Abstract:Microbial anaerobic oxidation of methane coupled to the reduction of Mn(IV)-oxides, typically MnO 2 , derived from continental weathering and riverine transport has been proposed as a globally important sink of methane. However, the potential role of hydrothermal Mn(IV) oxide-rich sediments as a methane sink, and the mechanisms of underpinning methane oxidation at high-temperature hydrothermal fields remain poorly understood. Here, we report the occurrence of almost pure rhodochrosite with extremely negative δ 13 C PDB values (as low as –76.4‰) in direct association with hausmannite formed through hydrothermal activity in the Late Triassic Heqing Mn deposit in Southern China. Based on detailed petrography and geochemistry, this rhodochrosite is interpreted as the result of the thermochemical oxidation of methane by hausmannite during early diagenesis. Given high hydrothermal Mn 2+ and CH 4 release coupled to low sulfate concentration observed in the Archean oceans, we propose that hydrothermal Mn(IV) oxides may have been effective methane sinks in Earth’s ancient oceans.

Abstract:Methane oxidation affects hydrocarbon accumulation and carbon cycling with important geological and paleoclimatic responses. However, the petrological and geochemical evidences that can clearly discern anaerobic (AOM) and thermochemical (TOM) oxidations in petroliferous basins are unclear, causing the disputes if these two processes can take place in specific conditions. Here, the Baikouquan Formation (T(1)b) in the Mahu Sag, Junggar Basin, China, was used as the first case study for comprehensive petrological and geochemical analyses to explore this scientific issue. Results indicate that the two main types of T(1)b calcite cement record different methane oxidation mechanisms. Calcite cements filling intergranular pores were formed during early diagenesis in relatively shallow-burial stages, through AOM with high-valence Mn oxides as electron acceptors, and with compositions of -47.5 parts per thousand < delta C-13 < -30.9 parts per thousand, 1.1 wt% < MnO < 5.8 wt%, and 0.02 wt% < FeO < 0.13 wt%. Calcite cements filling intragranular dissolution pores were formed through TOM with high-valence Mn oxides as electron acceptors during mesogenesis during relatively deep-burial stages, with compositions of -39.7 parts per thousand < delta C-13 < -14.3 parts per thousand, 0.43 wt% < MnO < 11.00 wt%, and 0.03 wt% < FeO < 0.36 wt%. Thus, methane oxidation underwent a transition from AOM to TOM with increasing depth, as recorded by the calcite cements with different occurrences. This transition may be a common feature of clastic strata in petroliferous basins.

Abstract:Methane bio-oxidation plays an important role in the global methane balance and warming mitigation, while copper has a crucial function in methane bio-oxidation. On one side, copper is known to be a key factor in regulating the expression of the genes encoding the two forms of methane monooxygenases (MMOs) and is the essential metal element of the particulate methane monooxygenase (pMMO). On the other side, the content and fractionation of copper in the environment have great effects on the distribution of methanotrophs and their metabolic capability of methane and non-methane organic compounds, as well as on the copper-specific uptake systems in methanotrophs. Thus, it is meaningful to know the role of copper in methane bio-oxidation for comprehensive understanding of this process and is valuable for guiding the application of methanotrophs in greenhouse gas removal and pollution remediation. In this paper, the roles of copper in methane oxidation were reviewed, including the effect of copper on methanotrophic community structure and activity, the expression and activity of MMOs as well as the copper uptake systems in methanotrophs. The future studies of copper and methane oxidation were also discussed.

Abstract:Strongly 13C-depleted authigenic carbonates (e.g., δ13CVPDB <−30‰; VPDB—Vienna Peedee belemnite) in nature are generally believed to form by sulfate-dependent anaerobic oxidation of methane (AOM). However, we demonstrate using geochemical data and thermodynamic calculation that such calcites are most likely derived from biogenic oxidation of methane in sulfate-poor, nonmarine environments during early diagenesis, as observed in the Triassic sandy conglomerates from the Junggar Basin, northwestern China. This process operated through preferential oxidation of 13C-depleted methane by Mn oxides in closed conditions, producing calcites with higher Mn contents and δ13C values in association with more 13C-enriched residual methane as a result of kinetic isotope fractionation. Thus, the Mn-rich and 13C-depleted carbonates are proposed as tracers of Mn-dependent AOM, which should have served as an important sink of greenhouse methane in low-sulfate early Earth’s oceans.

