Abstract:Critical network states and neural plasticity are essential for flexible behavior in dynamic environments, allowing for efficient information processing and experience-dependent learning. Synaptic-weight-based Hebbian plasticity and homeostatic synaptic scaling were considered the key mechanisms in enabling memory while stabilizing network dynamics. However, the role of structural plasticity as a homeostatic mechanism is less consistently reported, especially under activity inhibition, leading to an incomplete understanding of its functional impact. In this study, we combined live-cell microscopy of eGPF-labeled neurons in organotypic entorhinal-hippocampal tissue cultures with computational modeling to investigate the response of spine-number-based structural plasticity to activity perturbations and its interaction with homeostatic synaptic scaling. Tracking individual dendritic segments, we demonstrated that inhibiting excitatory neurotransmission does not monotonically regulate dendritic spine density. Specifically, inhibition of AMPA receptors with a low concentration of 2,3-dioxo-6-nitro-7-sulfamoyl-benzo[f]quinoxaline (NBQX, 200 nM) significantly increased spine density, while complete AMPA receptors blockade with 50 μM NBQX reduced spine density. Motivated by these findings, we developed network simulations incorporating a bi-phasic structural plasticity rule governing activity-dependent synapse formation. We showed that this biphasic rule maintained neural activity homeostasis under stimulation and permitted either synapse formation or synapse loss, depending on the degree of activity deprivation. Homeostatic synaptic scaling affected the recurrent connectivity, modulated the network activity, and influenced the outcome of structural plasticity. Specifically, it reduced stimulation-triggered synapse loss by downscaling synaptic weights and rescued silencing-induced synapse loss by upscaling recurrent inputs, thus reactivating silent neurons. Our interaction between these mechanisms offers an explanation for divergent findings in the existing literature. In summary, calcium-based synaptic scaling and homeostatic structural plasticity rules compete and compensate for one another, ensuring efficient and robust control of firing rate homeostasis.

Abstract:Synaptic connections in neuronal circuits are modulated by pre- and post-synaptic spiking activity. Heuristic models of this process of synaptic plasticity can provide excellent fits to results from in-vitro experiments in which pre- and post-synaptic spiking is varied in a controlled fashion. However, the plasticity rules inferred from fitting such data are inevitably unstable, in that given constant pre- and post-synaptic activity the synapse will either fully potentiate or depress. This instability can be held in check by adding additional mechanisms, such as homeostasis. Here we consider an alternative scenario in which the plasticity rule itself is stable. When this is the case, net potentiation or depression only occur when pre- and post-synaptic activity vary in time, e.g. when driven by time-varying inputs. We study how the features of such inputs shape the recurrent synaptic connections in models of neuronal circuits. In the case of oscillatory inputs, the resulting structure is strongly affected by the phase relationship between drive to different neurons. In large networks, distributed phases tend to lead to hierarchical clustering. Our results may be of relevance for understanding the effect of sensory-driven inputs, which are by nature time-varying, on synaptic plasticity, and hence on learning and memory.

Abstract:The brain is believed to operate in part by making predictions about sensory stimuli and encoding deviations from these predictions in the activity of "prediction error neurons." This principle defines the widely influential theory of predictive coding. The precise circuitry and plasticity mechanisms through which animals learn to compute and update their predictions are unknown. Homeostatic inhibitory synaptic plasticity is a promising mechanism for training neuronal networks to perform predictive coding. Homeostatic plasticity causes neurons to maintain a steady, baseline firing rate in response to inputs that closely match the inputs on which a network was trained, but firing rates can deviate away from this baseline in response to stimuli that are mismatched from training. We combine computer simulations and mathematical analysis systematically to test the extent to which randomly connected, unstructured networks compute prediction errors after training with homeostatic inhibitory synaptic plasticity. We find that homeostatic plasticity alone is sufficient for computing prediction errors for trivial time-constant stimuli, but not for more realistic time-varying stimuli. We use a mean-field theory of plastic networks to explain our findings and characterize the assumptions under which they apply.

