Abstract:The objective was to develop naturally derived polymer-based hydrogels with high mechanical strength and a controlled delivery of drug for extended period of time. Here, we report the fabrication of chemically cross-linked polyvinyl alcohol-graft-poly(acrylic acid)/sodium alginate hydrogel as a semi-interpenetrating polymer network (SIPN). For the preparation of SIPN hydrogels, SA and PVA were cross-linked with AA monomer in the presence of co-monomer EGDMA through free-radical polymerization reaction, using APS as an initiator. Loxoprofen sodium was loaded as a model drug. FTIR, XRD, TGA, and DSC were performed for the characterization of copolymer. Surface morphology was studied by SEM. Swelling studies were carried out at low and higher pH to evaluate pH-dependent swelling of formed SIPN hydrogels. FTIR, XRD, TGA, and DSC studies confirmed the formation of a new copolymer. Developed SIPN hydrogels showed maximum swelling, drug loading, and drug release at pH 7.4 while low at pH 1.2. Moreover, formulations with higher AA contents showed maximum swelling at 7.4 pH. High drug loading and higher drug release have been observed at pH 7.4. In vitro release profile of loxoprofen sodium was found dependent on pH, concentration of monomers, and cross-linking agent. Gel% and yield% for the prepared SIPN hydrogels were determined and found that gel% or yield% is directly proportional to the concentration of polymers, i.e., SA and PVA, due to their behavior as macromolecule radicals for monomer. The results from FTIR analysis showed that both SA and PVA react with the acrylic acid monomer during the polymerization process and result into the formation of SIPN. The formation of semi-IPN structure significantly improved the surface morphology of SIPN hydrogels as evident by SEM, which corresponds to their improved swelling ability and mechanical strength. Drug release mechanism from the formed SIPN was explained by kinetic modeling and found that first-order, Higuchi model, and Korsmeyer–Peppas model are the best fit models to explain drug release from hydrogels. Conclusively, prepared hydrogels were highly pH-responsive and showed good mechanical strength and time-dependent drug release. SIPN hydrogels could be a potential carrier network for controlled delivery of loxoprofen sodium for extended period of time.

Abstract:The aim of the current study was to prepare glutamic acid/polyvinyl alcohol based hydrogels by the free radical polymerization technique. The FTIR analysis of glutamic acid, polyvinyl alcohol, acrylic acid, and ketorolac tromethamine indicated the development and loading of the drug by the prepared hydrogels. DSC and TGA revealed the increase in the thermal stability of glutamic acid and polyvinyl alcohol after crosslinking and polymerization process, thus revealed high thermal stability for the fabricated hydrogel as compared to hydrogel contents. XRD depicted a reduction in the crystallinity of the reagents, which indicated the strong crosslinking and formation of a new polymeric network of hydrogel. SEM indicated a hard and dense surface of the prepared hydrogel, which may be correlated with the strong polymerization among hydrogel contents. Similarly, the effects of cross-linked polymers and monomer on polymer volume fraction, dynamic swelling and in-vitro drug release studies were investigated in three different pH values i.e., pH 1.2, 4.6, and 7.4, respectively. High swelling and drug release were observed as the pH of the medium was enhanced from 1.2 to 4.6 and 7.4, thus, revealed a pH-responsive nature of the prepared hydrogels. Moreover, percent porosity and drug loading were performed for fabricated hydrogels. Increase in percent porosity and drug loading was seen with the increasing composition of glutamic acid and acrylic acid while a decrease was perceived with the increase in the composition of polyvinyl alcohol. Results of sol-gel analysis revealed an increase in the cross-linking degree of the hydrogels with the increasing composition of glutamic acid, polyvinyl alcohol, and acrylic acid, while a decrease in un-crosslinking was observed, respectively. The degradation rate of hydrogel was decreased as the compositions of glutamic acid, polyvinyl alcohol, and acrylic acid was increased, which indicated a compatible network formation of hydrogel with the incorporation of high hydrogel contents. Similarly, in-vivo study was performed on rabbits and the results of pharmacokinetic parameters indicated a controlled release of the drug from the developed hydrogels for a long period of time. HET-CAM test revealed no toxic effect of the polymeric hydrogel on the chorioallantoic membrane of the fertilized chicken eggs. Therefore, regarding the results of all studies, we can conclude that the prepared hydrogels could be used for the controlled drug delivery system.

