Abstract:BACKGROUND:Herbivore-induced defence responses are often specific - different herbivores induce different defence responses in plants - and their specificity is largely mediated by chemical cues (herbivore-associated elicitors: HAEs) in insect oral or oviposition secretions. However, the specificity and the mechanisms of HAE-induced defence have not been investigated in the context of the evolutionary relationships among plant species. Here we compare the responses of six closely related Nicotiana species to a synthetic elicitor, N-linolenoyl-glutamic acid (C18:3-Glu) and HAE of two insect herbivores (the Solanaceae specialist Manduca sexta and generalist Spodoptera littoralis).RESULTS:HAE-induced defences are highly specific among closely related Nicotiana species at three perspectives. 1) A single Nicotiana species can elicit distinct responses to different HAEs. N. pauciflora elicited increased levels of JA and trypsin proteinase inhibitors (TPI) in response to C18:3-Glu and the oral secretions of M. sexta (OS Ms ) but not to oral secretions of S. littoralis (OS Sl ). In contrast, N. miersii only responded to OS Sl but not to the other two HAEs. The specific responses to different HAEs in Nicotiana species are likely due to the perception by the plant of each specific component of the HAE. 2) One HAE can induce different defence responses among closely related Nicotiana species. OS Ms and C18:3-Glu induced JA and TPI accumulations in N. linearis, N. attenuata, N. acuminata and N. pauciflora, but not in N. miersii and N. obtusifolia. 3) The effect of HAE-induced defences differ for the Solanaceae specialist M. sexta and the generalist S. littoralis. Among the four tested Nicotiana species, while the growth rate of M. sexta was only reduced by the induced defences elicited by C18:3-Glu; the growth rate of S. littoralis can be reduced by the induced defences elicited by all three HAEs. This is likely due to differences in the susceptibility of the specialist M. sexta and generalist S. littoralis to induced defences of their host.CONCLUSIONS:Closely related Nicotiana species elicit highly specific defence responses to herbivore associated elicitors and provide an ideal framework for investigating the molecular mechanisms and evolutionary divergence of induced resistance in plants.
Species‐specific Regulation of Herbivory‐induced Defoliation Tolerance is Associated with Jasmonate Inducibility

Abstract:Summary Induced changes in root carbohydrate pools are commonly assumed to determine plant defoliation tolerance to herbivores. However, the regulation and species specificity of these two traits are not well understood. We determined herbivory‐induced changes in root carbohydrates and defoliation tolerance in seven different solanaceous plant species and correlated the induced changes in root carbohydrates and defoliation tolerance with jasmonate inducibility. Across species, we observed strong species‐specific variation for all measured traits. Closer inspection revealed that the different species fell into two distinct groups: Species with a strong induced jasmonic acid ( JA ) burst suffered from a reduction in root carbohydrate pools and reduced defoliation tolerance, while species with a weak induced JA burst maintained root carbohydrate pools and tolerated defoliation. Induced JA levels predicted carbohydrate and regrowth responses better than jasmonoyl‐L‐isoleucine ( JA ‐Ile) levels. Our study shows that induced JA signaling, root carbohydrate responses, and defoliation tolerance are closely linked, but highly species specific, even among closely related species. We propose that defoliation tolerance may evolve rapidly via changes in the plant's defense signaling network.

Abstract:Plants rely on the perception of a multitude of herbivory-associated cues (HACs) to activate their defense response to insect herbivores. These stimuli are mainly derived from three functional components, namely, mechanical damage, insect-associated microbe, and insect's chemical cues. While simulated herbivory integrating these stimuli is widely exploited for complementing actual herbivory in clarifying the details of plant-herbivore interaction, breaking down these stimuli and identifying the mechanisms of plant responses associated with them have been less explored. In this study, the components of potato tuber moth (Phthorimaea operculella, PTM) herbivory were reorganized in a cumulative way and their impacts on the early defense responses of potato leaf were characterized. We found that simulated and actual herbivory of PTM triggered similar patterns of phytohormonal and transcriptomic responses in potato leaf. Moreover, the microbe in the PTM herbivory stimuli is associated with the regulation of the phytohormones jasmonic acid (JA) and abscisic acid (ABA) since reducing the microbe in HAC could reduce JA while increasing ABA. In addition, seven robust gene modules were identified to illustrate how potato plants respond to different PTM herbivory stimuli when herbivory components increased. Significantly, we found that mechanical damage mainly activated JA-mediated signaling; PTM-derived HACs contributed much more to potato early-defense response and induced signaling molecules such as multiple protein kinases; orally secreted bacteria stimuli could antagonize PTM-derived HACs and modulate plant defense, including repressing phenylpropanoid biosynthesis. Our study broadened the understanding of how potato plants integrate the responses to a multitude of stimuli upon PTM herbivory and evidenced that insect-associated microbes greatly modulated the plants response to insect herbivory.

