Abstract:Strontium ions (Sr(2+)) are known to prevent osteoporosis and also encourage bone formation. Such twin requirements have motivated researchers to develop Sr-substituted biomaterials for orthopaedic applications. The present study demonstrates a new concept of developing Sr-substituted Mg3(PO4)2 - based biodegradable scaffolds. In particular, this work reports the fabrication, mechanical properties with an emphasis on strength reliability as well as in vitro degradation of highly biodegradable strontium-incorporated magnesium phosphate cements. These implantable scaffolds were fabricated using three-dimensional powder printing, followed by high temperature sintering and/or chemical conversion, a technique adaptable to develop patient-specific implants. A moderate combination of strength properties of 36.7MPa (compression), 24.2MPa (bending) and 10.7MPa (tension) were measured. A reasonably modest Weibull modulus of up to 8.8 was recorded after uniaxial compression or diametral tensile tests on 3D printed scaffolds. A comparison among scaffolds with varying compositions or among sintered or chemically hardened scaffolds reveals that the strength reliability is not compromised in Sr-substituted scaffolds compared to baseline Mg3(PO4)2. The micro-computed tomography analysis reveals the presence of highly interconnected porous architecture in three-dimension with lognormal pore size distribution having median in the range of 17.74-26.29μm for the investigated scaffolds. The results of extensive in vitro ion release study revealed passive degradation with a reduced Mg(2+) release and slow but sustained release of Sr(2+) from strontium-substituted magnesium phosphate scaffolds. Taken together, the present study unequivocally illustrates that the newly designed Sr-substituted magnesium phosphate scaffolds with good strength reliability could be used for biomedical applications requiring consistent Sr(2+)- release, while the scaffold degrades in physiological medium. Statement of significance: The study investigates the additive manufacturing of scaffolds based on different strontium-substituted magnesium phosphate bone cements by means of three-dimensional powder printing technique (3DPP). Magnesium phosphates were chosen due to their higher biodegradability compared to calcium phosphates, which is due to both a higher solubility as well as the absence of phase changes (to low soluble hydroxyapatite) in vivo. Since strontium ions are known to promote bone formation by stimulating osteoblast growth, we aimed to establish such a highly degradable magnesium phosphate ceramic with an enhanced bioactivity for new bone ingrowth. After post-processing, mechanical strengths of up to 36.7MPa (compression), 24.2MPa (bending) and 10.7MPa (tension) could be achieved. Simultaneously, the failure reliability of those bioceramic implant materials, measured by Weibull modulus calculations, were in the range of 4.3-8.8. Passive dissolution studies in vitro proved an ion release of Mg(2+) and PO4(3-) as well as Sr(2+), which is fundamental for in vivo degradation and a bone growth promoting effect. In our opinion, this work broadens the range of bioceramic bone replacement materials suitable for additive manufacturing processing. The high biodegradability of MPC ceramics together with the anticipated promoting effect on osseointegration opens up the way for a patient-specific treatment with the prospect of a fast and complete healing of bone fractures.
Effect of Foreign Ion Substitution and Micropore Tuning in Robocasting Single-Phase Bioceramic Scaffolds on the Physicochemical Property and Vascularization

Abstract:The inorganic powder slurry extrusion printing technique known as robocasting is an interesting method to fabricate complex porous architectures whereby feedstocks containing organic binders and powders are printed and the resulting scaffolds are subjected to sintering. A major limiting factor of this technique is the simultaneous tailoring of vascularization efficacy and osteogenic activity, usually done by adding the secondary phase in the organic slurry before the writing step. Mechanical mixing of biphasic powders is required to avoid compromising the biological performance and physical defects caused by significantly different physicochemical properties. This study addresses this issue by developing a selective ion doping and microstructure tuning for the production of bioceramic scaffolds with a binozzle robocasting process. Different metal ions (Sr2+, Mg2+) were doped into wollastonite (CaSiO3; CSi) powders considering the mechanical stability and bioactive enhancement of the bioceramic scaffolds. Subsequently, the Mg-doped CSi slurries were used as shell-nozzle feedstocks added with 5, 10, and 15 μm diameter polystyrene microbeads that allowed shell-layer micropore production in pore struts during sintering. Finally, the most promising pore-strut microstructures and mechanical evolution of scaffolds were evaluated, and especially the enhanced fibrovascularization potential was confirmed in dorsal muscle embedding model in rabbits. This study may open an avenue to designing multiproperty-tuned macro- and microporous bioceramics for bone regenerative medicine, especially in challenging bone defect conditions.
3D-Printed Bioceramic Scaffolds with High Strength and High Precision