Abstract:Self-sustained neural activity maintained through local recurrent connections is of fundamental importance to cortical function. Converging theoretical and experimental evidence indicates that cortical circuits generating self-sustained dynamics operate in an inhibition-stabilized regime. Theoretical work has established that four sets of weights ( W E←E , W E←I , W I←E , and W I←I ) must obey specific relationships to produce inhibition-stabilized dynamics, but it is not known how the brain can appropriately set the values of all four weight classes in an unsupervised manner to be in the inhibition-stabilized regime. We prove that standard homeostatic plasticity rules are generally unable to generate inhibition-stabilized dynamics and that their instability is caused by a signature property of inhibition-stabilized networks: the paradoxical effect. In contrast, we show that a family of "cross-homeostatic" rules overcome the paradoxical effect and robustly lead to the emergence of stable dynamics. This work provides a model of how—beginning from a silent network—self-sustained inhibition-stabilized dynamics can emerge from learning rules governing all four synaptic weight classes in an orchestrated manner.

Neuromodulation and homeostasis: complementary mechanisms for robust neural function

Abstract:Neurons depend on two interdependent mechanisms-homeostasis and neuromodulation-to maintain robust and adaptable functionality. Homeostasis stabilizes neuronal activity by adjusting ionic conductances, whereas neuromodulation dynamically modifies ionic properties in response to external signals. Combining these mechanisms in conductance-based models often produces unreliable outcomes, particularly when sharp neuromodulation interferes with homeostatic tuning. This study explores how a biologically inspired neuromodulation controller can harmonize with homeostasis to ensure reliable neuronal function. Using computational models of stomatogastric ganglion and dopaminergic neurons, we demonstrate that controlled neuromodulation preserves neuronal firing patterns while maintaining intracellular calcium levels. Unlike sharp neuromodulation, the neuromodulation controller integrates activity-dependent feedback through mechanisms mimicking G-protein-coupled receptor cascades. The interaction between these controllers critically depends on the existence of an intersection in conductance space, representing a balance between target calcium levels and neuromodulated firing patterns. Maximizing neuronal degeneracy enhances the likelihood of such intersections, enabling robust modulation and compensation for channel blockades. We further show that this controller pairing extends to network-level activity, reliably modulating central pattern generators in crustaceans. These findings suggest that targeting neuromodulation pathways-rather than ion channels directly-may offer safer pharmacological strategies to manage neuronal dysfunctions. This study highlights the complementary roles of homeostasis and neuromodulation, proposing a unified control framework for maintaining robust and adaptive neural activity under physiological and pathological conditions.

Abstract:Rich, spontaneous brain activity has been observed across a range of different temporal and spatial scales. These dynamics are thought to be important t for efficient neural functioning. Experimental evidence suggests that these neural dynamics are maintained across a variety of different cognitive states, in response to alterations of the environment and to changes in brain configuration (e.g., across individuals, development and in many neurological disorders). This suggests that the brain has evolved mechanisms to stabilize dynamics and maintain them across a range of situations. Here, we employ a local homeostatic inhibitory plasticity mechanism, balancing inhibitory and excitatory activity in a model of macroscopic brain activity based on white-matter structural connectivity. We demonstrate that the addition of homeostatic plasticity regulates network activity and allows for the emergence of rich, spontaneous dynamics across a range of brain configurations. Furthermore, the presence of homeostatic plasticity maximises the overlap between empirical and simulated patterns of functional connectivity. Therefore, this work presents a simple, local, biologically plausible inhibitory mechanism that allows stable dynamics to emerge in the brain and which facilitates the formation of functional connectivity networks.

Abstract:Bangla handwriting recognition is becoming a very important issue nowadays. It is potentially a very important task specially for Bangla speaking population of Bangladesh and West Bengal. By keeping that in our mind we are introducing a comprehensive Bangla handwritten character dataset named BanglaLekha-Isolated. This dataset contains Bangla handwritten numerals, basic characters and compound characters. This dataset was collected from multiple geographical location within Bangladesh and includes sample collected from a variety of aged groups. This dataset can also be used for other classification problems i.e: gender, age, district. This is the largest dataset on Bangla handwritten characters yet.

Abstract:Understanding the relationship between large-scale structural and functional brain networks remains a crucial issue in modern neuroscience. Recently, there has been growing interest in investigating the role of homeostatic plasticity mechanisms, across different spatiotemporal scales, in regulating network activity and brain functioning against a wide range of environmental conditions and brain states (e.g., during learning, development, ageing, neurological diseases). In the present study, we investigate how the inclusion of homeostatic plasticity in a stochastic whole-brain model, implemented as a normalization of the incoming node's excitatory input, affects the macroscopic activity during rest and the formation of functional networks. Importantly, we address the structure-function relationship both at the group and individual-based levels. In this work, we show that normalization of the node's excitatory input improves the correspondence between simulated neural patterns of the model and various brain functional data. Indeed, we find that the best match is achieved when the model control parameter is in its critical value and that normalization minimizes both the variability of the critical points and neuronal activity patterns among subjects. Therefore, our results suggest that the inclusion of homeostatic principles lead to more realistic brain activity consistent with the hallmarks of criticality. Our theoretical framework open new perspectives in personalized brain modeling with potential applications to investigate the deviation from criticality due to structural lesions (e.g. stroke) or brain disorders.

