Abstract:Traumatic spinal cord injury (SCI) is one of the most serious injuries, which will cause severe sensory and motor obstacles. The current effective treatment methods remain a worldwide challenge for both clinic and research. The lack of electrical signal conduction and cell growth microenvironment in SCI is one of the main obstacles that inhibit spinal cord regeneration. Herein, we report a multifunctional polycitrate-based nanocomposite hydrogel (PMEAC) scaffold with spinal cord-biomimetic mechanical and electrical properties for promoting spinal cord repair and regeneration. PMEAC scaffolds were prepared using poly(citrate-maleic)-epsilon-polylysine (PME) and multi-walled carbon nanotubes (MWCNT) by the facile self-crosslinking route. PMEAC scaffolds possessed multifunctional properties including injectability, self-healing ability, tissue-adhesiveness, broad-spectrum antibacterial property and UV-shielding performance, especially the biomimetic mechanical modulus (G' = 291.69 Pa) and electroconductivity (0.15 S/m) with spinal cord tissue. PMEAC scaffolds also exhibited good cytocompatibility, hemocompatibility and biodegradability. In vivo results from a spinal cord defect model showed that PMEAC scaffolds could significantly improve locomotion recovery (BBB score 6-7), reduce inflammation, promote remyelination and axon regeneration after SCI. This study showed that multifunctional bioactive biomaterials with mechanics-electro-biomimetic properties should be the effective strategy to promote SCI recovery and regeneration.
3D Bioprinted Conductive Spinal Cord Biomimetic Scaffolds for Promoting Neuronal Differentiation of Neural Stem Cells and Repairing of Spinal Cord Injury

Abstract:Spinal cord injury (SCI) is a terrible injury of the central nervous system for which there is no ideal treatment in clinic so far. The spinal cord biomimetic scaffolds which mimic the shape and structure of native spinal cord tissue, could effectively promote the repair of SCI. However, current biomimetic scaffolds do not mimic the biological functions, especially the electrical conductivity, of spinal cord tissue, which largely limits the therapeutic effect of SCI. In this study, we have developed novel conductive hydrogels based on gelatin methacrylate (GelMA), hyaluronic acid methacrylate (HAMA) and poly(3,4-ethylenedioxythiophene): sulfonated lignin (PEDOT:LS). The incorporation of PEDOT:LS into GelMA/HAMA hydrogel matrix significantly improved the conductivity of the hydrogels. By precisely regulating the light curing time, the conductive hydrogels showed mechanical properties similar to native spinal cord tissues. Then, the conductive biomimetic scaffolds were fabricated by 3D bioprinting, and the neural stem cells (NSCs) encapsulated in the scaffolds exhibited good survival rate (higher than 90%). Compared to the non-conductive scaffolds reported, the conductive scaffolds remarkably promoted neuronal differentiation of NSCs in vitro. In a rat spinal cord complete transection model, the conductive biomimetic scaffold dramatically promoted the recovery of hindlimb motor function. Immunofluorescence staining results further showed that the conductive biomimetic scaffold effectively promoted the regeneration of neurons at the injury site, reduced the deposition of glial scar, and promoted nerve axon regeneration and myelination. Overall, the 3D bioprinting of the conductive spinal cord biomimetic scaffolds developed in this work represents a promising approach for the stem cell-based treatment of SCI.