Abstract:The occurrence of high-valence Mn/Fe oxides in deeply buried oxidized reservoir rocks promotes thermochemical oxidation of hydrocarbons (TOH). This study aimed to better understand this process and its effects on hydrogen isotopes via sampling natural gases from the Lower Triassic red beds with depths of >3.5 km in the Junggar Basin, NW China. The gas compositions, hydrogen and carbon isotopes of C1–C4 alkanes, and carbon isotopes of carbon dioxide were analyzed. The gases are almost mature thermogenic gases generated by sapropelic kerogen from the deeper Lower Permian source rock. The hydrocarbon gases display a unique reversed trend of δ2HCH4 > δ2HC2H6 < δ2HC3H8 > δ2Hn-C4H10, whereas the carbon isotopic compositions (δ13C) of C1–C4 alkanes mostly follow the normal trend of δ13CCH4 < δ13CC2H6 < δ13CC3H8 < δ13CC4H10. The average δ13C values of C1–C4 increased by 10.0‰, 6.0‰, 4.4‰, and 5.1‰, respectively, compared with the initial gases from source rocks, and exhibit a gradually declining trend. In contrast, the δ2H values of C1–C4 increased more from those of normal thermogenic gas, and the size of the 2H-enrichment varies, eventually resulting in the unique reversals. The δ2H and δ13C increments of methane are much higher than those of C2–C4. The reversals cannot be explained by current models for hydrocarbon gas generation, such as mixing of gases from different sources and thermal maturities. Instead, the results are consistent with a model where TOH caused a systematic 13C- and 2H-enrichment of C1–C4 after hydrocarbons charged the reservoir beds enriched in high-valence Mn/Fe oxides. Increase of C1 content accompanied by decreasing of C2–C4, heavy isotopic enrichment of C1–C4, and differential hydrogen isotopic fractionation of C1 and C2–C4 relative to theoretical calculation, suggest C2–C4 were preferentially oxidized during TOH, and C1 had been consumed in reaction. As TOH proceeded the 13C- and 2H-enrichment of C1–C4 was mainly caused by the Rayleigh fractionation. Involvement of formation water in the reaction intermediates of TOH is likely another factor causing the abnormal 2H-enrichment of C1 and the reversals. This study demonstrates that in some circumstances the TOH reactions induced by high-valence metal oxides cause large differences in the hydrogen isotopic compositions of natural hydrocarbons, and the process can be identified via their molecular and hydrogen isotopic signatures.
Denitrifying Anaerobic Methane Oxidation: A Previously Overlooked Methane Sink in Intertidal Zone

Abstract:The intertidal zone is an open ecosystem rich in organic matter and plays an important role in global biogeochemical cycles. It was previously considered that methane was mainly removed by sulfate-dependent anaerobic methane oxidation (sulfate-AOM) process in marine ecosystems while other anaerobic methane oxidation processes were ignored. Recent researches have demonstrated that denitrifying anaerobic methane oxidation (DAMO), consisting of nitrite-dependent anaerobic methane oxidation (nitrite-AOM) and nitrate-dependent anaerobic methane oxidation (nitrate-AOM), can also oxidize methane. In this work, the community structure, quantity and potential methane oxidizing rate of DAMO archaea and bacteria in the intertidal zone were studied by high-throughput sequencing, qPCR and stable isotope tracing method. The results showed that nitrate-AOM and nitrite-AOM were both active in the intertidal zone and showed approximate methane oxidation rates. The copy number of 16S rRNA gene of DAMO archaea and DAMO bacteria were 104 ∼ 105 copies g-1 (dry sediment), whereas NC10 bacteria were slightly higher. The contribution rate of DAMO process to total anaerobic methane removal in the intertidal zone reached 65.6% ∼ 100%, which indicates that DAMO process is an important methane sink in intertidal ecosystem. Laboratory incubations also indicated that DAMO archaea were more sensitive to oxygen and preferred a more anoxic environment. These results help us draw a more complete picture of methane and nitrogen cycles in natural habitats.
Clumped and in situ carbon and oxygen isotopes of calcite as tracers for oxidation of hydrocarbons in deep siliciclastic strata

