Abstract:In this study, cell biosensor chip with micro-electrode arrays was used to monitor the culture behaviour of mammalian cancer cells and evaluate the chemosensitivity of anti-cancer drugs using electrochemical impedance spectroscopy. The electrode arrays were fabricated by semiconductor technology with diameter from 50 pm to 10 pm of each electrode. The human oesophageal cancer cell lines (KYSE 30) were cultured on the surface of the electrode with the help of fibronectin. Morphology changes of cells adhesion, spreading, and proliferation can be detected by impedimetric analysis in a real time and non-invasive way. Anti-caner drug of cisplatin was added to cells for potential drug screening applications. The experimental results show that drug's chemosensitivity effects can be detected by this cell based biosensor.

Abstract:A novel ECL biosensor for cytosensing and cell surface carbohydrate expression evaluation was developed, by the integration of the peptide modified interface for highly specific carbohydrate recognition and sodium alginate loaded glucose oxidase as the signal probe with high signal amplification efficiency. A cysteine-terminated peptide self-assembled on the electrode through Au–S bond to construct a functional interface for cell capture, with decent biocompatibility and high affinity for the human breast cancer cell MCF-7. Concanavalin A lectin modified gold nanoparticles specifically recognized the cell surface carbohydrates and were absorbed on the electrode, followed by the immobilization of multiple glucose oxidase conjugated sodium alginate, which could remarkably increase the sensitivity of the biosensor with enhanced catalysis. The as-proposed ECL cytosensor was successfully applied for the detection of the MCF-7 tumor cells, whose glycans on the cell membranes are over-expressed. A low detection limit of 150cellsmL−1 was obtained, with a wide dynamic linear range from 5.0×102 to 5.0×105cellsmL−1. Due to the excellent sensitivity, stability and biocompatibility, the ECL biosensor would be promising in reliable diagnostics of glycan relevant biomarkers for cancer and other diseases.

Abstract:For use in the voltammetric sensing of galactose-dependent proteins, we modified magnetic beads with a peptide that had both electroactive- and molecular recognition properties. The peptide consisted of a YXY sequence and behaved as an electron-transfer carbohydrate-mimetic peptide that would combine with proteins. With this tool, the protein could be detected via a label-free system. We synthesized several penta- and hexa-peptides with a cysteine residue on the C-terminals to examine the properties of peptides. These peptides contained amino acid residues (X) of alanine, serine, or tyrosine. The peptides were immobilized on magnetic beads via N-(8-maleimidocapryloxy) succinimide. Soybean agglutinin(SBA), the in vivo function of which has been well established in animals, was selected as a model protein. The protein was detected via the changes in electrode response due to the oxidation of tyrosine residues from the phenol group to quinone. As a result, SBA was selectively accumulated on the beads modified with YYYYC. The calibration curve of SBA was linear and ranged from 2.5 × 10-12 to 1.0 × 10-10 M. With this system, SBA was recovered in human serum at values that ranged from 98 to 103%. Furthermore, the beads with peptides were regenerated five times using a protein denaturant. Accordingly, this electrochemical system was simple and could be rapidly applied to the detection of galactose-recognition proteins.

Abstract:Abstract One of the major challenges in the design of biosensors for cancer diagnosis is to introduce a low-cost and selective probe that can recognize cancer cells. In this paper, we combined the phage display technology and electrochemical impedance spectroscopy (EIS) to develop a label-free cytosensor for the detection of cancer cells, without complicated purification of recognition elements. Fabrication steps of the cytosensing interface were monitored by EIS. Due to the high specificity of the displayed octapeptides and avidity effect of their multicopy display on the phage scaffold, good biocompatibility of recombinant phage, the fibrous nanostructure of phage and the inherent merits of EIS technology, the proposed cytosensor demonstrated a wide linear range (2.0 × 10 2 − 2.0 × 10 8 cells mL −1 ), a low limit of detection (79 cells mL −1 , S/N = 3), high specificity, good inter-and intra-assay reproducibility and satisfactory storage stability. This novel cytosensor designing strategy will open a new prospect for rapid and label-free electrochemical platform for tumor diagnosis.

