Abstract:ABSTRACT Gordonia polyisoprenivorans strain VH2, a potent rubber-degrading actinomycete, harbors two latex clearing proteins (Lcps), which are known to be essential for the microbial degradation of rubber. However, biochemical information on the exact role of this protein in the degradation of polyisoprene was lacking. In this study, the gene encoding Lcp1VH2 was heterologously expressed in strains of Escherichia coli, the corresponding protein was purified, and its role in rubber degradation was examined by measurement of oxygen consumption as well as by chromatographic and spectroscopic methods. It turned out that active Lcp1VH2 is a monomer and is responsible for the oxidative cleavage of poly(cis-1,4-isoprene) in synthetic as well as in natural rubber by the addition of oxygen (O2) to the cis double bonds. The resulting oligomers possess repetitive isoprene units with aldehyde (CHO-CH2—) and ketone (—CH2-CO-CH3) functional groups at the termini. Two fractions with average isoprene contents of 18 and 10, respectively, were isolated, thus indicating an endocleavage mechanism. The activity of Lcp1VH2 was determined by applying a polarographic assay. Alkenes, acyclic terpenes, or other rubber-like polymers, such as poly(cis-1,4-butadiene) or poly(trans-1,4-isoprene), are not oxidatively cleaved by Lcp1VH2. The pH and temperature optima of the enzyme are at pH 7 and 30°C, respectively. Furthermore, it was demonstrated that active Lcp1VH2 is a Cu(II)-containing oxygenase that exhibits a conserved domain of unknown function which cannot be detected in any other hitherto-characterized enzyme. The results presented here indicate that this domain might represent a new protein family of oxygenases.

Abstract:Purpose Electron spin resonance (ESR) is used to measure oxygen partial pressure (pO2) in biological media with many clinical applications. Traditional clinical ESR involves large magnets that encompass the subject of measurement. However, certain applications might benefit from a scanner operating within local static magnetic fields. Our group recently developed such a compact scanner for transcutaneous (surface) pO2 measurements of skin tissue. Here we extend this capability to subsurface (subcutaneous) pO2 measurements and verify it using an artificial tissue emulating (ATE) phantom. Methods We introduce a new scanner, tailored for subcutaneous measurements up to 2 mm beneath the skin's surface. This scanner captures pulsed ESR signals from embedded approximate 1‐mm oxygen‐sensing solid paramagnetic implant, OxyChip. The scanner features a static magnetic field source, producing a uniform region outside its surface, and a compact microwave resonator, for exciting and receiving ESR signals. Results ESR readings derived from an OxyChip, positioned approximately 1.5 mm from the scanner's surface, embedded in ATE phantom, exhibited a linear relation of 1/T2 versus pO2 for pO2 levels at 0, 7.6, 30, and 160 mmHg, with relative reading accuracy of about 10%. Conclusion The compact ESR scanner can report pO2 data in ATE phantom from an external position relative to the scanner. Implementing this scanner in preclinical and clinical applications for subcutaneous pO2 measurements is a feasible next phase for this development. This innovative design also has the potential to operate in conjunction with artificial skin graft for wound healing, combining therapeutic and pO2 diagnostic features.

Phosphorescent Microneedle Array for the Measurement of Oxygen Partial Pressure in Tissue

Abstract:The knowledge of the exact oxygen partial pressure in tissue is crucial for patient care and in the treatment of ischemic medical conditions. However, current methods to assess oxygen partial pressure in tissue suffer from a variety of disadvantages, including complex equipment and procedures that necessitate trained personnel. Additionally, the barrier function of the stratum corneum reduces oxygen exchange and can consequently hamper surface measurements of rapidly changing oxygen partial pressure in tissue. To overcome these challenges, a novel, easy-to-use technique to monitor the oxygen partial pressure in tissue using microneedle arrays (MNAs) has been developed. The MNAs can be made from poly(ethyl methacrylate) and poly(propyl methacrylate) and overcome the skin's barrier function to measure oxygen in the capillary bed and interstitial fluid of the skin. The MNAs' tips are embedded with an oxygen-sensitive phosphorescent metalloporphyrin, where the oxygen partial pressure inversely correlates to changes in both emission intensity and phosphorescence lifetime of the in-house developed red emitting Pt-core porphyrin. It was demonstrated that the oxygen-sensing MNAs are sufficiently robust to puncture human skin via rupture of the stratum corneum, and that the MNAs can detect changes in oxygen partial pressure in skin within the physiologically relevant range (0-160 mmHg). Additionally, the MNAs can be combined with a wearable wireless optical readout system, making these oxygen-sensing MNAs a novel wearable and portable method for user-friendly monitoring of oxygen partial pressure in skin.
Research on the Best Measurement Situation Between Optical Probe and Tissue Surfaces in Non-Invasive Detection.