Abstract:Due to the increasing cases of bone damage and bone graft demand, bone-repair technology has great social and economic benefits and the manufacturing of artificial bone implants has become a focus in the domain of regenerative therapy. Considering that the traditional manufacturing process cannot effectively control the overall size of the scaffold, the diameter and shape of micropores, and the interoperability of micropores, 3D printing technology has emerged as a focal point of research within the realm of bone tissue engineering. However, the printing accuracy of extrusion-based biological 3D printing techniques is low. In this research, we utilized three-dimensional printing technology to develop high-precision magnesium-containing silicate (CSi-Mg) scaffolds. The precision of this innovative method was scrutinized and the influence of pore size on scaffold strength was systematically analyzed. Furthermore, the influence of the pore architecture on the sidewalls of these 3D-printed scaffolds was evaluated in terms of mechanical properties. The CSi-Mg scaffold, post a 3-week immersion in a simulated body of fluid, demonstrated a high modulus of elasticity (exceeding 404 MPa) and significant compressive strength (beyond 47 MPa). Furthermore, it exhibited commendable bioactivity and biodegradability. These results suggest that the high-precision 3D-printed CSi-Mg scaffolds hold great promise for addressing challenging bone defect cases.
3D printing of strontium-enriched biphasic calcium phosphate scaffolds for bone regeneration

Abstract:Calcium phosphate (CaP) scaffolds doping with therapeutic ions are one of the focuses of recent bone tissue engineering research. Among the therapeutic ions, strontium stands out for its role in bone remodeling. This work reports a simple method to produce Sr-doped 3D-printed CaP scaffolds, using Sr-doping to induce partial phase transformation from β-tricalcium phosphate (β-TCP) to hydroxyapatite (HA), resulting in a doped biphasic calcium phosphate (BCP) scaffold. Strontium carbonate (SrCO3) was incorporated in the formulation of the 3D-printing ink, studying β-TCP:SrO mass ratios of 100:0, 95:5, and 90:10 (named as β-TCP, β-TCP/5-Sr, and β-TCP/10-Sr, respectively). Adding SrCO3 in the 3D-printing ink led to a slight increase in viscosity but did not affect its printability, resulting in scaffolds with a high printing fidelity compared to the computational design. Interestingly, Sr was incorporated into the lattice structure of the scaffolds, forming hydroxyapatite (HA). No residual SrO or SrCO3 were observed in the XRD patterns of any composition, and HA was the majority phase of the β-TCP/10-Sr scaffolds. The addition of Sr increased the compression strength of the scaffolds, with both β-TCP/5-Sr and β-TCP/10-Sr performing better than the β-TCP. Overall, β-TCP/5-Sr presented higher mineralized nodules and mechanical strength, while β-TCP scaffolds presented superior cell viability. The incorporation of SrCO3 in the ink formulation is a viable method to obtain Sr-BCP scaffolds. Thus, this approach could be explored with other CaP scaffolds aiming to optimize their performance and the addition of alternative therapeutic ions.

Abstract:Mechanical strength of bioceramic scaffolds is a problem to treat the load bearing bone defects. We developed the Mg-doping wollastonite (CSi-Mg)-based scaffolds with high strength via 3D printing technology. The effect of pore size, β-tricalcium phosphate (β-TCP) content (x%), and heating schedule on the strength of scaffolds were investigated systematically. Incorporation of β-TCP could readily adjust the sintering properties of the CSi-Mg scaffolds and the scaffolds with high (20–30%) and low (10–20%) β-TCP possess much high strength (80–100MPa or 120–140MPa) after undergoing one- or two-step sintering. Meanwhile, the CSi-Mg/TCPx (x=10, 20) with medium-pore (∼320μm) had over 100MPa in compression and ∼52% in porosity. In particular, the composite scaffolds maintained appreciable strength (over 50MPa) after immersion in Tris buffer for a long time stage (6 weeks). These findings demonstrate that the CSi-Mg/TCPx scaffolds are promising for treating some challengeable bone defects, especially for load-bearing bone repair.