Abstract:To perform computations with the efficiency necessary for animal survival, neocortical microcircuits must be capable of reconfiguring in response to experience, while carefully regulating excitatory and inhibitory connectivity to maintain stable function. This dynamic fine-tuning is accomplished through a rich array of cellular homeostatic plasticity mechanisms that stabilize important cellular and network features such as firing rates, information flow, and sensory tuning properties. Further, these functional network properties can be stabilized by different forms of homeostatic plasticity, including mechanisms that target excitatory or inhibitory synapses, or that regulate intrinsic neuronal excitability. Here we discuss which aspects of neocortical circuit function are under homeostatic control, how this homeostasis is realized on the cellular and molecular levels, and the pathological consequences when circuit homeostasis is impaired. A remaining challenge is to elucidate how these diverse homeostatic mechanisms cooperate within complex circuits to enable them to be both flexible and stable. Abstract:Many neural computations emerge from self-sustained patterns of activity in recurrent neural circuits, which rely on balanced excitation and inhibition. Neuromorphic electronic circuits that use the physics of silicon to emulate neuronal dynamics represent a promising approach for implementing the brain's computational primitives, including self-sustained neural activity. However, achieving the same robustness of biological networks in neuromorphic computing systems remains a challenge, due to the high degree of heterogeneity and variability of their analog components. Inspired by the strategies used by real cortical networks, we apply a biologically-plausible cross-homeostatic learning rule to balance excitation and inhibition in neuromorphic implementations of spiking recurrent neural networks. We demonstrate how this learning rule allows the neuromorphic system to work in the presence of device mismatch and to autonomously tune the spiking network to produce robust, self-sustained, fixed-point attractor dynamics with irregular spiking in an inhibition-stabilized regime. We show that this rule can implement multiple, coexisting stable memories, with emergent soft-winner-take-all (sWTA) dynamics, and reproduce the so-called ''paradoxical effect'' widely observed in cortical circuits. In addition to validating neuroscience models on a substrate that shares many similar properties and limitations with biological systems, this work enables the construction of ultra-low power, mixed-signal neuromorphic technologies that can be automatically configured to compute reliably, despite the large on-chip and chip-to-chip variability of their analog components.

Abstract:This paper presents a comprehensive mechanistic model of a neuron with plasticity that explains how information input as time-varying signals is processed and stored. Additionally, the model addresses two long-standing, specific biological challenges: Integrating Hebbian and homeostatic plasticity, and identifying a concise synaptic learning rule. A biologically accurate electric-circuit equivalent is derived through a one-to-one mapping from the known properties of ion channels. The often-overlooked dynamics of the synaptic cleft is essential in this process. Analysis of the model reveals a simple and succinct learning rule, indicating that the neuron functions as an internal-feedback adaptive filter, a common concept in signal processing. Simulations confirm the model’s functionality, stability, and convergence, demonstrating that even a single neuron without external feedback can act as a potent signal processor. The model replicates several key characteristics typical of biological neurons, which are seldom captured in other neuron models. It can encode time-varying functions, learn without risking instability, and bootstrap from a state where all synaptic weights are zero. This interdisciplinary work spans a broad range of subjects within the realm of neurobiophysics, including neurobiology, electronics, and signal processing.

Abstract:Determining the sites and directions of plasticity underlying changes in neural activity and behavior is critical for understanding mechanisms of learning. Identifying such plasticity from neural recording data can be challenging due to feedback pathways that impede reasoning about cause and effect. We studied interactions between feedback, neural activity, and plasticity in the context of a closed-loop motor learning task for which there is disagreement about the loci and directions of plasticity: vestibulo-ocular reflex learning. We constructed a set of circuit models that differed in the strength of their recurrent feedback, from no feedback to very strong feedback. Despite these differences, each model successfully fit a large set of neural and behavioral data. However, the patterns of plasticity predicted by the models fundamentally differed, with the direction of plasticity at a key site changing from depression to potentiation as feedback strength increased. Guided by our analysis, we suggest how such models can be experimentally disambiguated. Our results address a long-standing debate regarding cerebellum-dependent motor learning, suggesting a reconciliation in which learning-related changes in the strength of synaptic inputs to Purkinje cells are compatible with seemingly oppositely directed changes in Purkinje cell spiking activity. More broadly, these results demonstrate how changes in neural activity over learning can appear to contradict the sign of the underlying plasticity when either internal feedback or feedback through the environment is present.