Abstract:Axon degeneration and neurons after spinal cord injury (SCI) can lead to the serious sensory and motor dysfunctions. The effective treatment of SCI still remains a grand clinical challenge for current medicine. Development of biological scaffolds with drugs are of great significance to provide a supportive environment for re-establishment of cellular connections and growth of new tissues after SCI. In this study, we presented the synthesis of strontium-doped hydroxyapatite (Sr-HAP) with high biocompatibility and osteoconductivity, which was a promising biomaterial for repair of hard/soft tissues. The Sr-HAP nanoparticles was obtained by a classical precipitation process followed by the loading on macroporous chitosan (CS) scaffold. The CS/Sr-HAP nanocomposite was characterized with SEM, XRD, and FT-IR techniques, which was used to carry drugs for guiding nerve regeneration. We have performed the in vivo experiment by transplanting the scaffolds carried with chymopapain into an acute rat with SCI model. The robust neurogenesis of neural stem cells (NSCs) was observed, which then transformed into the functional neurons and the injured gap can be reconnected. This study suggests that the CS/Sr-HAP nanocomposite scaffold has great potential for promoting the spinal cord repair. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Spinal cord injury (SCI) results in devastating consequences for the motor and sensory function of patients due to neuronal loss and disrupted neural circuits, confronting poor prognosis and lack of effective therapies. A new therapeutic strategy is urgently required. Here, human amniotic epithelial cells (hAEC), featured with immunocompatibility, non-tumorgenicity and no ethical issues, were induced into neural-like cells by a compound cocktail, as evidenced with morphological change and the expression of neural cell markers. Interestingly, the hAEC-neural-like cells maintain the characteristic of low immunogenicity as hAEC. Aiming at SCI treatment in vivo, we constructed a 3D-printed GelMA hydrogel biomimetic spinal cord scaffold with micro-channels, in which hAEC-neural-like cells were well-induced and grown. In a rat full transection SCI model, hAEC-neural-like cell scaffolds that were implanted in the lesion demonstrated significant therapeutic effects; the neural circuit and hindlimb locomotion were partly recovered compared to little affection in the SCI rats receiving an empty scaffold or a sham implantation operation. Thus, the establishment of hAEC-neural-like cell biomimetic scaffolds may provide a safe and effective treatment strategy for SCI.
Integrated printed BDNF/collagen/chitosan scaffolds with low temperature extrusion 3D printer accelerated neural regeneration after spinal cord injury

Abstract:Abstract Recent studies have shown that 3D printed scaffolds integrated with growth factors can guide the growth of neurites and promote axon regeneration at the injury site. However, heat, organic solvents or cross-linking agents used in conventional 3D printing reduce the biological activity of growth factors. Low temperature 3D printing can incorporate growth factors into the scaffold and maintain their biological activity. In this study, we developed a collagen/chitosan scaffold integrated with brain-derived neurotrophic factor (3D-CC-BDNF) by low temperature extrusion 3D printing as a new type of artificial controlled release system, which could prolong the release of BDNF for the treatment of spinal cord injury (SCI). Eight weeks after the implantation of scaffolds in the transected lesion of T10 of the spinal cord, 3D-CC-BDNF significantly ameliorate locomotor function of the rats. Consistent with the recovery of locomotor function, 3D-CC-BDNF treatment could fill the gap, facilitate nerve fiber regeneration, accelerate the establishment of synaptic connections and enhance remyelination at the injury site.

Abstract:Tissue engineering has brought new possibilities for the treatment of spinal cord injury. Two important components for tissue engineering of the spinal cord include a suitable cell source and scaffold. In our study, we investigated induced mouse embryonic fibroblasts (MEFs) directly reprogrammed into neural stem cells (iNSCs), as a cell source. Three-dimensional (3D) electrospun poly (lactide-co-glycolide)/polyethylene glycol (PLGA-PEG) nanofiber scaffolds were used for iNSCs adhesion and growth. Cell growth, survival and proliferation on the scaffolds were investigated. Scanning electron microscopy (SEM) and nuclei staining were used to assess cell growth on the scaffolds. Scaffolds with iNSCs were then transplanted into transected rat spinal cords. Two or 8 weeks following transplantation, immunofluorescence was performed to determine iNSC survival and differentiation within the scaffolds. Functional recovery was assessed using the Basso, Beattie, Bresnahan (BBB) Scale. Results indicated that iNSCs showed similar morphological features with wild-type neural stem cells (wt-NSCs), and expressed a variety of neural stem cell marker genes. Furthermore, iNSCs were shown to survive, with the ability to self-renew and undergo neural differentiation into neurons and glial cells within the 3D scaffolds in vivo. The iNSC-seeded scaffolds restored the continuity of the spinal cord and reduced cavity formation. Additionally, iNSC-seeded scaffolds contributed to functional recovery of the spinal cord. Therefore, PLGA-PEG scaffolds seeded with iNSCs may serve as promising supporting transplants for repairing spinal cord injury (SCI).