Abstract:One of the main abiotic stresses that strongly affects plant survival and the primary cause of crop loss around the world is drought. Drought stress leads to sequential morphological, physiological, biochemical and molecular changes that can have severe effects on plant growth, development and productivity. As a consequence of these changes, the interaction between plants and insects can be altered. Using cultivated Brassica oleracea plants, the parasitoid Microplitis mediator and its herbivorous host Mamestra brassicae, we studied the effect of drought stress on (1) the emission of plant volatile organic compounds (VOCs), (2) plant hormone titres, (3) preference and performance of the herbivore, and (4) preference of the parasitoid. Higher levels of jasmonic acid (JA) and abscisic acid (ABA) were recorded in response to herbivory, but no significant differences were observed for salicylic acid (SA) and indole-3-acetic acid (IAA). Drought significantly impacted SA level and showed a significant interactive effect with herbivory for IAA levels. A total of 55 VOCs were recorded and the difference among the treatments was influenced largely by herbivory, where the emission rate of fatty acid-derived volatiles, nitriles and (E)-4,8-dimethylnona-1,3,7-triene [(E)-DMNT] was enhanced. Mamestra brassicae moths preferred to lay eggs on drought-stressed over control plants; their offspring performed similarly on plants of both treatments. VOCs due to drought did not affect the choice of M. mediator parasitoids. Overall, our study reveals an influence of drought on plant chemistry and insect-plant interactions.

Abstract:Abstract Cultivated tomato Solanum lycopersicum ( Slyc ) is sensitive to water shortages, while its wild relative Solanum peruvianum L. ( Sper ), an herbaceous perennial small shrub, can grow under water scarcity and soil salinity environments. Plastic Sper modifies the plant architecture when suffering from drought, which is mediated by the replacement of leaf organs, among other changes. The early events that trigger acclimation and improve these morphological traits are unknown. In this study, a physiological and transcriptomic approach was used to understand the processes that differentiate the response in Slyc and Sper in the context of acclimation to stress and future consequences for plant architecture. In this regard, moderate (MD) and severe drought (SD) were imposed, mediating PEG treatments. The results showed a reduction in water and osmotic potential during stress, which correlated with the upregulation of sugar and proline metabolism-related genes. Additionally, the senescence-related genes FTSH6 protease and asparagine synthase were highly induced in both species. However, GO categories such as “protein ubiquitination” or “endopeptidase inhibitor activity” were differentially enriched in Sper and Slyc , respectively. Genes related to polyamine biosynthesis were induced, while several cyclins and kinetin were downregulated in Sper under drought treatments. Repression of photosynthesis-related genes was correlated with a higher reduction in the electron transport rate in Slyc than in Sper . Additionally, transcription factors from the ERF, WRKY and NAC families were commonly induced in Sper . Although some similar responses were induced in both species under drought stress, many important changes were detected to be differentially induced. This suggests that different pathways dictate the strategies to address the early response to drought and the consequent episodes in the acclimation process in both tomato species.