Abstract:The objective of the current study work was to fabricate sodium poly(styrene sulfonate-co-poly acrylic acid) (SPSPAA) hydrogels by using a free radical co-polymerization method for controlled delivery of ketorolac tromethamine (KT). Polymer (sodium poly(styrene sulfonate) (SPS) polymerized with monomer acrylic acid (AA) in the presence of initiator ammonium peroxodisulfate (APS) and cross-linker N′,N′-Methylene bisacrylamide (MBA). Different combinations of polymer, cross-linker and monomer, were employed for development of polymeric hydrogels. Various studies such as sol-gel, drug loading, dynamic swelling, and drug release studies were carried out to know the sol and gel portion of SPSPAA, swelling behavior of hydrogels at different pH media (1.2 and 7.4), quantification of drug loaded by fabricated hydrogels, and amount release of KT at pH 1.2 and 7.4. Higher dynamic swelling was found at pH 7.4 compared to pH 1.2, and as a result, greater percent release of drug was perceived at pH 7.4. Thermal stability, crystallinity, confirmation of functional groups and development of a new polymeric system, and surface morphology were evaluated via Thermogravimetric Analysis (TGA), Differential Scanning Calorimetry (DSC), Powder X-ray Diffraction (PXRD), Fourier Transform Infrared Spectroscopy (FTIR) and Scanning Electron Microscopy (SEM) respectively. The results showed that the present work could be used as a potential candidate for controlled delivery of KT.

Abstract:This research was carried out to create a pH-responsive polymeric system for the targeted drug delivery of Diloxanide furoate. It relied on sodium alginate (Na-Alg) and Carbopol 934P as building blocks. Using an aqueous free radical polymerization method, SCH1-SCH12 was created with varying polymer, MAA, and MBA input ratios. Positive outcomes were seen in the swelling and release profiles at higher pH levels. Hydrogel formation, as well as component compatibility, thermal stability, and Diloxanide furoate loading, were all validated by instrumental characterization. A drug loading percentage of 83.56% was determined, with the swelling reaching 743.19%. For the formulation with MBA, the gel fraction was 94.58%. The release of diloxanide furoate increased to 91.77% at neutral pH. The formulation containing Carbopol 934P provided the highest mucoadhesion force (3993.42 dynes/cm2). The created hydrogel has been shown to be biocompatible by toxicological testing of the network. Based on the findings, the created polymeric nexus proved promising for pH-dependent localized and regulated delivery of Diloxanide furoate.

Abstract:The aim of the current study was to fabricate naturally derived polymer based hydrogels for controlled release of diclofenac sodium (DS) for a long duration of time. In this research work, sodium alginate-co-poly(2-acrylamido-2-methyl propane sulphonic acid) (SA-co-poly(AMPS)) hydrogels were prepared by the free radical polymerization technique, where sodium alginate (SA) and 2-acrylamido-2-methyl propane sulphonic acid (AMPS) were used as the polymer and monomer while ammonium peroxodisulfate (APS) and N,N′-Methylene bisacrylamide (MBA) were used as the initiator and cross-linker, respectively. A swelling study was performed to determine the swelling index of developed hydrogels in both acidic (pH 1.2) and basic (pH 7.4) media and pH-independent swelling was observed due to the presence of AMPS. An in vitro release study was conducted to evaluate the percentage of drug released, and a high release of the drug was found at the higher pH of 7.4. Sol–gel analysis was performed to analyze the crosslinked and uncrosslinked part of the hydrogels, and results showed a rise in gel fraction as the composition of SA, AMPS and MBA increased while the sol fraction decreased and vice versa. This work demonstrated a potential for sustained delivery of diclofenac sodium by employing various concentration of SA, AMPS and MBA.