Abstract:The oxidation of hydrocarbons, including methane, is part of interrelated hydrogeochemical reactions affecting the carbon budget in Earth’s crust. To investigate these processes in deep siliciclastic strata, we analyzed core samples from Lower Triassic red beds in the Mahu Sag (Junggar Basin, northwest China) by coupling petrological observations with high-resolution in situ secondary ion mass spectroscopy stable carbon and oxygen isotope analyses and clumped isotopes (Δ47) of authigenic calcite. The strata contain variable oil and gas content as well as abundant high-valence Fe and/or Mn oxides. Three sequential generations of cement occur, which are characterized as (1) non-luminescent, early diagenetic calcite (MnO <0.3%, δ13CVPDB [Vienna Peedee belemnite] = −5.6‰ to −4.1‰); (2) bright-orange luminescent late-stage I calcite (0.75%−5.23% MnO, δ13C = −51.4‰ to −25.8‰); and (3) dull-orange late-stage II calcite (4.10%−12.93% MnO, δ13C = −91.4‰ to −30.9‰). Clumped isotopic thermometry reveals that the calcite precipitation temperature increases successively from <40 °C, to 81−107 °C, to finally 107−132 °C, corresponding to three precipitation time periods: before the Late Triassic, from the Early Jurassic to the Early Cretaceous, and from the Early Cretaceous to the present, respectively. δ13C values of −55.7‰ to −25.8‰ indicate that late-stage I calcite is the final product of oxidation of both methane and C2+ hydrocarbons, whereas δ13C values as low as −91‰ indicate that late-stage II calcite is mainly derived from the thermochemical oxidation of methane (δ13C = −46.8‰ to −39.3‰) induced by high-valence Mn and/or Fe oxides. For late-stage I calcite, hydrocarbon oxidation was most likely promoted by high temperatures, although microbial oxidation cannot be completely ruled out. The higher precipitation temperature of late-stage II calcite demonstrates that the oxidation of methane requires higher activation energies than oxidation of C2+ hydrocarbons. We provide reliable geochemical evidence for thermally induced sequential oxidation of hydrocarbons within deep siliciclastic strata.

Abstract:Climate warming is a hot environmental issue of global concern. As one of the major methane sinks, the process of methane oxidation coupled with denitrification (MOD) reduces the environmental impact brought by the greenhouse effect and water eutrophication. In addition, as an energy substance, methane can also improve its economic value by transforming into other liquid chemicals. Previous studies on the mechanism of the process have mainly focused on the extracellular electron transfer between species. However, in recent years, the production of intermediates influenced by different factors, and the existence of a large number of acid-producing bacteria and methanogens under anaerobic conditions, has led some researchers to pursue research into a new mechanism of the process. Moreover, the discovery of CO2 as a potential electron acceptor in products is certainly exciting, being a big opportunity under the ‘carbon neutral’ policy. This review looks back at the development of the MOD process and describes its functional microorganism and mechanism in detail when summarizing the types of microorganisms and intermediates at different oxygen levels, and introduces some traditional and novel biotechnologies, such as metagenomics, meta-transcriptomics, and meta-proteomics, etc., to help explore the novel mechanism of the process of MOD mediated by intermediates.