Abstract:The synergy between drought-responsive traits across different organs is crucial in the whole-plant mechanism influencing drought resilience. These organ interactions, however, are poorly understood, limiting our understanding of drought response strategies at the whole-plant level. Therefore, we need more integrative studies, especially on herbaceous species that represent many important food crops but remain underexplored in their drought response. We investigated inflorescence stems and rosette leaves of six Arabidopsis thaliana genotypes with contrasting drought tolerance, and combined anatomical observations with hydraulic measurements and gene expression studies to assess differences in drought response. The soc1ful double mutant was the most drought-tolerant genotype based on its synergistic combination of low stomatal conductance, largest stomatal safety margin, more stable leaf water potential during non-watering, reduced transcript levels of drought stress-marker genes, reduced loss of chlorophyll content in leaves, in combination with stems showing highest embolism resistance, most pronounced lignification and thickest intervessel pit membranes. In contrast, the most sensitive Cvi ecotype shows the opposite extreme of the same set of traits. The remaining four genotypes show variations in this drought syndrome. Our results reveal that anatomical, ecophysiological and molecular adaptations across organs are intertwined, and multiple (differentially combined) strategies can be applied to acquire a certain level of drought tolerance.

Abstract:Plants have acquired a repertoire of mechanisms to combat biotic stressors, which may vary depending on the feeding strategies of herbivores and the plant species. Hormonal regulation crucially modulates this malleable defense response. Jasmonic acid (JA) and salicylic acid (SA) stand out as pivotal regulators of defense, while other hormones like abscisic acid (ABA), ethylene (ET), gibberellic acid (GA) or auxin also play a role in modulating plant-pest interactions. The plant defense response has been described to elicit effects in distal tissues, whereby aboveground herbivory can influence belowground response, and vice versa. This impact on distal tissues may be contingent upon the feeding guild, even affecting both the recovery of infested tissues and those that have not suffered active infestation.

Abstract:Abstract The effect of temperature on insect-plant interactions in the face of changing climate is complex as the plant, its herbivores and their interactions are usually affected differentially leading to an asymmetry in response. Using experimental warming and a combination of biochemical and herbivory bioassays, the effects of elevated temperatures and herbivore damage ( Helicoverpa zea) on resistance and tolerance traits of Solanum lycopersicum var. Better boy (tomato), as well as herbivory performance and salivary defense elicitors were examined. Insects and plants were differentially sensitive towards warming within the experimental temperature range. Herbivore growth rate increased with temperature, whereas plants growth as well as the ability to tolerate stress measured by photosynthesis recovery and regrowth ability were compromised at the highest temperature regime. In particular, temperature influenced the caterpillars’ capacity to induce plant defenses due to changes in the amount of a salivary defense elicitor, glucose oxidase (GOX). This was further complexed by the temperature effects on plant inducibility, which was significantly enhanced at an above-optimum temperature; this paralleled with an increased plants resistance to herbivory but significantly varied between previously damaged and undamaged leaves. Elevated temperatures produced asymmetry in species’ responses and changes in the relationship among species, indicating a more complicated response under a climate change scenario.

Abstract:Breeding for drought-tolerant crops is a pressing issue due to the increasing frequency and duration of droughts caused by climate change. Although important sources of variation for drought tolerance exist in wild relatives, the mechanisms and the key genes controlling tolerance in tomato are little known. The aim of this study is to determine the drought response of the tomato wild relative Solanum pennellii (Sp) compared with the cultivated tomato Solanum lycopersicum (Sl). The paper investigates the physiological and molecular responses in leaves of Sp and Sl plants without stress and moderate drought stress. Significant physiological differences between species were found, with Sp leaves showing greater ability to avoid water loss and oxidative damage. Leaf transcriptomic analysis carried out when leaves did not as yet show visual dehydration symptoms revealed important constitutive expression differences between Sp and Sl species. Genes linked to different physiological and metabolic processes were induced by drought in Sp, especially those involved in N assimilation, GOGAT/GS cycle and GABA-shunt. Up-regulation in Sp of genes linked to JA/ET biosynthesis and signaling pathways was also observed. In sum, genes involved in the amino acid metabolism together with genes linked to ET/JA seem to be key actors in the drought tolerance of the wild tomato species.