Abstract:Thermosensitive hydrogels were prepared by free radical polymerization of N-isopropylacrylamide (NIPAM) and 2-acrylamido-2-methylpropanesulphonic acid (AMPS) in presence of polyethylene glycol (PEG) as macroinitiator. The incorporation of functional monomer in the hydrogel was confirmed by infrared spectroscopy. The equilibrium swelling ratio (ESR), deswelling rate, volume transition temperature, and surface morphology of synthesized hydrogels are found to be dependent on concentration of AMPS monomer in the feed composition. The equilibrium swelling ratio (ESR) and water retention properties increase with increasing AMPS concentration in hydrogels and decrease with increasing salt concentration in external medium. The hydrogel with high dose of AMPS has high loading efficiency of theophylline drug, whereas the in-vitro release of the drug is delayed for a prolonged period in swollen gels above LCST of PNIPAM.

Abstract:This study aimed to fabricate Gelatin-PVA-co-poly(2-acrylamido-2-methylpropane sulfonic acid)-based hydrogels by applying a free-radical polymerization process to achieve extended-release of sitagliptin and metformin. Gelatin-PVA-co-poly(AMPS)-based hydrogels were developed applying Box–Behnken design of response surface methodology. Gelatin and PVA have been chemically cross-linked with monomer (AMPS) with the help of EGDMA (cross-linker) in the presence of sodium hydrogen sulfate and ammonium peroxo-disulfate as initiators. Sitagliptin and metformin were loaded as the model drug. Sol–gel fraction, FTIR, TGA, DSC, and XRD were performed for the fabricated hydrogel. Properties such as swelling studies as well as in vitro drug release studies were carried out in buffers of pH 1.2 and 6.8. The independent variables such as different concentrations of polymers, cross-linking agent, and monomer affect the dependent variables as swelling and drug release of sitagliptin and metformin from gelatin-PVA-co-Poly(AMPS)-based hydrogels. Successful cross-linking of constituents and formulation of stable hydrogels was confirmed by FTIR and XRD. While the DSC and TGA confirmed the thermodynamic stability of hydrogels. The fabricated hydrogel showed a pH-independent swelling index and drug release was swelling controlled. Gel content increased by increasing polymer concentration. Drug release from formulated hydrogels was compared with drug release from a marketed product which showed extended release of drugs from formulated hydrogel as compared with a marketed product. The fabricated hydrogel could be a potential carrier for extended-release of sitagliptin and metformin for a prolonged course of treatment which can prevent multiple dosing and side effects thus enhancing patient compliance.

Abstract:Abstract The present study aimed to prepare hydrogel based on polyvinyl alcohol (PVA) and gelatin (Ge) and characterization of PVA/Ge hydrogel for their potential use as a sustained drug delivery system. Polyvinyl Alcohol (PVA) and-Gelatin (Ge) were cross-linked using glutaraldehyde (GA) as a crosslinking agent and hydrochloric acid (HCl) as a catalyst. Different feed polymer ratio and crosslinking agent concentration were used to prepare a series of PVA/Ge hydrogels. The obtained PVA/Ge hydrogels were investigated for dynamic and equilibrium swelling studies. The effect of polymers ratio, degree of crosslinking and pH of the medium on swelling of PVA/Ge hydrogels was investigated. Furthermore, the values of diffusion coefficient (D), volume fraction, polymer-solvent interaction parameter, molecular weight between crosslink and crosslink density were calculated. For swelling studies, 0.05M USP phosphate buffer solutions of different pH (1.2, 5.5, 6.5 and 7.5) were used. For the drug release study, ciprofloxacin HCl was loaded into selected samples as a model drug. The release of drug from these samples was performed for 12 hours in USP phosphate buffers of pH 1.2, 5.5 and 7.5. The release data from these samples were fitted into various kinetic models like zero order, first order, Higuchi and Peppas models to investigate the release mechanism. It was found that by varying the composition of PVA/Ge hydrogel and GA concentration, a significant difference was observed in drug release kinetics. FTIR spectroscopy and X-ray diffraction were used for the characterization of hydrogels. PVA/Ge hydrogel showed sustained release of the model drug at various pH values suggesting its potential use as a sustained drug delivery system.