Abstract:Abstract Background Bone scaffold is one of the most effective methods to treat bone defect. The ideal scaffold of bone tissue should not only provide space for bone tissue growth, but also have sufficient mechanical strength to support the bone defect area. Moreover, the scaffold should provide a customized size or shape for the patient’s bone defect. Methods In this study, strontium-containing Mg-doped wollastonite (Sr-CSM) bioceramic scaffolds with controllable pore size and pore structure were manufactured by direct ink writing 3D printing. Biological properties of Sr-CSM scaffolds were evaluated by apatite formation ability, in vitro proliferation ability of rabbit bone-marrow stem cells (rBMSCs), and alkaline phosphatase (ALP) activity using β-TCP and Mg-doped wollastonite (CSM) scaffolds as control. The compression strength of three scaffold specimens was probed after completely drying them while been submerged in Tris–HCl solution for 0, 2,4 and 6 weeks. Results The mechanical test results showed that strontium-containing Mg-doped wollastonite (Sr-CSM) scaffolds had acceptable initial compression strength (56 MPa) and maintained good mechanical stability during degradation in vitro. Biological experiments showed that Sr-CSM scaffolds had a better apatite formation ability. Cell experiments showed that Sr-CSM scaffold had a higher cell proliferation ability compared with β-TCP and CSM scaffold. The higher ALP activity of Sr-CSM scaffold indicates that it can better stimulate osteoblastic differentiation and bone mineralization. Conclusions Therefore, Sr-CSM scaffolds not only have acceptable compression strength, but also have higher osteogenesis bioactivity, which can be used in bone tissue engineering scaffolds.

Abstract:Background. Damage to the intestine culminating in NEC may occur by reduced mesenteric blood flow, bacterial overgrowth, enteral feeding or a combination of these factors. Antenatal steroids, human milk, lower pH formula, enteral IgA and antibiotics have each been reported to lower the incidence of NEC. In this study, the inclusion of EPL in a PT formula led to an apparent reduction in the incidence of NEC but not other diseases in hospitalized PT infants. Methods. In a randomized, double-masked trial, PT infants (725-1375 g birth weight, n=119) were fed either a control formula (CF) or an egg phospholipid-supplemented formula (EPLF) made by replacing some coconut oil in CF with EPL (75% phosphatidylcholine, 20% phosphatidylethanolamine, 5% other PL). PT and T versions of CF and EPLF were fed from -3 mo to 2 mo and 2 mo to 12 mo, respectively (40 wks gestation = 0 mo). EPLF, but not CF, contained ARA (0.4%) and DHA (0.13%, total fatty acids). Infants were assigned to one of three treatments that were balanced for gender in three stratified weight ranges (725-925 g; 926-1150 g; 1151-1375 g): Control (CF from -3 mo to 12 mo, n=41); Late Supplementation (CF from -3 mo to -1 mo; EPLF from -1 mo to 12 mo, n=44); and Early Supplementation (EPLF from -3 mo to 12 mo, n=34). Subjects lost before 4 mo corrected age were replaced to ensure study of at least 30 infants/group through that age. Infants fed CF and EPLF had similar neonatal, perinatal and demographic characteristics. Results. Fifteen of 85 infants (17.6%) fed CF in hospital (Control and Late Supplementation) and 1 of 34 (2.9%) fed EPLF (Early Supplementation) developed NEC stage II or III (p<0.05). Diet had no apparent effect on other diseases including sepsis (CF, 28.2%; EPLF, 26.5%), BPD (CF,23.4%; EPLF, 23.5%), and ROP (CF,40.0; EPLF,38.2%). The incidence of NEC was higher among male than female infants (11/60 vs 5/59, p,<0.05). All 11 males who developed NEC were fed CF (11/43, 25.6%) (p<0.01 vs males fed EPLF). Summary. EPLF appeared to decrease the incidence of NEC but not other common diseases of PT infants. Conclusion. We speculate that EPL or some component of EPL (e.g., ARA, DHA or choline) protected the immature intestine by one of several plausible mechanisms. A prospective randomized trial to test the effect of EPLF on NEC seems warranted, because NEC remains a major cause of morbidity and mortality among PT infants.
In-Vivo Degradation Behavior and Osseointegration of 3D Powder-Printed Calcium Magnesium Phosphate Cement Scaffolds