Indeed, cell transplantation and bio-scaffold implantation are considered to be effective methods for neural regeneration. This study was designed to fabricate a type of three-dimensional collagen/silk fibroin scaffold (3D-CF) with cavities that simulate the anatomy of normal spinal cord. This scaffold allows cell growth in vitro and in vivo. To observe the effects of combined transplantation of neural stem cells (NSCs) and 3D-CF on the repair of spinal cord injury. Forty Sprague-Dawley rats were divided into four groups: sham (only laminectomy was performed), spinal cord injury (transection injury of T10 spinal cord without any transplantation), 3D-CF (3D scaffold was transplanted into the local injured cavity), and 3D-CF + NSCs (3D scaffold co-cultured with NSCs was transplanted into the local injured cavity. Neuroelectrophysiology, imaging, hematoxylin-eosin staining, argentaffin staining, immunofluorescence staining, and western blot assay were performed. Apart from the sham group, neurological scores were significantly higher in the 3D-CF + NSCs group compared with other groups. Moreover, latency of the 3D-CF + NSCs group was significantly reduced, while the amplitude was significantly increased in motor evoked potential tests. The results of magnetic resonance imaging and diffusion tensor imaging showed that both spinal cord continuity and the filling of injury cavity were the best in the 3D-CF + NSCs group. Moreover, regenerative axons were abundant and glial scarring was reduced in the 3D-CF + NSCs group compared with other groups. These results confirm that implantation of 3D-CF combined with NSCs can promote the repair of injured spinal cord. Abstract:OBJECTIVES:Biomaterials are used to aid in the regeneration of damaged tissue and in promotion of axonal guidance following spinal cord injury (SCI). In the present study, electrospun composite poly(hydroxybutyrate-cohydroxyvalerate) (PHBV), poly(lactic acid) (PLA), and collagen (Col) nanofibrous scaffolds were applied to determine their roles in neural regeneration and recovery in a rat model of SCI.METHODS:The morphological and chemical properties of the electrospun scaffolds were investigated. The growth and proliferation of astrocytes on the scaffolds were assessed by MTT assay. The differentiation and gene expression of astrocytes on the scaffolds were measured by immunofluorescence and quantitative real-time polymerase chain reaction (q-PCR) assays. In a rat spinal cord hemisection model with 3-mm defects, 80 Sprague-Dawley rats were randomly divided into five groups: Sham group, SCI group, SCI+PHBV/PLA group, SCI+PHBV/PLA/Col (70:30) group, and SCI+PHBV/PLA/Col (50:50) group. The Basso-Beattie-Bresnahan (BBB) scores were evaluated every week postsurgery, and (immuno) histological and protein analyses were performed on specimens at 8 weeks.RESULTS:PHBV/PLA/Col scaffolds strongly inhibited the activation of astrocytes without decreasing their proliferation. qPCR assays revealed significant increases in the expression of brain lipid-binding protein (BLBP), glutamate transporter 1 (GLT-1) and S100 calcium-binding protein B (S100-β), but decreases in the expression of glial fibrillary acidic protein (GFAP), chondroitin sulphate sulfate proteoglycan (CSPG), neurocan, and phosphacan in the PHBV/PLA/Col scaffold group. In a series of in vivo experiments, PHBV/PLA/Col scaffold-treated SCI groups showed significant reductions in the numbers of CD68- and GFAP-immunopositive astrocytes within the interface of the remodeled tissue layer, but increased expression of NF-200 in residual neurons with better locomotor functional recovery. However, there were no significant differences between the PHBV/PLA/Col (70:30) and PHBV/PLA/Col (50:50) groups, except in BBB scores.CONCLUSIONS:PHBV/PLA/Col nanofibrous scaffolds were biocompatible and significantly promoted astrocyte differentiation but decreased astrocyte activation. The topographic structures of the PHBV/PLA/Col (70:30 and 50:50) nanofibers were favorable for neural regeneration due to a decrease in astrogliosis in SCI rats.

Abstract:Abstract Accumulating research has indicated that the transplantation of combined stem cells and scaffolds is an effective method for spinal cord injury (SCI). The development of three-dimensional (3D) bioprinting technology can make the 3D scaffolds combined with cells more accurate and effective for SCI treatment. However, unmyelinated newborn nerve fibers have no nerve signaling conduction, hampering recovery of motor function. In this study, we designed and printed a type of sodium alginate/gelatin scaffold loaded with neural stem cells and oligodendrocytes, which were involved in the formation of the myelin sheaths of neural cell axons. In order to observe the effectiveness of this 3D bioprinting scaffold, we transplanted it into the completely transected rat spinal cord, and then immunofluorescence staining, hematoxylin–eosin staining and behavioral assessment were performed. The results showed that this 3D bioprinting scaffold markedly improved the hindlimb motor function and promoted nerve regeneration. These findings suggested that this novel 3D bioprinting scaffold was a good carrier for cells transplantation, thereby enhancing spinal cord repair following injury.