Abstract:Drought stress poses adverse influence on plant growth and further alters plant-herbivore interactions. Such effect is enhanced as drought occurrence is reported to increase due to global warming. Although dioecious plant species have shown sex-specific response to drought stress through the changes in growth performance and stress tolerance, whether such changes will drive sex-specific differences in defense against herbivores between male and female plant conspecifics is barely studied. In the current study, female and male poplar full-siblings were submitted to moderate (75 % field water capacity) and severe drought (50 % field water capacity) stresses, followed by herbivore growth and feeding bioassays to test the effect of plant gender on herbivore growth and feeding performance of two specialist and two generalist leaf herbivores. The results showed that although the growth of both plant sexes was inhibited by the two drought levels, male plants performed better than female conspecifics. In the paired-choice bioassays, the specialist herbivores preferred female plants while the generalist herbivores fed more on the male plants without drought stress. Both the moderate and severe drought stresses reversed such preferences. In the triple-choice bioassays, the specialist herbivores preferred female control plants while the generalist herbivores fed more on female plants under severe drought. In addition, the specialist herbivores fed on female plants from severe drought stress grew the worst while the generalist herbivores gained the highest fresh weight. The transcriptomic and metabolomic profiling revealed that female plant leaves contained higher levels of flavonoids than males under control condition while severe drought stress remarkably reduced the levels of defensive metabolites such as flavonoids, isoflavonoids, neoflavonoids and alkaloids in female but not in male plant leaves.

Abstract:Drought alone causes more annual loss in crop yield than all pathogens combined. To adapt to moisture gradients in soil, plants alter their physiology, modify root growth and architecture, and close stomata on their aboveground segments. These tissue-specific responses modify the flux of cellular signals, resulting in early flowering or stunted growth and, often, reduced yield. Physiological and molecular analyses of the model plant Arabidopsis thaliana have identified phytohormone signaling as key for regulating the response to drought or water insufficiency. Here we discuss how engineering hormone signaling in specific cells and cellular domains can facilitate improved plant responses to drought. We explore current knowledge and future questions central to the quest to produce high-yield, drought-resistant crops.

Abstract:Spittlebugs cause large production losses that affect agribusiness worldwide. Understanding plant-herbivore interactions at the molecular level may be the key to developing resistant cultivars. After a nymph survival experiment, root transcriptomes were assembled from two genotypes (BGP 248 and 344) with different first-line defense strategies, with no infestation and at two times after the initial attack of the spittlebug ( nymph, thus integrating differential expression analysis and biological network modeling supplemented by root anatomical analysis. Gene Ontology terms related to different stress responses, such as salicylic acid catabolic process, were enriched in BGP 248, while some specific to spittlebugs, such as response to herbivores, were enriched in BGP 344. Enriched pathways were related to structural differences between genotypes, such as those related to cutin, suberin and wax biosynthesis. BGP 344 also presented pathways related to induced defense, such as glutathione metabolism. Metabolic networks highlighted kinases, and coexpression networks demonstrated a complex cascade response that included lncRNAs. This study provides the first molecular insights into the defense mechanisms of against The genotype with the fastest response to insect attack and highest nymph mortality (BGP 344) presented kinases and an enriched glutathione pathway, in addition to constitutive barriers, such as lignin, which can make it difficult for the insect to colonize the plant.

Abstract:Drought stress is a key factor limiting the growth and tuber yield of potatoes (Solanum tuberosum L.). Brassinosteroids (BRs) have been shown to alleviate drought stress in several plant species; however, little is known about the physiological and molecular mechanisms by which BRs enhance drought resistance in potatoes. Here, we characterized changes in the physiology and transcriptome of the tetraploid potato variety 'Xuanshu-2′ in response to drought stress after 24-epibrassinolide (EBR) pretreatment. The abscisic acid (ABA) content, photosynthetic capacity, and the activities of antioxidant enzymes were increased; the intercellular CO2 concentration, relative conductivity, reactive oxygen species, malondialdehyde, proline, and soluble sugar content were decreased after EBR pretreatment compared with plants under drought stress. Transcriptome analysis revealed 1330 differently expressed genes (DEGs) involved in the response to drought stress after EBR pretreatment. DEGs were enriched in plant hormone signal transduction, starch and sucrose metabolism, circadian rhythm, flavonoid biosynthesis, and carotenoid biosynthesis. DEGs associated with the BR signaling and biosynthesis pathways, as well as ABA metabolic pathways were identified. Our findings provide new insights into the mechanisms by which BRs enhance the drought resistance of potatoes.