Abstract:The aim of the current study was to fabricate a pH-sensitive hydrogels for controlled release of Bovine serum albumin (BSA) protein. In this research work, polyvinylalcohol grafted poly(acrylic acid-co-2-acrylamido-2-methylpropane sulfonic acid) (PVOH-g-poly(AA-co-AMPS)) hydrogels with varying AA/AMPS molar ratio were synthesized through free radical graft copolymerization of partially neutralized acrylic monomers and characterized by Fourier transform infrared spectroscopy, Elemental analysis, X-ray diffraction, and Scanning electron microscopy, thermogravimetric analysis. Then, mechanical proprieties of hydrogels were investigated, and drug release behavior with Bovine serum albumin (BSA) as model drug was studied in vitro. The effect of AA/AMPS molar ratio, pH media on the swelling property of hydrogels was optimized. Results indicated that the strong interaction in the hydrogels resulted in the formation of a more stable copolymer. The high values of the grafting percentage which exceeds 800% confirmed the high performance of the grafting and the successful synthesis. Grafting AMPS onto PVOH/AA improved both thermal stability and mechanical properties of hydrogel. The hydrogel is simultaneous sensitive to pH and ionic strength; the swelling ratio maximizes at pH 6.8 medium and shrinks at pH 1.2, and decreases as the ionic strength increases; the swelling dynamic mechanism was explained well by Fickian diffusion and Schott's pseudo-second-order models. Different kinetic models were applied, and it has been observed that release profile of BSA best followed the Hixson–Crowell for the release of drug in all release media. However, Zero order and Korsmeyer–peppas models only for pH 1.2 and pH 7.4. The swelling of the hydrogels and release of BSA from the drug loaded hydrogels occurred through non-Fickian diffusion mechanism. It is believed that this hydrogel could be potentially used for localized drug delivery.

Abstract:This study investigates hydrogels based on 2-Acrylamido-2-methyl-1-propanesulfonic acid sodium salt (AMPS) copolymers, incorporating N-hydroxyethyl acrylamide (HEA) and 3-sulfopropyl acrylate potassium salt (SPA). The addition of HEA and SPA is designed to fine-tune the hydrogels' water absorption and mechanical properties, ultimately enhancing their characteristics and expanding their potential for biomedical applications. A copolymer of AMPS, 2-carboxyethyl acrylate (CEA) combined with methacrylic acid (MAA) as poly(AMPS-stat-CEA-stat-MAA, PACM), was preliminarily synthesized. CEA and MAA were modified with allyl glycidyl ether (AGE) through ring-opening, yielding macromers with pendant allyl groups (PACM-AGE). Copolymers poly(AMPS-stat-HEA-stat-CEA-stat-MAA) (PAHCM) and poly(AMPS-stat-SPA-stat-CEA-stat-MAA) (PASCM) were also synthesized and modified with AGE to produce PAHCM-AGE and PASCM-AGE macromers. These copolymers and macromers were characterized by 1H NMR, FT-IR, and GPC, confirming successful synthesis and functionalization. The macromers were then photocrosslinked into hydrogels and evaluated for swelling, water content, and mechanical properties. The results revealed that the PASCM-AGE hydrogels exhibited superior swelling ratios and water retention, achieving equilibrium water content (~92%) within 30 min. While the mechanical properties of HEA and SPA containing hydrogels show significant differences compared to PACM-AGE hydrogel (tensile strength 2.5 MPa, elongation 47%), HEA containing PAHCM-AGE has a higher tensile strength (5.8 MPa) but lower elongation (19%). In contrast, SPA in the PASCM-AGE hydrogels led to both higher tensile strength (3.7 MPa) and greater elongation (92%), allowing for a broader range of hydrogel properties. An initial study on drug delivery behavior was conducted using PACM-AGE hydrogels loaded with photosensitizers, showing effective absorption, release, and antibacterial activity under light exposure. These AMPS-based macromers with HEA and SPA modifications demonstrate enhanced properties, making them promising for wound management and drug delivery applications.