Abstract:The disability rate of spinal cord injury (SCI) is extremely high, and stem cell inhibition is one of the most effective schemes in treating the spinal cord, but the survival rate is extremely low after stem cell transplantation, so it cannot be widely used in clinic. Studies have revealed that loading stem cells with biological scaffolds can effectively improve the survival rate and effect after stem cell transplantation. Therefore, this research was devised to analyze the repair effect of thiolated chitosan nanocarriers scaffold carrying de-epithelized human amniotic epithelial cells (HAECs) on SCI. And we used thiolated chitosan as nanocarriers, aiming to provide a reliable theoretical basis for future clinical practice. Through experiments, we concluded that the Tarlov and BBB scores of rats with SCI were raised under the intervention of thiolated chitosan carrying HAECs, while the inflammatory factors in serum, oxidative stress reaction in spinal cord tissue, apoptosis rate of nerve cells, and autophagy protein expression were all suppressed. Thus, the thiolated chitosan carrying HAECs may be applied to treat SCI by suppressing autophagy protein expression, oxidative stress response, and release of inflammatory factors in spinal cord tissue, which may be a new clinical therapy for SCI in the future. Even though we cannot understand exactly the therapeutic mechanism of thiolated chitosan carrying HAECs for SCI, the real clinical application of thiolated chitosan carrying HAECs needs to be confirmed by human experiments.

Abstract:Biotechnology products offer both problems and opportunities for pharmacists. On one hand, they are expensive to purchase and keep on inventory. Typical compensation for outpatient prescriptions does not offset the cost of maintaining the inventory, or providing the education and training that patients often need to use biotechnology products properly. On the other hand, there are issues related to proper storage, preparation, and administration for which pharmacists are well prepared to address. Pharmacists are also convenient, trusted, and provide service at relatively low cost. Examples of special services that pharmacists can provide to improve the use of biotechnology products include patient education, injection clinics, provision of medical supplies, and predrawing syringes for patients. Patients are often sent to many providers for these services, resulting in inconvenience and fragmentation of care. If new compensation methods can be established to support more comprehensive pharmacy services, the use of biotechnology products by patients will improve.

Abstract:Abstract Although implantation of biomaterials carrying mesenchymal stem cells (MSCs) is considered as a promising strategy for ameliorating neural function after spinal cord injury (SCI), there are still some challenges including poor cell survival rate, tumorigenicity and ethics concerns. The performance of the secretome derived from MSCs was more stable, and its clinical transformation was more operable. Cytokine antibody array demonstrated that the secretome of MSCs contained 79 proteins among the 174 proteins analyzed. Three-dimensional (3D) printed collagen/silk fibroin scaffolds carrying MSCs secretome improved hindlimb locomotor function according to the Basso–Beattie–Bresnahan scores, the inclined-grid climbing test and electrophysiological analysis. Parallel with locomotor function recovery, 3D printed collagen/silk fibroin scaffolds carrying MSCs secretome could further facilitate nerve fiber regeneration, enhance remyelination and accelerate the establishment of synaptic connections at the injury site compared to 3D printed collagen/silk fibroin scaffolds alone group according to magnetic resonance imaging, diffusion tensor imaging, hematoxylin and eosin staining, Bielschowsky’s silver staining, immunofluorescence staining and transmission electron microscopy. These results indicated the implantation of 3D printed collagen/silk fibroin scaffolds carrying MSCs secretome might be a potential treatment for SCI.