Understanding the response in Pugionium cornutum (L.) Gaertn. seedling leaves under drought stress using transcriptome and proteome integrated analysis

Abstract:Background Drought is one of the crucial constraints limiting horticultural plant’s production and development around the world. Pugionium cornutum is an annual or biennial xerophyte with strong environmental adaptability and drought resistance; however, the mechanisms with respect to response to drought stress remain largely unclear. Methods After seedling emergence, the gravimetric method was used to control soil relative water content (SRWC). Drought stress was applied to the six-leaf stage P. cornutum seedlings. The soil water content of different drought stress levels (L) was controlled by gravimetric method as follows: control (L1): 70–75% SRWC; moderate drought level (L2): 40–45% SRWC; severe drought level (L3): 30–35% SRWC, and the water was added to different drought stress levels at about 18:00 p.m. every day. The experiment ended when the leaves of P. cornutum showed severe wilting (10-leaf stage). Samples were harvested and stored at −80 °C for physiological determination, and transcriptomic and proteomic sequencing. Results Compared with L1, the leaves of P. cornutum seedlings were increasingly wilted after drought treatment; the SRWC of the drought-stress leaves decreased notably while the leaf water potential was rose; the proline, malondialdehyde (MDA) content increased with the continuous drought treatment but peroxidase (POD) activity decreased. Besides, 3,027 differential genes (DGs) and 196 differential proteins (DPs), along with 1,943 DGs and 489 DPs were identified in L2-L1 and L3-L1, respectively. The transcriptome and proteome integrated analysis manifested that only 30 and 70 were commonly regulated both in L2-L1 and L3-L1, respectively. Of which, 24 and 61 DGs or DPs showed the same trend including sHSPs, APX2, GSTU4, CML42, and POD, etc . However, most of DGs or DPs were regulated only at the transcriptome or proteome level mainly including genes encoding signal pathway (PYR1, PYLs, SnRK2J, PLC2, CDPK9/16/29, CML9, MAPKs), transcription factors (WRKYs, DREB2A, NAC055, NAC072, MYB and, HB7) and ion channel transporters (ALMT4, NHX1, NHX2 and TPK2). These genes or proteins were involved in multiple signaling pathways and some important metabolism processes, which offers valuable information on drought-responsive genes and proteins for further study in P. cornutum .

Abstract:Root traits respond to drought in a species-specific manner, but little is known about how soil fungal communities and root traits respond to drought in concert. In a glasshouse experiment, we determined the response of soil pathogens, saprotrophs, mutualistic and all fungi associated with the roots of 24 plant species subjected to drought. At harvest, soil fungal communities were characterized by sequencing. Data on root traits were extracted from a previously published work. Differences in fungal beta diversity between drought and control were plant species-specific. For some species, saprotrophic fungi increased in relative abundance and richness with drought while mutualistic fungi showed the opposite pattern. Community structure of pathogenic fungi was plant-species specific but was slightly affected by drought. Pathogen composition was correlated with specific root surface area and root:shoot, saprotroph abundance with root tissue density, while mutualist composition was correlated with root:shoot. All these were the fungal attributes that best predicted shoot mass. Fungal response to drought highly depended on the fungal group and was related to root trait adjustments to water scarcity. This provides new insights into the role that root trait adjustments to drought may have in modulating plant-fungal interactions in grasslands ecosystems.