Abstract:Copolymeric hydrogel was prepared by crosslinking polyvinyl alcohol (PVA) with 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS). The crosslinking of polymers was assisted by microwave radiation using N,N′-methylenebisacrylamide (MBA) as crosslinker and very low quantities of potassium persulfate (KPS) as initiator. The prepared hydrogels were loaded with captopril and subjected to in vitro and in vivo evaluation. The swelling studies were performed at pH 2 and pH 7.4 to determine water absorption at lower and higher pH. The hydrogel formulations with higher proportions of AMPS and appropriate dose of radiation had shown maximum swelling. The prepared PVA-co-poly (AMPS) hydrogels were evaluated by FT-IR, SEM, XRD, and thermal analysis (DSC and TGA). The crosslinking in components was confirmed by FT-IR, XRD, TGA and DSC analysis. The in vitro drug release was measured at wavelength of 205 nm using UV spectrophotometer. Drug release observed was higher at pH 2 than pH 7.4, due to relatively more swelling capacity at lower pH in aqueous medium. For determination of pharmacokinetic behavior, the hydrogel formulations were orally administered to rabbits. Tmax of captopril-loaded hydrogels was increased to 4 h, while Cmax was reduced to 661.853 ng/ml. The prepared polyvinyl alcohol and AMPS polymeric hydrogels may have ability to release captopril for longer duration.

Abstract:Artemisia vulgaris seed mucilage and mucin polymers are cross‐linked in the presence of methylene bisacrylamide and ammonium persulfate via aqueous free radical polymerized with tuned controlled drug release properties. To cope with the constraints of conventional drug delivery systems, site‐specific drug delivery systems are the major focus of researchers. The present research developed water‐swellable, pH‐responsive methacrylic acid‐based hydrogel scaffolds of Artemisia vulgaris seed mucilage with mucin and loaded with acyclovir sodium as a model drug. The developed hydrogel discs are evaluated for diverse parameters. Drug loading efficiency in all formulations ranges from 63% to 75%. The hydrogels exhibited pH‐dependent swelling, displaying optimum swelling in a phosphate buffer (pH 7.4), and insignificant swelling in an acidic buffer (pH 1.2), in addition, they responded well to electrolyte concentrations. The sol‐gel fraction is estimated ranging from 60 to 95%. Dissolution studies unveiled sustained drug release for 24 h in a phosphate buffer of pH 7.4, exhibiting zero‐order release kinetics. Moreover, FTIR spectra confirmed the drug‐excipient compatibility. SEM photomicrographs revealed a rough and porous surface of hydrogel discs with several pores and channels. The PXRD diffractograms exposed the amorphous nature of the polymeric blends. The findings of acute toxicity studies proved the developed hydrogel network is biocompatible. Therefore, these outcomes connote the newly created network as a smart delivery system, able to dispatch acyclovir sodium into the intestinal segment for a prolonged period.