Abstract:Calcium magnesium phosphate cements (CMPCs) are promising bone substitutes and experience great interest in research. Therefore, in-vivo degradation behavior, osseointegration and biocompatibility of three-dimensional (3D) powder-printed CMPC scaffolds were investigated in the present study. The materials Mg225 (Ca0.75Mg2.25(PO4)2) and Mg225d (Mg225 treated with diammonium hydrogen phosphate (DAHP)) were implanted as cylindrical scaffolds (h = 5 mm, Ø = 3.8 mm) in both lateral femoral condyles in rabbits and compared with tricalcium phosphate (TCP). Treatment with DAHP results in the precipitation of struvite, thus reducing pore size and overall porosity and increasing pressure stability. Over 6 weeks, the scaffolds were evaluated clinically, radiologically, with Micro-Computed Tomography (μCT) and histological examinations. All scaffolds showed excellent biocompatibility. X-ray and in-vivo μCT examinations showed a volume decrease and increasing osseointegration over time. Structure loss and volume decrease were most evident in Mg225. Histologically, all scaffolds degraded centripetally and were completely traversed by new bone, in which the remaining scaffold material was embedded. While after 6 weeks, Mg225d and TCP were still visible as a network, only individual particles of Mg225 were present. Based on these results, Mg225 and Mg225d appear to be promising bone substitutes for various loading situations that should be investigated further.

Abstract:Obliteration of the fat that surrounds the celiac axis or superior mesenteric artery is a common CT feature of pancreatic cancer. In its typical form, this finding is so characteristic of local extension of pancreatic adenocarcinoma that it has come to be regarded as almost diagnostic of that condition [1 -3]. We report a case of pancreatitis in which obliteration of the fat at the margins of the celiac axis and superior mesenteric artery, together with a focal mass in the pancreas, produced a CT appearance indistinguishable from that typically caused by pancreatic carcinoma.

Abstract:Due to the positive effects of magnesium substitution on the mechanical properties and the degradation rate of the clinically well-established calcium phosphate cements (CPCs), calcium magnesium phosphate cements (CMPCs) are increasingly being researched as bone substitutes. A post-treatment alters the materials' physical properties and chemical composition, reinforcing the structure and modifying the degradation rate. By alkaline post-treatment with diammonium hydrogen phosphate (DAHP, (NH 4 ) 2 HPO 4 ), the precipitation product struvite is formed, while post-treatment with an acidic phosphate solution [e.g., phosphoric acid (PA, H 3 PO 4 )] results in precipitation of newberyite and brushite. However, little research has yet been conducted on newberyite as a bone substitute and PA post-treatment of CMPCs has not been described in the accessible literature so far. Therefore, in the present study, the influence of an alkaline (DAHP) or acid (PA) post-treatment on the biocompatibility, degradation behavior, and osseointegration of cylindrical scaffolds ( h = 5.1 mm, Ø = 4.2 mm) produced from the ceramic cement powder Ca 0.75 Mg 2.25 (PO 4 ) 2 by the advantageous manufacturing technique of three-dimensional (3D) powder printing was investigated in vivo . Scaffolds of the material groups Mg225d (DAHP post-treatment) and Mg225p (PA post-treatment) were implanted into the cancellous part of the lateral femoral condyles in rabbits. They were evaluated up to 24 weeks by regular clinical, X-ray, micro-computed tomographic (μCT), and histological examinations as well as scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX) analysis and compared with tricalcium phosphate (TCP). All materials showed excellent biocompatibility and rapid osseointegration. While TCP degraded only slightly, the CMPCs showed almost complete degradation. Mg225d demonstrated significantly faster loss of form and demarcability from surrounding bone, scaffold volume reduction, and significantly greater degradation on the side towards the bone marrow than to the cortex than Mg225p. Simultaneously, numerous bone trabeculae have grown into the implantation site. While these were mostly located on the side towards the cortex in Mg225d, they were more evenly distributed in Mg225p and showed almost the same structural characteristics as physiological bone after 24 weeks in Mg225p. Based on these results, the acid post-treated 3D powder-printed Mg225p is a promising degradable bone substitute that should be further investigated.