Experimental Investigations on the Thermochemical Oxidation of N-Alkane and Alcohol Compounds by MnO2 and Fe2O3 at Temperatures Up to 325 °C

Abstract:Thermochemical oxidation of organic components (TOOC) coupled to the reduction of Mn(IV) or Fe(III) can occur in a spectrum of geologic settings. Examples of geologic case studies implied that the TOOC can steadily occur at temperatures below 200 degrees C. However, existing relevant experimental work was conducted at temperatures above 350 degrees C. Therefore, uncertainties remain over the reaction at lower temperatures. This study mainly adopted Raman spectroscopy combined with fused silica capillary capsules (FSCCs) to explore the thermochemical oxidation of methane, n-alkanes (here C13H25) and alcohol compounds (here CH3OH and C2H5OH) by MnO2 or Fe2O3 at relatively low temperatures. The results showed that the TOOC reaction can proceed efficiently at temperatures below 200 degrees C, which, to a large extent, decreased the temperature gap between experiments and geological cases. The onset temperature of TOOC is largely dependent on the species of organic matter. Expectedly, long chain n-alkane (C13H25), methanol (CH3OH) and ethanol (C2H5OH) show higher reactivity than CH4 and thus were readily oxidized by oxidants at lower temperatures. The results also implied that either the oxidation of organic components or the reduction of MnO2 proceeds by steps, during which a series of intermediate products are generated. The activation energy of the thermochemical oxidation of CH4 by manganese oxides was also estimated based on quantitative Raman spectroscopic analyses. Finally, we discussed the potential contributions of TOOC to the alteration of organic components, the modification of reservoir properties, the carbon cycles and other geological processes.
13C-enriched carbonate precipitates reveal intense methanogenic oil degradation in the upper Wuerhe Formation, Northwest China

Abstract:Microbial methanogenesis from crude oil is an important source of CH4 for gas reservoirs and atmospheric greenhouse gases. However, its petrological records have not been found in natural environments, and the geological conditions under which it may occur remain unclear. Here, we provide the first petrological evidence of Fe(III)-mediated methanogenic hydrocarbon degradation in a deep subsurface oil reservoir in Northwest China. The major findings are as follows: (1) highly positive δ13C values (up to +16‰) of secondary calcite attributed to methanogenesis; (2) paragenetic relation of high-δ13C calcite to biodegraded hydrocarbon; and (3) remarkably high FeO contents (up to 8 wt%) and heavy δ56Fe ratios (up to +0.52‰) in calcite, indicative of microbial Fe(III) reduction. Our study shows that methanogenic hydrocarbon degradation can occur in Fe(III)-reducing environments. This process transformed hydrocarbons into CO2 and CH4, where the former mostly precipitated as Fe-rich calcite (the carbon sink), while the latter, representing an estimated ⁓1968 Tg, might have escaped into the overburden and atmosphere from the Permian reservoir during the Late Triassic to Early Jurassic, which may have acted as an important CH4 source in changing global climate in the geological past.

Abstract:Direct oxidation of methane into value‐added chemicals is considered as one of the holy grails in chemistry and catalysis. Considerable efforts have been devoted to developing advanced catalyst systems operated under mild conditions, and mononuclear metal species, which are currently in the spotlight of methane oxidation, have shown great advantages and addressed significant attention. In this Concept, we highlight the recent breakthroughs and developments in monomeric metal species for low‐temperature (<100 °C) methane oxidation. Emphasis is put on the identification and validation of monomeric metal species as the intrinsic active sites. Promising characterization approaches for identifying catalytically relevant features of physicochemical properties of monomeric metal species are discussed, as well as the emerging insights on the development of in situ characterization technologies. Finally, the open challenges and outlooks for rational development of monomeric metal catalysts for methane valorization are examined for guiding further developments and inspiring the future work.