Abstract:Drought tolerance varies among different peanut (Arachis hypogaea L.) cultivars. Here, drought responses of two cultivars, Huayu 22 (HY22) with drought tolerance and Fuhua 18 (FH18) with drought sensitivity, were compared at the morphological, physiological, biochemical, photosynthetic, and transcriptional levels. Drought stress caused wilting and curling of leaves, bending of stems, and water loss in both cultivars. There was an increase in malondialdehyde (MDA) content under prolonged drought stress, more so in FH18. But the levels of reactive oxygen species (H2O2) and lipid peroxidation were low in HY22. The activities of superoxide dismutase (SOD), peroxidase (POD), and glutathione reductase (GR) were considerably elevated, corresponding with rapid increases in the accumulation of soluble proteins, soluble sugars, and proline. Transcriptional sequencing showed gene expression varied seriously in HY22, which was upregulated in both stems of two cultivars, though downregulation was less pronounced in HY22. KEGG pathway analysis revealed significant enrichment in four leaf and six stem pathways. Additionally, core genes relating to photosynthesis, carbon fixation, proline synthesis, and sucrose and starch synthesis pathways were identified by correlation analysis. Those gene expressions were variously upregulated in stems of two cultivars, especially in HY22, giving a novel view of the shoot as a whole participating in stress response.

Abstract:Abstract Background and Aims Anthropogenic disturbances are causing a co-occurring increase in biotic (ungulate herbivory) and abiotic (drought) stressors, threatening plant reproduction in oak-dominated ecosystems. However, we wonder whether herbivory could compensate for the adverse impact of drought by reducing evapotranspiration. Thus, we investigate the isolated and joint effects of herbivory and drought on oak seedlings of two contrasting Mediterranean species that differ in leaf habit and drought resistance. Methods California oak seedlings from the evergreen, and more drought-resistant, Quercus agrifolia and the deciduous Q. lobata (n=387) were assigned to a fully crossed factorial design with herbivory and drought as stress factors. Seedlings were assigned in a greenhouse to 3-4 clipping levels simulating herbivory and 3-4 watering levels, depending on the species. We measured survival, growth, and leaf attributes (chlorophyll, secondary metabolites, leaf area and weight) once a month (May-Sep) and harvested above- and below-ground biomass at the end of the growing season. Key Results For both oak species, simulated herbivory enhanced seedling survival during severe drought or delayed its adverse effects, probably due to reduced transpiration resulting from herbivory-induced leaf area reduction and compensatory root growth. Seedlings from the deciduous, and less drought-resistant species, benefitted from herbivory at lower levels of water stress, suggesting different response across species. We also found complex interactions between herbivory and drought on their impact on leaf attributes. In contrast to chlorophyll content which was not affected by herbivory, anthocyanins increased with herbivory – although water stress reduced differences in anthocyanins due to herbivory. Conclusions Herbivory seems to facilitate Mediterranean oak seedlings to withstand summer drought, potentially alleviating a key bottleneck in the oak recruitment process. Our study highlights the need to consider ontogenetic stages and species-specific traits in understanding complex relationships between herbivory and drought stressors for the persistence and restoration of multi-species oak savannas.

Abstract:Drought-induced metabolic dysregulation significantly enhances the production of reactive oxygen species (ROS), which, in turn, exerts a substantial influence on the oxidation–reduction regulatory status of cells. These ROS, under conditions of drought stress, become highly reactive entities capable of targeting various plant organelles, metabolites, and molecules. Consequently, disruption affects a wide array of metabolic pathways and eventually leads to the demise of the cells. Given this understanding, this study aimed to investigate the effects of different drought stress levels on the growth and development of the invasive weed Wedelia trilobata and its co-responding native counterpart Wedelia chinensis. Both plants evolved their defense mechanisms to increase their antioxidants and hormone contents to detoxify ROS to avoid oxidative damage. Still, the chlorophyll content fluctuated and increased in a polyethylene-glycol-simulated drought. The proline content also rose in the plants, but W. chinensis showed a significant negative correlation between proline and malondialdehyde in different plant parts. Thus, W. trilobata and W. chinensis exhibited diverse or unlike endogenous hormone regulation patterns under drought conditions. Meanwhile, W. trilobata and W. chinensis pointedly increased the content of indole acetic acid and gibberellic acid in a different drought stress environment. A positive correlation was found between endogenous hormones in other plant parts, including in the roots and leaves. Both simulated and natural drought conditions exerted a significant influence on both plant species, with W. trilobata displaying superior adaptation characterized by enhanced growth, bolstered antioxidant defense mechanisms, and heightened hormonal activities.