Abstract:Ionic substitutions within the hydroxyapatite lattice are a widely used approach to mimic the chemical composition of the bone mineral. In this work, Sr-substituted and Mg- and Sr-co-substituted calcium phosphate (CaP) scaffolds, with various levels of strontium and magnesium substitution, were prepared using the hydrothermal method at 200 °C. Calcium carbonate skeletons of cuttlefish bone, ammonium dihydrogenphosphate (NH4H2PO4), strontium nitrate (Sr(NO3)2), and magnesium perchlorate (Mg(ClO4)2) were used as reagents. Materials were characterized by means of X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). Whole powder pattern decomposition refinements of XRD data indicated that increased magnesium content in the Mg- and Sr-co-substituted scaffolds was related to an increased proportion of the whitlockite (WH) phase in the biphasic hydroxyapatite (HAp)/WH scaffolds. In addition, refinements indicate that Sr2+ ions have replaced Ca2+ sites in the WH phase. Furthermore, PCL-coated Mg-substituted and Sr- and Mg-co-substituted scaffolds, with the HAp:WH wt. ratio of 90:10 were prepared by vacuum impregnation. Results of compression tests showed a positive impact of the WH phase and PCL coating on the mechanical properties of scaffolds. Human mesenchymal stem cells (hMSCs) were cultured on composite scaffolds in an osteogenic medium for 21 days. Immunohistochemical staining showed that Mg-Sr-CaP/PCL scaffold exhibited higher expression of collagen type I than the Mg-CaP/PCL scaffold, indicating the positive effect of Sr2+ ions on the differentiation of hMSCs, in concordance with histology results. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) analysis confirmed an early stage of osteogenic differentiation.

Abstract:Beyond the traditional phase conversion or biphase mixing hybrid, we developed the dilute magnesium-doped wollastonite inks and three-dimensional (3D) printing approaches to fabricate the ultrahigh strength bioceramic porous scaffolds. The mechanical strength (>120 MPa) of the porous bioceramics was an order of magnitude higher than the pure wollastonite and other stoichiometric Ca-Mg silicate porous bioceramics. This abnormal but expected improvement in strength in bioceramic scaffolds is equivalent or even superior to the mechanical requirement in load-bearing bone defects. The breakthrough is totally unexpected, and it quickly opens the door for the 3D printing bioceramics manufacture and large-area segmental bone defect repair applications.
Degradation of 3D-printed magnesium phosphate ceramics in vitro and a prognosis on their bone regeneration potential

Abstract:Regenerative bone implants promote new bone formation and ideally degrade simultaneously to osteogenesis. Although clinically established calcium phosphate bone grafts provide excellent osseointegration and osteoconductive efficacy, they are limited in terms of bioresorption. Magnesium phosphate (MP) based ceramics are a promising alternative, because they are biocompatible, mechanically extremely stable, and degrade much faster than calcium phosphates under physiological conditions. Bioresorption of an implant material can include both chemical dissolution as well as cellular resorption. We investigated the bioresorption of 3D powder printed struvite and newberyite based MP ceramics in vitro by a direct human osteoclast culture approach. The osteoclast response and cellular resorption was evaluated by means of fluorescence and TRAP staining, determination of osteoclast activities (CA II and TRAP), SEM imaging as well as by quantification of the ion release during cell culture. Furthermore, the bioactivity of the materials was investigated via SBF immersion, whereas hydroxyapatite precipitates were analyzed by SEM and EDX measurements. This bioactive coating was resorbed by osteoclasts. In contrast, only chemical dissolution contributed to bioresorption of MP, while no cellular resorption of the materials was observed. Based on our results, we expect an increased bone regeneration effect of MP compared to calcium phosphate based bone grafts and complete chemical degradation within a maximum of 1.5-3.1 years.
The Calcium Channel Affect Osteogenic Differentiation of Mesenchymal Stem Cells on Strontium-Substituted Calcium Silicate/Poly-ε-Caprolactone Scaffold

