Abstract:Carbon monoxide (CO) can be endogenously produced in mammalian cells and regarded as an essential cell signaling molecule in the body. And it is closely related with a range of physiological and pathological processes. Although some fluorescence probes for tracking CO are reported, near-infrared (NIR) fluorescence probe for CO with a large Stokes shift remains scarce. Herein, a NIR fluorescence probe named DCX-CO is designed and synthesized. In the probe, the allyl formate group is chosen as recognizing moiety and benzopyranonitrile-xanthene-based dye is selected as fluorophore. Upon the addition of CO, the probe shows NIR emission at 762 nm with a large Stokes shift (164 nm). Moreover, the probe displays high sensitivity for CO with 16-fold fluorescence enhancement and excellent selectivity to CO over other potential interference in biological sample. Based on the low cytotoxicity, the probe can be used for the detection of exogenous and endogenous CO in HepG2 cells and HCT116 cells. In addition, taking advantage of NIR emission with a large Stokes shift, DCX-CO is applied for monitoring CO in vivo.

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.

Abstract:Carbon monoxide (CO), as one of significant gas transmitter, is closely associated with a variety of physiological and pathological processes. Although plenty of fluorescent probes have been prepared for detecting CO, most of them suffer from water-soluble fluorophores and short emission wavelength, which tends to diffuse and is limited to apply in vivo. Herein, a novel water-soluble fluorescent probe (HPQ-MQ-CO) is prepared to detect CO by releasing a precipitating fluorochrome (HPQ-MQ-OH), which is developed by introducing the 1-ethyl-2-methylquinoline group into HPQ to obtain long emission wavelength and good diffusion resistant ability. Allyl formate, as the identification unit of CO, has good water solubility and quenches the fluorescence of HPQ-MQ-CO. When the probe reacts with CO and Pd2+, an long-emission and solid-state fluorescence signal at 650 nm can be observed, which is based on excited-state intramolecular proton transfer (ESIPT) mechanism. When the concentration of CO is raised to 100.0 μM, the fluorescence is increased 29 times, indicating the sensitivity of the probe. Moreover, this probe shows prominent selectivity for CO compared with other interfering species. Given these advantages, HPQ-MQ-CO can be used for CO detection in HepG2 cells and zebrafish by in-situ and long-term fluorescence imaging. In addition, this probe can monitor the up-regulation of CO in HepG2 cells and zebrafish during drug-induced liver injury (DILI).

Abstract:In this paper, we constructed a label-free ratiometric fluorescence nanoprobe for ascorbic acid (AA) on the basis of the change in dual-emission signals by the specific reaction between cobalt oxyhydroxide (CoOOH) and AA. CoOOH nanoflakes were used as the recognition unit while polymer dots (PFO dots) and gold nanoclusters (AuNCs) served as luminescent units of the nanoprobe. After electrostatic assembly of PFO dots and AuNCs on the surface of CoOOH nanoflakes, the energy transfer between the PFO dots and CoOOH could occur and thereby quench the fluorescence of the PFO dots. When AA was present, CoOOH nanoflakes were reduced to Co2+, resulting in a recovery of the fluorescence of the PFO dots. In the meantime, the generated Co2+ could combine with the surface ligands of AuNCs, accompanied by a reduction in the luminescence intensity of the AuNCs. Therefore, the fluorescence intensity ratio of PFO dots to AuNCs would be related to the concentration of AA. The nanoprobe enabled highly sensitive analysis of AA with a detection limit of 1.9 μM and showed great performance in human serum samples and living cells. This report may not only endow a CoOOH-based nanoprobe with improved ability but also offer a novel strategy for the construction of ratiometric nanoprobes for AA and have potential applications in research into and clinical diagnosis of AA-related diseases.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:The sensitive and accurate detection of intracellular gasotransmitter carbon monoxide (CO) is of great significance for unraveling its diverse cellular functions. Herein, we report a simple highly specific and sensitive 4-hydroxynaphthalimide-based ratiometric fluorescent probe Ratio-CO for monitoring CO levels in living cells. Probe Ratio-CO could quantitatively detect CO in the range of 0-50 M by the ratiometric fluorescence spectroscopy method and the detection limit was measured to be about 17.9 nM. Additionally, probe Ratio-CO has been proven to possess preeminent selectivity towards CO over other bioactive species. Most importantly, the excellent response properties of probe Ratio-CO enable its applications in the monitoring of the fluctuations of CO levels in living cells by the ratiometric fluorescence imaging technique, and we thus anticipate that this probe would be a novel tool for further elucidating the biological functions of CO in living systems.

Abstract:As an important gaseous signal molecule, carbon monoxide (CO) plays a major role in multiple physiological and pathological processes. Therefore, it is significantly meaningful to develop fluorescent probes with features such as in situ imaging to detect CO in biological samples. However, the reported CO fluorescent probes still suffer from low sensitivity, poor selectivity, and low water solubility. In particular, due to the very low concentration of CO in organisms, these probes must have high sensitivity to achieve the detection of CO. To address this problem, we constructed a CO fluorescence probe ( HS-CO ) with a reasonable design, which can greatly increase the rigid plane of the probe through spontaneous intramolecular cyclization after the reaction with CO. The substantial increase in the fluorescence quantum yield after the reaction enabled it to achieve a strong change in the fluorescence signal and exhibit high sensitivity for detecting CO (LOD = 4.5 nM). Importantly, probe HS-CO was successfully used to detect endogenous and exogenous CO in HeLa cells. In addition, probe HS-CO successfully detected CO in zebrafish. These results together identified the great potential of the probe as a tool for functional analysis of CO in organisms.

