery Sleep and Arousal Threshold in Drosophila.

Abstract:STUDY OBJECTIVES:Physiological studies show that aging affects both sleep quality and quantity in humans, and sleep complaints increase with age. Along with knowledge about the negative effects of poor sleep on health, understanding the enigmatic relationship between sleep and aging is important. Because human sleep is similar to Drosophila (fruit fly) sleep in many ways, we addressed the effects of aging on sleep in this model organism.METHODS:Baseline sleep was recorded in five different Drosophila genotypes raised at either 21°C or 25°C. The amount of sleep recovered was then investigated after a nighttime of sleep deprivation (12 h) and after chronic sleep deprivation (3 h every night for multiple nights). Finally, the effects of aging on arousal, namely, sensitivity to neuronal and mechanical stimuli, were studied.RESULTS:We show that fly sleep is affected by age in a manner similar to that of humans and other mammals. Not only do older flies of several genotypes have more fragmented sleep and reduced total sleep time compared to young flies, but older flies also fail to recover as much sleep after sleep deprivation. This suggests either lower sleep homeostasis and/or a failure to properly recover sleep. Older flies also show a decreased arousal threshold, i.e., an increased response to neuronal and mechanical wake-promoting stimuli. The reduced threshold may either reflect or cause the reduced recovery sleep of older flies compared to young flies after sleep deprivation.CONCLUSIONS:Further studies are certainly needed, but we suggest that the lower homeostatic sleep drive of older flies causes their decreased arousal threshold.
In vitro studies of the interactions of PuIV with blood proteins.

Abstract:The interaction between Pu239 and the constituents of the blood is a subject that has been pursued by a number of workers (1-4). It has been established that Pu239 is bound to plasma proteins (1-4). The identities of the particular blood proteins that bind plutonium are only tentatively known (1, 2), and they are thought to be A-globulins. The separation and identification of the proteins to which plutonium is bound is difficult, since neither the chemistry of proteins nor that of plutonium is simple. Plutonium can exist in four oxidation states in aqueous solution. The studies reported herein are limited to one oxidation state, PuIV, whereas earlier studies used PuVI (1, 2). This small highly charged ion (PuIV) essentially does not exist as such in systems of biological interest. Instead it binds with many anions, including OH(hydrolysis), to form chemical entities of lower net charge. Blood serum or plasma offers an enormous number of ionic binding sites. Thus, PuIV in blood is undoubtedly involved in an unknown number of unidentified competitive equilibria. These characteristics exclude the use of ion exchange in these studies, for the ion exchange system would introduce new competitive equilibria. Electrophoretic separation of the proteins is also excluded, since PuIV could be released from the protein and move independently under the appropriate (unknown) voltage gradient (5; also F. W. Bruenger, unpublished observations). The advent of gel-filtration as described by Flodin and Killander (6) has provided a new approach to this problem. This method of separating proteins is primarily mechanical in nature, in contrast with the chemical and electrical methods mentioned above, and hence it is much less likely to perturb the PuIV-protein system. The work reported here is a preliminary study of the separation of the proteins that bind PuIV, using the method of gel filtration.
Paired nanoinjection and electrophysiology assay to screen for bioactivity of compounds using the Drosophila melanogaster giant fiber system