Abstract:Cellulose acetate phthalate-based pH-responsive hydrogel was synthesized for fabrication of polymeric matrix tablets for gastro-protective delivery of loxoprofen sodium. Cellulose acetate phthalate (CAP) was cross-linked with methacrylic acid (MAA) using free radical polymerization technique. Fourier transform infrared (FTIR) spectra confirmed the formation of cross-linked structure of CAP-co-poly(methacrylic acid). Thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) confirmed the thermal stability of polymeric networks, and scanning electron microscopy (SEM) and energy-dispersive X-ray spectrum (EDS) images unveiled that the prepared formulations were porous in nature and thus the developed formulations had shown better diffusibility. Swelling and in vitro drug release was performed at various pHs and maximum swelling and release was obtained at pH 7.4, while swelling and release rate was very low at pH 1.2 which confirmed the pH-responsive behavior of CAP-co-poly(MAA). CAP-co-poly(MAA) copolymer prevents the release of loxoprofen sodium into the stomach due to reduced swelling at gastric pH while showing significant swelling and drug release in the colon. Cytotoxicity studies revealed higher biocompatibility of fabricated hydrogel. Acute oral toxicity studies were performed for the evaluation and preliminary screening of safety profile of the developed hydrogels. Matrix tablets were evaluated for release behavior at simulated body pH. The investigations performed for analysis of hydrogels and fabricated matrix tablets indicated the controlled drug release and gastro-protective drug delivery of CAP-co-poly(MAA) hydrogels and pH-sensitive matrix tablets for targeted delivery of gastro-sensitive/irritative agents. Abstract:The prime objective of present study was to fabricate novel pH sensitive chemically cross-linked interpenetrating network (IPN) of gelatin/polyvinylpyrrolidone-co-poly (acrylic acid) for controlled delivery of 5-fluorouracil at colon-specific site by combining the properties of natural and synthetic polymers. IPN was prepared through free radical polymerization method by using gelatin and polyvinylpyrrolidone (PVP) as polymers, acrylic acid as monomer and Ethylene glycol dimethacrylate (EGDMA) as a cross-linker. Ammonium peroxodisulphate/sodium hydrogen sulphite were utilized to initiate polymerization process. Series of hydrogels were prepared by altering concentration of gelatin, PVP, acrylic acid and EGDMA. Evaluations with TGA, DSC, XRD, SEM, FTIR and sol–gel analysis were performed to investigate thermal stability, amorphous or crystalline nature, surface morphology and existence of functional groups, which confirm stability and effective cross-linking of hydrogels. Swelling and in vitro drug release studies were performed at both acidic (1.2) and basic (7.4) pH. Hydrogels exhibited higher swelling and drug release at pH 7.4 as compared to at pH 1.2, which confirms the pH sensitive behaviour of fabricated hydrogels. However, it was observed that by increasing polymers, monomer and cross-linker ratios, swelling and drug release of fabricated hydrogels decreases while gel fraction increases. Fabricated hydrogels exhibited biocompatibility with biological system as probed through acute oral toxicity study upon rabbits. Therefore, fabricated IPN could act as a potential controlled drug delivery system for 5-Fluorouracil.

Abstract:Hydrogel is one of the most interesting and excellent candidates for oral drug delivery. The current study focuses on formulation development of hydrogels for controlled oral delivery of esomeprazole. The hydrogels were prepared by solution casting method by dissolving polymers in Polyvinyl alcohol (PVA) solution. Calcium alginate, Hydroxyl propyl methylcellulose (HPMC), acrylic acid and chondroitin sulfate were used in the preparation of hydrogels. Fourier transform infrared (FTIR) analysis showed no incompatibilities between drug and excipients used in the preparation of formulations. The hydrogels were characterized for size and surface morphology. Drug encapsulation efficiency was measured by Ultraviolet-visible (UV-VIS) spectroscopy. In vitro release studies were carried out using dissolution apparatus. The formulated hydrogels were then compared with the marketed product in vivo using rabbits. The result indicates that prepared hydrogels have a uniform size with a porous surface. The esomeprazole encapsulation efficiency of the prepared hydrogels was found to be 83.1 ± 2.16%. The esomeprazole-loaded hydrogel formulations showed optimum and Pharmacopeial acceptable range swelling behavior. The release of esomeprazole is controlled for 24 h (85.43 ± 0.32% in 24 h). The swelling and release of drug results make the prepared hydrogels a potential candidate for the controlled delivery of esomeprazole. The release of the drug from prepared hydrogel followed the super case transport-2 mechanism. The in vivo studies showed that prepared hydrogel formulations showed controlled and prolonged release of esomeprazole as compared to drug solution and marketed product. The formulations were kept for stability studies; there was no significant change observed in physical parameters, i.e., (appearance, color change and grittiness) at 40 °C ± 2/75% ± RH. There was a negligible difference in the drug content observed after the stability study suggested that all the formulations are stable under the given conditions for 60 days. The current study provides a valuable perspective on the controlled release profile of Hydroxyl propyl methylcellulose (HPMC) and calcium alginate-based esomeprazole hydrogels.