Abstract:A simple method for the fabrication of tunable surfaces with wettability that can be manipulated rapidly and reversibly from superhydrophilicity to superhydrophobicity is reported. A polyelectrolyte multilayer (PEM) was deposited on a rough substrate, and via ion exchange of the counterion in the PEM, the water contact angle of the surface was switched between <5 degrees and 164 degrees. By controlling the ion-exchange kinetics, on the same surface, completely erasable, rewritable gradient wettability from superhydrophilicity to superhydrophobicity was conveniently achieved.
Implantable 3D printed multiplexed microtunnels for spinal cord injury treatment

Abstract:3D printed scaffolds offer a promising strategy for treating spinal cord injury (SCI). Here we present an innovative biotechnological approach for free-form 3D printing of scaffolds with a biomimetic architecture at a spatial resolution of up to a micrometer, designed for implantation in treatment of SCI in Wistar rats. The fabrication of scaffolds was based on 2-photon photopolymerization of organic polymers and was scalable to lesion geometries. The scaffolds were implemented as multiple densely packed squared parallel microtunnels (50 um per side) running their entire length. These microtunnels are separated by thin walls (5-10 um), rendering the scaffolds nearly hollow while maximizing their internal surface area. This design provides an optimal substrate, spatially aligned in the rostro-caudal direction, to support axonal and vascular ingrowth. We have found that the scaffolds, implanted in the excision of the lateral half-fragment of the spinal cord at the low thoracic level demonstrated excellent integration with surrounding tissue without the formation of a significant gliofibrous scar. Myelinated axons and oligodendrocytes, as well as vessels were observed in each microtunnel of the implanted scaffolds in 12 weeks after the operation with at least 1000 axons regenerating in the scaffold throughout its whole length. The treatment significantly improved motor function and reduced spasticity in the ipsilateral paretic limb by 8th week, with recovery sustained for at least 20 weeks. Thus, 3D oriented hollow scaffolds having a large internal surface area and direct continues microtunnels, effectively reducing axonal dispersion, mimic natural structure of the recipient tissue and create conditions for enhancing spinal cord regeneration and recovery of the motor function of the paretic limb.

Functional Multichannel Poly(Propylene Fumarate)-Collagen Scaffold with Collagen-Binding Neurotrophic Factor 3 Promotes Neural Regeneration after Transected Spinal Cord Injury

Abstract:Many factors contribute to the poor axonal regrowth and ineffective functional recovery after spinal cord injury (SCI). Biomaterials have been used for SCI repair by promoting bridge formation and reconnecting the neural tissue at the lesion site. The mechanical properties of biomaterials are critical for successful design to ensure the stable support as soon as possible when compressed by the surrounding spine and musculature. Poly(propylene fumarate) (PPF) scaffolds with high mechanical strength have been shown to provide firm spatial maintenance and to promote repair of tissue defects. A multichannel PPF scaffold is combined with collagen biomaterial to build a novel biocompatible delivery system coated with neurotrophin-3 containing an engineered collagen-binding domain (CBD-NT3). The parallel-aligned multichannel structure of PPF scaffolds guide the direction of neural tissue regeneration across the lesion site and promote reestablishment of bridge connectivity. The combinatorial treatment consisting of PPF and collagen loaded with CBD-NT3 improves the inhibitory microenvironment, facilitates axonal and neuronal regeneration, survival of various types of functional neurons and remyelination and synapse formation of regenerated axons following SCI. This novel treatment strategy for SCI repair effectively promotes neural tissue regeneration after transected spinal injury by providing a regrowth-supportive microenvironment and eventually induces functional improvement.

Abstract:This paper investigates the experimental implementation and detection of rotor faults in permanent magnet synchronous machines. Methods are shown how to experimentally introduce static and dynamic eccentricities and broken magnet cases. A new magnet flux estimation that does not require the measurement of the rotor position or speed is developed. The detection of these and other rotor faults by measuring only the stator currents and voltages are shown experimentally. The paper concludes by contributing a description of a condition-monitoring scheme for detection rotor faults

Abstract:In spinal cord injury (SCI) therapy, glial scarring formed by activated astrocytes is a primary problem that needs to be solved to enhance axonal regeneration. In this study, we developed and used a collagen scaffold for glial scar replacement to create an appropriate environment in an SCI rat model and determined whether neural plasticity can be manipulated using this approach. We used four experimental groups, as follows: SCI-collagen scaffold, SCI control, normal spinal cord-collagen scaffold, and normal control. The collagen scaffold showed excellent in vitro and in vivo biocompatibility. Immunofluorescence staining revealed increased expression of neurofilament and fibronectin and reduced expression of glial fibrillary acidic protein and anti-chondroitin sulfate in the collagen scaffold-treated SCI rats at 1 and 4 weeks post-implantation compared with that in untreated SCI control. This indicates that the collagen scaffold implantation promoted neuronal survival and axonal growth within the injured site and prevented glial scar formation by controlling astrocyte production for their normal functioning. Our study highlights the feasibility of using the collagen scaffold in SCI repair. The collagen scaffold was found to exert beneficial effects on neuronal activity and may help in manipulating synaptic plasticity, implying its great potential for clinical application in SCI.