Abstract:Near-infrared spectroscopy is often used for the non-invasive detection of composition in the human body, such as that of blood glucose and haemoglobin, due to its high penetration depth into tissues. Although it is feasible to position the optical probe precisely, contact situation between probe and human tissues is a difficult problem to determine because of physiological tremor and mechanical performance of bio-soft tissue. Here, we proposed a novel estimation method for the situation between the optical probe and tissue surfaces based on the dynamic auto-correlation matrix of two-dimensional correlation spectroscopy (2DCOS) and radar chart. The diffuse reflectance spectra from the left palm of 4 healthy volunteers were collected while the optical probe gradually approached and pressed bio-tissues with a custom-design controlling device. 2DCOS in the wavelength with lower absorption (1000-1400 nm) was calculated under the perturbation of relative-distance and contact pressure between the optical probe and tissue surface. The synchronous 2DCOS showed that the surface reflection and diffuse reflectance were greatly affected by the contact conditions in 1100 nm, 1220 nm, and 1300 nm. Then the dynamic auto-correlation matrix of 2DCOS was established for the adjacent spectra, and the significant difference wavelengths were used to build radar charts to determine the critical contact situation visually. Results showed that the maximum variations of dynamic auto-correlation matrix appeared at near 1300 nm, and the relative distance between the probe and tissue corresponding to the critical contact state can be easily observed with radar charts with 0.25 mm uncertainty, which was consistent with the self-feeling of each volunteer. So this method can be applied to exactly determine the optimal measurement status for the non-invasive body composition detection in vivo. It is important for the design of human-machine interface and the accuracy improvement of body composition measurements.
A Handheld Technology for Optical Monitoring of Transcutaneous Blood Oxygen Saturation

Abstract:This paper presents the use of a handheld technology for noninvasive prediction of one’s transcutaneous blood oxygen saturation, StO2, via an in-house developed skin oxygenation system. The quantification strategy involved the use of wavelength dependent Modified Lambert Beer law and is based on light signals of wavelengths 532 nm, 560 nm and 650 nm reflected from the selected skin site. This study performed at rest and arterial blood occlusion experiment on left palm of the hand of five healthy Asian volunteers to evaluate the performance of the system and to verify the validity of the predicted results. The preliminary results revealed a considerable decrease in the predicted mean percent StO2 value from 63.7 ± 13.2 % for at rest condition to 52.2 ± 11.4 % after a pressure of 140 mmHg was applied on upper left arm of these recruits for 120 seconds. This work concluded that the developed optical system is able to provide comprehensive information on spatially dependent StO2 and it has unlimited skin access, hence may be potentially used in field applications to assess the skin oxygen level of those in workforce whose job is at risk of exposure to poisonous gases.

Abstract:Evaluation of the oxygen-mediated effects of clinical and daily activities demands an on-skin device that can track multi-vital regional tissue hemodynamics simultaneously. For example, peripheral arterial disease (PAD) is the third most prevalent cardiovascular disease, but the means of diagnosing and monitoring this disease are limited because the affected area is usually in the non-pulsatile area away from the heart. Herein, we report on an ultrathin and ultralight multi-vital near-infrared optoelectronic biosensor for the diagnosis and rehabilitation monitoring of regional tissue hemodynamics, which is suitable for mounting on the skin for long-term measurement. The device can simultaneously detect tissue oxygen saturation, heart rate, arterial blood oxygen, and tissue perfusion and shows potential for various hypoxia monitoring applications. Moreover, the tissue hemodynamics detected by this device showed a highly accordance with the ankle-brachial index and CT angiography obtained by traditional clinical methods. Therefore, our design was able to accurately diagnose and effectively evaluate PAD patients before and after surgery. The on-skin optoelectronic biosensor shows potential in biological oxygen-mediated behavior evaluation, injury-state monitoring, PAD clinical diagnosis optimization, and after surgery care.