Discovering plasticity rules that organize and maintain neural circuits

Abstract:Intrinsic dynamics within the brain can accelerate learning by providing a prior scaffolding for dynamics aligned with task objectives. Such intrinsic dynamics should self-organize and self-sustain in the face of fluctuating inputs and biological noise, including synaptic turnover and cell death. An example of such dynamics is the formation of sequences, a ubiquitous motif in neural activity. The sequence-generating circuit in zebra finch HVC provides a reliable timing scaffold for motor output in song and demonstrates a remarkable capacity for unsupervised recovery following perturbation. Inspired by HVC, we seek a local plasticity rule capable of organizing and maintaining sequence-generating dynamics despite continual network perturbations. We adopt a meta-learning approach introduced by Confavreux et al, which parameterizes a learning rule using basis functions constructed from pre- and postsynaptic activity and synapse size, with tunable time constants. Candidate rules are simulated within initially random networks, and their fitness is evaluated according to a loss function that measures the fidelity with which the resulting dynamics encode time. We use this approach to introduce biological noise, forcing meta-learning to find robust solutions. We first show that, in the absence of perturbation, meta-learning identifies a temporally asymmetric generalization of Oja’s rule that reliably organizes sparse sequential activity. When synaptic turnover is introduced, the learned rule incorporates an additional form of homeostasis, better maintaining sequential dynamics relative to other previously proposed rules. Additionally, inspired by recent findings demonstrating plasticity in synapses from inhibitory interneurons in HVC, we explore the role of inhibitory plasticity in sequence-generating circuits. We find that learned plasticity adjusts both excitation and inhibition in response to manipulations, outperforming rules applied only to excitatory connections. We demonstrate how plasticity acting on both excitatory and inhibitory synapses can better shape excitatory cell dynamics to scaffold timing representations.

Abstract:In neural circuits, synaptic strengths influence neuronal activity by shaping network dynamics, and neuronal activity influences synaptic strengths through activity-dependent plasticity. Motivated by this fact, we study a recurrent-network model in which neuronal units and synaptic couplings are interacting dynamic variables, with couplings subject to Hebbian modification with decay around quenched random strengths. Rather than assigning a specific role to the plasticity, we use dynamical mean-field theory and other techniques to systematically characterize the neuronal-synaptic dynamics, revealing a rich phase diagram. Adding Hebbian plasticity slows activity in already chaotic networks and can induce chaos in otherwise quiescent networks. Anti-Hebbian plasticity quickens activity and produces an oscillatory component. Analysis of the Jacobian shows that Hebbian and anti-Hebbian plasticity push locally unstable modes toward the real and imaginary axes, respectively, explaining these behaviors. Both random-matrix and Lyapunov analysis show that strong Hebbian plasticity segregates network timescales into two bands, with a slow, synapse-dominated band driving the dynamics, suggesting a flipped view of the network as synapses connected by neurons. For increasing strength, Hebbian plasticity initially raises the complexity of the dynamics, measured by the maximum Lyapunov exponent and attractor dimension, but then decreases these metrics, likely due to the proliferation of stable fixed points. We compute the marginally stable spectra of such fixed points as well as their number, showing exponential growth with network size. Finally, in chaotic states with strong Hebbian plasticity, a stable fixed point of neuronal dynamics is destabilized by synaptic dynamics, allowing any neuronal state to be stored as a stable fixed point by halting the plasticity. Abstract:In recent years, there have been many computational simulations of spontaneous neural dynamics. Here, we explore a model of spontaneous neural dynamics and allow it to control a virtual agent moving in a simple environment. This setup generates interesting brain-environment feedback interactions that rapidly destabilize neural and behavioral dynamics and suggest the need for homeostatic mechanisms. We investigate roles for both local homeostatic plasticity (local inhibition adjusting over time to balance excitatory input) as well as macroscopic task negative activity (that compensates for task positive, sensory input) in regulating both neural activity and resulting behavior (trajectories through the environment). Our results suggest complementary functional roles for both local homeostatic plasticity and balanced activity across brain regions in maintaining neural and behavioral dynamics. These findings suggest important functional roles for homeostatic systems in maintaining neural and behavioral dynamics and suggest a novel functional role for frequently reported macroscopic task-negative patterns of activity (e.g., the default mode network).