Abstract:Carbon monoxide (CO) has captivated great attention in part due to the discovery of the therapeutic and cell-signalling role that CO plays in biological systems. The ever-increasing interest in CO has resulted in the development of rapid and facile approaches for the sensitive and selective detection of CO, which still remains challenging. Herein, the first example of a colorimetric and near-infrared fluorescent dual probe for carbon monoxide (CO) has been developed by assembling an allyl chloroformate moiety with a naphthofluorescein fluorophore, which enables the label-free and visual detection of CO. The living cell imaging results indicate that the probe shows great potential in sensing intracellular CO.

Abstract:A colorimetric and fluorescent dual probe for CO is designed and synthesized. The probe is capable of sensitive and specific detection of CO with naked-eye through colorimetric assay in phosphate buffer from colorless to yellow, which also features 75-fold turn-on fluorescence enhancement upon exciting at 480 nm with the detection limit of 26.3 nM. Abstract:Exploring techniques for monitoring the intracellular signaling molecule carbon monoxide (CO) in biosystems is important to help understand its various cellular functions. Therefore, a simple long-wavelength colorimetric fluorescent probe LW-CO was designed for selectively and sensitively detecting intracellular CO in living systems. Probe LW-CO is ultrasensitive and can track CO levels in the range of 0–1 μM, with a detection limit of about 3.2 nM. Additionally, the obvious color changes of probe LW-CO with CO (yellow to pink) provide a convenient way for on-site detection of CO with the naked eye. Probe LW-CO was applied to track the exogenous levels of CO in RAW264.7 cells. Probe LW-CO proved to be an efficient method for investigating various cellular functions of CO.

Abstract:Carbon monoxide (CO), a significant gas transmitter, plays a vital role in the intricate functioning of living systems and is intimately linked to a variety of physiological and pathological processes. To comprehensively investigate CO within biological system, researchers have widely adopted CORM-3, a compound capable of releasing CO, which serves as a surrogate for CO. It aids in elucidating the physiological and pathological effects of CO within living organisms and can be employed as a therapeutic drug molecule. Therefore, the pivotal role of CORM-3 necessitates the development of effective probes that can facilitate the visualization and tracking of CORM-3 in living systems. However, creating fluorescent probes for real-time imaging of CORM-3 in living species has proven to be a persisting challenge that arises from factors such as background interference, light scattering and photoactivation. Herein, the BNDN fluorescent probe, a brand-new near-infrared is proposed. Remarkably, the BNDN probe offers several noteworthy advantages, including a substantial Stokes shift (201 nm), heightened sensitivity, exceptional selectivity, and an exceedingly low CORM-3 detection limit (0.7 ppb). Furthermore, the underlying sensing mechanism has been meticulously examined, revealing a process that revives the fluorophore by reducing the complex Cu2+ to Cu+. This distinctive NIR fluorescence "turn-on" character, coupled with its larger Stokes shift, holds great promise for achieving high resolution imaging. Most impressively, this innovative probe has demonstrated its efficacy in detecting exogenous CORM-3 in living animal. It is important to underscore that these endeavors mark a rare instance of a near-infrared probes successfully detecting exogenous CORM-3 in vivo. These exceptional outcomes highlighted the potential of BNDN as a highly promising new tool for in vivo detection of CORM-3. Considering the impressive imaging capabilities demonstrated by BNDN presented in this study, we anticipate that this tool may offer a compelling avenue for shedding light on the roles of CO in future research endeavors.

Abstract:Carbon monoxide (CO) is one of the signaling molecules that are ubiquitous in humans, which involves in the regulation of human physiology and pathology. In this work, the probe PEC was designed and synthesized based on BODIPY fluorophore that can selectively detect CO through reducing the nitro group to amino group, resulting in a "turn-on" fluorescence response with a simultaneous increase in the concentration of CO. The response is selective over a variety of relevant reactive free radicals, ions, and amino acid species. PEC has the advantages of good stability, good water solubility, and obvious changes in fluorescence signals. In addition, PEC can be used to detect and track endogenous CO in living cells.

Abstract:Carbon monoxide (CO) is a highly emitted atmospheric pollutant that can pose a serious health risk when its concentration is too high. However, endogenous CO plays an important role in maintaining human health. The development of tools capable of detecting CO in environmental samples and biological samples is therefore essential. Although CO fluorescent probes have been reported, their further applications are limited by factors such as low sensitivity and emission wavelength. Here, we have constructed a CO probe with simple structure, high sensitivity, good selectivity and near-infrared (NIR) emission characteristics. The probe uses the dicyanoisophorone derivative as fluorophore and connects allyl moiety as the reaction site. After the response, the intramolecular charge transfer (ICT) process turns on and releases strong near-infrared fluorescence at 675 nm. The linear range of fluorescence intensity of the probe for CO concentration is 1-10 mu M and the detection limit is as low as 58 nM. The high sensitivity and NIR emission give the probe excellent imaging of cellular exogenous and endogenous CO. Moreover, the probe can also be used to image CO in zebrafish. There is reason to believe that the probe can be a powerful tool for detecting CO concentrations in environmental samples and for analyzing the biological role of CO in organisms.