Abstract:Measurement of intramuscular oxygen could play a key role in the early diagnosis of acute compartment syndrome, a common condition occurring after severe trauma leading to ischemia and long-term consequences including rhabdomyolysis, limb loss, and death. However, to date, there is no existing oxygen sensor approved for such a purpose. To address the need to improve the assessment of compartment syndrome, a portable fiber-optic device for intramuscular oxygen measurements was developed. The device is based on phosphorescence quenching, where the tip of an optical fiber was coated with a poly(propyl methacrylate) (PPMA) matrix containing a brightly emitting Pt(II)-core porphyrin. The optoelectronic circuit is highly portable and is based on a microspectrometer and a microcontroller readout with a smartphone. Results from an in vivo tourniquet porcine model show that the sensor is sensitive across the physiological oxygen partial pressure range of 0-880 mmHg and exhibits an appropriate and reproducible response to changes in intramuscular oxygen. A commercial laboratory oxygen sensor based on a lifetime measurement did not respond as expected.

Abstract:Characterization of the thermal properties of the surface and subsurface structures of the skin can reveal the degree of hydration, the rate of blood flow in near‐surface micro‐ and macrovasculature, and other important physiological information of relevance to dermatological and overall health status. Here, a soft, stretchable thermal sensor, based on the so‐called three omega (i.e., 3ω) method, is introduced for accurate characterization of the thermal conductivity and diffusivity of materials systems, such as the skin, which can be challenging to measure using established techniques. Experiments on skin at different body locations and under different physical states demonstrate the possibilities. Systematic studies establish the underlying principles of operation in these unusual systems, thereby allowing rational design and use, through combined investigations based on analytical modeling, experimental measurements, and finite element analysis. The findings create broad opportunities for 3ω methods in biology, with utility ranging from the integration with surgical tools or implantable devices to noninvasive uses in clinical diagnostics and therapeutics.
Non-invasive method and experimental study for measurement of oxygen saturation rate in wide range

Abstract:This paper describes a nonlinear method of measuring the blood oxygen saturation rate (SpO 2) in wide range on the basis of spectrophotometry, uses the CAN bus communication and fast digital signal processing to implement the non-invasive pulse blood oxygen saturation monitor in wide range and introduces the components and functions of the monitor in detail. This method solves the nonlinear problem in measuring the blood oxygen saturation in wide range, improves the applying range and the precision. Besides, according to the experiments, the oxygen saturation rate can be correctly measured by the pulse oximeter we designed in the wide range of 35% to 100% within ±3% accuracy and the absorption coefficient of the hemoglobin is also analyzed in the discussion. The pulse oximeter based on the method can be used for patients in clinic and the cosmonauts in the field of spaceflight. © 2008 IEEE.
Transcutaneous regional venous oximetry: a feasibility study

Abstract:Background: The arterial pulse oximeter, which was introduced clinically in the 1970s, is a convenient, useful, and now ubiquitous anesthesia monitor. Unfortunately, although percent saturation of arterial hemoglobin is, along with cardiac output and concentration of hemoglobin, one of 3 components of oxygen delivery, it does not indicate whether oxygen delivery to a region of interest is adequate. Knowledge of peripheral or regional venous oxygen saturation (Sxvo₂) may lend insight into analysis of regional oxygen supply and demand. Our goal was to assess the suitability of 3 anatomic sites for the transcutaneous assessment of Sxvo₂. Methods: Using a Nonin reflectance oximetry probe (provided by Nonin Medical, Plymouth, MN) placed directly over the antecubital, external jugular, and internal jugular veins in 10 volunteers, we measured the absorbance of red and infrared electromagnetic radiation. We performed fast Fourier transformation on these absorbance waveforms. The ratio of pulsatile absorbance of red and infrared radiation at different frequencies was compared with nonpulsatile absorption, and Sxvo₂ was calculated based on previously derived empiric correlations. Results: Estimates of transcutaneous Sxvo₂ ranged from 41% to 97%, with mean values of 75%, 80%, and 80% at the antecubital, external jugular, and internal jugular veins, respectively. Overall, 93% of predicted Sxvo₂ values were < 90%. Conclusion: Validation and subsequent improvement of this technique requires correlation of our results with venous blood gas measurements, followed by incorporation of technologies from related fields in oximetry (fetal reflectance oximetry and near-infrared spectroscopy), as well as the development of advanced signal processing techniques.