Abstract:Peptides possess many appealing and desirable features, which have attracted increasing attention in the field of electrochemical biosensing. However, peptides hardly produce noticeable electronic signals in response to target binding events. In this work, amphipathic peptides FFFGGGGRGDS with both target recognition and self-assembly capabilities are designed to be co-assembled with the electroactive species ferrocenecarboxylic acid (FcCOOH). Furthermore, the resultant electroactive peptide nanoprobes (ePNPs) are applied for sensitive electrochemical analysis of tumor cells. Specifically, tumor cells are captured by the electrode modified with the corresponding DNA aptamers, and ePNPs can then selectively bind to integrin proteins on the cell surface, thereby accompanied by a remarkable increase of electrochemical signal. Taking the assay of MDA-MB-231 cells, the fabricated biosensor can detect cancer cells with a detection limit of 7 cells mL(-1). Moreover, the ePNPs can act as a universal probe for the detection of different cell lines. Given the merits of easy synthesis, convenient operation, and favorable analytical performance, the proposed biosensor exhibits great potential in developing peptide-based electrochemical biosensing for clinical applications.

Abstract:The identification of biocompatible nanomaterials with high conductivities as sensing interfaces is important in developing novel electrochemical cytosensors. We prepared a novel protein-inorganic nanomaterial-bovine serum albumin (BSA) incorporated Ag nanoflowers with three-dimensional porous architectures, using a simple biomimetic method. The BSA-incorporated Ag nanoflowers were modified on a glassy carbon electrode (GCE) surface and conjugated with a targeting lectin molecule, i.e., Sambucus nigra agglutinin (SNA), for sensing DLD-1 human colon cancer cells. The BSA-incorporated Ag nanoflowers were a suitable platform, and showed improved cell-immobilization capacity, and good biocompatibility, with retention of activity of the immobilized cells. These properties are attributed to the large surface area of the porous structure and the natural BSA layer acting as a biocompatible support. The attachment of DLD-1 cells to the GCE increased the electron-transfer resistance, with a good correlation with the logarithm of the concentration from 1.35×102 to 1.35×107cellsmL−1, with a low detection limit of 40cellsmL−1. Based on the affinity between SNA and sialic acid (SA), the UV–vis absorption spectrum of the one-step reaction between SA and acidic ninhydrin indicated that the average number of SA molecules on a single living DLD-1 cell surface was approximately 2.16×1012. This proposed cytosensing strategy had good reproducibility, acceptable precision, and high specificity for SA-over-expressed cells, indicating that it has potential applications for the early monitoring of tumor cells and convenient evaluation of SA on living cells.

Abstract:A novel reusable and dual-potential responsive electrogenerated chemiluminescence (ECL) biosensor was fabricated for synchronous detection of cancer cells and their surface N-glycan. In this strategy, a cancer cell recognized aptamer hybridized with a capture DNA was immobilized on electrochemically reduced MoS2 nanosheets, and Ru(phen)3(2+) as ECL probes was intercalated into the grooves of the double-strand DNA. In the presence of target cells, the capture DNA and Ru(phen)3(2+) were released from the electrode interface owing to the specific interaction between cancer cells and the aptamer. Meanwhile, concanavalin A (Con A), a mannose binding protein, and a conjugated gold nanoparticle modified graphite-C3N4 (Con A@Au-C3N4) was used as a negative ECL nanoprobe and applied for the cell surface N-glycan evaluation owing to the excellent ECL properties of g-C3N4 at negative potential. The cytosensing and cell surface N-glycan evaluation could be simultaneously realized with high sensitivity and excellent selectivity based on the ratio of ECL intensity between the negative potential and positive potential (ΔECLn/ΔECLp), avoiding the traditional routing cell counting procedures. Moreover, the aptamer modified electrode can be regenerated in the presence of capture DNA solutions for cyclic utilization. As a proof-of-concept, the ECL cytosensor showed excellent performances for the analysis of the MCF-7 cancer cell and its surface N-glycan evaluation in human serum samples. The reusable and dual potential response ECL biosensor endows a feasibility tool for clinical diagnosis and drug screening especially in complex biological systems.
A Simple Electrochemical Lectin-Probe For In Situ Homogeneous Cytosensing And Facile Evaluation Of Cell Surface Glycan

