Abstract:Commercially pure titanium specimens are subjected to three different treatments, and their bioactivity are evaluated by immersing the specimens in a simulated body fluid (SBF, Kokubo's recipe) for various periods up to 7 days, with particular attention being paid to the differences in apatite deposition between surfaces open to SBF and surfaces in contact with the container's bottom. The treatment with a H(2)O(2)/HCl solution at 80 degrees C for 30 min followed by heating at 400 degrees C for 1 h produces an anatase titania gel layer on the specimen surface. This gel layer deposits apatite both on the contact and on open surfaces, and apatite deposition ability does not change with pre-staking in distilled water. The treatment with a NaOH solution at 60 degrees C for 3 days produces a sodium titanate gel layer. This gel layer can deposit apatite only on the contact surface, and the apatite deposition ability is completely lost after 1 day of pre-staking in distilled water. It is concluded, therefore, that the bioactivity of the titania gel originates from the favorable structure of the gel itself while the bioactivity of the sodium titanate gel depends heavily on ion release from the gel. The third treatment, a simple heat treatment at 400 degrees C for 1 h, produces a dense (not porous) oxide layer on the specimen surface. The specimens can deposit apatite on the contact surface after only 3 days of staking in SBF, but they cannot deposit apatite on the open surface for up to 2 months of staking. The implications of such apatite deposition behavior have been discussed in relation to the environments of titanium implants in bone as well as to the methodology of the SBF staking experiment.

Abstract:The purpose of this study was to evaluate the bone-bonding ability of alkali-treated titanium with and without heat treatment. Three groups of smooth titanium plate were prepared: control, or pure titanium, alkali-treated titanium, and alkali- and heat-treated titanium. The plates were inserted transcortically into the proximal metaphyses of bilateral rabbit tibiae. The tensile failure loads between implants and bones were measured at two time intervals using a detaching test. The tensile failure loads of the alkali- and heat-treated titanium group were 2.71 and 4.13 kgf, at 8 and 16 weeks, respectively, and significantly higher than those of the other titanium groups. Histological examination revealed that alkali- and heat-treated titanium was in direct contact with bone, but the other titanium groups had a thin intervening fibrous tissue. This result indicated that the alkali-treated titanium without heat treatment had no bone-bonding ability due to the unstable reactive surface layer of alkali-treated titanium. In conclusion, both alkali and heat treatment are essential for preparing bioactive titanium and this bioactive titanium is thought to be useful for orthopedic implants with cementless fixation.

Abstract:Objective: Alkali treatment and bioactive glass (BG) sol dip-coating are well-known individual methods for titanium (Ti) surface modification. In this study, a unique combination of alkali treatment and bioactive glass sol dip coating was applied to the Ti substrate, then the mechanical properties and cell responses were investigated. Methods: Based on the methods introduced above, the Ti substrate was treated by 6 mL of an NaOH 5 M aqueous solution for 24 h at 60 ̊C; this was followed by adding 1.2 mL of a BG 58S sol to form a novel combined nanostructure network covered by a thin BG layer. For the assessment of the formed coating layer, the morphology, elemental analysis, phase structure, adhesion property and the cell response of the untreated and treated surfaces were investigated. Results: The BG coating layer was reinforced by the nanostructure, fabricated through the alkali treatment. The results obtained by applying the combined modification method confirmed that the mechanical and biological properties of the fabricated surface demonstrated the highest performance compared to that of the unmodified and individually modified surfaces. Significance: The achieved upgrades for this method could be gained from the demanded porous nanostructure and the apatite transformation ability of the alkali treatment. Therefore, the hybridized application of the alkali-BG treatment could be introduced as a promising surface modification strategy for hard-tissue replacement applications.