Abstract:Screening compounds for in vivo activity can be used as a first step to identify candidates that may be developed into pharmacological agents. We developed a novel nanoinjection/electrophysiology assay that allows the detection of bioactive modulatory effects of compounds on the function of a neuronal circuit that mediates the escape response in Drosophila melanogaster. Our in vivo assay, which uses the Drosophila Giant Fiber System (GFS, Figure 1) allows screening of different types of compounds, such as small molecules or peptides, and requires only minimal quantities to elicit an effect. In addition, the Drosophila GFS offers a large variety of potential molecular targets on neurons or muscles. The Giant Fibers (GFs) synapse electrically (Gap Junctions) as well as chemically (cholinergic) onto a Peripheral Synapsing Interneuron (PSI) and the Tergo Trochanteral Muscle neuron (TTMn. The PSI to DLMn (Dorsal Longitudinal Muscle neuron) connection is dependent on Dα7 nicotinic acetylcholine receptors (nAChRs). Finally, the neuromuscular junctions (NMJ) of the TTMn and the DLMn with the jump (TTM) and flight muscles (DLM) are glutamatergic. Here, we demonstrate how to inject nanoliter quantities of a compound, while obtaining electrophysiological intracellular recordings from the Giant Fiber System and how to monitor the effects of the compound on the function of this circuit. We show specificity of the assay with methyllycaconitine citrate (MLA), a nAChR antagonist, which disrupts the PSI to DLMn connection but not the GF to TTMn connection or the function of the NMJ at the jump or flight muscles. Before beginning this video it is critical that you carefully watch and become familiar with the JoVE video titled "Electrophysiological Recordings from the Giant Fiber Pathway of D. melanogaster" from Augustin et al, as the video presented here is intended as an expansion to this existing technique. Here we use the electrophysiological recordings method and focus in detail only on the addition of the paired nanoinjections and monitoring technique.
Feedback inhibition by a descending GABAergic neuron regulates timing of escape behavior in Drosophila larvae

Abstract:Escape behaviors help animals avoid harm from predators and other threats in the environment. Successful escape relies on integrating information from multiple stimulus modalities (of external or internal origin) to compute trajectories toward safe locations, choose between actions that satisfy competing motivations, and execute other strategies that ensure survival. To this end, escape behaviors must be adaptive. When a Drosophila melanogaster larva encounters a noxious stimulus, such as the focal pressure a parasitic wasp applies to the larval cuticle via its ovipositor, it initiates a characteristic escape response. The escape sequence consists of an initial abrupt bending, lateral rolling, and finally rapid crawling. Previous work has shown that the detection of noxious stimuli primarily relies on class IV multi-dendritic arborization neurons (Class IV neurons) located beneath the body wall, and more recent studies have identified several important components in the nociceptive neural circuitry involved in rolling. However, the neural mechanisms that underlie the rolling-escape sequence remain unclear. Here, we present both functional and anatomical evidence suggesting that bilateral descending neurons within the subesophageal zone of D. melanogaster larva play a crucial role in regulating the termination of rolling and subsequent transition to escape crawling. We demonstrate that these descending neurons (designated SeIN128) are inhibitory and receive inputs from a second-order interneuron upstream (Basin-2) and an ascending neuron downstream of Basin-2 (A00c). Together with optogenetic experiments showing that co-activation of SeIN128 neurons and Basin-2 influence the temporal dynamics of rolling, our findings collectively suggest that the ensemble of SeIN128, Basin-2, and A00c neurons forms a GABAergic feedback loop onto Basin-2, which inhibits rolling and thereby facilitates the shift to escape crawling.

Inhibitory control of locomotor statistics in walking Drosophila

Abstract:In order to forage for food, many animals regulate not only specific limb movements but the statistics of locomotor behavior over time, switching between long-range dispersal and localized search depending on resource availability. How pre-motor circuits regulate such locomotor statistics is not clear. Here we analyze and model locomotor statistics in walking Drosophila, and their modulation by attractive food odor. Odor evokes three motor regimes in flies: baseline walking, upwind running during odor, and search behavior following odor loss. During search behavior, we find that flies adopt higher angular velocities and slower ground speeds, and tend to turn for longer periods of time in one direction. We further find that flies spontaneously adopt periods of different mean ground speed, and that these changes in state influence the length of odor-evoked runs. We next developed a simple model of neural locomotor control that suggests that contralateral inhibition plays a key role in regulating the statistical features of locomotion. As the fly connectome predicts decussating inhibitory neurons in the lateral accessory lobe (LAL), a pre-motor structure, we gained genetic access to a subset of these neurons and tested their effects on behavior. We identified one population of neurons whose activation induces all three signature of search and that bi-directionally regulates angular velocity at odor offset. We identified a second group of neurons, including a single LAL neuron pair, that bi-directionally regulate ground speed. Together, our work develops a biologically plausible computational architecture that captures the statistical features of fly locomotion across behavioral states and identifies potential neural substrates of these computations.