Abstract:This work constructed a novel electrochemical team-probe, ferrocene-concanavalin A (Fc-ConA), for in situ monitoring of cell surface glycan by incorporating the specific recognition ability of lectin to glycan and favorable electrochemical property of ferrocenyl group. The covalent conjugation of ConA with ferrocenyl group was achieved by a carbodiimide coupling reaction and proved with UV-vis absorption spectroscopy and infrared spectroscopy. Cyclic voltammetric behavior of Fc-ConA at glassy carbon electrode demonstrated a reversible diffusion-controlled process A facile homogeneous cytosensing strategy was then developed using Fc-ConA probe for detection of K562 cells. The suspending cells specifically captured Fc-ConA via membrane mannosyl groups and decreased the concentration of free Fc-ConA, producing a response correlative with cell number and the content of cell surface glycan A wide linear response to cells ranging from 1 x 10(4) to 1 x 10(7) cells mL(-1) with a calculated detection limit of 3000 cells mL(-1) was obtained. The lectin-probe could be conveniently used to in situ evaluate cell surface glycan The average number of mannose moieties on single living K562 cell was detected to be 3.0 x 10(10), while this value increased by 81% on drug-treated cells. These results agreed with those from flow cyto-metric detection. Abstract:A new strategy for assessing cell surface carbohydrates and P-glycoprotein (P-gp) expression status and quantifying the cell numbers with an electrochemical immunoassay was designed. In order to construct the base of the cytosensor, a novel 3-D architecture was initially fabricated by combining nitrogen-doped carbon nanotubes, thionine, and gold nanoparticles via a simple layer-by-layer method. The formed architecture provided an effective matrix for concanavalin A (Con A) binding and made the immobilized Con A hold high stability and bioactivity. On the basis of the specific recognition of cell surface mannosyl groups to Con A, the Con A/3-D architecture interface showed a predominant capability for cell capture. With another coupled signal amplification based on a enzymatic catalytic reaction of HRP toward the oxidation of thionine by the H2O2, which was induced by two-step immunoreactions, the proposed cytosensor showed an excellent analytical performance for the detection of He La cells ranging from 8.0 x 10(2) to 2.0 x 10(7) cells mL(-1) with a limit of detection of 500 cells mL(-1). Moreover, with the use of preblocking procedures, the mannosyl groups and P-gp on single He La cell could be further detected to be (4 +/- 2) x 10(10) molecules of mannose moieties and 8.47 x 10(6) molecules of P-gp. This strategy offers great promise for sensitive detection of cancer cells and cell surface receptors and thus may help improve cancer diagnosis and treatment.

Abstract:A multivalent recognition and alkaline phosphatase (ALP)-responsive electrogenerated chemiluminescence (ECL) biosensor for cancer cell detection and in situ evaluation of cell surface glycan expression was developed on a poly(amidoamine) (PAMAM) dendrimer-conjugated, chemically reduced graphene oxide (rGO) electrode interface. In this strategy, the multivalency and high affinity of the cell-targeted aptamers on rGO provided a highly efficient cell recognition platform on the electrode. The ALP and concanavalin A (Con A) coated gold nanoparticles (Au NPs) nanoprobes allowed the ALP enzyme-catalyzed production of phenols that inhibited the ECL reaction of Ru(bpy)(3)(2+) on the rGO electrode interface, affording fast and highly sensitive ECL cytosensing and cell surface glycan evaluation. Combining the multivalent aptamer interface and ALP nanoprobes, the ECL cytosensor showed a detection limit of 38 CCRF-CEM cells per mL in human serum samples, broad dynamic range and excellent selectivity. In addition, the proposed biosensor provided a valuable insight into dynamic profiling of the expression of different glycans on cell surfaces, based on the carbohydrates recognized by lectins applied to the nanoprobes. This biosensor exhibits great promise in clinical diagnosis and drug screening.