Abstract:Background: Nowadays, Cardiovascular disease is a severe threat to human health. It is necessary to monitor the cardiovascular parameters such as blood oxygen saturation reliably and conveniently. Non-contact measurement has been widely studied. However, some inevitable factors limit the use of the platform and even lead to inaccurate estimation. Hence, this study proposed a new method to estimate blood oxygen saturation. Methods: This paper introduced a new IPPG device consisting of a dual-wavelength light source system, an enclosed illumination chamber and an RGB camera. In order to get a high-precision SpO2 calculation model, the data obtained and processed from the system are compared to the output of a traditional figure blood volume pulse sensor that was employed on 20 subjects at the same time. The most suitable region of interest (ROI) area was determined based on previous research. Findings: The comparison showed that the data from the new IPPG system is consistent and comparable with the traditional pulse oximeter. Compared to other IPPG-based measuring methods, our new IPPG device can remove the motion artifacts caused by body movement well. What's more, the consistency of the environment was maintained and reduce the interference of the external environment during the experiment because of the device.
Growth of isolated amastigotes of Trypanosoma cruzi in cell-free medium.

Abstract:Amastigotes of Trypanosoma cruzi were purified from overlays of infected Vero cell cultures by centrifugation over a discontinuous gradient of metrizamide. Pure amastigote preparations were usually recovered from the pellet under the layer of specific gravity 1.086. The isolated amastigotes grew in cell-free ML-15HA medium. Growth rate for the different strains of T. cruzi were in the order Y greater than Tulahuen greater than CL. The generation time of amastigotes in ML-15HA medium was 16.8, 18.0 and 26.4 h for the Y, Tulahuen, and CL strains, respectively, in the presence of 5% CO2, and 16.8, 31.2, and 36.4 h, respectively, in the absence of CO2. Intracellular amastigotes did not differ ultrastructurally from amastigotes from either the density-gradient fractionation or culture in cell-free medium.
Validation of a Novel Noninvasive Technology to Estimate Blood Oxygen Saturation Using Green Light: Observational Study

Abstract:Background: Pulse oximeters work within the red-infrared wavelengths. Therefore, these oximeters produce erratic results in dark-skinned subjects and in subjects with cold extremities. Pulse oximetry is routinely performed in patients with fever; however, an elevation in body temperature decreases the affinity of hemoglobin for oxygen, causing a drop in oxygen saturation or oxyhemoglobin concentrations. Objective: We aimed to determine whether our new investigational device, the Shani device or SH1 (US Patent 11191460), detects a drop in oxygen saturation or a decrease in oxyhemoglobin concentrations. Methods: An observational study (phase 1) was performed in two separate groups to validate measurements of hemoglobin and oxygen concentrations, including 39 participants recruited among current university students and staff aged 20-40 years. All volunteers completed baseline readings using the SH1 device and the commercially available Food and Drug Administration-approved pulse oximeter Masimo. SH1 uses two light-emitting diodes in which the emitted wavelengths match with absorption peaks of oxyhemoglobin (hemoglobin combined with oxygen) and deoxyhemoglobin (hemoglobin without oxygen or reduced hemoglobin). Total hemoglobin was calculated as the sum of oxyhemoglobin and deoxyhemoglobin. Subsequently, 16 subjects completed the "heat jacket study" and the others completed the "blood donation study." Masimo was consistently used on the finger for comparison. The melanin level was accounted for using the von Luschan skin color scale (VLS) and a specifically designed algorithm. We here focus on the results of the heat jacket study, in which the subject wore a double-layered heated jacket and pair of trousers including a network of polythene tubules along with an inlet and outlet. Warm water was circulated to increase the body temperature by 0.5-0.8 °C above the baseline body temperature. We expected a slight drop in oxyhemoglobin concentrations in the heating phase at the tissue level. Results: The mean age of the participants was 24.1 (SD 0.8) years. The skin tone varied from 12 to 36 on the VLS, representing a uniform distribution with one-third of the participants having fair skin, brown skin, and dark skin, respectively. Using a specific algorithm and software, the reflection ratio for oxyhemoglobin was displayed on the screen of the device along with direct hemoglobin values. The SH1 device picked up more minor changes in oxyhemoglobin levels after a change in body temperature compared to the pulse oximeter, with a maximum drop in oxyhemoglobin concentration detected of 6.5% and 2.54%, respectively. Conclusions: Our new investigational device SH1 measures oxygen saturation at the tissue level by reflectance spectroscopy using green wavelengths. This device fared well regardless of skin color. This device can thus eliminate racial disparity in these key biomarker assessments. Moreover, since the light is shone on the wrist, SH1 can be readily miniaturized into a wearable device.
A Wearable Device to Monitor the Partial Pressure of Transcutaneous Carbon Dioxide