Abstract:A study was undertaken in rabbit tibiae to determine the effects of chemical treatments and/or surface-induced bonelike apatite on the bone-bonding ability of titanium (Ti) implants. Smooth-surfaced plates (10 x 10 x 2 mm) of pure Ti, alkalil- and heat-treated Ti, and bonelike apatite-formed Ti after the treatments were implanted into the tibial metaphyses of mature rabbits. The tibiae containing the implants were harvested at 4, 8, and 16 weeks after implantation and subjected to a tensile testing and histologic evaluation. Biomechanical results showed that both treated implants exhibited significantly higher failure loads compared with untreated Ti implants at all time periods. Histologic examination by Giemsa surface staining, contact microradiography (CMR), and scanning electron microscopy (SEM) in backscatter mode revealed that both treated Ti implants directly bonded to bone tissue during the early postimplantation period, whereas untreated Ti implants formed direct contact with the bone only at 16 weeks. SEM-electron-probe microanalysis (EPMA) examination showed a Ca-P-rich layer at the interface between the treated implants and bone, although the Ca-P-rich layer was not detected on the surface of untreated implants during observation periods. The results of this study suggest that chemical treatments may accelerate the bone-bonding behavior of titanium implants and enhance the strength of bone-implant bonding by inducing a bioactive surface layer on Ti implants.
Biomimetic Deposition of Hydroxyapatite by Mixed Acid Treatment of Titanium Surfaces

Abstract:A simple chemical method was established for inducing bioactivity of Ti metal. In the present study, two kinds of mixed acid solutions were used to treat Ti specimens to induce Ca-P formation. Following a strong mixed acid activation process, Ca-P coatings successfully formed on the Ti surfaces in the simulated body fluid. Strong mixed acid etching was used to increase the roughness of the metal surface, because the porous and rough surfaces allow better adhesion between Ca-P coatings and substrate. Nano-scale modification of titanium surfaces can alter cellular and tissue responses, which may benefit osseointegration and dental implant therapy. Some specimens were treated with a 5 M NaOH aqueous solution, and then heat treated at 600 °C in order to form an amorphous sodium titanate layer on their surface. This treated titanium metal is believed to form a dense and uniform bone-like apatite layer on its surface in a simulated body fluid (SBF). This study proved that mixed acid treatment is not only important for surface passivation but is also another bioactive treatment for titanium surfaces, an alternative to alkali treatment. In addition, mixed acid treatment uses a lower temperature and shorter time period than alkali treatment.

Abstract:Objective To investigate the biocompatibility of pure titanium (Ti) and Ti-6Al-7Nb surface treated by sanding acid etch (SLA) on rat osteoblasts. Methods Experiments were divided into four groups, Ti mechanical grinding group (S1 group), Ti sand-blasting acid group (SLA1 group), Ti-6Al-7Nb mechanical grinding group (S2 group) and Ti-6Al-7Nb sand-blasting acid group (SLA2 group). The surface topography of samples was examined by microscope. The contact angle measurement instrument was used to analyse surface hydrophily of SLA1 and SLA2 groups. The surface sediment mor?phology and phase were observed by scanning electron microscopy (SEM) and X-ray diffraction (XRD) in two groups after be?ing soaked into simulated body fluid (SBF) for 7 d,14 d and 21 d. Osteoblasts extracted from rats were seeded on titanium sheets, and the osteoblast cells on different titanium surfaces were observed by inverted microscope. MTT colorimetric meth?od was used to measure the proliferation of osteoblasts. Results Compared with S1 and S2 groups, there were more holes on sample surface of SLA1 and SLA2 groups. The sample surface was hydrophilic structure in SLA1 and SLA2 groups. The con?tact angle was smaller in SLA2 group than that of SLA1 group. The hydroxyapatite coating was firstly observed in SLA2 group at 14 d. The hydroxyapatite coating was found in samples of two groups after 21 d. The proliferative ability of osteo?blasts was stronger in SLA1 and SLA2 groups than that of S1 and S2 groups. And the proliferative ability of osteoblasts was stronger in sample surface of SLA2 group than that of SLA1 group (P<0.05). Conclusion Ti-6Al-7Nb by SLA has good biological compatibility, which is helpful to promote the combination of implant and bone tissue.

Abstract:The present study aimed to evaluate the bioactivity of titanium surfaces sandblasted with large-grit corundum and acid etched (SLA) plus further alkali or hydrogen peroxide and heat treatment for dental implant application. Pure titanium disks were mechanically polished as control surface (Ti-control) and then sandblasted with large-grit corundum and acid etched (SLA). Further chemical modifications were conducted using alkali and heat treatment (ASLA) and hydrogen peroxide and heat treatment (HSLA) alternatively. The surface properties were characterized by scanning electron microscopy (SEM), x-ray photoelectron spectroscopy (XPS), and contact angle and roughness measurements. Further evaluation of surface bioactivity was conducted by MC3T3-E1 cell attachment, proliferation, morphology, alkaline phosphatase (ALP) activity and calcium deposition on the sample surfaces. After insertion in the beagle's mandibula for a specific period, cylindrical implant samples underwent micro-CT examination and then histological examination. It was found that ASLA and HSLA surfaces significantly increased the surface wettability and MC3T3-E1 cell attachment percentage, ALP activity and the quality of calcium deposition in comparison with simple SLA and Ti-control surfaces. Animal studies showed good osseointegration of ASLA and HSLA surfaces with host bone. In conclusion, ASLA and HSLA surfaces enhanced the bioactivity of the traditional SLA surface by integrating the advantages of surface topography, composition and wettability.