Abstract:Quantifying the glycan expression status on cell surfaces is of vital importance for insight into the glycan function in biological processes and related diseases. Here we developed a versatile aptasensor for electrochemical quantification of cell surface glycan by taking advantage of the cell-specific aptamer, and the lectin-functionalized gold nanoparticles acting as both a glycan recognition unit and a signal amplification probe. To construct the aptasensor, amine-functionalized mucin 1 protein (MUC1) aptamer was first covalently conjugated to carboxylated-magnetic beads (MBs) using the succinimide coupling (EDC-NHS) method. On the basis of the specific recognition between aptamer and MUC1 protein that overexpressed on the surface of MCF-7 cells, the aptamer conjugated MBs showed a predominant capability for cell capture with high selectivity. Moreover, a lectin-based nanoprobe was designed by noncovalent assembly of concanavalin A (ConA) on gold nanoparticles (AuNPs). This nanoprobe incorporated the abilities of both the specific carbohydrate recognition and the signal amplification based on the gold-promoted reduction of silver ions. By coupling with electrochemical stripping analysis, the proposed sandwich-type cytosensor showed an excellent analytical performance for the ultrasensitive detection of MCF-7 cells and quantification of cell surface glycan. More importantly, taking advantage of Con A-gold nanoprobe catalyzed silver enhancement, the proposed method was further used for naked-eye tracking glycolytic inhibition in living cells. This aptasensor holds great promise as a new point-of-care diagnostic tool for analyzing glycan expression on living cells and further helps cancer diagnosis and treatment.

Abstract:A novel electrochemical cytosensing strategy was designed based on the specific recognition of integrin receptors on a cell surface to arginine-glycine-aspartic acid-serine (RGDS)-functionalized single-walled carbon nanotubes (SWNTs). The covalent conjugation of the RGDS tetrapeptide to SWNTs was proved with Raman and FF-IR spectra. The conjugated RGDS showed a predominant ability to capture cells on the electrode surface by the specific combination of RGD domains with integrin receptors. With the use of BGC-823 human gastric carcinoma cells (BGC cells) as a model, the cell surface mannosyl groups could specifically bind with horseradish peroxidase labeled concanavalin A, producing an electrochemical cytosensor. On the basis of the dual signal amplification of SWNTs and enzymatic catalysis, the cytosensor could respond down to 620 cells mL(-1) of BGC cells with a linear calibration range from 1.0 x 10(3) to 1.0 X 10(7) cells mL-1, showing very high sensitivity. The dual signal amplification could be further used to evaluate the mannosyl groups on the cell surface, and the mannosyl groups on a single living intact BGC cell were detected to correspond to 5.3 x 10(7) molecules of mannose. This strategy presents a promising platform for highly sensitive cytosensing and convenient evaluation of surface carbohydrates on living cells.

Abstract:<p>Simple, sensitive and rapid detection of circulating tumor cells (CTCs) is of great importance for early diagnosis and therapy of cancers. Overexpression of sugar units on cell surface is related to the phenotypes of many cancers. Based on the boronate ester interaction, we reported the electrochemical and colorimetric detection of CTCs with high simplicity and sensitivity. Specifically, ferroceneboronic acid (FcBA) can be measured by differential pulse voltammetry and 4-mercaptophenylboronic acid (MPBA) can induce the aggregation and color change of gold nanoparticles (AuNPs). CTCs captured by the aptamer-modified magnetic beads (Apt-MBs) can sequestrate FcBA or MPBA molecules by the formation of boronate ester bonds, thus leading to the decrease in the electrochemical signal of FcBA or preventing the MPBA-triggered aggregation of AuNPs. Due to the overexpression of sugar groups on the surface of CTCs, the amplification-free methods exhibited high sensitivity and obviated the use of additional antibody or aptamer for the recognition of captured cells. With MCF-7 cancer cell as the model, 50 cells can be readily determined by the electrochemical and colorimetric methods. The proposed strategy is valuable for probing of cell glycosylation and designing of novel sensing devices for detection of sugar-containing biological macromolecules and cells.</p>

