Abstract:Dwarfism is one of the most important agronomic traits of rice.The genes underlying participate a series of important physiological and biochemical process.Cloning and functional analysis of the dwarfism related genes reveal the mechanisms for high yielding rice production.In the review,the concept of dwarfism and class of dwarf gene of rice were introduced,and the structure,function and relation with classical genetics and breeding values of the cloned genes were summarized.

Furthermore, due to special characteristics of transposons, transposons techniques will be a powerful tool for new gene discovery and gene functional analysis. This paper reviewed the developments of plant transposons in gene isolating and cloning, as well as its use in studying gene function in plant.

Abstract:Since the beginning of this century, microRNAs (miRNAs), which are tiny RNA molecules, have become one of the major research topics on gene expression regulation in both animals and plants. The major task of miRNA study is to elucidate how the miRNAs are expressed in vivo, how they exert regulatory effects on their targets, and how they can be qualitatively or quantitatively cloned. For these purposes, the methodology of miRNA study has been developed and significantly improved in recent years. The focus here is on a number of powerful methods for plant miRNA research including bioinformatics tools and experimental approaches being used for upstream or downstream analysis of miRNAs or miRNA cloning. Some discrepancies exist in the miRNA research methodology between plants and animals, for example, 5' modified RACE (Rapid Amplification of cDNA Ends) can be used for cleavage target validation only in plants. However, numerous common methods are shared by these two miRNA research areas. Thus, this review will enhance our understanding of miRNA research methodology in organisms.

Abstract:Fibrinogen is the blood coagulation factor I,with the catalysis of thrombin,it plays an important role in coagulation and hemostasis.Our rearch group successfully cloned fibrinogen α,β,γ gene by RT-PCR from human hepatic cells,then inseted it into prokaryotic expression vector,respectivelly.Expressed and purified of human fibrinogen α,β,γ protein are successful.The purified protein then was identified by SDS-PAGE and western blot.Our research result will promote the study of Chinese Han population fibrinogen gene polymorphism and haplotypes.The study provides an important experimental basis for the industrialization and technological base for the development of effective medical clinical trial of fibrin sealant.

It has been well recognized as a natural gene bank that conserves a lot of specific genes presently not available for extinct in the cultivated rice. There is an urgent need to explore these specific genes. Abstract:Plant disease resistance gene (R gene) is one of the hot spots in the field of molecular plaint pathology and plant genetic engineering. Cloning and functional analysis of R gene provide the basis for revealing the mechanisms of resistance and controlling the plaint disease. This article mainly summarized strategies of R gene cloning and the functions of R gene in resistance. It also discussed the development of strategies to optimize R gene mediated resistance through genetic engineering.

Abstract:In the past 20 years,cloning and functional studies on the genes causing human genetic disorders has been developing repidly.However,the function of most disease genes still remains unknown.This paper briefly describes the basic characteristics of inherited diseases,then reviews the progress in disease gene cloning and pathogenic mechanism,and two achievements were presented,one is that the fibroblast growth factor 9(FGF9) missense mutation(S99N) causes the multiple synostoses syndrome,another is that the disruption of palladin results in neural tube closure defects,herniation of intestine and liver,fetal liver atrophy,defects in definitive erythropoiesis,and embryonic lethality in mice.

Abstract:To clone genes of interest into suitable vectors is the first step to investigate their functions in vitro and in vivo. At the genome era, the sequences of more and more genes were decoded and available gradually. Therefore, it is critical to develop high efficient strategies for cloning genes of interest into different vectors to facilitate the functional analyses of them. In our previous studies, we created âCombinatorial strategyâ for DNA recombination. Here, I theoretically analyzed the procedure of DNA recombination, the mechanism of this strategy, and further gave suggestions and predictions for various ligation-dependent molecular cloning experiments.

Abstract:Gene therapy is of great promise to replace or counteract a defective gene thus curing a disease at its genetic origins.However, there have been many hur-dles to put this simple concept into practice for a long term.Since the end of last century, prosperous devel-opment of genetic engineering and vector technologies have advanced gene therapy in a diversity of diseases, including genetic disorders, cancer, and chronic infec-tions,etc.Recently,clinical trials of gene therapy have achieved breakthrough progress worldwide with remarka-ble therapeutic effects and safety record.These evi-dence demonstrate that gene therapy has the potential to bring innovation into traditional medicine for some pre-viously incurable conditions.In addition, gene editing and stem cell technologies are also contributing to revi-talization and advances of gene therapy.In this context we review the efficacy and safety achievements during the development of gene therapy,as well as ethical con-cerns related to current clinical trials, and further dis-cuss the research hotspots,development prospects,and challenges for gene therapy.

Abstract:Microbial natural products (NPs) are a major source of pharmacological agents. Most NPs are synthesized from specific biosynthetic gene clusters (BGCs). With the rapid increase of sequenced microbial genomes, large numbers of NP BGCs have been discovered, regarded as a treasure trove of novel bioactive compounds. However, many NP BGCs are silent in native hosts under laboratory conditions. In order to explore their therapeutic potential, a main route is to activate these silent NP BGCs in heterologous hosts. To this end, the first step is to accurately and efficiently capture these BGCs. In the past decades, a large number of effective technologies for cloning NP BGCs have been established, which has greatly promoted drug discovery research. Herein, we describe recent advances in strategies for BGC cloning, with a focus on the preparation of high-molecular-weight DNA fragment, selection and optimization of vectors used for carrying large-size DNA, and methods for assembling targeted DNA fragment and appropriate vector. The future direction into novel, universal, and high-efficiency methods for cloning NP BGCs is also prospected.

Abstract:Carotenoids have a range of diverse biological functions and actions, especially playing an important role in human health with provitamin A activity, anti-cancer activity, enhancing immune ability and so on. Human body can't synthesis carotenoids by itself and must absorb them from outside. However, carotenoid contents in many plant are very low, and many kinds of carotenoid are difficult to produce by chemical ways. With the elucidation of carotenoid biosynthetic pathway and cloning genes of relative enzymes from microorganisms and higher plants, it is possible to regulate carotenoid biosynthesis via genetic engineering. This article reviews gene cloning of carotenoid biosynthetic enzymes in microorganisms and higher plants, and advances in the studies of carotenoid production in heterologous microorganisms and crop plants using gene-manipulated carotenoid biosynthesis.

Abstract:Since the discovery of penicillin, natural products and their derivatives have been a valuable resource for drug discovery. With recent development of genome mining approaches in the post-genome era, a great number of natural product biosynthetic gene clusters (BGCs) have been identified and these can potentially be exploited for the discovery of novel natural products that can find application as pharmaceuticals. Since many BGCs are silent or do not express in native hosts under laboratory conditions, heterologous expression of BGCs in genetically tractable hosts becomes an attractive route to activate these BGCs to discover the corresponding products. Here, we highlight recent achievements in cloning and discovery of natural product biosynthetic pathways via intact BGC capturing, and discuss the prospects of high-throughput and multiplexed cloning of rational-designed gene clusters in the future.