Abstract:pH-responsive hydrogels based on alginic acid grafted with acrylic acid and ethylene glycol dimethylacrylate in the presence of ammonium persulfate were developed for controlled delivery of Ketorolac tromethamine. The alginic acid based hydrogels were prepared by free radical polymerization technique. Increase in gel fraction was observed with the increase in alginic acid, acrylic acid, and ethylene glycol dimethylacrylate content. The dynamic swelling and drug release studies were conducted at two different pH values (pH 1.2 and 7.4). Maximum swelling and drug release were observed at pH 7.4. The characterization of prepared hydrogels was carried out by using Fourier transform infrared spectroscopy, thermogravimetric analysis, differential scanning calorimetry, powder x-ray diffraction, and scanning electron microscopy. Similarly, in-vivo study was performed on rabbits and greater plasma drug concentration was achieved by fabricated hydrogels as compared to drug solution and commercial product Keten. Conclusively, the fabricated hydrogels can be considered as a potential candidate for controlled delivery of Ketorolac tromethamine.

Abstract:Combining natural polysaccharides with synthetic materials improves their functional properties which are essential for designing sustained-release drug delivery systems. In this context, the Aloe vera leaf mucilage/hydrogel (ALH) was reacted with acrylic acid (AA) to synthesize a copolymerized hydrogel, i.e., ALH-grafted-Polyacrylic acid (ALH-g-PAA) through free radical copolymerization. Concentrations of the crosslinker N,N'-methylene-bis-acrylamide (MBA), and the initiator potassium persulfate (KPS) were optimized to study their effects on ALH-g-PAA swelling. The FTIR and solid-state NMR (CP/MAS 13C NMR) spectra witnessed the formation of ALH-g-PAA. Scanning electron microscopy (SEM) analysis revealed superporous nature of ALH-g-PAA. The gel fraction (%) of ALH-g-PAA was directly related to the concentrations of AA and MBA whereas the sol fraction was inversely related to the concentrations of AA and MBA. The porosity (%) of ALH-g-PAA directly depends on the concentration of AA and MBA. The ALH-g-PAA swelled admirably at pH 7.4 and insignificantly at pH 1.2. The ALH-g-PAA offered on/off switching properties at pH 7.4/1.2. The metoprolol tartrate was loaded on different formulations of ALH-g-PAA. The ALH-g-PAA showed pH, time, and swelling-dependent release of metoprolol tartrate (MT) for 24 h following the first-order kinetic and Korsmeyer-Peppas model. Haemocompatibility studies ascertained the non-thrombogenic and non-hemolytic behavior of ALH-g-PAA.

Abstract:There has been significant progress in the last few decades in addressing the biomedical applications of polymer hydrogels. Particularly, stimuli responsive hydrogels have been inspected as elegant drug delivery systems capable to deliver at the appropriate site of action within the specific time. The present work describes the synthesis of pH responsive semi-interpenetrating network (semi-IPN) hydrogels of N-succinyl-chitosan (NSC) via Schiff base mechanism using glutaraldehyde as a crosslinking agent and Poly (acrylamide-co-acrylic acid)(Poly (AAm-co-AA)) was embedded within the N-succinyl chitosan network. The physico-chemical interactions were characterized by Fourier transform infrared (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), and field emission scanning electron microscope (FESEM). The synthesized hydrogels constitute porous structure. The swelling ability was analyzed in physiological mediums of pH 7.4 and pH 1.2 at 37°C. Swelling properties of formulations with various amounts of NSC/ Poly (AAm-co-AA) and crosslinking agent at pH 7.4 and pH 1.2 were investigated. Hydrogels showed higher swelling ratios at pH 7.4 while lower at pH 1.2. Swelling kinetics and diffusion parameters were also determined. Drug loading, encapsulation efficiency, and in vitro release of 5-fluorouracil (5-FU) from the synthesized hydrogels were observed. In vitro release profile revealed the significant influence of pH, amount of NSC, Poly (AAm-co-AA), and crosslinking agent on the release of 5-FU. Accordingly, rapid and large release of drug was observed at pH 7.4 than at pH 1.2. The maximum encapsulation efficiency and release of 5-FU from SP2 were found to be 72.45% and 85.99%, respectively. Kinetics of drug release suggested controlled release mechanism of 5-FU is according to trend of non-Fickian. From the above results, it can be concluded that the synthesized hydrogels have capability to adapt their potential exploitation as targeted oral drug delivery carriers.