Abstract:There had been a paradigm shift in tissue engineering studies over the past decades. Of which, part of the hype in such studies was based on exploring for novel biomaterials to enhance regeneration. Strontium ions have been reported by others to have a unique effect on osteogenesis. Both in vitro and in vivo studies had demonstrated that strontium ions were able to promote osteoblast growth, and yet at the same time, inhibit the formation of osteoclasts. Strontium is thus considered an important biomaterial in the field of bone tissue engineering. In this study, we developed a Strontium-calcium silicate scaffold using 3D printing technology and evaluated for its cellular proliferation capabilities by assessing for protein quantification and mineralization of Wharton’s Jelly mesenchymal stem cells. In addition, verapamil (an L-type of calcium channel blocker, CCB) was used to determine the mechanism of action of strontium ions. The results found that the relative cell proliferation rate on the scaffold was increased between 20% to 60% within 7 days of culture, while the CCB group only had up to approximately 10% proliferation as compared with the control specimen. Besides, the CCB group had downregulation and down expressions of all downstream cell signaling proteins (ERK and P38) and osteogenic-related protein (Col I, OPN, and OC). Furthermore, CCB was found to have 3–4 times lesser calcium deposition and quantification after 7 and 14 days of culture. These results effectively show that the 3D printed strontium-contained scaffold could effectively stimulate stem cells to undergo bone differentiation via activation of L-type calcium channels. Such results showed that strontium-calcium silicate scaffolds have high development potential for bone tissue engineering.

Abstract:Calcium silicate cements have been considered as alternative bone substitutes owing to its extraordinary bioactivity and osteogenicity. Unfortunately, the major disadvantage of the cements was the slow degradation rate which may limit the efficiency of bone regeneration. In this study, we proposed a facile method to synthesize degradable calcium silicate cements by incorporating strontium into the cements through solid-state sintering. The effects of Sr incorporation on physicochemical and biological properties of the cements were evaluated. Although, our findings revealed that the incorporation of strontium retarded the hardening reaction of the cements, the setting time of different cements (11–19 min) were in the acceptable range for clinical use. The presence of Sr in the CS cements would hampered the precipitation of calcium phosphate products on the surface after immersion in SBF, however, a layer of precipitated calcium phosphate products can be formed on the surface of the Sr-CS cement within 1 day immersion in SBF. More importantly, the degradation rate of the cements increased with increasing content of strontium, consequentially raised the levels of released strontium and silicon ions. The elevated dissolving products may contribute to the enhancement of the cytocompatibility, alkaline phosphatase activity, osteocalcin secretion, and mineralization of human Wharton's jelly mesenchymal stem cells. Together, it is concluded that the strontium-incorporated calcium silicate cement might be a promising bone substitute that could accelerate the regeneration of irregularly shaped bone defects.

Abstract:Bone defects persist as a significant challenge in the field of clinical orthopedics. This study focuses on the fabrication and characterization of 3D-printed composite hydrogel scaffolds composed of sodium alginate, gelatin, and α-tricalcium phosphate (α-TCP) with varying ratios of Strontium ions (Sr2+). These scaffolds aim to address the clinical challenges associated with bone defect repair by providing mechanical support and promoting bone formation and vascularization. The degradation, swelling, mechanical properties, and release profiles of Sr2+ from the hydrogel scaffolds were comprehensively characterized. In vitro tests were conducted to assess cell viability and proliferation, as well as osteogenic and angiogenic gene expression, to investigate the osteogenic and pro-angiogenic potential of the composite hydrogel scaffolds. Furthermore, skull defect simulations were performed, and composite scaffolds with varying Sr2+ ratios were implanted to evaluate their effectiveness in bone repair. This research establishes a foundation for advancing bone tissue engineering through composite scaffolds containing biological macromolecules and strontium, with alginate serving as a key element in enhancing performance and expanding clinical applicability.
A novel calcium phosphate-based ceramic scaffolds with unexpected high osteogenic activity by strontium doping

Abstract:The effective restoration of local bone homeostasis is a key factor in successfully treating pathological bone defects, which can be attributed to conditions such as osteoporosis or bone tumors. To address this challenge, we prepared a strontium calcium phosphate (SrCaP) ceramic using an ion doping method that demonstrated both osteogenic promotion and osteoclastic inhibition. In this study, we systematically evaluated the physicochemical properties, in vitro cell responses and in vivo osteogenesis for this material. Our results demonstrate that a higher purity SrCaP can be obtained when the strontium doping content in hydroxyapatite is equal to or greater than 25 mol.%. The long-term release of strontium ions from SrCaP significantly promoted osteogenic differentiation of stem cells. Furthermore, in vitro experiments revealed that SrCaP scaffolds doped with 30 mol.% of strontium (Sr30–S) demonstrated the most effective performance in promoting osteogenic differentiation and inhibiting osteoclast formation, thus providing effective support for the restoration of local bone homeostasis. Although Sr30–S inhibited the formation of osteoclasts, which hindered active material absorption, its faster passive degradation rate ensured a more rapid in vivo degradation rate than that of hydroxyapatite scaffold, thereby promoting the generation and growth of new bones. In summary, the innovative SrCaP ceramic scaffolds synthesized in this study demonstrate superior capabilities in promoting osteogenic differentiation and inhibiting osteoclastic resorption activity. These scaffolds not only hold potential in facilitating the restoration of local bone homeostasis but also exhibit a degradation rate that aligns with new bone growth. As a result, they represent an excellent candidate for repairing pathological bone defects characterized by high osteoclastic absorption activity, offering promising clinical applications.