Abstract:The electrochemical approach with CO2 electrodes is the gold standard for the measurement of transcutaneous CO2 (PtCO2). However, this system requires the continuous calibration of the device, the remembranization of the electrodes, an infrastructure with specialized personnel, and a still position during the measurement. This article presents a wearable device for PtCO2 measurement with an optical approach (nondispersive infrared sensor). The device first measures the environmental CO2, then brings the skin temperature to 42 °C, and finally measures the actual PtCO2. The 30 volunteers (20 men and ten women, mean age 27.2 ± 7.0) were enrolled to validate the new device against the gold standard Sentec Digital monitor. The protocol consisted in 10 min of spontaneous breathing, 2 min of rebreathing in a 2-L bag to induce hypercapnia, and 12 min of recovery. This was repeated for each volunteer because the gold standard probe was placed once on the forearm and once at the earlobe. Also, a conversion method from ppm readings of the new device to mmHg was implemented and evaluated. The new device detects a response to the stimulus of hypercapnia with a time delay comparable to that of the gold standard. Before reaching a baseline, both devices require an assessment time approximatively in the order of minutes. The Sentec device goes back to the baseline after the end of the stimulus, while the new device saturates. After conversion of values into mmHg, the intraclass correlation coefficient (ICC) analysis on median values results in a coefficient of 0.75. The new device has a median root mean square error of 2.38 mmHg (IQR = 1.59 mmHg). The median of the mean absolute error is 2.05 mmHg (IQR = 1.64 mmHg) and the uncertainty is ±4.63 mmHg (IQR = 2.38 mmHg). This work demonstrates that a wearable device to measure PtCO2 based on an optical sensor is technically feasible.

Abstract:There is an essential need to develop wearable multimodality technologies that can continuously measure both blood flow and oxygenation in deep tissues to investigate and manage various vascular/cellular diseases. To develop a wearable dual-wavelength diffuse speckle contrast flow oximetry (DSCFO) for simultaneous measurements of blood flow and oxygenation variations in deep tissues. A wearable fiber-free DSCFO probe was fabricated using 3D printing to confine two small near-infrared laser diodes and a tiny CMOS camera in positions for DSCFO measurements. The spatial diffuse speckle contrast and light intensity measurements at the two different wavelengths enable quantification of tissue blood flow and oxygenation, respectively. The DSCFO was first calibrated using tissue phantoms and then tested in adult forearms during artery cuff occlusion. Phantom tests determined the largest effective source-detector distance (15 mm) and optimal camera exposure time (10 ms) and verified the accuracy of DSCFO in measuring absorption coefficient variations. The DSCFO detected substantial changes in forearm blood flow and oxygenation resulting from the artery occlusion, which meet physiological expectations and are consistent with previous study results. The wearable DSCFO may be used for continuous and simultaneous monitoring of blood flow and oxygenation variations in freely behaving subjects.

Abstract:Traditional methods for measuring blood oxygen use multiple wavelengths, which produce an intrinsic error due to ratiometric measurements. These methods assume that the absorption changes with the wavelength, but in fact the scattering changes as well and cannot be neglected. We found that if one measures in a specific angle around a cylindrical tissue, called the iso-pathlength (IPL) point, the reemitted light intensity is unaffected by the tissue's scattering. Therefore, the absorption can be isolated from the scattering, which allows the extraction of the subject's oxygen saturation. In this work, we designed an optical biosensor for reading the light intensity reemitted from the tissue, using a single light source and multiple photodetectors (PDs), with one of them in the IPL point's location. Using this bio-device, we developed a methodology to extract the arterial oxygen saturation using a single wavelength light source. We proved this method is not dependent on the light source and is applicable to different measurement locations on the body, with an error of 0.5%. Moreover, we tested thirty-eight males and females with the biosensor under normal conditions. Finally, we show the results of measuring subjects in a hypoxic chamber that simulates extreme conditions with low oxygen.