Abstract:A facile electrochemiluminescent (ECL) strategy for in situ label-free monitoring of carbohydrate expression on living cells was designed by integrating the specific recognition of lectin to carbohydrate with a carbohydrate-functionalized CdS nanocomposite. The mercaptopropionic acid-capped CdS quantum dots were firstly immobilized on carbon nanotubes modified electrode and then functionalized with carbohydrate using mannan as a model on the surface. The carbohydrate-functionalized CdS nanocomposite showed high ECL sensitivity and good stability, and could be used for competitive recognition to concanavalin A with the target cells in solution, which led to a change of ECL intensity due to the resistance of concanavalin A. The change depended on both the cell number and the expression level of cell surface carbohydrate. A wide linear response to cells ranging from 2×103 to 1×107cellsmL−1 with a detection limit of 1.2×103cellsmL−1 was obtained. The proposed biosensor could be used to in situ evaluate cell surface glycan, and the average number of mannose moieties on single living BGC cell was detected to be 8.7×107. This sensitive strategy was further used for facile monitoring of dynamic carbohydrate expression on living cells in response to drugs. The proposed method could be further expanded to high-throughput detection with the addition of more specific glycan–lectin pairs to the repertoire.
Fluorescent visual quantitation of cell-secreted sialoglycoconjugates by chemoselective recognition and hybridization chain reaction.

Abstract:Sialic acid (SA), usually located at the termini of glycan chains, is one of the most important monosaccharide blocks for glycosylation of proteins. The expression level of sialoglycoconjugates (SiaGCs) in cellular secretome is of great significance in diagnosis of tumor malignancy. This work developed a fluorescent visual method for the detection of SiaGCs secreted from living cells by a boronic acid modified chip based chemoselective recognition and hybridization chain reaction. The cell-secreted SiaGCs, which were labeled with the azide group through a metabolic labeling technique during cell culture, were captured by the chip through chemoselective recognition of boronic acid toward SA. After further conjugating the azide group with an alkyne modified DNA probe, the captured SiaGCs could be conveniently endowed with the amplified fluorescent signal through a hybridization chain reaction of a pair of dye-labeled DNA hairpins, which led to a quantitative imaging method for detection of SiaGCs. The average amount of metabolically labeled SiaGCs secreted from a single HeLa cell and MCF-7 cell was 2.18 x 10(-17) and 3.98 x 10(-17) mol, respectively. The proposed method could be utilized to monitor the variation of the secreted SiaGCs during drug treatment, providing a useful tool for investigating the glycosylation and glycan-related biological processes.

Abstract:Circulating tumor cells (CTCs), as potential cancer biomarkers, play a crucial role in cancer diagnosis, prognosis and treatment. However, there are still significant challenges due to their rarity. In this paper, a label-free and ultrasensitive electrochemical impedance cytosensor was developed by a galactosylated gold-nanoisland (Gal-GNI) biointerface for the capture and detection of rare cancer cells (HepG-2) in whole blood samples. By mimic the natural multivalent biorecognition systems, the biointerface was designed to capture the target cancer cells by the specific binding between galactosyl ligands of Gal-GNI and over-expressed asialoglycoprotein receptors (ASGP-R) on the surface of HepG-2. From the results of electrochemical impedance spectroscopy (EIS) and fluorescence spectroscopy, the nanostructured Gal-GNI provides an improved electron-transfer rate, specific detection for HepG-2, and good biocompatibility for viable captured cells. The capture of HepG-2 to the EIS cytosensor increased the electron-transfer resistance, with a good correlation with the logarithm of the concentration from 1.0 × 102 to 1.0 × 105 cells mL− 1, with a low detection limit of 30 cells mL− 1. This cytosensing strategy has good reproducibility with the RSD of 1.9% in blood samples, indicating that it can be used as a potential noninvasive assay tool for the early diagnosis and therapy of tumor.
Highly Sensitive Fluorescent Analysis of Dynamic Glycan Expression on Living Cells Using Glyconanoparticles and Functionalized Quantum Dots