Abstract:In this study, porous magnesium (Mg) scaffolds were investigated with varying strontium (Sr) and constant zinc (Zn) concentrations through the powder metallurgy process. All samples were examined at room temperature to evaluate their microstructure, mechanical andin-vitrodegradation behaviour and biological properties. Results indicated that adding Sr was associated with fine average grain size, increased mechanical strength, and a decreased corrosion rate. All samples show tiny isolated and open interconnected pores (porosities: 18%-30%, pores: 127-279 µm) with a suitable surface roughness of less than 0.5 µm. All the provided samples possess mechanical and hemocompatible properties that closely resemble natural bone. Mg-4Zn-2Sr has the highest hardness (102.61 ± 15.1 HV) and compressive strength (24.80 MPa) than Mg-4Zn-0.5Sr (85 ± 8.5 HV, 22.14 MPa) and Mg-4Zn-1Sr (97.71 ± 11.2 HV, 18.06 MPa). Immersion results revealed that samples in phosphate-buffered saline solutions have excellent degradability properties, which makes them a promising biodegradable material for orthopaedic applications. The scaffold with the highest Sr concentration shows the best optimised mechanical and degradation behaviour out of the three porous scaffolds, with a 2.7% hemolysis rate.
Fabrication of Amorphous Strontium Polyphosphate Microparticles That Induce Mineralization of Bone Cells in Vitro and in Vivo

Abstract:Here we describe the fabrication process of amorphous strontium-polyphosphate microparticles ("Sr-a-polyP-MP"). The effects of these particles on growth and gene expression were investigated with SaOS-2 cells as well as with human mesenchymal stem cells (MSC) and compared with those particles prepared of amorphous calcium-polyphosphate ("Ca-a-polyP-MP") and of strontium salt. The results revealed a markedly higher stimulation of growth of MSC by "Sr-a-polyP-MP" compared to "Ca-a-polyP-MP" and a significant increase in mineralization of SaOS-2 cells, as well as an enhanced upregulation of the expression of the genes encoding for alkaline phosphatase and the bone morphogenetic protein 2 (BMP-2), likewise performed with SaOS-2 cells. On the other hand, "Sr-a-polyP-MP" only slightly changes the expression of the osteocyte-specific sclerostin, a negative regulator of the canonical Wnt signaling pathway and an inhibitor of bone cell differentiation as well as of mineralization in SaOS-2 cells. In contrast, "Ca-a-polyP-MP" strongly increased the steady-state expression of the SOST (sclerostin) gene. In animal studies poly(D,L-lactide-co-glycolide (PLGA) microspheres, containing polyP particles, were implanted into critical-size calvarial defects in rats. The results show that the amorphous Sr-polyP-containing microspheres caused an increased healing/mineralization of the bone defect even after short implantation periods of 8-12 weeks, if compared to the B-tri-calcium phosphate control as well as to Ca-polyP. It is proposed that "Sr-a-polyP-MP" might elicit suitable properties to be applied as a regeneratively active implant material for bone repair.Statement of SignificanceIn this manuscript, we fabricated amorphous strontium-polyphosphate microparticles ("Sr-a-polyP-MP") and studied their effects on bone mineral formation in vitro as well as in vivo. In vitro, those particles substantially increased the expression of the genes encoding for alkaline phosphatase, the bone morphogenetic protein 2 and the mineralization. In vivo, the "Sr-a-polyP-MP" packed into PLGA microspheres and implanted into critical-size calvarial defects in rats resulted in a speeded up of the healing/mineralization of the bone defect.