Abstract:Achieving fast and long-term osseointegration is one of the major goals in dental implant design. Physicchemical surface modification is a key to osseointegration improvement. Two types of alkaline modifications, 1 M sodium bicarbonate and 0.05 M sodium hydroxide, were applied on the sand-blasting and acid etching (SLA) titanium surface. After the alkaline treatments, superhydrophilic SLA surfaces were produced. The mRNA expression level of IL-1b of Raw264.7 cells cultured on the sodium bicarbonate treated SLA surface was inhibited compared with SLA and sodium hydroxide treated surfaces, while MMP-9 expression on the sodium hydroxide treated SLA surface was increased with comparison to SLA and sodium bicarbonate treated surfaces. Cell adhesion of osteoblast-like MG63 cells on surfaces with alkaline treatments were moderately more pronounced than that on the SLA surface. With comparison to SLA surface, the proliferation of MG63 cells was decreased, while the alkaline phosphatase (ALP) activity was enhanced on the sodium bicarbonate treated SLA surface after 3 days. An increased mRNA expression level of integrin av of MG63 cells were observed on the alkali treated surfaces compared with the SLA surface. Integrin b1, osteocalcin (OCN) and bone morphogenetic protein-2 (BMP-2) were enhanced on the sodium bicarbonate treated SLA surface when compared to the SLA surface. Abstract:Objective. The aim of this study was to investigate the effects of nano- or submicro topography and fluoride ion on the biology of osteoblasts.Study design. Pure Ti plates were sandblasted, etched with an HCl/H2SO4 solution (control surface) and then etched in a diluted HF/HNO3 solution (test surface). MC3T3-E1 cells attached, spread, and proliferated on both surfaces.Results. The alkaline phosphatase activity was evidently higher for the test surface than for control surface after 4 and 7 days of cell culture. Real-time PCR showed significant increases in type I collagen and osteocalcin gene expression in osteoblast growth on the test surface after 4 days of culture compared with the control surface.Conclusion. With nanotopography and fluoride, hydrogen ions might improve MC3T3-E1 cell proliferation and differentiation during the early stages of cell culture. Development of a Bioactive Titanium Surface via Alkalinization and Naringenin Coating for Peri-Implant Repair: In Vitro Study

Abstract:This study assessed the effects of titanium (Ti) surface modification with sodium hydroxide (NaOH) associated or not with Naringenin (NA) citrus flavonoid-coating on osteoblastic-like cells (Ob) metabolism. Ti discs were submitted to alkalinization by NaOH solution (5 M, 60 °C) for 24 h; then, the discs were impregnated or not with 100 μg/mL of NA and dried for 1 h at room temperature. The chemical composition, surface topography, and NA release were evaluated. For the biological assays, the discs were placed on 24-well cell culture plates and Ob (Saos-2; ATCC HTB-85) was seeded onto the discs. After different periods, cell adhesion and viability, alkaline phosphatase activity (ALP), and mineralized nodules deposition (MND) were assessed. In addition, cells stimulated with tumor necrosis factor-alpha (TNF-α) were submitted to matrix metalloproteinase (MMP)-2 synthesis and ALP gene expression assessment. Since data presented normal distribution and homogeneity (Shapiro-Wilk; Levene), Student's t-test or one-way ANOVA/post-hoc tests were selected for data analysis (α = 0.05). Higher roughness was observed on Ti discs submitted to NaOH treatment, while the chemical and NA release evaluations indicated the successful adsorption of NA to alkali-treated Ti surface. Higher cell adhesion, cell viability (after 7 days of culture), ALP activity, and MND were observed on Ti NaOH coated with NA compared to the control group (Ti NaOH) (p < 0.05). Moreover, NA coating also promoted decreased MMP-2 synthesis and increased ALP gene expression in the presence of the inflammatory stimulus TNF-α (p < 0.05). The modification of Ti disks with NaOH associated with NA-coating enhanced bone cell metabolism, suggesting that this type of surface modification has a promising potential to accelerate bone repair and formation around dental implants.