Abstract:A double signal amplification strategy was designed for highly sensitive and selective in situ monitoring of carbohydrate on living cells. The double signal amplification included the multiplex sandwich binding of functionalized quantum dots (QDs) to both glycan groups on the cell surface and glyconanoparticles and a cadmium cation sensitized fluorescence emission of Rhod-5N. Using the sialic acid-phenylboronic acid recognition system as a model, the 3-aminophenylboronic acid functionalized QDs (APBA-QDs) were synthesized by covalently binding APBA to mercaptopropionic acid capped CdS QDs, and the glyconanoparticles, polysialic acid stabilized gold nanoparticles (PSA-AuNPs), were prepared by a one-pot procedure. The APBA-QDs first recognized the sialic acid (SA) groups on BGC-823 human gastric carcinoma (BGC) cells and then the PSA on AuNPs, which were further used to bind more APBA-QDs on the cell surface for signal amplification. After the bound QDs were dissolved to release the Cd2+, a Cd2+ sensitized fluorescence method was developed for the detection of BGC cells in a linear range from 5.0 x 10(2) to 1.0 x 10(7) cells rnL(-1) with a limit of detection down to 210 cells mL(-1) (8 cells in 40 mu L of solution) and the dynamic monitoring of SA expression variation on the cell surface. The monitoring result was identical with that from flow cytometric analysis. This approach showed high specificity and acceptable reproducibility. This strategy provided a promising platform for highly sensitive cytosensing and cytobiologic study.

Abstract:A dual-functionalized nanoprobe was designed for highly sensitive and selective in situ evaluation of carbohydrates on living cells by integrating the specific carbohydrate recognition and enzymatic signal amplification of proteins on Au nanoparticles. A nanoscaffold of nanohorns functionalized with arginine-glycine-aspartic acid-serine tetrapeptide was also prepared on an electrode surface for cell capture and enhancing the electrical connectivity. Combined with the nanoprobe and peptide-nanohorns, a highly sensitive electrochemical strategy was developed for cytosensing, which showed a detection limit down to 15 cells, broad dynamic range, acceptable rapidity, and low cost. The proposed method was further used for monitoring of dynamic variation of carbohydrate expression on cancer cells in response to drugs, which obviated the destruction or labeling of cells and the covalent tagging of lectin and enzyme. The one-pot conjugation of three components was very convenient and could be extended for preparation of other lectin-functionalized nanoprobes. Further development of this technique would contribute considerably to meeting the challenges in comprehensive understanding of the glycomic codes.
Sensitive Electrochemical Aptamer Biosensor for Dynamic Cell Surface N-Glycan Evaluation Featuring Multivalent Recognition and Signal Amplification on A Dendrimer-Graphene Electrode Interface

Abstract:We demonstrate a multivalent recognition and highly selective aptamer signal amplification strategy for electrochemical cytosensing and dynamic cell surface N-glycan expression evaluation by the combination of concanavalin A (Con A), a mannose binding protein, as a model, conjugated poly(amidoamine) dendrimer on a chemically reduced graphene oxide (rGO-DEN) interface, and aptamer- and horseradish peroxidase-modified gold nanoparticles (HRP-aptamer-AuNPs) as nanoprobes. In this strategy, the rGO-DEN can not only enhance the electron transfer ability but also provide a multivalent recognition interface for the conjugation of Con A that avoids the weak carbohydrate protein interaction and dramatically improves the cell capture efficiency and the sensitivity of the biosensor for cell surface glycan. The high-affinity aptamer- and HRP-modified gold nanoparticles provide an ultrasensitive electrochemical probe with excellent specificity. As proof-of-concept, the detection of CCRF-CEM cell (human acute lymphoblastic leukemia) and its surface N-glycan was developed. It has demonstrated that the as-designed biosensor can be used for highly sensitive and selective cell detection and dynamic evaluation of cell surface N-glycan expression. A detection limit as low as 10 cells mL(-1) was obtained with excellent selectivity. Moreover, this strategy was also successfully applied for N-glycan expression inhibitor screening. These results imply that this biosensor has potential in clinical diagnostic and drug screening applications and endows a feasibility tool for insight into the N-glycan function in biological processes and related diseases.