Abstract:Abiotic methane has been increasingly detected at the surface of Earth and other terrestrial planets, exerting a strong effect on the study of chemolithoautotrophic life and thus astrobiology. In contrast, abiotic methane generation in subduction zones, which is intimately linked to questions such as the mechanisms of deep carbon mobility, has received scarce attention. Experiments elucidated the significant production of abiotic methane through reduction of carbonate minerals under subduction zone conditions, whereas detailed geological conditions and processes for the reduction in natural rocks are hitherto poorly understood. Here, we report carbonate reduction and genesis of abiotic methane in dolomitized serpentinites (referred to as ophidolomites) from a fossil subduction zone (SW Tianshan, China). Detailed petrological, Raman spectroscopic, strontium and carbon isotopic, and thermodynamic results provide evidence for dolomite reduction into the phase assemblage of calcite + brucite + methane, likely associated with retrograde serpentinization starting at 7-9 kbar and 410-430 degrees C in the subduction zone. Microthermometric data for dolomite-hosted fluid inclusions are consistent with petrographic observations, indicative of fluid entrapment postdating the onset of dolomite reduction during exhumation. Model calculations suggest that water-rich fluids characterized by relatively high hydrogen fugacities can create favorable conditions for the reduction process, which, however, do not exclude the possibility of carbonate methanation by hydrogen-rich fluids as reported in previous studies. The widespread occurrence of methane in these rocks gives credence to the intricate redox transformations of subducted carbon, implying that the elevated hydrogen fugacities may facilitate abiotic synthesis of methane through dolomite reduction at convergent plate boundaries. Our work shows that alteration of dolomite-bearing lithologies represents a potential source for abiotic methane in subduction zones, which may have implications for the transfer of subducted carbon. Abstract:Trivalent iron (Fe 3+ ) could participate in methane (CH 4 ) oxidation as an electron acceptor under anaerobic conditions, but the kinetic process remains unknown in landfills, and the understanding of metabolic pathway of Fe 3+ -dependent anaerobic methane oxidation (Fe-DAMO) is still limited. In this study, the dual-substrate (CH 4 -Fe 3+ ) kinetic model of CH 4 oxidation is obtained with V max (7.35 ± 0.4184)μmol/(kg d), half-saturation constant (16.6699 ± 2.3940)ppmv, and (0.00107 ± 0.0002g/g). Microbial community analysis shows that Methanobacteriales and Clostridia are dominant microorganisms of Fe-DAMO. PICRUSt analysis confirms that the metabolic pathway of AMO is the reverse CH 4 production pathway. The results provide a new perspective for CH 4 biodegradation in landfills and offer a better understanding of Fe-DAMO process.

Abstract:Anoxic sediments and digested sewage sludge anaerobically oxidized methane to carbon dioxide while producing methane. This strictly anaerobic process showed a temperature optimum between 25 and 37°C, indicating an active microbial participation in this reaction. Methane oxidation in these anaerobic habitats was inhibited by oxygen. The rate of the oxidation followed the rate of methane production. The observed anoxic methane oxidation in Lake Mendota and digested sewage sludge was more sensitive to 2-bromoethanesulfonic acid than the simultaneous methane formation. Sulfate diminished methane formation as well as methane oxidation. However, in the presence of iron and sulfate the ratio of methane oxidized to methane formed increased markedly. Manganese dioxide and higher partial pressures of methane also stimulated the oxidation. The rate of methane oxidation in untreated samples was approximately 2% of the CH 4 production rate in Lake Mendota sediments and 8% of that in digested sludge. This percentage could be increased up to 90% in sludge in the presence of 10 mM ferrous sulfate and at a partial pressure of methane of 20 atm (2,027 kPa).