Abstract:The synaptic connectivity of cortical networks features an overrepresentation of certain wiring motifs compared to simple random-network models. This structure is shaped, in part, by synaptic plasticity that promotes or suppresses connections between neurons depending on their spiking activity. Frequently, theoretical studies focus on how feedforward inputs drive plasticity to create this network structure. We study the complementary scenario of self-organized structure in a recurrent network, with spike timing-dependent plasticity driven by spontaneous dynamics. We develop a self-consistent theory that describes the evolution of network structure by combining fast spiking covariance with a fast-slow theory for synaptic weight dynamics. Through a finite-size expansion of network dynamics, we obtain a low-dimensional set of nonlinear differential equations for the evolution of two-synapse connectivity motifs. With this theory in hand, we explore how the form of the plasticity rule drives the evolution of microcircuits in cortical networks. When potentiation and depression are in approximate balance, synaptic dynamics depend on the frequency of weighted divergent, convergent, and chain motifs. For additive, Hebbian STDP, these motif interactions create instabilities in synaptic dynamics that either promote or suppress the initial network structure. Our work provides a consistent theoretical framework for studying how spiking activity in recurrent networks interacts with synaptic plasticity to determine network structure.

Abstract:Biological neurons have adaptive nature and perform complex computations involving the filtering of redundant information. However, most common neural cell models, including biologically plausible, such as Hodgkin-Huxley or Izhikevich, do not possess predictive dynamics on a single-cell level. Moreover, the modern rules of synaptic plasticity or interconnections weights adaptation also do not provide grounding for the ability of neurons to adapt to the ever-changing input signal intensity. While natural neuron synaptic growth is precisely controlled and restricted by protein supply and recycling, weight correction rules such as widely used STDP are efficiently unlimited in change rate and scale. The present article introduces new mechanics of interconnection between neuron firing rate homeostasis and weight change through STDP growth bounded by abstract protein reserve, controlled by the intracellular optimization algorithm. We show how these cellular dynamics help neurons filter out the intense noise signals to help neurons keep a stable firing rate. We also examine that such filtering does not affect the ability of neurons to recognize the correlated inputs in unsupervised mode. Such an approach might be used in the machine learning domain to improve the robustness of AI systems.

Homeostatic regulation of brain activity: from endogenous mechanisms to homeostatic nanomachines

Abstract:After the initial concepts of the constancy of the internal milieu or homeostasis, put forward by Claude Bernard and Walter Cannon, homeostasis emerged as a mechanism to control oscillations of biologically meaningful variables within narrow physiological ranges. This is a primary need in the central nervous system that is continuously subjected to a multitude of stimuli from the internal and external environments that affect its function and structure, allowing to adapt the individual to the ever-changing daily conditions. Preserving physiological levels of activity despite disturbances that could either depress neural computation or excessively stimulate neural activity is fundamental, and failure of these homeostatic mechanisms can lead to brain diseases. In this review, we cover the role and main mechanisms of homeostatic plasticity involving the regulation of excitability and synaptic strength from the single neuron to the network level. We analyze the relationships between homeostatic and Hebbian plasticity and the conditions under which the preservation of the excitatory/inhibitory balance fails, triggering epileptogenesis and eventually epilepsy. Several therapeutic strategies to cure epilepsy have been designed to strengthen homeostasis when endogenous homeostatic plasticity mechanisms have become insufficient or ineffective to contrast hyperactivity. We describe "on demand" gene therapy strategies including optogenetics, chemogenetics, and chemo-optogenetics, and particularly focus on new closed loop sensor-actuator strategies mimicking homeostatic plasticity that can be endogenously expressed to strengthen the homeostatic defenses against brain diseases.

Abstract:We propose that a regulation mechanism based on Hebbian covariance plasticity may cause the brain to operate near criticality. We analyze the effect of such a regulation on the dynamics of a network with excitatory and inhibitory neurons and uniform connectivity within and across the two populations. We show that, under broad conditions, the system converges to a critical state lying at the common boundary of three regions in parameter space; these correspond to three modes of behavior: high activity, low activity, oscillation.
