Abstract:This Perspective is dedicated to the 25th Anniversary of Biomacromolecules. It provides a personal view on the developing field of the polymer and biology interface over the 25 years since the journal was launched by the American Chemical Society (ACS). This Perspective is meant to bridge an article published in the first issue of the journal and recent bioinspired developments in the laboratory of the corresponding author. The discovery of supramolecular spherical helices self-organizing into Frank-Kasper and quasicrystals as models of icosahedral viruses, as well as of columnar helical assemblies that mimic rodlike viruses by supramolecular dendrimers, is briefly presented. The transplant of these assemblies from supramolecular dendrimers to block copolymers, giant surfactants, and other self-organized soft matter follows. Amphiphilic self-assembling Janus dendrimers and glycodendrimers as mimics of biological membranes and their glycans are discussed. New concepts derived from them that evolved in the in vivo targeted delivery of mRNA with the simplest one-component synthetic vector systems are introduced. Some synthetic methodologies employed during the synthesis and self-assembly are explained. Unraveling bioinspired applications of novel materials concludes this brief 25th Anniversary Perspective of Biomacromolecules.

An Innovation 10 Years in the Making: The Stories in the Pages of ACS Medicinal Chemistry Letters

Abstract:Innovation in medicinal chemistry has been at the heart of ACS Medicinal Chemistry Letters since the journal's founding 10 years ago. In his inaugural editorial, Editor-in-Chief Dennis Liotta laid out a vision for the journal to become the "premier international journal for rapid communication of cutting-edge studies," and, after 10 years, it has become exactly that. The great hope of drug discovery scientists is that their innovations will lead to new therapeutics to treat unmet medical needs. In the spirit of innovation and in celebration of the recent 10th anniversary of ACS Med. Chem. Lett., we highlight five therapeutics that were first reported or first comprehensively characterized within ACS Med. Chem. Lett.. This overview also serves to introduce the expansion of the scope of the Innovations article type to include Topical Innovations. With this extension, the journal hopes to provide a forum to showcase concise (rather than comprehensive) reviews of topics that are both timely and of great interest to the medicinal chemistry community. Moreover, these articles will emphasize the next steps to move the field toward new areas of interest in medicinal chemistry. Appropriate topics might include case studies of clinical candidates or approved drugs, new assay technologies in drug discovery, novel target classes, and innovative new approaches towards modulation of human physiology. Since its founding 10 years ago, ACS Med. Chem. Lett. has established itself as a venue for the rapid communication of studies in medicinal chemistry and drug discovery. There have been several drugs and clinical candidates that were first reported or first comprehensively characterized in ACS Med. Chem. Lett. In celebration of the 10th anniversary of ACS Med. Chem. Lett. this Topical Innovations article highlights five of these compounds: Ivosidenib, Siponimod, Glasdegib, Parsaclisib, and Dabrafenib.

Abstract:Supramolecular materials held together by noncovalent interactions, such as hydrogen bonding, host-guest interactions, and electrostatic interactions, have great potential in material science. The unique reversibility and adaptivity of noncovalent intreractions have brought about fascinating new functions that are not available by their covalent counterparts and have greatly enriched the realm of functional materials. This review article aims to highlight the very recent and important progresses in the area of functional supramoleuclar materials, focusing on adaptive mechanical materials, smart sensors with enhanced selectivity, soft luminescent and electronic nanomaterials, and biomimetic and biomedical materials with tailored structures and functions. We cannot write a complete account of all the interesting work in this area in one article, but we hope that it can in a way reflect the current situation and future trends in this prosperously developing area of functional supramolecular materials.

Abstract:Covering: 1998 up to the end of 2023 Since its initial disclosure in 1951, the Kornblum DeLaMare rearrangement has proved an important synthetic transformation and has been widely adopted as a biomimetic step in natural product synthesis. Utilising the base catalysed decomposition of alkyl peroxides to yield a ketone and alcohol has found use in many syntheses as well as a key strategic step, including the unmasking of furans, as a biomimetic synthetic tool, and the use of the rearrangement to install oxygen enantioselectively. Since ca. 1998, its impact as a synthetic transformation has grown significantly, especially given the frequency of use in natural product syntheses, therefore this 25 year time period will be the focus of the review.

Biocatalysis as a Versatile Tool for Macrolactonization: Comparative Evaluation of Catalytic and Stoichiometric Approaches

Abstract:Macrolactonization is a challenging process where high dilution and temperatures can extend reaction times and promote reagent degradation. Biocatalysis is a versatile strategy for synthesis but not traditionally associated within the toolbox of organic chemists for macrocyclization. Macrolactonization has been investigated using modern methods employing both catalysis and stoichiometric activation strategies on 20 different substrates with differing ring sizes and types (cyclophanes, macrolactones, macrodiolides) and structural features at the reaction site (central vs. planar chirality; primary vs. secondary alcohols). The data demonstrates that of all protocols examined, the biocatalytic route was superior, providing the highest average yields across all classes of macrocycles studied. From the stoichiometric activation strategies investigated, the Yamaguchi macrolactonization was the most versatile in terms of ring size and nature. Despite the advantages of biocatalytic macrolactonization, advances in developing supported, versatile non-enantioselective lipases would actually represent a useful tool in molecular synthesis .