Abstract:Background. The aims of this study were to deposit a zinc-hydroxyapatite (Zn-HA) coating on titanium surfaces by using the electrochemical process and investigate the cell response to the Zn-HA-coated titanium surface.Study Design. Surface characteristics were evaluated by scanning electron microscopy (SEM) and inductively coupled plasma atomic emission spectroscopy (ICP-AES). Murine preosteoblast cell (MC3T3-E1) proliferation, alkaline phosphatase (ALP) activity, and osteocalcin release on Zn-HA-coated surfaces were compared with HA-coated surfaces.Results. Field-emission SEM observation showed rod-like HA crystals with a hexagonal cross-section on the HA-coated surface, although the hexagon of the cross-section of Zn-HA crystals became irregular. ICP-AES analysis showed that Zn was present in the Zn-HA coatings at a Zn/(Ca+Zn) molar ratio of 1.04%. Significant increases in cell proliferation, ALP activity on day 7, and osteocalcin produnction on day 14 (P < .05) were observed for Zn2+-containing HA-coated surfaces.Conclusions. The present study showed that a Zn-HA coating deposited by using the electrochemical process enhances proliferation and differentiation of osteoblasts, which has the potential benefit to enhance implant osseointegration. Abstract:Objectives. This study aims to investigate the effects of anatase-structured titania surface on the response of osteoblast-like cells.Methods. Three kinds of titanium (Ti) disks were prepared: the polished Ti (PT), the roughened Ti (RT) formed by sandblasting and acid etching, and the anatase-coated roughened Ti (ART) formed on the RT via H2O2/HCl solution treatment followed by heating at 400 degrees C. The surface topography of Ti disks was observed under a scanning electron microscope. MC3T3-E1 cells were cultured on the Ti disks. Cell morphology was observed by fluorescence microscopy. Cell adhesion and proliferation were assessed by MTT testing. To evaluate the cell differentiation on RT and ART, the expressions of core-binding factor alpha, osterix, bone sialoprotein, collagen type I????, osteocalcin and alkaline phosphatase-2 were measured by real-time reverse transcriptase polymerase chain reaction.Results. The three kinds of Ti disks had distinct surface topographic characteristics. The cells on the ART more rapidly displayed well-defined stress fibers and numerous processes and pseudopodi than the cells on the PT and RT. At 1 and 3 h after culture, there were more cells adhering to the ART than to PT and RT. Cell proliferation did not differ between groups after 1-5 days of culture. In addition, ART generally enhanced the osteogenic gene expression in a time-dependent manner compared with RT.Significance. Anatase-structured titania promotes initial adhesion and spreading of osteoblast-like cells, but does not influence cell proliferation. Abstract:The aim of this study was to evaluate the influence of a roughened H2O2/HCl heat-treated titanium surface on peri-implant bone formation at an early stage in vivo. 24 Ti6Al4V alloy implants were used; half were treated by sandblasted and dual acid-etched treatments (control group), while the others were treated by sandblasted, dual acid-etched and H2O2/HCl heat treatments (test group). The morphology and roughness were analyzed by field emission SEM and atomic force microscopy. The implants were inserted into the femora of 12 adult white rabbits. After 2 and 4 weeks, femora block specimens were prepared for histological and histomorphometric analysis. SEM micrographs showed that multilevel and different sized pits were formed on both surfaces. New bone formation was observed on both implant surfaces. Test implants demonstrated a greater mean percentage of bone-implant contact as compared with controls at 2 (46.84 vs. 41.81, p=0.000) and 4 weeks (49.43 vs. 44.87, p=0.006) of healing. It is concluded that the H2O2/HCl heat-treated rough titanium surface promoted enhanced bone apposition during the early stages of new bone formation around the implant.