Abstract:Regenerative healing of spinal cord injury poses an ongoing medical challenge by causing persistent neurological impairment and a significant socioeconomic burden. The complexity of spinal cord tissue presents hurdles to successful regeneration following injury, due to the difficulty of forming a biomimetic structure that faithfully replicates native tissue using conventional tissue engineering scaffolds. 3D bioprinting is a rapidly evolving technology with unmatched potential to create 3D biological tissues with complicated and hierarchical structure and composition. With the addition of biological additives such as cells and biomolecules, 3D bioprinting can fabricate preclinical implants, tissue or organ-like constructs, and in vitro models through precise control over the deposition of biomaterials and other building blocks. This review highlights the characteristics and advantages of 3D bioprinting for scaffold fabrication to enable spinal cord injury repair, including bottom-up manufacturing, mechanical customization, and spatial heterogeneity. This review also critically discusses the impact of various fabrication parameters on the efficacy of spinal cord repair using 3D bioprinted scaffolds, including the choice of printing method, scaffold shape, biomaterials, and biological supplements such as cells and growth factors. High-quality preclinical studies are required to accelerate the translation of 3D bioprinting into clinical practice for spinal cord repair. Meanwhile, other technological advances will continue to improve the regenerative capability of bioprinted scaffolds, such as the incorporation of nanoscale biological particles and the development of 4D printing.

Abstract:Current methods for bioprinting functional tissue lack appropriate biofabrication techniques to build complex 3D microarchitectures essential for guiding cell growth and promoting tissue maturation1. 3D printing of central nervous system (CNS) structures has not been accomplished, possibly owing to the complexity of CNS architecture. Here, we report the use of a microscale continuous projection printing method (μCPP) to create a complex CNS structure for regenerative medicine applications in the spinal cord. μCPP can print 3D biomimetic hydrogel scaffolds tailored to the dimensions of the rodent spinal cord in 1.6 s and is scalable to human spinal cord sizes and lesion geometries. We tested the ability of µCPP 3D-printed scaffolds loaded with neural progenitor cells (NPCs) to support axon regeneration and form new ‘neural relays’ across sites of complete spinal cord injury in vivo in rodents1,2. We find that injured host axons regenerate into 3D biomimetic scaffolds and synapse onto NPCs implanted into the device and that implanted NPCs in turn extend axons out of the scaffold and into the host spinal cord below the injury to restore synaptic transmission and significantly improve functional outcomes. Thus, 3D biomimetic scaffolds offer a means of enhancing CNS regeneration through precision medicine.

Abstract:Spinal cord injury is a permanent destructive disease that causes devastating neurologic deficits and disability. Long-term complications are associated with low prognosis, mortality, and decreased quality of life. The functional recovery depends on the regeneration of neurons and the growth of medullated axons. Single treatment strategies, including cell transplantation, cannot adapt to a changeable microenvironment. Patients with spinal cord injuries need more effective, long-term, and stable treatment options. Therefore, we investigated the benefit of a combined-tissue engineering strategy by loading homologous bone mesenchymal stem cells (BMSCs) and Schwann cells in three-dimensional (3D) scaffolds. We placed BMSCs and Rat Schwann cells (RSCs) in specific spatial arrangements using cell gravity and the diffusion effect to promote the formation of intercellular connections and cell-directed differentiation. This novel bioengineering system allowed us to control multiple factors, including cell types, cell relative position, and axon growth direction in the scaffold. Our system facilitated motor function recovery by enhancing tissue mimicry and allowing the reconstruction of medullated axons. This new 3D-integrated printing platform is multi-function and can simulate biomimetic tissue using different types of materials and multi-cells scaffolds. We believe that this study can help promote the clinical development and application of 3D printing in the field of regenerative medicine.