Abstract:Coastal wetlands are widely recognized as atmospheric methane sources. However, recent field studies suggest that some coastal wetlands could also act as methane sinks, but the mechanism is not yet clear. Here, we investigated methane oxidation with different electron acceptors (i.e., oxygen, nitrate/nitrite, sulfate, Fe(III) and Mn(IV)) in four coastal wetlands in China using a combination of molecular biology methods and isotopic tracing technologies. The geochemical profiles and in situ Gibbs free energies suggest that there was significant nitrite-dependent anaerobic oxidation of methane (nitrite-AOM) in the sub-surface sediments; this was subsequently experimentally verified by both the microbial abundance and activity. Remarkably, the methanotrophic communities seemed to exist in the sediments as layered structures, and the surface aerobic methane-oxidizing bacteria were able to take up atmospheric methane at a rate of 0.10-0.18 nmol CH4 day-1 cm-2, while most, if not all, sedimentary methane was being completely consumed by anaerobic methanotrophs (23-58% by methane oxidizers in phylum NC10). These results suggest that coastal methane sinks might be governed by diverse microbial communities where NC10 methane oxidizers contributed significantly. This finding helps to better understand and predict the coastal methane cycle and reduce uncertainties in the estimations of the global methane flux.
Characteristics of Authigenic Minerals around the Sulfate-Methane Transition Zone in the Methane-Rich Sediments of the Northern South China Sea: Inorganic Geochemical Evidence

Abstract:Sediments at marine cold seep areas provide potential archives of past fluid flow, which allow insights into the evolution of past methane seepage activities. However, signals for anaerobic oxidation of methane (AOM) might be obscured in bulk sediments in cold-seep settings due to several factors, especially flood and turbidite deposition. Comprehensive inorganic data were gathered in this study to explore the availability of related records at cold seeps and to provide insights into the evolution of past methane seepage activities. Sediments collected from the site 973-4 in the Taixinan Basin on the northern slope of the South China Sea were characterized in terms of total carbon and sulfur, δ13C values of total organic carbon (δ13CTIC), δ34S values of chromium reducible sulfur (δ34SCRS), and foraminiferal oxygen and carbon isotopes. The results confirmed a strong correlation between formation of authigenic minerals and AOM. Moreover, the 34S enrichments and abundant chromium reducible sulfur (CRS) contents in the authigenic sulfides in the sulfate–methane transition zone (SMTZ) within 619–900 cm below seafloor (cmbsf) reflected past high methane fluxes supported by constant methane seepages. Lithological distribution and AMS (Accelerator Mass Spectra) 14C dating of planktonic foraminifera show that the turbidite (~35.14 ka) was related to a foraminifera-rich interval (Unit II: 440-619 cmbsf) and increased carbonate productivity during the last glacial maximum (LGM). Enrichment of Mo and U was observed accompanied by low contents of nutrient metals (Al, Ti, V, Ni, Fe, Mn, and Cu) in Unit II. The foraminifera-rich interval (Unit II) of cold seep sediments was probably linked to the phenomenon of inconsecutive sedimentary sequence due to the turbidites, which resulted in the lack of Fe, Mn, and Ba enrichment. There is no U enrichment but only Mo enrichment within Unit III, which might be related to H2S produced by AOM during the methane seepages. Based on the above results, it can be speculated that this area has experienced multiple-episodes of methane seep events. Further exploration of AOM should focus on the risks of rapid deposition, especially the impact of turbidity current on sediments.

Abstract:Methane（CH 4 ） is a critical but overlooked component in the study of the deep carbon cycle.Abiotic CH 4 produced by serpentinization of ultramafic rocks has received extensive attention,but its formation and flux in mafic rocks during subduction remain poorly understood.Here,we report massive CH 4 -rich fluid inclusions in well-zoned garnet from eclogites in Western Tianshan,China.Petrological characteristics and carbon-hydrogen isotopic compositions confirm the abiotic origin of this CH 4 .Reconstructed P-T-fO 2 -fluid trajectories and Deep Earth Water modeling imply that massive abiotic CH 4 was generated during cold subduction at depths of 50-120 km,whereas CO 2 was produced during exhumation.The massive production of abiotic CH 4 in eclogites may result from multiple mechanisms during prograde high pressure-ultrahigh pressure metamorphism.Our flux calculation proposes that abiotic CH 4 that has been formed in HP-UHP eclogites in cold sub duction zones may represent one of the largest,yet overlooked,sources of abiotic CH 4 on Earth.