Abstract:The large surface area of highly porous titanium structures produced by additive manufacturing can be modified using biofunctionalizing surface treatments to improve the bone regeneration performance of these otherwise bioinert biomaterials. In this longitudinal study, we applied and compared three types of biofunctionalizing surface treatments, namely acid-alkali (AcAl), alkali-acid-heat treatment (AlAcH), and anodizing-heat treatment (AnH). The effects of treatments on apatite forming ability, cell attachment, cell proliferation, osteogenic gene expression, bone regeneration, biomechanical stability, and bone-biomaterial contact were evaluated using apatite forming ability test, cell culture assays, and animal experiments. It was found that AcAl and AnH work through completely different routes. While AcAl improved the apatite forming ability of as-manufactured (AsM) specimens, it did not have any positive effect on cell attachment, cell proliferation, and osteogenic gene expression. In contrast, AnH did not improve the apatite forming ability of AsM specimens but showed significantly better cell attachment, cell proliferation, and expression of osteogenic markers. The performance of AlAcH in terms of apatite forming ability and cell response was in between both extremes of AnH and AsM. AcAl resulted in significantly larger volumes of newly formed bone within the pores of the scaffold as compared to AnH. Interestingly, larger volumes of regenerated bone did not translate into improved biomechanical stability as AnH exhibited significantly better biomechanical stability as compared to AcAl suggesting that the beneficial effects of cell-nanotopography modulations somehow surpassed the benefits of improved apatite forming ability. In conclusion, the applied surface treatments have considerable effects on apatite forming ability, cell attachment, cell proliferation, and bone ingrowth of the studied biomaterials. The relationship between these properties and the bone-implant biomechanics is, however, not trivial.

Abstract:Titanium (Ti) and Ti-based alloys are commonly used in dental implants, and surface modifications of dental implants are important for achieving osseointegration (i.e., direct connection between the implant surface and bone). This study investigated the effect of an eco-friendly etching solution—a hydrogen peroxide–sodium bicarbonate mixture—on the surface properties and contact angles of osteoblast adhesion and proliferation on Ti surfaces. Disk-shaped Ti specimens were prepared using different surface treatments (machining, sandblasting, and sandblasting/acid-etching), and they were immersed in the etching solution and ultrasonically cleaned. Surface characterization was performed using scanning electron microscopy, digital microscopy, contact angle analysis, and X-ray photoelectron spectroscopy. MG-63 osteoblasts were cultured on the specimens, and their adhesion to the specimen surface and proliferation were examined using staining and the MTT assay, respectively. Additional etching with the etching solution caused the formation of nano/micro hierarchical structures, increased surface roughness, and enhanced hydrophilicity. Osteoblast adhesion and proliferation were found to improve on the modified surfaces. The eco-friendly etching method has the potential to enhance the biological properties of Ti implant surfaces and thereby improve dental implant performance.

In vivo evaluation of plasma-sprayed titanium coating after alkali modification

Abstract:In this paper, plasma-sprayed titanium coatings were modified by alkali treatment. The changes in chemical composition and structure of coatings were examined by SEM and AES. The results obtained indicated that a net-like microscopic texture feature, which was full of the interconnected fine porosity, appeared on the surface of alkali-modified titanium coatings. The surface chemical composition was also altered by alkali modification. A sodium titanate compound was formed on the surface of the titanium coating and replaced the native passivating oxide layer. Its thickness was measured as about 150 nm which was about 10 times of that of the as-sprayed coating. The bone bonding ability of titanium coatings were investigated using a canine model. The histological examination and SEM observation demonstrated that more new bone was formed on the surface of alkali-modified implants and grew more rapidly into the porosity. The alkali-modified implants were found to appose directly to the surrounding bone. In contrast, a gap was observed at the interface between the as-sprayed implants and bone. The push-out test showed that alkali-modified implants had a higher shear strength than as-sprayed implants after 1 month of implantation. An interfacial layer, containing Ti, Ca and P, was found to form at the interface between bone and the alkali-modified implant by EDS analysis.

Abstract:Two new modified sandblasted and acid etched (SLA) surface methods had been used on commercially pure Ti (Cp-Ti) surface. Scanning electron microscopy investigation showed that modified SLA surfaces had micro- and nano-structure surface topography. Contact angle test showed that surface hydrophilicity was significantly increased after modified SLA surface modification. Human osteoblast-like MG63 cell attachment test showed that modified SLA surfaces would attach more cells than simple SLA surface. The alkaline phosphatase (ALP) activity assay indicated that ALP activity was enhanced on two modified SLA surfaces relative to SLA and mechanically polished Cp-Ti surface at early stage. Thus, subsequent chemical modification of SLA surface seems to be a promising method to generate better bioactive surface properties.