Abstract:The continuous measurement of percutaneous oxygen saturation (SpO2) enables diseases that cause hypoxemia to be detected early and patients' conditions to be monitored. Currently, SpO2 is mainly measured using a pulse oximeter, which, owing to its simplicity, can be used in clinical settings and at home. However, the pulse oximeter requires a sensor to be in contact with the skin; therefore, prolonged use of the pulse oximeter for neonates or patients with sensitive skin may cause local inflammation or stress due to restricted movement. In addition, owing to COVID-19, there has been a growing demand for the contactless measurement of SpO2. Several studies on measuring SpO2 without contact used skin video images have been conducted. However, in these studies, the SpO2 values were estimated using a linear regression model or a look-up table that required reference values obtained using a contact-type pulse oximeter. In this study, we propose a new technique for the contactless measurement of SpO2 that does not require reference values. Specifically, we used certain approaches that reduced the influence of non-pulsating components and utilized different light wavelengths of video images that penetrated subcutaneously to different depths. We experimentally investigated the accuracy of SpO2 measurements using the proposed methods. The results indicate that the proposed methods were more accurate than the conventional method.

Abstract:In article number 1601013, Xue Feng and co-workers report a systematic strategy to design an epidermal inorganic optoelectronic device by using specific strain-isolation design, nano-diamond thinning and hybrid transfer printing. This device has similar mechanical properties to epidermis and it is totally conformal to skin and keeps the constant light transmission between emitting element and photodetector as well as the signal stability due to skin deformation. It is also utilized to quantitatively detect human peripheral capillary oxygen saturation and cardiac pulse rate in vitro experiment. This strategy opens a route for traditional inorganic optoelectronics to achieve flexibility and stretchability and improve the performance of optoelectronics for biomedical application.
Skin temperature influence on transcutaneous carbon dioxide (CO2) conductivity and skin blood flow in healthy human subjects at the arm and wrist

Abstract:Objective: present transcutaneous carbon dioxide (CO 2 )—tcpCO 2 —monitors suffer from limitations which hamper their widespread use, and call for a new tcpCO 2 measurement technique. However, the progress in this area is hindered by the lack of knowledge in transcutaneous CO 2 diffusion. To address this knowledge gap, this study focuses on investigating the influence of skin temperature on two key skin properties: CO 2 permeability and skin blood flow. Methods: a monocentric prospective exploratory study including 40 healthy adults was undertaken. Each subject experienced a 90 min visit split into five 18 min sessions at different skin temperatures—Non-Heated (NH), 35, 38, 41, and 44°C. At each temperature, custom sensors measured transcutaneous CO 2 conductivity and exhalation rate at the arm and wrist, while Laser Doppler Flowmetry (LDF) assessed skin blood flow at the arm. Results: the three studied metrics sharply increased with rising skin temperature. Mean values increased from the NH situation up to 44°C from 4.03 up to 8.88 and from 2.94 up to 8.11 m·s −1 for skin conductivity, and from 80.4 up to 177.5 and from 58.7 up to 162.3 cm 3 ·m −2 ·h −1 for exhalation rate at the arm and wrist, respectively. Likewise, skin blood flow increased elevenfold for the same temperature increase. Of note, all metrics already augmented significantly in the 35–38°C skin temperature range, which may be reached without active heating— i.e. only using a warm clothing. Conclusion: these results are extremely encouraging for the development of next-generation tcpCO 2 sensors. Indeed, the moderate increase (× 2) in skin conductivity from NH to 44°C tends to indicate that heating the skin is not critical from a response time point of view, i.e. little to no skin heating would only result in a doubled sensor response time in the worst case, compared to a maximal heating at 44°C. Crucially, a skin temperature within the 35–38°C range already sharply increases the skin blood flow, suggesting that tcpCO 2 correlates well with the arterial paCO 2 even at such low skin temperatures. These two conclusions further strengthen the viability of non-heated tcpCO 2 sensors, thereby paving the way for the development of wearable transcutaneous capnometers.

Abstract:A novel method (Sophia) is presented to track oxygen saturation changes in a controlled environment using an RGB camera placed approximately 1.5 m away from the subject. The method is evaluated on five healthy volunteers (Fitzpatrick skin phenotypes II, III, and IV) whose oxygen saturations were varied between 80% and 100% in a purpose-built chamber over 40 minutes each. The method carefully selects regions of interest (ROI) in the camera image by calculating signal-to-noise ratios for each ROI. This allows it to track changes in oxygen saturation accurately with respect to a conventional pulse oximeter (median coefficient of determination, 0.85).
